Abstract

The combination of sulfate radical-mediated advanced oxidation process (SA-AOP) and photocatalysis is researched as a highly efficient strategy for pollutant degradation in recent decades. To systematically study the synergistic effect between photocatalysis and peroxymonosulfate (PMS) induced SA-AOP, graphene oxide (GO) coupled graphitic carbon nitride (CN) composites were synthesized as catalyst. About 98% of bisphenol A was decomposed in CNGO/PMS/light system, which was significantly higher than that in CNGO/PMS or CNGO/light system, demonstrating the strong synergistic effect between photocatalysis and SA-AOP. CNGO was deactivated in catalyst/PMS system, but not in catalyst/PMS/light system, showing that the deactivation of CNGO in the presence of sulfate radicals was inhibited by the photocatalysis. The photogenerated electrons may directly reduce the oxidized GO, or react with the oxidative species which may oxidize GO, and thus maintain the activity of CNGO. In addition, the system temperature was elevated due to the light-to-heat conversion on GO. The increased reaction temperature accelerated the degradation reaction due to the improved the charge separation and molecular collision. This work explains the synergy effect between photocatalysis and SA-AOP from different perspectives, and provides a new idea to improve the reusability of carbonaceous catalyst.

24. Engineered Escherichia colias a Controlled-Release Biocarrier for Electrochemical Immunoassay

Abstract

Micro/nanocarriers hold great potential in bioanalysis for molecular recognition and signal amplification but are frequently hampered by harsh synthesis conditions and time-consuming labeling processes. Herein, we demonstrate that Escherichia coli(Ec) can be engineered as an efficient biocarrier for electrochemical immunoassay, which can load ultrahigh amounts of redox indicators and simultaneously be decorated with detection antibodies via a facile polydopamine (PDA)-mediated coating approach. Compared with conventional carrier materials, the entire preparation of the Ec biocarrier is simple, highly sustainable, and reproducible. Moreover, immune recognition and electrochemical transduction are performed independently, which eliminates the accumulation of biological interference on the electrode and simplifies electrode fabrication. Using human epidermal growth factor receptor 2 (HER2) as the model target, the proposed immunosensor exhibits excellent analytical performance with a low detection limit of 35 pg/mL. The successful design and deployment of Ec biocarrier may provide new guidance for developing biohybrids in biosensing applications.

Abstract

Sulfate radical-based advanced oxidation process is a promising technology to degrade organic pollutants for water remediation. Photo-activation of peroxymonosulfate (PMS) has received increasing attention. From the aspect of solar energy, the utilization of near-infrared (NIR) light which accounts for about 50% of the solar energy in PMS activation is still challenging. Herein, we designed and synthesized few-layered graphite modified graphitic carbon nitride (GrCN) for whole-spectrum photo-activation of PMS. During the reaction, NIR light was converted to heat on few-layered graphite through photothermal effect to accelerate the chemical reactions via raising the system temperature. In addition to the photothermal effect, few-layered graphite also acted as cocatalyst to alter the band structure and promote the charge separation. Therefore, GrCN exhibited excellent activity in the degradation of bisphenol A with the addition of PMS under illumination. In addition, GrCN presented high reusability in the presence of sulfate radicals under illumination.

39. Target-Triggered Aggregation of Modified E. colifor Diagnosis of Ovarian Cancer

Abstract

Bacteria inherently possess the capability of quorum sensing in response to the environment. In this work, we have proposed a strategy to confer bacteria with the ability to recognize targets with quorum-sensing behavior. Meanwhile, we have successfully achieved artificial control over the target-triggered aggregation of Escherichia coli(E. coli) by modifying the bacteria surface in a new way. Furthermore, by making use of green fluorescent protein (GFP) expressed by E. colias the output signal, the aggregation of modified E. colican be observed with the naked eye. Therefore, via the detection of the target, MUC1, an ovarian cancer biomarker, a simple and conveniently operated method to diagnose ovarian cancer is developed in this work. Experimental results show that the developed low-background and enzyme-free amplification method enables the highly sensitive detection of MUC1, achieving a remarkable limit of detection (LOD) of 5.47 fM and a linear detection range spanning from 1 pM to 50 nM and 50 nM to 100 nM, respectively. Clinical samples from healthy donors and patients can give distant assay results, showing great potential for clinical applications of this method.

Abstract

Asperalins represent a novel class of viridicatin natural products with potent inhibitory activities against fish pathogens. In this study, we elucidated the biosynthesis of asperalins in the Aspergillus oryzae NSAR1 heterologous host and identified the FAD-dependent monooxygenase AplB stereoselectively hydroxylates viridicatin to yield a unique 3 R ,4 S configuration. The monomodular NRPS AplJ catalyzes a rare intramolecular ester bond formation reaction using dihydroquinoline as a nucleophile. Subsequent modifications by cytochrome P450 AplF, chlorinase AplN, and prenyltransferase AplE tailor the anthranilic acid portion, leading to the formation of asperalins. Additionally, we explored the potential of AplB for the hydroxylation of viridicatin analogs, demonstrating its relaxed substrate specificity. This finding suggests that AplB could be developed as a biocatalyst for the synthesis of viridicatin derivatives.

Abstract

CsPbX 3 perovskite nanocrystals (NCs), with excellent optical properties, have drawn considerable attention in recent years. However, they also suffer from inherent vulnerability and hydrolysis, causing the new understanding or new applications to be difficultly explored. Herein, for the first time, it is discovered that the phospholipid membrane (PM)-coated CsPbX 3 NCs have intrinsic biocatalytic activity. Different from other peroxidase-like nanozymes relying on extra chromogenic reagents, the PM-CsPbX 3 NCs can be used as a self-reporting nanoprobe, allowing an "add-to-answer" detection model. Notably, the fluorescence of PM-CsPbX 3 NCs can be rapidly quenched by adding H 2 O 2 and then be restored by removing excess H 2 O 2 . Initiated from this unexpected observation, the PM-CsPbX 3 NCs can be explored to prepare multi-color bioinks and metabolite-responsive paper analytical devices, demonstrating the great potential of CsPbX 3 NCs in bioanalysis. This is the first report on the discovery of nanozyme-like property of all-inorganic CsPbX 3 perovskite NCs, which adds another piece to the nanozyme puzzle and opens new avenues for in vitro disease diagnostics., (© 2021 Wiley-VCH GmbH.)

Abstract

With the development of microarray techniques, there is an increasing need of information processing methods to analyze the high throughput data. Clustering is one of the most promising candidates because of its simplicity, flexibility and robustness. However, there is no "perfect" clustering approach outperforming its counterparts, and it is hard to evaluate and combine the results from different techniques, especially in a field without much prior knowledge, such as bioinformatics. This paper proposes a meta-clustering approach to extract the information from results of different clustering techniques, so that a better interpretation of the data distribution can be obtained. A special distance measure is defined to represent the statistical "signal" of each cluster produced by various clustering techniques. The algorithm is applied on both artificial and real data Simulations show that the proposed approach is able to extract the information efficiently and accurately from the input clustering structure.
