- Abstract
The emergence of new SARS-CoV-2 variants poses challenges to global surveillance efforts, necessitating swift actions in their detection, evaluation, and management. Among the most recent variants, Omicron BA.2.86 and its sub-lineages have gained attention due to their potential immune evasion properties. This study describes the development of a digital PCR assay for the rapid detection of BA.2.86 and its descendant lineages, in wastewater samples. By using this assay, we analyzed wastewater samples collected in Italy from September 2023 to January 2024. Our analysis revealed the presence of BA.2.86 lineages already in October 2023 with a minimal detection rate of 2% which then rapidly increased, becoming dominant by January 2024, accounting for a prevalence of 62%. The findings emphasize the significance of wastewater-based surveillance in tracking emerging variants and underscore the efficacy of targeted digital PCR assays for environmental monitoring.

The ESPRI project relies on the astrometric capabilities offered by the PRIMA facility of the Very Large Telescope Interferometer for the discovery and study of planetary systems. Our survey consists of obtaining high-precision astrometry for a large sample of stars over several years and to detect their barycentric motions due to orbiting planets. We present the operation principle, the instrument's implementation, and the results of a first series of test observations. A comprehensive overview of the instrument infrastructure is given and the observation strategy for dual-field relative astrometry is presented. The differential delay lines, a key component of the PRIMA facility which was delivered by the ESPRI consortium, are described and their performance within the facility is discussed. Observations of bright visual binaries are used to test the observation procedures and to establish the instrument's astrometric precision and accuracy. The data reduction strategy for astrometry and the necessary corrections to the raw data are presented. Adaptive optics observations with NACO are used as an independent verification of PRIMA astrometric observations. The PRIMA facility was used to carry out tests of astrometric observations. The astrometric performance in terms of precision is limited by the atmospheric turbulence at a level close to the theoretical expectations and a precision of 30 micro-arcseconds was achieved. In contrast, the astrometric accuracy is insufficient for the goals of the ESPRI project and is currently limited by systematic errors that originate in the part of the interferometer beamtrain which is not monitored by the internal metrology system. Astrophysics - Abstract
We describe and summarize the optical challenges for future instrumentation for Extremely Large Telescopes (ELTs). Knowing the complex instrumental requirements is crucial for the successful design of 30-60m aperture telescopes. After all, the success of ELTs will heavily rely on its instrumentation and this, in turn, will depend on the ability to produce large and ultra-precise optical components like light-weight mirrors, aspheric lenses, segmented filters, and large gratings. New materials and manufacturing processes are currently under study, both at research institutes and in industry. In the present paper, we report on its progress with particular emphasize on volume-phase-holographic gratings, photochromic materials, sintered silicon-carbide mirrors, ion-beam figuring, ultra-precision surfaces, and free-form optics. All are promising technologies opening new degrees of freedom to optical designers. New optronic-mechanical systems will enable efficient use of the very large focal planes. We also provide exploratory descriptions of "old" and "new" optical technologies together with suggestions to instrument designers to overcome some of the challenges placed by ELT instrumentation., Comment: (Proc. - Abstract
The SARS-CoV-2 Omicron variant emerged in South Africa in November 2021, and has later been identified worldwide, raising serious concerns. A real-time RT-PCR assay was designed for the rapid screening of the Omicron variant, targeting characteristic mutations of the spike gene. The assay was used to test 737 sewage samples collected throughout Italy (19/21 Regions) between 11 November and 25 December 2021, with the aim of assessing the spread of the Omicron variant in the country. Positive samples were also tested with a real-time RT-PCR developed by the European Commission, Joint Research Centre (JRC), and through nested RT-PCR followed by Sanger sequencing. Overall, 115 samples tested positive for Omicron SARS-CoV-2 variant. The first occurrence was detected on 7 December, in Veneto, North Italy. Later on, the variant spread extremely fast in three weeks, with prevalence of positive wastewater samples rising from 1.0% (1/104 samples) in the week 5–11 December, to 17.5% (25/143 samples) in the week 12–18, to 65.9% (89/135 samples) in the week 19–25, in line with the increase in cases of infection with the Omicron variant observed during December in Italy. Similarly, the number of Regions/Autonomous Provinces in which the variant was detected increased from one in the first week, to 11 in the second, and to 17 in the last one. The presence of the Omicron variant was confirmed by the JRC real-time RT-PCR in 79.1% (91/115) of the positive samples, and by Sanger sequencing in 66% (64/97) of PCR amplicons. In conclusion, we designed an RT-qPCR assay capable to detect the Omicron variant, which can be successfully used for the purpose of wastewater-based epidemiology. We also described the history of the introduction and diffusion of the Omicron variant in the Italian population and territory, confirming the effectiveness of sewage monitoring as a powerful surveillance tool.

- Abstract
The Advanced GAmma Tracking Array (AGATA) has been installed at Laboratori Nazionali di Legnaro (LNL), Italy. In this installation, AGATA will consist, at the beginning, of 13 AGATA triple clusters (ATCs) with an angular coverage of 1n,and progressively the number of ATCs will increase up to a 2 pi angular coverage. This setup will exploit both stable and radioactive ion beams delivered by the Tandem-PIAVE-ALPI accelerator complex and the SPES facility. The new implementation of AGATA at LNL will be used in two different configurations, firstly one coupled to the PRISMA large-acceptance magnetic spectrometer and lately a second one at Zero Degrees, along the beam line. These two configurations will allow us to cover a broad physics program, using different reaction mechanisms, such as Coulomb excitation, fusion-evaporation, transfer and fission at energies close to the Coulomb barrier. These setups have been designed to be coupled with a large variety of complementary detectors such as charged particle detectors, neutron detectors, heavy-ion detectors, high-energy gamma-ray arrays, cryogenic and gasjet targets and the plunger device for lifetime measurements. We present in this paper the conceptual design, characteristics and performance figures of this implementation of AGATA at LNL.

- Abstract
The nucleus $^{212}$Po has been produced through the fragmentation of a $^{238}$U primary beam at 1 GeV/nucleon at GSI, separated with the FRagment Separator, FRS, and studied via isomer γ-decay spectroscopy with the RISING setup. Two delayed previously unknown γ rays have been observed. One has been attributed to the E3 decay of a 21$^{−}$ isomeric state feeding the α-emitting 45-s (18$^{+}$) high-spin isomer. The other γ-ray line has been assigned to the decay of a higher-lying 23$^{+}$ metastable state. These are the first observations of high-spin states above the $^{212}$Po (18$^{+}$) isomer, by virtue of the selectivity obtained via ion-by-ion identification of $^{238}$U fragmentation products. Comparison with shell-model calculations points to shortfalls in the nuclear interactions involving high-j proton and neutron orbitals, to which the region around Z∼100 is sensitive.

The measurement of β-decay rates in plasma, simulating stellar-like conditions, is of high interest for the investigation of radionuclides involved in nuclear astrophysics processes. In the new PANDORA plasma trap, to be built at the INFN—Laboratori Nazionali del Sud of Catania (Italy), the β-decay rates will be estimated by detecting the γ-rays emitted by the daughter nuclei trapped in the confined plasma. The present work describes the high efficiency High Purity Germanium (HPGe) detector array that will be placed around the magnetic trap for this purpose, as well as the front-end electronics and acquisition system suitable for the array operation in presence of a high counting rate background originating from Bremsstrahlung radiation.

White matter hyperintensities (WMH) are the most common brain-imaging feature of cerebral small vessel disease (SVD), hypertension being the main known risk factor. Here, we identify 27 genome-wide loci for WMH-volume in a cohort of 50,970 older individuals, accounting for modification/confounding by hypertension. Aggregated WMH risk variants were associated with altered white matter integrity (p=2.5x10-7) in brain images from 1,738 young healthy adults, providing insight into the lifetime impact of SVD genetic risk. Mendelian randomization suggested causal association of increasing WMH-volume with stroke, Alzheimer-type dementia, and of increasing blood pressure (BP) with larger WMH-volume, notably also in persons without clinical hypertension. Transcriptome-wide colocalization analyses showed association of WMH-volume with expression of 39 genes, of which four encode known drug targets. Finally, we provide insight into BP-independent biological pathways underlying SVD and suggest potential for genetic stratification of high-risk individuals and for genetically-informed prioritization of drug targets for prevention trials. White matter hyperintensities (WMH) are a common brain-imaging feature of cerebral small vessel disease. Here, the authors carry out a GWAS and followup analyses for WMH-volume, implicating several variants with potential for risk stratification and drug targeting.

- Abstract
White matter hyperintensities (WMH) are the most common brain-imaging feature of cerebral small vessel disease (SVD), hypertension being the main known risk factor. Here, we identify 27 genome-wide loci for WMH-volume in a cohort of 50,970 older individuals, accounting for modification/confounding by hypertension. Aggregated WMH risk variants were associated with altered white matter integrity (p = 2.5×10-7) in brain images from 1,738 young healthy adults, providing insight into the lifetime impact of SVD genetic risk. Mendelian randomization suggested causal association of increasing WMH-volume with stroke, Alzheimer-type dementia, and of increasing blood pressure (BP) with larger WMH-volume, notably also in persons without clinical hypertension. Transcriptome-wide colocalization analyses showed association of WMH-volume with expression of 39 genes, of which four encode known drug targets. Finally, we provide insight into BP-independent biological pathways underlying SVD and suggest potential for genetic stratification of high-risk individuals and for genetically-informed prioritization of drug targets for prevention trials.

Fusion cross sections of the
28 Si +100 Mo system have been measured near and below the Coulomb barrier by detecting the evaporation residues at forward angles. The excitation function has an overall smoother trend than what obtained in a previous experiment, and a large discrepancy is found for the lowest-energy region, where we observe a tendency of the S factor to develop a maximum, which would be a clear indication of hindrance. The results have been compared with the theoretical prediction of coupled-channels calculations using a Woods–Saxon nuclear potential, and including the low-energy excitation modes of both nuclei. Good agreement with data is found by including, in the coupling scheme, the three lowest members of the ground state rotational band of the oblate deformed28 Si, and two-phonons of the strong quadrupole vibration of100 Mo. The additional coupling, in a schematic way, of the two-neutron pick-up between ground states (Q-value = +4.86 MeV) has a minor effect on calculated cross sections, and does not essentially improve the data fit. The excitation function of28 Si +100 Mo has been compared with that of (1) the heavier system60 Ni +100 Mo having analogous features, and (2) several near-by28 Si,32 S + Zr, Mo systems with various nuclear structures and transfer Q-values. The role of quadrupole and octupole excitation modes, as well as of transfer channels, in affecting the fusion dynamics, are clarified to some extent. Systematic measurements of fusion barrier distributions and CC calculations properly including transfer couplings, are necessary, in order to shed full light on the influence of the various coupled channels on the fusion cross sections. International audience; In right-handers (RH), an increase in the pace of dominant hand movement results in increased ipsilateral deactivation of the primary motor cortex (M1). By contrast, an increase in non-dominant hand movement frequency is associated with reduced ipsilateral deactivation. This pattern suggests that inhibitory processes support right hand dominance in right-handers and raises the issues of whether this phenomenon also supports left hand preference in left-handers (LH), and/or whether it relates to asymmetry of manual ability in either group. Thanks to the BIL&GIN, a database dedicated to the investigation of hemispheric specialization (HS), we studied the variation in M1 activity during right and left finger tapping tasks (FTT) in a sample of 284 healthy participants balanced for handedness. An M1 fMRI localizer was defined for each participant as an 8 mm diameter sphere centered on the motor activation peak. RH exhibited significantly larger deactivation of the ipsilateral M1 when moving their dominant hand than their non-dominant hand. In contrast, LH exhibited comparable ipsilateral M1 deactivation during either hand movement, reflecting a bilateral cortical specialization. This pattern is likely related to left-handers' good performances with their right hand and consequent lower asymmetry in manual ability compared with RH. Finally, inter-individual analyses over the whole sample demonstrated that the larger the difference in manual skill across hands, the larger the difference in ipsilateral deactivation. Overall, we propose that difference in ipsilateral deactivation is a marker of difference in manual ability asymmetry reflecting differences in the strength of transcallosal inhibition when a given hand is moving.
