This study echoes the rising demand for bio-based material in concrete composite structures in the race to accelerate carbon neutrality in construction. Noticing that most previous studies are focused on straight timber or engineered bamboo-to-concrete composite beams, this study focuses on: 1) developing unique and practical curved laminated bamboo-concrete (LBC) composite beams; 2) experimentally investigating and comparing the structural performances of the curved and straight LBC composite beams; 3) developing a novel and proper in-member interface shear slip calibration method that captures the longitudinal after-slip strain redistribution, validated by stain gauge measurements. This study also highlights the interlayer shear bonding strength of laminated bamboo as the thresholding parameter that determines the composite beams' overall flexural strength, evidenced by detailed failure mode analysis. The proposed interface shear slip calibration method can be extended to the other types of shear connectors such as screws, shear plates, etc.

The control method of deformation of large sections of soft rock formations in coal mines is a major problem in mining practice. Taking Wangzhuang Coal Mine in Shanxi Province, China, for example, the support method of large sections of soft rock formations was studied to control the deformation of the top and bottom plates and two gangs, and to obtain the damage of the surrounding rocks at different depths through drilling imaging. In addition, the displacement, plastic area and stress field distribution patterns of surrounding rocks are obtained by simulating in FLAC3D. Coupling support for broken, plastic and elastic zones is achieved using bolts, grout anchors, and long anchor cables. The bottom of the road is effectively controlled by floor grout and concrete reverse arches. The results show that under the combination of anchorage and grout, the fault depth and degree of the surrounding rocks are less than the original support method, which can effectively reduce the subsidon of the roof and the deformation of the two gangs. It is important to note that anchorage cable set support can effectively reduce shear fault areas in floor and two gang plastic areas, and reduce the weight of the pavement floor. This study provides a technical reference for the support of large sections of soft rock roadways in coal mines.

The marine bacterium Vibrio alginolyticus forms only a single flagellum at the cell pole. In Vibrio, two proteins (GTPase FlhF and ATPase FlhG) regulate flagellar number at the cell pole. We previously isolated a mutant strain characterized as NMB155 that forms multiple flagella despite the absence of mutations in flhF and flhG. NMB155 also exhibited straight swimming without a directional change in flagellar rotation. Whole-genome sequencing of NMB155 identified an E9K mutation in FliM that is a component of the C-ring in the flagellar rotor. Mutations in FliM result in defects in flagellar formation (fla) and flagellar rotation (che or mot); however, there are few reports indicating that FliM mutations increase the number of flagella. Here, we determined that the E9K mutation confers the multi-flagellar phenotype and also the che phenotype. The co-expression of wild-type FliM and FliM-E9K indicated that they were competitive in regard to determining the flagellar number. It had been shown that the ATPase activity of FlhG corresponds to the flagellar number. We observed that the ATPase activity of FlhG was increased by the addition of FliM but not by the addition of FliM-E9K. This indicates that the N-terminal region of FliM that includes the E9 residue interacts with FlhG to increase its ATPase activity, and the E9K mutation may inhibit this interaction. We concluded that FliM downregulate FlhG activity to inhibit the formation of additional flagella.ImportanceThe flagellar rotor generates a driving force to rotate the flagellum and is not involved in controlling the number of flagella in Vibrio. However, we observed that the E9K mutation in the rotor protein FliM confers multiple flagella. Our findings reveal a novel regulatory mechanism controlling flagellar number.

SummaryGTPase FlhF and ATPase FlhG are two key factors involved in regulating the flagellum number in Vibrio alginolyticus. FlhG is a paralog of the Escherichia coli cell division regulator MinD, which has a longer N-terminal region. Deleting the N-terminal region of FlhG results in cells with multiple flagella. The Q9A mutation in the DQAxxLR motif of the N-terminal region prevents it from activating the GTPase activity of FlhF in vitro and results in a multi-flagellation phenotype. The mutant FlhG protein was remarkably reduced compared to that of the wild-type protein in vivo. When the mutant FlhG was expressed at the same level as the wild-type FlhG, the number of flagella was restored to the wild-type level. Once synthesized in Vibrio cells, the N-terminal region mutation in FlhG seems not to affect the protein stability. We speculated that the flhG translation efficiency is decreased by N-terminal mutation. Our results suggest that the N-terminal region of FlhG controls the number of flagella by adjusting the FlhF activity and the amount of FlhG in vivo. We speculate that the regulation by FlhG, achieved through transcription by the master regulator FlaK, is affected by the mutations, resulting in reduced flagellar formation by FlhF.

Severe dynamic disturbance in extrathick coal seam mining has become one of the main factors threatening the stability of roadway surrounding rock. In this article, the #6 thick coal seam of Buliangou mine in Inner Mongolia, China, is taken as the engineering background. A mechanical model of the roadway roof structure is established to obtain an analytic formula of the key block subsidence. A three-dimensional discrete element model is established and used to verify the field measurement results. The fracture characteristics of the main roof above the F6104 transport roadway and the deformation and damage evolution law of the surrounding rock during thick coal seam mining are analyzed. The results show that because of the long-term breaking and falling of the roof rocks during extrathick coal seam mining, the F6104 transport roadway will undergo two severe mining disturbances at the locations of 10∼30 m and 50∼70 m ahead of the F6103 working face. During the two disturbance periods, the roadway roof displacement settles to 300∼350 mm and 750∼800 mm, and the deformation of the solid coal wall reaches 650∼700 mm and 1350∼1450 mm, respectively. The energy change curve of the total length of the fractured key roof is obtained, and when mining at 50 m, the basic roof is close to its tensile strength, and the strain energy can reach the peak value of 5.2 × 10 4 kJ, which easily leads to rock burst. The plastic damage zones on both sides of the roadway develop to the roof central area and eventually coalesce, and the deformation of the surrounding rock is obvious. When mining at 50∼70 m, the basic roof breaks and unloads, and elastic strain energy of 3.57 × 10 4 kJ is instantaneously released. These two dynamic disturbances are the main reasons for the instability of the roadway surrounding rock. The results clarify that the failure mechanism investigation of roadways in thick coal seam mining conditions can be effectively applied to control the stability of the roadway surrounding rock under strong mining disturbance.
