Objective To invettigate the therapeutic effects of recombinant adenovirus (Ad)-mediated expresiion of NT4p53(N15) Ant, which is specifically cytotoxical to tumor cells, against transplanted H22 hepatoma carcinoma cell in ICR. Methods Totally 16 ICR mice were randomly divided into two groups: infection wíth Ad-NT4p53(N15) Ant (8 in treatment group) and infection with blank adenovirus (8 in control group). Mice in both groups were infected in peritoneal cavity with Ad-NT4p53(N15) Ant or blank adenovirus, respectively, in advance. Then, H22 cell suspensions were injected into the abdominal cavity of ICR mice to establish the models of extensive cancer metastasis to abdominal cavity. The efficacy of Ad-NT4p53(N15) Ant on tumors was evaluated according to survival time of the mice, tumor weight, tumor inhibition rate, morphous of ascite cells, and apoptosis and necrosis rates of tumor cells. Results The models of extensive cancer metastasis to abdominal cavity were successfully established. Compared to the control group, Ad-NT4p53 (N15) Ant significantly prolonged the survival time, reduced tumor weight, decreased the number of tumor cells wth pathologic karyokinesis, and increased apoptosis and necrosis rates of tumor cells. Conclusion Ad-NT4p53 (15) Ant can induce apoptosis of the H22 tumor cells transplanted in IRC mice, thus inhibiting tumor growth and prolong survival of the mice. [ABSTRACT FROM AUTHOR]

Objective: This study was to examine the anti-inflammatory effect of sappanone A on interleukin- (IL-) 1 β -stimulated osteoarthritis (OA) chondrocytes., Methods: Chondrocytes were pretreated with sappanone A for 2 h before subsequent IL-1 β stimulation. The mRNA expression levels of iNOs, COX-2, aggrecan, and collagen-II were measured with qRT-PCR. The levels of TNF- α , IL-6, IL-8, MMP-3, and MMP-13 were determined by ELISA. The protein levels of iNOs, COX-2, ADAMTS-4, ADAMTS-5, aggrecan, collagen-II, p-p65, p65, I κ B α , Nrf2, and HO-1 were assessed by Western blot., Results: Sappanone A inhibited the IL-1 β -stimulated production of NO, PGE2, iNOS, COX-2, TNF- α , IL-6, and IL-8 in OA chondrocytes. In addition, sappanone A suppressed the expression of MMP-3, MMP-13, ADAMTS-4, and ADAMTS-5 in IL-1 β -stimulated OA chondrocytes. The degradation of ECM components was reversed by sappanone A. Objective: To explore the expression and clinical significance of VEGF, IL-17, β2-MG and IL-35 in patients with multiple myeloma., Methods: A total of 83 patients with multiple myeloma (MM) from January 2012 to December 2016 were enrolled in MM group, 36 healthy subjects were enrolled in control group. The levels of IL-17, IL-35 and VEGF in serum were detected by ELISA. The levels of β2-MG in serum were measured by radioimmunoassay. The differences of different indexes between 2 groups were compared., Results: The serum levels of IL-17, VEGF and β2-MG in serum of III stage were higher than that in II stage, which was higher than that in I stage and control group (P<0.05). The levels of IL-35 in the control group were significantly higher than those in the I,II,III stage group (P<0.05). The levels of IL-17, VEGF and β2-MG in serum of progress period were higher than those in stable phase and control group, level of IL-35 in serum of control group was significantly higher than that in the stable phase and progress period group (P<0.05). The correlation analysis showed that the level of serum IL-17 positively correlated with VEGF, β2-MG expression (r=0.65, 0.58, P<0.05); and the serum IL-17 levels were negatively correlated with IL-35 levels (r=-0.42, P<0.05)., Conclusion: The anomalous levels of IL-17, IL-35, IVEGF and β2-MG expressions correlate with the progression and prognosis of patients with multiple myeloma.
