Groundwater is an important water supply source in Xining City. Previous works have found that iron-rich groundwater limits water development and utilization. However, few studies focus on the distribution characteristics and mechanism of iron-rich groundwater in different types of aquifers in Xining City. Based on the hydrochemical data of 144 groundwater samples, combined with the geological and hydrogeological conditions, the distribution characteristics and influencing factors of iron content in groundwater in different aquifers were analyzed by methods of statistics and geostatistics. The results show that the exceeding standard ratio of iron in groundwater of unconsolidated sediments aquifer (Area Ⅰ) is 27. 85%, which is 1. 6 times and 2. 7 times higher than that of clastic rocks fissure-pore aquifer (Area Ⅱ) and bedrock aquifer (Area Ⅲ), respectively. The proportion of iron-rich groundwater (concentration greater than 0. 3 mg/L) in construction land area is significantly higher than in areas with other land use types. The redox environment and human activities (such as industrial wastewater, domestic sewage, and infiltration of iron-rich river water) may be the main factors affecting the increase of iron content in groundwater in Area I. In addition to the redox environment, Agricultural nitrogen fertilizer use and domestic sewage discharge are also important factors affecting iron-rich groundwater in Area Ⅱ. The iron-rich groundwater in Area Ⅲ is mainly controlled by redox conditions. On the regional scale, the distribution of iron-rich groundwater presents in bands or spots along the river. The iron deposits releasing in the primary strata and human activities (industrial wastewater) are the main source of iron ions in the shallow groundwater in the study area. The migration and enrichment of iron in groundwater are mainly controlled by redox conditions, and affected by the runoff condition, without relation to pH and salt effect. This study can provide scientific basis for urban water environment management in Xining City and similar arid and semi-arid areas.

Highlights Ag nanoparticles are evenly plated on BN-embedded fibers as a thermally conductive sheath, bridging the insulating interface between fibers to construct a 3D heat transfer network. The LM-printed e-textile shows outstanding capability for the monitoring of human ECG, sEMG, and EEG signals even under intense EM interference, when commercial electrodes cannot work properly. The e-textile simultaneously manifests excellent EMI shielding (SET > 65, X-band) and UV protection (UPF = 143.1) performance, thus protecting the device and skin from harmful radiation.

Abstract Fullerene derivatives are highly attractive materials in solar cells, organic thermoelectrics, and other devices. However, the intrinsic low electron mobility and electrical conductivity restrict their potential device performance, such as perovskite solar cells (PSCs). Herein, we successfully enhanced the electric properties and morphology of phenyl‐C61‐butyric acid methyl ester (PCBM) by n‐doping it with a benzimidazoline derivative, 9‐(1,3‐dimethyl‐2,3‐dihydro‐1H‐benzoimidazol‐2‐yl)‐julolidine (JLBI‐H) via a solution process. We found the n‐doping can not only improve the conductivity and optimize the band alignment but also enable the PCBM to have a constantly strong charge extraction ability in a wide temperature from 173 to 373 K, which guarantees a stable photovoltaic performance of the corresponding PSCs under a wide range of operating temperatures. With the JLBI‐H‐doped PCBM, we improved the efficiency from 17.9% to 19.8%, along with enhanced stability of the nonencapsulated devices following the aging protocol of ISOS‐D‐1.

Abstract Immune checkpoint blockade (ICB) has shown considerable promise for treating various malignancies, but only a subset of cancer patients benefit from immune checkpoint inhibitor therapy because of immune evasion and immune-related adverse events (irAEs). The mechanisms underlying how tumor cells regulate immune cell response remain largely unknown. Here we show that hexokinase domain component 1 (HKDC1) promotes tumor immune evasion in a CD8+ T cell-dependent manner by activating STAT1/PD-L1 in tumor cells. Mechanistically, HKDC1 binds to and presents cytosolic STAT1 to IFNGR1 on the plasma membrane following IFNγ-stimulation by associating with cytoskeleton protein ACTA2, resulting in STAT1 phosphorylation and nuclear translocation. HKDC1 inhibition in combination with anti-PD-1/PD-L1 enhances in vivo T cell antitumor response in liver cancer models in male mice. Clinical sample analysis indicates a correlation among HKDC1 expression, STAT1 phosphorylation, and survival in patients with hepatocellular carcinoma treated with atezolizumab (anti-PD-L1). These findings reveal a role for HKDC1 in regulating immune evasion by coupling cytoskeleton with STAT1 activation, providing a potential combination strategy to enhance antitumor immune responses.

Abstract Background The pathophysiological mechanisms of diabetic retinopathy (DR), a blinding disease, are intricate. DR was thought to be a microvascular disease previously. However, growing studies have indicated that the retinal microglia-induced inflammation precedes microangiopathy. The binary concept of microglial M1/M2 polarization paradigms during inflammatory activation has been debated. In this study, we confirmed microglia had the most significant changes in early DR using single-cell RNA sequencing. Methods A total of five retinal specimens were collected from donor SD rats. Changes in various cells of the retina at the early stage of DR were analyzed using single-cell sequencing technology. Results We defined three new microglial subtypes at cellular level, including two M1 types (Egr2 + M1 and Egr2 − M1) and one M2 type. We also revealed the anatomical location between these subtypes, the dynamic changes of polarization phenotypes, and the possible activation sequence and mutual activation regulatory mechanism of different cells. Furthermore, we constructed an inflammatory network involving microglia, blood-derived macrophages and other retinal nonneuronal cells. The targeted study of new disease-specific microglial subtypes can shorten the time for drug screening and clinical application, which provided insight for the early control and reversal of DR. Conclusions We found that microglia show the most obvious differential expression changes in early DR and reveal the changes in microglia in a high-glucose microenvironment at the single-cell level. Our comprehensive analysis will help achieve early reversal and control the occurrence and progression of DR.

Tree peony (Paeonia suffruticosa Andrews) is a well-known ornamental plant with high economic value, but the short fluorescence is a key obstacle to its ornamental value and industry development. High temperature accelerates flower senescence and abscission, but the associated mechanisms are poorly understood. In this study, the tandem mass tag (TMT) proteome and label-free quantitative ubiquitome from tree peony cut flowers treated with 20 °C for 0 h (RT0), 20 °C or 28 °C for 60 h (RT60 or HT60) were examined based on morphological observation, respectively. Totally, 6970 proteins and 1545 lysine ubiquitinated (Kub) sites in 844 proteins were identified. Hydrophilic residues (such as glutamate and aspartate) neighboring the Kub sites were in preference, and 36.01% of the Kub sites were located on the protein surface. The differentially expressed proteins (DEPs) and Kub-DEPs in HT60 vs RT60 were mainly enriched in ribosomal protein, protein biosynthesis, secondary metabolites biosynthesis, flavonoid metabolism, carbohydrate catabolism, and auxin biosynthesis and signaling revealed by GO and KEGG analysis, accompanying the increase of endogenous abscisic acid (ABA) accumulation and decrease of endogenous indoleacetic acid (IAA) level. Additionally, the expression patterns of six enzymes (SAMS, ACO, YUC, CHS, ANS and PFK) putatively with Kub modifications were analyzed by proteome and real-time quantitative RT-PCR. The cell-free degradation assays showed PsSAMS and PsACO proteins could be degraded via the 26 S proteasome system in tree peony flowers. Finally, a working model was proposed for the acceleration of flower senescence and abscission by high temperature. In summary, all results contributed to understanding the mechanism of flower senescence induced by high temperature and prolonging fluorescence in tree peony.

Silicosis is a disease characterized by lung inflammation and fibrosis caused by long-term inhalation of free silicon dioxide (SiO2). Recent studies have found that a large number of lymphatic hyperplasia occurs during the occurrence and development of silicosis. miRNAs play an important role in lymphangiogenesis. However, the regulation and mechanism of miRNAs on lymphangiogenesis in silicosis remain unclear. In this study, lymphangiogenesis was observed in silicosis rats, and VEGF-C-targeted miRNAs were screened, and the effect of miRNAs on the formation of human lymphatic endothelial cells (HLECs) tubular structure was investigated in vitro. The results showed that SiO2 promoted the expressions of Collagen Ι and α-SMA, TNF-α, IL-6 and VEGF-C increased first and then decreased, and promoted the formation of lymphatic vessels. Bioinformatics methods screened miR-455–3p for targeted binding to VEGF-C, and dual luciferase reporter genes confirmed VEGF-C as the target gene of miR-455–3p, and miR-455–3p was down-regulated in the lung tissue of silicosis rats. Transfection of miR-455–3p Inhibitors down-regulated the expression level of miR-455–3p and up-regulated the expression levels of VEGF-C and VEGFR-3 in HLECs, enhanced migration ability and increased tube formation. Transfection of miR-455–3p Mimics showed an opposite trend. These results suggest that miR-455–3p further regulates the tubular structure formation of HLECs by regulating VEGF-C/VEGFR3. Therefore, targeting miR-455–3p may provide a new therapeutic strategy for SiO2-induced silicosis injury.

PIN-FORMED (PIN) proteins, which function as efflux transporters, play many crucial roles in the polar transportation of auxin within plants. In this study, the exogenous applications of auxin IAA and TIBA were found to significantly prolong and shorten the florescence of tree peony (Paeonia suffruticosa Andr.) flowers. This finding suggests that auxin has some regulatory influence in petal senescence and abscission. Further analysis revealed a total of 8 PsPINs distributed across three chromosomes, which could be categorized into two classes based on phylogenetic and structural analysis. PsPIN1, PsPIN2a-b, and PsPIN4 were separated into the “long” PIN category, while PsPIN5, PsPIN6a-b, and PsPIN8 belonged to the “short” one. Additionally, the cis-regulatory elements of PsPIN promoters were associated with plant development, phytohormones, and environmental stress. These genes displayed tissue-specific expression, and phosphorylation sites were abundant throughout the protein family. Notably, PsPIN4 displayed distinct and elevated expression levels in roots, leaves, and flower organs. Expression patterns among the abscission zone (AZ) and adjacent areas during various flowering stages and IAA treatment indicate that PsPIN4 likely influences the initiation of peony petal abscission. The PsPIN4 protein was observed to be co-localized on both the plasma membrane and the cell nucleus. The ectopic expression of PsPIN4 reversed the premature flower organs abscission in the Atpin4 and significantly protracted florescence when introduced to Col Arabidopsis. Background: Family involvement and comfort are equally important in palliative care. Dignity undertook a new meaning and novel challenges as a result of restrictions on visits and companionship during the pandemic. Family-centered family dignity interventions have been shown to be effective in increasing patients’ sense of dignity, increasing levels of hope, and reducing psychological distress; however, the effectiveness in enhancing family adaptability and intimacy in the survivor-caregiver binary and reducing expected grief have been inconclusive. Objectives: The primary objective of this study was to assess the efficacy of family dignity interventions on family adaptability and cohesion. The secondary objective was to explore the effects of the interventions on anticipatory grief and psychological distress, and the lasting effect 1 month after the intervention. Design: A single-blinded, two-arm parallel group, randomized controlled trial was conducted in China. Settings: and methods: Ninety-eight dyads who met the inclusion criteria were randomly assigned to the family dignity intervention (n = 51) or standard palliative care group (n = 47) between June and August 2022. Study outcomes were measured at baseline, immediately post-intervention, and at the 1-month follow-up post-intervention evaluation. Data were analyzed using the Kolmogorov-Smirnov test, chi-square test, Fisher's exact test, independent sample t-test, Wilcoxon rank-sum test, and generalized estimation equations. The Intention-To-Treat analysis was performed for all available data. Results: In comparison to the control group, significant improvements in family adaptability and cohesion and anticipatory grief over post-intervention and 1-month follow-up were demonstrated among the patients in the intervention group. Background: Given that the circadian rhythm is intricately linked to cardiovascular physiological functions, the objective of this investigation was to employ bibliometric visualization analysis in order to scrutinize the trends, hotspots, and prospects of the circadian rhythm and cardiovascular disease (CVD) over the past two decades. Methods: A thorough exploration of the literature related to the circadian rhythm and CVD was conducted via the Web of Science Core Collection database spanning the years 2002–2022. Advanced software tools, including citespace and VOSviewer, were employed to carry out a comprehensive analysis of the co-occurrence and collaborative relationships among countries, institutions, journals, references, and keywords found in this literature. Furthermore, correlation mapping was executed to provide a visual representation of the data. Results: The present study encompassed a total of 3399 published works, comprising of 2691 articles and 708 reviews. The publications under scrutiny were primarily derived from countries such as the United States, Japan, and China. The most prominent research institutions were found to be the University of Vigo, University of Minnesota, and Harvard University. Notably, the journal Chronobiology International, alongside its co-cited publications, had the most substantial contribution to the research in this field. Following an exhaustive analysis, the most frequently observed keywords were identified as circadian rhythm, blood pressure, hypertension, heart rate, heart rate variability, and melatonin. Furthermore, a nascent analysis indicated that future research might gravitate towards topics such as inflammation, metabolism, oxidative stress, and autophagy, thereby indicating new directions for investigation. Conclusion: This analysis represents the first instance of bibliometric scrutiny pertaining to circadian rhythm and its correlation with cardiovascular disease (CVD) through the use of visualization software. Notably, this study has succeeded in highlighting the recent research frontiers and prominent trajectories in this field, thereby providing a valuable contribution to the literature.

The supra-permafrost water is a vital water source to support the ecosystem and an important link in maintaining the hydrothermal cycle in the permafrost area. Under the intensification of climate change, scientific understanding of the effect of permafrost degradation on the supra-permafrost water is of great significance to water resources and ecological protection. Focusing on the hydrological effects of permafrost degradation in the source areas of the Yellow River, this study analyzed the degradation characteristics of permafrost, and revealed the response of the depth of groundwater level and recharge process of the supra-permafrost water to the permafrost degradation, on the basis of the temperature and moisture content of frozen soil at typical monitoring points and the runoff change at the Huangheyan Hydrologic Station. The results show that the average temperature increased 0.42 °C at the 0−2.4 m profile of the monitoring point from 2010 to 2020. The depth of the upper interface of the permafrost reduced from 2.1m to 2.5 m, with an average decrease rate of 4cm/a. After 2018, the depth of supra-permafrost water level reduced from less than 0.9 m to 0.9−1.8 m. The permafros degradation led to the runoff process in the melting period of active layer (May−October) being advanced, the ratio of extreme value being reduced，and the runoff hydrograph in January being more prominent. Ground temperature is the dominant factor in controlling the changes of depth of the supra-permafrost water. Under the condition of warm and humid climate change, Permafrost degradation would change the dynamic characteristics of the water above the frozen layer and its hydraulic relationship with surface water, affecting the hydrological and ecological processes in the source area of the Yellow River.

Abstract Resveratrol, curcumin, and quercetin are the secondary metabolites from medicinal food homology plants, that have been proven their potency in cancer treatment. However, the antitumor effect of a single component is weak. So, herein, we designed an antitumor compound named RCQ composed of resveratrol, curcumin, and quercetin. This study examined the effect on tumorigenesis and development of 4T1 breast cancer-bearing mice following administering RCQ by intragastric administration. RCQ increased the recruitment of T cells and reduced the accumulation of neutrophils and macrophages in the tumor microenvironment. Meanwhile, RCQ suppressed the development of tumor-infiltrating lymphocytes into immunosuppressive cell subpopulations, including CD4+ T cells to T helper Type 2 type (Th2), tumor-associated neutrophils (TANs) to the N2 TANs, and tumor-associated macrophages (TAMs) cells to M2 TAMs. RCQ reversed the predominance of immunosuppressive infiltrating cells in the tumor microenvironment and tipped the immune balance toward an immune activation state. In vitro the study showed that RCQ significantly increased reactive oxygen species (ROS), reduce mitochondrial membrane potentials in cancer cells, and modulate pro-apoptotic Bcl-2 family members. In conclusion, RCQ can promote the ROS apoptosis mechanism of tumor cells and alleviate immunosuppression of the tumor microenvironment to enhance the anti-tumor effect.

Highlights Efficient sunlight reflectivity and high mid-infrared radiation emissivity are simultaneously realized in a nonwoven metafabric via PTFE microparticle impregnation and thermal-fusion. The metafabric achieves a maximum cooling effect of 17 °C and fully retains its passive cooling performance even under 50% stretching. High-quality electrophysiological monitoring of ECG, sEMG and EEG is realized through compact and homogeneous encapsulation of liquid metal on the elastomeric fibers.

Abstract Genetic compensation responses (GCRs) can be induced by deleterious mutations in living organisms in order to maintain genetic robustness. One type of GCRs, homology-dependent GCR (HDGCR), involves transcriptional activation of one or more homologous genes related to the mutated gene. In zebrafish, ~80% of the genetic mutants produced by gene editing technology failed to show obvious phenotypes. The HDGCR has been proposed to be one of the main reasons for this phenomenon. It is triggered by mutant mRNA bearing a premature termination codon and has been suggested to depend on components of both the nonsense mRNA-mediated degradation (NMD) pathway and the complex of proteins associated with Set1 (COMPASS). However, exactly which specific NMD factor is required for HDGCR remains disputed. Here, zebrafish leg1 deleterious mutants are adopted as a model to distinguish the role of the NMD factors Upf1 and Upf3a in HDGCR. Four single mutant lines and three double mutant lines were produced. The RNA-seq data from 71 samples and the ULI-NChIP-seq data from 8 samples were then analyzed to study the HDGCR in leg1 mutants. Our results provide strong evidence that Upf3a, but not Upf1, is essential for the HDGCR induced by nonsense mutations in leg1 genes where H3K4me3 enrichment appears not to be a prerequisite. We also show that Upf3a is responsible for correcting the expression of hundreds of genes that would otherwise be dysregulated in the leg1 deleterious mutant.

Abstract The ICT service industry has become a burgeoning industry at a high and stable speed. Their equitable distribution can improve national and global positive peace. This paper aimed to verify the characteristics of spatio-temporal evolution and its influencing factors in the ICT service industry. Based on the data from 31 Provinces in China from 2015 to 2019, this paper uses location quotient, spatial autocorrelation methods and spatial econometric analysis to explore the development characteristics, evolution and influencing factors of the ICT service industry, respectively. The main results are shown as follows: (1) China's ICT service industry is mainly concentrated in Beijing, Shanghai, Zhejiang, Tibet, and Guangdong, with a trend of specialisation development. They are not only distributed in cities with relatively superior overall development but also those with superior industrial and development carrier elements. Technological relevance, aggregation, and political difference might have an impact on promoting the emergence and development of these industries. (2) ICT service industry is characterised by stable and highly concentrated development. Numbers between three to five significant provinces and types with high-high (HH) and high-low (HL) clusters of local spatio-temporal association kept stable in the period. The HH was in eastern coastal areas, including Zhejiang, Shanghai, Jiangsu, and Shandong, and the HL was in Guangdong in 2015. There is a definite spatial correlation in spatial distribution with constant strengthening. (3) TUR, NDN, MIAT and the area were shown to have a significant role in promoting the ICT service industry, while NW, GDP and ICT Employment were shown to have a significant negative impact on this industry. Correspondingly, two strategies were put forward here: (1) accelerating the inter-provincial networking development of the ICT service industry, and (2) strengthening government policy guidance for the ICT service industry. These outcomes can not only provide a scientific basis and theoretical support for the distribution of strategies and resources for these industries at the theoretical level but also improve resource integration from the national perspective and the efficiency of resource use at the practical level.

In remote sensing image (RSI) object detection, the oriented bounding box (OBB) can accurately locate objects with arbitrary orientation and obtain orientation information. The detection based on OBB is still a challenging task. In RSI, the distribution of objects is extremely uneven, which causes aggregation to occur. Some researchers believe that the characteristic of dense distribution is a reason for the difficulty of object detection. However, there are no in-depth experimental studies on this. This paper proposes an OBB-based dense object determination method, which determines the dense objects in datasets by two conditions consisting of interclass distance, intraclass distance, minimum distance between objects, and minimum edge length of objects. The experimental results of dense and non-dense object detection concludes that the characteristics of dense distribution in RSI do not easily cause the objects to be more difficult to detect. To make full use of the object features, we propose a second-stage detection head named RoIF-Net, in which we extract region of interest (RoI) from the input image and fuse it with the RoI extracted from feature maps to add detail features, and construct a feature induction module based on self-attention mechanism to achieve position regression and category classification. This structure can be used in any two-stage network to enhance detection capabilities. Using our method on three credible and challenging datasets, DOTA, DIOR-R, and UCAS-AOD, we obtained 81.80%, 68.49%, and 90.25% mAP, respectively, reaching SOTA based on OBB detection, proving the effectiveness and advancement of our method.

The growing number of renewable energy replacing conventional generators results in a loss of the reserve for frequency control in power systems, while many industrial power grids often have excess energy supply due to abundant wind and solar energy resources. This paper proposes a secondary frequency control (SFC) strategy that allows industrial power grids to provide emergency high-voltage direct current (HVDC) power support (EDCPS) for emergency to a system requiring power support through a voltage source converter (VSC) HVDC link. An architecture including multiple model predictive control (MPC) controllers with periodic communication is designed to simultaneously obtain optimized EDCPS capacity and minimize adverse effects on the providing power support (PPS) system. Moreover, a model of a virtual power plant (VPP) containing aluminum smelter loads (ASLs) and a high penetration of wind power is established for the PPS system. The flexibility and controllability of the VPP are improved by the demand response of the ASLs. The uncertainty associated with wind power is considered by chance constraints. The effectiveness of the proposed strategy is verified by simulation results using the data of an actual industrial power grid in Inner Mongolia, China. The DC voltage of the VSCs and the DC in the potlines of the ASLs are also investigated in the simulation.

The brief longevity of tree peony blossoms constrains its ornamental value and economic worth. Gibberellins (GAs) are crucial in the modulation of flower senescence, and GA 20-oxidase (GA20ox), GA 3-oxidase (GA3ox), and GA 2-oxidase (GA2ox) catalyze the synthesis and deactivation of bioactive GAs. In Paeonia ostii, a total of three PoGA20ox, ten PoGA3ox, and twelve PoGA2ox proteins were identified and comprehensively analyzed. The analysis of the gene structures, conserved domains, and motifs revealed structural similarities and variances among the GA20ox, GA3ox, GA2ox-A, and GA2ox-B subfamilies. The synteny analysis indicated a scarcity of collinear blocks within the P. ostii genome, with no tandem or whole-genome duplication/segmental duplications found in PoGAoxs. The investigation into the binding of transcription factors to PoGAox promoters and the assessments of the expression levels suggest that PoGA2ox1 and PoGA2ox8.1 are promising candidate genes implicated in the regulation of floral senescence. Further, Pos.gene61099 (BPC6) and Pos.gene61094 (CIL2) appear to modulate PoGA2ox1 transcription in a positive and negative manner, respectively, while Pos.gene38359 (DDF1) and Pos.gene17639 (DREB1C) likely enhance PoGA2ox8.1’s expression. This study lays a foundation for an in-depth understanding of PoGAox functions and the development of strategies to delay flower senescence in tree peony.

Introduction: Systemic lupus erythematosus (SLE) is an autoimmune disorder characterized by the production of autoantibodies, immune complex deposition, and tissue/organ damage. In this study, we aimed to identify molecular features and signaling pathways associated with SLE severity using RNA sequencing (RNA-seq), single-cell RNA sequencing (scRNA-seq), and clinical parameters.Methods: We analyzed transcriptome profiles of 45 SLE patients, grouped into mild (mSLE, SLEDAI ≤ 9) and severe (sSLE, SLEDAI > 9) based on SLE Disease Activity Index (SLEDAI) scores. We also collected clinical data on anti-dsDNA, ANA, ESR, CRP, snRNP, AHA, and anti-Smith antibody status for each patient.Results: By comparing gene expression across groups, we identified 12 differentially expressed genes (DEGs), including 7 upregulated (CEACAM6, UCHL1, ARFGEF3, AMPH, SERPINB10, TACSTD2, and OTX1) and 5 downregulated (SORBS2, TRIM64B, SORCS3, DRAXIN, and PCDHGA10) DEGs in sSLE compared to mSLE. Furthermore, using the CIBERSORT algorithm, we found that Treg cells were significantly decreased in sSLE and negatively correlated with AMPH expression, which was mainly expressed in Treg cells from SLE patients according to public scRNA-seq data (GSE135779).Discussion: Overall, our findings shed light on the molecular mechanisms underlying SLE severity and provide insight into potential therapeutic targets.

Myocardial ischemia-reperfusion injury is a common condition in cardiovascular diseases, and the mechanism of its occurrence involves multiple complex metabolic pathways and signaling pathways. Among these pathways, glucose metabolism and lipid metabolism play important roles in regulating myocardial energy metabolism. Therefore, this article focuses on the roles of glucose metabolism and lipid metabolism in myocardial ischemia-reperfusion injury, including glycolysis, glucose uptake and transport, glycogen metabolism and the pentose phosphate pathway; and triglyceride metabolism, fatty acid uptake and transport, phospholipid metabolism, lipoprotein metabolism, and cholesterol metabolism. Finally, due to the different alterations and development of glucose metabolism and lipid metabolism in myocardial ischemia-reperfusion, there are also complex interregulatory relationships between them. In the future, modulating the equilibrium between glucose metabolism and lipid metabolism in cardiomyocytes and ameliorating aberrations in myocardial energy metabolism represent highly promising novel strategies for addressing myocardial ischemia-reperfusion injury. Therefore, a comprehensive exploration of glycolipid metabolism can offer novel theoretical and clinical insights into the prevention and treatment of myocardial ischemia-reperfusion injury.

Abstract In this article, we solve the key positions prediction problem of engineering projects in smart grid, which pays more attention to the spatial-temporal distribution of projects. Many studies show that the projects are affected by multi-dimensional features such as time, space, correlation etc. However, few work can accurately predict the key positions of projects based on multi-dimensional features. In order to solve this problem, we propose the idea of multi-feature extraction, and make use of the real-world records trace to conduct multi-dimensional modeling. Then we introduce a multi-dimensional features extraction model: Multi-Feature-based GCN-LSTM (MF-GCN-LSTM) to take the effect of time, space and correlation for predicting the key positions of projects. Experiments on different datasets with various project types have proved that our model can complete the key positions prediction task efficiently. Compared with the other traditional method and non-linear models, our model shows higher prediction accuracy and robustness. Moreover, we show that the whole prediction framework MF-GCN-LSTM can be split and deployed in a distributed manner to accelerate the inference of the model under the cloud edge system.

Abstract To compare the survival benefit between preoperative and postoperative radiotherapy for stage I–III non-inflammatory breast cancer patients, we conducted a retrospective cohort study using surveillance, epidemiology and end results databases. Our study recruited patients who had been diagnosed with stage I–III breast cancer and underwent surgery and radiotherapy. The overall survival was calculated by Kaplan–Meier method. Cox risk model was used to determine the impact of radiotherapy according to stage, molecular subtype and other risk factors. Propensity score matching was used to balance measurable confounding factors. Of all the 411,279 enrolled patients varying from 1975 to 2016, 1712 patients received preoperative radiotherapy, and 409,567 patients received postoperative radiotherapy. Compared with the postoperative radiotherapy group, the preoperative radiotherapy group showed significantly higher risks of overall mortality and breast cancer-specific mortality. Survival differences in treatment sequences were correlated with stage, molecular subtypes and other risk factors. According to the results of this study, preoperative radiotherapy did not show a survival advantage, and postoperative radiotherapy is still the primary treatment. However, preoperative radiotherapy also has some theoretical advantages, such as phase reduction and recurrence reduction. Therefore, it is still worthy of further exploration.

In response to COVID-19, governments worldwide are implementing public health and social measures (PHSM) that substantially impact many areas beyond public health. The new field of PHSM data science collects, structures, and disseminates data on PHSM; here, we report the main achievements, challenges, and focus areas of this novel field of research.

Introduction: Aconite is a form of traditional Chinese medicine (TCM) that has been widely used to treat diarrhea for thousands of years. However, it is not clear whether the anti-diarrhea role of aconite aqueous extract (AA) is associated with regulation of the gut microbiota or with bile acid (BA) metabolism. This study aimed to confirm whether AA exerts its anti-diarrhea effects by regulating the gut microbiota and BA metabolism.Methods: The therapeutic effect of AA in a mouse model of diarrhea was measured based on analysis of body weight, fecal water content, diarrhea scores, intestinal propulsion rate, colonic pathology, and colonic immunohistochemistry. In addition, 16S rRNA high-throughput sequencing was conducted to analyze the effect of AA on the gut microbiota, and targeted metabolomics was employed to analyze the effect of AA on metabolism of BAs.Results: The results showed that treatment with AA reduced fecal water content and diarrhea scores, inhibited intestinal propulsion rate and pathological changes in the colon, and increased AQP3 and AQP4 content in the colon. In addition, AA was found to be capable of regulating the gut microbiota. Effects included increasing its richness (according to the ACE and Chao1 indices); altering the gut microbiota community structure (PCA, PCoA, and NMDS); increasing the relative abundance of norank_f_Muribaculaceae, Ruminococcus, Lachnospiraceae_NK4A136_group, Prevotellaceae_UCG-001, and norank_f_norank_o_Clostridia_UCG-014; and decreasing the relative abundance of Escherichia-Shigella, unclassified_f_Ruminococcaceae, Ruminococcus_torques_group, and Parasutterella. More importantly, AA significantly increased fecal TCA (a primary BA) and DCA, LCA, GDCA, dehydro-LCA, and 12-keto-LCA (secondary BAs), thus restoring BA homeostasis. Moreover, AA increased the ratios of DCA/CA, DCA/TCA, and LCA/CDCA and decreased the ratios of TLCA/LCA, GLCA/LCA, and TUDCA/UDCA.Conclusion: The anti-diarrhea effect of AA was associated with restoration of the gut microbiota and BA metabolism-related homeostasis. The results of this study provide insights into the application of AA and the treatment of diarrhea.

Abstract Glioma is the most common malignant tumors in the brain. Previous studies have revealed that, as the innate immune cells in nervous system, microglia cells were involved in glioma pathology. And, the resident microglia displayed its specific biological roles which distinguished with peripheral macrophages. In this study, an integrated analysis was performed based on public resource database to explore specific biological of microglia within glioma. Through comprehensive analysis, the biological characterization underlying two conditions, glioma microglia compared to glioma macrophage (MicT/MacT) as well as glioma microglia compared to normal microglia (MicT/MicN), were revealed. Notably, nine core MicT/MicN genes displayed closely associations with glioma recurrence and prognosis, such as P2RY2, which was analyzed in more than 2800 glioma samples from 25 studies. Furthermore, we applied a random walk based strategy to identify microglia specific subpathways and developed SubP28 signature for glioma prognostic analysis. Multiple validation data sets confirmed the predictive performance of SubP28 and involvement in molecular subtypes. The associations between SuP28 score and microglia M1/M2 polarization were also explored for both GBM and LGG types. Finally, a comprehensive drug-subpathway network was established for screening candidate medicable molecules (drugs) and identifying therapeutic subpathway targets. In conclusions, the comprehensive analysis of microglia related gene and functional signatures in glioma pathobiologic events by large-scale data sets displayed a framework to dissect inner connection between microglia and glioma, and identify robust signature for glioma clinical implications.

Abstract Esophageal squamous cell carcinoma (ESCC) is one of the most common cancers worldwide, with high incidence and mortality rates and low survival rates. However, the detailed molecular mechanism of ESCC progression remains unclear. Here, we first showed significantly higher WNT5A and SNAIL expression in ESCC samples than in corresponding paracancerous samples. High WNT5A and SNAIL expression levels correlated positively with lymphatic metastasis and poor prognosis for patients with ESCC based on immunohistochemical (IHC) staining of 145 paired ESCC samples. Spearman’s correlation analyses confirmed the strong positive correlation between WNT5A and SNAIL expression, and patients with ESCC presenting coexpression of WNT5A and SNAIL had the worst prognosis. Then, we verified that the upregulation of WNT5A promoted ESCC cell metastasis in vivo and in vitro, suggesting that WNT5A might be a promising therapeutic target for the prevention of ESCC. Furthermore, WNT5A overexpression induced the epithelial-mesenchymal transition via histone deacetylase 7 (HDAC7) upregulation, and HDAC7 silencing significantly reversed WNT5A-induced SNAIL upregulation and ESCC cell metastasis. In addition, we used HDAC7 inhibitors (SAHA and TMP269) to further confirm that HDAC7 participates in WNT5A-mediated carcinogenesis. Based on these results, HDAC7 is involved in WNT5A-mediated ESCC progression, and approaches targeting WNT5A and HDAC7 might be potential therapeutic strategies for ESCC.

Abstract Background Most nurses in China have not been trained to take care of end-of-life patients appropriately due to lack of educational resources and insufficient training. A palliative care program was launched by the Jiangsu Nursing Association (JNA training program) and to identify gaps in palliative care training. The main aim of this study was to evaluate the training effects of the JNA training program on nurses’ knowledge and attitudes to palliative care. Methods A cross-sectional study was conducted with 10 048 registered nurses in all regions of Jiangsu. All participants completed an online questionnaire using the Chinese version of The Palliative Care Quiz for Nursing (PCQN-C) and the Frommelt Attitude Toward Care of the Dying scale (FATCOD-B-C). A propensity score matched analysis was performed between the nurses who had attended the JNA training program and whose who hadn’t. Results The average score of PCQN-C among all nurses was 8.79, while the mean score of the FATCOD-B-C was 103.62. Those participants who attended the JNA training program had significantly better scores than those who did not. Propensity score matching analysis showed that the palliative care training program failed to improve nurses’ knowledge in psychosocial and spiritual care or their attitudes towards the necessity of family support although there was positive impact on other aspects of palliative care. Conclusions Knowledge of palliative care among Chinese nurses remains low. Training programs may improve general knowledge and attitudes to palliative care. However, important aspects of knowledge such as communication skills, family support, and psychosocial aspects of care, are missing. These gaps should be filled in future palliative care training programs targeting nurses with oriental culture background.

Under the background of global climate change, rainstorm and flood disasters have become the most serious cataclysm. Under the circumstances of an increasingly severe risk situation, it is necessary to enhance urban disaster resilience. Based on the disaster resilience process of prevention, absorption, and enhancement, and considering the safety factors such as personnel, facility, environment and management, this paper forms a dual dimension of the urban disaster resilience assessment model covering the key elements of urban disaster response and the core capacity of urban disaster recovery. Furthermore, if taking into account the characteristics of rainstorm and flood disasters, the paper screens the key indicators to build up an assessment index system of an urban rainstorm and flood disaster. The practical application was implemented in Beijing to have an assessment of the ability to recover from rainstorm and flood disasters in all districts of Beijing. And then, some pertinent suggestions for enhancing the resilience of Beijing to rainstorm and flood disasters were proposed.

Psoriasis is a chronic inflammatory skin disease occurring worldwide, with multiple systemic complications, which seriously affect the quality of life and physical and mental health of patients. The pathogenesis of psoriasis is related to the environment, genetics, epigenetics, and dysregulation of immune cells such as T cells, dendritic cells (DCs), and nonimmune cells such as keratinocytes. Absent in melanoma 2 (AIM2), a susceptibility gene locus for psoriasis, has been strongly linked to the genetic and epigenetic aspects of psoriasis and increased in expression in psoriatic keratinocytes. AIM2 was found to be activated in an inflammasome-dependent way to release IL-1β and IL-18 to mediate inflammation, and to participate in immune regulation in psoriasis, or in an inflammasome-independent way by regulating the function of regulatory T(Treg) cells or programming cell death in keratinocytes as well as controlling the proliferative state of different cells. AIM2 may also play a role in the recurrence of psoriasis by trained immunity. In this review, we will elaborate on the characteristics of AIM2 and how AIM2 mediates the development of psoriasis.

Abstract Titanium alloys, in particular, medical-grade Ti-6Al-4 V, are heavily used in orthopaedic applications due to their high moduli, strength, and biocompatibility. Implant infection can result in biofilm formation and failure of prosthesis. The formation of a biofilm on implants protects bacteria from antibiotics and the immune response, resulting in the propagation of the infection and ultimately resulting in device failure. Recently, slippery liquid-infused surfaces (LIS) have been investigated for their stable liquid interface, which provides excellent repellent properties to suppress biofilm formation. One of the current limitations of LIS coatings lies in the indistinctive repellency of bone cells in orthopaedic applications, resulting in poor tissue integration and bone ingrowth with the implant. Here, we report a chitosan impregnated LIS coating that facilitates cell adhesion while preventing biofilm formation. The fabricated coating displayed high contact angles (108.2 ± 5.2°) and low sliding angles (3.56 ± 4.3°). Elemental analysis obtained using X-ray photoelectron spectroscopy (XPS) confirmed the availability of fluorine and nitrogen, indicating the presence of fluorosilane and chitosan in the final coating. Furthermore, our results suggest that chitosan-conjugated LIS increased cell adhesion of osteoblast-like SaOS-2 cells and significantly promoted proliferation (a fourfold increase at 7-day incubation) compared to conventional titanium liquid-infused surfaces. Furthermore, the chitosan conjugated LIS significantly reduced biofilm formation of methicillin-resistant Staphylococcus aureus (MRSA) by up to 50% and 75% when compared to untreated titanium and chitosan-coated titanium, respectively. The engineered coating can be easily modified with other biopolymers or capture molecules to be applied to other biomaterials where tissue integration and biofilm prevention are needed.

Recently, object detection in remote sensing images (RSIs) have received extensive attention and made significant progress. Nonetheless, the arbitrary orientations of objects in RSIs make their detection a challenging task. Most of the existing detection methods are difficult to extract the orientation features of objects due to the lack of directionality of conventional convolutions. In addition, the boundary discontinuity in angle regression affects the detection of object orientations. In response to these problems, this article proposes an orientation-first refinement detector (OFRDet), which is based on a strategy that enables the detector to detect the angle of an object ahead of others and presets oriented anchors. In OFRDet, we propose an angle encoding regression module (AERM) and an angle channel attention module (ACAM). AERM transforms angle detection into multiparameter regression, which eliminates boundary discontinuities. ACAM uses convolution kernels with different angles to extract directional features purposefully according to the preset oriented anchors. After these two modules, more accurate bounding boxes are generated and sent to the refined stage to obtain the final detection results. We evaluate our method and demonstrate the effectiveness of it by conducting experiments on two challenging and credible datasets, DOTA, HRSC2016. OFRDet achieves competitive results 79.56%, 96.29% mAP on the two datasets, respectively.

In deep learning-based hyperspectral remote sensing image classification tasks, random sampling strategies are typically used to train model parameters for testing and evaluation. However, this approach leads to strong spatial autocorrelation between the training set samples and the surrounding test set samples, and some unlabeled test set data directly participate in the training of the network. This leaked information makes the model overly optimistic. Models trained under these conditions tend to overfit to a single dataset, which limits the range of practical applications. This paper analyzes the causes and effects of information leakage and summarizes the methods from existing models to mitigate the effects of information leakage. Specifically, this paper states the main issues in this area, where the issue of information leakage is addressed in detail. Second, some algorithms and related models used to mitigate information leakage are categorized, including reducing the number of training samples, using spatially disjoint sampling strategies, few-shot learning, and unsupervised learning. These models and methods are classified according to the sample-related phase and the feature extraction phase. Finally, several representative hyperspectral image classification models experiments are conducted on the common datasets and their effectiveness in mitigating information leakage is analyzed.

The diagnosis of bearing faults is an important guarantee for the healthy operation of mechanical equipment. Due to the time-varying working conditions of mechanical equipment, it is necessary to achieve bearing fault diagnosis under time-varying working conditions. However, the superposition of the two-dimensional working conditions of speed and acceleration brings great difficulties to diagnosis via data-driven models. The long short-term memory (LSTM) model based on the infinitesimal method is an effective method to solve this problem, but its performance still has certain limitations. On this basis, this article proposes a model for fault diagnosis under time-varying operating conditions that combines a residual network model (ResNet) and a gate recurrent unit (model) (GRU). Firstly, the samples were segmented, and feature extraction was performed using ResNet. We then used GRU to process the information. Finally, the classification results were output through the output network. This model could ignore the influence of acceleration and achieve high fault diagnosis accuracy under time-varying working conditions. In addition, we used t-SNE to reduce the dimensionality of the features and analyzed the role of each layer in the model. Experiments showed that this method had a better performance compared with existing bearing fault diagnosis methods.

Abstract Background Cotton is not only a major textile fiber crop but also a vital oilseed, industrial, and forage crop. Oleosins are the structural proteins of oil bodies, influencing their size and the oil content in seeds. In addition, the degradation of oleosins is involved in the mobilization of lipid and oil bodies during seed germination. However, comprehensive identification and the systematic analysis of the Oleosin gene (OLEOs) family have not been conducted in cotton. Results An in-depth analysis has enabled us to identify 25 and 24 OLEOs in tetraploid cotton species G. hirsutum and G. barbadense, respectively, while 12 and 13 OLEOs were identified in diploid species G. arboreum and G. raimondii, respectively. The 74 OLEOs were further clustered into three lineages according to the phylogenetic tree. Synteny analysis revealed that most of the OLEOs were conserved and that WGD or segmental duplications might drive their expansion. The transmembrane helices in GhOLEO proteins were predicted, and three transmembrane models were summarized, in which two were newly proposed. A total of 24 candidate miRNAs targeting GhOLEOs were predicted. Three highly expressed oil-related OLEOs, GH_A07G0501 (SL), GH_D10G0941 (SH), and GH_D01G1686 (U), were cloned, and their subcellular localization and function were analyzed. Their overexpression in Arabidopsis increased seed oil content and decreased seed germination rates. Conclusion We identified OLEO gene family in four cotton species and performed comparative analyses of their relationships, conserved structure, synteny, and gene duplication. The subcellular localization and function of three highly expressed oil-related OLEOs were detected. These results lay the foundation for further functional characterization of OLEOs and improving seed oil content.

Backgroud: Protein kinases play an important role in cell proliferation, differentiation, mobility and cell cycle arrest etc. These enzymes act as important targets in developing anticancer agents. Over the years, a large number of protein kinase inhibitors have been discovered and developed as anticancer agents for the treatment of cancers clinically. However, the drug-resiatance and off-targeting limit their effeciancy for the treatment of human cancer. Materials and methods: Alkaloids are an important class of natural products with broad spectrum biological activities. In the past decades, numerus alkaloids with significant anticancer activity by inhibiting protein kinases were identified. In the present mini-review, we will present the key enzymes including mitogen-activated protein kinase (MAPK), phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/AKT/mTOR) and janus-activated kinases/signal transducer and activator of transcription (JAK/STAT) targeted by alkaloids and highlight the special sites targeted by alkaloids on protein kinases and/or reversing drug resistance. Additionally, the challenge and prospect of developing alkaloids as new anticancer agents are also discussed. Conclusion: Alkaloids suppressed tumor growth through targeting different signaling pathways mediated by protein kinases of cancer cells. It is conceivable that novel alkaloids anticancer agents with promising clinical value will be developed in the future.

IntroductionMunro's microabscess is a typical pathological feature in the early psoriatic lesion, mainly characterized by the accumulation of neutrophils in the epidermis. DNA methylation microenvironment of Munro's microabscess and the crosstalk with transcription and its effect on neutrophils have not yet been revealed.MethodsPerformed genome-wide DNA methylation analysis and further differential methylation analysis of psoriatic skin lesions with and without Munro's microabscess from two batch samples consisting of 114 former samples in the discovery stage and 21 newly-collected samples in the validation stage. Utilized GO, MEME, and other tools to conduct downstream analysis on differentially methylated sites (DMSs). Correlation analysis of methylation level and transcriptome data was also conducted.ResultsWe observed 647 overlapping DMSs associated with Munro's microabscess. Subsequently, GO pathway analysis revealed that DNA methylation might affect the physical properties associated with skin cells through focal adhesion and cellsubstrate junction and was likely to recruit neutrophils in the epidermis. Via the MEME tool, used to investigate the possible binding transcription factors (TFs) of 20 motifs around the 647 DMSs, it was found that DNA methylation regulated the binding of AP1 family members and the recruitment of neutrophils in the epidermis through the TGF-beta pathway and the TH17 pathway. Meanwhile, combined with our earlier transcriptome data, we found DNA methylation would regulate the expressions of CFDP, SIRT6, SMG6, TRAPPC9, HSD17B7, and KIAA0415, indicating these genes would potentially promote the process of Munro's microabscess.DiscussionIn conclusion, DNA methylation may affect the course of psoriasis by regulating the progression of Munro's microabscess in psoriatic skin lesions.

As the basic pathological changes of diabetic retinopathy (DR), the destruction of the blood-retina barrier (BRB) and vascular leakage have attracted extensive attention. Without timely intervention, BRB damage will eventually lead to serious visual impairment. However, due to the delicate structure and complex function of the BRB, the mechanism underlying damage to the BRB in DR has not been fully clarified. Here, we used single-cell RNA sequencing (RNA-seq) technology to analyze 35,910 cells from the retina of healthy and streptozotocin (STZ)-induced diabetic rats, focusing on the degeneration of the main cells constituting the rat BRB in DR and the new definition of two subpopulations of Müller cells at the cell level, Ctxn3+Müller and Ctxn3−Müller cells. We analyzed the characteristics and significant differences between the two groups of Müller cells and emphasized the importance of the Ctxn3+Müller subgroup in diseases. In endothelial cells, we found possible mechanisms of self-protection and adhesion and recruitment to pericytes. In addition, we constructed a communication network between endothelial cells, pericytes, and Müller subsets and clarified the complex regulatory relationship between cells. In summary, we constructed an atlas of the iBRB in the early stage of DR and elucidate the degeneration of its constituent cells and Müller cells and the regulatory relationship between them, providing a series of potential targets for the early treatment of DR.

BackgroundThere are numerous published studies on the association between RET polymorphisms and susceptibility to Hirschsprung disease (HSCR). However, some of the results are inconsistent and the studies were conducted with small sample sizes. Therefore, we performed a meta-analysis to clarify the relationship.MethodsRelevant data were retrieved from PubMed, Web of Science, Cochrane Library, EMBASE, CNKI, and Google Scholar according to PRISMA guidelines. Odds ratios (OR) were calculated to assess susceptibility to HSCR. Meanwhile, heterogeneity and publication bias were also calculated by R software package (version 4.2.1). The protocol was published in PROSPERO (CRD42022348940).ResultsA total of 12 studies were included in the meta-analysis and comprised 12 studies on the RET polymorphism rs2435357 (1,939 subjects and 3,613 controls) and 7 studies on the RET polymorphism rs2506030 (1,849 patients with HSCR and 3,054 controls). The analysis revealed that rs2435357 [A vs. G: odds ratio (OR) = 3.842, 95% confidence interval (CI) 2.829–5.220; AA vs. GG: OR = 2.597, 95% CI 1.499–4.501; AA + AG vs. GG: OR = 6.789, 95% CI 3.0711–14.9973; AA vs. AG + GG: OR = 8.156, 95%CI 5.429–12.253] and rs2506030 (A vs. G: OR = 0.519, 95% CI 0.469–0.573; AA vs. GG: OR = 0.543, 95% CI 0.474–0.623; AA + AG vs. GG: OR = 0.410, 95% CI 0.360–0.468; AA vs. AG + GG: OR = 0.361, 95%CI 0.292–0.447) were significantly associated with susceptibility to HSCR.ConclusionsThe polymorphisms rs2435357 and rs2506030 in the RET may be related to susceptibility to HSCR, of which rs2435357 (T > C) is the causal locus and rs2506030 (A > G) is the protective locus. Background: Colon adenocarcinoma (COAD) is a common malignant tumor of the digestive tract with poor clinical outcomes. Cuproptosis is a novel cell death mechanism and linked to mitochondrial respiration. However, the role of cuproptosis in colon cancer tumor microenvironment (TME) and immune responses remains unknown.Methods: We conducted difference analysis to identify the differential expressed cuproptosis-related genes (CRGs). According to the CRGs, the TCGA-COAD samples were categorized using consensus clustering. The LASSO regression analysis was utilized to develop the cuproptosis-related signature. We then verified the model reliability by Kaplan–Meier, PCA, and ROC analysis. The GES39582 cohort served as the validation set. GO and KEGG functional analyses were conducted to investigate the underlying mechanism. We compared the infiltration levels of immune cells, the expression levels of immune checkpoints, and microsatellite instability (MSI) status between the high- and low-risk groups. Additionally, the relationships between the risk signature and immune cells and cancer stem cell (CSC) were analyzed.Results: Finally, we identified 9 differentially expressed CRGs in COAD. According to the expression of CRGs, the TCGA-COAD samples were separated into two clusters. The 11-gene signature was established by LASSO, and it had excellent predictive power for COAD prognosis. Besides, we used the GSE39582 cohort to validate the prognostic value of the model. GO and KEGG results demonstrated that the survival differences between two risk groups was mainly linked to the extracellular matrix (ECM). Further immune characterization analysis showed the significant differences in the immune cell infiltration and immune responses between two risk groups.Conclusion: Overall, the novel cuproptosis-related signature was able to accurately predict COAD prognosis and played important roles in COAD tumor microenvironment and immune responses.

Due to the highly evolved industrialization and modernization, air quality has deteriorated in most countries. As reported by the World Health Organization (WHO), air pollution is now considered as one of the major threats to global health and a principal risk factor for noncommunicable diseases. Meanwhile, the increasing worldwide prevalence of overweight and obesity is attracting more public attentions. Recently, accumulating epidemiological studies have provided evidence that overweight and obesity may be partially attributable to environmental exposure to air pollution. This review summarizes the epidemiological evidence for the correlation between exposure to various outdoor and indoor air pollutants (mainly particulate matter (PM), nitrogen oxides (NOx), ozone (O3), and polycyclic aromatic hydrocarbons (PAHs)) and overweight and obesity outcomes in recent years. Moreover, it discusses the multiple effects of air pollution during exposure periods throughout life and sex differences in populations. This review also describes the potential mechanism underlying the increased risk of obesity caused by air pollution, including inflammation, oxidative stress, metabolic imbalance, intestinal flora disorders and epigenetic modifications. Finally, this review proposes macro- and micro-measures to prevent the negative effects of air pollution exposure on the obesity prevalence.

Implanted biomaterials elicit an immune-mediated foreign body reaction (FBR) that results in the fibrous encapsulation of the implant and can critically impact the performance of some implants. Consequently, understanding the molecular mechanisms that underpin cell-materials interactions that initiate biomaterial-induced inflammation and fibrosis is critical to improving the performance of biomaterial implants negatively impacted by the FBR. Damage-associated molecular patterns (DAMPs) are endogenous mediators of inflammation that are released upon tissue injury and induce sterile inflammation via Toll-like receptors (TLRs). However, the prevalence of DAMPs within the adsorbed protein layer on material surfaces and their role mediating cell-material interactions is unclear. Previously, our group demonstrated that molecules in fibroblast lysates adsorbed to various biomaterials and induced a potent TLR2-dependent inflammatory response in macrophages at 24 h. In this study, we examined the extended response of RAW-Blue reporter macrophages on lysate or serum-adsorbed Teflon™ AF surfaces to understand the potential role of adsorbed DAMPs in macrophage-material interactions at later time points. Lysate-conditioned surfaces maintained increased nuclear factor kappa B (NF-κB) and activator protein 1 (AP-1) transcription factor activity and increased expression Regulated upon Activation, Normal T Cell Expressed and Presumably Secreted (RANTES/CCL5) at 72 h and 120 h, compared to FBS-conditioned surfaces. In contrast, monocyte chemoattractant protein 1 (MCP-1/CCL2) was only elevated at 72 h in lysate conditions. Transforming growth factor beta 1 (TGF-β1) secretion was significantly increased on lysate-conditioned surfaces, while conditioned media from macrophages on lysate-conditioned surfaces induced alpha smooth muscle actin (αSMA) expression in 3T3 fibroblasts. TLR2 neutralizing antibody treatment significantly decreased NF-κB/AP-1 activity and attenuated TGF-β1 expression at both time points, and MCP-1 and RANTES at 72 h. Finally, multinucleated cells were observed on lysate-conditioned surfaces at 72 h, indicating adsorbed DAMPs induced a fusion permissive environment for adherent macrophages. This study demonstrates that adsorbed DAMPs continue to influence macrophage-material responses beyond the initial 24-h period and maintain a pro-inflammatory and fibrotic response that models aspects of the early FBR. Furthermore, the transient inhibition of TLR2 continued to exert an effect at these later time points, suggesting TLR2 may be a target for therapeutic interventions in FBR.

Attaining optimal eco-efficiency is of paramount importance in promoting the sustainable and harmonious development of the economy and environment within urban agglomerations. Firstly, this paper utilizes the Super-SBM model with undesirable output to measure the eco-efficiency (EE) of 64 cities in the Beijing–Tianjin–Hebei metropolitan region (BTHMR), the Yangtze River Delta (YRD), the Pearl River Delta (PRD), and the Chengdu–Chongqing Economic Zone (CCEZ) from 2006 to 2019. Secondly, this study puts forth a novel and comprehensive index system aimed at evaluating the urbanization efficiency and sheds light on the spatiotemporal changes in EE and urbanization efficiency. Finally, the STIRPAT model is used to examine the influencing factors of EE and to investigate the correlation between EE and urbanization efficiency. The study found that the overall EE of the four typical urban agglomerations is high, but the trend varies with a decrease of about 12.9% from 2006 to 2019. The mean EE is in the order of CCEZ > PRD > BTHMR > YRD, with mean values of 0.941, 0.909, 0.842, and 0.732, respectively. The level of science and technology and the urbanization efficiency have a significant positive impact on EE, while population, industrial structure, FDI, and greening level have an inhibitory effect on urban eco-efficiency. Based on the results, policy suggestions such as paying attention to regional heterogeneity and giving full play to the government’s macro-regulatory role in shaping the economic and industrial structure are proposed to serve as a guide for the coordinated development of urban agglomerations under the Dual Carbon Target.

Hyperspectral image (HSI) super-resolution (SR) is a classical computer vision task that aims to accomplish the conversion of images from lower to higher resolutions. With the booming development of deep learning (DL) technology, more and more researchers are dedicated to the research of image SR techniques based on DL and have made remarkable progress. However, no scholar has provided a comprehensive review of the field. As a response, in this paper we aim to supply a comprehensive summary of the DL-based SR techniques for HSI, including upsampling frameworks, upsampling methods, network design, loss functions, representative works with different strategies, and future directions, in which we design several sets of comparative experiments for the advantages and limitations of two-dimensional convolution and three-dimensional convolution in the field of HSI SR and analyze the experimental results in depth. In addition, the paper also briefly discusses the secondary foci such as common datasets, evaluation metrics, and traditional SR algorithms. To the best of our knowledge, this paper is the first review on DL-based HSI SR.

The two-dimensional discriminant locally preserved projections (2DDLPP) algorithm adds a between-class weighted matrix and a within-class weighted matrix into the objective function of the two-dimensional locally preserved projections (2DLPP) algorithm, which overcomes the disadvantage of 2DLPP, i.e., that it cannot use the discrimination information. However, the small sample size (SSS) problem still exists, and 2DDLPP processes the whole original image, which may contain a large amount of redundant information in the retained features. Therefore, we propose a new algorithm, two-dimensional exponential sparse discriminant local preserving projections (2DESDLPP), to address these problems. This integrates 2DDLPP, matrix exponential function and elastic net regression. Firstly, 2DESDLPP introduces the matrix exponential into the objective function of 2DDLPP, making it positive definite. This is an effective method to solve the SSS problem. Moreover, it uses distance diffusion mapping to convert the original image into a new subspace to further expand the margin between labels. Thus more feature information will be retained for classification. In addition, the elastic net regression method is used to find the optimal sparse projection matrix to reduce redundant information. Finally, through high performance experiments with the ORL, Yale and AR databases, it is proven that the 2DESDLPP algorithm is superior to the other seven mainstream feature extraction algorithms. In particular, its accuracy rate is 3.15%, 2.97% and 4.82% higher than that of 2DDLPP in the three databases, respectively.

A CB/CCE electrochemical sensor for Cr(VI) detection was prepared by ultrasonic dispersion of Ketjen Black and then the coating of modified carbon cloth electrode. Material morphology and composition were characterized based on XPS, TEM, BET, BJH; electrochemical performance of the electrode was studied based on LSV, EIS and CV. The results show that the high conductivity of Ketjen Black accelerates charge transfer on the electrode surface, while the abundant mesoporous structure and large specific surface area enhance Cr(VI) adsorption and reduction by the electrode. Under the optimal experimental conditions, the relationship between the reduction peak current of Cr(VI) and its concentration in the sample was studied by the i-t method. The reduction peak current intensity and its concentration demonstrate a good linear relationship in the range of 0.025-57?M and 57-483?M. The linear equations are: Ip(?A) = -0.2452C(?M)-0.0303 (R = 0.9908), Ip(?A) = -0.0329C(?M)-15.9212 (R = 0.9853), LOD is 9.88nM (S/N = 3). Compared with other methods, the sensor displays outstanding detection performance advantages. Plus the advantages of low cost and environmental protection, it has good application prospects in detecting Cr(VI) in water.

BackgroundProstate cancer is the second most common cancer with a high risk of biochemical recurrence (BCR) among men. Recently, 5-methylcytosine (m5C) modification has attracted more attention as a new layer of RNA post-transcriptional regulation. Hence, we aimed at investigating the potential roles of m5C modification regulators in the BCR of prostate adenocarcinoma (PRAD).MethodsCNV data, mutation annotation data, mRNA expression profiles, and clinical data were downloaded from TCGA and GEO databases. Kaplan-Meier curves analysis, log-rank test, univariate and multivariate Cox regression, and time-dependent ROC curves analysis were performed to evaluate the prognostic factors. Principal components analysis (PCA) was applied to validate the distinction between subgroups. Gene set variation analysis (GSVA) was used to investigate the underlying pathways associated with m5C modification patterns. Single sample gene set enrichment analysis (ssGSEA) was utilized to assess the infiltration of distinct immune cells. Tumor Immune Dysfunction and Exclusion (TIDE) prediction was carried out to assess the potential response to immune checkpoint blockade (ICB) therapy. The m5C modification signature was constructed via LASSO Cox’s proportional hazards regression method.ResultsAfter comprehensively analyzing various types of data from TCGA dataset, and exploring the differential expression and prognostic value of each m5C regulator, we identified m5C modification patterns based on 17 m5C regulators. Two patterns presented a significant difference in the risk of BCR, the tumor microenvironment (TME), and immunotherapy response in PRAD. We found that TET2, which was highly expressed in adjacent normal tissues compared to tumor tissues, was closely associated with many infiltrating immune cells. The m5C modification signature was constructed for the clinical application. Risk score calculated by m5C signature was associated with T stage, N stage, Gleason score, and the possibility of BCR (HR, 4.197; 95% CI, 3.016-5.842; p < 0.001). A higher risk score also represented the possibility of immunotherapy response. Finally, the potential roles of m5C modification signature were validated in the testing dataset.ConclusionsOur study revealed the potential roles of m5C modification in the PRAD BCR and TME diversity, which may provide new insight into the field of prostate cancer in future research.

Severe tracheomalacia (TM) patients with respiratory symptoms need surgical intervention, including aortopexy, internal stents or external splint. While some patients continue to have respiratory symptoms after tracheal relief, and there is no evidence to support any one surgery therapy over another. Here we introduce a clinical safety and efficacy of the three-dimensional (3D)-printed bioresorbable airway external splints in treating congenital heart disease (CHD) patients with severe TM. From May 2019 to September 2020, nine patients with severe TM were enrolled. The median age was 5 months (range, 3–25 months), and the median weight was 7.5 kg (range, 3–15 kg). All patients had wheezing, and two patients were assisted by machine ventilation (MV) preoperatively. The median length of TM was 1.5 cm (range, 1.0–3.0 cm). All patients underwent suspension of a “C”-shaped lumen airway external splint, which were designed in SOLIDWORKS and made of polycaprolactone (PCL). The airway external splint could provided effective support for at least 6 months and was completely degraded into carbon dioxide and water within 2–3 years. The median time of postoperative machine assisted ventilation was 23.7 h (range, 3.3–223.4 h), and the median time of ICU stay was 9 days (range, 4–25 days). The median follow-up time was 18 months (range, 12–24 months). Respiratory symptoms were all relieved, and no external splint-associated complications occurred. The 3D computed tomography reconstruction showed no airway stenosis. Personalized 3D-printed bioresorbable airway external splint can not only limit external compression and prevent airway collapse but also ensure the growth potential of the airway, which is a safe, reliable and effective treatment for CHD with TM.

Glioma is the most common malignant tumor of the central nervous system. Tumor purity is a source of important prognostic factor for glioma patients, showing the key roles of the microenvironment in glioma prognosis. In this study, we systematically screened functional characterization related to the tumor immune microenvironment and constructed a risk model named Glioma MicroEnvironment Functional Signature (GMEFS) based on eight cohorts. The prognostic value of the GMEFS model was also verified in another two glioma cohorts, glioblastoma (GBM) and low-grade glioma (LGG) cohorts, from The Cancer Genome Atlas (TCGA). Nomograms were established in the training and testing cohorts to validate the clinical use of this model. Furthermore, the relationships between the risk score, intrinsic molecular subtypes, tumor purity, and tumor-infiltrating immune cell abundance were also evaluated. Meanwhile, the performance of the GMEFS model in glioma formation and glioma recurrence was systematically analyzed based on 16 glioma cohorts from the Gene Expression Omnibus (GEO) database. Based on multiple-cohort integrated analysis, risk subpathway signatures were identified, and a drug–subpathway association network was further constructed to explore candidate therapy target regions. Three subpathways derived from Focal adhesion (path: 04510) were identified and contained known targets including platelet derived growth factor receptor alpha (PDGFRA), epidermal growth factor receptor (EGFR), and erb-b2 receptor tyrosine kinase 2 (ERBB2). In conclusion, the novel functional signatures identified in this study could serve as a robust prognostic biomarker, and this study provided a framework to identify candidate therapeutic target regions, which further guide glioma patients’ clinical decision.

Natural killer (NK) cells are a type of innate lymphoid cell that are involved in the progression of acute myocardial infarction and ischemic stroke. Although multiple forms of programmed cell death are known to play important roles in these diseases, the correlation between NK cells and apoptosis-related genes during acute myocardial infarction and ischemic stroke remains unclear. In this study, we explored the distinct patterns of NK cell infiltration and apoptosis during the pathological progression of acute myocardial infarction and ischemic stroke using mRNA expression microarrays from the Gene Expression Omnibus database. Since the abundance of NK cells correlated positively with apoptosis in both diseases, we further examined the correlation between NK cell abundance and the expression of apoptosis-related genes. Interestingly, APAF1 and IRAK3 expression correlated negatively with NK cell abundance in both acute myocardial infarction and ischemic stroke, whereas ATM, CAPN1, IL1B, IL1R1, PRKACA, PRKACB, and TNFRSF1A correlated negatively with NK cell abundance in acute myocardial infarction. Together, these findings suggest that these apoptosis-related genes may play important roles in the mechanisms underlying the patterns of NK cell abundance and apoptosis in acute myocardial infarction and ischemic stroke. Our study, therefore, provides novel insights for the further elucidation of the pathogenic mechanism of ischemic injury in both the heart and the brain, as well as potential useful therapeutic targets.
