There were no major advances in conventional treatments in inhibiting tumor progression and increasing patient survival time. In order to suppress mechanisms responsible for tumor cell development such as those with oncogenic roles, more advanced therapeutic strategies should be sought. One of the most important oncogenes of pancreatic cancer is the MYC gene. The overexpression of MYC can activate many tumorigenic processes such as cell proliferation and pancreatic cancer cell invasion. MiRNAs are important molecules that are confirmed by targeting mRNA transcripts to regulate the expression of the MYC gene. Therefore, restoring MYC-repressing miRNAs expression tends to be an effective method of treating MYC-driven cancers.Objective: The purpose of this study was to identify all validated microRNAs targeting C-MYC expression to inhibit PDAC progression by conducting a systematic review.Methods: In this systematic review study, the papers published between 2000 and 2020 in major online scientific databases including PubMed, Scopus, and Web of Science were screened, following inclusion and exclusion criteria. We extracted all the experimental studies that showed miRNAs could target the expression of the MYC gene in PDAC.Results: Eight papers were selected from a total of 89 papers. We found that six miRNAs (Let-7a, miR-145, miR-34a, miR-375, miR-494, and miR-148a) among the selected studies were validated for targeting MYC gene and three of them confirmed Let-7a as a direct MYC expression regulator in PC cells. Finally, we summarized the latest shreds of evidence of experimentally validated miRNAs targeting the MYC gene with respect to PDAC’s therapeutic potential.Conclusion: Restoring the expression of MYC-repressing miRNAs tends to be an effective way to treat MYC-driven cancers such as PDAC. Several miRNAs have been proposed to target this oncogene via bioinformatics tools, but only a few have been experimentally validated for pancreatic cancer cells and models. Further studies should be conducted to find the interaction network of miRNA-MYC to develop more successful therapeutic strategies for PC, using the synergistic effects of these miRNAs.Keywords: micro RNA, pancreatic cancer, MYC, cancer therapy

The method of data collection was done by interviewing farmers directly (respondents) using questionnaires. There were 4 subdistricts those were being the targets of the population, namely from 13 subdistricts in Pamekasan District were the locations for maintaining sonok cattle. Cluster proporsive sampling was determined by the sample of respondents, that was the method of sampling by setting characteristics those were in accordance with the objectives and criteria, namely all of the Sonok cattle those were kept by farmers in 4 subdistricts in Pamekasan District which were the centers for Sonok cattle breeding. The reproduction status of Sonok cattle in Pamekasan District was quite good. The first age of marriage was 23.40 ± 4.17, S / C 1.59 ± 0.53 times, the distance between young calf 14.50 ± 1.83 months, and the value of reproductive efficiency (ER) is 90.51%. The natural increase (NI) value of Sonok cattle in Pamekasan District in 2014 was 9.55%, the value of net replacement rate (NRR) was 101, 48%. The estimation result of Sonok cattle output was 8.24%. Madura cattle population dynamics at the Sonok cattle Nursery Center in Pamekasan District in the period of 2013 to 2017 experienced an average increase of 7.98% annually. The estimated results of the population dynamics from 2018 to 2022 were expected to increase annually by 9.18%. It was concluded that Sonok cattle in Pamekasan District still had the potential to increase the population and the output value was lower than NI (5.06% vs. 15.81%).

Adenocarcinoma is the most fatal cancer in Iran and a patient with this kind of cancer, has a lower lifetime than others. In this research, the survival of patients with gastric carcinoma who were registered at Taleghani Hospital, were studied."n"nMethods: 291 patients with Gastric carcinoma who had received care, chemotherapy or chemoradiotherapy, at Taleghani Hospital in Tehran from 2002 to 2007 were studied as a historical cohort. Their survival rates and its relationship with 12 risk factors were assessed."n"nResults: Of the 291 patients with Gastric carcinoma, 70.1 percent were men and others (29.9%) were women. The mean age of men was 62.26 years and of women was 59.32 years at the time of diagnosis. Most of patients (93.91%) were advanced stage and metastasis. The Cox proportional hazards model showed that age at diagnosis, tumor stage and histology type with survival time had significant relationships (p=0.039, p=0.042 and p=0.032 respectively)."n"n Conclusion: The five-year survival rate and median lifetime of gastric cancer patients who underwent chemotherapy or chemoradiotherapy are very low and seems that one of the important reasons for this situation is delayed diagnosis. The scheme of public education about the early warning signs of the disease and diagnosis and administration of periodic examinations is unavoidable.

Three-dimensional (3D) bio-printing has been shown up as a state of the art and creative technological solution to nowadays tissue engineering and stem cell research challenges toward human tissue regeneration and construction of artificial living organs. Thereby, using hydrogel-based bio-inks to 3D print living microenvironments is a crucial strategy to reconstruct basically functional living scaffolds in order to shape human living organs based on digital 3D computer-aided design (CAD) inputs. The focus of this paper lies on design and development of a portable multi-functional 3D bio-printing extruder for fabrication of hydrogel-based highly complex living tissues using advanced methods invented along this study. The presented article, precisely optimizes the process of fabricating 3D printed scaffolds by redesigning of an integrated gel-extrusion system capable of controlling the adjustability of thermal condition of bio-inks. Also a UV crosslink module is utilized in the bottom of the extruder to cure hydrogel scaffolds consisting of photo-reacting agents to provide a novel bio-printing experience for end users. As a result, the integrated extrusion system is easily portable and compatible with almost any computer numerical control (CNC) machine. Therefore, it could be simply installed on or removed from any CNC machine or fused deposition modeling (FDM) 3D printing system considering that all the control units remain adjustable. The whole system parameters and the performance of tissue fabrication regarding this developed portable multi-functional 3D bio-printing extruder have been tested and practically confirmed. The thermal control system performance has also been simulated using finite element analysis (FEA) and computational fluid dynamics (CFD) methods. Thus, certified documents have been provided and depicted in this paper.

- Abstract
High blood cholesterol is typically considered a feature of wealthy western countries1,2. However, dietary and behavioural determinants of blood cholesterol are changing rapidly throughout the world³ and countries are using lipid-lowering medications at varying rates. These changes can have distinct effects on the levels of high-density lipoprotein (HDL) cholesterol and non-HDL cholesterol, which have different effects on human health4,5. However, the trends of HDL and non-HDL cholesterol levels over time have not been previously reported in a global analysis. Here we pooled 1,127 population-based studies that measured blood lipids in 102.6 million individuals aged 18 years and older to estimate trends from 1980 to 2018 in mean total, non-HDL and HDL cholesterol levels for 200 countries. Globally, there was little change in total or non-HDL cholesterol from 1980 to 2018. This was a net effect of increases in low- and middle-income countries, especially in east and southeast Asia, and decreases in high-income western countries, especially those in northwestern Europe, and in central and eastern Europe. As a result, countries with the highest level of non-HDL cholesterol—which is a marker of cardiovascular risk—changed from those in western Europe such as Belgium, Finland, Greenland, Iceland, Norway, Sweden, Switzerland and Malta in 1980 to those in Asia and the Pacific, such as Tokelau, Malaysia, The Philippines and Thailand. In 2017, high non-HDL cholesterol was responsible for an estimated 3.9 million (95% credible interval 3.7 million–4.2 million) worldwide deaths, half of which occurred in east, southeast and south Asia. Aim: To perform a simple, rapid and sensitive Real-time PCR based SYBR Green method to determine the human leukocyte antigen (HLA)-DQ 2/8 alleles in celiac disease (CD) patients. Background: Many molecular techniques are available to determine the HLA-DQ2 and DQ8 alleles, but they are too expensive and have many steps that make them difficult to use. Methods: To determine the HLA-DQ 2/8 alleles we have developed a new real-time PCR assay, using SYBR Green technique with melting curve analysis on genomic DNA isolated from 75 CD patients and 94 healthy controls. The specific primers to examine HLA-DQA1*05, HLA-DQB1*02 and HLA-DQB1*0302 alleles were used and results were compared with commercially available kits. Results: Using this method, the presence of HLA-DQ2 and HLA-DQ8 alleles were determined with sensitivity and specificity 80% and 100% respectively and compared to low resolution commercially available kits, the results of this method were more efficient. The frequency of DQ2 and DQ8 in patients was 76% and 29%, respectively and overall 96% of patients were carries DQ2 and/or DQ8 alleles. Conclusion: The result of this study showed that Real-time PCR using SYBR Green method with melting curve analysis has good efficiency to identify the HLA-DQ2/8 risk alleles.

Background and Objective: One of the most important effects of nanomaterials on cells is the induction of programmed cell death (apoptosis). The aim of this study is to investigate the toxicity of graphene oxide-gold nanoparticles and to evaluate the expression of caspase 3 gene in HepG2 liver cancer cell line. Methods: In this experimental study, hepatocellular carcinoma cell line (HepG2) was prepared from Pasteur Institute of Iran. Cells were treated with concentrations of 10 to 500 µg/ml Gold-rGO and untreated cells were used as control. The level of caspase 3 gene expression and cell viability were investigated and compared within 24 and 48 hours in three groups of A, B and control (C). The effect of concentration was investigated by XTT method and acridine orange ethidium bromide staining. Caspase 3 gene expression was measured by relative quantification using real time PCR test. Findings: Based on XTT results, EC20, EC50, EC80 values were calculated as 386.420, 90.680, 24.151 µg/ml in 24 hours and 358.146, 89.536, 22.384 µg/ml in 48 hours. Examining the level of caspase 3 gene expression change in these concentrations compared to untreated cells showed mean fold changes (mean±SEM) of 3.436±1.022 in 24 hours and 4.054±0.02 in 48 hours, according to which the level of gene expression changes significantly. The viability of control cells (C) compared to cells treated with concentrations higher than 50 µg/ml (B) has a relative increase, which was statistically significant (p<0.0001). Examination with AO/EB staining to determine the level of apoptosis showed a higher level of apoptosis with increasing dose. Conclusion: The results of the study showed that shorter times and lower concentrations should be used in the application of this nanoparticle. [ABSTRACT FROM AUTHOR]

Micro-organisms contain 90% of cells in human body and trillions foreign genes versus less than 30 thousand of their own. The human colon host various species of microorganisms, appraised at more than 1014 microbiota and contained of over a thousand species. Although each one’s profile is separable, the relative abundance and distribution of bacterial species is the same between healthy ones, causing conservation of each person’s overall health. Germline DNA mutations have been attributed to the less than 5% of CRC occurrence while more than 90% is associated with the epigenetic regulation. The most ubiquitous environmental factor in epigenetic modification is gut microbiota. Disruptive changes in the gut microbiome strongly contributed to the improvement of colorectal cancer. Gut microbiota may play critical role in progression of CRC via their metabolite or their structural component interacting with host intestinal epithelial cell (IEC). Herein we discuss the mechanism of epigenetic modification and its implication in CRC development, progression even metastasis by gut microbiota induction.

BACKGROUND AND OBJECTIVE: Colorectal cancer is one of the growing cancers in Iran and one of the few preventable cancers in the World Health Organization. One of the most important ways to early diagnose this malignancy and its underlying lesions (polyps) and subsequently to prevent it is screening. Colorectal cancer is one of the growing cancers in Iran and one of the few preventable cancers in the World Health Organization. One of the most important ways of early detection of this malignancy and its underlying lesions (polyps) and consequently its prevention is screening. Different protocols have been developed for the diagnosis of first degree relatives of known hereditary cases such as HNPCC and FAP, but the diagnosis plan for first degree relatives has not been defined as a group of familial colorectal cancers without known mutations (type X colorectal cancer). The purpose of this study was to evaluate the results of colonoscopy screening in first-degree relatives of these patients. METHODS: This cross-sectional study was performed on the families of patients with suspected Lynch syndrome who had no deficiency in the expression of repair proteins. Data were collected from the medical records system of colorectal cancer patients who had been enrolled in the system for prevention and early detection of colorectal cancer at Shahid Beheshti University of Medical Sciences Research Center from 1387 to 1397. FINDINGS: The results of colonoscopy revealed that 18 out of 77 subjects had 23% polyps or tumors in their colon, out of which 55% were polyps and the rest were adenomas. About 60% of polyps were in the rectum and sigmoid and the rest in the colon and cecum. Also, 4 patients in this screening showed tumors in their colon, all of which were newly diagnosed, and 2 patients in this screening were diagnosed with inflammatory bowel disease. CONCLUSION: According to the results of this study, in the case of colon polyps, the families of these patients appear to be at high risk like those of Lynch syndrome families and need regular colonoscopy care. This review discusses on diagnostic and therapeutic function of colonoscopy. Colonoscopy is a diagnostic and therapeutic procedure that allows the physician to examine and treatment of the rectum, colon and portion of the terminal ileum. Also colonoscopy is considered the gold standard for colon cancer screening and surveillance. Colonoscopy is the best screening test for early detection and prevention of Colorectal Cancer (CRC). Colonoscopy with removal of detected polyps has been shown to reduce the incidence and mortality of subsequent CRC. The growing importance of colonoscopy function in diagnostic and treatment of colon disorders and also the prevention of CRC have stimulated an effort to identify and to optimize the potential of colonoscopy as well as its new role in public health. Therefore today is needed to expand and improve the quality of colonoscopy and strengthen systems to provide colonoscopy services continuously.

Background: Cryptosporidium is a worldwide protozoan parasite and one of the most common causes of infection and diarrhea in humans and cattle. The aim of the present study was determination of subtypes of Cryptosporidium among children with diarrhea in Tehran by sequence analysis of the highly polymorphic 60-kDa glycoprotein (GP60) gene.Methods: Fecal samples were collected from 794 diarrheic children. Initial identification of Cryptosporidium was carried out on stool samples by Ziehl-Neelsen acid-fast staining method. DNA was extracted from positive microscopically samples and Cryptosporidium genotypes and subtypes were determined, accordingly."nResults: Out of 794 collected samples, 19 (2.40 %) were positive for Cryptosporidium oocysts. Sequences analysis of GP60 gene showed that 17 (89.47 %) of the positive isolates were Cryptosporidium parvum and 2 (10.52 %) were C. hominis. All subtypes of C. parvum isolates belonged to allele families IIa (6/17) and IId (11/17). The most common allele in all 17 isolates belonged to IId A20G1a (41.18%). A22G1 (IF) subtype was detected in two C. hominis isolates of the children."nConclusion: The predominancy of C. parvum species (specially, IId A20G1a subtype) in current study underlines the importance of zoonotic Cryptosporidium transmission in Iran.

BACKGROUND AND OBJECTIVE: Transforming growth factor beta (TGF-β) has an inhibitory role in homeostasis of T-cell response and regulatory T cells and regulation of immune response against viral infections. Codon 10 of protein is located in the signal peptide and involved in secretion of cytokine. The aim of this study was to investigate the association of leu-pro polymorphism of codon 10 and hepatitis C susceptibility in patients.METHODS: This case-control study was performed on 112 chronic hepatitis C patients and 122 healthy control subjects. TGF-β1 gene was amplified with PCR method and genotypes were determined using Restriction Fragment Length Polymorphism (RFLP) with MspA1I restriction enzyme.FINDINGS: The frequency of CT genotype in both groups was higher than CC and TT. Genotyping results for CC, CT and TT states in patients was 50%, 19.6% and 30.4% and in healthy controls was 59%, 13.9% and 27.1% respectively. We found no significant difference between patients and healthy controls according to codon 10 polymorphism. CONCLUSION: According to the results of this study, no relationship was found between this protein’s genetic variations and chronic hepatitis C infection susceptibility.

Introduction: Cryptosporidium is an Apicomplexa parasite that infects humans and a wide range of domestic and wild animals. However, the sub-genotypes of the species infecting animals in Iran are unclear. The aim of the present study isto identify DNA fingerprinting of bovine Cryptosporidium in Qazvin province using sequences of GP60 gene. Materials and Methods : In this study we investigated 25 C. parvum isolated form bovine in Qazvin animal husbandries. Subgenotypes were determined by DNA sequencing of 60-kDa glycoprotein gene. Results: Using DNA sequencing of GP60 gene, two subtype families within the C. parvum included IId (15/22) and IIa (7/22) were recognized. Also three subtypes in these two subtype families included IIa A15G2R1 (22/25), IIa A16G3R (1/25), IId A15G1 (2/25) were determined. Conclusion: Today, new zoonose strains are identified which based on severity of infection compared with human strains are more sever and the source and transmission of their infection are unclear. Therefore, determination of C .parvum strains, genotypes and sub-genotypes for epidemiological studies are quite necessary specially to identify the animal sources and to improve the control and prevention programs due to the lake of effective drugs for this infection.

50. Intestinal Leishmaniasis in Acquired Immunodeficiency Syndrome

In endemic regions, visceral leishmaniasis is one of the most common opportunistic infections in HIV positive patients. Simultaneous infection with Leishmania and HIV has been reported in some countries but this is the first report of such a case in Iran. Our patient was a 27 years old man with intermittent night fever, abdominal pain, loss of appetite, vomiting, watery diarrhea and severe weight loss for 6 months. He had low socio-economic status with an imprisonment history. The patient was quite cachectic and had low grade fever. Physical exam and upper GI endoscopy revealed oropharyngeal candidiasis. Microscopic evaluation of duodenal biopsy material showed Leishmania amastigotes in macrophages of lamina propria. Leishman bodies were also observed in bone marrow aspiration specimen. Serologic tests were positive for Leishmania infantum. HIV antibody was also positive with a CD4+cell count of 80/μl. The diagnosis was acquired immunodeficiency syndrome with simultaneous visceral leishmaniasis involving intestinal mucosa.
