The aim is to translate and cross-culturally adapt the HAAS into a Chinese version (HAAS-C) and to evaluate the psychometric properties of HAAS-C in patients after primary total knee arthroplasty (TKA).Methods: A total of 104 patients diagnosed with knee osteoarthritis who had undergone TKA at least 12 months prior were recruited. A forward and backward translation procedure was performed for developing a culturally acceptable HAAS-C. Internal consistency was assessed using Cronbach’s α, and test-retest reliability was measured using the intraclass correlation coefficient (ICC) within a 10-day interval. Construct validity was assessed by examining the correlations between HAAS-C and the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC), EuroQoL Group’s five-dimension questionnaire (EQ-5D-5L), and Oxford knee score (OKS).Results: HAAS-C demonstrated adequate Internal consistency reliability, as indicated by Cronbach’s α coefficient of 0.75. Test-retest reliability yielded excellent results, with an ICC value of 0.98. Content validity indices were high, with a scale-level validity index of 0.9 and item-level validity indices greater than or equal to 0.8. HAAS-C showed a strong correlation with WOMAC (r = 0.69), a moderate correlation with EQ-5D-5L (r = 0.43), and OKS (r = 0.53) while exhibiting no floor or ceiling effects.Conclusion: The validated HAAS-C questionnaire is a valid instrument for assessing patients undergoing TKA in mainland China.Keywords: high activity arthroplasty score, total knee arthroplasty, Chinese validation, reliability, validity

Abstract The Central Plains Han Chinese (CPHC) is the typical agricultural population of East Asia. Investigating the genome of the CPHC is crucial to understanding the genetic structure and adaptation of the modern humans in East Asia. Here, we perform whole genome sequencing of 492 CPHC individuals and obtained 22.65 million SNPs, 4.26 million INDELs and 41,959 SVs. We found the CPHC has a higher level of genetic diversity and the glycolipid metabolic genes show strong selection signals, e.g. LONP2, FADS2, FGF21 and SLC19A2. Ancient DNA analyses suggest that the domestication of crops, which drove the emergence of the candidate mutations. Notably, East Asian-specific SVs, e.g., DEL_21699 (LINC01749) and DEL_38406 (FAM102A) may be associated with the high prevalence of esophageal squamous carcinoma and primary angle-closure glaucoma. Our results provide an important genetic resource and show that dietary adaptations play an important role in phenotypic evolution in East Asian populations.

Abstract Ru single-atom catalysts have great potential to replace toxic mercuric chloride in acetylene hydrochlorination. However, long-term catalytic stability remains a grand challenge due to the aggregation of Ru atoms caused by over-chlorination. Herein, we synthesize an asymmetric Ru-In atomic pair with vinyl chloride monomer yield (>99.5%) and stability (>600 h) at a gas hourly space velocity of 180 h−1, far surpassing those of the Ru single-atom counterparts. A combination of experimental and theoretical techniques reveals that there is a strong d-p orbital interaction between Ru and In atoms, which not only enables the selective adsorption of acetylene and hydrogen chloride at different atomic sites but also optimizes the electron configuration of Ru. As a result, the intrinsic energy barrier for vinyl chloride generation is lowered, and the thermodynamics of the chlorination process at the Ru site is switched from exothermal to endothermal due to the change of orbital couplings. This work provides a strategy to prevent the deactivation and depletion of active Ru centers during acetylene hydrochlorination.

Objectives To compare the surgical and functional outcomes of the 120 W HPS and 180 W XPS GreenLight laser vaporization systems in the treatment of benign prostatic hyperplasia (BPH). Methods In January 2024, a comprehensive search across PubMed, Cochrane Library, and EMBASE was conducted following PRISMA guidelines, registered under PROSPERO (CRD42024531176). Studies comparing 120 W and 180 W GreenLight lasers in BPH treatment were assessed for clinical outcomes. Results Eight studies were included. The 180 W XPS system improved operation time (MD: 12.70, 95% CI [5.29–20.11], p = 0.0008), lasing duration (MD: 10.09, 95% CI [0.85–19.33], p = 0.03), and catheterization duration (MD: 0.43, 95% CI [0.12–0.74], p = 0.007). No significant differences in energy consumption, energy density, or length of hospital stay were found. Functional outcomes such as International Prostate Symptom Score and maximum urinary flow rate showed no significant differences, except in quality of life (MD: 0.43, 95% CI [0.06–0.80], p = 0.02) and prostate-specific antigen levels (MD: −0.77, 95% CI [−1.28 to −0.25], p = 0.003). The 180 W system exhibited a lower rate of overall (OR: 1.52, 95% CI [1.14–2.04], p = 0.005) and minor complications (OR: 1.84, 95% CI [1.27–2.66], p = 0.001), with no significant differences in major complications or other adverse events. Conclusions The 180 W XPS system demonstrates enhanced efficiency and reduced complication rates, offering a favorable option for BPH treatment, particularly for larger prostates. Future studies should focus on randomized trials to confirm these findings and assess long-term outcomes.

Abstract Capturing gaseous mercury (Hg0) from sulfur dioxide (SO2)-containing flue gases remains a common yet persistently challenge. Here we introduce a low-temperature sulfur chemical vapor deposition (S-CVD) technique that effectively converts SO2, with intermittently introduced H2S, into deposited sulfur (Sd 0) on metal sulfides (MS), facilitating self-sustained adsorption of Hg0. ZnS, as a representative MS model, undergoes a decrease in the coordination number of Zn–S from 3.9 to 3.5 after Sd 0 deposition, accompanied by the generation of unsaturated-coordinated polysulfide species (Sn 2–, named Sd *) with significantly enhanced Hg0 adsorption performance. Surprisingly, the adsorption product, HgS (ZnS@HgS), can serve as a fresh interface for the activation of Sd 0 to Sd * through the S-CVD method, thereby achieving a self-sustained Hg0 adsorption capacity exceeding 300 mg g−1 without saturation limitations. Theoretical calculations substantiate the self-sustained adsorption mechanism that S8 ring on both ZnS and ZnS@HgS can be activated to chemical bond S4 chain, exhibiting a stronger Hg0 adsorption energy than pristine ones. Importantly, this S-CVD strategy is applicable to the in-situ activation of synthetic or natural MS containing chalcophile metal elements for Hg0 removal and also holds potential applications for various purposes requiring MS adsorbents.

The accuracy of traditional topic models may be compromised due to the sparsity of co-occurring vocabulary in the corpus, whereas conventional word embedding models tend to excessively prioritize contextual semantic information and inadequately capture domain-specific features in the text. This paper proposes a hybrid semantic representation method that combines a topic model that integrates conceptual knowledge with a weighted word embedding model. Specifically, we construct a topic model incorporating the Probase concept knowledge base to perform topic clustering and obtain topic semantic representation. Additionally, we design a weighted word embedding model to enhance the contextual semantic information representation of the text. The feature-based information fusion model is employed to integrate the two textual representations and generate a hybrid semantic representation. The hybrid semantic representation model proposed in this study was evaluated based on various English composition test sets. The findings demonstrate that the model presented in this paper exhibits superior accuracy and practical value compared to existing text representation methods.

Highlights A facile chelated coupled pyrolysis strategy was reported to prepare Mn single-cluster Hg0 adsorbent. The nitrogen-containing group in biomass determined the size of atomically dispersed manganese. The presence of oxygen and small size Mn clusters in biochar promoted Hg0 adsorption.

The present study examined the relationships between soil characteristics, microbial community structure and function in the forests artificially planted with exotic Sonneratia apetala at stand ages of 1-, 2-, 7-, 10- and 14-years and Sonneratia caseolaris of 1-, 4-, 7-, 10- and 14-years in Futian National Nature Reserve, Shenzhen Bay, China. The 7-years old forests of both Sonneratia species reached peak growth and had the highest content of nitrogen and phosphorus, enzymatic activities, including dehydrogenase, cellulase, phosphatase, urease and β-glucosidase, except arylsulphatase which increased continuously with stand ages. The microbial community structure reflected by phospholipid fatty acid (PLFA) profiles also reached the maximum value in the 7-years old forests and soil bacterial PLFAs in both forests were significantly higher than fungal PLFAs. The canonical correlation analysis revealed that differences in microbial structural variables were significantly correlated to the differences in their functional variables, and the highest correlation was found between the soil enzymatic activities and the content of carbon and nitrogen. The effect of different concentrations of NaCl, 0, 100, 200, 300 and 400 mM, on the anatomical features and physiology of Myoporum bontioides was investigated. The photosynthetic rates (Pn) were significantly reduced by salt stress, with the lowest values at 400 mM NaCl. The content of malondialdehyde (MDA), proline and soluble sugar, as well as the activities of peroxidase (POD) and catalase (CAT) increased at the beginning, but became similar to the control as the experiment proceeded. The NaCl effect on superoxide dismutase (SOD) was different from the other parameters, with a significant reduction at 400 mM NaCl at Day 7. Salt glands were found in both upper and lower epidermis, and the ratios of the thickness of palisade to spongy mesophyll tissues increased with NaCl concentrations. The medullary ray was clearly damaged by NaCl at levels of 200 and 300 mM. These results demonstrated that M. bontioides could adapt to a relatively low salinity, and was not a halophilous species. PPy-Fe3O4/Kaolin was prepared with polypyrrole functionalized magnetic Kaolin by a simple, green, and low cost method to improve the agglomeration and low adsorption capacity of Kaolin. PPy-Fe3O4/Kaolin was employed to remove Hg2+ and the results were characterized by various methods. Relevant factors, including solution pH, dosage of adsorbent, concentration (C0), and temperature (T), were optimized by Response Surface Methodology (RSM) and Central Composite Designs (CCD). The optimal results show that the importance for adsorption factors is pH > T > C0 > dosage, and the optimal adsorption conditions of PPy-Fe3O4/Kaolin are pH = 7.2, T = 315 K, C0 = 50 mg/L, dosage of 0.05 g/L, and the capacity is 317.1 mg/g. The adsorption process conforms to the pseudo-second-order and Langmuir models. Dubinin–Radushkevich model shows that adsorption process is spontaneous and endothermic. Moreover, the adsorption of mercury by PPy-Fe3O4/Kaolin was achieved mainly through electrostatic attraction, pore diffusion, and chelation between amino functional groups and Hg2+. PPy-Fe3O4/Kaolin has excellent reproducibility, dispersity, and chemical stability, and it is easy to be separated from solution through an external magnetic field. The experiments show that PPy-Fe3O4/Kaolin is an efficient and economical adsorbent towards mercury.

In order to avoid using toxic or harmful operational conditions, shorten synthesis time, enhance adsorption capacity, and reduce operational cost, a novel magnetic nano-adsorbent of CoFe2O4@SiO2 with core–shell structure was successfully functionalized with polypyrrole (Ppy). The physical and chemical properties of CoFe2O4@SiO2-Ppy are examined by various means. The as-prepared CoFe2O4@SiO2-Ppy nanomaterial was used to adsorb Hg2+ from water. During the process, some key effect factors were studied. The adsorption process of Hg2+ onto CoFe2O4@SiO2-Ppy was consistent with the pseudo-second-order kinetic and Langmuir models. The Langmuir capacity reached 680.2 mg/g, exceeding those of many adsorbents. The as-prepared material had excellent regeneration ability, dispersibility, and stability. The fitting of kinetics, isotherms, and thermodynamics indicated the removal was endothermic and spontaneous, and involved some chemical reactions. The application evaluation of electroplating wastewater also shows that CoFe2O4@SiO2-Ppy is an excellent adsorbent for Hg2+ ions from water.

To avoid the dangerous operational conditions, shorten the preparation time, and improve the adsorption performance of amino-functionalized nanomagnetic materials with a core–shell structure, a magnetic nanocomposite of CoFe2O4@SiO2 was successfully functionalized with amino group (−NH2) through a mild and facile hydrothermal method without the use of any toxic or harmful solvents at a relatively low temperature. The preparation time of the key steps of amino functionalization was shortened from 30 h to about 10 h. The core-shell structure and successful grafting were confirmed by various means. The amino-functionalized CoFe2O4@SiO2 was used for the removal mercury (Hg(II)), a heavy metal, and exhibited excellent magnetic properties and a high Langmuir adsorption capacity of 149.3 mg Hg(II)/g. The adsorption of Hg(II) onto CoFe2O4@SiO2–NH2 followed the pseudo-second-order kinetic equation and Langmuir model. The thermodynamic data showed that the Hg(II) adsorption process was achieved through spontaneous exothermic and monolayer adsorption with electrostatic adsorption and chemisorption. In addition, the as-prepared CoFe2O4@SiO2–NH2 nanoparticles had a good reusable value, good application performance and stability, and can provide a mild and facile way to remove heavy metals from aqueous solution.

Purpose: The purpose of this investigation is to assess and contrast the effectiveness of the two EuroQol five dimensions questionnaire (EQ-5D) versions-EQ-5D-3L and EQ-5D-5L-in assessing one-year quality of life outcomes for patients with knee osteoarthritis (KOA) undergoing unicompartmental knee arthroplasty (UKA)., Material and Method: From the medical records at the Honghui Hospital, Xi'an Jiaotong University, 402 individuals aged 50 and above, who were one-year post-operation, were selected to fill out survey questionnaires during their return hospital visits. Of these, 231 respondents (57.5%) completed the questionnaire; 228 completed both versions, and 56 completed the EQ-5D retest questionnaire. The assessment included missing data, ceiling effects, informativity and discriminatory power, as well as response consistency, redistribution properties, and inconsistency. Reliability and validity were also evaluated., Results: The results indicate that the EQ-5D-5L surpasses the EQ-5D-3L in construct validity, informativity, detection precision, and discriminatory power. Consistency reliability is also better in the EQ-5D-5L than in the EQ-5D-3L. Both instrument versions maintained reliable levels of test-retest reliability., Conclusion: In patients with KOA undergoing UKA, the EQ-5D-5L has proven superior in measurement capabilities when compared with the EQ-5D-3L one-year post-operation. The early diagnosis of liver injury and in situ real-time monitoring of tumor therapy efficacy are important for the enhancement of personalized precision therapy but remain challenging due to the lack of reliable in vivo visualization tools with integrated diagnostic, therapeutic, and efficacy monitoring functions. Herein, a smart second near-infrared window (NIR-II) molecule (BITX-OH) is rationally designed for diagnosis and therapy by vinyl-bridging hydroxyl diphenyl xanthine unit and benzo[cd]indolium skeleton. BITX-OH exhibits high selectivity and sensitivity toward viscosity, exhibiting a significant enhancement (1167-fold) in NIR-II fluorescence at 962 nm. With the assistance of BITX-OH and NIR-II fluorescence imaging, early diagnosis and therapeutic evaluation of non-alcoholic fatty liver (NAFL), as well as in-site real-time monitoring of hepatic fibrosis (HF) in live mice have been successfully achieved, which is at least several hours earlier than the typical clinical test. Notably, BITX-OH displays excellent photothermal conversion efficiency when exposed to an 808 nm laser, which can induce tumor ablation and increase viscosity, thereby enhancing NIR-II fluorescence for the real-time evaluation of photothermal therapy (PTT). This viscosity-based "self-monitoring" strategy provides a convenient and reliable platform for timely obtaining therapeutic feedback to avoid over- or under-treatment, thus enabling personalized precision therapy., (© 2024 Wiley‐VCH GmbH.)

Mitophagy is a key pathway for regulating mitochondrial quality and quantity which is essential for the preservation of cellular homeostasis. Mitophagy process may be accompanied by changes of the mitochondrial microenvironments. The multifunctional fluorescent probe is crucial for the precise detection of multiple microenvironments, which is vital for the visualization of mitophagy. Herein, a mitochondria-immobilized fluorescent probe DPP was designed and fabricated to visualize mitophagy by monitoring polarity and viscosity in dual-channel. The DPP is characterized by "D-π-A″ structure, which provides the basis for the intramolecular charge transfer (ICT) and twisted intramolecular charge transfer (TICT) platform, enabling dual-channel responses to polarity and viscosity at emission wavelengths of 487 nm and 656 nm, respectively. The significant wavelength gap (169 nm) between the above channels prevents signal crosstalk. Additionally, the incorporation of 1, 4-dibenzyl chloride grants the probe mitochondrial immobilization capabilities, avoiding the leak of probe due to mitochondrial depolarization during autophagy. The DPP accumulates in mitochondria and monitors polarity and viscosity changes in green and red channels, respectively. Notably, the investigation of the relationship between polarity and viscosity revealed that an increase in viscosity is accompanied by a decrease in polarity. The mitophagy was effectively observed through the induction of DPP by rapamycin, with a particular emphasis on the increase in viscosity and decrease in polarity. Purpose: To investigate the clinical efficacy and prognostic factors associated with autologous osteoperiosteal transplantation for the treatment of single cystic osteochondral lesions of the talus (OLT)., Methods: The clinical data of patients with single cystic OLT undergoing autologous osteoperiosteal transplantation at the Department of Foot and Ankle Surgery of our hospital between 2018 and 2022, including complete follow-up, were retrospectively analyzed. Imaging data from each patient were imported into Mimics software to measure the surface area, volume and depth of the lesions. Then, the talus nine-compartment partitioning method was used to partition the injury site. Preoperative and final follow-up assessments were performed using the American Orthopaedic Foot and Ankle Society (AOFAS) score, visual analogue scale (VAS) for pain and 36-item Short-Form Health Survey (SF-36) to evaluate treatment efficacy and analyze prognostic factors., Results: Of the 31 patients with single cystic OLT with a complete set of follow-up data, there were 17 males and 14 females, with a mean age of 43.3 ± 13.6 years, a mean follow-up time of 30.1 ± 14.0 months and a mean illness duration of 30.4 ± 20.0 months. The postoperative final follow-up AOFAS score was 90.7 ± 5.5; this represented significant improvement when compared to the preoperative score of 57.0 ± 8.5 (P < 0.001). The final postoperative follow-up VAS score was 18.5 ± 8.3; this was significantly better than the preoperative score of 57.8 ± 8.7 (P < 0.001). The physical component summary (PCS) score and mental component summary (MCS) score on the SF-36 scale showed significant improvement at the final postoperative follow-up when compared to preoperative scores (p < 0.001). No other complications were observed during follow-up, such as wound infection or pain at the donor site. One of the patients showed less improvement, which may be related to premature weight-bearing or re-sprained ankle after surgery. There was no significant correlation between the duration of illness, gender and the location, depth, surface area and volume of the OLT and the postoperative scores. However, patient age showed a significant negative correlation with the postoperative SF-36 PCS and MCS scores., Conclusion: Autologous osteoperiosteal transplantation for single cystic OLT demonstrated good clinical efficacy with a low incidence of complications. Purpose: This study aims to conduct a network meta-analysis to compare the clinical efficacy of seven distinct non-pharmacological therapies for knee osteoarthritis. We hope that our research findings can provide reference for clinical practitioners in formulating treatment plans., Methods: Through a computer-based search, we systematically retrieved randomized controlled trials (RCTs) on non-pharmacological therapies for knee osteoarthritis from eight databases, including CNKI, Wanfang, VIP, PubMed, Web of Science, Embase, Scopus, and The Cochrane Library. Following screening, data extraction, and methodological quality assessment, relevant data were included and analyzed using R 4.2.3 software., Results: A comprehensive analysis of 24 RCTs involving 2582 patients encompassed seven diverse non-pharmacological therapies. The efficacy rankings, based on Visual Analog Scale (VAS) scores, were as follows: shock wave therapy > needle-knife > laser therapy > acupuncture > ultrasound > exercise > transcutaneous electrical nerve stimulation. Similarly, based on Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) total scores, the efficacy rankings were as follows: shock wave therapy > needle-knife > laser therapy > acupuncture > ultrasound > transcutaneous electrical nerve stimulation > exercise. Among the three WOMAC subscales, the efficacy rankings for non-pharmacological therapies were as follows: For stiffness: laser therapy > exercise > shock wave therapy > acupuncture > needle-knife > ultrasound > transcutaneous electrical nerve stimulation; For daily activities: shock wave therapy > laser therapy > needle-knife > acupuncture > ultrasound > transcutaneous electrical nerve stimulation > exercise; For pain: shock wave therapy > needle-knife > laser therapy > acupuncture > exercise > transcutaneous electrical nerve stimulation > ultrasound., Conclusion: Based on the currently limited research, we can prioritize the use of shockwave therapy to treat patients with knee osteoarthritis. Hydrogen peroxide (H 2 O 2 ) overexpressed in mitochondria has been regarded as a key biomarker in the pathological processes of various diseases. However, there is currently a lack of suitable mitochondria-targetable near-infrared (NIR) probes for the visualization of H 2 O 2 in multiple diseases, such as PM 2.5 exposure-induced lung injury, hepatic ischemia-reperfusion injury (HIRI), nonalcoholic fatty liver (NAFL), hepatic fibrosis (HF), and malignant tumor tissues containing clinical cancer patient samples. Herein, we conceived a novel NIR fluorescent probe ( HCy-H 2 O 2 ) by introducing pentafluorobenzenesulfonyl as a H 2 O 2 sensing unit into the NIR hemicyanine platform. HCy-H 2 O 2 exhibits good sensitivity and selectivity toward H 2 O 2 , accompanied by a remarkable "turn-on" fluorescence signal at 720 nm. Meanwhile, HCy-H 2 O 2 has stable mitochondria-targetable ability and permits monitoring of the up-generated H 2 O 2 level during mitophagy. Furthermore, using HCy-H 2 O 2 , we have successfully observed an overproduced mitochondrial H 2 O 2 in ambient PM 2.5 exposure-induced lung injury, HIRI, NAFL, and HF models through NIR fluorescence imaging. Significantly, the visualization of H 2 O 2 has been achieved in both tumor-bear mice as well as surgical specimens of cancer patients, making HCy-H 2 O 2 a promising tool for cancer diagnosis and imaging-guided surgery.

Purpose: This study aims to explore the clinical value of autogenous tibial periosteal bone grafting in the treatment of osteochondral lesions of the talus (OLT) and analyze the three-dimensional factors in the necrotic zone of the talus., Methods: A retrospective analysis was performed on 36 patients who underwent autogenous tibial periosteal bone grafting in the Foot and Ankle Surgery Department of our hospital between September 2018 and September 2022. The American Orthopaedic Foot and Ankle Society (AOFAS), Visual Analogue Scale (VAS), and Chinese Short-Form 36 Health Survey (SF-36) were used to evaluate treatment efficacy prior to surgery and at the last follow-up. Furthermore, Mimics 21.0 software was employed to measure the three-dimensional data of the necrotic area, including surface area, volume, and depth, in order to investigate their potential impact on patient prognosis., Results: Among the 36 OLT patients who obtained complete follow-up, there were 22 males and 14 females. No complications such as surgical site infection, non-union of cartilage, post-traumatic arthritis, or donor site pain were observed. The AOFAS, VAS, and Chinese SF-36 scores of all patients at the last follow-up showed significant improvement compared to preoperative values. There was no significant correlation between the AOFAS, VAS, and Chinese SF-36 scores at the last follow-up and the depth, surface area, and volume of the necrotic zone., Conclusion: The use of autogenous tibial periosteal bone grafting can safely and effectively treat Hepple V OLT. Purpose: This study aims to investigate the psychological status of patients with Hepple V osteochondral lesions of the talus (OLT) and evaluate the effect of autologous osteoperiosteal transplantation (AOPT) on their psychological well-being., Methods: Fifty patients with Hepple V OLT who underwent AOPT at the Comprehensive Foot and Ankle Surgery Ward of Xi'an Honghui Hospital from November 2021 to May 2023 were included in this study. The patients were divided into two groups based on the presence or absence of preoperative symptoms of anxiety/depression. Group A comprised patients with preoperative symptoms, while Group B included patients without such symptoms. Preoperative and final follow-up assessments included the Hospital Anxiety and Depression Scale for evaluating anxiety and depression, the visual analogue scale for pain assessment, and the American Orthopaedic Foot and Ankle Society scores for assessing ankle and hindfoot function., Results: Among the 50 Hepple V OLT patients who obtained complete follow-up, twenty-four had preoperative symptoms of anxiety/depression, with an incidence rate of up to 48%. As an important endogenous gasotransmitter, hydrogen sulfide (H 2 S) plays a critical role in various physiological functions and has been regarded as a biomarker of cancer due to its overexpression in cancer cells. In addition, the early stages of cancer are often accompanied by abnormalities in the intracellular microenvironments, and distinguishing between cancer cell/tissues and normal cell/tissues is of great significance to the accuracy of cancer diagnosis. However, deep insights into the simultaneous detection of H 2 S and viscosity/polarity variations in cancer cells/tissues are rarely reported. In this work, we designed and synthesized a mitochondria-targeting fluorescent probe PDQHS, which exhibits high selectivity for H 2 S with an emission peak around 632 nm and excellent response (17-fold) to viscosity/polarity beyond 706 nm. Meanwhile, PDQHS shows good biocompatibility and can specifically accumulate into mitochondria. Using PDQHS, the visual distinguishing of cancer cells from normal cells was achieved via dual-channel detection of H 2 S and viscosity/polarity. More importantly, PDQHS has been successfully applied to visualize endogenous and exogenous H 2 S in living cells and tumor tissue. Obviously, compared to the detection of a single biomarker, monitoring multiple biomarkers simultaneously through dual-channel response is conducive to amplifying the detection signal, providing a more sensitive and reliable imaging tool in the tumor region, which is beneficial for cancer prediction., (© 2024. Background: In recent decades, there have been notable advancements in the field of analgesia and total knee arthroplasty (TKA). This study aims to employ bibliometric analysis to elucidate the prevailing research focal points and trends within analgesia and TKA from 1990 to 2022., Material and Methods: Relevant publications were retrieved from the Web of Science Core Collection. CiteSpace, VOSviewer, and Scimago Graphica were used for visualization and bibliometric analysis of countries, institutions, authors, journals, references, and keywords., Results: A total of 2776 publications on analgesia and TKA were identified, with the United States having the highest number of publications. The University of Copenhagen was the most productive institution, and Kehlet, Henrik was the most prolific author. The Journal of Arthroplasty had the most publications and citations. The most common keywords were "TKA," "pain management," "postoperative pain," "Total hip arthroplasty (THA)," and "postoperative management." Keyword burst detection demonstrated that adductor canal block (ACB) was a recent research hotspot., Conclusion: Our study revealed a sharp increase in global publications on analgesia and TKA, and this trend is expected to continue. Hydrogen sulfide (H 2 S), a significant gas signal molecule, is closely related to various physiological/pathological processes. The monitoring of H 2 S is crucial in understanding the occurrence and development of diseases such as cancers. Emerging evidence suggests that abnormal regulation of Lipid droplets (LDs) is associated with many human diseases. For example, cancer cells are characterized by the abnormal accumulation of LDs. Therefore, understanding the relationship between LDs and cancer is of great significance for developing therapies against cancer. To address this challenge, we designed and developed a LD-targeting and H 2 S-activated probe (BTDA-DNB) by engineering a 2,4-dinitrophenyl ether (DNBE) as the H 2 S reactive site. In the presence of H 2 S, a strongly fluorescent emitter, 3-(benzo[d]thiazol-2-yl)-N,N-diethyl-2-imino-2H-chromen-7-amine (BTDA) was obtained with the leaving of DNBE group. BTDA-DNB displayed favorable sensitivity, selectivity and functioning well at physiological pH. The probe features excellent LD-targeting specificity and low cellular toxicity. The practical applications of LD-targeting probe BTDA-DNB as H 2 S probe in living cells, cancer tissues and Arabidopsis seedling have been evaluated. The excellent imaging performance demonstrates a potential ability for cancer diagnosis. Purpose: Frailty is a difficult-to-measure condition that is susceptible to adverse outcomes. The Japan Frailty Scale (JFS) is a tool for assessing frailty status in older adults. This study aimed to translate and culturally adapt the JFS into a Chinese version (JFS-C)., Materials and Methods: The study included 160 older adults as participants. Internal consistency was assessed using Cronbach's alpha, and test-retest reliability was conducted using the intraclass correlation coefficient (ICC). Convergent validity was evaluated by assessing the correlation between JFS-C and the Barthel Index, the Frail scale, and the 36-item Short-Form Health Survey (SF-36). Criterion validity was assessed by comparing JFS-C scores with the Frail scale., Results: JFS-C demonstrated adequate internal consistency (Cronbach's alphas = 0.711) and excellent test-retest reliability over a 7 to 10-day interval (ICC = 0.949). Correlation analysis showed a strong positive correlation between JFS-C and the Frail scale ( r  = 0.786, p  < 0.001), a moderate negative correlation with the Barthel Index ( r  = -0.598, p  < 0.001), and moderate correlations with various subscales of SF-36 ( r  = -0.574 to -0.661). However, no significant correlations were found between JFS-C and SF-36 mental health ( r  = -0.363, p  < 0.001) or role emotional ( r  = -0.350, p  < 0.001). The present study aimed to introduce the research status and hotspots and explore the field of robotic-assisted knee arthroplasty from a bibliometric perspective. The Web of Science Core Collection database was utilized to retrieve articles and reviews on robotic-assisted knee arthroplasty published between 1993 and 2023. CiteSpace, VOSviewer, Scimago Graphica, Pajek, and a bibliometric online analysis platform (http://bibliometric.com/) were employed to analyze the regions, institutions, journals, authors, and keywords, aiming to predict the latest trends in research related to robotic-assisted knee arthroplasty. This study encompasses 697 records. The annual publication count pertaining to robotic-assisted knee arthroplasty demonstrates consistent growth. The United States leads with the highest number of studies (298), trailed by the United Kingdom (110) and France (49). The Hospital for Special Surgery emerges as the most prolific institution, while Professor Mont, Michael A holds significant author influence. The Journal of Arthroplasty reigns supreme in this field, boasting the highest publication and citation figures. Funding sources predominantly include Stryker (34), Smith Nephew (19), and the National Natural Science Foundation of China (17). Lysosomal viscosity is an essential microenvironment parameter in lysosomes, which is closely associated to the occurrence and development of various diseases, including cancer. Thus, accurately quantifying lysosomal viscosity changes is highly desirable for a better understanding of the dynamics and biological functions of lysosomes. In this study, lysosome self-targetable orange-red emissive carbon dots (OR-CDs) were rationally designed and developed for monitoring lysosomal viscosity fluctuations. The enhanced fluorescence of OR-CDs could be obviously observed as the viscosity increased from 1.07 to 950 cP. Moreover, the as-prepared OR-CDs could quickly enter cells for lysosome-targeting imaging and visualize viscosity variations in living cells and zebrafish. More importantly, by utilizing OR-CDs, we successfully achieved tracing the variations in lysosomal viscosity during the autophagy process. Additionally, as cancer cells possess high viscosity than normal cells, the OR-CDs have been effectively utilized for cancer imaging from cell, tissue, and organ to in vivo levels. It is expected that the developed OR-CDs not only provide a meaningful tool for visualizing investigations of lysosome viscosity-related diseases but also shed light on the development based on the nanomaterial for the clinical diagnosis of cancer.

Abnormal changes occurring in the mitochondrial microenvironment are important markers indicating mitochondrial and cell dysfunction. Herein, we designed and synthesized a multifunctional fluorescent probe DPB that responds to polarity, viscosity, and peroxynitrite (ONOO - ). DPB is composed of an electron donor (diethylamine group) and electron acceptor (coumarin, pyridine cations, and phenylboronic acid esters), in which the pyridine group with a positive charge is responsible for targeting to mitochondria. D-π- A structure with strong intramolecular charge transfer (ICT) and twisted intramolecular charge transfer (TICT) properties give rise to respond to polarity and viscosity. The introduction of cyanogroup and phenylboronic acid esters increases the electrophilicity of the probe, which is prone to oxidation triggered by ONOO - . The integrated architecture satisfies the multiple response requirements. As the polarity increases, the fluorescence intensity of probe DPB at 470 nm is quenched by 97%. At 658 nm, the fluorescence intensity of DPB increases with viscosity and decreases with the concentration of ONOO - . Furthermore, the probe is not only successfully used to monitor mitochondrial polarity, viscosity, and endogenous/exogenous ONOO - level fluctuations but also to distinguish cancer cells from normal cells by multiple parameters. Therefore, as-prepared probe provides a reliable tool for better understanding of the mitochondrial microenvironment and also a potential approach for the diagnosis of disease.

Monitoring the variations of lipid droplet (LD) polarity is of great significance for the investigation of LD-related cellular metabolism and function. We hereby report a lipophilic fluorescent probe (BTHO) with the feature of intramolecular charge transfer (ICT) for imaging the LD polarity in living cells. BTHO exhibits an obvious attenuation of fluorescence emission in response to the increase of environmental polarity. The linear response range of BTHO to polarity ( ε , the dielectric constant of solvents) is derived to be 2.21-24.40, and the fluorescence of BTHO in glyceryl trioleate falls in this range. Furthermore, BTHO has high molecular brightness, which may effectively improve the signal to noise ratio, along with the decrease of phototoxicity. BTHO exhibits excellent photostability and targeting capability to LDs with low cytotoxicity, which is satisfactory in long-term imaging in live cells. The probe was successfully applied for imaging LD polarity variation in live cells caused by oleic acid (OA), methyl-β-cyclodextrin (MβCD), H 2 O 2 , starvation, lipopolysaccharide (LPS), nystatin, and erastin. The low crosstalk caused by viscosity to BTHO measuring the LD polarity was confirmed from a calculation result.

Intracellular peroxynitrite anions (ONOO - ) and microenvironments (such as viscosity and polarity) play an important role in maintaining redox homeostasis, regulating diffusion, transportation, and signal transduction in living cells. The abnormality of these factors is often closely related to various physiological/pathological processes. However, owing to the lack of suitable probes, the simultaneous visualization of ONOO - , viscosity, and polarity in ferroptosis and cancer models has not been achieved. To meet urgent needs, we presented a multifunctional near-infrared (NIR) fluorescent probe, named MQA-P , for simultaneously detecting ONOO - , viscosity, and polarity within mitochondria. The probe exhibited a remarkable turn-on response to ONOO - with the far-red emission of about 645 nm and was highly sensitive to viscosity/polarity in the NIR channel with λ em > 704 nm. Facilitated by MQA-P , for the first time, we revealed that erastin-induced ferroptosis was accompanied by a significant upregulation of ONOO - and an increase of viscosity (or decrease of polarity) at the same time. Moreover, the concurrent use of ONOO - , viscosity, and polarity for the diagnosis of cancer has been successfully achieved not only at cell/tissue levels but also in tumor mice models. Compared with detecting only one factor, this simultaneous detection of multimarkers provides a more sensitive and reliable method/tool for tracking ferroptosis-related pathological processes and cancer diagnosis, holding great potential in preclinical research, medical diagnosis, and imaging-guided surgery.

Ethyl carbamate (EC) is mainly found in fermented foods and fermented alcoholic beverages, which could cause carcinogenic potential to humans. Reducing EC is one of the key research priorities to address security of fermented foods. Enzymatic degradation of EC with EC hydrolase in food is the most reliable and efficient method. However, poor tolerance to ethanol severely hinders application of EC hydrolase. In this study, the mutants of EC hydrolase were screened by diphasic high pressure molecular dynamic simulations (dHP-MD). The best variant with remarkable improvement in specific activity and was H68A/K70R/S325N, whose specific activity was approximately 3.42-fold higher than WT, and relative enzyme activity under 20% (v/v) was 5.02-fold higher than WT. Moreover, the triple mutant increased its stability by acquiring more hydration shell and forming extra hydrogen bonds. Furthermore, the ability of degrading EC of the immobilized triple mutant was both detected in mock wine and under certain reaction conditions. The stability of immobilized triple mutant and WT were both improved, and immobilized triple mutant degraded nearly twice as much EC as that of immobilized WT. In order to conduct in-depth research on the mechanisms of cancer diagnosis and treatment, it is very important to develop fluorescent probes to study the interactions between different organelles and understand the relationship between various organelles and cell viability. However, the lack of fluorescent probes to visualize two or more targets has resulted in limited studies of intracellular interactions between different organelles. To this end, in this work, we developed a near-infrared (NIR) AIE probe with dual-color emission, NAP-Py-E, for mitochondria and lipid droplets imaging. The probe NAP-Py-E consists of lipophilic fraction, pyridine cation structure and esterase hydrolysis site. Interestingly, NAP-Py-E first targets mitochondria and emits red fluorescence; after partially hydrolyed by esterase in living cells, the hydrolysate NAP-Py accumulates in lipid droplets and emits green fluorescence. Protein-glutaminase plays a significant role in future food (e.g., plant-based meat) processing as a result of its ability to improve the solubility, foaming, emulsifying, and gel properties of plant-based proteins. However, poor stability, activity, high pressure, and high shear processing environments hinder its application. Therefore, we developed an application-oriented method isothermal compressibility perturbation engineering strategy to improve enzyme performance by simulating the high-pressure environment. The best variant with remarkable improvement in specific activity and half-time, N16M/Q21H/T113E, exhibited a 4.28-fold increase compared to the wild type in specific activity (117.18 units/mg) and a 1.23-fold increase in half-time (472 min), as one of the highest comprehensive performances ever reported. The solubility of the soy protein isolate deaminated by the N16M/Q21H/T113E mutant was 55.74% higher than that deaminated by the wild type, with a tinier particle size and coarser texture. Overall, this strategy has the potential to improve the functional performance of enzymes under complex food processing conditions.

Ferroptosis, as a new regulated mode of cell death, is mainly characterized by iron-dependent accumulation of lipid peroxides related to reactive oxygen species (ROS). However, the changes of peroxynitrite (ONOO - ) during ferroptosis are not very clear due to the limited reports. On the other hand, ONOO - has become an endogenous toxicant leading to cell apoptosis and necrosis. Thus, it is urgent to develop molecular tools with high selectivity and sensitivity to monitor intracellular ONOO - . Herein, we presented a fluorescent probe, BTMO-PN, by introducing a phenylboronic acid ester as the ONOO - recognition site into benzothiazolyl derivative. BTMO-PN exhibited a rapid and marked fluorescence enhancement signal toward ONOO - , owing to the ONOO - -triggered the cleavage of phenylboronic acid ester to release strongly fluorescent BTMO. Moreover, BTMO-PN could image endogenous and exogenous ONOO - changes in live cells. Importantly, using BTMO-PN, we demonstrated the up-generation of ONOO - levels in cancer cells during ferroptosis. A water-soluble fluorescent probe BPN, by introducing a piperazine as the pH-sensitive fluorescence signaling motif to the hydrophilic propionic acid-substituted 1,8-naphthalimide fluorophore, is highly sensitive to pH changes within cytoplasm matrix in living cells, as well as pH-related diseases models. Owing to the protonation-induced inhibition of the photoinduced electron transfer (PET) from piperazine to naphthalimide fluorophore, BPN displayed a significant fluorescence enhancement (more than 131-fold) upon the pH decreasing from 11.0 to 3.0. The linear range was between pH 6.4 to 8.0 with a pK a value of 6.69 near the physiological pH, which was suitable for cytosolic pH research. Furthermore, BPN exhibited a large Stokes shift (142 nm), good water solubility, excellent photostability, high selectivity and low cytotoxicity. All these advantages were particularly beneficial for intracellular pH imaging. Using BPN, we demonstrated the real-time monitoring of cytosolic pH changes in living cells. Lipid droplets (LDs) have recently been reported as an attractive target for cancer diagnosis and treatment, owing to their special structure or microenvironment changes in cancer development and resistance. However, the relationship between the biothiol level of LDs and cancer is still poorly understood, partially owing to the absence of effective molecular tools. Herein, we developed a LD-targetable and biothiol-sensitive fluorescent probe, BTDA-RSS, by introducing 2,4-dinitrobenzenesulfonyl (DNBS) as the biothiol reaction group into a benzothiazolyl derivative. BTDA-RSS displayed a marked and rapid fluorescence turn-on response toward biothiols, due to the biothiol-triggered cleavage of DNBS to yield the highly fluorescent benzothiazolyl iminocoumarin BTDA. In addition, the probe shows significant LD-targetable ability, and has been applied for imaging endogenous/exogenous biothiol changes in LDs. Importantly, BTDA-RSS has successfully been utilized to distinguish cancerous cells/tissues from normal cells/tissues with excellent contrast. Surprisingly, we demonstrated for the first time the visualization of LD biothiols in surgical specimens from cancer patients, thereby holding great potential for the application of BTDA-RSS in the clinical diagnosis of human cancers.

Elucidating the intrinsic relationship between diseases and lipid droplet (LD) polarity remains a great challenge owing to the lack of the research on multiple disease models. Until now, the visualization of abnormal LD polarity in models of inflammation and clinical cancer patient samples has not been achieved. To meet the urgent challenge, we facilely synthesized a robust LD-specific and polarity-sensitive fluorescent probe ( LD - TTP ), which consists of a triphenylamine segment as an electron-donor group (D) and a pyridinium as an electron-acceptor moiety (A), forming a typical D-π-A molecular configuration. Owing to the unique intramolecular charge transfer effect, LD-TTP exhibits high sensitivity to polarity change in the linear range from Δ f = 0.258 to 0.312, with over 278-fold fluorescence enhancement. Moreover, we revealed that LD-TTP possessed satisfactory ability for sensitively monitoring LD-polarity changes in living cells. Using LD-TTP , we first demonstrated the detection of LD-polarity changes in fatty liver tissues and inflammatory living mice via confocal laser scanning fluorescence imaging. Surprisingly, the visualization of LD polarity has been achieved not only at the cellular levels and living organs but also in surgical specimens from cancer patients, thus holding great potential in the clinical diagnosis of human cancer. All these features render LD-TTP an effective tool for medical diagnosis of LD polarity-related diseases.

Heat stroke is a fatal condition which usually results in central nervous system dysfunction, organism damage and even death. The relationship between heat stroke and mitochondria is still relatively unknown due to a lack of suitable tools. Herein, an aggregation-induced emission (AIE) probe CSP, by introducing a pyridinium cation as the mitochondria-targeted group to an AIE active core cyanostilbene skeleton, is highly sensitive to viscosity changes due to the restriction of intramolecular motion (RIM) and inhibition of twisted intramolecular charge transfer (TICT) in high-viscosity systems. As expected, with the viscosity increasing from 0.903 cP (0% glycerol) to 965 cP (99% glycerol), CSP exhibited a significant enhancement (more than 117-fold) in fluorescence intensity at 625 nm, with an excellent linear relationship between log I 625 nm and log η (R 2  = 0.9869, slope as high as 0.6727). More importantly, using CSP we have successfully monitored the decreased mitochondrial viscosity during heat stroke for the first time. All these features render the probe a promising candidate for further understanding the mechanism underlying mitochondria-associated heat stroke.

The aim of this paper was to investigate the preventive and therapeutic effects of Xiaoer Feike Granules(XEFK) on chronic bronchitis in rats and its mechanism. Except for 10 rats in the blank group, the remaining 50 of the 60 SD rats were used to establish a model of chronic bronchitis induced by LPS. On the 22 nd day, the model rats were randomly divided into 5 groups according to their body weight, and administrated with purified water, Keteling Capsules 0.11 g·kg~(-1), XEFK 3.2, 1.6 and 0.8 g·kg~(-1)(the dosing concentrations were 0.32, 0.16, 0.08 g·mL~(-1), respectively). These rats took the corresponding drug orally once a day, for consecutive 21 days. The rats were anesthetized 1 hour after the last administration, and the lavage bronchus and alveoli were collected. Then, after the fixation of the smear, neutrophils were counted microscopically, and the contents of glutathione peroxidase(GSH-Px), superoxide dismutase(SOD) and malondialdehyde(MDA) in the bronchoalveolar lavage fluid(BALF) were detected by colorimetric method. Flow cytometry was used to detect the content changes of T cell subsets CD4~+, CD8~+, CD4~+/CD8~(+ )in serum. Hemorheology related indexes were detected by automatic hemorheology. Enzyme-linked immunosorbent assay(ELISA) was used to detect the contents of tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), IL-2, IL-6 and IL-10 in serum. The expression of TNF-α and IL-10 mRNA in lung was detected by Real-time quantitative PCR(RT-qPCR). HE staining was used to observe the pathological changes in the bronchitis tissues. Compared with the model group, XEFK high and medium dose groups could significantly reduce the contents of neutrophils and MDA in bronchial lavage fluid, and increase the activities of GSH-Px and SOD in BALF, and repair the chronic inflammatory cell infiltration and lymphoid tissue hyperplasia in the bronchial mucosal layer and submucosal layer. The high-dose group could reduce the plasma viscosity of rats, but there was no statistical difference in other hemorheological indexes. CD4~+, CD8~+, CD4~+/CD8~+, IL-2 and IL-10 contents in each dose group were significantly increased, and TNF-α, IL-1β and IL-6 contents were significantly decreased in serum. Each dose group could significantly down-regulate the expression level of TNF-α mRNA in the lung and increase the expression of IL-10 mRNA. XEFK could reduce lipid peroxidation, increase the content of peripheral blood T cell subsets, regulate the release and secretion of inflammatory factors, and repair the morphological and pathological changes of bronchial tissue. Its mechanism might be related to the improvement of inflammatory response and the enhancement of immune function.

Mitophagy plays a crucial role in maintaining intracellular homeostasis through the removal of dysfunctional mitochondria and recycling their constituents in a lysosome-degradative pathway, which leads to microenvironmental changes within mitochondria, such as the pH, viscosity, and polarity. However, most of the mitochondrial fluorescence viscosity probes only rely on electrostatic attraction and readily leak out from the mitochondria during mitophagy with a decreased membrane potential, thus easily leading to an inaccurate detection of viscosity changes. In this work, we report a mitochondria-immobilized NIR-emissive aggregation-induced emission (AIE) probe CS-Py-BC , which allows for an off-on fluorescence response to viscosity, thus enabling the real-time monitoring viscosity variation during mitophagy. This system consists of a cyanostilbene skeleton as the AIE active core and viscosity-sensitive unit, a pyridinium cation for the mitochondria-targeting group, and a benzyl chloride subunit that induces mitochondrial immobilization. As the viscosity increased from 0.903 cP (0% glycerol) to 965 cP (99% glycerol), CS-Py-BC exhibited an about 92-fold increase in fluorescence intensity at 650 nm, which might be attributed to the restriction of rotation and inhibition of twisted intramolecular charge transfer in a high viscosity system. We also revealed that CS-Py-BC could be well immobilized onto mitochondria, regardless of the mitochondrial membrane potential fluctuation. Most importantly, using CS-Py-BC , we have successfully visualized the increased mitochondrial viscosity during starvation or rapamycin-induced mitophagy in real time. All these features render CS-Py-BC a promising candidate to investigate mitophagy-associated dynamic physiological and pathological processes.

Near-infrared spectroscopy(NIRS) combined with band screening method and modeling algorithm can be used to achieve the rapid and non-destructive detection of the traditional Chinese medicine(TCM) production process. This paper focused on the ginkgo leaf macroporous resin purification process, which is the key technology of Yinshen Tongluo Capsules, in order to achieve the rapid determination of quercetin, kaempferol and isorhamnetin in effluent. The abnormal spectrum was eliminated by Mahalanobis distance algorithm, and the data set was divided by the sample set partitioning method based on joint X-Y distances(SPXY). The key information bands were selected by synergy interval partial least squares(siPLS); based on that, competitive adaptive reweighted sampling(CARS), successive projections algorithm(SPA) and Monte Carlo uninformative variable(MC-UVE) were used to select wavelengths to obtain less but more critical variable data. With selected key variables as input, the quantitative analysis model was established by genetic algorithm joint extreme learning machine(GA-ELM) algorithm. The performance of the model was compared with that of partial least squares regression(PLSR). The results showed that the combination with siPLS-CARS-GA-ELM could achieve the optimal model performance with the minimum number of variables. The calibration set correlation coefficient R&#95;c and the validation set correlation coefficient R&#95;p of quercetin, kaempferol and isorhamnetin were all above 0.98. The root mean square error of calibration(RMSEC), the root mean square error of prediction(RMSEP) and the relative standard errors of prediction(RSEP) were 0.030 0, 0.029 2 and 8.88%, 0.041 4, 0.034 8 and 8.46%, 0.029 3, 0.027 1 and 10.10%, respectively. Compared with the PLSR me-thod, the performance of the GA-ELM model was greatly improved, which proved that NIRS combined with GA-ELM method has a great potential for rapid determination of effective components of TCM.
