Alcohol dehydrogenase I from Zymomonas mobilis (zmADH1) is a zinc-dependent oxidoreductase that catalyses the oxidation of primary or secondary alcohols to the corresponding aldehydes or ketones using NAD+/NADH as a cofactor. Efforts to express zmADH1 in Escherichia coli in a soluble form have been laden with solubility difficulties. A soluble form of recombinant zmADH1 was achieved by the addition of 1 mM zinc into media. Zinc addition facilitates the proper folding of recombinant zmADH1 and significantly reduces the formation of inclusion bodies. The yield of recombinant zmADH1 represents approximately 30 mg/1 L Luria-Bertani media. Intensified production in fermenters showed a striking difference between the specific and total activities of zmADH1 produced at different zinc concentrations. The zmADH1 showed an affinity to medium-chain alcohols, especially 1-pentanol, which could be used in new greener routes for preparation of aldehydes and alcohols. • Zinc-dependent ADH1 enzyme from Zymomonas mobilis was successfully expressed. • High cell density fermentation process for ADH successfully applied. • Enzyme successfully applied for biotransformation of 1-pentanol to valeraldehyde. Zinc is the second most valuable microelement after iron, according to its abundance in the human body. Supplementation of this metal is not straightforward because of several factors like phytates and interference of other minerals during gastrointestinal digestion. For the last several years, intensive research has been developing new forms of zinc supplements based on zinc‐binding peptides from food products. Except for the advantages of supplementation, zinc‐peptide complexes are expected to be drug candidates against various diseases. Zinc supplementation can be improved by protein hydrolysates and peptides due to their zinc‐binding ability. They enhance zinc supplementation and contribute to preventing conditions leading to zinc deficiency that cause various diseases. Research on food‐derived zinc‐binding peptides is progressing in two directions: the isolation and identification of individual peptides (1) and the preparation of zinc complexes of protein hydrolysates (2). Both approaches are primarily aimed at developing effective mineral supplements, although some work on the second approach is also related to nutrition and therapy. Because zinc–protein hydrolysate complexes are nonstandardized mixtures of peptides, their biological activity mechanisms can be difficult to study. Therefore, it is important to focus more research on the biological activity of individual zinc‐binding complexes and their zinc complexes. This work reviewed recent advances in isolating and identifying zinc‐binding peptides from food sources, preparing protein hydrolysate–zinc complexes, and their biological activities. The established sequences of zinc‐binding peptides have been compiled into a table to review their amino‐acid composition and sequence. We also highlighted approaches for isolating and determining the zinc‐binding capacity of peptides in this class. A field experiment was conducted during the kharif season (June-August, 2018) at Bihar Agricultural University, Sabour, Bihar, India, to assess the growth and quality of baby corn as influenced by NPK and zinc fertility approaches under varying spatial geometries in Indo-gangetic plain zone of Bihar. The experiment was laid out in split-split-plot design with three levels of spatial geometry, viz. 40×20 cm2 spacing, 50×15 cm2 spacing and paired row planting (30 cm spacing in between two lines in a paired-row; 50 cm between the two paired-rows and 10 cm between plants within a row) in main plot; three levels of fertility viz. 120:60:60, 150:75:75 and 180:90:90 kg N:P2O5:K2O ha-1 in sub-plots; and two levels of zinc viz., 2.5 and 5.0 kg Zn ha-1 in sub-sub plots, replicated thrice. Paired row planting recorded enhanced and significant increase in growth attributes, thereby registering highest baby corn yield (1.43 t ha-1), green fodder yield (26.78 t ha-1) and dry fodder yield (11.46 t ha-1), as compared to conventional planting. 180:90:90 kg N:P2O5:K2O ha-1 and 5.0 kg Zn ha-1 enhanced the growth parameters as well as baby corn, green fodder and dry fodder yield. Paired row planting, NPK fertility @180:90:90 kg N:P2O5:K2O ha-1 and Zn fertility levels @5.0 kg Zn ha-1 also recorded improvement in reducing sugar, protein and zinc content of baby corn as well as nutritional quality of fodder. AbstractApplication of growth regulators like paclobutrazol and gibberellic acid is a suitable way to prevent excessive nitrogen consumption. Therefore, this 2-year research was conducted in Karaj, Iran. This research was laid out in a randomized complete block design with three replications in a split-plot arrangement in 2019–2021. Treatments included nitrogen at three levels: (60, 105 and 150 kg ha−1) which were placed in the main plots. The second factor was the foliar application of growth regulators at three levels: [gibberellic acid (100 ppm), paclobutrazol (200 ppm) and control]. The results showed that application of paclobutrazol and gibberellic acid raised super wheat yield and quality compared to control. The highest biological yield (27.67 ton ha−1), was observed in the interaction of nitrogen 150 kg ha−1 + gibberellic acid in the second year, which was the same with nitrogen 150 kg ha−1 + gibberellic acid in the first year and also interaction of nitrogen 150 kg ha−1 + paclobutrazol in the second year. The highest grain protein (14.69%) was seen in nitrogen 150 kg ha−1 + gibberellic acid in the second year and nitrogen 150 kg ha−1 + gibberellic acid in the first year. Growth regulators usage and increasing the nitrogen content (150 kg ha−1), improved the yield and grain quality of wheat. Therefore, the interaction of nitrogen 150 kg ha−1 + gibberellic acid and nitrogen 150 kg ha−1 +paclobutrazol, were the best treatments of this study, due to the creation of suitable environmental conditions for wheat, and these treatments can be suggested to the farmers. RCPD-1, RCPD-6, and RCPD-15 are key for biofortification. Growing health awareness has stimulated a focus on natural immune enhancement. Lablab beans { Lablab purpureus (L.) Sweet}, abundant in bioactive compounds, were investigated for protein, mineral, and vitamin profiles. This research specifically conducted a comparative analysis between two distinct botanical forms: Lablab purpureus var. typicus L. and L. purpureus var. lignosus L, focusing on their nutritional profiles. Among L. purpureus var. typicus L. genotypes, RCPD-6 exhibited the highest protein content (25.46 ± 0.29 g/100 g) followed by RCPD-1 (23.74 ± 0.26 g/100 g). In contrast, L. purpureus var. lignosus L., L. purpureus var. lignosus L. particularly genotype RCPD-22, had lower protein content (19 ± 0.15 g/100 g). Significant variations in Fe content were observed within L. purpureus var. typicus L., ranging from 5.97 ± 0.01 to 10.5 ± 0.02 mg/100 g, with RCPD-2 reported the highest. Zn content ranged from 1.7 ± 0.01 to 3.65 ± 0.02 mg/100 g in both forms. The calcium content in L. purpureus var. typicus L. genotypes are found to be high, ranging from 372.72 ± 0.02 mg/100 g to 458.75 ± 0.03 mg/100 g, suggesting that var. typicus holds promise as a significant dietary source of Ca. P content exhibited variability, ranging from 317.12 ± 0.01 to 460.07 ± 0.06 mg/100 g. L. purpureus var. typicus L. shows correlated levels of protein/Zn, vitamins A/C, and antioxidant activities (DPPH/ABTS) suggesting targeted breeding for co-improved health benefits. It is noteworthy that L. purpureus var. typicus L., whose pods are consumed as vegetables, exhibits higher nutritional content in terms of protein, iron, calcium, and vitamin A. This clearly suggests that L. purpureus var. typicus L. surpasses L. purpureus var. lignosus L. in nutritional value. To strengthen findings, chemometric techniques such as principal component analysis (PCA) and agglomerative hierarchical clustering (AHC) were employed for differentiation between the two botanical forms. These methods highlight the disparities in their nutritional profiles and functional characteristics. PCA analysis demonstrated that the first three components encapsulated 80.67 % of the overall variability. Meanwhile, AHC grouped cultivars into two major clusters which can be divided into two sub-groups based on the evaluated parameters. Personal thermal regulation through passive radiative cooling is an effective strategy to achieve sustainable development. Although recent advances have introduced various cooling textile designs, the extensive use of materials unsuitable for common textiles, alongside coating or compositing technologies, not only compromises the wearability and safety of these textiles but also impacts green sustainability. Herein, it is shown that wool is a promising natural material for transferring mid‐infrared human body radiation, but the high absorption in the solar spectrum prohibits wool from achieving satisfactory daytime radiative cooling. Thus the natural wool impregnates zinc ions into the protein chains to form a zinc‐ion woolen (wool‐Zn) fabric. The wool‐Zn fabric can provide humans with a cooling sensation both indoors and daytime outdoors, showing a temperature reduction of 2 °C on human skin indoors and a reduction of 7 °C on artificial skin under direct sunlight, respectively, when covered by wool‐Zn fabrics compared to when covered with cotton. Moreover, the wool‐Zn fabric shows desirable antibacterial properties and durability against washing and abrasion, while maintaining desirable wearability. As a result, the wool‐Zn fabric shows great potential for commercial application in personal thermoregulation and provides an effective avenue toward sustainable energy and environment. ABSTRACT The synthesis of four distinct protein‐based metal nanocomposites has been fabricated by employing egg albumin and zinc metal through a simple, straightforward two‐step method. The Fourier transform infrared (FTIR), X‐ray diffraction (XRD), and scanning electron microscopy (SEM) analyses confirmed the successful incorporation of zinc nanoparticles into the protein matrix and revealed an irregular microporous structure with average crystallite sizes of less than 100 nm. Thermal stability was assessed via thermogravimetric (TG) and derivative thermal gravimetry (DTG) analysis, revealing a notable increase in thermal stability in a high‐temperature range (300°C°C–700°C), suggesting remarkable stability at elevated temperatures. Antimicrobial efficacy against both gram‐positive (B. cereus and Lactobacillus spp.) and gram‐negative bacteria (E. coli and K. pneumoniae) was evaluated using paper disk diffusion, pour plate, and minimum inhibitory concentration (MIC) methods. All samples exhibited antibacterial efficacy, with zone of inhibition (ZOI) diameters ranging from 12 to 35 mm, and demonstrated a bactericidal effect by eliminating 93 to 98% of bacteria within 8 to 20 h at their MICs. Notably, protein–ZnO–PANI exhibited potent antimicrobial activity against all four bacterial strains and among all the samples, demonstrated higher antimicrobial activity than the unpolymerized nanocomposites. The disk diffusion study confirmed that a concentration of 25 μg/mL of protein–ZnO–PANI resulted in ZOI measurements of 25 mm and 35 mm, whereas MIC values of 300 μg/mL and 200 μg/mL were observed against Bacillus cereus and E. coli, respectively, and killed 95% of B. cereus and 98% of E. coli within 24 h. The presented study sought to determine the foliar application of potassium nitrate effects and addition of black tea leaf wastage to soil on the biochemical qualities (zinc, protein, oil percentage, and carbohydrate) of wheat (Triticum aestivum L.) and the content of NH4N, Na, Mg, Ca, Cl, and pH of the soil. The pot experiment commenced in the spring of 2022 in a randomized complete block design (RCBD) with a factorial arrangement having two factors and four replications in Al-Hindia city, Kerbala, Iraq. The first factor was a potassium nitrate compound comprising three levels (control, 1000, and 3000 g 1000 l-1 ), and the second factor was black tea leaf waste mixed with soil in four doses (control, 25, 45, and 55 g 1000 l-1 ). The results showed the impact of black tea wastes mixed with the soil of higher significance, increasing the NH4N, Na, Mg, Ca, and Cl to 29.28 mg kg-1, 3.75, 3.88, 5.88, and 6.79 mm l-1, respectively, and higher impact in reducing the soil pH (7.54) at a concentration of 55 g 1000 l-1 ). The black tea leaf waste (55 g 1000 l-1 ) helped raise the treated wheat’s zinc, protein, oil, and carbohydrate percentages. Zinc (Zn) is crucial for both plant metabolism and human nutrition, with its deficiency being a global health concern. Strategies to increase its availability in food, such as agronomic biofortification, have gained prominence. This study evaluated the impact of foliar spraying of Zn [at full bloom stage: 0 (control) and 600 g ha−1, as ZnSO4⋅7H2O] on the nutritional quality of cowpea (Vigna unguiculata L. Walp.) grains. Field experiments involving 20 cowpea genotypes were carried out over two seasons in a Typic Quartzipsamment under a no-tillage system. The photosynthetic responses of cowpea plants and the concentrations of Zn, amino acids, sucrose, total sugars, and storage proteins (glutelin, albumin, prolamin, and globulin) in grains were analyzed. All genotypes showed enrichment of Zn in grains in response to ZnSO4⋅7H2O application compared to untreated plants. Foliar spraying of ZnSO4⋅7H2O during initial grain filling was ideal for increasing Zn concentration in grains and improving plant physiological processes. Additionally, Zn fertilization led to higher concentrations of storage and total amino acids and proteins in the grains, supporting the rational application of Zn in cowpea production to improve the nutritional quality of grains and increase plant productivity. As an essential trace element for organisms, zinc participates in various physiological processes, such as RNA transcription, DNA replication, cell proliferation, and cell differentiation. The destruction of zinc homeostasis is associated with various diseases. Zinc homeostasis is controlled by the cooperative action of zinc transporter proteins that are responsible for the influx and efflux of zinc. Zinc transporter proteins are mainly categorized into two families: Zrt/Irt‐like protein (SLC39A/ZIP) family and zinc transporter (SLC30A/ZNT) family. ZIP transporters contain 14 members, namely ZIP1‐14, which can be further divided into four subfamilies. Currently, ZIP transporters‐regulated zinc homeostasis is one of the research hotspots. Cumulative evidence suggests that ZIP transporters‐regulated zinc homeostasis may cause physiological dysfunction and contribute to the onset and progression of diverse diseases, such as cancers, neurological diseases, and cardiovascular diseases. In this review, we initially discuss the structure and distribution of ZIP transporters. Furthermore, we comprehensively review the latest research progress of ZIP transporters‐regulated zinc homeostasis in diseases, providing a new perspective into new therapeutic targets for treating related diseases. Cucumber (Cucumis sativus L.) is a globally prevalent and extensively cultivated vegetable whose yield is significantly influenced by various abiotic stresses, including drought, heat, and salinity. Transcription factors, such as zinc finger-homeodomain proteins (ZHDs), a plant-specific subgroup of Homeobox, play a crucial regulatory role in stress resistance. In this study, we identified 13 CsZHDs distributed across all six cucumber chromosomes except chromosome 7. Phylogenetic analysis classified these genes into five clades (ZHDI-IV and MIF) with different gene structures but similar conserved motifs. Collinearity analysis revealed that members of clades ZHD III, IV, and MIF experienced amplification through segmental duplication events. Additionally, a closer evolutionary relationship was observed between the ZHDs in Cucumis sativus (C. sativus) and Arabidopsis thaliana (A. thaliana) compared to Oryza sativa (O. sativa). Quantitative real-time PCR (qRT-PCR) analysis demonstrated the general expression of CsZHD genes across all tissues, with notable expression in leaf and flower buds. Moreover, most of the CsZHDs, particularly CsZHD9-11, exhibited varying responses to drought, heat, and salt stresses. Virus-induced gene silencing (VIGS) experiments highlighted the potential functions of CsZHD9 and CsZHD10, suggesting their positive regulation of stomatal movement and responsiveness to drought stress. We reported that a 31-amino-acid Zfra protein (zinc finger-like protein that regulates apoptosis) blocks neurodegeneration and cancer growth. Zfra binds WW domain-containing oxidoreductase (WWOX) to both N- and C-termini, which leads to accelerated WWOX degradation. WWOX limits the progression of neurodegeneration such as Alzheimer's disease (AD) by binding tau and tau-hyperphosphorylating enzymes. Similarly, Zfra binds many protein targets and accelerates their degradation independently of ubiquitination. Furthermore, Zfra4-10 peptide strongly prevents the progression of AD-like symptoms in triple-transgenic (3xTg) mice during aging. Zfra4-10 peptide restores memory loss in 9-month-old 3xTg mice by blocking the aggregation of a protein cascade, including TPC6AΔ, TIAF1, and SH3GLB2, by causing aggregation of tau and amyloid β. Zfra4-10 also suppresses inflammatory NF-κB activation. Zfra-activated Hyal-2+ CD3- CD19- Z cells in the spleen, via Hyal-2/WWOX/Smad4 signaling, are potent in cancer suppression. Aquaporins (AQPs) constitute a wide family of water channels implicated in all kind of physiological processes. Zinc is the second most abundant trace element in the human body and a few studies have highlighted regulation of AQP0 and AQP4 by zinc. In the present work, we addressed the putative regulation of AQPs by zinc cations in silico through molecular dynamics simulations of human AQP0, AQP2, AQP4, and AQP5. Our results align with other scales of study and several in vitro techniques, hence strengthening the reliability of this regulation by zinc. We also described two distinct putative molecular mechanisms associated with the increase or decrease in AQPs' water permeability after zinc binding. Autoimmune Regulator 1 (AIRE1) and Forebrain Embryonic Zinc Finger-Like Protein 2 (FEZF2) play pivotal roles in orchestrating the expression of tissue-restricted antigens (TRA) to facilitate the elimination of autoreactive T cells. AIRE1's presence in the gonads of various vertebrates has raised questions about its potential involvement in gene expression control for germline cell selection. Nevertheless, the evolutionary history of these genes has remained enigmatic, as has the rationale behind their apparent redundancy in vertebrates. Furthermore, the origin of the elimination process itself has remained elusive. To shed light on these mysteries, we conducted a comprehensive evolutionary analysis employing a range of tools, including multiple sequence alignment, phylogenetic tree construction, ancestral sequence reconstruction, and positive selection assessment. Our investigations revealed intriguing insights. AIRE1 homologs emerged during the divergence of T cells in higher vertebrates, signifying its role in this context. Conversely, FEZF2 exhibited multiple homologs spanning invertebrates, lampreys, and higher vertebrates. Ancestral sequence reconstruction demonstrated distinct origins for AIRE1 and FEZF2, underscoring that their roles in regulating TRA have evolved through disparate pathways. Furthermore, it became evident that both FEZF2 and AIRE1 govern a diverse repertoire of genes, encompassing ancient and more recently diverged targets. Notably, FEZF2 demonstrates expression in both vertebrate and invertebrate embryos and germlines, accentuating its widespread role. Intriguingly, FEZF2 harbors motifs associated with autophagy, such as DKFPHP, SYSELWKSSL, and SYSEL, a process integral to cell selection in invertebrates. Our findings suggest that FEZF2 initially emerged to regulate self-elimination in the gonads of invertebrates. As organisms evolved toward greater complexity, AIRE1 likely emerged to complement FEZF2's role, participating in the regulation of cell selection for elimination in both gonads and the thymus. Streptococcus pneumoniae (pneumococcus) is a nasopharyngeal commensal and respiratory pathogen. This study characterises the immunoglobulin G (IgG) repertoire recognising pneumococci from birth to 24 months old (mo) in a prospectively-sampled cohort of 63 children using a panproteome array. IgG levels are highest at birth, due to transplacental transmission of maternal antibodies. The subsequent emergence of responses to individual antigens exhibit distinct kinetics across the cohort. Stable differences in the strength of individuals' responses, correlating with maternal IgG concentrations, are established by 6 mo. By 12 mo, children develop unique antibody profiles that are boosted by re-exposure. However, some proteins only stimulate substantial responses in adults. Integrating genomic data on nasopharyngeal colonisation demonstrates rare pneumococcal antigens can elicit strong IgG levels post-exposure. Quantifying such responses to the diverse core loci (DCL) proteins is complicated by cross-immunity between variants. In particular, the conserved N terminus of DCL protein zinc metalloprotease B provokes the strongest early IgG responses. DCL proteins' ability to inhibit mucosal immunity likely explains continued pneumococcal carriage despite hosts' polyvalent antibody repertoire. Yet higher IgG levels are associated with reduced incidence, and severity, of pneumonia, demonstrating the importance of the heterogeneity in response strength and kinetics across antigens and individuals. Background: Micronutrients significantly impact agricultural production and are necessary for human health. Therefore, a systematic programme on identification of sources with high concentration of iron and zinc is important for overcoming the problem of micronutrient malnutrition. Therefore, present study was undertaken with the objectives (1) to find out iron and zinc concentrations and (2) to identify stable urdbean genotypes. This will be further helpful to set desirable level of iron and zinc concentrations in the upcoming urdbean varieties. Methods: The present study was conducted to evaluate 30 urdbean genotypes for variability studies of iron, zinc and protein content. The experiment was laid out in the randomized complete block design and replicated thrice at the experimental farm of Punjab Agricultural University, Regional Research Station, Gurdaspur, Punjab (India) during spring 2019 and 2020. Result: Analysis of variance showed that effect due to genotypes was significant for iron, zinc and protein content as well Iron and Zinc content ranged from (28.75 ppm to 90.75 ppm) and (44.75 ppm to 60.0 ppm) respectively, while protein content ranged from (18.7 to 25.13%). The genotype PDU1 and OBG35 possessed high level of all three nutrients viz., Fe, Zn and protein. Fe content showed significant positive correlation with Zn content suggesting that both the nutrient can be improved simultaneously. Micronutrient dense genotypes identified can be used as donar to develop nutrient enriched varieties either through conventional breeding or by using biotechnological tool like marker assisted selection. The design of cereblon-binding molecular glues (MGs) that selectively recruit a desired protein while excluding teratogenic SALL4 is an area of significant interest when designing therapeutic agents. Previous studies show that SALL4 is degraded in the presence of IKZF1 degraders pomalidomide, and to a lesser extent by CC-220. To expand our understanding of the molecular basis for the interaction of SALL4 with cereblon, we performed biophysical and structural studies demonstrating that SALL4 zinc finger domains one and two (ZF1-2) interact with cereblon (CRBN) in a unique manner. ZF1 interacts with the N-terminal domain of cereblon and ZF2 binds as expected in the C-terminal IMiD-binding domain. Both ZF1 and ZF2 contribute to the potency of the interaction of ZF1-2 with CRBN:MG complexes and the affinities of SALL4 ZF1-2 for the cereblon:CC-220 complex are less potent than for the corresponding pomalidomide complex. Structural analysis provides a rationale for understanding the reduced affinity of SALL4 for cereblon in the presence of CC-220, which engages both ZF1 and ZF2. These studies further our understanding of the molecular glue-mediated interactions of zinc finger-based proteins with cereblon and may provide structural tools for the prospective design of compounds with reduced binding and degradation of SALL4. Cannabidiol (CBD) is a chemical obtained from Cannabis sativa; it has therapeutic effects on anxiety and cognition and anti-inflammatory properties. Although pharmacological applications of CBD in many types of tumors have recently been reported, the mechanism of action of CBD is not yet fully understood. In this study, we perform an mRNA-seq analysis to identify the target genes of CBD after determining the cytotoxic concentrations of CBD using an MTT assay. CBD treatment regulated the expression of genes related to DNA repair and cell division, with metallothionein (MT) family genes being identified as having highly increased expression levels induced by CBD. It was also found that the expression levels of MT family genes were decreased in colorectal cancer tissues compared to those in normal tissues, indicating that the downregulation of MT family genes might be highly associated with colorectal tumor progression. A qPCR experiment revealed that the expression levels of MT family genes were increased by CBD. Moreover, MT family genes were regulated by CBD or crude extract but not by other cannabinoids, suggesting that the expression of MT family genes was specifically induced by CBD. A synergistic effect between CBD and MT gene transfection or zinc ion treatment was found. In conclusion, MT family genes as novel target genes could synergistically increase the anticancer activity of CBD by regulating the zinc ions in human colorectal cancer cells. Zinc is crucial for physiological processes; however, deficiency persists globally. Binding zinc to plant proteins enhances absorption, minimizing toxicity risks and offering a potential solution for deficiency. Mineral binding efficiency of the unmodified protein is limited; hence, dual modification (succinylation and ultrasonication) is potentially used to achieve higher binding efficiency. Enhancing zinc uptake is crucial for cellular health due to its vital roles in various biological processes including enzymatic activity, DNA repair, immune function, antioxidant defense, hormone regulation, brain function, signaling, growth, gene expression, and reproduction. Therefore, this research aimed to develop a chickpea protein-zinc complex and to evaluate the influence of dual modification on their physiochemical, bioavailability, and cellular mineral uptake attributes. Succinylation exhibited significant improvements in the water-holding capacity by 28.73%, oil-holding capacity by 34.09%, and solubility by 5.46% of the chickpea protein-zinc complex as compared to the native complex. Mineral bioavailability increased by 8.32%, and there were notable increases in cellular uptake of zinc by 2.10%, retention by 5.80%, and transport by 3.96%, respectively. Furthermore, the dual modification approach resulted in a notable decrease in the particle size of the chickpea protein-zinc complex, with a substantial reduction of 73.25% and an increased zeta potential value of −21 mV compared to the succinylated complex. As well, dual modification concurrently led to a substantial decline of 48.04% in the sulfhydryl (SH) content, coupled with a marked increase of 21.92% in the surface hydrophobicity. In addition, zinc bioavailability, cellular uptake, retention, and transport were further enhanced by 1.89, 3.34, and 4.8% through dual modification. Our findings highlight that the dual modification of the chickpea protein-zinc complex shows a promising strategy for enhancing the techno-functional characteristics, bioavailability, and cellular uptake of zinc, which could be a better platform for developing vegan foods. Soybean (Glycine max) productivity is significantly reduced by drought stress. Breeders are aiming to improve soybean grain yields both under well-watered (WW) and drought-stressed (DS) conditions, however, little is known about the genetic architecture of yield-related traits. Here, a panel of 188 soybean germplasm was used in a genome wide association study (GWAS) to identify single nucleotide polymorphism (SNP) markers linked to yield-related traits including pod number per plant (PN), biomass per plant (BM) and seed weight per plant (SW). The SLAF-seq genotyping was conducted on the population and three phenotype traits were examined in WW and DS conditions in four environments. Based on best linear unbiased prediction (BLUP) data and individual environmental analyses, 39 SNPs were significantly associated with three soybean traits under two conditions, which were tagged to 26 genomic regions by linkage disequilibrium (LD) analysis. Of these, six QTLs qPN-WW19.1, qPN-DS8.8, qBM-WW1, qBM-DS17.4, qSW-WW4 and qSW-DS8 were identified controlling PN, BM and SW of soybean. There were larger proportions of favorable haplotypes for locus qPN-WW19.1 and qSW-WW4 rather than qBMWW1, qBM-DS17.4, qPN-DS8.8 and qSW-DS8 in both landraces and improved cultivars. In addition, several putative candidate genes such as Glyma.19G211300, Glyma.17G057100 and Glyma.04G124800, encoding E3 ubiquitin-protein ligase BAH1, WRKY transcription factor 11 and protein zinc induced facilitator-like 1, respectively, were predicted. Objective: The purpose of this study was to assess the protective effects of zinc against injury to sperm parameters and male reproductive hormones brought on by toxic protein supplement constituents. Methods: 30 male Sprague Dawley rats were used in the study, of which 10 were in Group 1 (given standard laboratory food), 10 were in Group 2, fed standard laboratory food nutritional supplement powder and 10 were in Group 3, fed standard laboratory food, nutritional supplement powder and zinc. Semen Analysis of Male Sprague Dawley rats of all 3 groups was evaluated after 8 weeks. Sperm parameters and serum levels of Male Reproductive Hormones were measured at the beginning and the end of the study. Results: There was a significant decrease (P<0.05) in Mean ± SEM of sperm count millions /ml, sperm motility (%), and normal sperm morphology (%) in Group 2 rats when compared with Group 1. Zinc supplement in Group 3 caused a significant increase (P<0.05) in Mean ± SEM of sperm parameters including sperm count millions /ml, sperm motility (%) and normal sperm morphology (%) as compared to Group 2 rats. Mean ± SEM of serum level of testosterone Hormone, Follicle Stimulating Hormone(FSH), and Luteinizing Hormone(LH) of Group 2 rats showed a significant (P<0.05) decrease when compared with Group 1 rats. However, the use of Zinc in Group 3 rats significantly (P<0.05) increased serum Testosterone, FSH, and LH levels when compared to Group 2. No significant difference was noted between serum levels of Testosterone, in Group 1 and Group 3. Serum levels of FSH and LH in Group 3 rats (484.81+ 78.17 ng/ml) were significantly increased (P<0.05) as compared to Group 1 rats. Iron-sulfur clusters are prosthetic groups of proteins involved in various biological processes. However, details of the immature state of the iron-sulfur cluster into proteins have not yet been elucidated. We report here the first structural analysis of the Zn-containing form of a Rieske-type iron-sulfur protein, PetA, from Thermochromatium tepidum (TtPetA) by X-ray crystallography and small-angle X-ray scattering analysis. The Zn-containing form of TtPetA was indicated to be a dimer in solution. The zinc ion adopts a regular tetra-coordination with two chloride ions and two cysteine residues. Only a histidine residue in the cluster-binding site exhibited a conformational difference from the [2Fe-2S] containing form. The Zn-containing structure indicates that the conformation of the cluster binding site is already constructed and stabilized before insertion of [2Fe-2S]. The binding mode of ZnCl2, similar to the [2Fe-2S] cluster, suggests that the zinc ions might be involved in the insertion of the [2Fe-2S] cluster. Iron–sulfur clusters are important cofactors involved in a wide range of protein functions, however, the incorporation of FeS clusters into proteins remains underexplored. Rhodomyrtus tomentosa is a source of a novel antibiotic, rhodomyrtone. Because of the increasing industrial demand for this compound, germplasm with a high rhodomyrtone content is the key to sustainable future cultivation. In this study, rhodomyrtone genotypes were verified using the plastid genomic region marker matK and nuclear ribosomal internal transcribed spacer ITS. These two DNA barcodes proved to be useful tools for identifying different rhodomyrtone contents via the SNP haplotypes C569T and A561G, respectively. The results were correlated with rhodomyrtone content determined via HPLC. Subsequently, R. tomentosa samples with high- and low-rhodomyrtone genotypes were collected for de novo transcriptome and gene expression analyses. A total of 83,402 unigenes were classified into 25 KOG classifications, and 74,102 annotated unigenes were obtained. Analysis of differential gene expression between samples or groups using DESeq2 revealed highly expressed levels related to rhodomyrtone content in two genotypes. semiquantitative RT-PCR further revealed that the high rhodomyrtone content in these two genotypes correlated with expression of zinc transporter protein (RtZnT). In addition, we found that expression of RtZnT resulted in increased sensitivity of R. tomentosa under ZnSO4 stress. Petal size is a critical factor in plant reproduction and horticulture, and is largely determined by cell expansion. Gerbera hybrida is an important horticultural plant and serves as a model for studying petal organogenesis. We have previously characterized GhWIP2, a Trp-Ile-Pro (WIP)-type zinc protein, that constrains petal size by suppressing cell expansion. However, the underlying molecular mechanism remains largely unclear. Using yeast two-hybrid screening, bimolecular fluorescence complementation, and co-immunoprecipitation, we identified a TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR (TCP) family transcription factor, GhTCP7, that interacts with GhWIP2 both in vitro and in vivo. Using reverse genetic approaches, we elucidated the function of the GhTCP7-GhWIP2 complex in controlling petal expansion. GhTCP7 overexpression severely reduced cell expansion and petal size, whereas GhTCP7 silencing resulted in increased cell expansion and petal size. GhTCP7 showed similar expression patterns to GhWIP2 in various types of G. hybrida petals. We further identified GhIAA26 , which encodes an auxin signalling regulator, that is activated by the GhTCP7-GhWIP2 complex, leading to the suppression of petal expansion. Our findings reveal a previously unknown transcriptional regulatory mechanism that involves protein-protein interactions between two different transcription factor families to activate a negative regulator of petal organogenesis. Childhood malnutrition persists in low-income countries due to inadequate diet diversity and nutrient density. For seasonal crops, consumers make food substitutions based on price variations which has dietary implications and can cause episodes of nutritional deficiencies. Locally available foodstuffs can be used to formulate low-cost nutritionally adequate food mixtures. Design-Expert® and Nutrisurvey software were used to generate nutrient-dense formulations for the dry season (n=2) and wet season (n=5) from low-cost locally available foods in Eastern Uganda (sweet potatoes, sorghum, soybeans, beans, sesame, groundnuts and maize). Composite flours of the formulations were prepared and cooked following the World Food Program (WFP) recommendations and consumer acceptability determined using a consumer panel (n=43). The most acceptable formulation for the dry season (D2) and the most acceptable formulation for the wet season (R5) were selected for the determination of functional properties (dispersibility, bulk density and water absorption index (WAI) and water solubility index (WSI)), pasting properties (peak viscosity, breakdown viscosity, final viscosity, setback viscosity and peak time), color and nutrient density (energy content, sugars, starch, protein, crude fat, fiber, ash, iron and zinc). D2 contained 25.35g of sorghum, 1.31g of soybeans, 4.34g of beans, 33.11g of sesame and 35.89g of groundnuts per 100g of formulation. R5 contained 4.95g of maize, 20.98g of sorghum, 5.49g of beans, 29.39g of sesame and 39.19g of groundnuts per 100g of formulation. The nutrient densities of D2 and R5 when cooked according to WFP recommendations were also determined. D2 and R5 had high dispersibility (77.2-76.8%), low water absorption index (1.7-2.0g/g) and high water solubility index (0.2-0.3g/g). The pasting properties indicated that the formulations form stable low viscosity pastes that can withstand breakdown during cooking and have high resistance to retrogradation on cooling. Cooked D2 (100g) contained 87.2kcal, 9.5g starch, 2.3g sugars, 5.8g protein, 1.6g fat, 1.7g fiber, 2.3mg iron and 1.6mg zinc. Cooked R5 (100g) had 71.4kcal, 7.1g starch, 2.6g sugars, 4.2g protein, 0.9g fat, 1.0g fiber, 1.9mg iron and 1.4mg zinc. The cooked samples provided more than 50% of the Recommended Nutrient Intake (RNI) for children aged 1-5 years for protein and zinc per serving (200g for children aged 1-3 years and 250g for children aged 4-5 years). The Zfp296 gene encodes a zinc finger-type protein. Its expression is high in mouse embryonic stem cells (ESCs) but rapidly decreases following differentiation. Zfp296-knockout (KO) ESCs grew as flat colonies, which were reverted to rounded colonies by exogenous expression of Zfp296. KO ESCs could not form teratomas when transplanted into mice but could efficiently contribute to germline-competent chimeric mice following blastocyst injection. Transcriptome analysis revealed that Zfp296 deficiency up- and down-regulates a distinct group of genes, among which Dppa3, Otx2, and Pou3f1 were markedly downregulated. Chromatin immunoprecipitation sequencing demonstrated that ZFP296 binding is predominantly seen in the vicinity of the transcription start sites (TSSs) of a number of genes, and ZFP296 was suggested to negatively regulate transcription. Consistently, chromatin accessibility assay clearly showed that ZFP296 binding reduces the accessibility of the TSS regions of target genes. Zfp296-KO ESCs showed increased histone H3K9 di- and trimethylation. Co-immunoprecipitation analyses revealed interaction of ZFP296 with G9a and GLP. These results show that ZFP296 plays essential roles in maintaining the global epigenetic state of ESCs through multiple mechanisms including activation of Dppa3, attenuation of chromatin accessibility, and repression of H3K9 methylation, but that Zfp296-KO ESCs retain a unique state of pluripotency while lacking the teratoma-forming ability. More than two billion people around the world are affected by zinc deficiency, mainly due to the inadequate intake and absorption of zinc. Based on recent research findings, the bioactive peptides could potentially be used to combat zinc deficiency particularly due to their Zinc chelating ability. The main aim of this review was to present current findings, supporting the potential use of bioactive peptides based on their ability to enhance zinc absorption. In-vivo, in-vitro, and ex-vivo studies have demonstrated that zinc chelating peptides can enhance the retention, transportation, and absorption of zinc. Comparative studies on zinc bioavailability from protein hydrolysates and zinc salts have demonstrated that the protein hydrolysates-zinc complexes are more bioavailable than the zinc salts. Data from the structure-function relationship of zinc chelating peptides suggest that the zinc chelating capacities of peptides increase in the following order; the position of zinc chelator > zinc chelator strength > abundance of zinc chelators > net charge > molecular weight. in addition, the transport mechanism of peptide-zinc complex is hypothesized, and the potential use of bioactive peptides based on their safety and taste and limitations to their commercialization are also discussed. NVS-ZP7-4 was identified as a novel chemical reagent targeting the zinc input protein ZIP7, which accounts for the zinc surge from the apparatus to the cytoplasm. Since zinc dysregulation is related to multiple diseases, in this study, we aimed to identify the anti-tumor effects of NVS-ZP7-4 and explore the molecular mechanisms of NVS-ZP7-4 in hepatocellular carcinoma (HCC) progression. We found that NVS-ZP7-4 inhibited cell viability, caused cell cycle arrest, induced apoptosis, and inhibited the proliferation, migration, and invasion of HCCLM3 and Huh7 cells. We further investigated the inhibited activation of the phosphatidylinositol 3-kinase (PI3K)/Akt pathway was involved in the antitumor effect of NVS-ZP7-4 in HCC. Furthermore, NVS-ZP7-4 inhibited HCC tumor growth in vivo. The present study demonstrated that NVS-ZP7-4 is a promising therapeutic target for HCC by regulating PI3K/AKT signaling. The dissolution of zinc oxide is investigated using spectroscopic ellipsometry to investigate its suitability as a platform for biosensing applications. The results indicate that once the ZnO surface has been functionalised, it is suitably protected, and no significant dissolving of the ZnO occurs. The binding kinetics of the SARS-CoV-2 spike protein on aptamer-functionalised zinc oxide surfaces are subsequently investigated. Values are extracted for the refractive index and associated optical constants for both the aptamer layer used and the protein itself. It is shown that upon an initial exposure to the protein, a rapid fluctuation in the surface density is observed. After around 20 min, this effect stabilises, and a fixed increase in the surface density is observed, which itself increases as the concentration of the protein is increased. Retrotransposons have generated about half of the human genome and LINE-1s (L1s) are the only autonomously active retrotransposons. The cell has evolved an arsenal of defense mechanisms to protect against retrotransposition with factors we are only beginning to understand. In this study, we investigate Zinc Finger CCHC-Type Containing 3 (ZCCHC3), a gag-like zinc knuckle protein recently reported to function in the innate immune response to infecting viruses. We show that ZCCHC3 also severely restricts human retrotransposons and associates with the L1 ORF1p ribonucleoprotein particle. We identify ZCCHC3 as a bona fide stress granule protein, and its association with LINE-1 is further supported by colocalization with L1 ORF1 protein in stress granules, dense cytoplasmic aggregations of proteins and RNAs that contain stalled translation pre-initiation complexes and form when the cell is under stress. Our work also draws links between ZCCHC3 and the anti-viral and retrotransposon restriction factors Mov10 RISC Complex RNA Helicase (MOV10) and Zinc Finger CCCH-Type, Antiviral 1 (ZC3HAV1, also called ZAP). Furthermore, collective evidence from subcellular localization, co-immunoprecipitation, and velocity gradient centrifugation connects ZCCHC3 with the RNA exosome, a multi-subunit ribonuclease complex capable of degrading various species of RNA molecules and that has previously been linked with retrotransposon control. Author summary: Retrotransposons are mobile DNA elements that duplicate themselves by a "copy and paste" mechanism using an RNA intermediate. Their misregulation has been linked with some cancers, neuropathologies, and cellular aging. Consequently, the cell has evolved a battery of defenses to protect against retrotransposition. The ancient origin of endogenous retrotransposons suggests their inhibition may have been an evolutionary driver for some host restriction factors, and these may later have been co-opted for defense against exogenous viruses. Zinc Finger CCHC-Type Containing 3, ZCCHC3, protein is a putative host cell restriction factor whose antiviral and innate immune response actions have recently been identified, and that we here show interacts with and limits activity of human retrotransposons in cell culture. In this work we also draw connections between ZCCHC3 and the RNA helicase MOV10 and zinc-finger protein ZAP, host proteins known to restrict viral infection in mammals and previously shown by ourselves and others to potently inhibit human retrotransposition. We also consider that ZCCHC3 may be functionally associated with the cytoplasmic RNA exosome as well as cytoplasmic stress granules, membraneless ribonucleoprotein aggregates that are important for both mRNA metabolism and viral infectivity. Our experiments improve understanding of ZCCHC3 restriction function and may in the future have potential application to the therapeutic treatment of human infectious diseases. Zinc deficiency is a global public health problem in developing countries, including Bangladesh, where rice is the primary consumed staple food. To combat zinc deficiency in Bangladesh, BRRI dhan84 (paddy) has been released as a zinc-biofortified and high-yielding rice variety. Therefore, the objective of this study was to evaluate the changes in physicochemical properties, nutritional attributes, and contents of zinc in newly released rice variety after parboiling and cooking using standard methods. Significant differences were noted among different properties. The study revealed that the parboiling method increased fat (2.36%), protein (9.42%), and zinc (2.741%), whereas those not using parboiling methods resulted in 1.72% fat, 6.68% protein and 2.732% zinc. In contrast, other components like fiber, carbohydrates, ash, and iron decreased after parboiling. Physicochemical analysis showed that parboiled rice was found to be slightly higher in length (6.507 mm), broader in width (2.032 mm), lower in solid gruel loss (0.090%), and also higher in head rice yield (80.09%) than non-parboiled rice. A prolonged cooking time of approximately 39.41 minutes was observed in parboiled rice as compared to non-parboiled rice. However, the cooking process negatively affected the nutrient contents of both parboiled and non-parboiled rice than uncooked or raw rice. Zinc content was generally similar between parboiled and non-parboiled rice after cooking (2.264-2.344 mg/100g). The sensory test further revealed that the parboiled rice obtained more overall acceptability, although the rice color was darker than that of non-parboiled rice. The study concluded that, depending on desired physicochemical and nutritional properties, the zincbiofortified BRRI dhan84 rice variety could be the important option for improving rice grain nutritional quality to overcome the micronutrient problem in Bangladesh. Systematic studies have revealed interactions between components of the Hsp90 chaperone system and Fe/S protein biogenesis or iron regulation. In addition, two chloroplast‐localized DnaJ‐like proteins, DJA5 and DJA6, function as specific iron donors in plastidial Fe/S protein biogenesis. Here, we used Saccharomyces cerevisiae to study the impact of both the Hsp90 chaperone and the yeast DJA5‐DJA6 homologs, the essential cytosolic Ydj1, and the mitochondrial Mdj1, on cellular iron‐related processes. Despite severe phenotypes induced upon depletion of these crucial proteins, there was no critical in vivo impact on Fe/S protein biogenesis or iron regulation. Importantly, unlike the plant DJA5‐DJA6 iron chaperones, Ydj1 and Mdj1 did not bind iron in vivo, suggesting that these proteins use zinc for function under normal physiological conditions. Metal ions are essential cofactors for many proteins and play a crucial role in many applications such as enzyme design or design of protein-protein interactions because they are biologically abundant, tether to the protein using strong interactions, and have favorable catalytic properties. Computational design of metalloproteins is however hampered by the complex electronic structure of many biologically relevant metals such as zinc. In this work, we develop two tools - Metal3D (based on 3D convolutional neural networks) and Metal1D (solely based on geometric criteria) to improve the location prediction of zinc ions in protein structures. Comparison with other currently available tools shows that Metal3D is the most accurate zinc ion location predictor to date with predictions within 0.70 ± 0.64 Å of experimental locations. Metal3D outputs a confidence metric for each predicted site and works on proteins with few homologes in the protein data bank. Metal3D predicts a global zinc density that can be used for annotation of computationally predicted structures and a per residue zinc density that can be used in protein design workflows. Currently trained on zinc, the framework of Metal3D is readily extensible to other metals by modifying the training data. Zinc is an essential metal for many proteins. Introduction: Burning Mouth Syndrome (BMS) is defined by a burning sensation or intraoral dysesthesia without obvious causal lesion. Despite the development of research, diagnostic aids for this syndrome are non-existent and treatments are partially ineffective. Some studies have shown changes in salivary composition, viscosity or flow in patients with BMS. The evaluation of the salivary biochemical characteristics of patients with BMS could then help to better understand the pathogenesis of this disease. The objective of this meta-analysis is to perform a qualitative and quantitative synthesis of the literature concerning the salivary biomarkers present in patients with BMS in comparison to healthy subjects. Materials and methods: The PubMed, Web of Science and Cochrane databases were searched to identify articles corresponding to the defined inclusion criteria. Review Manager 5.4.1 software was used to perform the quantitative analysis. Results: The quantitative analysis included 15 articles and found a significant increase in salivary concentrations of cortisol and immunoglobulin A (IgA) with a standardized mean difference SMD = 0.53 and a 95% confidence interval CI [0.33 to 0.74] and SMD = 0.32 to 95% CI [0.10 to 0.55] respectively. For calcium and copper, the analysis found no significant difference in patients with BMS with SMD = 0.06 at 95% CI [–0.19 to 0.32] and SMD = -0.19 at 95% CI [–0.44 to 0.06]. Finally, for magnesium, the analysis found a probable decrease with SMD = -0.29 at 95% CI [–0.52 to -0.06]. Four other biomarkers (potassium, alpha amylase, zinc and total protein) showed too high levels of heterogeneity (I² > 44%) to be able to interpret the results with confidence. Discussion: This strong heterogeneity can be explained by different saliva sampling methods or biomarker measurement techniques that are not strictly identical between the studies. In addition, some saliva samples may have undergone blood contamination which may bias the results. Conclusion: This meta-analysis nevertheless confirms all the interest of focusing on salivary biomarkers in BMS patients; the measurement of cortisol and/or salivary IgA could be a line of research for the establishment of a standardized biological assessment. Nevertheless, the number of available studies being low and of variable methodological quality with a limited number of patients, additional studies are necessary to give a firm and definitive conclusion. In this paper, we report results of using molecular modeling to assign specific Zn(II) binding affinities to the known binding sites of the YiiP–zinc transporter. YiiP is a cation-diffusion facilitator. It facilitates the transmembrane exchange of Zn(II) ions and protons. The crystal structure of this protein is known. There are several zinc binding sites, and some of the Zn(II) binding affinities have been measured, but the value of all the binding/dissociation constants and the exact assignment of the sites with these affinities are not completely understood. We have recently developed a fast and accurate coarse-grain framework for predicting protein pKa shifts named PKA17. In this paper, we report extending of the same technique to produce a methodology capable of quickly predicting metal–protein binding affinities. The new software has been named M21. It has been tested on several zinc–protein binding cases, and the average unsigned error in the binding energies has been found to be 2.17 kcal/mol vs. the AMBER average error of 3.49 kcal/mol ( K d ratio of ca. 30 vs. the AMBER one of 330). We have then applied the M21 methodology to calculate and assign the YiiP–Zn(II) binding constants of − 2.31 − 13.28 kcal/mol ( K d values from 2. 0 4 × 1 0 − 2 to 1. 8 5 × 1 0 − 1 0 ). We have also undertaken additional modifications of parameters. On one hand, we have included another 11 zinc binding proteins in our target fitting set. These were the Uno Ferro single chain (UFsc) and its modifications created by the Professor Olga Makhlynets group. On the other hand, we have significantly reduced the number of fittable parameters in order to further reduce the possibility of overfitting and to demonstrate the stability of the technique. The final parameter set has only eight adjustable parameters (as opposed to the above case with 17 independent parameters). The average error for the binding cases compared with the same AMBER test set as above did not change much and was still very low at 2.17 kcal/mol. We believe that these results not only further validate the presented methodology but also point out a promising direction for potential multiple joint experimental and computational collaborative projects. Multiple proteins bind to telomeric DNA and are important for the role of telomeres in genome stability. A recent study established a broad-complex, tramtrack and bric-à-brac - zinc finger (BTB-ZF) protein, ZBTB10 (zinc finger and BTB domain-containing protein 10), as a telomeric variant repeat-binding protein at telomeres that use an alternative method for lengthening telomeres). ZBTB10 specifically interacts with the double-stranded telomeric variant repeat sequence TTGGGG by employing its tandem C2H2 zinc fingers (ZF1-2). Here, we solved the crystal structure of human ZBTB10 ZF1-2 in complex with a double-stranded DNA duplex containing the sequence TTGGGG to assess the molecular details of this interaction. Combined with calorimetric analysis, we identified the vital residues in TTGGGG recognition and determined the specific recognition mechanisms that are different from those of TZAP (telomere zinc finger-associated protein), a recently defined telomeric DNA-binding protein. Following these studies, we further identified a single amino-acid mutant (Arg767Gln) of ZBTB10 ZF1-2 that shows a preference for the telomeric DNA repeat TTAGGG sequence. This research determined the nutritional composition, antioxidant activity and total phenolic compound content of sweet potato flour from different cultivars: Beauregard cv. sweet potato (BF) and the common sweet potato Brazlândia Rosada cv. (CF). Total lipids, proteins, moisture, carbohydrates, pH, titratable acidity, fiber, ash, iron, zinc, beta-carotene, vitamin A and determination of caloric value were analyzed. Antioxidant activity and total phenolic compounds were also determined by the DPPH (2,2-diphenyl-1-picryl-hydrazyl), ABTS (2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) and FRAP (Ferric Reducing Antioxidant Power) methods. CF showed higher levels of carbohydrates, ash, fiber, pH and antioxidant activity by the ABTS method. Protein and zinc contents were similar between the two flours. BF had higher moisture content, caloric value, lipids, iron, DPPH radical scavenging capacity and antioxidant activity by the FRAP method, as well as total phenolic compounds, beta-carotene and vitamin A content. The data revealed good nutritional composition for both analyzed flours. The Beauregard flour can be highlightedregarding its higher beta-carotene and iron content. Moreover, the Beauregard sweet potato flour is a good source of provitamin A, in addition to being a source of fiber. [ABSTRACT FROM AUTHOR]

Zinc-finger proteins play different roles in cancer; however, the function of zinc-finger protein ZNF575 in cancer remains unclear. In the present study, we aimed to determine the function and expression of ZNF575 in colorectal cancer. Proliferation assay, colony formation assay, and tumor model in mice were used to investigate the function of ZNF575 after ectopic expression of ZNF575 in colorectal cancer (CRC) cells. RNA sequencing, ChIP, and luciferase assays were used to investigate the mechanism behind ZNF575 regulation of CRC cell growth. The expression of ZNF575 was determined by IHC staining in 150 pairs of malignant CRC tissues, followed by prognosis analysis. We indicated that ectopic expression of ZNF575 inhibited CRC cell proliferation, colony formation and promoted cell apoptosis in vitro. Tumor growth in CRC was also impaired by ZNF575 in mice. RNA sequencing, follow-up western blotting, and qPCR results demonstrated the increase of p53, BAK, and PUMA in ZNF575- expressing CRC cells. Further results indicated that ZNF575 directly targeted the p53 promoter and promoted the transcription of p53. Downregulation of ZNF575 was confirmed in malignant tissues, and ZNF575 expression was positively correlated with the prognosis of CRC patients. The present study demonstrated the function, underlying mechanism, expression, and the prognosis-predicting role of ZNF575 in CRC, which indicated that ZNF575 would be a potential prognostic predictor and therapeutic target for CRC and other cancers. Zinc homeostatic medicine is of great potential for cancer chemo‐immunotherapy; however, there are few reports on antitumor compounds that can trigger Zn2+‐mediated immune responses. In this work, we developed a novel cyclometalated PtIV‐terthiophene complex, Pt3, that not only induces DNA damage and cellular metabolism dysregulation, but also disrupts zinc homeostasis as indicated by the abnormal transcriptional level of zinc regulatory proteins, excess accumulation of Zn2+ in cytoplasm, and down‐regulation of metallothioneins (MTs), which further caused redox imbalance. The simultaneous disruption of zinc and redox homeostasis in response to Pt3 treatment activated gasdermin‐D mediated pyroptosis accompanied by cytoskeleton remodeling, thus releasing pro‐inflammatory cytokines to promote dendritic cell (DC) maturation and T cell tumor‐infiltration, eventually eliminating both primary and distant tumors in vivo. Background: In an effort to develop micronutrient-rich chickpea lines, a study to examine the variability in yield and nutritional traits was conducted. Methods: 99 genotypes were studied, the data was recorded and analyzed on yield traits, protein, iron and zinc content in Rabi 2019-20. Result: Maximum variability was recorded in plant yield followed by iron concentration (mg/kg), hundred seed weight and number of pods per plant. Of the total entries, nine entries namely, RGH4, RGH56, RG2016-84, ICC251762, RGH53, IPC98-12, RG2016-03, ICC1053 and RGH58 recorded high protein content (>20%). All these entries possessing high protein had pink flower. One chickpea accession (RGH53) accumulated the highest concentration for both protein and zinc, with an average of 21.86% and 73.00 ppm, respectively, but it showed low iron concentration (20.77 ppm). The first six principal components provided a reasonable summary of the data and explained 80.19% of the total variation. Ninety nine genotypes were grouped into ten clusters. Maximum inter cluster distance was observed between clusters VI and IX; VI and VII and IV and VI. The genotypes of these clusters are suggested for utilization in the crossing programs to breed varieties of chickpea for high yield with amenability to nutrients. [ABSTRACT FROM AUTHOR]

CCCH-type zinc figure proteins (ZFP) are small cellular proteins that are structurally maintained by zinc ions. Zinc ions coordinate the protein structure in a tetrahedral geometry by binding to cystine-cystine or cysteines-histidine amino acids. ZFP's unique structure enables it to interact with a wide variety of molecules including RNA; thus, ZFP modulates several cellular processes including the host immune response and virus replication. CCCH-type ZFPs have shown their antiviral efficacy against several DNA and RNA viruses. However, their role in the human coronavirus is little explored. We hypothesized that ZFP36L1 also suppresses the human coronavirus. To test our hypothesis, we used OC43 human coronavirus (HCoV) strain in our study. We overexpressed and knockdown ZFP36L1 in HCT-8 cells using lentivirus transduction. Wild type, ZFP36L1 overexpressed, and ZFP36L1 knockdown cells were each infected with HCoV-OC43, and the virus titer in each cell line was measured over 96 hours post-infection (p.i.). Our results show that HCoV-OC43 replication was significantly reduced with ZFP36L1 overexpression while ZFP36L1 knockdown significantly enhanced virus replication. ZFP36L1 knockdown HCT-8 cells started producing infectious virus at 48 hours p.i. which was an earlier timepoint as compared to wild -type and ZFP36L1 overexpressed cells. Wild-type and ZFP36L1 overexpressed HCT-8 cells started producing infectious virus at 72 hours p.i. ATP binding cassette (ABC) transporters are the primary means by which bacteria acquire trace elements from the environment. They rely on solute binding proteins (SBPs) to bind the relevant substrate and deliver it to the integral membrane permease for ATP-powered import into the cytoplasm. SBPs of cluster A-I are known to facilitate the transport of essential metals zinc, manganese, and iron, and many have been characterized to date. A group of ABC transporter operons dubbed zinc-regulated genes (zrg) have recently been shown to transport zinc with putative SBPs (zrgA) bearing no homology to the classical cluster A-I family, and a recent crystal structure of a representative protein from Pseudomonas aeruginosa shows no structural similarity to classical SBPs. Thus, the ZrgA proteins appear to represent a newly discovered family of zinc SBPs widespread among Gram-negative bacteria, including human pathogens. Here, we have determined the crystal structure of ZrgA from Vibrio cholerae and characterized its zinc binding in vitro and function in vivo. We also assessed the role of a histidine-rich sequence that appears to be a hallmark of ZrgA proteins that is particularly long in V. cholerae ZrgA. The results show that the zrgA gene is critical to the function of the operon, consistent with a function as an SBP in this system. Further, the His-rich region is not essential to the function of ZrgA, but it does provide additional zinc binding sites in vitro. The structure and zinc binding data for ZrgA reveal interesting differences between it and its homologue from P. aeruginosa, illustrating diversity within this little-studied protein family. Barley is one of the most consumed cereals, with many different cultivars available worldwide. Like other crops, its yield has been affected by climate change and soil degradation. This work proposes controlled-release protein-based matrices with incorporated zinc to improve barley seed germination and zinc content in the plant. Thus, the main objective of this study was to investigate the use of controlled-release protein-based matrices for massive crops, such as barley. Different barley cultivars of barley were studied: Barke, Golden Promise, Morex, WB-200, WB379, and WB-446. The seeds of each cultivar were also analyzed in order to explain the behavior of plants observed during the growth. To this end, the physico-chemical (FT-IR, Raman spectroscopy, and Zn concentration) and microstructural (SEM) properties of the different seeds were firstly evaluated to establish differences between the studied cultivars. In addition, the use of controlled-release soybean protein-based matrices without zinc (M) or with zinc incorporated (MZ) was evaluated as fertilizers in the different barley cultivars. In this sense, the use of these matrices as a zinc carrier improved seed germination and zinc content in the plants, indicating that the use of matrices improves the amount of zinc assimilated by the crops (up to 30 and 50% with M and MZ, respectively) and allows the proper root growth of all cultivars of barley. Zinc is an essential micronutrient that participates in a multitude of cellular and biochemical processes. It is indispensable for normal growth and the maintenance of physiological functions. As one of the most significant trace elements in the body, zinc fulfills three primary biological roles: catalytic, structural, and regulatory. It serves as a cofactor in over 300 enzymes, and more than 3000 proteins require zinc, underscoring its crucial role in numerous physiological processes such as cell division and growth, immune function, tissue maintenance, as well as synthesis protein and collagen synthesis. Zinc deficiency has been linked to increased oxidative stress and inflammation, which may contribute to the pathogenesis of a multitude of diseases, like neurological disorders and cancer. In addition, zinc is a key constituent of zinc-binding proteins, which play a pivotal role in maintaining cellular zinc homeostasis. This review aims to update and expand upon the understanding of zinc biology, highlighting the fundamental roles of zinc in biological processes and the health implications of zinc deficiency. This work also explores the diverse functions of zinc in immune regulation, cellular growth, and neurological health, emphasizing the need for further research to fully elucidate the therapeutic potential of zinc supplementation in disease prevention and management. Zinc (Zn) plays a crucial role in metabolism in both plant and animal life. Zn deficiency is a worldwide problem that has recently gotten worse. This micronutrient shortage can be largely attributed to eating foods that are poor in zinc. If biofortification methods were widely used, Zn enrichment of the organ or tissue of interest would increase dramatically. However, Zn absorption mechanisms in rice plants must be understood on a fundamental level before these methods can be used effectively. Plant systems' Zn transporters and metal chelators play a major role in regulating this intricate physiological characteristic. The Zn efficiency of specific species is affected by a variety of factors, including the plant's growth stage, edaphic conditions, the time of year, and more. Both old and new ways of breeding plants can be used for biofortification. We have highlighted the significance of recombinant and genetic approaches to biofortifying in rice. In this review, we have the metabolic role of zinc in rice, and the different transporter families involved in the transportation of zinc in rice. We have also discussed the combined approaches of agronomic and genetic in zinc biofortification in rice and potential outcomes and future predictions. Background and Objectives: ZBTB48 is a telomere-related protein that has been renamed telomeric zinc finger-associated protein (TZAP). It favorably binds to elongated telomeres to regulate their appropriate length. However, TZAP expression has not been investigated in hepatocellular carcinomas (HCC). Materials and Methods: The clinical significance of TZAP expression in 72 HCC was investigated. Additionally, its findings were supported by open big data and cancer cell lines. Results: TZAP expression level was not associated with the clinical parameters of HCC. TZAP expression induced a poorer survival result (overall survival, p = 0.020; disease-free survival, p = 0.012). TCGA data showed TZAP expression was more frequently found in HCCs with hepatitis C infection (p = 0.023). However, TCGA data revealed that TZAP expression did not predict HCC prognosis. In a cell line study, TZAP inhibition via siRNA suppressed PLC/PRF/5 cell growth; however, cell viability was increased in HepG2 cells. Conclusions: We presented the clinical and prognostic values of TZAP expression in HCC tissues and cancer cell lines. Aeromonas hydrophila infections are common in aquaculture. Our previous studies found that the A. hydrophila B11 strain can survive in fish macrophages for at least 24 h and the two‐component system EnvZ/OmpR may be involved in intracellular survival. To reveal the role and mechanism of the two‐component system EnvZ/OmpR in intracellular survival of A. hydrophila, the genes of envZ/ompR were silenced by shRNAi. The results showed that the survival rates of the envZ‐RNAi and ompR‐RNAi strains were only 2.05% and 3.75%, respectively, which were decreased by 91% and 83.6% compared with that of the wild‐type strain. The escape ability of envZ‐RNAi and ompR‐RNAi was also decreased by 51.4% and 19.7%, respectively. The comparative transcriptome analysis revealed that the functional genes directly related to bacterial intracellular survival mainly included the genes related to anti‐stress capacity, and the genes related to Zn2+ and Mg2+ transport. Further research confirmed that two‐component system EnvZ/OmpR can regulate the expression of the important molecular chaperones, such as groEL, htpG, dnaK, clpB and grpE. The expression of these molecular chaperones in wild‐type strain was up‐regulated with the increase in H2O2 concentrations, while the expression of these molecular chaperones in silent strains did not change significantly. Cells that phagocytosed wild‐type strain had higher ROS content than cells that phagocytosed silent strains. Two‐component system EnvZ/OmpR could also regulate zinc transporter (znuA, znuB, znuC) and zinc efflux protein (zntA) to maintain zinc homeostasis in cells, thus affecting the ability of bacteria to survive in phagocytes. Moreover, two‐component system EnvZ/OmpR could affect the growth and intracellular survival of A. hydrophila by regulating the expression of MgtA, MgtC and MgtE and participating in bacterial Mg2+ homeostasis in fish macrophages.
