BACKGROUND: Alternaria can infest pears to produce metabolites, which can contaminate pears and their processed products. Pear paste, one of the most important pear‐based products, is popular among Chinese consumers especially for its cough relieving and phlegm removal properties. Although people are concerned about the risk of Alternaria toxins in many agro‐foods and their products, little is known about the toxins in pear paste. RESULTS: A method was developed for the determination of tenuazonic acid, alternariol, alternariol menomethyl ether, altenuene and tentoxin in pear paste by ultra‐performance liquid chromatography tandem mass spectrometry with saturated sodium sulphate dissolution and acidified acetonitrile extraction. The mean recoveries of the five toxins were 75.3–113.8% with relative standard deviations of 2.8–12.2% at spiked levels of 1.0–100 μg kg−1. Alternaria toxins were detected in 53 out of 76 samples, with a detection rate of 71.4%. Tenuazonic acid (67.1%), alternariol (35.5%), tentoxin (23.7%) and alternariol monomethyl ether (7.9%) were detected in all samples at concentrations of < limit of quantification (LOQ)–105.0 μg kg−1, < LOQ–32.1 μg kg−1, < LOQ–74.2 μg kg−1 and < LOQ–15.1 μg kg−1, respectively. Altenuene was never found in pear paste samples. Tenuazonic acid, alternariol, tentoxin and alternariol menomethyl ether should be focused on due to their toxicity and detection rates. CONCLUSION: To the best of our knowledge, this is the first report on the detection method and residue levels of Alternaria toxins in pear paste. The proposed method and research data can provide technical support for the Chinese government to continuously monitor and control Alternaria toxins in pear paste, especially tenuazonic acid. Cavity-free air lasing offers a promising route towards the realization of atmospheric lasers for various applications such as remote sensing and standoff spectroscopy; however, achieving efficient generation and control of air lasing in ambient air is still a challenge. Here we show the experimental realization of a giant lasing enhancement by three to four orders of magnitude in ambient air for the self-seeded N 2+ lasing at 428 nm, assigned to the B 2 Σ u +( ν ' =0) and X 2 Σ g +( ν ' ' =1) emission, by modulating the spatiotemporal overlap of ultrashort near-infrared control-pump pulses in a filamentary plasma grating; meanwhile, the spontaneous emission from the same transition is only enhanced by three to four times. We find that this enhancement is sensitive to the relative polarization and interference time of the two pulses, and reveal that the formation of the plasma grating induces different population variations in the B 2 Σ u +( ν ' =0) and X 2 Σ g +( ν ' ' =1) levels, resulting in an enormous population inversion between the two levels, thereby a higher gain for the giant enhancement of N 2+ lasing in ambient air.

We experimentally demonstrate an asymmetric enhancement of the N 2 + lasing at 391 nm for the transition between the B 2 Σu + (v = 0) and X 2 Σg + (v" = 0) states in an intense laser field with the ellipticity, ε, modulated by a 7-order quarter-wave plate (7-QWP). It is found that when the 7-QWP is rotated from α = 0 to 90°, where α is the angle between the polarization direction of the input laser and the slow axis of the 7-QWP, the intensity of the 391-nm lasing is optimized at ε ∼ 0.3 with α∼ 10°-20° and 70°-80° respectively, but the optimization intensity at α∼ 10°-20° is about 4 times smaller than that at α∼ 70°-80°. We interpret the asymmetric enhancement based on a post-ionization coupling model, in which the birefringence of the 7-QWP induces an opposite change in the relative amplitudes of the ordinary (Eo) and extraordinary (Ee) electric components under the two conditions, so that the same temporal separation of Eo and Ee leads to a remarkably different coupling strength for the population transfer from the X 2 Σg + (v "=0) to A 2 Π u (v '=2) states.

Context: The DUOX/DUOXA systems play a key role in H 2 O 2 generation in thyroid cells, which is required for iodine organification and thyroid hormone synthesis. DUOX2/DUOXA2 defects can cause congenital hypothyroidism (CH), but it is unknown whether DUOX1/DUOXA1 mutations can also cause CH. Objective: We aimed to identify DUOX1/DUOXA1 mutations and explore their role in the development of CH by investigating their functional impacts on H 2 O 2 generation. Patients and Methods: Forty-three children with CH with goiter were enrolled, in whom all exons and flanking intronic regions of DUOX1/DUOXA1 were directly sequenced. We characterized the functional effects of identified mutations on the expression of DUOX1 and DUOXA1 and H 2 O 2 generation. Results: We identified a heterozygous DUOX1 missense mutation (G > A base substitution at nucleotide 3920 in exon 31) that changed a highly conserved arginine to glutamine at residual 1307 (p.R1307Q) in patient 1. A heterozygous-missense mutation (c.166 C>T; p.R56W) was identified in DUOXA1 in patient 2. Functional studies demonstrated that both p.R1307Q mutant or p.R56W mutant decreased the DUOX1 expression at mRNA and protein levels, with a corresponding impairment in H 2 O 2 generation ( P < 0.01). The results also showed that intact DUOXA1 was required for full activity of DUOX1 and H 2 O 2 generation. Conclusions: We have identified two heterozygous missense mutations in DUOX1 and DUOXA1 in two patients that can cause CH through disrupting the coordination of DUOX1 and DUOXA1 in the generation of H 2 O 2 . This study for the first time demonstrates that the DUOX1/DUOXA1 system, if genetically defective, can cause CH.

Background: Thyroid dysgenesis (TD) is the most frequent cause of congenital hypothyroidism (CH), but its pathogenesis remains unclear. As a thyroid transcription factor, paired box transcription factor 8 (PAX8) is essential for thyroid organogenesis and development., Aim: To screen PAX8 mutations and characterize the features of these mutations in Chinese TD patients., Materials and Methods: Blood samples were collected from 63 TD patients in Shandong Province, China, and genomic DNA was extracted from peripheral blood leukocytes. Exon 3~4 of PAX8 were analyzed by PCR and direct sequencing., Results: Direct sequencing of PAX8 revealed a heterozygous missense mutation (c.155G/C, P.Arg52Pro) in one child with agenesis. Genetic screening of the child's family revealed that the clinically unaffected parents do not carry the mutation, suggesting that the identified sequence change is a de novo mutation., Conclusion: We report a heterozygous missense de novo mutation in PAX8 in one out of 63 unrelated Chinese TD patients, showing that the PAX8 mutation rate is very low in TD patients in China. However, de novo mutation and epigenetic mechanisms need to be considered in the future study.
