Summary: The integrity of wheat (Triticum aestivum) production is increasingly jeopardized by the fungal pathogen Blumeria graminis f. sp. tritici (Bgt), particularly amid the vicissitudes of climate change.Here, we delineated the role of a wheat transcription factor, TaNAC1, which precipitates cellular apoptosis and fortifies resistance against Bgt. Utilizing BiFC, co‐immunoprecipitation, protein quantification, luciferase report assays, we determined that cytoplasmic TaNAC1‐7A undergoes phosphorylation at the S184/S258 sites by TaCDPK20, facilitating its nuclear translocation. This migration appears to prime further phosphorylation by TaMPK1, thereby enhancing transcriptional regulatory activity. Notably, the apoptotic activity of phosphorylated TaNAC1‐7A is negatively modulated by the nuclear protein phosphatase PP2Ac.Furthermore, activation of TaNAC1 phosphorylation initiates transcription of downstream genes TaSec1a and TaCAMTA4, through binding to the C[T/G]T[N7]A[A/C]G nucleic acid motif. Suppression of TaNAC1, TaCDPK20, and TaMPK1 in wheat compromises its resistance to Bgt strain E09, whereas overexpression of TaNAC1 and silencing of PP2Ac markedly elevate resistance levels.Our results reveal the pivotal role of TaNAC1 in basal resistance which is mediated by its effects on homotypic fusion, vacuolar protein sorting, and the expression of defense‐related genes. The morphological analysis of bulk heterojunction (BHJ) active layer stands as a critical imperative for advancing the performance of future organic solar cells. Conventional characterization tools employed for morphological investigation often require substantial resources, both in cost and physical space, thereby imposing restraints on research endeavors in this domain. Here, we extend the application of charge carrier transport characterization beyond conventional mobility assessments, utilizing it as a table-top method for preliminary morphological screening in organic thin films. The investigation focuses on several high-performance BHJ systems that utilize typical "Y" non-fullerene acceptors. It involves in-depth transport studies, including temperature- and field-dependent transport characterizations. The resulting transport data are analyzed in detail using the Gaussian disorder model to extract key transport parameters, specifically the high-temperature limited mobility (μ∞) and positional disorder (∑). Integrating these transport parameters with morphological insights obtained through various characterization tools—including x-ray scattering, sensitive spectroscopy, and quantum chemistry simulation—provides a deep understanding of the intricate interplay between charge transport properties and morphological characteristics. The results reveal explicit relationships, associating μ∞ with the degree of molecular stacking in BHJs and ∑ with the structural disorder in molecule skeleton. Our findings point to the promising potential of utilizing a simple transport characterization technique for the early stage evaluation of thin film packing and geometric properties of organic materials. Background: N6-methyladenosine (m6A) modification is the most abundant type of RNA modification in eukaryotic cells, playing pivotal roles in multiple plant growth and development processes. Yet the potential role of m6A in conferring the trait of male sterility in plants remains unknown. Results: In this study, we performed RNA-sequencing (RNA-Seq) and m6A-sequencing (m6A-Seq) of RNAs obtained from the anther tissue of two wolfberry lines: 'Ningqi No.1' (LB1) and its natural male sterile mutant 'Ningqi No.5' (LB5). Based on the newly assembled transcriptome, we established transcriptome-wide m6A maps for LB1 and LB5 at the single nucleus pollen stage. We found that the gene XLOC_021201, a homolog of m6A eraser-related gene ALKBH10 in Arabidopsis thaliana, was significantly differentially expressed between LB1 and LB5. We also identified 1642 and 563 m6A-modified genes with hypermethylated and hypomethylated patterns, respectively, in LB1 compared with LB5. We found the hypermethylated genes significantly enriched in biological processes related to energy metabolism and lipid metabolism, while hypomethylation genes were mainly linked to cell cycle process, gametophyte development, and reproductive process. Among these 2205 differentially m6A methylated genes, 13.74% (303 of 2205) were differentially expressed in LB1 vis-à-vis LB5. Conclusions: This study constructs the first m6A transcriptome map of wolfberry and establishes an association between m6A and the trait of male sterility in wolfberry. [ABSTRACT FROM AUTHOR]
