IntroductionTuberculosis, caused by Mycobacterium tuberculosis complex (MTBC), remains a global health concern in both human and animals. However, the absence of rapid, accurate, and highly sensitive detection methods to differentiate the major pathogens of MTBC, including M. tuberculosis, M. bovis, and BCG, poses a potential challenge.MethodsIn this study, we have established a triplex droplet digital polymerase chain reaction (ddPCR) method employing three types of probe fluorophores, with targets M. tuberculosis (targeting CFP-10-ESAT-6 gene of RD1 and Rv0222 genes of RD4), M. bovis (targeting CFP-10-ESATs-6 gene of RD1), and BCG (targeting Rv3871 and Rv3879c genes of ΔRD1), respectively.ResultsBased on optimization of annealing temperature, sensitivity and repeatability, this method demonstrates a lower limit of detection (LOD) as 3.08 copies/reaction for M. tuberculosis, 4.47 copies/reaction for M. bovis and 3.59 copies/reaction for BCG, without cross-reaction to Mannheimia haemolytica, Mycoplasma bovis, Haemophilus parasuis, Escherichia coli, Pasteurella multocida, Ochrobactrum anthropi, Salmonella choleraesuis, Brucella melitensis, and Staphylococcus aureus, and showed repeatability with coefficients of variation (CV) lower than 10%. The method exhibits strong milk sample tolerance, the LOD of detecting in spike milk was 5 × 103 CFU/mL, which sensitivity is ten times higher than the triplex qPCR. 60 clinical DNA samples, including 20 milk, 20 tissue and 20 swab samples, were kept in China Animal Health and Epidemiology Center were tested by the triplex ddPCR and triplex qPCR. The triplex ddPCR presented a higher sensitivity (11.67%, 7/60) than that of the triplex qPCR method (8.33%, 5/60). The positive rates of M. tuberculosis, M. bovis, and BCG were 1.67, 10, and 0% by triplex ddPCR, and 1.67, 6.67, and 0% by triplex qPCR, with coincidence rates of 100, 96.7, and 100%, respectively.DiscussionOur data demonstrate that the established triplex ddPCR method is a sensitive, specific and rapid method for differentiation and identification of M. tuberculosis, M. bovis, and BCG.

LaB6/TiC/TiB was incorporated into a Ti–Al–Zr–Sn alloy to achieve an efficient improvement in acid-resistance. The incorporation of LaB6/TiC/TiB deteriorated the performance of the passive layer, but impeded the penetration of the corrosive medium into Ti matrix. The incorporation of the conductive reinforcements promoted galvanic corrosion, leading to a decrease in barrier effect of the passive film. Delayed passivation in less aggressive media was achieved by controlling the formation and growth of corrosion pits. The distribution of LaB6/TiC/TiB determined the corrosion properties after complete failure of the passive layer. An appropriate approach for enhancing the acid-resistance of Ti-based materials was suggested.

Single-phase body-centered cubic (BCC) lightweight Ti–V–Nb–Zrx solid-solution alloys were fabricated by reducing the Zr concentration within the range of 0–5 at% to improve the mechanical properties of equimolar TiVNbZr alloy at elevated temperatures. The fabricated Zrx alloys indeed exhibited greater strength above 800 °C than TiVNbZr, but some special mechanical responses occurred at 800 °C: (ⅰ) superficial cracks with different lengths and amounts along the high-angle grain boundaries (HAGBs) on the compressed sample surface, and (ⅱ) near-brittle fracture of the tensile stress-strain curves. Based on the analysis of the deformed and undeformed surface, local stress concentration and phase transformation were responsible for the cracks and difference in compression behavior, and atmospheric oxygen, deformation localization and phase transformation were mainly responsible for the tensile fracture difference at 800 °C. Phase transformation promote stress dispersion, combined with better oxidation resistance, which make the Zr-containing alloys fractured via the transgranular mode in tensile test. In particular, the Zr3 alloy exhibited work hardening and tensile ductility. Based on the tensile and compression properties of the Zrx alloys, it was found that at RT and 600 °C, atomic size difference (δ) governed alloy strength; from 800 to 1000 °C, δ and melting temperature (Tm) co-operatively governed alloy strength; at 1200 °C, alloy strength was independent of δ, but dependent on Tm. The fabricated Ti–V–Nb–Zrx alloys have medium mechanical properties but a small strength loss at high temperatures, and are suitable for various RT deformation processing methods.

The development of ship technology and information technology has been driving the continuous improvement of ship intelligence, with safety being an inevitable requirement in the shipping industry. A machine vision-based ship collision warning method is proposed for high monitoring system cost and limited information acquisition in safety design of autonomous ship navigation. The method combines machine learning with image algorithms. Firstly, the backbone of YOLOv7 detector is replaced by EfficientFormerV2 network to achieve model lightweight while ensuring detection accuracy. Public datasets SeaShips, Flow and self-made ship pictures are combined, and the network is trained on this dataset. StrongSORT is used for target tracking. Secondly, a data fusion algorithm is introduced to determine the target point at the bow-bottom of the ship based on the time-varying attitude of the camera and the time-series features of the bounding boxes. Ship navigation trajectory estimation is performed using image algorithms. Finally, a collision evaluation model is established to calculate the collision risk index. Experimental results demonstrate that the improved YOLOv7 network maintains similar mAP.5 and Recall compared to the original model, while reducing the parameters by 31.2 % and GFLOPs by 58.4 %. The accuracy of target ship trajectory estimation is high, with MAE values below 1.5 % and RMSE values below 2 % in experiments. In ship collision warning experiments, the proposed method accurately identifies navigating vessels, estimates the trajectories, and provides timely warnings for imminent collision accidents. Compared to traditional ship collision warning methods, this paper offers a more intelligent and lightweight solution.

Abstract For the purpose of object detection, numerous key points based methods have been suggested. To alleviate the imbalance problem that some objects may be missing when a single-center-point based network is used for object detection, we propose a brand-new multiple space based cascaded center point network (MSCCPNet) for object detection. Particularly, we first bulid a novel structure to alleviate the imbalance problem in detecting different scale objects by scanning more objects in different scale spaces. We then propose a cascaded center point structure to predict the category and confidence of the object by integrating the results of the two centers with the idea of choosing the high confidence and discarding the low confidence. Finally, we determine the object’s location by predicting the center point deviation as well as the width and height of the object. Our MSCCPNet shows competitive accuracy when compared with many sample classical object detection algorithms on GeForce RTX 2080Ti, according to the results of experiments on PASCAL VOC datasets and COCO datasets.

Inflow forecast plays an indispensable role in reservoir operation. Accuracy and effectiveness of model prediction play a decisive role in it. In this paper, the certainty coefficient, root mean square error (RMSE), mean absolute deviation (MAE) and Nash-Suthcliffe coefficient (NSE) are used to consider the effect of Soil and Water Assessment Tool (SWAT) and Xin'anjiang (XAJ) model on the inflow prediction of Jinxi Reservoir. Results indicate that the certainty coefficient of XAJ model is 90.95% and SWAT model is 84.98%. The NSE of SWAT model is 84.42% and the NSE of XAJ model is 82.33% in which the NSE of the SWAT model is superior to the XAJ model. In view of this, based on the two hydrological models results, Bayes Model Averaging (BMA) method is used to study the inflow combination forecast. The SWAT-XAJ coupling model based on BMA method is proposed. During the calibration period and the verification period, compared with single model, the maximum increment of the deterministic coefficient of BMA is 2.87%, the maximum decrement of RMSE is 82.00, and the maximum increment of NSE is 6.80%. This shows that the new model can effectively overcome the shortcomings of single hydrological model and give full play to the advantages of different forecasting models. Accurate forecasting of ship motion is of great significance for ensuring maritime operational safety and working efficiency. A data-driven ship motion forecast method is proposed in this paper, aiming at the problems of low generalization of a single forecast model and insufficient forecast accuracy under unknown conditions. First, the fluid dynamics simulations of the ship are carried out under multiple node conditions based on overset mesh technology, and the obtained motion data is used for training the Bidirectional Long Short-term Memory network models. One or more pre-trained forecast models would be selected based on the correlation of condition nodes when forecasting ship motion under non-node conditions. The Golden Jackal Optimization Algorithm is used to compute the regression coefficient of each node model in real time, and finally, the dynamic model average is calculated. The results show that the method proposed in this study can accurately forecast the pitch and heave of the KCS ship in 5 s, 10 s, and 15 s of forecast duration. The accuracy of the multi-order forecast model improves more in longer forecast duration tasks compared with the first-order model. When forecasting ship motion under non-node conditions, the method shows stronger model generalization capabilities.

Abstract Many residual network-based methods have been proposed to perform object detection. However, most of them may lead to overfitting or cannot perform well in small object detection and alleviate the problem of overfitting. We propose a multiple spatial residual network (MSRNet) for object detection. Particularly, our method is based on central point detection algorithm. Our proposed MSRNet employs a residual network as the backbone. The resulting features are processed by our proposed residual channel pooling module. We then construct a multi-scale feature transposed residual fusion structure consists of three overlapping stacked residual convolution modules and a transpose convolution function. Finally, we use the Center structure to process the high-resolution feature image for obtaining the final prediction detection result. Experimental results on PASCAL VOC dataset and COCO dataset confirm that the MSRNet has competitive accuracy compared with several other classical object detection algorithms, while providing a unified framework for training and reasoning. The MSRNet runs on GeForce RTX 2080Ti.

The Shanghai high-repetition-rate XFEL and extreme light facility (SHINE) plans to install several elliptically polarizing undulators (EPUs) as afterburners behind the planar undulator section to obtain nearly saturated circularly polarized free-electron laser (FEL) radiation. Therefore, the SHINE R&D project needs to develop a 4-m-long EPU with an effective magnetic field of 1.5 T, a period of 68 mm, and a minimum gap of 3 mm. A magnetic force compensation EPU prototype has recently been built and tested at the Shanghai Synchrotron Radiation Facility (SSRF) based on the addition of permanent magnets. This prototype can realize magnetic compensation under various polarization modes in all working gaps, thereby reducing the deformation of the girders and maintaining sufficient rigidity of the mechanical structure. A girder deformation monitoring system was established to obtain real force compensation feedback. The final magnetic field measurement results meet the physical requirements, and the proposed scheme can be used as an alternative to the EPU with a high magnetic field and large magnetic force.

In recent years, events of transmission line tripping caused by wildfires have occurred frequently in many mountain areas. In order to understand the mechanism of such events, an experimental system for simulating breakdown discharge processes of model wires induced by a simulated fire source is designed. In this experiment, propane jet flame (somewhat like eruptive fires in real wild fires) is used as a simulated flame source. By setting three different gas flow rates and five different wire-wire gaps, discharge breakdown behaviors and characteristics between two ACSR (aluminum conductors steel reinforced) segments in propane flames with three flame zones are explored. The influences of these different factors on the breakdown characteristics and mechanisms of wire-wire air gaps are analyzed. Results show that breakdown voltages increase almost linearly with the increase of wire-wire gaps in continuous zone of the flames for three gas flow rates. For a short wire-wire gap like 5.0 cm, the pilot discharge channels can be formed more easily, stably and completely under a lower voltage and weaker external ionization environment. The propane jet flame conditions are complex and affected by multiple actions like initial jet force (affected by gas flow rate), temperature distribution, soot particle, ion wind. As a result, local electric field will be distorted, a streamer channel is established dependently in different cases. These factors play individual, competitive or synergistic roles at the same time in breakdowns of real ACSR wire-wire gaps.

A series of porous S-doped carbon nitride ribbons (PSCN) were prepared by one-pot hydrothermal and sonochemical synthesis techniques. The morphologies and nanostructures of the catalysts were characterized by SEM, XRD and IR, which confirmed the pristine graphitic structures of carbon nitrides retained in the products. Due to sonication treatment, PSCN has porous structures in the thin ribbon and larger specific surface areas (PSCN 43.5 m2/g, SCN 26.6 m2/g and GCN 6.5 m2/g). XPS and elemental mappings verified that sulfur atoms were successfully introduced into the carbon nitride framework. Diffuse reflectance spectroscopy (DRS) results showed S-doping in the carbon nitride reduced the bandgap energy and enhanced their capability of the utilization of visible light, which contributed to higher photo-generated current. Photoluminescence (PL) analysis indicates the recombination of photogenerated carriers was suppressed in PSCN. Moreover, the photocatalytic performance showed that S-doping and porous and thin ribbon nanostructures may effectively boost the CO2 reduction rate (to as much as 5.8 times of GCN) when illuminated by visible light (>420 nm) without the need of sacrificial materials. The preliminary mechanisms of the formation of PSCN and its applications in photocatalytic CO2 reduction are proposed. It highlights the potential of the current technique to produce effective, nonmetal-doped carbon nitride photocatalysts.

Regarding the need to decrease carbon emissions, the electric vehicle (EV) industry is growing rapidly in China; the charging needs of EVs require the number of EV charging stations to grow significantly. Therefore, many refueling stations have been modified to integrated energy stations, which contain photovoltaic systems. The key issue in current times is to figure out how to operate these integrated energy stations in an efficient way. Therefore, an effective scheduling model is needed to operate an integrated energy station. Photovoltaic (PV) and energy storage systems are integrated into EV charging stations to transform them into integrated energy stations (PE-IES). Considering the demand for EV charging during different time periods, the PV output, the loss rate of energy storage systems, the load status of regional grids, and the dynamic electricity prices, a multi-objective optimization scheduling model was established for operating integrated energy stations that are connected to a regional grid. The model aims to simultaneously maximize the daily profits of the PE-IES, minimize the daily loss rate of the energy storage system, and minimize the peak-to-valley difference of the load in the regional grid. To validate the effectiveness of the model, simulation experiments under three different scenarios for the PE-IES were conducted in this research. Each object weight was determined using the entropy weight method, and the optimal solution was selected from the Pareto solution set using an order-preference technique according to the similarity to an ideal solution (TOPSIS). The results demonstrate that, compared to traditional charging stations, the daily revenue of the PE-IES stations increases by 26.61%, and the peak-to-valley difference of the power load in the regional grid decreases by 30.54%, respectively. The effectiveness of PE-IES is therefore demonstrated. Furthermore, to solve the complex optimization problem for PE-IES, a novel multi-objective optimization algorithm based on multiple update strategies (MOMUS) was proposed in this paper. To evaluate the performance of the MOMUS, a detailed comparison with seven other algorithms was demonstrated. These results indicate that our algorithm exhibits an outstanding performance in solving this optimization problem, and that it is capable of generating high-quality optimal solutions.

It has been well established that Foxp3+ regulatory T cells (Treg cells) play a crucial role for immune repression and tolerance, protecting the body from autoimmunity and inflammation. Previous studies indicate that intestinal Treg cells are one specialized population of Treg cells, distinct from those in other organ compartments, both functionally and phenotypically. Specific external and internal signals, particularly the presence of microbiota, shape these Treg cells to better cooperate with the gut ecosystem, controlling intestinal physiology. The integrity of intestinal epithelial barrier represents a key feature of gut immune tolerance, which can be regulated by multiple factors. Emerging evidence suggests that bidirectional interactions between gut epithelium and resident T cells significantly contribute to intestinal barrier function. Understanding how Treg cells regulate intestinal barrier integrity provides insights into immune tolerance-mediated mucosal homeostasis, which can further illuminate potential therapeutic strategies for treating inflammatory bowel disease and colon cancer.

With the development of image segmentation technology, image context information plays an increasingly important role in semantic segmentation. However, due to the complexity of context information in different feature maps, simple context capture operations can easily cause context information omission. Rich context information can better classify categories and improve the quality of image segmentation. On the contrary, poor context information will lead to blurred image category segmentation and an incomplete target edge. In order to capture rich context information as completely as possible, we constructed a Multi-Pooling Context Network (MPCNet), which is a multi-pool contextual network for the semantic segmentation of images. Specifically, we first proposed the Pooling Context Aggregation Module to capture the deep context information of the image by processing the information between the space, channel, and pixel of the image. At the same time, the Spatial Context Module was constructed to capture the detailed spatial context of images at different stages of the network. The whole network structure adopted the form of codec to better extract image context. Finally, we performed extensive experiments on three semantic segmentation datasets (Cityscapes, ADE20K, and PASCAL VOC2012 datasets), which fully proved that our proposed network effectively alleviated the lack of context extraction and verified the effectiveness of the network.

Abstract Background NAC transcription factors (TFs) are plant-specific proteins encoded by a large gene family. They play important roles in diverse biological processes, such as plant growth and development, leaf senescence, and responses to biotic or abiotic stresses. Functions of a number of NAC TFs have been identified mainly in model plants. However, very few studies on NAC TFs have been conducted in the fruit tree of kiwifruit. Results Genome-wide NAC genes were identified and their phylogeny, genomic structure, chromosomal location, synteny relationships, protein properties and conserved motifs were analyzed. In addition, the fruit developmental process was evaluated in a new kiwifruit cultivar of Actinidia eriantha ‘Ganlu 1’. And expressions for all those NAC genes were analyzed by quantitative real-time PCR method in fruits of ‘Ganlu 1’ during its developmental process. Our research identified 142 NAC TFs which could be phylogenetically divided into 23 protein subfamilies. The genomic structures of those NAC genes indicated that their exons were between one and ten. Analysis of chromosomal locations suggested that 116 out of 142 NACs distributed on all the 29 kiwifruit chromosomes. In addition, genome-wide gene expression analysis showed that expressions of 125 out of 142 NAC genes could be detected in fruit samples. Conclusion Our comprehensive study provides novel information on NAC genes and expression patterns in kiwifruit fruit. This research would be helpful for future functional identification of NAC genes involved in kiwifruit fruit development.

Elucidation of the molecular mechanisms involving the initiation and progression of radiation-induced esophageal injury (RIEI) is important for prevention and treatment. Despite ongoing advances, the underlying mechanisms controlling RIEI remain largely unknown. In the present study, RNA-seq was performed to characterize mRNA profiles of the irradiated rat esophagus exposed to 0, 25, or 35 Gy irradiation. Bioinformatics analyses including dose-dependent differentially expressed genes (DEGs), Gene Ontology (GO), Kyoto Encyclopedia of Gene and Genome (KEGG) pathway, protein-protein interaction (PPI) network, and immune infiltration were performed. 134 DEGs were screened out with a dose-dependent manner (35 Gy > 25 Gy > control, or 35 Gy < 25 Gy < control). GO and KEGG analyses showed that the most significant mechanism was IL-17 signaling-mediated inflammatory response. 5 hub genes, Ccl11, Cxcl3, Il17a, S100a8, and S100a9, were identified through the intersection of the DEGs involved in inflammatory response, IL-17 pathway, and PPI network. Additionally, immune infiltration analysis showed the activation of macrophages, monocytes, T cells, NKT cells, and neutrophils, among which macrophages, monocytes, and neutrophils might be the main sources of S100a8 and S100a9. Thus, these findings further our understanding on the molecular biology of RIEI and may help develop more effective therapeutic strategies.

Papillary thyroid cancer (PTC) accounts for about 90% of thyroid cancer. There are approximately 20%–30% of PTC patients showing disease persistence/recurrence and resistance to radioactive iodine (RAI) treatment. For these PTC patients with RAI refractoriness, the prognosis is poor. In this study, we aimed to establish a comprehensive prognostic model covering multiple signatures to increase the predictive accuracy for progression-free survival (PFS) of PTC patients with RAI treatment. The expression profiles of mRNAs and miRNAs as well as the clinical information of PTC patients were extracted from TCGA and GEO databases. A series of bioinformatics methods were successfully applied to filtrate a two-RNA model (IPCEF1 and hsa-mir-486-5p) associated with the prognosis of RAI-therapy. Finally, the RNA-based risk score was calculated based on the Cox coefficient of the individual RNA, which achieved good performances by the time-dependent receiver operating characteristic (tROC) curve and PFS analyses. Furthermore, the predictive power of the nomogram, integrated with the risk score and clinical parameters (age at diagnosis and tumor stage), was assessed by tROC curves. Collectively, our study demonstrated high precision in predicting the RAI response of PTC patients.

Abstract Parkinson’s disease (PD) is the second most common neurodegenerative disease, and mild cognitive impairment (MCI) is a well-established risk factor for the development of dementia in PD. A growing body of evidence suggests that low expression of glucocerebrosidase (GBA) promotes the transmission of α-synuclein (α-Syn) interpolymers and the progression of PD. However, how GBA mutations affect the pathogenesis of PD via abnormal aggregation of α-Syn is unclear, and no clinically valid PD-MCI genetic markers have been identified. Here, we first located a GBA eQTL, rs12411216, by analysing DHS, eQTL SNP, and transcription factor binding site data using the UCSC database. In this study, bacterial pathogens were isolated from the liver, spleen, kidney, heart, and muscle of diseased goldfish (Carassius auratus) after a massive outbreak of disease in an aquaculture farm in Anshan, Liaoning Province, North China (N41°05′58.42″ E122°53′31.89″). Based on physiological and biochemical characterization of the bacteria and 16S rDNA sequence analysis, the bacterial isolates were identified as Aeromonas salmonicida salmonicida. Virulence gene multilocus sequence typing analysis showed that the bacterial isolates from all the organs collected were of the same type, and clustered in the same phylogenetic group as A. salmonicida salmonicida. The pathogen, was named AS.17 and was resistant to rifampicin, bacitracin, vancomycin, penicillin and compound sulfamethoxazole, but was highly sensitive to nalidixic acid, lomefloxacin, spectinomycin, ofloxacin, and ciprofloxacin. Goldfish experimentally infected with AS.17 had similar symptoms to naturally infected fish, including gill filament anemia, intestinal bleeding, intra-abdominal fluid, and jaw bleeding. Histological analyses showed that the liver, the spleen, the kidney and the intestines of fish infected with AS.17 had severe pathological alterations. In summary, this study reports the isolation of A. salmonicida salmonicida from C. auratus in North China and provides a reference strain and a research foundation for further studies on A. salmonicida disease control and epidemiology. New ways of recycling fly ash are of great significance for reducing the environmental pollution. In this work, biodegradable hydrophobic poly (L-lactic acid)/fly ash composites for anti-icing application were successfully fabricated via a facile solvent-volatilization-induced phase separation approach. A silane coupling agent of 3-(Trimethoxysilyl) propyl methacrylate was used to decorate a fly ash surface (FA@KH570) for strengthening the interface bonding between fly ash and poly (L-lactic acid). Moreover, FA@KH570 could obviously enhance the crystallinity of poly (L-lactic acid) (PLLA)/FA@KH570 composites, which accelerated the conversion from the liquid-liquid to the liquid-solid phase separation principle. Correspondingly, the controllable surface morphology from smooth to petal-like microspheres was attained simply by adjusting the FA@KH570 content. After coating nontoxic candle grease, the apparent contact angle of 5 wt% PLLA/FA@KH570 composite was significantly increased to an astonishing 151.2°, which endowed the composite with excellent anti-icing property. This strategy paves the way for recycling waste fly ash and manufacturing hydrophobic poly (L-lactic acid) composite for potential application as an anti-icing material for refrigerator interior walls.

Kiwifruit (Actinidia eriantha) is a dioecious vine, and the pollen of its male cultivars has a direct effect on the quality of its fruits. In this study, to facilitate molecular breeding and gene identification, we performed genome-wide association studies (GWAS) on 11 traits of flower and leaf. A total of 946,337 highly consistent SNP markers were obtained in the whole genome. Phylogenetic tree analysis and population structure analysis showed that the 143 germplasms can be divided into two groups. The linkage disequilibrium analysis showed that A. eriantha have a relatively fast attenuation rate, and that the average attenuation distance of LD was 0.1–0.3 Kb. The MLM (QK) model was determined as best for correlation analysis, and eight and three SNPs associated with flower- and leaf-related traits were identified, respectively, at 0.01 significance level. However, SNP markers associated with stamen number per flower, pollen viability, total chlorophyll content, and total flavonoid content were not identified at the 0.01 significant level, although it is worth noting that one, one, five, and two SNPs were identified to be associated with these traits at the 0.05 significant level. This study provides insights into the complex flower- and leaf-related biology, and identifies genes controlling important traits in A. eriantha through GWAS, which extends the genetic resources and basis for facilitating molecular breeding in kiwifruits.

Obtaining optimal operation rules for cascade reservoirs is crucial for making the most of the comprehensive benefits of reservoirs. However, the operation of cascade reservoirs is generally complex, which involves with challenges including various decision variables, multiple conflicting objectives and constraints. In this paper, a new algorithm named multi-objective tangent algorithm (MOTA) is proposed for optimizing operation rules of cascade reservoirs with the objectives of hydropower generation, ecology and navigation. The performance of MOTA is firstly validated through several well-known benchmark problems. Then it is applied to a case study of cascade reservoirs optimization in China's Pearl River. Using observed inflow data (1956-2015) of cascade reservoirs, a uniform Pareto front is obtained eventually via MOTA after 1000 generations. The optimal operation rules fully considers the comprehensive benefits of hydropower generation, ecology and navigation in China's Pearl River. The optimal operation rules can be used as a guidance tool for decision makers, through the objectives' tradeoff without having to embed a priori preferences in the decision process. Finally, this paper use observed inflow data (2016) of cascade reservoirs for examining the operational rule to comprehend the analysis under different optimal operation rules. The obtained results show that MOTA can be a viable alternative for generating optimal operation rules for cascade reservoirs planning and management.

For the multi-energy power system composed of thermal power, wind power, and a pumped-storage power station aiming at minimizing coal consumption of the power grid, an optimal dispatch model is established in this paper. Its advantage is to allow the power grid to accept a high proportion of new energy while ensuring power demand. The dynamic programming method is used to solve the problem. In the solution process, the traditional dynamic programming method is improved by introducing the penalty function and the dynamic value of the state variable, which can ensure the reliability of the power supply while achieving the optimization goal, as well as realize the full utilization of energy. Using the example of a high proportion of wind power systems with a pumped-storage power station as the energy storage mode and considering the relevant constraints after the heating transformation of the thermal power plant, our built model solves these challenges. The results show that when the maximum pumping power of the pumped-storage power station reaches 1138 MW and the maximum generating power reaches 755 MW, the wind curtailment and power rationing during the off-peak period of heating can be reduced from the previous 58,158 MWH and 46,838 MWH to almost 0, and the wind curtailment and power rationing during the peak period of heating can be reduced from the previous 77,656 MWH and 53,780 MWH to almost 0, so as to realize the flexible operation of the power grid.

Sustainable conversion of CO2 to fuels using solar energy is highly attractive for fuel production. This work focuses on the synthesis of porous graphitic carbon nitride nanobelt catalyst (PN-g-C3N4) and its capability of photocatalytic CO2 reduction. The surface area increased from 6.5 m2·g−1 (graphitic carbon nitride, g-C3N4) to 32.94 m2·g−1 (PN-g-C3N4). C≡N groups and vacant N2C were introduced on the surface. PN-g-C3N4 possessed higher absorbability of visible light and excellent photocatalytic activity, which was 5.7 and 6.3 times of g-C3N4 under visible light and simulated sunlight illumination, respectively. The enhanced photocatalytic activity may be owing to the porous nanobelt structure, enhanced absorbability of visible light, and surface vacant N-sites. It is expected that PN-g-C3N4 would be a promising candidate for CO2 photocatalytic conversion.

Corticosteroid is commonly used to reduce damage from inflammatory reactions in coronavirus disease 2019 (COVID-19). We aim to determine the outcomes of corticosteroid use in critically ill COVID-19 patients. Ninety six critically ill patients, hospitalized in 14 hospitals outside Wuhan from January 16 to March 30, 2020 were enrolled in this study. Among 96 critical patients, 68 were treated with corticosteroid (CS group), while 28 were not treated with corticosteroids (non-CS group). Multivariable logistic regression were performed to determine the possible correlation between corticosteroid use and the treatment outcomes. Forty-six (68%) patients in the CS group died compared to six (21%) of the non-CS group. Corticosteroid use was also associated with the development of ARDS, exacerbation of pulmonary fibrosis, longer hospital stay and virus clearance time. On admission, no difference in laboratory findings between the CS and the non-CS group was observed. After corticosteroid treatment, patients treated with corticosteroids were associated with higher counts of white blood cells, neutrophils, neutrophil-to-lymphocyte ratio, alanine aminotransferase level and Sequential Organ Failure Assessment score. In conclusion, corticosteroid use in critically ill COVID-19 patients was associated with a much higher case fatality rate. Frequent incidence of liver injury and multi-organ failure in corticosteroid treated patients may have contributed to the adverse outcomes. The multi-organ failure is likely caused by more persistent SARS-CoV-2 infection and higher viral load, due to the inhibition of immune surveillance by corticosteroid.

The joint scheduling optimization of hydrothermal power is one of the most important optimization problems in the power system, which is a non-linear, multi-dimensional, non-convex complex optimization problem, and its difficulty in solving is increasing with the expansion of the grid-connected scale of hydropower systems in recent years. In this paper, three effective improvement strategies are proposed given the shortcomings of the standard collaborative search algorithm, which easily falls into local optimization and weakening of global search ability in later stages. Based on this, an elite collaborative search algorithm (ECSA) coupled with three improvement strategies is established. On this basis, taking the classic joint scheduling problem of a hydrothermal power system as an example, the optimization model with the goal of the least pollutant gas emission is constructed, and the system constraint treatment method is proposed. In addition, five algorithms, i.e., ECSA, CSA, PSO, GWO, and WOA are used to solve the model, respectively. Through the comparison of results, taking the median as an example, the emission of polluting gases of ESCA is reduced by about 1.8%, 13.1%, 38.2%, and 11.2%, respectively, and it can be found that ECSA has obvious advantages in the convergence speed and quality compared with the other four algorithms, and it has a strong ability for global search and jumps out of the local optimal.

Due to the inflexibility of cogeneration power plants and the uncertainty of wind power production, the excess power of the distribution network brings challenges to the power grid operation. This paper introduced an improved moth-flame optimization algorithm to meet the challenge of energy complementary dispatching. The proposed algorithm adopts three effective strategies, namely inertia weight, unified initialization, and the spiral position update strategy, which maintains a strong global search ability and a potent compromise between global and local search. The effectiveness of the proposed method was evaluated by benchmark functions. Furthermore, the proposed method was applied to combine heat and power system operation problems and economic dispatch in light load and wind power unpredictability. In order to verify the robustness of the algorithm and solve the complex constraints of power systems under extreme conditions, three different cases had been discussed. The experimental findings indicate that the proposed algorithm shows better performances in terms of convergence speed, ability to escape from a local optimum solution, and population diversity maintenance under different complexity conditions of engineering problems.

Summary: Obesity commonly co-exists with fatty liver disease with increasing health burden worldwide. Family with Sequence Similarity 13, Member A (FAM13A) has been associated with lipid levels and fat mass by genome-wide association studies (GWAS). However, the function of FAM13A in maintaining metabolic homeostasis in vivo remains unclear. Here, we demonstrated that rs2276936 in this locus has allelic-enhancer activity in massively parallel reporter assays (MPRA) and reporter assay. The DNA region containing rs2276936 regulates expression of endogenous FAM13A in HepG2 cells. In vivo, Fam13a−/− mice are protected from high-fat diet (HFD)-induced fatty liver accompanied by increased insulin sensitivity and reduced glucose production in liver. Mechanistically, loss of Fam13a led to the activation of AMP-activated protein kinase (AMPK) and increased mitochondrial respiration in primary hepatocytes. These findings demonstrate that FAM13A mediates obesity-related dysregulation of lipid and glucose homeostasis. In flood control scheduling of reservoirs, the usual conventional scheduling fails to obtain the optimal solution to the problem. The dynamic programming method is applied to the field. However, the problem of ‘dimensional disaster’ restricts its application. To solve these problems, the improved DP is constructed according to the principle of period inflow and water balance, which includes a variable dispersion mechanism of the retraction space and a calculation method for the smallest discrete points of each discrete range. Taking Dongjiang Reservoir as the research object, based on the maximum peak clipping criterion and the maximum flood control safety guarantee criterion, the improved DP is used for optimization. The results find that when the former criterion is used for scheduling, the discharge flow processes of the two floods are more uniform than conventional scheduling. When the latter criterion is used for scheduling, the minimum water levels of the two floods are lower than the conventional scheduling. As such, it is found that in the two flood dispatches, the calculation time after the first flood DP dimensionality reduction processing is reduced by about 65%, and the second flood is reduced by about 59%, which greatly improves the calculation efficiency of the DP.

Abstract Inhibition of amyloid β-protein (Aβ) aggregation is considered as a promising strategy for the prevention and treatment of Alzheimer’s disease. Epigallocatechin-3-gallate (EGCG) and curcumin have been recognized as effective inhibitors of Aβ aggregation. Herein, we proposed dual-inhibitor modification of hyaluronic acid (HA) to explore the synergistic effect of the two inhibitors. EGCG-modified HA (EHA) formed dispersed hydrogel structures, while EGCG-curcumin bi-modified HA (CEHA) self-assembled into nanogels like curcumin-modified HA (CHA). Thioflavin T fluorescent assays revealed that the inhibitory effect of CEHA was 69% and 55% higher than EHA and CHA, respectively, and cytotoxicity assays showed that the viability of SH-SY5Y cells incubated with Aβ and CEHA was 28% higher than that with Aβ and the mixture of EHA and CHA. These results clearly indicate the synergism of the two inhibitors. It is considered that the difference in the hydrophobicities of the two inhibitors made the bi-modification of HA provide a favorable CEHA nanostructure that coordinated different inhibition effects of the two inhibitors. This research indicates that fabrication of dual-inhibitor nanosystem is promising for the development of potent agents against Aβ aggregation and cytotoxicity.

Charitable crowdfunding provides a new channel for people and families suffering from unforeseen events, such as accidents, severe illness, and so on, to seek help from the public. Thus, finding the key determinants which drive the fundraising process of crowdfunding campaigns is of great importance, especially for those suffering. With a unique data set containing 210,907 crowdfunding projects covering a period from October 2015 to June 2020, from a famous charitable crowdfunding platform, specifically Qingsong Chou, we will reveal how many online donations are due to endogeneity, referring to the positive feedback process of attracting more people to donate through broadcasting campaigns in social networks by donors. For this aim, we calibrate three different Hawkes processes to the event data of online donations for each crowdfunding campaign on each day, which allows us to estimate the branching ratio, a measure of endogeneity. It is found that the online fundraising process works in a sub-critical state and nearly 70–90% of the online donations are endogenous. Furthermore, even though the fundraising amount, number of donations, and number of donors decrease rapidly after the crowdfunding project is created, the measure of endogeneity remains stable during the entire lifetime of crowdfunding projects. Our results not only deepen our understanding of online fundraising dynamics but also provide a quantitative framework to disentangle the endogenous and exogenous dynamics in complex systems.

This experiment was conducted to determine the toxicological effect of selenomethionine (Se-met) and bio-fermenting Selenium (Se-bio) supplementation with basal diet to investigate growth performance, bioaccumulation and non-specific immune function in sea cucumber, (Apostichopus japonicus). How the human brain differs from those of non-human primates is largely unknown and the complex drivers underlying such differences at the genomic level remain unclear. In this study, we selected 243 brain-related genes, based on Gene Ontology, and identified 184,113 DNaseI hypersensitive sites (DHSs) within their regulatory regions. To performed comprehensive evolutionary analyses, we set strict filtering criteria for alignment quality and filtered 39,132 DHSs for inclusion in the investigation and found that 2,397 (~6%) exhibited evidence of accelerated evolution (aceDHSs), which was a much higher proportion that DHSs genome-wide. Target genes predicted to be regulated by brain-aceDHSs were functionally enriched for brain development and exhibited differential expression between human and chimpanzee. Alignments indicated 61 potential human-specific transcription factor binding sites in brain-aceDHSs, including for CTCF, FOXH1, and FOXQ1. Furthermore, based on GWAS, Hi-C, and eQTL data, 16 GWAS SNPs, and 82 eQTL SNPs were in brain-aceDHSs that regulate genes related to brain development or disease. Among these brain-aceDHSs, we confirmed that one enhanced the expression of GPR133, using CRISPR-Cas9 and western blotting. The GPR133 gene is associated with glioblastoma, indicating that SNPs within DHSs could be related to brain disorders. These findings suggest that brain-related gene regulatory regions are under adaptive evolution and contribute to the differential expression profiles among primates, providing new insights into the genetic basis of brain phenotypes or disorders between humans and other primates.

A four-weeks feeding trial was conducted to evaluate the effects of dietary Bio-fermented selenium (Bio-Se) on zebrafish (Danio rerio). Effects of three Se diets containing basal diet (1.63 mg Se/kg), L-Se diet (3 mg Se/kg), and H-Se diet (10 mg Se/kg) on growth performance, nonspecific immune enzyme activities, proximate composition, and selenium accumulation of zebrafish were tested. The results indicate that body weight gain (BWG), specific growth rate (SGR), and nonspecific immune enzyme [glutathione peroxidase (GPx), superoxide dismutase (SOD), and alkaline phosphatase (ALP)] activities of fish fed with L-Se diet were significantly highest among all the groups. However, increased H2O2 content and decreased SOD activity indicates oxidative stress when fish was fed with H-Se diet. Dietary Se did not affect the proximate composition of zebrafish. The Se concentration in muscle increased in a dose-dependent manner, and the highest Se concentration was recorded in H-Se diet. In summary, the growth performance and nonspecific immunity of zebrafish fed with L-Se diet improved, but exposure to higher concentrations of Bio-Se severely affects the health status of fish. Hydrogel adhesives are attractive for applications in intelligent soft materials and tissue engineering, but conventional hydrogels usually have poor adhesion. In this study, we designed a strategy to synthesize a novel adhesive with a thin hydrogel adhesive layer integrated on a tough substrate hydrogel. The adhesive layer with positive charges of ammonium groups on the polymer backbones strongly bonds to a wide range of nonporous materials’ surfaces. The substrate layer with a dual hydrogen bond system consists of (i) weak hydrogen bonds between N,N-dimethyl acrylamide (DMAA) and acrylic acid (AAc) units and (ii) strong multiple hydrogen bonds between 2-ureido-4[1H]-pyrimidinone (UPy) units. The dual hydrogen-bond network endowed the hydrogel adhesives with unique mechanical properties, e.g., toughness, highly stretchability, and insensitivity to notches. The hydrogel adhesion to four types of materials like glass, 316L stainless steel, aluminum, Al2O3 ceramic, and two biological tissues including pig skin and pig kidney was investigated. The hydrogel bonds strongly to dry solid surfaces and wet tissue, which is promising for biomedical applications.

In order to analyze the year-end water level of multi-year regulating reservoir of the cascade hydropower system, this paper studied the joint operation optimization model of cascade reservoirs and its solving method based on multi-dimensional dynamic programming, and analyzed the power generation impact factors of cascade system that contains multi-year regulating reservoir. In particular, taking the seven reservoirs in the middle and lower reaches of Yalong River as an example, the optimal year-end water levels of multi-year regulating reservoir under the multi-year average situation and different inflow frequencies situation were studied. Based on the optimal calculation results of multi-dimensional dynamic programming, the inflow frequency difference considered operation rule of year-end water level of Lianghekou reservoir was extracted using the least square principle. The simulation results showed that, compared with the fixed year-end water level in multi-year, the extracted rule can improve the cascade power generation by more than 400 million kWh in an average year, representing an increase of 0.4%. This result means that the extracted rule can give full play to the regulation performance of multi-year regulating reservoir and improve the conversion efficiency of hydropower resources in cascade system. This is of great significance to the practical operation of cascade reservoirs system that contains multi-year regulating reservoir.

Studies on organic acid metabolism have been mainly concentrated on the fruit, whereas, few have focused on the mechanism of high organic acids content in the fruit of Actinidia eriantha. Fruits of ‘Ganmi 6’ harvested at eleven developmental periods were used as materials. The components and content of organic acids were determined by high-performance liquid chromatography (HPLC) system, the activities of the related enzyme were detected, and gene expression levels were measured by quantitative real-time PCR (qRT-PCR). Components of ascorbic acid (AsA) and eight kinds of organic acids were detected. These results showed that quinic acid and citric acid were the main organic acids in the fruit of ‘Ganmi 6’. Correlation analysis showed that NADP-Quinate dehydrogenase (NADP-QDH), NADP-Shikimate dehydrogenase (NADP-SDH), and Cyt-Aconitase (Cyt-Aco) may be involved in regulating organic acids biosynthesis. Meanwhile, the SDH gene may play an important role in regulating the accumulation of citric acid. In this study, the activities of NADP-SDH, Mit-Aconitase (Mit-Aco), and NAD-Isocitrate dehydrogenase (NAD-IDH) were regulated by their corresponding genes at the transcriptional level. The activity of Citrate synthase (CS) may be affected by post-translational modification. Our results provided new insight into the characteristics of organic acid metabolism in the fruit of A. eriantha.

In some studies, the metal materials with different magnetic permeability and same chemical composition need to be prepared to avoid the interference caused by different compositions and make the parameter of magnetic permeability become the key variable directly. In order to prepare the metal materials with different magnetic and same component, the paper has considered other two commonly schemes, such as the casting method by adding some ferromagnetic elements and the heat-treatment process to increase its phase content of martensite or ferrite. Finally, it has been found that the cold working process was preferable if the austenitic stainless steel 1Cr18Ni9Ti with paramagnetic was selected as a raw material, because it got better control in forging ratio. The method of magnetic balance has been used to measure the relative magnetic permeability of every specimen with different hardness HB 165 (A), HRC 20 (B), HRC 25 (C) and HRC 30 (D). The metallurgical microscope, XRD, TEM and SEM has been put to use to analyse their microstructure before and after forging. The results show, the cold working process improved magnetic properties of 1Cr18Ni9Ti continuously and largely within limits by controlling different forging rate, as well as the process was not complicated. The relative rate of magnetic permeability for 1Cr18Ni9Ti and W18Cr4V (high-speed steel) could be up to 1:2.435 at 0.03 T. The 20.1 % maximum reduction of the austenite phase should be a main reason to improving magnetic permeability of 1Cr18Ni9Ti. The real magneto-crystalline anisotropy should be due to martensite or ferrite phases with strong magnetic property. The 1Cr18Ni9Ti after cold working still maintained high corrosion resistance. High moisture content along with poor dewaterability are the main challenges for sewage sludge treatment and utilization. In this study, the effect of hydrothermal treatment at various temperature (120-200 ˚C) on the properties of sewage sludge derived solid fuel was investigated in the terms of mechanical dewatering character, drying character, calorific value and heavy metal distribution. Hydrothermal treatment (HT) followed by dewatering process significantly reduced moisture content and improved calorific value of sewage sludge with the optimum condition obtained at 140˚C. No significant alteration of drying characteristic was produced by HT. Heavy metal enrichment in solid particle was found after HT that highlighted the importance of further study regarding heavy metal behavior during combustion. However, it also implied the potential application of HT on sewage sludge for heavy metal removal from wastewater.

The lethal concentration of water-borne copper in Chinese mitten crab Eriocheir sinensis larvae was tested by exposing the animals to 0, 0.1, 0.2, 0.3, 0.5 and 0.8 mg Cu L-1 at 20°C for 96 h. The 96-h median lethal concentration (LC50) and its corresponding 95% confident interval was estimated on zoea 1 larvae and megalopa larvae, respectively. Acute dissolved copper toxicity was higher for zoea 1 larvae (0.16 mg L-1) than for megalopa larvae (0.21 mg L-1). The antioxidant status, metallothionein-I mRNA expression and physiological response of the crab to copper toxicity was further investigated by exposing the megalopa larvae to 0, 0.08 and 0.16 mg Cu L-1 for 96 h. The superoxide dismutase activity, catalase activity, glutathione S-transferase (GST) activity and lipid peroxidation content of megalopa larvae increased concomitantly with the exposure time and copper concentration. MT-I mRNA expression levels were positively correlated with both the concentration and duration of copper exposure. The oxygen consumption and respiratory quotient of megalopa larvae in response to 0.16 mg L-1 copper were significantly higher than those in the control group after 96 h of exposure (P < 0.05). The results of this study highlight the potential effects of copper as a common stressor in E. sinensis larvae. MT-I and GST appear to be suitable biomarkers of environmental copper exposure stress in E. sinensis larvae

In view of the problems that have not been solved or studied in the previous studies of cascade Energy Storage Operation Chart (ESOC), based on a brief description of the composition, principle, drawing methods, and simulation methods of ESOC, the following innovative work has been done in this paper. Firstly, considering the inconsistency of inflow frequency of upstream and downstream watershed in selecting the typical dry years, a novel optimization model for selecting the overall inflow process considering the integrity of watershed was proposed, which aimed at minimizing the sum of squares of inflow frequency differences. Secondly, aiming at the influence of output coefficients (including number and values) on the results of ESOC, this paper proposed a new method to construct the initial solution of output coefficients and established an optimization model of output coefficients based on progressive optimality algorithms. Thirdly, to the optimization of ESOC with multi-year regulating reservoir, a discrete optimization model of drawdown level was constructed based on the idea of ergodic optimization. On these bases, taking the seven reservoirs in the Yalong River basin of China as an example, the typical dry years considering the inflow frequency inconsistency, the optimal output coefficients of ESOC and the optimal end-of-year drawdown level of a multi-year regulating reservoir (Lianghekou) were obtained, and compared with the previous research results, the ESOC optimized in this paper can increase the total power generation of the cascade system by 9% under the condition that the guaranteed rate did not change much. Furthermore, the difference of the optimal end-of-year drawdown levels between the cascade joint operation and single reservoir operation was discussed for the Lianghekou reservoir at the end of the case study. The obtained results were of great significance for guiding the actual operation of cascade reservoirs.

A reservoir operation chart is an important tool in guiding actual reservoir operation at present. There are mainly two kinds of methods in drawing the operation chart, i.e., conventional methods and optimization methods, but each of them has some shortcomings, such as the repeated empirical inspection and correction of conventional methods, and the sensitivity to the initial trajectories of some optimization algorithms. In view of this, based on the principle of dynamic programming (DP), this paper coupled the reservoir operation chart drawing model and the DP model, and proposed a new reservoir operation chart drawing method which has no empirical inspection and correction, no requirement for initial solution, no problem of premature convergence and local convergence. In addition, this method can guarantee the global convergence of the results to a certain extent because of the global convergence of DP. Ya Yangshan reservoir in the Li Xianjiang River of China was selected as the research object to derive the operation chart by the drawing method. The simulation results show that the proposed method in this paper presents better performance compared with the conventional method on power generation, guaranteed output, and assurance rate, especially on the latter, which has a 2.68% increase. In addition, compared with the deterministic optimization results of DP, it is found that the results of the proposed method are very close to that of deterministic DP, the differences are only 1.8 GWh (0.36% decline) and 1.6 GWh (0.32% decline). So, the validity and rationality of the proposed method are further verified by the simulation results.

Single nucleotide polymorphisms (SNPs) have become the marker of choice for genome-wide association studies in many species. High-throughput sequencing of RNA was developed primarily to analyze global gene expression, while it is an efficient way to discover SNPs from the expressed genes. In this study, we conducted transcriptome sequencing of the swimbladder of Takifugu rubripes using Illumina HiSeq2000 platform to identify gene-associated SNPs in the swimbladder. A total of 30,312,181 unique-mapped-reads were obtained from 44,736,850 raw reads. A total of 62,270 putative SNPs were discovered, which were located in 11,306 expressed genes and 2,246 scaffolds. The average minor allele frequency (MAF) of the SNPs was 0.26. GO and KEGG pathway analysis were conducted to analyze the genes containing SNPs. Validation of selected SNPs revealed that 54% of SNPs (26/48) were true SNPs. The results suggest that RNA-Seq is an efficient and cost-effective approach to discover gene-associated SNPs. In this study, a large number of SNPs were identified and these data will be useful resources for population genetic study, evolution analysis, resource assessment, genetic linkage analysis and genome-wide association studies.

The fish gill, as one of the mucosal barriers, plays an important role in mucosal immune response. The fish swimbladder functions for regulating buoyancy. The fish swimbladder has long been postulated as a homologous organ of the tetrapod lung, but the molecular evidence is scarce. In order to provide new information that is complementary to gill immune genes, initiate new research directions concerning the genetic basis of the gill immune response and understand the molecular function of swimbladder as well as its relationship with lungs, transcriptome analysis of the fugu Takifugu rubripes gill and swimbladder was carried out by RNA-Seq. Approximately 55,061,524 and 44,736,850 raw sequence reads from gill and swimbladder were generated, respectively. Gene ontology (GO) and KEGG pathway analysis revealed diverse biological functions and processes. Transcriptome comparison between gill and swimbladder resulted in 3,790 differentially expressed genes, of which 1,520 were up-regulated in the swimbladder while 2,270 were down-regulated. In addition, 406 up regulated isoforms and 296 down regulated isoforms were observed in swimbladder in comparison to gill. By the gene enrichment analysis, the three immune-related pathways and 32 immune-related genes in gill were identified. In swimbladder, five pathways including 43 swimbladder-enriched genes were identified. This work should set the foundation for studying immune-related genes for the mucosal immunity and provide genomic resources to study the relatedness of the fish swimbladder and mammalian lung.
