2. Epidemiological investigation of goose circovirus based on a newly developed indirect ELISA method.

Abstract

Goose circovirus (GoCV) is a recently identified pathogen in geese that is known to cause slow growth, feather disorder syndrome, and immunosuppression. Infection with GoCV may increase the risk of coinfections with multiple pathogens, leading to significant economic losses in the goose industry. However, due to a lack of serological detection methods, analysis of viral nucleic acids has been widely used in GoCV epidemiological surveys, which has limited accurate monitoring of the prevalence of GoCV. In this study, we developed and optimized an indirect ELISA method based on the prokaryotic-expressed recombinant GoCV capsid protein (△Cap-iELISA). The △Cap-iELISA was then used to test 349 goose serum samples collected from Guangdong, Shandong, and Fujian provinces during 2023 and 2024. The results showed that the positive rate of GoCV antibodies in the sampled geese was 71.06%. Further analysis indicated that the positive rate of GoCV antibodies increased with the age of the geese. In conclusion, we have developed a novel iELISA method that is well-suited for large-scale clinical detection and early diagnosis of GoCV infection. 10. Immunomodulatory Effects of Yupingfeng Polysaccharides on Immunosuppressed Mice.

Abstract

[Objective] To discuss the protection and regulatory effects of Yupingfeng polysaccharides on immunosuppressed mice. [Methods] The immunosuppressive mice model was established by 80 mg/kg cyclophosphamide. The spleen index, spleen lymphocyte proliferation capability, serum hemolysin antibody generation and serum IL-2 level of mice were researched, so as to observe the effects of Yupingfeng polysaccharides at different dosages (100, 200, 400 mg/kg). [Results] Yupingfeng polysaccharides enhanced the spleen index of immunosuppressed mice, improved the proliferation capability of spleen lymphocyte, promoted the generation of serum hemolysin antibody in normal mice, and protected against the secretion reduction of serum hemolysin antibody induced by cyclophosphamide. Besides, Yupingfeng polysaccharides enhanced the IL-2 levels in serum of both immunosuppressed and normal mice, especially that the middle- and low-dosage groups had relatively significant effects. [Conclusions] Yupingfeng polysaccharides protected against the immunosuppression of mice induced by cyclophosphamide, enhanced the immune function and protected the spleen function of mice. 11. Ac and Dc insulation leakage in platinum resistance thermometers up to 750 °C

Abstract

The ac and dc insulation resistance of two open-circuited, commercial, standard platinum resistance thermometers is investigated over the range 0-750 °C and 2-4000 Hz, to determine the magnitude of temperature errors caused by the insulation shunt across the platinum sensor. It is found that ac insulation leakage is always greater than dc leakage; that ac leakage increases as the test frequency increases; and that both ac and dc leakage usually increase rapidly as the temperature increases above 500 °C. For example, the temperature errors at 630 °C caused by insulation leakage when a thermometer is measured with dc, 100 Hz and 1000 Hz excitation can reach about 1, 2 and 8 m °C respectively. With a metal-sheathed thermometer, it is confirmed experimentally that the more readily measured insulation resistance between the sheath and the leads gives a useful approximation to the true shunt across the sensor at high temperatures. Similarly, it is shown for a quartz-sheathed thermometer that the insulation resistance between the leads and a metal mesh placed over the sheath will also approximate the true shunt at high temperatures. Other properties investigated include: dc leakage as a function of the thermometer furnace immersion; ac leakage in a thermometer containing moist air; ac leakage in the external thermometer leads; and the ac capacitance of the thermometer and leads.
