Abstract

Background and aims The use of root-associated microorganisms emerge as a sustainable tool to enhance crop tolerance and productivity under climate change, particularly in drought-affected areas. Here, the impact of an inoculum based on arbuscular mycorrhizal fungi (AMF) was evaluated on pepper (Capsicum annuum L.) cultivation at varying water irrigation treatments (well-watered, reduced irrigation and rain-fed) under open-field conditions. Methods Agronomic and ecophysiological parameters, as well as biochemical analyses on stress markers and phytohormones in leaves and on fruit quality traits, were evaluated, along with the shifts in soil- and root-associated microbial communities. Results Rain-fed water treatment caused reduced fruit sizes, while no differences were detected among well-watered and reduced irrigation. Reduced irrigation did not cause a reduction in stomatal conductance. The highest AM fungal colonization rates were observed under reduced irrigation, and the enhanced flavonoid content and reduced oxidative stress markers in AMF-inoculated plants suggested a synergistic effect of AM fungal inoculation in boosting plant tolerance against stress. A shift in microbial community composition in the different irrigation treatments, associated with different enzymatic activity, highlighted the potential role of microbial dynamics in plant stress response under water-limited conditions. Conclusion The study suggests that a reduced irrigation comes along with beneficial impacts on pepper root associated microbes, while not impairing crop performance and yields, indicating a potential of saving water. All together, our results imply that optimization of irrigation and beneficial plant-microbe interactions, such as AM fungal symbiosis, can improve pepper physiological and productivity features under climate change.

Abstract

A major goal in ecology is to understand how interactions among organisms influence ecosystem services. This work compares the effects of two Lepidoptera defoliators, one non-native (Hyphantria cunea) and one native (Lymantria dispar) to Europe, on the colonization of black poplar (the Populus nigra clone “Jean Pourtet”) roots by an arbuscular mycorrhizal (AM) symbiotic fungus (Funneliformis mosseae) in a pot experiment. The effects of defoliation have also been assessed on the expression of fungal and plant genes playing a role during symbiosis. Both control and defoliated poplars have shown a low level of mycorrhization. Additionally, neither the non-native nor the native insect seem to strongly affect the AM colonization, at least at the time of observation (eight days from the end of the defoliation). Concerning the gene expression analysis, our results suggest that defoliation does not influence neither the expression of genes coding for a fungal and a plant phosphate transporter nor that of a gene coding for a fungal ATPase, and that there were no differences between defoliation carried out by the non-native and the native insect.

Abstract

Incidence and risk factors associated with the development of pressure ulcers in an Italian general intensive care unit. INTRODUCTION: Pressure ulcers (PU) represent one of the most frequent adverse event in intensive care units (ICU). Critical patients are at higher risk of developing a PU, with an incidence between 3.3-39.3%. AIM: To assess the incidence and risk factors for developing PUs in a general ICU of an Italian University hospital. METHODS: Retrospective observational study on a sample of 122 patients admitted to a general intensive care unit, from January to December 2015, with a length of ICU stay > 5 days. PUs were stadied according to the National Pressure Ulcer Advisory Panel. RESULTS: One hundred and twenty two patients were enrolled. The incidence of PUs was 33%, and the incidence rate was 23 PUs for 1.000 days of ICU stay; 29% of PUs were of stage I, 56% of stage II, 8% stage III and 6% stage IV. PUs mainly affected the sacrum (21%) with a very low incidence in the heels (1.6%). Despite several factors were associated to the development of PUs (being female, Sequential Organ Failure Assessment, length of stay, mechanical ventilation > 72 hours, administration of dobutamine, Admission pO 2/FiO2 ratio, mean Braden during ICU stay and admission diagnosis) only ICU lenght of day remained statistically significant. CONCLUSIONS: The incidence of PUs is comparable to other recent studies. Many factors are potentially involved in the onset of PUs. Identifying these factors can help nurses to plan interventions to prevent their development.

16. Who is out there? What are they doing? Application of metagenomics and metaproteomics to reveal soil functioning

Abstract

In order to link microbial community composition to ecological processes happening in the brûlé, a metaproteomics analysis was applied to a brûlé previously characterized by metagenomics. The metagenomics data had showed a reduced fungal biodiversity, a dominance of Tuber melanosporum and a reduced presence both of ectomycorrhizal Basidiomycota and of bacteria belonged to Pseudomonas and Flavobacteriaceae inside the brûlé. By metaproteomics analysis, the identified proteins revealed which biological processes were more represented or only present in the brûlé, and, among them, processes related to multiple stresses were identified in herbaceous plants. This study demonstrates that combining the data of metagenomics and metaproteomics gives the opportunity to potentially reveal the functioning of any environment.

20. A local adaptive method for the numerical approximation in seismic wave modelling

Abstract

We propose a new numerical approach for the solution of the 2D acoustic wave equation to model the predicted data in the field of active-source seismic inverse problems. This method consists in using an explicit finite difference technique with an adaptive order of approximation of the spatial derivatives that takes into account the local velocity at the grid nodes. Testing our method to simulate the recorded seismograms in a marine seismic acquisition, we found that the low computational time and the low approximation error of the proposed approach make it suitable in the context of seismic inversion problems.

21. Global Optimization Procedure to Estimate a Starting Velocity Model for Local Full Waveform Inversion

Abstract

Finding an efficient procedure to solve a seismic inversion problem, such as Full Waveform Inversion, still remains an open question. This is mainly due to the non linearity of the misfit function, characterized by the presence of multiple local minima, that cause unsuccessful results of local optimization strategies when the starting model is not in the basin of attraction of the global minimum. Therefore, the use of a global optimization strategy in order to estimate a suitable starting velocity model for Full Waveform Inversion is a crucial point. In this work we propose a new method which is based on the application of the Adaptive Simulated Annealing algorithm using a coarse inversion grid, different from the domain modelling one, that allows the reduction of the number of unknowns. Numerical results show that the application of our strategy to an acoustic Full Waveform Inversion provides a good starting model for a local optimization procedure, lying in the basin of attraction of the global minimum.

Abstract

More than 90% of terrestrial plants form root interactions with mycorrhizal fungi that provide mineral nutrients in exchange for carbon compounds. In particular, arbuscular mycorrhizal (AM) symbiosis involves Glomeromycota fungi and the majority of plants, including forest tree species such as poplars (Populus spp.). Plants can interact with defoliators that might affect carbon availability, thus influencing mycorrhizal symbiosis. The increasing threat of invasive species, among which are several defoliators, raises a question about their possible impact on the components of native ecosystems. This work compares the effect of two Lepidoptera defoliators, one invasive (Hyphantria cunea) and one native (Limantria dispar) on poplar colonization by an AM fungus (Funneliformis mosseae). In detail, we evaluated the effect of both partial and total defoliation by larvae of the two species i) on the colonization of black poplar plants (P. nigra Jean Pourtet) by F. mosseae and ii) on the expression of fungal genes playing a role during symbiosis: an amino acid permease (GmosAAP1); a phosphate transporter (GmosPT) and two different H -ATPases (GmHA5, GmPMA1). Both control and defoliated poplars showed a low level of mycorrhization, as already shown by previous works, and no significant differences have been found among the five considered treatments (control plants; partial and total defoliation by H. cunea; partial and total defoliation by L. dispar). Concerning gene expression in the mycorrhizae, GmosPT and GmHA5 were not differently expressed in control and defoliated plants (p>0.05). GmosAAP1, previously reported as expressed in the extra-radical mycelium, was detected in RT-PCR, although it was not possible to quantify its transcripts in quantitative PCR (qPCR). This may suggest the presence of a little amount of extra-radical mycelium in poplar roots. Similarly, GmPMA1 transcripts were detected in one replicate of the defoliated samples using RT-PCR, but not in qPCR. These results show that neither the invasive nor the native insect do not affect the AM colonization, at least after of the interval of considered time. We cannot exclude that a longer time after defoliation may be needed to affect fungal colonization. In addition, an analysis of further fungal genes (e.g., hexose transporter) could be useful to obtain a more complete picture, but, unfortunately, F. mosseae genome has not been sequenced and available sequences are still limited.

Abstract

Tunisian desert truffles have been identified on the basis of the internal transcribed spacer (ITS) locus as Tirmania nivea, Terfezia bouderii and T. arenaria. ITS and, for the first time, β-tubulin gene were employed in the phylogenetic analyses of these species to assess whether β-tubulin gene could be a complementary molecular tool for resolution of phylogenetic species. From their comparison, both markers showed to be suitable to distinguish Tirmania nivea from Terfezia bouderii and T. arenaria and, focusing on the T. boudieri complex, to assign the phylogenetic types previously described. In addition to give a first glimpse into the distribution of desert truffles from north to south of Tunisia, we then suggest to use the β-tubulin gene as a complementary marker to ITS. This indication can be useful above all for the phylogenetic reconstruction of the T. boudieri complex. [ABSTRACT FROM AUTHOR]

Abstract

The QDF (Quality of life, Depression and cognitive Function) is a project elaborated and implemented by a Research Network of the nursing personnel of the cardiological services which promoted and run the GISSI-HF trial. It is a "companion study" which included up to 1/3 of all the GISSI-HF centres.To describe the distribution and evaluation of QDF variables measured with standardised instruments (Kansas City Cardiomiopathy Quality of Life questionnaire, KCCQ; Geriaric Depression scale GDS, Mini Mental State Examination MMSE), to verify whether and how far their knowledge could influence clinical decisions; to assess the prognostic impact of QDF values measured at entrance on mortality and morbidity. The study protocol included also a comprehensive evaluation of QDF variables independent from the questionnaires, by the nurses in charge of the study patients (the results of this part of the protocol are not included in this report).The QDF data were collected at baseline, 6 months, 1, 2 and 3 years through two self administered questionnaires (KCCQ, GDS), while the MMSE only for patients70 years. The questionnaires were administered to the patients who accepted to participate. A specific goal of the project focuses on the degree of independence between the results of the questionnaires and the judgement of the nurses. This contribution presents the operational implementation of the study protocol and general framework.

Abstract

To document the degree of reliability of validated and mainly qualitative questionnaires most often used in research settings and/or projects, in a broad network of unselected cardiological centres; to assess whether the quality of data could be compatible with their use not only as descriptive but also in prognostic scores including mainly "hard" clinical variables.Eighty-three patients were included. Compliance with the self-administration of the Kansas City Cardiomiopathy Questionnaire (KCCQ) and the Geriatric Depression Scale (GDS) was very high throughout the study (KCCQ 97.7% baseline and 85.8% at three years). As expected more difficulties were encountered with the Mini Mental State Examination (administered to 80.6% patients baseline and to 54.3% at three years). Main reasons for not administering questionnaires was ascribed to organizational and patients problems.It is worth noticing that the study was conducted by nurses without economical incentives. The questionnaires were well completed with minimal missing data. If questionnaires are well presented to patients, a routine assessment of quality of life and depression could be feasible in everyday care.

Abstract

Aim of this paper is to explore and quanti-quantitatively assess whether QoL as measured with the Kansas City Cardiomiopathy Questionnaire (KCCQ) summary score could be considered as an independent relevant component of clinical prognostic score of morbidity and mortality and identify patients at risk for death or admissions.Sixty-three per cent NYHA II and 39.4% NYHA III-IV patients experience a good Qol (score75). Risk factors for "not good" (75) QoL are age, NYHA class III-IV, diabetes, COPD and previous hospitalizations. NYHA II and III-IV patients with not good QoL experience an higher mortality than patients with a good QoL in the same classes. NYHA II patients with not good QoL experience the same 1 year readmission rates as NYHA III-IV patients with good QoL.Quality of life scores identify patients with different risk of mortality and readmissions within the same NYHA class. The prognostic value of KCCQ summary scores could identify candidates for disease management in whom better targeted care strategies may reduce hospitalizations and prevent deaths.

Abstract

This contribution describes the patients included for the QDF (Quality of life, Depression and Cognitive functions) study and explores the distribution and time trends of scores of the three dimensions.The large majority of patients are males (78.3%), and mean age is 67.2 +/- 10.7 years (9.2%80 years). Patients in NYHA class III-IV experience more depression and a worse QoL as compared to the less severly clinically affected counterparts in NYHA class. Although QoL and depression are influenced by the same variables, not all patients with bad QoL are depressed and viceversa. Mean scores of cognitive functioning are not influenced by the same variables that influence QoL and depression.The regular measurement of QoL, depression and cognitive function is recommended by some authors. It seems to be confirmed that the three dimensions need to be explored to disentangle the specific informative value that each of them could add to clinical measures.

Abstract

The prevalence of cognitive impairment (CI) is reported to be higher in heart failure patients than in the general population.To describe the prevalence of CI and to assess its prognostic value on the increased risk of hospitalizations and death over the medium term (3 years).MMSE was administered to 620/745 heart failure patients70 years. Up to 35% of the tested population were classified as mild CI and 12% as moderate. The severity of baseline clinical conditions, as expressed as NYHA classes, do not appear to be associated to different degrees of CI. The subgroup of patients with moderate CI includes higher proportion of people with the lowest educational level, of feminine gender, of higher age (80). Even in a population closely monitored as the one included in the GISSI-HLF trial, it is worth to be underlined that the presence of moderate CI appears to be strongly prognostic of an increased burden of care (hospitalizations), and of death over the whole observation period of the study (3 years).The presence of even mild or moderate CI is prognostic for increased mortality and readmissions for patients of the same NYHA class. In the absence of effective intervention strategies, research should concentrate on the yield, if any, of a systematic screening for CI in heart failure patients.

Abstract

Depression is common in patients with heart failure but its causal and/or prognostic role with respect to the cardiac condition in still a matter of research.To describe the prevalence of depression and to assess its specific prognostic value in terms of increased risk of hospitalizations or death.Overall, 27.5% of 1495 patients experience symptoms of depression. Risk factors for depression are female sex, age70, higher NYHA class, previous hospitalizations and comorbidities. Depressed patients of the same NYHA class are at higher risk for 1 and 3 years mortality and all causes hospital admissions.Depression, assessed with a simple and easy to administer instrument, can add information to clinical variables in the identification of patients at increased risk of mortality and readmissions within the same NYHA class.

Abstract

The huge diversity of fungi may reflect both the heterogeneity of the niches they occupy and the diverse stresses they must cope with. In order to investigate the genetic and functional diversity in the ericoid mycorrhizal fungus Oidiodendron maius subjected to heavy metal stress, we isolated O. maius strains from a serpentine site naturally enriched by heavy metals. Despite the high Cr and Ni soil concentrations, a high level of diversity was found in the serpentine fungal community. The growth of these isolates in the presence of different metal contaminants identified some tolerant strains, suggesting a site-specific adaptation. To investigate within-species gene divergence in stressful environments, we then compared the sequence polymorphism of a neutral (internal transcribed spacer) and a functional (Cu,ZnSOD) gene in O. maius isolates derived from the serpentine site, from a site heavily polluted with industrial wastes and from unpolluted sites. For all isolates tested, the polymorphism was higher in the nucleotide sequence of the functional gene. However, when compared with isolates from the serpentine area, isolates from industrially polluted sites showed a significantly higher polymorphism in the Cu,ZnSOD promoter region, suggesting that environmental stress may influence the rate of mutations in specific regions of the Sod1 locus.

Abstract

Characterization of genomic variation among different microbial species, or different strains of the same species, is a field of significant interest with a wide range of potential applications. We have investigated the genomic variation in mycorrhizal fungal genomes through genomic suppressive subtractive hybridization.The comparison was between phylogenetically distant and close truffle species (Tuber spp.), and between isolates of the ericoid mycorrhizal fungus Oidiodendron maius featuring different degrees of metal tolerance. In the interspecies experiment, almost all the sequences that were identified in the Tuber melanosporum genome and absent in Tuber borchii and Tuber indicum corresponded to transposable elements. In the intraspecies comparison, some specific sequences corresponded to regions coding for enzymes, among them a glutathione synthetase known to be involved in metal tolerance. This approach is a quick and rather inexpensive tool to develop molecular markers for mycorrhizal fungi tracking and barcoding, to identify functional genes and to investigate the genome plasticity, adaptation and evolution.
