Background Transportation contributes most of the carbon emissions in dentistry. Therefore, this study investigates the travel-related carbon footprint at Harvard School of Dental Medicine (HSDM) and explores factors that impact it.Methods A cross-sectional survey, developed and administered to the HSDM community, examined the modes and frequency of transportation utilized prior to and since the start of the COVID-19 pandemic, factors influencing transportation choices, and perceptions of climate change. The Mann-Whitney U test and Chi-square test were used to test statistical significance.Results Since the start of the pandemic, respondents had significantly lower annual carbon footprint (U = 1181, p-value = 0.049) and fewer weekly travel days to/from HSDM (U = 4457, p-value = 0). Over 95% and 85% of respondents are concerned about climate change and the effect of transportation on climate change, respectively.Conclusions Since the onset of the COVID-19 pandemic, HSDM’s travel-related carbon emissions have been significantly reduced, likely due to the adoption of hybrid work schedules and resulting shifts in transportation habits.Practical Implications This study highlights a methodology for assessing travel carbon footprints in other dental schools or similar settings, which is a crucial first step in understanding these impacts.

Abstract Objective Fibrinogen, essential in primary hemostasis, platelet aggregation, and leukocyte-endothelial interactions, is also associated with a heightened risk of acute ischemic stroke (AIS). However, its influence on AIS patient outcomes is unclear. This study examines the correlation between fibrinogen levels and the risk of unfavorable outcomes three months post-AIS. Methods This is a secondary analysis of a prospective cohort study conducted in Korea. The sample consisted of 1851 AIS patients who received treatment at a Korean hospital between January 2010 and December 2016. Statistical models were established to understand the relationship between fibrinogen levels(mg/dL) and unfavorable outcomes(mRs ≥ 3), including logistic regression models, Generalized Additive Models (GAM), and smooth curve fitting (penalized splines). The log-likelihood ratio test has been utilized to evaluate the best fit. To ensure the robustness of the results, sensitivity analyses were conducted by reanalyzing the relationship after excluding participants with TG > 200 mg/dl and BMI > 25 kg/m2. Subgroup analyses were also performed to assess whether influencing factors modify the association between fibrinogen levels and unfavorable outcomes. Results After adjusting for multiple covariates including age, BMI, sex, LDL-c, TG, HGB, HDL-c, BUN, FPG, ALB, PLT, AF, hypertension, smoking, DM, mRs score at admission, the binary logistic regression model demonstrated revealed a significant positive association between fibrinogen levels and the risk of unfavorable outcomes in AIS patients (OR = 1.215, 95% CI: 1.032–1.429, p = 0.019). Abstract Background Polycystic ovary syndrome (PCOS) has unusual levels of hormones. The hormone receptors in the endometrium have a hostile effect and make the microenvironment unfavorable for embryo implantation. The use of gonadotropin stimulation during in vitro fertilization (IVF) may have an impact on embryo implantation and live birth rate. According to recent data, the clinical results of day 4 embryo transfer (D4 transfer) were on par with those of day 5 embryo transfer (D5 transfer) in IVF-ET. There are few studies comparing the outcomes of transplants with various etiologies and days. The purpose of this study was to determine which transfer day had the best result for PCOS patients undergoing IVF. Methods This retrospective cohort study was conducted in the Xingtai Infertility Specialist Hospital between January 2017 and November 2021. A total of 1,664 fresh ART cycles met inclusion criteria, including 242 PCOS transfers and 1422 tubal factor infertility transfers. Conclusions PCOS individuals had the highest live birth rate on D4 transferred. It was not need to culture embryos to blastocysts to optimize embryo transfer for PCOS women. This could be a novel approach to transplantation for PCOS.

In this study, we executed single-cell RNA sequencing of intestinal crypts. We analyzed the differentially expressed genes (DEGs) at different time points (the first, third, and fifth days) after 13 Gy and 15 Gy abdominal body radiation (ABR) exposure and then executed gene ontology (GO) enrichment analysis, RNA velocity analysis, cell communication analysis, and ligand‒receptor interaction analysis to explore the vital events in damage repair and the multiple effects of the Wnt3/β-catenin pathway on irradiated mice. Results from bioinformatics analysis were confirmed by a series of biological experiments. Results showed that the antibacterial response is a vital event during the damage response process after 13 Gy ABR exposure; ionizing radiation (IR) induced high heterogeneity in the transient amplification (TA) cluster, which may differentiate into mature cells and stem cells in irradiated small intestine (SI) crypts. Conducting an enrichment analysis of the DEGs between mice exposed to 13 Gy and 15 Gy ABR, we concluded that the Wnt3/β-catenin and MIF-CD74/CD44 signaling pathways may contribute to 15 Gy ABR-induced mouse death. Wnt3/β-catenin promotes the recovery of irradiated SI stem/progenitor cells, which may trigger macrophage migration inhibitory factor (MIF) release to further repair IR-induced SI injury; however, with the increase in radiation dose, activation of CD44 on macrophages provides the receptor for MIF signal transduction, initiating the inflammatory cascade response and ultimately causing a cytokine release syndrome. In contrast to previous research, we confirmed that inhibition of the Wnt3/β-catenin pathway or blockade of CD44 on the second day after 15 Gy ABR may significantly protect against ABR-induced death. This study indicates that the Wnt3/β-catenin pathway plays multiple roles in damage repair after IR exposure; we also propose a novel point that the interaction between intestinal crypt stem cells (ISCs) and macrophages through the MIF-CD74/CD44 axis may exacerbate SI damage in irradiated mice.

Abstract Background Artemisia vulgaris L. is often used as a traditional Chinese medicine with the same origin of medicine and food. Its active ingredient in leaves have multiple biological functions such as anti-inflammatory, antibacterial and insecticidal, anti-tumor, antioxidant and immune regulation, etc. It is confirmed that folium Artemisiae argyi has obvious anti-HBV activity, however, its antiviral activity and mechanism against herpesvirus or other viruses are not clear. Hence, we aimed to screen the crude extracts (Fr.8.3) isolated and extracted from folium A. argyi to explore the anti-herpesvirus activity and mechanism. Methods The antiherpes virus activity of Fr.8.3 was mainly characterized by cytopathic effects, real-time PCR detection of viral gene replication and expression levels, western blotting, viral titer determination and plaque reduction experiments. The main components of Fr.8.3 were identified by using LC–MS, and selected protein targets of these components were investigated through molecular docking. Results We collected and isolated a variety of A. vulgaris L. samples from Tangyin County, Henan Province and then screened the A. vulgaris L. leaf extracts for anti-HSV-1 activity. The results of the plaque reduction test showed that the crude extract of A. vulgaris L.-Fr.8.3 had anti-HSV-1 activity, and we further verified the anti-HSV-1 activity of Fr.8.3 at the DNA, RNA and protein levels. Moreover, we found that Fr.8.3 also had a broad spectrum of antiviral activity. Finally, we explored its anti-HSV-1 mechanism, and the results showed that Fr.8.3 exerted an anti-HSV-1 effect by acting directly on the virus itself. Then, the extracts were screened on HSV-1 surface glycoproteins and host cell surface receptors for potential binding ability by molecular docking, which further verified the phenotypic results. LC–MS analysis showed that 1 and 2 were the two main components of the extracts. Docking analysis suggested that compounds from extract 1 might similarly cover the binding domain between the virus and the host cells, thus interfering with virus adhesion to cell receptors, which provides new ideas and insights for clinical drug development for herpes simplex virus type 1. Conclusion We found that Fr.8.3 has anti-herpesvirus and anti-rotavirus effects. The main 12 components in Fr.8.3 were analyzed by LC–MS, and the protein targets were finally predicted through molecular docking, which showed that alkaloids may play a major role in antiviral activity.

Deep image prior is a classical unsupervised deep learning method that does not require plenty of training samples, because in some practical applications, like medical imaging, collecting tons of training samples is not always viable. At the same time, it is hard and time-consuming to construct a favorable training set, since it requires that the selected data must be sufficient and typical. In addition, due to the overfitting problem of the deep image prior method, it can lead to the loss of image texture details and the destruction of edge information. And the second-order total generalized variation can effectively protect the image edge information. Therefore, in this work, an image denoising method that combines a deep image prior with the second-order total generalized variation is proposed to remove noise, preserve the image edge and texture structuration a better way. To solve the new model efficiently, first, we transform the constrained problem into an unconstrained problem using the augmented Lagrangian method, and then solve the augmented Lagrangian function using the alternating direction method of multiplication. Experimental results show that the proposed method can removes noise more effectively, retain more image details, and obtain higher performance in terms of peak signal-to-noise ratio and structural similarity, and outperform other existing methods.

Abstract Hyperuricemia (HUA) seriously harms human health but the exact etiology and pathogenesis of HUA are not fully understood. Therefore, it is still of great significance to find effective biomarkers and explore the pathogenesis of HUA. Metabolomics reflects the influence of internal and external factors on system metabolism, explains the changes in metabolite levels during the development of diseases, and reveals the molecular mechanism of pathogenesis. Metabolomics is divided into untargeted metabolomics and targeted metabolomics according to different research modes. Each other's advantages can be fully utilized by combining the two so that the results of metabolomics research can be consummated. 20 HUA patients and 20 healthy individuals participated in the experiment, and untargeted metabolomics was employed to find 50 differential metabolites in HUA serum samples. Twelve candidate biomarkers were screened based on literature research and ROC Curve analysis for subsequent verification. Based on the UPLC-TQ-MS analysis platform, the targeted metabolomics detection methods were established and the content of 12 candidate biomarkers was precisely quantified. Compare with the results of untargeted metabolomics, the targeted metabolomics results were considered more reliable.

Abstract Three-dimensional (3D) ordered porous carbon is generally believed to be a promising electromagnetic wave (EMW) absorbing material. However, most research works targeted performance improvement of 3D ordered porous carbon, and the specific attenuation mechanism is still ambiguous. Therefore, in this work, a novel ultra-light egg-derived porous carbon foam (EDCF) structure has been successfully constructed by a simple carbonization combined with the silica microsphere template-etching process. Based on an equivalent substitute strategy, the influence of pore volume and specific surface area on the electromagnetic parameters and EMW absorption properties of the EDCF products was confirmed respectively by adjusting the addition content and diameter of silica microspheres. As a primary attenuation mode, the dielectric loss originates from the comprehensive effect of conduction loss and polarization loss in S-band and C band, and the value is dominated by polarization loss in X band and Ku band, which is obviously greater than that of conduction loss. Furthermore, in all samples, the largest effective absorption bandwidth of EDCF-3 is 7.12 GHz under the thickness of 2.13 mm with the filling content of approximately 5 wt%, covering the whole Ku band. Meanwhile, the EDCF-7 sample with optimized pore volume and specific surface area achieves minimum reflection loss (RLmin) of − 58.08 dB at 16.86 GHz while the thickness is 1.27 mm. The outstanding research results not only provide a novel insight into enhancement of EMW absorption properties but also clarify the dominant dissipation mechanism for the porous carbon-based absorber from the perspective of objective experiments.

Natural images generally contain both structure component and texture component. The existing image segmentation models based on piecewise smooth cannot handle such natural images containing texture well. For this reason, in this paper, a multi-region image segmentation model based on low-rank prior decomposition is proposed. We use a low-rank prior to characterize the texture component, and the total variational norm to characterize the piecewise smooth structure component so that the proposed method can perform image decomposition and image segmentation tasks simultaneously. Then, the decomposed structure image can be used for image segmentation, and thus we can improve the accuracy of segmentation. To solve the novel model, the alternating direction multiplier method is designed. The experimental results show that compared with the related classical models, the new model can significantly improve the subjective visual effect of image segmentation and the mean values of the precision rate, F1-measure, and Jaccard similarity coefficient for the new model on the test images are at least 3.29%, 1.74%, and 3.13% higher, respectively.

Abstract Background Colorectal cancer (CRC) is one of the most common malignant tumours. The recurrence and metastasis of CRC seriously affect the survival rate of patients. Angiogenesis is an extremely important cause of tumour growth and metastasis. Circular RNAs (circRNAs) have been emerged as vital regulators for tumour progression. However, the regulatory role, clinical significance and underlying mechanisms still remain largely unknown. Methods High-throughput sequencing was used to analyse differential circRNAs expression in tumour and non-tumour tissues of CRC. In situ hybridization (ISH) and qRT-PCR were used to determine the level of circ3823 in CRC tissues and serum samples. Then, functional experiments in vitro and in vivo were performed to investigate the effects of circ3823 on tumour growth, metastasis and angiogenesis in CRC. Sanger sequencing, RNase R and Actinomycin D assay were used to verify the ring structure of circ3823. Mechanistically, dual luciferase reporter assay, fluorescent in situ hybridization (FISH), RNA immunoprecipitation (RIP) and RNA pull-down experiments were performed to confirm the underlying mechanisms of circ3823. Results Circ3823 was evidently highly expressed in CRC and high circ3823 expression predicted a worse prognosis of CRC patients. Receiver operating characteristic curves (ROCs) indicated that the expression of circ3823 in serum showed high sensitivity and specificity for detecting CRC which means circ3823 have the potential to be used as diagnostic biomarkers. Functional experiments in vitro and in vivo indicated that circ3823 promote CRC cell proliferation, metastasis and angiogenesis. Mechanism analysis showed that circ3823 act as a competing endogenous RNA of miR-30c-5p to relieve the repressive effect of miR-30c-5p on its target TCF7 which upregulates MYC and CCND1, and finally facilitates CRC progression. In addition, we found that N6-methyladenosine (m6A) modification exists on circ3823. And the m6A modification is involved in regulating the degradation of circ3823. Conclusions Our findings suggest that circ3823 promotes CRC growth, metastasis and angiogenesis through circ3823/miR-30c-5p/TCF7 axis and it may serve as a new diagnostic marker or target for treatment of CRC patients. In addition, m6A modification is involved in regulating the degradation of circ3823.

Abstract Background Plantaginis Semen has been widely used as folk medicine and health care food against hyperuricemia (HUA) and gout, but its pharmacological mechanism remains unclear. This study investigated the therapeutic mechanism of Plantaginis Semen extract on potassium oxonate -induced HUA rats based on a lipidomics approach. Methods A model of HUA was established by potassium oxonate intragastric administration. 42 Sprague-Dawley (SD) male rats were randomly divided into the control group, model group, benzbromarone group (10 mg/kg) and three Plantaginis Semen groups (n = 7). The Plantaginis Semen groups were treated orally with Plantaginis Semen, 0.9375, 1.875 or 3.75 g/kg for 28 days. The levels of serum uric acid (UA), creatinine (Cr), triacylglycerol (TG) and tumor necrosis factor-α (TNF-α) were measured using enzyme-linked immunosorbent assay kits. Ultra performance liquid chromatography quadrupole time of flight mass spectrometry (UPLC-Q-TOF/MS) was used for the serum lipidomics analysis, multivariate statistical analysis and independent samples t-test were carried out for the pattern recognition and characteristic metabolites identification. The relative levels of critical regulatory factors were determined by quantitative real-time polymerase chain reaction (RT-qPCR). Image inpainting is a crucial task in computer vision that aims to restore missing and occluded parts of damaged images. Deep-learning-based image inpainting methods have gained popularity in recent research. One such method is the deep image prior, which is unsupervised and does not require a large number of training samples. However, the deep image prior method often encounters overfitting problems, resulting in blurred image edges. In contrast, the second-order total generalized variation can effectively protect the image edge information. In this paper, we propose a novel image restoration model that combines the strengths of both the deep image prior and the second-order total generalized variation. Our model aims to better preserve the edges of the image structure. To effectively solve the optimization problem, we employ the augmented Lagrangian method and the alternating direction method of the multiplier. Numerical experiments show that the proposed method can repair images more effectively, retain more image details, and achieve higher performance than some recent methods in terms of peak signal-to-noise ratio and structural similarity.

In this paper, we propose a curvature-guided model with divergence-free constraints to facilitate image decomposition. Since basic TV regularization has difficulty in processing edge geometry information of cartoon image, in order to preserve the edge features, we introduce a level set curvature term to smooth the uniform area, and use the edge indicator function as the weighted regularization to preserve the edge. In addition, to control the smoothness, a gradient function is proposed to balance the edge indicator function and the curvature term. On the other hand, we find that the existing partial decomposition models measure the oscillation function with the $H^{-1}$ functional, but this functional ignores the divergence-free vector field during the Hodge decomposition process, then the texture will lose some vector direction information, and then affects the decomposition results. By analyzing the theory of divergence-free vector field, a new decomposition model with the constraint of divergence-free vector field is proposed in this paper. Numerical experiments show that the proposed model can well preserves the edges of cartoon and protects the texture of repetitive patterns.

Use of chimeric antigen receptors (CARs), as an immune cell therapy, has generated excellent clinical outcomes against hematologic tumors in recent years. Among them, the CAR-NK (natural killer) therapy has shown better efficacy, and less toxicity, than chimeric antigen receptor T-cell (CAR-T) therapy. In our phase II clinical trials, administering chimeric costimulatory converting receptor (CCCR)-NK92 cells on advanced non-small cell lung cancer patients proved efficacious in cell and animal experiments. However, we observed occurrence of cytokine release syndrome (CRS), a rare and unexpected side effect, never reported before during CAR-NK therapy. Here, we provide a detailed report of the patient’s case, emphasize on the need to pay attention to CRS in NK cell therapy, and suggest improvements that will minimize potential toxicity.

Hyperoside is a natural flavonol glycoside, which has antioxidation, antitumor, and anticancer activities together with other healthy effects like improving cardiovascular function, protecting the liver, and regulating the immune system. It is a popular compound used in the traditional Chinese medicine and different studies on hyperoside are present in the literature. However, studies on the metabolism of hyperoside in vivo were not comprehensive. In this study, UPLC-Q-Exactive Orbitrap MS technology was used to establish a rapid and comprehensive analysis strategy to explore the metabolites and metabolic process of hyperoside in rats. The metabolites of hyperoside were systematically identified in rat plasma, urine, and feces. According to the hyperoside standard substance and relevant works of literature, a total of 33 metabolites were identified, including 16 in plasma, 31 in urine, and 14 in feces. Among them, the metabolites quercetin and dihydroquercetin were unambiguously confirmed by comparison with standard substances. In addition, 13 metabolites had not been reported in hyperoside metabolism-related articles at present. The metabolic reactions of hyperoside in vivo were further explored, including phase I metabolism (hydroxylation, dehydroxylation, glycoside hydrolysis, hydrogenation, and hydration) and phase II metabolism (methylation, acetylation, sulfation, and glucuronide conjugation). The fragment ions of hyperoside and its metabolites were usually produced by glucoside bond hydrolysis, the neutral loss of (CO + OH), COH, CO, O, and Retro-Diels Alder (RDA) cleavage. In conclusion, this study comprehensively characterized the metabolism of hyperoside in rats, providing a basis for exploring its various biological activities.

Cancer drug resistance has always been a major difficulty in cancer therapy. In the face of drug pressure, resistant cancer cells show complex molecular mechanisms including epigenetic changes to maintain survival. Studies prove that cancer cells exhibit abnormal m6A modification after acquiring drug resistance. m6A modification in the target RNA including non-coding RNA can be a controller to determine the fate and metabolism of RNA by regulating their stability, subcellular localization, or translation. In particular, m6A-modified non-coding RNA plays multiple roles in multiple drug-resistant cancer cells, which can be a target for cancer drug resistance. Here, we provide an overview of the complex regulatory mechanisms of m6A-modified non-coding RNA in cancer drug resistance, and we discuss its potential value and challenges in clinical applications.

Artemisia argyi H. Lév. and Vaniot is a traditional medical herb that has been used for a long time in China and other Asian counties. Essential oil is the main active fraction of Artemisia argyi H. Lév. and Vaniot, and its anti-inflammatory potential has been observed in vitro and in vivo. Here, we found that the essential oil of Artemisia argyi H. Lév. and Vaniot (EOAA) inhibited monosodium urate (MSU)- and nigericin-induced NLRP3 inflammasome activation. EOAA suppressed caspase-1 and IL-1β processing and pyroptosis. NF-κB p65 phosphorylation and translocation were also inhibited. In addition, EOAA suppressed nigericin-induced NLRP3 inflammasome activation without blocking ASC oligomerization, suggesting that it may inhibit NLRP3 inflammasome activation by preventing caspase-1 processing. Our study thus indicates that EOAA inhibits NLRP3 inflammasome activation and has therapeutic potential against NLRP3-driven diseases.

PMP-HPLC, FT-IR, and HPSEC fingerprints of 10 batches of polysaccharides from Saposhnikoviae Radix with different production areas and harvest times have been prepared, and the chemometrics analysis was performed. The anti-allergic activity of 10 batches of Saposhnikoviae Radix polysaccharide (SP) was evaluated, and the spectrum-effect relationship of the 10 batches of SP was analyzed by gray correlation degree with the chromatographic fingerprint as the independent variable. The results showed that the PMP-HPLC, HPSEC, and FT-IR fingerprints of 10 batches of SP had a high similarity. Two monosaccharides (rhamnose and galactose), the polysaccharide fragment Mn = 8.67 × 106~9.56 × 106 Da, and the FT-IR absorption peak of 892 cm−1 can be used as the quality control markers of SPs. All 10 batches of SP could significantly inhibit the release of β-HEX in RBL-231 cells, and the polysaccharides harvested from Inner Mongolia in the winter had the best anti-allergic activity. The spectrum-effect relationship model showed that the monosaccharide composition and molecular weight were related to the anti-allergic activity of the SPs. Multiple fingerprints combined with spectrum-effect relationship analysis can evaluate and control the quality of SPs from the aspects of overall quality and efficacy, which has more application value.

In order to better decompose the images and protect their edges, in this paper, we proposed a coupled variational system consisting of two steps. The first step, an improved weighted variational model is introduced to obtain the cartoon and texture. Using the obtained cartoon image, in the second step, a new vector function is obtained for describing the pseudo edge of the considered image by one Tikhonov regularization variational model. Because Tikhonov regularization model is equivalent to carrying out a Gaussian linear filtering, the obtained vector function is smoother. To solve the coupled system, we give the alternating direction method, primal-dual method and Gauss-Seidel iteration. Using the coupled system, we can not only separate out the cartoon and texture parts, but also extract the edge. Extensive numerical experiments are given to show the effectiveness of the proposed method compared with other variational methods.

Abstract The major components of vitelline membrane (VM) are ovomucin, VM outer (VMO) I and VMO II. At present, the distribution pattern of maternal cells on the VM has not been described in detail. In this study, the existence and distribution characteristics of maternal cells on VM were observed. There were more than 3.2 × 105 somatic cells on VM, which were uneven distributed. The calcein AM/PI staining of the maternal cells on the VM showed that the cells’ viability changed with the freshness of the eggs, and that the maternal cells gradually underwent apoptosis and became degraded. The results of morphology of different tissues indicated that the most of maternal cells on the VM were granulosa cells. Moreover, the karyotype of the cultured granulosa cells, which is the main source of cells on VM, were identified as the normal diploid karyotype of chicken. Furthermore, the VM DNA extracted from chickens and quails, which represent the eggs of different size, was adequate for further genetic analysis. The VM DNA was easily accessible and relatively constant, without cross-contamination. Therefore, the VM DNA could potentially be applied for the molecular traceability between eggs and chickens, and be beneficial in avian ecology research studies.

Objective: Our study aimed to investigate the underlying molecular mechanisms of selin-11-en-4α-ol purified from Artemisia vulgaris against anti-inflammation through network pharmacology, molecular docking, and in vitro experiments. Methods: Potential targets for selin-11-en-4α-ol and inflammation were obtained using Swiss Target Prediction, Pharm Mapper, and GeneCards databases. Venn diagrams were used to obtain the targets of selin-11-en-4α-ol for the treatment of inflammation, and the intersecting targets were uploaded to the STRING database to construct a protein-protein interaction (PPI) network of selin-11-en-4α-ol anti-inflammatory target proteins for further screening of the core targets. The Gene Ontology and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were conducted using the DAVID database. In addition, molecular docking validation of core target binding to selin-11-en-4α-ol was performed. The anti-inflammatory effects were verified by in vitro experiments of lipopolysaccharide-induced RAW264.7 cells. Results: PPI network analysis showed that mitogen-activated protein kinase 14 (MAPK14), SRC, and HSP90AA1 were predicted as potential anti-inflammatory targets, and KEGG enrichment analysis revealed that the MAPK signaling pathway presented the highest gene enrichment counts. Molecular docking and western blot results suggested that selin-11-en-4α-ol regulated MAPK and nuclear transcription factor-κB (NF-κB) signaling pathways to alleviate inflammation.
