14. Study on Ultrasonic Assisted Solvent Extraction of Total Flavonoids from the Calyx of Physalis alkekengi.

Abstract

The calyx of Physalis alkekengi is rich in various bioactive components. In this study, ultrasonic assisted ethanol solvent was used to extract the total flavonoids using calyx of Physalis alkekengi as materials, and the effects of ultrasonic power, extraction temperature, ultrasonic time, ethanol volume fraction and solid -liquid ratio on the yields of total flavonoids were investigated, then the L16(45) orthogonal test was used to further optimize the extraction parameters on the basis of single factor experiments, in order to efficiently and fully extract the total flavonoids. The results showed that the optimal extraction process was determined as follows: ultrasonic power of 180 W, extraction temperature of 60 °C, 60% ethanol solution soaked for 24 h with solid-liquid ratio of 1:20(g/mL), and ultrasonic time of 50 min. Under these conditions, the yield of total flavonoids was 2.982%. The process in this paper is simple with low preparation cost, which is convenient for industrial production and helpful to the comprehensive utilization and development for calyx of Physalis alkekengi. In the past 3 years, the COVID-19 pandemic has resulted in a dramatic increase in the occurrence of psychological symptoms such as depression, anxiety, and stress in global populations. Therefore, it is important to help pregnant women cope with major public emergencies and improve their physical and mental health.Objective: To investigate the status and influencing factors of anxiety, insomnia, and psychological stress disorder in pregnant women with COVID-19 to provide a theoretical basis for psychological intervention in pregnant women in the case of public health emergency events.Patients and methods: A cross-sectional survey was conducted with 435 pregnant women from multiple tertiary hospitals in Zhengzhou from November 6 to December 6, 2022. 40. Stochastic Optimal Intervention for Robot-Assisted Crowd Evacuation

Abstract

Robot-assisted interventions are often provided for crowd evacuation to prevent accidents and keep crowds safety, which is currently a hot topic of research. However, the existing work has ignored the impact of crowd motion chaos which is a fundamental factor determining the effective evacuation. To assist the crowd evacuation more efficiently and reduce the intervention costs, robots need to identify destructive individuals based on the randomly evolved chaos and intervene in them specifically. To this end, we propose a stochastic optimal intervention method to provide the optimal strategies for robot-assisted crowd evacuation. First, the definition of chaos is introduced to quantify the chaotic degree of crowd movement. Then, we study the stochastic evolution of chaos by exploring the chaos state transition with marked temporal point process (MTPP) and stochastic differential equations (SDEs). Next, the robot-assisted intervention is formulated as a stochastic optimal intervention problem that seeks to maximize the intervention utility. Fortunately, a closed-form solution is obtained by using the Bellman’s principle of optimality and solving the derived Hamilton-Jacobi-Bellman (HJB) equation. At last, we develop an event-driven crowd motion system to simulate crowd evacuation process. The simulation results show that our method can prevent the occurrence of chaos effectively, and improve the efficiency of crowd evacuation significantly. The proposed method is expected to provide guidance for emergency management.

42. Kaempferol induces mitochondrial dysfunction and mitophagy by activating the LKB1/AMPK/MFF pathway in breast precancerous lesions.

Abstract

Kaempferol has been suggested to be an effective anticancer agent in several malignant tumors. However, its function and mechanisms in breast precancerous lesions remain largely elusive. Here, we showed that kaempferol induced excessive mitochondrial fission and mitochondrial damage with activated mitochondrial fission factor (MFF)‐mediated dynamin‐related protein (DRP) 1 mitochondrial translocation. As a result, the PTEN‐induced putative kinase 1 (PINK1)/Parkin signaling pathway was activated, accompanied by excessive mitophagy and reduced mitochondrial mass in cells. We also revealed that kaempferol‐induced lethal mitophagy contributed to inhibiting breast precancerous lesion growth in vitro and in vivo. Furthermore, we verified serine/threonine kinase 11 (STK11/LKB1)/AMP‐activated protein kinase (AMPK) pathway deficiency in breast precancerous lesions. Moreover, LKB1/AMPK pathway reactivation by kaempferol was required for excessive mitochondrial fission and lethal mitophagy. Taken together, our findings shed new light on the molecular mechanisms related to breast cancer prevention by kaempferol and provide evidence for its potential clinical application.
