6. Olfactory Ensheathing Cell: a peculiar glial population for cell therapy

Abstract

Introduction. Olfactory mucosa is considered a potential source of neural stem cells, since supported by peculiar glial cells, Olfactory Ensheathing Cells (OECs). Studies have shown that OECs share some common features with astrocytes and Schwann Cells (Ramon-Cueto and Avila 1998). The finding that OECs are a source of multiple Growth Factors (GFs) is very important as they play a decisive role in regeneration, promoting functional restoration in lesioned area (Raisman and Li 2007). Immunocytochemical investigations have reported that OECs express different markers and adhesion molecules (Pellitteri et. al., 2010). Therefore, OECs have attracted attention because of their remarkable neural plasticity. Materials and Methods. Primary OECs were prepared from neonatal rat olfactory bulbs. In some experiments were used GFs. Cells were identified by immunocytochemical procedures, using different antibodies and the results were highlighted by fluorescent microscopy. Results. Our in vitro studies have reported the ability of OECs to stimulate neuronal survival and neurite sprouting mediated through secretion of GFs; moreover, OECs have shown the capability to exert a protective effect on neurons exposed to stressed condition, such as hypoxia, simulating secondary events during spinal cord injury. Conclusion. These data suggest that OECs might be a suitable tool in cellular therapy for their exceptional ability to promote axonal regeneration and functional restoration making them attractive candidates for neurogenesis, in different neurological disorders or injured CNS.

10. INFLUENCE OF R-LIPOIC ACID ON INTRACELLULAR GLUTATHIONE IN HUMAN DOPAMINERGIC NEUROBLASTOMA CELLS: IMPLICATIONS FOR PARKINSON’S DISEASE

Abstract

The consistent findings of decreased levels of the major antioxidant glutathione in substantia nigra of patients with idiopathic Parkinson's disease (PD) has provided most of the basis for the oxidative stress hypothesis of the etiology of PD. Oxidative stress and mitochondrial dysfunction signify two important biochemical events associated with the loss of dopaminergic neurons in PD. Studies using in vitro and in vivo PD models and in affected tissues from the disease itself have demonstrated a selective inhibition of mitochondrial complex I activity that appears to affect normal mitochondrial physiology leading to neuronal cell death. R-lipoic acid plays a fundamental role in mitochondrial metabolism as a coenzyme for pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase and as a substrate for the NADPH-dependent enzyme glutathione reductase. To address the question of the possible mechanism of R-lipoic acid-mediated protection, we have investigated its effect on human dopaminergic SH-SY5Y cells exposed to rotenone, a specific complex I inhibitor. We found that rotenone dose- and time- dependently altered SH-SY5Y cell viability associated with a decreased of glutathione (GSH) levels and ATP production. We observed a protective effect R-lipoic acid on SH-SY5Y cells against rotenone indicating that the replenishment of normal GSH levels within the cells may hold an important key to therapeutics for PD. 11. KINASE DEREGULATION BY OXIDATIVE STRESS: RELEVANCE TO ALZHEIMER’S DISEASE

Abstract

There are multiple lines of evidence showing that oxidative stress and aberrant mitogenic signaling play an important role in the pathogenesis of Alzheimer’s disease (AD). Given the critical role of mitogen-activated protein kinase (MAPK) pathways in regulating cellular processes that are affected in AD, the importance of MAPKs in disease pathogenesis is being increasingly recognized. All MAPK pathways, i.e., the extracellular signal-regulated kinase (ERK) and p38 pathways, are activated in vulnerable neurons in patients with AD suggesting that MAPK pathways are involved in the pathophysiology and pathogenesis of AD. In order to investigate the possibility that oxidative stress may activate ERK1/2 and/or p38MAPK pathways, we examined their activation in human neuroblastoma cells exposed to lipid peroxidation product 4-hydroxynonenal (HNE). Addition of the HNE to the medium did not induce cell toxicity, evaluated by water soluble tetrazolium salt-based assay. The protein phosphorylation levels were examined by immunoblot analysis with specific activity-dependent phosphoantibodies. ERK2 phosphorylation occurs in a time-dependent fashion, maximum activation being achieved within 3 hours after treatment with HNE; we didn’t observed changes of p38MAPK phosphorylation state. In order to investigate the possibility that activation of ERK might account for defence to oxidative stress , we examined its activation and the phosphorylation of the down-stream effector glycogen synthase kinase 3- beta (GSK3-beta). Pre-treatment of cells with U0126 (a selective inhibitor of MEK1/2, the up-stream kinases responsible for ERK1/2 phosphorylation) completely inhibited Ser9-GSK3-beta phosphorylation due to ERK2 activation. This study provide evidence for the important stress response pathway leading to activation of ERK2 and the phosphorylation of Ser9-GSK3-beta. Taken together, our findings indicate that MAPK pathways are differentially activated during the course of HNE response in viable human neuroblastoma cells. We suggest that regulation of ERK2 pathway may be a central facet to any potential treatment for the early stages of AD. Supported by: RFO-University of Bologna.

Background. Progressive supranuclear palsy (PSP) is a neurodegenerative disorder characterized by pure neurofibrillary tau pathology involving mainly basal ganglia and brain stem nuclei [1]. One of the kinases involved in tau phosphorylation is glycogen synthase 3 kinase (GSK3) [2]. In mammals GSK3 is present in two isoforms, a and b regulated by phosphorylation: phosphorylation of Ser9 in GSK3b or Ser21 in GSK3a leads to inactivation while phosphorylation of Tyr216 in GSK3b or Tyr279 in GSK3a leads to activation [3]. Objective. We analyzed the protein levels of GSK3a/b and of the phosphorylated forms GSK3b S9, GSK3b Y216, GSK3a Y279 in brain tissues of subjects with PSP. Methods. Twelve cases fulfilling the clinical and pathological NINCDS criteria for PSP (63-78 years old), and twelve age-matched controls (65-80 years old) were considered. Protein levels of all GSK3 isoforms were evaluated by western blot analysis on brain tissues homogenates. Results. The analysis failed to show significant differences among the three areas examined for all GSK3 isoforms in PSP in comparison to age-matched control cases. Conclusions. This negative result argues against the role of GSK3 in the pathogenesis of PSP. Thus, the hyperphosphorylation of tau occurring in PSP can be due to alterations of other enzymes, as the cyclin-dependent kinase 5, that we previously demonstrated to be elevated in PSP brain [4]. 1. Abstract

Olfactory Ensheathing Cells (OECs) are cells that display Schwann Cell or astrocyte-like properties. They are a source of growth factors and adhesion molecules which play a very important role as neuronal support enhancing cellular survival. Over the past 10 years, OECs have emerged as a leading reparative candidate, when transplanted into the injured spinal cord, having shown significant promise in regeneration of spinal cord lesions. In this study we assessed the efficacy of OECs on the survival and neurite outgrowth of hippocampal neurons in vitro. Co-cultures of OECs and hippocampal cells of postnatal rats were successfully established and cells were immunocytochemically characterized. Some hippocampal cultures were added with growth factors, as bFGF, NGF and GDNF. Furthermore, conditioned medium from OECs cultures was used to feed some hippocampal neurons. Our results show that in co-cultures of hippocampal neurons and OECs the number of neurons and their neurite outgrowth were significantly increased in comparison with controls. Moreover, we showed that NGF and GDNF promoted a more positive effect in both neuronal survival and neurite outgrowth than bFGF. OEC-conditioned media stimulated both the neuronal survival and dense neurite outgrowth. These data indicate that OECs, as a source of growth factors, can promote the survival and the neurite outgrowth of hippocampal neurons in vitro and that bFGF, NGF and GDNF support them differently. Therefore, as OECs and their secreted growth factors appear to exert a neuroprotective effect for functional restoration and for neural plasticity in neurodegenerative disorders, they might be considered an approach for functional recovery.
