Abstract Background Nitrate (NO3 −) is one of the two major forms of inorganic nitrogen absorbed by plant roots, and the tissue nitrate concentration in roots is considered important for optimizing developmental programs. Technologies to quantify the expression levels of nitrate transporters and assimilating enzymes at the cellular level have improved drastically in the past decade. However, a technological gap remains for detecting nitrate at a high spatial resolution. Using extraction-based methods, it is challenging to reliably estimate nitrate concentration from a small volume of cells (i.e., with high spatial resolution), since targeting a small or specific group of cells is physically difficult. Alternatively, nitrate detection with microelectrodes offers subcellular resolution with high cell specificity, but this method has some limitations on cell accessibility and detection speed. Finally, optical nitrate biosensors have very good (in-vivo) sensitivity (below 1 mM) and cellular-level spatial resolution, but require plant transformation, limiting their applicability. In this work, we apply Raman microspectroscopy for high-dynamic range in-vivo mapping of nitrate in different developmental zones of Arabidopsis thaliana roots in-situ. Results As a proof of concept, we have used Raman microspectroscopy for in-vivo mapping of nitrate content in roots of Arabidopsis seedlings grown on agar media with different nitrate concentrations. Our results revealed that the root nitrate concentration increases gradually from the meristematic zone (~ 250 µm from the root cap) to the maturation zone (~ 3 mm from the root cap) in roots grown under typical growth conditions used for Arabidopsis, a trend that has not been previously reported. This trend was observed for plants grown in agar media with different nitrate concentrations (0.5–10 mM). These results were validated through destructive measurement of nitrate concentration. Conclusions We present a methodology based on Raman microspectroscopy for in-vivo label-free mapping of nitrate within small root tissue volumes in Arabidopsis. Measurements are done in-situ without additional sample preparation. Our measurements revealed nitrate concentration changes from lower to higher concentration from tip to mature root tissue. Accumulation of nitrate in the maturation zone tissue shows a saturation behavior. The presented Raman-based approach allows for in-situ non-destructive measurements of Raman-active compounds.

Implementing a collaborative governance framework across administrative boundaries is crucial for preserving the Basin’s ecological integrity and mitigating institutional fragmentation. Based on 19 cases of inter-provincial collaborative governance of ecological environment in the Yangtze River Basin of China, this study uses the fuzzy set qualitative comparative analysis method to explore the influencing factors and complex causal mechanism of the effect of inter-provincial collaborative governance. The results show that technology empowers relationship driving, institutions reinforce interactive driving, and internal and external interaction driving modes promote collaborative governance. Perceived factors are essential in motivating provinces to participate in collaborative governance. External factors play a hygienic role in collaborative governance, and internal factors play a motivated role. The roles of the two types of factors are separate but complementary. In view of this, government should attention to regional environmental concerns, ensuring consistent alignment of internal and external factors and fostering synergies to improve governance effectiveness.

The low carbon living of residents is important for China to achieve the “double carbon goal.” This paper uses the natural language processing technique in machine learning to collect data from 2005 to 2020, constructs the government’s “green will” index and residents’ low-carbon life index, and conducts an empirical study through a static panel model and a dynamic panel model. The study shows that: (a) all cities in the Yangtze River Delta pay more and more attention to low-carbon publicity, but there are individual differences in the degree of attention; (b) in general, the government’s willingness to go green does drive residents to develop a low-carbon life; (c) affluence and human capital growth are positively related to the effect of government low-carbon publicity. Accordingly, the following measures are proposed: increase the government’s low-carbon publicity efforts, deepen education reform, attract high-end talents, strengthen scientific and technological innovation, promote new urbanization, and reduce the marginal cost of carbon emissions.

Abstract Ice-templating technology holds great potential to construct industrial porous materials from nanometers to the macroscopic scale for tailoring thermal, electronic, or acoustic transport. Herein, we describe a general ice-templating technology through freezing the material on a rotating cryogenic drum surface, crushing it, and then re-casting the nanofiber slurry. Through decoupling the ice nucleation and growth processes, we achieved the columnar-equiaxed crystal transition in the freezing procedure. The highly random stacking and integrating of equiaxed ice crystals can organize nanofibers into thousands of repeating microscale units with a tortuous channel topology. Owing to the spatially well-defined isotropic structure, the obtained Al2O3·SiO2 nanofiber aerogels exhibit ultralow thermal conductivity, superelasticity, good damage tolerance, and fatigue resistance. These features, together with their natural stability up to 1200 °C, make them highly robust for thermal insulation under extreme thermomechanical environments. Cascading thermal runaway propagation in a high-capacity lithium-ion battery module consisting of LiNi0.8Co0.1Mn0.1O2 cathode, with ultrahigh thermal shock power of 215 kW, can be completely prevented by a thin nanofiber aerogel layer. These findings not only establish a general production route for nanomaterial assemblies that is conventionally challenging, but also demonstrate a high-energy-density battery module configuration with a high safety standard that is critical for practical applications.

Under the challenge of global energy transition, coal-fired cogeneration systems are undergoing a technical revolution towards enhanced efficiency, heating capacity, and flexibility. In this paper, four schemes using a steam ejector integrated into a cogeneration system are designed. Considering operational safety, integrated locations are selected at the front and back of high- and medium-pressure turbines. Subsequently, the thermodynamic and operational characteristics under both design and off-design conditions are analyzed based on a model built in EBSILON Professional. Finally, a sensitivity analysis of the heating process is conducted. The results show that the integration of steam ejectors can increase the waste heat recovery ratio of exhaust steam by 18.42–45.61% under design conditions. The largest waste heat recovery ratio is obtained in System 4, resulting in the power generation efficiency (ηg) and gross energy utilization efficiency (ηp) of 81.95% and 65.53%, respectively. Meanwhile, the steam ejector can expand the power-load regulation range of the cogeneration system, and System 4 has the lowest lower power limit among all the systems. The ηp values of Systems 1–4 reach extreme values at different mixed steam pressures of the ejector. Increasing the pinch point temperature difference reduces the power load ηg and ηp of Systems 1–4. The results provide technical solutions for improving the heating capacity and efficient and flexible operation of cogeneration systems.

Abstract Precision medicine that takes individual variability into account is a new way to minimize the individual variability for precise diagnosis and to establish the certain strategies of efficient therapy. Due to miniaturization sensing area, remoting detection, and flexible operation of light, fiber optics have been successfully used to develop in vitro, wearable, and implanted biosensors, exhibiting high potential in precision medicine from the diagnosis in vitro to therapy in vivo. When modified with the recognition element, the fiber optic‐based biosensor enables the point of care testing for the detection of disease biomarkers. Meanwhile, the fiber optics packaged in a needle/catheter also monitor the medical Internet of things for the continuous assessment of human health. Moreover, the light from the cladding or leaky mode in the fiber optics excites the strong photothermal effect for therapy. The fiber optic‐based biosensor addressed the drawbacks of plasmonic photothermal therapy that limited penetration depths and non‐selectivity therapy. When integrated with a photothermal agent, the fiber optic‐based biosensors provide in situ information for the diagnosis as well as specifically and efficiently kill the tumor cells in deep tissues. In this review, we summarized the advances of fiber optic‐based biosensors in the diagnosis in vitro and therapy in vivo. The challenges and prospection of the fiber optic‐based biosensor application in precision medicine were also explored in depth.

During the opening and closing process of the contact, it will undergo repeated arc ablation and wear. After each ablation and wear, the composition, performance, and shape of the contact surface constantly change, leading to cracks, slag drop, melting, evaporation, and other phenomena on the contact surface, ultimately causing the contact to fail, resulting in the switch not being able to open and close normally. In order to study the failure mechanism of arc contacts caused by arc erosion after breaking short-circuit current, a pair of arc contacts for the cumulative breaking short circuit current test was selected as the research object. The metallographic, microscopic morphology, and energy spectrum analysis of the arc contacts were conducted, and the analysis results were compared with the new arc contacts to reveal the evolution and development process of arc contacts from cumulative arc erosion to failure during breaking.

In recent years, bird droppings flashover faults frequently occur on transmission lines. Based on the existing conditions of the laboratory, this paper explores the flashover mechanism of 110 kV bird droppings. A new type of bird droppings propulsion device was designed and developed, and the process of bird droppings falling in the natural state was simulated without voltage. According to several typical forms of bird droppings, the main findings are ① the bird droppings connected to the two poles are the most serious short circuit situation; ② the bird droppings flashover fault of the 110 kV line is actually a single-phase grounding fault of the line through a resistance, and the grounding resistance is between 20 and 300 kΩ; and ③ the conductivity is the most sensitive factor affecting bird droppings flashover.

Root system architecture (RSA) is the primary predictor of nutrient intake and significantly influences potassium utilization efficiency (KUE). Uncertainty persists regarding the genetic factors governing root growth in rapeseed. The root transcriptome analysis reveals the genetic basis driving crop root growth. In this study, RNA-seq was used to profile the overall transcriptome in the root tissue of 20 Brassica napus accessions with high and low KUE. 71,437 genes in the roots displayed variable expression profiles between the two contrasting genotype groups. The 212 genes that had varied expression levels between the high and low KUE lines were found using a pairwise comparison approach. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) functional classification analysis revealed that the DEGs implicated in hormone and signaling pathways, as well as glucose, lipid, and amino acid metabolism, were all differently regulated in the rapeseed root system. Additionally, we discovered 33 transcription factors (TFs) that control root development were differentially expressed. By combining differential expression analysis, weighted gene co-expression network analysis (WGCNA), and recent genome-wide association study (GWAS) results, four candidate genes were identified as essential hub genes. These potential genes were located fewer than 100 kb from the peak SNPs of QTL clusters, and it was hypothesized that they regulated the formation of the root system. Three of the four hub genes’ homologs—BnaC04G0560400ZS, BnaC04G0560400ZS, and BnaA03G0073500ZS—have been shown to control root development in earlier research. The information produced by our transcriptome profiling could be useful in revealing the molecular processes involved in the growth of rapeseed roots in response to KUE.

Stem lodging resistance is a serious problem impairing crop yield and quality. ZS11 is an adaptable and stable yielding rapeseed variety with excellent resistance to lodging. However, the mechanism regulating lodging resistance in ZS11 remains unclear. Here, we observed that high stem mechanical strength is the main factor determining the superior lodging resistance of ZS11 through a comparative biology study. Compared with 4D122, ZS11 has higher rind penetrometer resistance (RPR) and stem breaking strength (SBS) at flowering and silique stages. Anatomical analysis shows that ZS11 exhibits thicker xylem layers and denser interfascicular fibrocytes. Analysis of cell wall components suggests that ZS11 possessed more lignin and cellulose during stem secondary development. By comparative transcriptome analysis, we reveal a relatively higher expression of genes required for S-adenosylmethionine (SAM) synthesis, and several key genes (4-COUMATATE-CoA LIGASE, CINNAMOYL-CoA REDUCTASE, CAFFEATE O-METHYLTRANSFERASE, PEROXIDASE) involved in lignin synthesis pathway in ZS11, which support an enhanced lignin biosynthesis ability in the ZS11 stem. Moreover, the difference in cellulose may relate to the significant enrichment of DEGs associated with microtubule-related process and cytoskeleton organization at the flowering stage. Protein interaction network analysis indicate that the preferential expression of several genes, such as LONESOME HIGHWAY (LHW), DNA BINDING WITH ONE FINGERS (DOFs), WUSCHEL HOMEOBOX RELATED 4 (WOX4), are related to vascular development and contribute to denser and thicker lignified cell layers in ZS11. Taken together, our results provide insights into the physiological and molecular regulatory basis for the formation of stem lodging resistance in ZS11, which will greatly promote the application of this superior trait in rapeseed breeding.

Fabricating TC4 coatings on the carbon fiber reinforced plastic (CFRP) substrate surface will contribute to increasing the applicability of CFRP materials in the aerospace industry. However, chemical metallurgical bonding between the TC4 alloy and CFRP is crucial during the manufacturing process. Here, we studied the effect of Al powder content on the microstructural characteristics and mechanical properties of the laser cladded TC4+AlSi10Mg composite coating on the CFRP surface. Furthermore, the relationship between laser process parameters, interfacial morphologies, and mechanical properties is established. The amount of CFRP decomposed increases with a rise in the AlSi10Mg powder content, while augmenting the penetration of the transition layer into the CFRP material. Moreover, the TC4 coating with 10wt.% AlSi10Mg is composed of α′ martensite, TiC, Ti3Al, and Ti3AlC. The average nano-hardness of the TC4 coating increases from 6.00 to 6.86 GPa when the AlSi10Mg content rises from 5% to 20%. Meanwhile, the maximum shear strength of the CFRP/TC4 interface is 26.8 MPa. Based on the experimental observations and theoretical analysis, the interface reaction mechanism of the laser cladded TC4+AlSi10Mg composite coating on the CFRP surface is: Ti(s)+C(s)→TiC(s), Al(1)+3Ti(s)→Ti3Al(s).

Abstract Wheat grain protein content and composition are important for its end-use quality. Protein synthesis during the grain filling phase is supported by the amino acids remobilized from the vegetative tissue, the process in which both amino acid importers and exporters are expected to be involved. Previous studies identified amino acid importers that might function in the amino acid remobilization in wheat. However, the amino acid exporters involved in this process have been unexplored so far. In this study, we have curated the Usually Multiple Amino acids Move In and out Transporter (UMAMIT) family of transporters in wheat. As expected, the majority of UMAMITs were found as triads in the A, B, and D genomes of wheat. Expression analysis using publicly available data sets identified groups of TaUMAMITs expressed in root, leaf, spike, stem and grain tissues, many of which were temporarily regulated. Strong expression of TaUMAMITs was detected in the late senescing leaves and transfer cells in grains, both of which are the expected site of apoplastic amino acid transport during grain filling. Biochemical characterization of selected TaUMAMITs revealed that TaUMAMIT17 shows a strong amino acid export activity and might play a role in amino acid transfer to the grains.

Despite promising for anti-icing applications, structured superhydrophobic surfaces usually lose their hydrophobicity after a few icing/melting cycles. Here, authors investigate specific structured surfaces and air bubbles on frozen ice droplets to propose three criteria to enable dewetting transitions.

This study investigates the institutional quality effects on the foreign direct investment (FDI)–regional integration nexus in the Community of Sahel-Saharan States (CEN-SAD). The novelty of our approach extends the regional integration theory in Africa by employing the Africa Regional Integration Index (ARII) and utilizes a dynamic panel data model based on the one-step system generalized method of moments (SYS-GMM) estimation to explore the institutional quality effects on the relationship between FDI location advantages and inflows and dimensions of regional integration. Our study highlights macroeconomic integration as the strongest indicator of regional integration as well as the most crucial determinant of FDI in CEN-SAD. We also discovered that FDI inflows into the lower-middle income countries (LMICs) were only slightly higher than in low-income countries (LICs). Our results infer that institutional reforms augment FDI location advantages and enhance FDI inflows irrespective of the level of integration in the Community.

Abstract Background The energy generation efficiencies of thermal power and hydropower, which are the two main forces of electric power in China, are important factors affecting the energy conservation, emission reduction, and green development of the country’s whole power industry. Methods Considering regional differences and multiple efficient decision-making units (DMUs), this research uses the meta-Frontier super-efficiency slack-based measure (meta-SE-SBM) undesirable model to comprehensively evaluate the efficiencies of hydropower and thermal power generation in China. The CO2 emissions of thermal power generation are taken as the undesirable output. Results The ranking of the average meta-efficiency of thermal power generation in China is Eastern China > Central China > Western China, and all regions show an upward trend. However, the ranking of the average meta-efficiency of hydropower generation is Western China > Central China > Eastern China, and all these regions present a downward trend. In 2017, the technology gap ratio (TGR) values for the thermal power generation efficiency of the eastern and western regions showed a rising trend, while that for the central region showed a declining trend. The TGR values of the hydropower generation efficiency of the western region continued to increase, while those of the central and eastern regions decreased. The development trends of the TGR values of the thermal power or hydropower generation efficiencies of the three regions were not consistent with each other, indicating that technological convergence has not been achieved. In the three regions, the technology gaps in hydropower have slightly expanded, but the technology gaps in thermal power have gradually narrowed. The undesirable output CO2 of the thermal power energy efficiency of the three regions is in a surplus, and the generation of hydropower in the eastern and central regions is insufficient. Conclusions The government and power industry managers should fully consider regional heterogeneity in the efficiency of hydropower and thermal power to reduce the technology gap in China. The thermal power industry is relatively mature, but its CO2 emissions should be controlled. The hydropower industry needs further policy support to promote an efficiency improvement in it under the condition of resource endowments.

Purpose. Given the efforts towards achieving the United Nations’ Sustainable Development Goals (SDGs) for food security by 2030, this study investigates the moderating impact of public investments in agriculture on the agricultural producer price – agricultural sustainability nexus in 40 African countries covering the period from 2000 till 2019. Methodology / approach. In this study we used multiple regression techniques to explore a dynamic panel data model based on the one-step system Generalized Method of Moments (System-GMM). Proposed by Arellano and Bover and further developed by Blundell and Bond, the System-GMM estimator was preferred over other techniques because of its efficiency in eliminating the simultaneous biases that are associated with regression model estimations. Specifically, the one-step System-GMM was preferred over the two-step System-GMM for our estimation due to the efficiency of its optimal weighting matrices. Results. The study discovered that although the interactions between public investments on agriculture and agricultural sustainability amplify the positive impact of a set of explanatory variables on agricultural producer price to an extent, the impact on food security in Africa is insignificant. The study also discovers that the net effects of a set of interactive terms on producer price in the developing countries in Africa are slightly lower than in the least-developed countries. The weighted average food security index for the period under study was abysmal 44.54%, indicating moderate food insecurity in Africa. Originality / scientific novelty. In the context of food security literature in Africa, this study is the first attempt at exploring the agricultural producer price – agricultural sustainability nexus based on the moderating impact of public investments on agriculture with the Global Food Security Index (GFSI), a composite food security model developed by the Economist Intelligence Unit (EIU). Among our study proposals it is a call for a detour from the current agricultural investment and producer price policies especially the current ‘one-size-fits-all’ regional frameworks which have proved to be less progressive and less transformative to more robust country/sector-specific frameworks that have the potential to better the fortunes of agriculture and improve food security. Practical value / implications. The current state of agricultural producer price in most African countries is ample proof that the role and importance of the producer price have been gravely diminished. Despite governments’ efforts towards improving food security, the evidence as presented in this study supports the fact that those efforts have not achieved much success. The study, which contains a number of recommendations, highlights agricultural producer price as a potentially important driver of agricultural sustainability and sustainable food security in Africa.

Abstract Background In terms of global demand, rapeseed is the third-largest oilseed crop after soybeans and palm, which produces vegetable oil for human consumption and biofuel for industrial production. Roots are vital organs for plant to absorb water and attain mineral nutrients, thus they are of great importance to plant productivity. However, the genetic mechanisms regulating root development in rapeseed remain unclear. In the present study, seven root-related traits and shoot biomass traits in 280 Brassica napus accessions at five continuous vegetative stages were measured to establish the genetic basis of root growth in rapeseed. Results The persistent and stage-specific genetic mechanisms were revealed by root dynamic analysis. Sixteen persistent and 32 stage-specific quantitative trait loci (QTL) clusters were identified through genome-wide association study (GWAS). Root samples with contrasting (slow and fast) growth rates throughout the investigated stages and those with obvious stage-specific changes in growth rates were subjected to transcriptome analysis. A total of 367 differentially expressed genes (DEGs) with persistent differential expressions throughout root development were identified, and these DEGs were significantly enriched in GO terms, such as energy metabolism and response to biotic or abiotic stress. Totally, 485 stage-specific DEGs with different expressions at specific stage were identified, and these DEGs were enriched in GO terms, such as nitrogen metabolism. Four candidate genes were identified as key persistent genetic factors and eight as stage-specific ones by integrating GWAS, weighted gene co-expression network analysis (WGCNA), and differential expression analysis. These candidate genes were speculated to regulate root system development, and they were less than 100 kb away from peak SNPs of QTL clusters. The homologs of three genes (BnaA03g52990D, BnaA06g37280D, and BnaA09g07580D) out of 12 candidate genes have been reported to regulate root development in previous studies. Conclusions Sixteen QTL clusters and four candidate genes controlling persistently root development, and 32 QTL clusters and eight candidate genes stage-specifically regulating root growth in rapeseed were detected in this study. Our results provide new insights into the temporal genetic mechanisms of root growth by identifying key candidate QTL/genes in rapeseed.

Branch architecture is an important factor influencing rapeseed planting density, mechanized harvest, and yield. However, its related genes and regulatory mechanisms remain largely unknown. In this study, branch angle (BA) and branch dispersion degree (BD) were used to evaluate branch architecture. Branch angle exhibited a dynamic change from an increase in the early stage to a gradual decrease until reaching a stable state. Cytological analysis showed that BA variation was mainly due to xylem size differences in the vascular bundle of the branch junction. The phenotypic analysis of 327 natural accessions revealed that BA in six environments ranged from 24.3° to 67.9°, and that BD in three environments varied from 4.20 cm to 21.4 cm, respectively. A total of 115 significant loci were detected through association mapping in three models (MLM, mrMLM, and FarmCPU), which explained 0.53%-19.4% of the phenotypic variations. Of them, 10 loci were repeatedly detected in different environments and models, one of which qBAD.A03-2 was verified as a stable QTL using a secondary segregation population. Totally, 1066 differentially expressed genes (DEGs) were identified between branch adaxial- and abaxial- sides from four extremely large or small BA/BD accessions through RNA sequencing. These DEGs were significantly enriched in the pathways related to auxin biosynthesis and transport as well as cell extension such as indole alkaloid biosynthesis, other glycan degradation, and fatty acid elongation. Four known candidate genes BnaA02g16500D (PIN1), BnaA03g10430D (PIN2), BnaC03g06250D (LAZY1), and BnaC06g20640D (ARF17) were identified by both GWAS and RNA-seq, all of which were involved in regulating the asymmetric distribution of auxins. Our identified association loci and candidate genes provide a theoretical basis for further study of gene cloning and genetic improvement of branch architecture.

Plants rely on root systems for nutrient uptake from soils. Marker-assisted selection helps breeders to select desirable root traits for effective nutrient uptake. Here, 12 root and biomass traits were investigated at the seedling stage under low nitrogen (LN), low phosphorus (LP), and low potassium (LK) conditions, respectively, in a recombinant inbred line (RIL) population, which was generated from Brassica napus L. Zhongshuang11 and 4D122 with significant differences in root traits and nutrient efficiency. Significant differences for all the investigated traits were observed among RILs, with high heritabilities (0.43–0.74) and high correlations between the different treatments. Quantitative trait loci (QTL) mapping identified 57, 27, and 36 loci, explaining 4.1–10.9, 4.6–10.8, and 4.9–17.4% phenotypic variances under LN, LP, and LK, respectively. Through QTL-meta analysis, these loci were integrated into 18 significant QTL clusters. Four major QTL clusters involved 25 QTLs that could be repeatedly detected and explained more than 10% phenotypic variances, including two NPK-common and two specific QTL clusters (K and NK-specific), indicating their critical role in cooperative nutrients uptake of N, P, and K. Moreover, 264 genes within the four major QTL clusters having high expressions in roots and SNP/InDel variations between two parents were identified as potential candidate genes. Thirty-eight of them have been reported to be associated with root growth and development and/or nutrient stress tolerance. These key loci and candidate genes lay the foundation for deeper dissection of the NPK starvation response mechanisms in B. napus.

China is the world's largest electricity producer and consumer, and the healthy development of the electricity industry contributes to its economic growth and social stability. Yangtze River Economic Belt (YREB) and non-YREB are two major economic engines of great strategic significance for China with distinct differences in the external development environment of their power industry. A cross-regional comparison of electricity generation efficiency in China is meaningful to obtain a more accurate understanding of the power generation efficiency of YREB and non-YREB, but there is currently no literature in this area as well as none for comparing electricity generation efficiency between different energy sources. Therefore, this research uses the meta-frontier epsilon-based measure (EBM) data envelopment analysis (DEA) model to calculate the efficiency of different electricity generation energy in YREB and non-YREB for the period 2013–2017. The empirical results show that: 1) YREB's electricity industry has higher electricity generation efficiency than non-YREB's, and the efficiency gap is continuing to widen; 2) YREB's average thermal electricity generation efficiency is higher than non-YREB's; 3) YREB's clean energy power generation efficiency is always higher than non-YREB's; 4) regional differences in YREB are smaller than those in non-YREB; and 5) the efficiency value of clean energy power generation is lower than that of thermal power generation. From our analysis results, this paper presents some policy recommendations.

The rise of energy consumption has also increased emissions of the “three wastes” (wastewater, waste gas, and industrial solid waste), and environmental emergencies caused by pollutants, natural disasters, and production safety accidents have aroused social concerns. As few scholars have combined treatment efficiency of the three wastes with environmental emergencies to explore their relationships, this research thus uses a two-stage undesirable Data Envelopment Analysis (DEA) method to explore the situations of 4 regions and 30 provinces in China from 2013 to 2017 based on such interactive perspectives. The study finds that the overall regional environmental efficiency in China is generally low, and in terms of regional differences, the eastern and northeastern regions are better than the central and western regions. The efficiency values of the three wastes in China have also fluctuated greatly from 0.7 down to 0.2 in recent years. The efficiency of environmental emergencies in China is greatly impacted by the efficiency of environmental governance inputs. Based on the results, the study proposes that the eastern provinces can be an example for promoting balanced regional development and offers policy recommendations such as taking precautions against environmental emergencies.

China has become the largest wind power installation market in the world, and on such a large scale its wind power industry contributes to the sustainability of electricity generation and reduction of carbon emissions, yet has problems such as wind curtailment, insufficient wind power consumption, and regional disparities. Thus, this research uses an epsilon-based measure (EBM) data envelopment analysis (DEA) model to evaluate and compare wind power electricity generation efficiency and CO2 emission reduction efficiency in the eastern, central, western, and northeastern regions of China for the period 2013–2017. The empirical results show that the nation’s overall wind power efficiency presents a significant upward trend, in which the western and northeastern regions have increased the most, while the east region has increased the least. Technical inefficiency is mainly due to diseconomies of scale in China’s wind power industry. Moreover, CO2 emission reduction in the four regions exhibits high efficiency, and the regions’ efficiencies are very consistent with that of installed capacity efficiency. Finally, this study’s policy implications are that industry development plans should be made according to local conditions as well as cross-regional trade of wind power electricity and that the upgrading of wind power generation capacity should be encouraged.

Roots are essential organs for a plant’s ability to absorb water and obtain mineral nutrients, hence they are critical to its development. Plants use root architectural alterations to improve their chances of absorbing nutrients when their supply is low. Nine root traits of a Brassica napus association panel were explored in hydroponic-system studies under low potassium (K) stress to unravel the genetic basis of root growth in rapeseed. The quantitative trait loci (QTL) and candidate genes for root development were discovered using a multilocus genome-wide association study (ML-GWAS). For the nine traits, a total of 453 significant associated single-nucleotide polymorphism (SNP) loci were discovered, which were then integrated into 206 QTL clusters. There were 45 pleiotropic clusters, and qRTA04-4 and qRTC04-7 were linked to TRL, TSA, and TRV at the same time, contributing 5.25–11.48% of the phenotypic variance explained (PVE) to the root traits. Additionally, 1360 annotated genes were discovered by examining genomic regions within 100 kb upstream and downstream of lead SNPs within the 45 loci. Thirty-five genes were identified as possibly regulating root-system development. As per protein–protein interaction analyses, homologs of three genes (BnaC08g29120D, BnaA07g10150D, and BnaC04g45700D) have been shown to influence root growth in earlier investigations. The QTL clusters and candidate genes identified in this work will help us better understand the genetics of root growth traits and could be employed in marker-assisted breeding for rapeseed adaptable to various conditions with low K levels.

This study employs 29 provincial-level administrative regions in China from 2013 to 2017 to evaluate the overall efficiency score of hydropower electricity generation in the three main regions of China. The EBM (Epsilon-based Measure) model with the DEA (Data Envelopment Analysis) method uses installed capacity data, labor force data, and equipment utilization hours as input indicators and electricity generation and CO2 emission reduction as output indicators. By comparing the efficiency values of the two indices of installed capacity and CO2 emission reduction, we are able to analyze the differences between installed hydropower electricity generation capacity efficiency and carbon emission reduction efficiency in various provinces and cities. The findings show that the western region is the best, followed by the central region and then the eastern region in terms of the input-output index level, total efficiency score of hydropower, and comparison of installed capacity-carbon emission reduction efficiency. Natural water resources and geographical advantages have a great positive effect on hydropower efficiency, while economic development has little effect on it. China should promote the sustainable development of hydropower according to local conditions and formulate and adopt countermeasures in line with the different circumstances between regions.

Agriculture is one of the three major industries in China, and its level of development occupies an important position in the development of the national economy. At present, China vigorously promotes the strategy of rural revitalization, which provides new opportunities for agricultural modernization. Since modern agriculture and regional economy are coupled in a mutually influencing and constraining relationship, this paper constructs a coupled and coordinated evaluation index system of agricultural modernization and regional economic development. Using the entropy value method and coupled coordination degree model, the comprehensive development level and coordinated development degree of agricultural modernization and regional economic development of 31 provinces, autonomous regions, and cities in China from 2011 to 2020 were evaluated, revealing the time evolution and spatial distribution characteristics of the two systems. The results show that: (1) In the past decade, China’s agricultural modernization and regional economy have made some progress in terms of their respective development levels and the degree of coupling and coordination between them. (2) In terms of spatial evolution, there are obvious regional differences between the two systems, and the coupling coordination degree shows a “ladder” distribution pattern from the east to the central, northeastern, and western parts of the country. (3) Regions with a high level of regional economic development also have a relatively high degree of coupling and coordination between agricultural modernization and regional economic development. Our results provide insights into the economic effects of agricultural modernization in China and provide useful insights into the future direction of agricultural development programs in countries around the world.

Rapeseed is the third largest oil crop in the world and the largest oil crop in China. The multi-functional development and utilization of rapeseed is an effective measure for the high-quality development of rapeseed industry in China. In this study, several basic nutrients of eight rapeseed sprouts and five bean sprouts (3–5 varieties each) were determined, including sugar, crude protein, crude fiber, vitamin E, minerals, fatty acids, amino acids, and glucosinolates. Data analysis revealed that compared with bean sprouts, rapeseed sprouts were nutritionally balanced and were richer in active nutrients such as glucose, magnesium, selenium, vitamin E, and glucosinolate. Moreover, rapeseed sprouts exhibited reasonable amino acid composition and abundant unsaturated fatty acids (accounting for 90.32% of the total fatty acids). All these results indicated the potential of rapeseed sprout as a functional vegetable. Subsequently, three dominant nutrients including vitamin E, glucosinolate, and selenium were investigated in seeds and sprouts of 44 B. napus L. varieties. The results showed that germination raised the ratio of α-tocopherol/γ-tocopherol from 0.53 in seeds to 9.65 in sprouts, greatly increasing the content of α-tocopherol with the strongest antioxidant activity among the eight isomers of vitamin E. Furthermore, germination promoted the conversion and accumulation of glucosinolate components, especially, glucoraphanin with strong anti-cancer activity with its proportion increased from 1.06% in seeds to 1.62% in sprouts. In addition, the contents of selenium, vitamin E, and glucosinolate in rapeseed sprouts were highly correlated with those in seeds. Furthermore, these three dominant nutrients varied greatly within B. napus varieties, indicating the great potential of rapeseed sprouts to be further bio-enhanced. Our findings provide reference for the multi-purpose development and utilization of rapeseed, lay a theoretical foundation for the development of rapeseed sprout into a functional vegetable, and provide a novel breeding direction.

Roots are complicated quantitative characteristics that play an essential role in absorbing water and nutrients. To uncover the genetic variations for root-related traits in rapeseed, twelve mature root traits of a Brassica napus association panel were investigated in the field within three environments. All traits showed significant phenotypic variation among genotypes, with heritabilities ranging from 55.18% to 79.68%. Genome-wide association studies (GWAS) using 20,131 SNPs discovered 172 marker-trait associations, including 103 significant SNPs (−log10 (p) > 4.30) that explained 5.24–20.31% of the phenotypic variance. With the linkage disequilibrium r2 > 0.2, these significant associations were binned into 40 quantitative trait loci (QTL) clusters. Among them, 14 important QTL clusters were discovered in two environments and/or with phenotypic contributions greater than 10%. By analyzing the genomic regions within 100 kb upstream and downstream of the peak SNPs within the 14 loci, 334 annotated genes were found. Among these, 32 genes were potentially associated with root development according to their expression analysis. Furthermore, the protein interaction network using the 334 annotated genes gave nine genes involved in a substantial number of interactions, including a key gene associated with root development, BnaC09g36350D. This research provides the groundwork for deciphering B. napus’ genetic variations and improving its root system architecture.

With the rapid development of its economy, environmental governance is becoming more important in China. The Yangtze River Economic Belt (YREB), as the world’s largest inland shipping channel, can lead the country’s regional green economy development. As most research on China’s environmental efficiency focuses on provinces or the east and west regions, this paper examines its energy input and output and environmental effects from the aspects of YREB and non-YREB, breaking through the limitations of previous studies that only used cross-section or panel data for environmental assessment. This paper employs the meta-frontier dynamic SBM model, selects fixed assets as carry-over indicators, and considers the interrelationships between the dynamics variables during 2014⁻2016. The results are as follows: The overall energy efficiency and CO2 emission efficiency of YREB are higher than those of non-YREB. The difference in energy consumption, CO2, and AQI efficiency is large, but the performance of YREB is generally better than that of non-YREB. After setting the meta-frontier, non-YREB is better than YREB, for the main reason that the technology gap values of YREB are smaller than those of non-YREB. Our findings thus suggest that YREB should strengthen technical exchanges and promotion within its region, thereby decreasing regional technology differences, while non-YREB should address environment protection and CO2 emissions and advocate a low-carbon production mode.

The role of the novel costimulatory molecule TIM4 in anti-islet response is unknown. We explored TIM4 expression and targeting in Th1 (BALB/c islets into C57BL/6 mice) and Th2 (BALB/c islets into Tbet -/- C57BL/6 mice) models of anti-islet alloimmune response and in a model of anti-islet autoimmune response (diabetes onset in NOD mice). The targeting of TIM4, using the monoclonal antibody RMT4-53, promotes islet graft survival in a Th1 model, with 30% of the graft surviving in the long term; islet graft protection appears to be mediated by a Th1 to Th2 skewing of the immune response. Differently, in the Th2 model, TIM4 targeting precipitates graft rejection by further enhancing the Th2 response. The effect of anti-TIM4 treatment in preventing autoimmune diabetes was marginal with only minor Th1 to Th2 skewing. B-Cell depletion abolished the effect of TIM4 targeting. TIM4 is expressed on human B-cells and is upregulated in diabetic and islet-transplanted patients. Our data suggest a model in which TIM4 targeting promotes Th2 response over Th1 via B-cells. The targeting of TIM4 could become a component of an immunoregulatory protocol in clinical islet transplantation, aiming at redirecting the immune system toward a Th2 response.

During an investigation of antitumor substances from Nigella glandulifera Freyn et Sint. (Ranunculaceae) the cytotoxicity of two oleanane triterpene saponins isolated from the seeds of this species, kalopanaxsaponins A and I, was evaluated against HepG2, drug resistant HepG2 (R-HepG2) (two hepatocyte cell lines) and primary cultured normal mouse hepatocytes. Evident cytotoxic activities were observed. Morphological observations and cell cycle analysis suggest that these compounds inhibit the proliferation of hepatoma by inducing apoptosis and consequently kalopanaxsaponins A and I may be potential therapeutic agents for the treatment of parental and drug resistant hepatoma.

Platelet-derived growth factor receptors (PDGFRs) have been implicated in a wide array of human malignancies, including medulloblastoma (MB), the most common brain tumor of childhood. Although significant progress in MB biology and therapeutics has been achieved during the past decades, MB remains a horrible challenge to the physicians and researchers. Therefore, novel inhibitors targeting PDGFR signaling pathway may offer great promise for the treatment of MB. In the present study, we investigated the cytotoxicity and mechanisms of cambogin in Daoy MB cells. Our results show that cambogin triggers significant S phase cell cycle arrest and apoptosis via down regulation of cyclin A and E, and activation of caspases. More importantly, further mechanistic studies demonstrated that cambogin inhibits PDGFR signaling in Daoy and genetically defined mouse embryo fibroblast (MEF) cell lines. These results suggest that cambogin is preferentially cytotoxic to cells expressing PDGFR. Our findings may provide a novel approach by targeting PDGFR signaling against MB.

The high rates of failure in oncology drug clinical trials highlight the problems of using pre-clinical data to predict the clinical effects of drugs. Patient population heterogeneity and unpredictable physiology complicate pre-clinical cancer modeling efforts. We hypothesize that gene networks associated with cancer outcome in heterogeneous patient populations could serve as a reference for identifying drug effects. Here we propose a novel in vivo genetic interaction which we call 'synergistic outcome determination' (SOD), a concept similar to 'Synthetic Lethality'. SOD is defined as the synergy of a gene pair with respect to cancer patients' outcome, whose correlation with outcome is due to cooperative, rather than independent, contributions of genes. The method combines microarray gene expression data with cancer prognostic information to identify synergistic gene-gene interactions that are then used to construct interaction networks based on gene modules (a group of genes which share similar function). In this way, we identified a cluster of important epigenetically regulated gene modules. By projecting drug sensitivity-associated genes on to the cancer-specific inter-module network, we defined a perturbation index for each drug based upon its characteristic perturbation pattern on the inter-module network. Finally, by calculating this index for compounds in the NCI Standard Agent Database, we significantly discriminated successful drugs from a broad set of test compounds, and further revealed the mechanisms of drug combinations. Thus, prognosis-guided synergistic gene-gene interaction networks could serve as an efficient in silico tool for pre-clinical drug prioritization and rational design of combinatorial therapies.

Although microarray technology has become the most common method for studying global gene expression, a plethora of technical factors across the experiment contribute to the variable of genome gene expression profiling using peripheral whole blood. A practical platform needs to be established in order to obtain reliable and reproducible data to meet clinical requirements for biomarker study.We applied peripheral whole blood samples with globin reduction and performed genome-wide transcriptome analysis using Illumina BeadChips. Real-time PCR was subsequently used to evaluate the quality of array data and elucidate the mode in which hemoglobin interferes in gene expression profiling. We demonstrated that, when applied in the context of standard microarray processing procedures, globin reduction results in a consistent and significant increase in the quality of beadarray data. When compared to their pre-globin reduction counterparts, post-globin reduction samples show improved detection statistics, lowered variance and increased sensitivity. More importantly, gender gene separation is remarkably clearer in post-globin reduction samples than in pre-globin reduction samples. Our study suggests that the poor data obtained from pre-globin reduction samples is the result of the high concentration of hemoglobin derived from red blood cells either interfering with target mRNA binding or giving the pseudo binding background signal.We therefore recommend the combination of performing globin mRNA reduction in peripheral whole blood samples and hybridizing on Illumina BeadChips as the practical approach for biomarker study.

Despite the growing understanding of pdgf signaling, studies of pdgf function have encountered two major obstacles: the functional redundancy of PDGFRalpha and PDGFRbeta in vitro and their distinct roles in vivo. Here we used wild-type mouse embryonic fibroblasts (MEF), MEF null for either PDGFRalpha, beta, or both to dissect PDGF-PDGFR signaling pathways. These four PDGFR genetically defined cells provided us a platform to study the relative contributions of the pathways triggered by the two PDGF receptors. They were treated with PDGF-BB and analyzed for differential gene expression, in vitro proliferation and differential response to pharmacological effects. No genes were differentially expressed in the double null cells, suggesting minimal receptor-independent signaling. Protean differentiation and proliferation pathways are commonly regulated by PDGFRalpha, PDGFRbeta and PDGFRalpha/beta while each receptor is also responsible for regulating unique signaling pathways. Furthermore, some signaling is solely modulated through heterodimeric PDGFRalpha/beta.
