Abstract Background Network modeling has been proposed as an effective approach to examine complex associations among antecedents, mediators and symptoms. This study aimed to investigate whether the severity of depressive symptoms affects the multivariate relationships among symptoms and mediating factors over a 2-year longitudinal follow-up. Methods We recruited a school-based cohort of 1480 primary and secondary school students over four semesters from January 2020 to December 2021. The participants (n = 1145) were assessed at four time points (ages 10–13 years old at baseline). Based on a cut-off score of 5 on the 9-item Patient Health Questionnaire at each time point, the participants were categorized into the non-depressive symptom (NDS) and depressive symptom (DS) groups. We conducted network analysis to investigate the symptom-to-symptom influences in these two groups over time. Results The global network metrics did not differ statistically between the NDS and DS groups at four time points. However, network connection strength varied with symptom severity. The edge weights between learning anxiety and social anxiety were prominently in the NDS group over time. The central factors for NDS and DS were oversensitivity and impulsivity (3 out of 4 time points), respectively. Moreover, both node strength and closeness were stable over time in both groups. Conclusions Our study suggests that interrelationships among symptoms and contributing factors are generally stable in adolescents, but a higher severity of depressive symptoms may lead to increased stability in these relationships.

Aiming at the problems of low equipment utilization and the high-capacity requirements of existing arc-suppression devices, a multi-functional reactive power compensation device with the capability of grounding fault regulation (MF-RPCD) is proposed. Firstly, the topology and operation mechanism of MF-RPCD are introduced in this paper. MF-RPCD works in a reactive power compensation mode when a power grid is under normal operation to ensure the unit power factor operation of the power-grid side. When a single line-to-ground fault occurs in the power grid, MF-RPCD works in a fault-regulation mode to effectively suppress the ground-fault current. Secondly, the parameters of the active and passive parts of MF-RPCD are optimally designed to ensure the stable operation of MF-RPCD. Finally, the correctness, feasibility and effectiveness of the proposed topology and functions are verified using simulations and experiments.

Parthenocarpy and male sterility are highly desirable traits in tomato breeding and molecular study. The stamen carpelloid mutant generally displays male sterility. A natural mutant displaying carpelloid stamen and parthenocarpy (csp) was identified in our research group. In this study, the csp locus was finely mapped to a 65 kb interval, which contained six putative genes. One of them, Solyc04g081000, encodes the tomato class B MADS box gene TAP3 (syn. SlDEF). Sequencing data revealed that a copia long terminal repeat retrotransposon was inserted in the first intron of the TAP3 gene of the csp mutant. qRT-PCR showed that the expression of TAP3 was significantly down-regulated in the petals and stamens of the csp mutant. A phenotypic analysis of the TAP3 gene-edited mutants and allelism tests indicated that TAP3 was the gene underlying csp, and csp was a novel allelic mutation of TAP3. The results of this study will lay the foundation for a further analysis of the function of TAP3 and provide materials and a basis for a further study of the functional differentiation of tomato B-class genes.

Male sterility is a valuable trait for hybrid seed production in tomato (Solanum lycopersicum). The mutants male sterile-30 (ms-30) and ms-33 of tomato exhibit twisted stamens, exposed stigmas, and complete male sterility, thus holding potential for application in hybrid seed production. In this study, the ms-30 and ms-33 loci were fine-mapped to 53.3 kb and 111.2 kb intervals, respectively. Tomato PISTILLATA (TPI, syn. SlGLO2), a B-class MADS-box transcription factor gene, was identified as the most likely candidate gene for both loci. TPI is also the candidate gene of tomato male sterile mutant 7B-1 and sl-2. Allelism tests revealed that ms-30, ms-33, 7B-1, and sl-2 were allelic. Sequencing analysis showed sequence alterations in the TPI gene in all these mutants, with ms-30 exhibiting a transversion (G to T) that resulted in a missense mutation (S to I); ms-33 showing a transition (A to T) that led to alternative splicing, resulting in a loss of 46 amino acids in protein; and 7B-1 and sl-2 mutants showing the insertion of an approximately 4.8 kb retrotransposon. On the basis of these sequence alterations, a Kompetitive Allele Specific PCR marker, a sequencing marker, and an Insertion/Deletion marker were developed. Phenotypic analysis of the TPI gene-edited mutants and allelism tests indicated that the gene TPI is responsible for ms-30 and its alleles. Transcriptome analysis of ms-30 and quantitative RT-PCR revealed some differentially expressed genes associated with stamen and carpel development. These findings will aid in the marker-assisted selection for ms-30 and its alleles in tomato breeding and support the functional analysis of the TPI gene.

In order to optimize the efficiency of pineapple harvesting robots in recognition and target detection, this paper introduces a lightweight pineapple detection model, namely MSGV-YOLOv7. This model adopts MobileOne as the innovative backbone network and uses thin neck as the neck network. The enhancements in these architectures have significantly improved the ability of feature extraction and fusion, thereby speeding up the detection rate. Empirical results indicated that MSGV-YOLOv7 surpassed the original YOLOv7 with a 1.98% increase in precision, 1.35% increase in recall rate, and 3.03% increase in mAP, while the real-time detection speed reached 17.52 frames per second. Compared with Faster R-CNN and YOLOv5n, the mAP of this model increased by 14.89% and 5.22%, respectively, while the real-time detection speed increased by approximately 2.18 times and 1.58 times, respectively. The application of image visualization testing has verified the results, confirming that the MSGV-YOLOv7 model successfully and precisely identified the unique features of pineapples. The proposed pineapple detection method presents significant potential for broad-scale implementation. It is expected to notably reduce both the time and economic costs associated with pineapple harvesting operations.

Plant cuticular wax plays an important role in resistance to environmental stresses. Eceriferum (CER) genes are involved in wax synthesis. However, little information is available for tomato species. In this study, 26 SlCER genes were identified in tomato (S. lycopersicum), and they were classified into four clades. The physicochemical properties and conserved motifs of their proteins were predicted. These SlCERs were mainly expressed in leaves, flowers or fruits, and most SlCERs played roles in response to abiotic stresses, especially drought stress. Furthermore, the changes in haplotypes indicated that SlCERs might have been involved in adapting to the environments for wild species S. pimpinellifolium before domestication. These findings would lay a foundation for future functional studies of SlCERs and also provide insights for anti-stress improvement in tomato in the near future.

Tomato (Solanum lycopersicum) is a widely consumed vegetable, and the tomato fruit weight is a key yield component. Many quantitative trait loci (QTLs) controlling tomato fruit weight have been identified, and six of them have been fine-mapped and cloned. Here, four loci controlling tomato fruit weight were identified in an F2 population through QTL seq.; fruit weight 6.3 (fw6.3) was a major-effect QTL and its percentage of variation explanation (R2) was 0.118. This QTL was fine-mapped to a 62.6 kb interval on chromosome 6. According to the annotated tomato genome (version SL4.0, annotation ITAG4.0), this interval contained seven genes, including Solyc06g074350 (the SELF-PRUNING gene), which was likely the candidate gene underlying variation in fruit weight. The SELF-PRUNING gene contained a single-nucleotide polymorphism that resulted in an amino acid substitution in the protein sequence. The large-fruit allele of fw6.3 (fw6.3HG) was overdominant to the small-fruit allele fw6.3RG. The soluble solids content was also increased by fw6.3HG. These findings provide valuable information that will aid the cloning of the FW6.3 gene and ongoing efforts to breed tomato plants with higher yield and quality via molecular marker-assisted selection.

Applying nitrogen fertilization is an important way to improve the yield and quality of autumn tea (Camellia sinensis L.), but the effects of nitrogen application rate and nitrogen form still remain unclear. Field experiments were conducted in a drip-irrigated tea garden in Rizhao City, China in 2020 and 2021. The effects of nitrogen application levels (N: 0 kg·hm−2, CK; N: 45 kg·hm−2, U1; 75 kg·hm−2, U2; and 105 kg·hm−2, U3) and nitrogen application forms (ammonium bicarbonate, AB; ammonium bicarbonate + urea, UAB; and urea, U) on soil moisture, as well as nitrogen spatiotemporal change, and autumn tea yield and quality, were studied. Results showed that applying ammonium bicarbonate or urea through a drip irrigation system can significantly increase the tea plant evapotranspiration and the autumn tea yield and quality (including free amino acids and tea polyphenols). With the same nitrogen application, the urea fertilization treatment had the higher ammonium nitrogen content within the 0–60 cm soil layer. The application form of nitrogen fertilizer had a significant impact on the yield of autumn tea, and the yield increasing ability was U > UAB > AB. The partial factor productivity of applied nitrogen under the AB treatment was the lowest. The yield-increasing effect of nitrogen fertilizer can be observed only 16–18 days after topdressing through the drip irrigation system. In 2020 and 2021, the yield of autumn tea under the U3 treatment increased by 40.6% and 23.0%, respectively, compared with the CK treatment. In conclusion, the topdressing with urea 105 kg·hm−2 with drip irrigation for tea plants in autumn is recommended. This recommendation will provide a theoretical basis for efficient irrigation and yield increase in tea gardens.

Abstract The tomato (Solanum lycopersicum) male sterile 32 (ms32) mutant has been used in hybrid seed breeding programs largely because it produces no pollen and has exserted stigmas. In this study, histological examination of anthers revealed dysfunctional pollen and tapetum development in the ms32 mutant. The ms32 locus was fine mapped to a 28.5 kb interval that encoded four putative genes. Solyc01g081100, a homolog of Arabidopsis bHLH10/89/90 and rice EAT1, was proposed to be the candidate gene of MS32 because it contained a single nucleotide polymorphism (SNP) that led to the formation of a premature stop codon. A codominant derived cleaved amplified polymorphic sequence (dCAPS) marker, MS32D, was developed based on the SNP. Real-time quantitative reverse-transcription PCR showed that most of the genes, which were proposed to be involved in pollen and tapetum development in tomato, were downregulated in the ms32 mutant. These findings may aid in marker-assisted selection of ms32 in hybrid breeding programs and facilitate studies on the regulatory mechanisms of pollen and tapetum development in tomato.

Plant height is an important agronomic trait in crops. Several genes underlying tomato (Solanum lycopersicum) plant height mutants have been cloned. However, few quantitative trait genes for plant height have been identified in tomato. In this study, seven quantitative trait loci (QTLs) controlling plant height were identified in tomato. Of which, qtph1.1 (QTL for tomato plant height 1.1), qtph3.1 and qtph12.1 were major QTLs and explained 15, 16, and 12% of phenotypic variation (R2), respectively. The qtph1.1 was further mapped to an 18.9-kb interval on chromosome 1. Based on the annotated tomato genome (version SL2.50, annotation ITAG2.40), Solyc01g098390 encoding GA receptor SlGID1a was the putative candidate gene. The SlGID1a gene underlying the qtph1.1 locus contained a single nucleotide polymorphism (SNP) that resulted in an amino acid alteration in protein sequence. The near-isogenic line containing the qtph1.1 locus (NIL-qtph1.1) exhibited shorter internode length and cell length than the wild type (NIL-WT). The dwarf phenotype of NIL-qtph1.1 could not be rescued by exogenous GA3 treatment. Transcriptome analysis and real-time quantitative reverse transcription PCR (qPCR) showed that several genes related to biosynthesis and signaling of GA and auxin were differentially expressed in stems between NIL-qtph1.1 and NIL-WT. These findings might pave the road for understanding the molecular regulation mechanism of tomato plant height.

A new kind of molecularly imprinted polymer (MIP) microspheres for the selective extraction of kaempferol was prepared by precipitation polymerization using 4-vinylpridine (4-VP) and ethylene glycol dimethacrylate (EDMA) as functional monomer and cross-linker respectively. The synthesis conditions, such as ratios of 4-VP/EDMA and polymerization time were discussed in detail. Results showed that the 2% was the optimal concentration of co-monomers to obtain monodisperse MIP microspheres, the best ratio of 4-VP/EDMA was 1:2, and 24 h was considered as the proper polymerization time. Compared with the MIP agglomeration or coagulum particles, monodisperse MIP microspheres showed the better adsorption capacity: the saturated adsorption capacity of monodisperse MIP microspheres was 7.47 mg g−1, the adsorption equilibrium could be obtained in 30 min. Finally, the adsorption performances of the optimal MIP microspheres were evaluated by kinetic adsorption, adsorption isotherm, and selective adsorption experiments, which indicated that the adsorption mechanism were chemical single layer adsorption and the separation factor was up to 3.91 by comparing with the structure similar compound (quercetin). The MIP microspheres exhibit prospects in the kaempferol efficient and selective separation.

Abstract Improving yield by increasing the size of produce is an important selection criterion during the domestication of fruit and vegetable crops. Genes controlling meristem organization and organ formation work in concert to regulate the size of reproductive organs. In tomato, lc and fas control locule number, which often leads to enlarged fruits compared to the wild progenitors. LC is encoded by the tomato ortholog of WUSCHEL (WUS), whereas FAS is encoded by the tomato ortholog of CLAVATA3 (CLV3). The critical role of the WUS‐CLV3 feedback loop in meristem organization has been demonstrated in several plant species. We show that mutant alleles for both loci in tomato led to an expansion of the SlWUS expression domain in young floral buds 2–3 days after initiation. Single and double mutant alleles of lc and fas maintain higher SlWUS expression during the development of the carpel primordia in the floral bud. This augmentation and altered spatial expression of SlWUS provided a mechanistic basis for the formation of multilocular and large fruits. Our results indicated that lc and fas are gain‐of‐function and partially loss‐of‐function alleles, respectively, while both mutations positively affect the size of tomato floral meristems. In addition, expression profiling showed that lc and fas affected the expression of several genes in biological processes including those involved in meristem/flower development, patterning, microtubule binding activity, and sterol biosynthesis. Several differentially expressed genes co‐expressed with SlWUS have been identified, and they are enriched for functions in meristem regulation. Our results provide new insights into the transcriptional regulation of genes that modulate meristem maintenance and floral organ determinacy in tomato.

Introgression lines are convenient populations for the identification, fine-mapping, and functional analysis of genes that are responsible for variations in traits, particularly quantitative trait genes. An introgression line population of Solanum pennellii LA0716 in a fresh market tomato inbred line 1052 was developed by our group. This population was composed of 214 lines. In the present study, five quantitative trait loci (QTLs) for fruit weight, two QTLs for soluble solids content (SSC), three QTLs for plant height, and one QTL for leaf size were identified using this introgression line population. Among these, fw3a and fw4a for fruit weight, ssc7a for SSC, h4t2a, h4t3a, and h4t7a for plant height, and lz12a for leaf size were determined to be novel loci. These results serve as a foundation for fine-mapping and functional analysis of genes underlying these QTLs.

To improve the economic efficiency of gas pipelines, core equipment such as compressor sets and large-diameter valves must be localized. For this purpose, in alliance with other related enterprises, PetroChina Company Limited established an equipment localization R&D system and a new product testing system and successfully developed a 20 MW class motor-driven compressor set, a 30 MW-class gas turbine-driven compressor unit, and a high-pressure and large-diameter welded ball valve. First, the motor-driven compressor R&D focuses on three main units. The developed frequency-control device structure is a cascaded multilevel with a capacity of 25 MVA. The developed anti-explosion dynamo with a motor speed of 4800 rpm can produce a power of 22 MW. The developed compressor is PCL800 with features of a high efficiency and a wide flow-operating point-adjustment range. Second, there are two steps of the R&D of a GT-driven compressor unit (product A + product B): auxiliary supporting systems and control systems are developed for the imported GT25000 gas turbine, together with China-made compressors, to constitute product A; simultaneously, the R&D of product B of a gas turbine is carried out, which would replace the imported one. Third, aiming to solve the problems of sealing and welding, we developed the high-pressure and large-diameter all-welded ball valves in full replace of the same kind of imported products with three different sizes: NPS40 Class 600, NPS48 Class 600, and NPS48 Class 900.

Increases in fruit weight of cultivated vegetables and fruits accompanied the domestication of these crops. Here we report on the positional cloning of a quantitative trait locus (QTL) controlling fruit weight in tomato. The derived allele of Cell Size Regulator (CSR-D) increases fruit weight predominantly through enlargement of the pericarp areas. The expanded pericarp tissues result from increased mesocarp cell size and not from increased number of cell layers. The effect of CSR on fruit weight and cell size is found across different genetic backgrounds implying a consistent impact of the locus on the trait. In fruits, CSR expression is undetectable early in development from floral meristems to the rapid cell proliferation stage after anthesis. Expression is low but detectable in growing fruit tissues and in or around vascular bundles coinciding with the cell enlargement stage of the fruit maturation process. CSR encodes an uncharacterized protein whose clade has expanded in the Solanaceae family. The mutant allele is predicted to encode a shorter protein due to a 1.4 kb deletion resulting in a 194 amino-acid truncation. Co-expression analyses and GO term enrichment analyses suggest association of CSR with cell differentiation in fruit tissues and vascular bundles. The derived allele arose in Solanum lycopersicum var cerasiforme and appears completely fixed in many cultivated tomato's market classes. This finding suggests that the selection of this allele was critical to the full domestication of tomato from its intermediate ancestors.

The hydrophobic cuticle that covers the surface of tomato (Solanum lycopersicum) fruit plays key roles in development and protection against biotic and abiotic stresses, including water loss, mechanical damage, UV radiation, pathogens, and pests. However, many details of the genes and regulatory mechanisms involved in cuticle biosynthesis in fleshy fruits are not well understood. In this study, we describe a novel tomato fruit phenotype, characterized by epidermal reticulation (ER) of green fruit and a higher water loss rate than wild type (WT) fruit. The ER phenotype is controlled by a single gene, ER4.1, derived from an introgressed chromosomal segment from the wild tomato species S. pennellii (LA0716). We performed fine mapping of the single dominant gene to an ~300 kb region and identified Solyc04g082540, Solyc04g082950, Solyc04g082630, and Solyc04g082910as potential candidate genes for the ER4.1 locus, based on comparative RNA-seq analysis of ER and WT fruit peels. In addition, the transcriptome analysis revealed that the expression levels of genes involved in cutin, wax and flavonoid biosynthesis were altered in the ER fruit compared with WT. This study provides new insights into the regulatory mechanisms and metabolism of the fruit cuticle.

Internode length is an important agronomic trait affecting plant architecture and crop yield. However, few genes for internode elongation have been identified in tomato. In this study, we characterized an elongated internode inbred line P502, which is a natural mutant of the tomato cultivar 05T606. The mutant P502 exhibits longer internode and higher bioactive GA concentration compared with wild-type 05T606. Genetic analysis suggested that the elongated internode trait is controlled by quantitative trait loci (QTL). Then, we identified a major QTL on chromosome 2 based on molecular markers and bulked segregant analysis (BSA). The locus was designated as EI (Elongated Internode), which explained 73.6% genetic variance. The EI was further mapped to a 75.8-kb region containing 10 genes in the reference Heinz 1706 genome. One single nucleotide polymorphism (SNP) in the coding region of solyc02g080120.1 was identified, which encodes gibberellin 2-beta-dioxygenase 7 (SlGA2ox7). SlGA2ox7, orthologous to AtGA2ox7 and AtGA2ox8, is involved in the regulation of GA degradation. Overexpression of the wild EI gene in mutant P502 caused a dwarf phenotype with a shortened internode. The difference of EI expression levels was not significant in the P502 and wild-type, but the expression levels of GA biosynthetic genes including CPS, KO, KAO, GA20ox1, GA20ox2, GA20ox4, GA3ox1, GA2ox1, GA2ox2, GA2ox4, and GA2ox5, were upregulated in mutant P502. Our results may provide a better understanding of the genetics underlying the internode elongation and valuable information to improve plant architecture of the tomato.

Anthocyanin pigments play many roles in plants, including providing protection against biotic and abiotic stresses. Many of the genes that mediate anthocyanin accumulation have been identified through studies of flowers and fruits; however, the mechanisms of genes involved in anthocyanin regulation in seedlings under low-temperature stimulus are less well understood. Genetic characterization of a tomato inbred line, FMTT271, which showed no anthocyanin pigmentation, revealed a mutation in a bHLH transcription factor (TF) gene, which corresponds to the ah (Hoffman's anthocyaninless) locus, and so the gene in FMTT271 at that locus was named ah. Overexpression of the wild type allele of AH in FMTT271 resulted in greater anthocyanin accumulation and increased expression of several genes in the anthocyanin biosynthetic pathway. The expression of AH and anthocyanin accumulation in seedlings was shown to be developmentally regulated and induced by low-temperature stress. Additionally, transcriptome analyses of hypocotyls and leaves from the near-isogenic lines seedlings revealed that AH not only influences the expression of anthocyanin biosynthetic genes, but also genes associated with responses to abiotic stress. Furthermore, the ah mutation was shown to cause accumulation of reactive oxidative species and the constitutive activation of defense responses under cold conditions. These results suggest that AH regulates anthocyanin biosynthesis, thereby playing a protective role, and that this function is particularly important in young seedlings that are particularly vulnerable to abiotic stresses.

Lodging is a major yield-limiting factor of quinoa production. In 2018 and 2019, the orthogonal field experiments were conducted to investigate the responses of quinoa lodging risk and yield to irrigation threshold (soil matric potential of -15, -25 and -55 kPa), nitrogen rate (80, 160 and 240 kg·ha-1) and planting density (20, 30 and 40 plants m-2). Results showed that high irrigation thresholds and nitrogen rates significantly (P < 0.05) increased plant height and fresh weight per plant, and high planting densities reduced stem diameter and strength, all of those led to significantly (P < 0.05) high lodging risks. The -15 and -55 kPa treatments gave the lowest actual yield (P < 0.05) in 2018 and 2019, respectively. Higher lodging rate with a nitrogen rate of 240 kg·ha-1 resulted in a lower actual yield than 80 and 160 kg·ha-1 in both years. Planting density of 30 plants m-2 gave a significantly (P < 0.05) greater estimated yield than 20 plants m-2 and had a lower lodging rate than 40 plants m-2, resulting in the maximum actual yield among planting densities. In conclusion, a moderate irrigation threshold of -25 kPa, a nitrogen rate of 80-160 kg·ha-1 and an intermediate planting density of 30 plants m-2 were determined to be best for quinoa cultivation in North-western China. In addition, the lower-stem lodging index (quarter plant height) could evaluate lodging risk more accurately than middle-stem (half plant height) or upper-stem (three quarters plant height) lodging indexes.
