Diving vertebrates represent unique models for the study of the physiological responses to reactive oxygen species (ROS) production and oxidative stress because of their adaptability to cope with dive-derived ROS production. We hypothesized that in the skeletal muscle of a diving mammal, the hooded seal (Cystophora cristata (Erxleben, 1777)), ROS production increases with maturation but the accumulation of oxidative damage does not. To test this, we analyzed the tissue capacity to produce ROS, the accumulation of oxidative damage, and the activity and protein content of the cooper, zinc, and manganese dependent superoxide dismutases (Cu, ZnSOD, MnSOD) in skeletal muscle from neonates, weaned pups, and adult hooded seals. Our results showed higher tissue capacity to produce ROS, higher Cu, ZnSOD and MnSOD activities, and higher MnSOD protein content in adult seals than in pups. No differences in oxidative damage to lipids, proteins, or DNA were detected among groups. Results suggest that increased SOD activity likely counters the oxidative damage commonly associated with increased ROS production. These findings highlight the unusual tolerance of skeletal muscle of seals to increased ROS production. Afin de tester l'hypothese, nous analysons la capacite du tissu a produire des ROS, l'accumulation de dommages oxydatifs, ainsi que l'activite et le contenu proteinique des superoxydes dismutases dependantes du cuivre, du zinc et du manganese (Cu, ZnSOD, MnSOD) dans le muscle squelettique chez des phoques a capuchon nouveau nes, des petits sevres et des adultes. Bisphenol A is one of the most used components of the polycarbonate plastic industry in the word. This contaminant has disrupting effect in cells in in vitro and in vivo in fish. This study evaluated for the first time the cytotoxicity, oxidative stress and apoptosis induced by bisphenol A (BPA) in head-kidney and spleen leukocytes isolated from Pacific red snapper Lutjanus peru. Head-kidney and spleen leukocytes were exposed to 100, 1000 and 10,000 µg/mL of BPA at 2 and 24 h. Results showed cytotoxicity of BPA at 1000 and 10,000 µg/mL. Cell viability > 80% was observed in leukocytes exposed to 100 µg/mL for 2 h; thus, this concentration was selected for the remainder of the study. Reactive oxygen species (ROS) production, analyzed by DCF-DA and NBT assays, significantly increased in those leukocytes exposed to BPA compared to controls after 2 or 24 h. Superoxide dismutase and catalase activities increased in head-kidney leukocytes after 24 h of BPA exposure. Apoptosis was inferred from caspase (casp-1 and casp-3), granzyme A (granz-A) and perforin 1 (perf-1) gene expression, which was significantly up-regulated, at 2 h BPA exposure in head-kidney leukocytes, and from granz-A and perf-1, which were up-regulated, after 24 h BPA exposure in spleen leukocytes. Short cytoplasmic prolongations and membrane blebs, suggestive of apoptosis, were observed by scanning electron microscopy. Freezing, dehydration, salinity variations, hypoxia or anoxia are some of the environmental constraints that many organisms must frequently endure. Organisms adapted to these stressors often reduce their metabolic rates to maximize their chances of survival. However, upon recovery of environmental conditions and basal metabolic rates, cells are affected by an oxidative burst that, if uncontrolled, leads to (oxidative) cell damage and eventually death. Thus, a number of adapted organisms are able to increase their antioxidant defenses during an environmental/functional hypoxic transgression; a strategy that was interpreted in the 1990s as a "preparation for oxidative stress" (POS). Since that time, POS mechanisms have been identified in at least 83 animal species representing different phyla including Cnidaria, Nematoda, Annelida, Tardigrada, Echinodermata, Arthropoda, Mollusca and Chordata. Coinciding with the 20th anniversary of the postulation of the POS hypothesis, we compiled this review where we analyze a selection of examples of species showing POS-mechanisms and review the most recent advances in understanding the underlying molecular mechanisms behind those strategies that allow animals to survive in harsh environments., (Copyright © 2019 Elsevier Inc. Aging is associated with a gradual decline in cognitive and motor functions, the result of complex biochemical processes including pre- and posttranslational modifications of proteins. Sirtuins are NAD(+) dependent protein deacetylases. These enzymes modulate the aging process by lysine deacetylation, which alters the activity and stability of proteins. Exercise can increase mean life-span and improve quality of life. Data from our laboratories revealed that 4 weeks of treadmill running improves performance in the Morris Maze test for young (4 months, old) but not old (30 months, old) male rats, and the exercise could not prevent the age-associated loss in muscle strength assessed by a gripping test. The positive correlation between protein acetylation and the gripping test suggests that the age-dependent decrease in relative activity of SIRT1 in the cerebellum impairs motor function. Similarly to the acetylation level of total proteins, the acetylation of ά -tubulin is also increased with aging, while the effect of exercise training was not found to be significant. Moreover, the protein content of nicotinamide phosphoribosyltransferase, one of the key enzymes of NAD biosynthesis, decreased in the young exercise group. These data suggest that aging results in decreased specific activity of SIRT1 in cerebellum, which could lead to increased acetylation of protein residues, including ά-tubulin, that interfere with motor function.

The aim of this study was to evaluate the use of metallothionein (MT) concentrations in tissues of the clam Megapitaria squalida as biomarkers of environmental cadmium (Cd) pollution from phosphorite enrichments in the marine environment, which resulted from mining activities in La Paz Bay, Baja California Sur, Mexico. Cd and MT were quantified in gills, digestive gland, and kidney of clams exposed to 0.2 or 0.5 mg Cd l(-1) for 10, 20, or 30 days. In addition, clams from four strategically selected natural sites of La Paz Bay were collected for analysis. In tissues of bioassayed and untreated clams, the gradient of Cd concentrations was digestive gland>>gills>kidney, whereas that of MT was digestive gland>gills>kidney. Digestive gland of the clams exposed to 0.5 mg Cd l(-1) for 30 days showed the highest concentrations of Cd (16.3+/-3.9 microg Cd g(-1)). The highest statistically significant MT concentrations were found in digestive gland at 10 days of exposure to Cd. In the untreated clams, one of the highest Cd concentrations, but not MT levels, was found in digestive glands of the organisms collected from the area close to phosphorite mining activities. For environmental monitoring, MT levels in digestive gland can be used as a first approximation of the presence of high levels of divalent metals in the environment. However, in this study, MT levels did not correlate with high Cd levels in clams that had been collected from areas associated with phosphorite enrichment.

Coral reefs are impacted by a range of environmental variables that affect their growth and survival, the main factors being the high irradiance and temperature fluctuations. Specimens of Pocillopora capitata Verrill 1864 were exposed to photosynthetically active radiation (PAR) and ultraviolet radiation (UVR) for 32 h under laboratory conditions. We examined lipid peroxidation (MDA), antioxidant enzyme activities (SOD, CAT, GPx and GST), chlorophyll a (Chl a), carotenoid pigments (CPs), mycosporine-like amino acids (MAAs), and expulsion of zooxanthellae. Our results revealed that corals exposed to UVR had relatively low levels of carotenoids and antioxidant enzyme activities compared to those exposed to PAR, as well as lower CPs/Chl a ratios. Although MAAs and CPs are rapidly produced as non-enzymatic antioxidants in response to UVR in corals, these were not sufficient, even in the dark phase of the experiment, to mitigate the damage caused by formation of reactive oxygen species (ROS), which caused breakdown of the symbiotic relationship between the zooxanthellae and the host animal to an extent 33 times greater than in the PAR treatment. In this study, it could be possible to distinguish that, parallel to the short-term adjustments, such as the amount of pigment in the algae or the sensitivity of the photosynthetic response reported in other species of coral, P. capitata exhibits at the enzymatic level a series of responses oriented to resist the effects derived from the propagation of ROS and, thus, to adapt to and maintain its reproductive capacity in shallow oceanic environments that commonly exhibit high UVR levels. Nevertheless, as a result of the inappropriate location of the artificial intercommunication structure of the Juluapan Lagoon with respect to the arrecifal area of study and therefore of the tides influence, other variables, such as the changes in short-term in turbidity, sediment inputs, nutrients, temperature and osmolarity, can act in combination and cause irreversible damage. The implementation of a management plan for the coralline reefs of the Mexican Pacific coast is required.

In isolated perfused guinea pig hearts, coronary flow causes a positive inotropic effect [positive coronary flow-induced effect (+CFIE)] that could be altered by dextrans (Dx) in the coronary perfusion solution. To test this possibility, Dx of 20, 40, 70, and 500 kDa were infused and found to modulate +CFIE; however, when Dx infusion was terminated, the effect persisted, i.e., was irreversible/nonwashable, suggesting that Dx may bind to luminal endothelial lectinic structures. This hypothesis was tested when Dx [with fluorescent traces (D*)] bound to the vessel wall was hydrolyzed by dextranase infusion and washout of D* fragments completely reverted the +CFIE, and it was found that bound D* to be displaced by free Dx required concentrations 50-100 times that used during binding. In addition, dose-response curves for Dx on +CFIE show that the higher the Dx molecular mass, the lesser the concentration required to have an effect. Because a large Dx molecule has a greater number polymeric glucose branches, it can bind to a larger number of endothelial lectinic sites, requiring a lower concentration to affect +CFIE. Our results suggest that luminal endothelial lectinic structures are part of the flow-sensing assembly.

In this review, we consider comparative aspects of the biology and pathology of oxygen radicals in neurodegenerative disease and how these findings have influenced our concept of oxidative stress. The common definition of oxidative stress is a breach of antioxidant defenses by oxygen radicals leading to damage to critical molecules and disrupted physiology. Inherent in this definition is that oxidative stress is an unstable situation, for if there is net damage, viability of the system decreases with time, leading to disequilibria and death. While this circumstance defines acute conditions, such as stroke and head trauma which result in dysfunction and death, it does not fit physiological situations or chronic diseases closely aligned to normal physiology. Therefore, we propose that oxidative modifications in Alzheimer disease may actually serve as a homeostatic response to stress resulting in a shift of neuronal priority from normal function to basic survival. This phenomenon is comparable to normal physiological conditions of metabolic decrease, such as those seen in hibernation and estivation. Thus, Alzheimer disease could be seen as part of normal aging that includes additional pathology due to inadequate homeostatic response.

We used radioimmunoassay methods to quantify arginine vasopressin (AVP), atrial natriuretic peptide (ANP), and angiotensin II (Ang II) in plasma samples from harbor seals (Phoca vitulina richardsii), Weddell seals (Leptonychotes weddellii), northern elephant seals (Mirounga angustirostris), ringed seals (Phoca hispida), California sea lions (Zalophus californianus), and Steller sea lions (Eumetopias jubatus). Plasma concentrations of AVP, ANP, and Ang II in these pinniped species were within the ranges reported for other vertebrates under resting conditions. However, there were species, geographic and developmental variations in these hormones: Levels of AVP in plasma samples from adult Steller sea lions and harbor seals were higher than in pups of the same species; higher levels of plasma ANP were found in wild captured Alaskan Steller sea lions and in hunted ringed seals; differences in plasma levels of all three hormones were found throughout the geographic distribution of harbor seals and Steller sea lions in Alaska. This is the first report on circulating concentrations of vasoactive hormones in pinnipeds, and demonstrates that further studies are needed to ascertain the natural variability in these levels with the impact of molting, fasting, diving and environmental factors in seals and sea lions.

Prolonged and repetitive breath-hold periods (apnea) during and sleep are a routine component in the ecological physiology of marine mammals. Seals are among the few mammals in which control of heart rate (HR) can be studied independent of respiration, without pharmacochemical manipulation. We hypothesized that the vasoactive hormones angiotensin II (Ang II), arginine vasopressin (AVP), and atrial natriuretic peptide (ANP) were involved in the control of cardiovascular function in seals, and that the relationship was dependent upon input from the respiratory system. Venous plasma samples were collected and electrocardiograms were recorded from Northern elephant seal (Mirounga angustirostris) and Weddell seal (Leptonychotes weddellii) pups during both spontaneous breathing (eupnea) and apnea. Instantaneous HR and simultaneous plasma levels of ANP, AVP, and Ang II from periods of eupnea and apnea were compared. In these seal pups, apnea was associated with bradycardia, increased ANP, and decreased AVP and Ang II. The results support the hypothesis of a complex involvement between the vasoactive hormones and the control of cardiovascular function, and provide evidence for differential levels of control during periods of eupnea and apnea.

We evaluated the plasma concentration of the acute phase protein haptoglobin (Hp) from Steller sea lions (Eumetopias jubatus) and harbor seals (Phoca vitulina) in regions of Alaska (USA) where the populations of these pinnipeds were declining and compared the values with concentrations of Hp from the same species in areas where the populations were stable. Samples were collected from 1992 through 1994 at sites in Southeast Alaska, Prince William Sound, the Gulf of Alaska, and the Aleutian Islands. Significantly higher levels of Hp were found in the samples from the areas of decline compared to those from stable populations. Based on these findings, we propose that one may be able to distinguish these compromised pinniped populations using Hp as a biomedical indicator.
