As the most common cause of bacillary dysentery or shigellosis, Shigella sonnei (S nonnei) has spread throughout the world. Invasion of the colorectal epithelial cells by this facultative intracellular bacterium occurs via various virulence factors. The increase in the resistance rate highlights the need for novel interventions, particularly increasing the urgency of the development of Shigella vaccines that may offer an effective solution. A multiepitope protein vaccine (MEPV) construct previously designed using bioinformatics tools against Shigella species, was applied in vivo in BALB/C mice. The designed vaccine construct was expressed in a bacterial host, purified, and finally confirmed by Western blot analysis. The immunogenicity of the purified MEPV was assessed against S sonnei via intranasal and subcutaneous administration routes, followed by evaluating its protective efficiency. We observed that interferon-gamma, interleukin-4, and immunoglobulin G levels were increased in all experimental groups. Therefore, The MEPV effectively protected the mice against S sonnei. The design, synthesis and biological study of several novel 1,2,3-triazolyl $$\upbeta $$ -hydroxy alkyl/carbazole hybrid molecules as a new type of antifungal agent has been described. In this synthesis, the N-alkylation reaction of carbazol-9-ide potassium salt with 3-bromoprop-1-yne afforded 9-(prop-2-ynyl)-9 H-carbazole. The 'Click' Huisgen cycloaddition reaction of 9-(prop-2-ynyl)-9 H-carbazole with diverse $$\upbeta $$ -azido alcohols in the presence of copper-doped silica cuprous sulphate led to target molecules in excellent yields. The in vitro antifungal and antibacterial activities of title compounds were screened against various pathogenic fungal strains, Gram-positive and/or Gram-negative bacteria. In particular, 1-(4-((9 H-carbazol-9-yl) methyl)-1 H-1,2,3-triazol-1-yl)-3-butoxypropan-2-ol ( 10e) proved to have potent antifungal activity against all fungal tests compared with fluconazole and clotrimazole as studied reference drugs. Our molecular docking analysis revealed an appropriate fitting and a potential powerful interaction between compound 10e and an active site of the Mycobacterium P450DM enzyme. The strong hydrogen bondings between $$\upbeta $$ -hydroxyl and ether groups in 10e were found to be the main factors that drive the molecule to fit in the active site of enzyme. While aging inevitably changes our skin, this complex biological process involves much more than just getting older. As the body's largest organ, the skin constantly safeguards us from harmful environmental pathogens and plays a key role in overall well-being. This study investigated the development and evaluation of a nanogel containing Rosa damascena essential oil for its potential anti-aging properties. The nanogel was prepared from a primary nanoemulsion with a particle size of 86 ± 4 nm. Antioxidant activity, measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, was found to be 76% ± 3%. Furthermore, the nanogel demonstrated superior anti-collagenase and anti-elastase activities (60% ± 2% and 51% ± 0.1%, respectively) compared to the essential oil alone. The antibacterial efficacy of the nanogel was tested against Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa, revealing potent inhibitory effects. To simulate the aging process, rats were subjected to UVB irradiation on both legs 4 days a week before or after treatment. In vivo studies conducted on mice showed that the nanogel effectively reduced the formation of deep wrinkles in treated groups compared to pretreated ones.
