Abstract

Coix lacryma-jobi L. belongs to family Poaceae, is widely cultivated in tropical Asian countries for its nutritional and medicinal values. Coix is often threatened by lepidopteran such as Mythimna separata during its life cycle, resulting in severe yield reduction. Insect feeding can trigger defense signaling and increased defense responses in many other crops, yet little is known about whether simulated armyworm feeding on Coix leaves could induce anti-herbivory responses and whether armyworm feeding could activate priming in systemic leaves. In this study, Mythimna separata simulated herbivory elicited increased jasmonic acid (JA) level, JA-Ile (JA-isoleucine conjugate) and altered transcriptome in the Coix leaves. Meanwhile, M. separata simulated herbivory in local leaves primed the systemic leaves for increased accumulation of jasmonic acid and enhanced resistance to M. separata. Consistently, transcriptome analysis showed the systemic leaves were primed, which were up- or down-regulated comparing with the non-primed systemic leaves. In this study, we first reported Mythimna separata simulated herbivory induced increased defense response in leaves of Coix, also Mythimna separata herbivory primed Coix resistance in systemic leaves. 15. A surface-based approach to the handling of uncertainties in an urban-orientated spatial database

Abstract

The work presented in this thesis is built on three assertions: (1) uncertainties should be perceived as integral components of GIS spatial databases; (2) as such and given the importance of uncertainties at all stages of processing spatial data by digital methods, an integrating strategy is needed to provide more direct access to the uncertainties of spatial data during data collection, update, spatial analysis and during the creation of output products; (3) surface-based models and methods are capable of such an integral strategy, by which many kinds of uncertainties of spatial data can be well represented and handled. A cumulative description is given of various uncertainties occurring in geographical abstraction and spatial data acquisitions with special reference to one common area of geographical studies, that is, land cover mapping. Two alternative forms of geographical abstraction or spatial data modelling are introduced: discrete objects and continuous fields. The uncertainties are then discussed with respect to their description, estimation and representation under object and field perspectives. For categorical data, in particular, uncertainties are represented as fuzzy surfaces, whose derivation and analysis are described in detail. To provide an evaluation of the integrated approach and to show how such an integrated strategy can be used to advantage, a case study is developed in the context of suburban land cover mapping, based on a local Edinburgh area. The case study begins with the construction of a co-registered hierarchy of test data with a corresponding hierarchy of accuracies, and continues to the generation and analysis of fuzzy surfaces using the suite of methods introduced previously. The various graphical maps and quantitative data produced show that surface based approaches are well suited to the representation and handling of uncertainties of spatial data, because they are effective and flexible.

25. Service Deployment Model on Shared Virtual Network Functions With Flow Partition

Abstract

Network operators can operate services in a flexible way with virtual network functions thanks to the network function virtualization technology. Flow partition allows aggregated traffic to be split into multiple parts, which increases the flexibility. This paper proposes a service deployment model with flow partition to minimize the service deployment cost with meeting service delay requirements. A virtual network function of a service is allowed to have several instances, each of which hosts a part of flows and can be shared among different services, to reduce the initial and proportional cost. We provide the mathematical formulation for the proposed model and transform it to a special case as a mixed integer second-order cone programming (MISOCP) problem. A heuristic algorithm, which is called a flow partition heuristic (FPH), is introduced to solve the original problem in practical time by decomposing it into several steps; each step handles a convex problem. We compare the performances of proposed model with flow partition and conventional model without flow partition. We consider the formulated MISOCP problem with adopting a strategy of even splitting to divide flows in a special case, which is called an even spitting heuristic (ESH). The performances of FPH and ESH are compared in a realistic scenario. We also consider the formulated MISOCP problem as an original problem and compare it to an FPH-based heuristic algorithm with the even-splitting strategy (FPH-ES), in both realistic and synthetic scenarios. The numerical results reveal that the proposed model saves the service deployment cost compared to the conventional one. It improves the maximum admissible traffic scale by 23% in average in our examined cases. We observe that FPH outperforms ESH and ESH outperforms FPH-ES in terms of the service deployment cost in their own focused problems, respectively.

Abstract

Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural dines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale.

Abstract

Herbivory activates responses in local and systemic leaves, and the glutamate receptor-like genes GLR3.3 and GLR3.6 are critical in leaf-to-leaf systemic signalling. However, whether and how these genes mediate plant systemic resistance to insects remain largely unexplored. We show that a piercing-sucking insect Myzus persicae (green peach aphid, GPA) or chewing insect Spodoptera litura (cotton leafworm, CLW) feeding-induced systemic defences were attenuated in the glr3.3 glr3.6 mutants. In response to herbivory from either insect, glr3.3 glr3.6 mutants exhibited reduced accumulation of the hormone jasmonic acid (JA) and defensive metabolites glucosinolates (GSs) in systemic (but not local) leaves. Transcriptome analysis indicated that GLR3.3 and GLR3.6 play an important role in regulating the transcriptional responses to GPA and simulated CLW feeding in both local and systemic leaves, including JA- and GS-related genes. Metabolome analysis also revealed that in response to GPA or simulated CLW feeding, GLR3.3 and GLR3.6 are involved in the regulation of various metabolites locally and systemically, including amino acids, carbohydrates, and organic acids. Taken together, this study provides new insights into the function of GLR3.3 and GLR3.6 in mediating transcripts and metabolites in local and systemic leaves under insect attack, and highlights their role in regulating insect resistance in systemic leaves. 45. An Adaptive Sampling Strategy for Land Cover Change Information and Its Accuracy Characterization

Abstract

An adaptive sampling strategy is proposed for location-specific characterization of accuracy in land cover change information. The local accuracy characterization strategy was established based on local patterns of land cover change maps (e.g land cover change classes, patch size, heterogeneity and dominance), which include exploring covariates significantly relate to accuracy. Standard error of prediction accuracy was used for identifying the area which needs to improve the reliability of prediction accuracy and locating samples adaptively and progressively. The performance of different sampling methods for accuracy prediction was evaluated at the same testing samples in Wuhan. It was indicated that 100 more training samples selected by adaptive sampling strategy lead to about 50.66% increase in prediction accuracy, as measured by sums-of-squares. In comparison, for random sampling, the same increase in training sample size led to about 17.22% increase in prediction accuracy, as measured by sums-of-squares. This confirms that adaptive sampling strategy improves the sampling efficiency while reduces the uncertainty in local accuracies prediction. Model selection reveals that land cover change classes and dominance are the highest significant covariates.
