Foot rot, also known as stem rot or tuber-root rot syndrome, on sweet potato occurs in sweet potato fields in Okinawa, Japan. Isolates of one fungal species that was consistently isolated from affected sweet potato produced pycnidia having alpha and gamma conidia. The same symptoms as those in the field were produced in vitro after inoculation of test plants with a representative isolate. Based on morphological and molecular phylogenetic evidence, we identified the fungus as Diaporthe destruens. This is the first report of the occurrence of foot rot on sweet potato caused by D. destruens in Japan.

In June 2017, deer-horn-like galls covered with a whitish, powdery fungal layer were found on the trunks of Cinnamomum pseudopedunculatum (Lauraceae) in Hahajima, Bonin Islands, Japan. The galls were similar to those of horn gall disease of Ci. tenuifolium caused by Laurobasidium hachijoense (Laurobasidiaceae, Exobasidiales) on Hachijo Island, approximately 700 km north of Hahajima. Based on morphological observation and molecular analyses of ITS and LSU regions, the fungus isolated from the surface of the galls was identified as L. hachijoense. This is the first report of the horn gall disease of Ci. pseudopedunculatum.

A new combination, Clinoconidium inouyei, is proposed to accommodate Uredo inouyei (syn. Ustilago inouyei, Melanopsichium inouyei, Ustilago machili) from Japan, producing galls on shoot buds of Machilus japonica (Lauraceae). This taxonomic treatment is based on observations of gastroid sporulation in peripheral lacunae of the host galls and basidiospore morphology. This is also supported by phylogenetic analyses with LSU and ITS regions of rDNA. This species is morphologically and phylogenetically similar to C. onumae producing galls on shoot buds of Cinnamomum tenuifolium reported from Japan, but has larger basidiospores.

In November 2015, leaf blight on Schima mertensiana was found in Chichijima, Bonin Islands, Tokyo, Japan. The causal fungus was isolated from the leaf symptoms and identified as Colletotrichum sp. based on morphological examination and molecular analysis using ITS, ACT, CHS-1, CAL, and TUB2 sequences. Leaf blight similar to the natural symptoms developed after leaves were inoculated with the isolate. This is the first report of leaf blight on S. mertensiana caused by Colletotrichum species in Japan, and anthracnose, “tanso-byo” in Japanese, is proposed as the disease name.

In September 2015, stem blight was found on Joseph’s coat amaranth (Amaranthus tricolor) in Kanagawa, Japan. A fungus was isolated from brown spots on the stems and leaves. After our inoculation tests using the isolated fungus, the same symptoms were observed in the field. The pathogen was identified as Diaporthe amaranthophila based on morphological and molecular analyses. This is the first report of D. amaranthophila as a pathogen of stem blight on Joseph’s coat amaranth in Japan.

Since 2010, an unknown fungus in the Gnomoniaceae has been found on overwintered leaves and petioles of Styrax obassia (Styracaceae) in Japan. This fungus is characterized by dark brown immersed or partially erumpent ascomata with long necks and fusiform to obovoid asci each with an acute or long tapering stipe. Each ascus bears eight fusiform to filiform ascospores. Our morphological observation and phylogenetic analyses based on the markers LSU, rpb2, and tef-1α indicated that this is a new monotypic genus in the Gnomoniaceae (Diaporthales), and Tenuignomonia styracis gen. et sp. nov. was descried herein. Members of the Gnomoniaceae are commonly isolated as endophytes, saprobes, and plant pathogens from a broad diversity of herbaceous, shade tree, and agriculturally significant plants. We thus carried out a pathogenicity test to determine if T. styracis is the causative agent of leaf blotch on S. obassia. One week after inoculation, this fungus produced small necrotic spots on the leaves and petioles, and all leaves having necrotic spots were abscised in a short time. We thus confirmed that this fungus has weak pathogenicity on S. obassia. This new species may promote early defoliation of S. obassia during the fall.

By 2016, fruit rot and two different leaf diseases (leaf spot and leaf blight) were found on Synsepalum dulcificum (miracle fruit) in Tokyo, Kanagawa and Kagoshima prefectures of Japan. From the lesions, abundant conidial masses and acervuli of three Colletotrichum species, two of which produced sexual state, were observed. We conducted a pathogenicity assay using these Colletotrichum species on healthy fruits and leaves of S . dulcificum . Our artificial inoculation tests showed symptoms of disease on tested fruit and leaf and indicated all three Colletotrichum species as causal agents of anthracnose on S . dulcificum . Based on morphological characters and molecular phylogenetic analyses using ITS, GAPDH , ACT , CAL and TUB2 loci, these species were identified as Colletotrichum aenigma (MAFF 246750), C . siamense (MAFF 246751) and C . karstii (MAFF 245966). They have been previously reported as plant pathogenic fungi elsewhere in the world. This is the first report of fruit rot, leaf blight and leaf spot on S . dulcificum caused by these three Colletotrichum species .

An international survey of house dust collected from eleven countries using a modified dilution-to-extinction method yielded 7904 isolates. Of these, six strains morphologically resembled the asexual morphs of Aureobasidium and Hormonema (sexual morphs ?Sydowia), but were phylogenetically distinct. A 28S rDNA phylogeny resolved strains as a distinct clade in Dothideales with families Aureobasidiaceae and Dothideaceae their closest relatives. Further analyses based on the ITS rDNA region, β-tubulin, 28S rDNA, and RNA polymerase II second largest subunit confirmed the distinct status of this clade and divided strains among two consistent subclades. As a result, we introduce a new genus and two new species as Zalaria alba and Z. obscura, and a new family to accommodate them in Dothideales. Zalaria is a black yeast-like fungus, grows restrictedly and produces conidiogenous cells with holoblastic synchronous or percurrent conidiation. Zalaria microscopically closely resembles Hormonema by having only one to two loci per conidiogenous cell, but species of our new genus generally has more restricted growth. Comparing the two species, Z. obscura grows faster on lower water activity (aw) media and produces much darker colonies than Z. alba after 7 d. Their sexual states, if extant, are unknown.

Based on prior taxonomic work, these fungi were determined to be highly host specific. We suspected that the association of Cosmospora and their hosts could not be a result of random chance, and tested the cospeciation of Cosmospora and the their hosts with contemporary methods (e.g., ParaFit, PACo, and Jane). The cophylogeny of Cosmospora and their hosts was found to be congruent, but only host-parasite links in more recent evolutionary lineages of the host were determined as coevolutionary. Reconciliation reconstructions determined at least five host-switch events early in the evolution of Cosmospora. Additionally, the rates of evolution between Cosmospora and their hosts were unequal. This pattern is more likely to be explained by pseudocospeciation (i.e., host switches followed by cospeciation), which also produces congruent cophylogenies.

During a global investigation of fungi in house dust, we isolated six novel arthroconidial fungi. Phylogenies from combined analysis of nuc rDNA 18S, 28S and internal transcribed spacers sequences demonstrated that these fungi and two species preserved in culture collections represent undescribed species of Spiromastigaceae, Onygenales. Seven of the eight species lacked sexual states and only characters of asexual states and growth rates on different media could be used to characterize them. The eighth species produced ascomata only on water agar. We introduce six new species and one new combination in Spiromastix and validate the recently proposed family Spiromastigaceae, genus Pseudospiromastix and combination Ps. tentaculata. The new genus Sigleria is proposed for two new species that differ from Spiromastix by conidiophore branching patterns, slower growth and a limited ability to utilize nitrate as a sole N source. A key to the three genera of Spiromastigaceae, Spiromastix, Pseudospiromastix and Sigleria, is provided. Phylogenetic analyses support the placement of Spiromastigaceae within Onygenales.

The co-existence of males, females and hermaphrodites, a rare mating system known as trioecy, has been considered as an evolutionarily transient state. In nematodes, androdioecy (males/hermaphrodites) as found in Caenorhabditis elegans, is thought to have evolved from dioecy (males/females) through a trioecious intermediate. Thus, trioecious species are good models to understand the steps and requirements for the evolution of new mating systems. Here we describe two new species of nematodes with trioecy, Auanema rhodensis and A. freiburgensis. Along with molecular barcodes, we provide a detailed analysis of the morphology of these species, and document it with drawings and light and SEM micrographs. Based on morphological data, these free-living nematodes were assigned to a new genus, Auanema, together with three other species described previously. Auanema species display convergent evolution in some features with parasitic nematodes with complex life cycles, such as the production of few males after outcrossing and the obligatory development of dauers into self-propagating adults.

A new disease found on garland chrysanthemum in Osaka, Japan in 2009 and its causal agent were identified and characterized. Light brown spots first appeared on lower leaves of seedlings, and the leaves blighted or rotted. A fungus isolated from diseased plants, described recently as Gibellulopsis chrysanthemi, was demonstrated to reproduce the natural symptoms in inoculation tests. Sufficiently long, moist periods after inoculation promoted the infection even after lengthy dry periods. The pathogen also caused the disease on chrysanthemum and lettuce, but not on seven other vegetables. The fungus caused the disease after 6 months of dry storage. The disease was termed seedling rot (“nae-fuhai-byo” in Japanese).

With the recent changes concerning pleomorphic fungi in the new International Code of Nomenclature for algae, fungi, and plants (ICN), it is necessary to propose the acceptance or protection of sexual morph-typified or asexual morph-typified generic names that do not have priority, or to propose the rejection or suppression1 of competing names. In addition, sexual morph-typified generic names, where widely used, must be proposed for rejection or suppression in favour of asexual morph-typified names that have priority, or the latter must be proposed for conservation or protection. Some pragmatic criteria used for deciding the acceptance or rejection of generic names include: the number of name changes required when one generic name is used over another, the clarity of the generic concept, their relative frequencies of use in the scientific literature, and a vote of interested mycologists. Here, twelve widely used generic names in three families of Hypocreales are proposed for acceptance, either by conservation or protection, despite their lack of priority of publication, or because they are widely used asexual morph-typified names. Each pair of generic names is evaluated, with a recommendation as to the generic name to be used, and safeguarded, either through conservation or protection. Four generic names typified by a species with a sexual morph as type that are younger than competing generic names typified by a species with an asexual morph type, are proposed for use. Eight older generic names typified by species with an asexual morph as type are proposed for use over younger competing generic names typified by a species with a sexual morph as type. Within Bionectriaceae, Clonostachys is recommended over Bionectria; in Hypocreaceae, Hypomyces is recommended over Cladobotryum, Sphaerostilbella over Gliocladium, and Trichoderma over Hypocrea; and in Nectriaceae, Actinostilbe is recommended over Lanatonectria, Cylindrocladiella over Nectricladiella, Fusarium over Gibberella, Gliocephalotrichum over Leuconectria, Gliocladiopsis over Glionectria, Nalanthamala over Rubrinectria, Nectria over Tubercularia, and Neonectria over Cylindrocarpon.

Gibellulopsis chrysanthemi sp. nov. is described and illustrated from specimens on rotten leaves of garland chrysanthemum (Chrysanthemum coronarium L. var. spatiosum L.H. Bailey), collected in three different sites of Osaka Prefecture, Japan. This species is characterized by having short and long verticillium-like conidiophores, 1-septate, long-cylindrical conidia with tapering ends and slightly brownish chlamydospores. Compared morphologically with other species in the Plectosphaerellaceae (sister to the Glomerellales), G. chrysanthemi is similar to Gibellulopsis nigrescens, from which it differs in the formation of long conidiophores and 1-septate, long-cylindrical conidia with tapering ends. In addition, the molecular analyses of G. chrysanthemi and other members of the Plectosphaerellaceae based on a phylogenetic tree with three-loci (ITS, D1/D2, tef1-α) sequences reveal that G. chrysanthemi is located as a sister group of G. nigrescens.

We examined the taxonomic position of Leptosphaerulina japonica, a parasitic fungus on male strobili of Japanese cedar (Cryptomeria japonica) using morphology and molecular phylogeny. Because neither specimens nor illustrations were designated in the original paper, this name is neotypified with a fresh specimen collected from the type locality, Yamagata, Japan. Leptosphaerulina japonica has asci produced in a broad layer arising from the entire base of the locule, stipitate acervuli, and a hormonema-like synanamorph. These characteristics are not typical of the genus Leptosphaerulina (Didymellaceae, Pleosporales). In a molecular phylogenetic analysis of ITS sequences, L. japonica was placed into the Dothideaceae/Dothioraceae, Dothideales. Although muriform ascospores and an anamorph with long stipitate acervuli have not been reported for species in Sydowia, the other morphology and phylogeny support placement of L. japonica in that genus. Thus, a new combination, Sydowia japonica, is proposed.

The region of the mitochondrial cytochrome c oxidase subunit 1 used as the animal barcode was excluded as a potential marker, because it is difficult to amplify in fungi, often includes large introns, and can be insufficiently variable. Three subunits from the nuclear ribosomal RNA cistron were compared together with regions of three representative proteincoding genes (largest subunit of RNA polymerase II, second largest subunit of RNA polymerase II, and minichromosome maintenance protein). Although the protein-coding gene regions often had a higher percent of correct identification compared with ribosomal markers, low PCR amplification and sequencing success eliminated them as candidates for a universal fungal barcode. Among the regions of the ribosomal cistron, the internal transcribed spacer (ITS) region has the highest probability of successful identification for the broadest range of fungi, with the most clearly defined barcode gap between inter- and intraspecific variation. The nuclear ribosomal large subunit, a popular phylogenetic marker in certain groups, had superior species resolution in some taxonomic groups, such as the early diverging lineages and the ascomycete yeasts, but was otherwise slightly inferior to the ITS. The nuclear ribosomal small subunit has poor species-level resolution in fungi. ITS will be formally proposed for adoption as the primary fungal barcode marker to the Consortium for the Barcode of Life, with the possibility that supplementary barcodes may be developed for particular narrowly circumscribed taxonomic groups.

Although Nectria is the type genus of Nectriaceae (Hypocreales, Sordariomycetes, Pezizomycotina, Ascomycota), the systematics of the teleomorphic and anamorphic state of Nectria sensu Rossman has not been studied in detail. The objectives of this study are to 1) provide a phylogenetic overview to determine if species of Nectria with Gyrostroma, Tubercularia, and Zythiostroma anamorphs form a monophyletic group; 2) define Nectria, segregate genera, and their species using morphologically informative characters of teleomorphic and anamorphic states; and 3) provide descriptions and illustrations of these genera and species. To accomplish these objectives, results of phylogenetic analyses of DNA sequence data from six loci (act, ITS, LSU, rpb1, tef1 and tub), were integrated with morphological characterisations of anamorphs and teleomorphs. Results from the phylogenetic analyses demonstrate that species previously regarded as the genus Nectria having Gyrostroma, Tubercularia, and Zythiostroma anamorphs belong in two major paraphyletic clades. The first major clade regarded as the genus Pleonectria contains 26 species with ascoconidia produced by ascospores in asci, perithecial walls having bright yellow scurf, and immersed or superficial pycnidial anamorphs (Zythiostroma = Gyrostroma). A lineage basal to the Pleonectria clade includes Nectria miltina having very small, aseptate ascospores, and trichoderma-like conidiophores and occurring on monocotyledonous plants. These characteristics are unusual in Pleonectria, thus we recognise the monotypic genus Allantonectria with Allantonectria miltina. The second major clade comprises the genus Nectria sensu stricto including the type species, N. cinnabarina, and 28 additional species. Within the genus Nectria, four subclades exist. One subclade includes species with sporodochial anamorphs and another with synnematous anamorphs. The other two paraphyletic subclades include species that produce abundant stromata in which the large perithecia are immersed, large ascospores, and peculiar anamorphs that form pycnidia or sporodochia either on their natural substrate or in culture. In this study the evolution of species, morphology, and ecology of the three genera, Allantonectria, Nectria, and Pleonectria, are discussed based on the phylogenetic analyses. In addition, descriptions, illustrations, and keys for identification are presented for the 56 species in Allantonectria, Nectria, and Pleonectria. Species of Clonostachys and Trichoderma isolated from the surface of bananas were highly antagonistic to crown rot-causing fungal pathogens of banana, such as Lasiodiplodia theobromae, Thielaviopsis paradoxa, Colletotrichum musae, and Fusarium verticillioides. Cultural and morphological examinations revealed that the fungal antagonists were similar to C. byssicola and T. harzianum, with some characters overlapping with closely related species. Molecular identification is recommended since the fungal isolates could not be differentiated adequately by cultural and morphological methods. Accurate taxonomy of these fungal antagonists was essential for the subsequent biological control studies. The tub2 region of β-tubulin genes of Clonostachys and 5.8S rDNA with the ITS regions of Trichoderma isolates were analyzed to determine their phylogenic placement. Neonectria is a cosmopolitan genus and it is, in part, defined by its link to the anamorph genus Cylindrocarpon. Neonectria has been divided into informal groups on the basis of combined morphology of anamorph and teleomorph. Previously, Cylindrocarpon was divided into four groups defined by presence or absence of microconidia and chlamydospores. Molecular phylogenetic analyses have indicated that Neonectria sensu stricto and Cylindrocarpon sensu stricto are phylogenetically congeneric. In addition, morphological and molecular data accumulated over several years have indicated that Neonectria sensu lato and Cylindrocarpon sensu lato do not form a monophyletic group and that the respective informal groups may represent distinct genera. In the present work, a multilocus analysis (act, ITS, LSU, rpb1, tef1, tub) was applied to representatives of the informal groups to determine their level of phylogenetic support as a first step towards taxonomic revision of Neonectria sensu lato. Results show five distinct highly supported clades that correspond to some extent with the informal Neonectria and Cylindrocarpon groups that are here recognised as genera: (1) N. coccinea-group and Cylindrocarpon groups 1 & 4 (Neonectria/Cylindrocarpon sensu stricto); (2) N. rugulosa-group (Rugonectria gen. nov.); (3) N. mammoidea/N. veuillotiana-groups and Cylindrocarpon group 2 (Thelonectria gen. nov.); (4) N. radicicola-group and Cylindrocarpon group 3 (Ilyonectria gen. nov.); and (5) anamorph genus Campylocarpon. Characteristics of the anamorphs and teleomorphs correlate with the five genera, three of which are newly described. The genus Nectria is typified by N. cinnabarina , a wood-inhabiting fungus common in temperate regions of the Northern Hemisphere. To determine the diversity within N. cinnabarina , specimens and cultures from Asia, Europe, and North America were obtained and examined. Their phylogeny was determined using sequences of multiple loci, specifically act, ITS, LSU, rpb1, tef1, and tub. Based on these observations, four species are recognised within the N. cinnabarina complex. Each species is delimited based on DNA sequence analyses and described and illustrated from specimens and cultures. The basionym for N. cinnabarina, Sphaeria cinnabarina, is lectotypified based on an illustration that is part of the protologue, and an epitype specimen is designated. Nectria cinnabarina s. str. is recircumscribed as having 2-septate ascospores and long stipitate sporodochia. Nectria dematiosa , previously considered a synonym of N. cinnabarina , has up to 2-septate ascospores and sessile sporodochia or no anamorph on the natural substrate. A third species, Nectria nigrescens, has up to 3-septate ascospores and short to long stipitate sporodochia. One newly described species, Nectria asiatica with a distribution restricted to Asia, has (0–)1-septate ascospores and short stipitate sporodochia. Young and mature conidia developing on SNA were observed for each species. Mature conidia of N. asiatica, N. cinnabarina , and N. nigrescens but not N. dematiosa bud when the mature conidia are crowded. On PDA the optimal temperature for growth for N. dematiosa is 20 °C, while for the other three species it is 25 °C. Based on our phylogenetic analyses, three subclades are evident within N. dematiosa . Although subtle culture and geographical differences exist, these subclades are not recognised as distinct species because the number of samples is small and the few specimens are insufficient to determine if morphological differences exist in the natural environment. Rotting of roots and stem bases and wilting of entire plants were found on a gentianaceous flowering plant, prairie gentian (Eustoma grandiflorum), grown in Kagawa Prefecture in the southwest region of Japan in April 2001. A mitosporic fungus, isolated repeatedly from the diseased plants, was identified as a species belonging to the clade 3 of Fusarium solani species complex based on the morphology and the sequence of the translation elongation factor gene. It was demonstrated to cause the disease by inoculating potted plants and reisolating the fungus from the diseased plants. We propose the name “Fusarium root rot of prairie gentian” for this disease.

Foliar wilt as well as crown and root rot with sclerotia formation has affected potted liver leaf (Hepatica nobilis var. japonica f. magna) in Ojiya, Niigata Prefecture, Japan, since 2006. Apothecia developed from the sclerotia on soil surface of pots with the diseased plants in March. A fungus forming the apothecia was identified as Dumontinia tuberosa (Sclerotiniaceae) based on its morphology and demonstrated to cause the disease. We coined the name “Dumontinia root rot (Dumontinia-negusare-byo in Japanese) of liver leaf” for the new disease.

Verrucostoma freycinetiae gen. et sp. nov. is described and illustrated from specimens on dead leaves of Freycinetia boninensis (Pandanaceae) collect- ed in Hahajima, Bonin (Ogasawara) Islands, Japan. The genus is characterized by pale orange perithecia with protuberances around the perithecial apex, no color change in 3% potassium hydroxide and lactic acid, unitunicate asci, spinulose ascospores and an Acremonium-like anamorph. Morphological charac- ters were compared with other genera in the Bionectriaceae and Nectriaceae (Hypocreales). Verru- costoma is morphologically similar to Bionectria (Bionectriaceae) from which it differs in the forma- tion of conspicuous protuberances around the perithecial apex and the Acremonium-like anamorph. Moreover molecular analyses of Verrucostoma and other members of the Bionectriaceae and Nectriaceae based on a-actin, large subunit nuclear ribosomal DNA and RNA polymerase II subunit 1 sequences support the conclusions based on morphological data. Our results confirm that V. freycinetiae is distinct from other genera among the Nectria-like fungi and represents a new genus belonging to the Bionectria- ceae.

Based on inoculation experiments and morphological and molecular phylogenetic studies, Cylindrocladium damping-off of strawberry and Cylindrocladium blight of coral bells in Japan were demonstrated to be caused by Cylindrocladium canadense. This is the first report of a disease on strawberry and coral bells caused by this fungus.

Seven species of the genus Cosmospora collected in Japan are reported in this article. Among them, Cosmospora japonica is described as a new species. Cosmospora henanensis, C. rishbethii, and C. triqua, all of which are known only from their type localities, are added to the Japanese mycobiota. The other species, C. chaetopsinaecatenulatae, C. diminuta, and C. peponum, are new records for Japan. Additional distribution records are given for Cosmospora species hitherto known in Japan.

Brown leaf spot disease caused by Cylindrocladium was found on Howea belmoreana on Hachijojima Island, Tokyo, Japan, in December 2001. Typical symptoms were incited after artificial inoculation. A culture of white mycelia, isolated from leaf spot symptoms, produced reddish perithecia of a nectriaceous fungus. Based on morphological and molecular analyses, this fungus was identified as Calonectria ilicicola (anamorph: Cylindrocladium parasiticum). Pathogenicity of this fungus on five plants cultivated on Hachijojima Island was confirmed by artificial inoculation. This report is the first on Cylindrocladium brown leaf spot of H. belmoreana caused by C. ilicicola (anamorph: Cy. parasiticum).

A ring spot disease of Aloe vera was found on leaves of potted seedlings of Aloe vera in Hachijojima and Chichijima Islands, Tokyo. From tissue of ring spot lesions, a fungus producing Fusarium-type conidia was consistently isolated. After 1 month, reddish perithecia of nectriaceous fungus had formed on the colonies of this isolate on PDA. These nectriaceous and Fusarium fungi were identified as Haematonectria haematococca and Fusarium sp., respectively. From a single ascospore isolation, the former was confirmed to be the teleomorph of the Fusarium sp. Typical ring spot lesions were reproduced by artificial inoculations using single ascospore and single conidium isolates. Inoculations of five species of genus Aloe revealed that they were highly susceptible except for A. arborescens. This is the first report of a disease on Aloe caused by H. haematococca (anamorph: Fusarium sp.) in Japan, and it was named aloe ring spot.

Based on the result of morphological and the phylogenetic analyses, three Bionectria (Bionectriaceae: Hypocreales) species are added to the mycobiota of Japan. Among them, one found in Okinawa and Kagoshima Prefectures is described as a new species, Bionectria pseudostriatopsis (anamorph: Clonostachys pseudostriatopsis). The other two, B. grammicospora and B. sporodochialis, are new records from Japan. Additional distribution records are given for Bionectria species hitherto known in Japan.

Basidioascus undulatus is a soil basidiomycete belonging to the order Geminibasidiales. The taxonomic status of the order was unclear as originally it was only tentatively classified in the class Wallemiomycetes. The fungi in Geminibasidiales have an ambiguously defined sexual cycle. In this study, we sequenced the genome of B. undulatus to gain insights into its sexuality and evolutionary origins. The assembled genome draft was approximately 32 Mb in size, had a median nucleotide coverage of 24X, and contained 6123 predicted genes. Previous morphological descriptions of B. undulatus relied on interpretation of putative sexual structures. In this study, nuclear staining and confocal microscopy showed meiosis occurring in basidia and genome analysis confirmed the existence of genes involved in meiosis and mating. Using 35 protein-coding genes extracted from genomic information, phylogenomic and molecular dating analyses confirmed that B. undulatus indeed belongs to a lineage distantly related to Wallemia while retaining a basal position in Agaricomycotina. These results, combined with differences in septal pore morphology, led us to move the order Geminibasidiales out of the Wallemiomycetes and into the new class Geminibasidiomycetes cl. nov. Finally, the concept of Agaricomycotina is emended to include both Wallemiomycetes and Geminibasidiomycetes.

A cleistothecial fungus, known only from the shells of giant land snails of the family Rhytidae, is described as a new genus and species within Onygenales, Harorepupu aotearoa gen. sp. nov. Known only from the sexual morph, this fungus is characterized morphologically by a membranous ascoma with no appendages and ascospores with a sparse network of ridges. Ribosomal DNA sequences place the new species within Onygenales, but comparison with the known genetic diversity within the order linked it to no existing genus or family. It is the first species of Onygenales reported from the shells of terrestrial snails. This fungus has been listed as Critically Endangered in New Zealand and has been previously referred to as ‘Trichocomaceae gen. nov.’ in those threat lists.

Thelonectria discophora (Thelonectria, Nectriaceae, Hypocreales) is a conspicuous group of saprobic fungi on decaying plant material, characterized by red perithecia each with a broad mammiform (nipple-like) apex. The anamorphic state is characterized by a cylindrocarpon-like morphology, with 3?5 septate macroconidia, unicellular microconidia and chlamydospores are rarely produced in culture. In the past, T. discophora was regarded as one species with a wide geographic distribution. However, a recent study rejected the monophyly and cosmopolitanism of this species, and showed the existence of at least sixteen cryptic species based solely on molecular data. In the present paper, we revise the taxonomy of T. discophora species complex by describing twelve new species and four new combinations based on historic names. Individual diagnostic morphological characters for each species could not be identified; however, discrete morphological traits corresponding to each one of the three main groups of species were discovered. Lineages could be differentiated by average values of morphological traits as well as presence/absence of characteristic asexual propagules and colony growth at 30 C. A new nectrioid fungus with its sporodochial Cylindrocarpon anamorph, collected on dead bark of Luchu pines (Pinus luchuensis) in the southern part of Japan (Kagoshima and Okinawa), having perithecia slightly constricted just below the papilla and conidia with a strongly hooked and acute apical cell, belongs to the genus Neonectria according to the recent concept of the Nectriaceae (Hypocreales). Molecular phylogenetic analysis based on the tub2 region of β-tubulin genes also supports the morphological consideration. This fungus is described as Neonectria amamiensis (anamorph: Cylindrocarpon amamiense).

During a survey of seed diseases of Fagus crenata, a new fungal disease of the seeds was found with high frequency in Akita, northern Japan. Main symptoms are often expressed as browning of the cracked parts from exposed cotyledons and complete loss of viability of infected seeds. Reddish perithecia and whitish yellow sporodochia were occasionally observed symptoms and determined that they were anamorph–telemorph relationship on the basis of both cultural observations. Inoculation studies confirmed that this fungus was the cause of seed rot. The fungus is morphologically identical with Neonectria ramulariae (anamorph: Cylindrocarpon obtusiusculum) that is well known as the soil-born fungi around the world. Sequences between the authentic isolate of Neo. ramulariae (CBS 151.29) and the pathogenic fungus based on ITS, LSU and tub showed high similarity. Thus, ‘seed rot’, the new disease of beech seeds caused by Neo. ramulariae (anamorph: Cyl. obtusiusculum) was proposed in this study.

Severe blight of potted seedlings of monkshood caused by Plectosporium tabacinum was found in glasshouses in Kagawa Prefecture in southwest Japan in May 2001. Root rot and browning of stem bases were followed by wilting and yellowing of leaves, then blighting of leaves, flower buds and stems. A fungus was isolated from diseased plants and confirmed to cause the disease. The new disease was named “Plectosporium blight of monkshood”.

[EN] Novel species of microfungi described in the present study include the following from South Africa: Cercosporella dolichandrae from Dolichandra unguiscati, Seiridium podocarpi from Podocarpus latifolius, Pseudocercospora parapseudarthriae from Pseudarthria hookeri, Neodevriesia coryneliae from Corynelia uberata on leaves of Afrocarpus falcatus, Ramichloridium eucleae from Euclea undulata and Stachybotrys aloeticola from Aloe sp. Novel species from the Netherlands include: Stagonospora trichophoricola, Keissleriella trichophoricola and Dinemasporium trichophoricola from Trichophorum cespitosum, Phaeosphaeria poae, Keissleriella poagena, Phaeosphaeria poagena, Parastagonospora poagena and Pyrenochaetopsis poae from Poa sp., Septoriella oudemansii from Phragmites australis and Dendryphion europaeum from Hedera helix (Germany) and Heracleum sphondylium (the Netherlands). Taphrina fungi are biotrophic plant pathogens that cause plant deformity diseases. We sequenced the genomes of four Taphrina species-Taphrina wiesneri, T. deformans, T. flavorubra, and T. populina-which parasitize Prunus, Cerasus, and Populus hosts with varying severity of disease symptoms. High levels of gene synteny within Taphrina species were observed, and our comparative analysis further revealed that these fungi may utilize multiple strategies in coping with the host environment that are also found in some specialized dimorphic species. These include species-specific aneuploidy and clusters of highly diverged secreted proteins located at subtelomeres. We also identified species differences in plant hormone biosynthesis pathways, which may contribute to varying degree of disease symptoms. The genomes provide a rich resource for investigation into Taphrina biology and evolutionary studies across the basal ascomycetes clade.

[Background] Horizontal gene transfer (HGT) has been suggested as the mechanism by which various plant parasitic nematode species have obtained genes important in parasitism. In particular, cellulase genes have been acquired by plant parasitic nematodes that allow them to digest plant cell walls. Unlike the typical glycoside hydrolase (GH) family 5 cellulase genes which are found in several nematode species from the order Tylenchida, members of the GH45 cellulase have only been identified in a cluster including the families Parasitaphelenchidae (with the pinewood nematode Bursaphelenchus xylophilus) and Aphelenchoididae, and their origins remain unknown., [Results] In order to investigate the distribution and evolution of GH45 cellulase genes in nematodes and fungi we performed a wide ranging screen for novel putative GH45 sequences. This revealed that the sequences are widespread mainly in Ascomycetous fungi and have so far been found in a single major nematode lineage. Close relationships between the sequences from nematodes and fungi were found through our phylogenetic analyses. An intron position is shared by sequences from Bursaphelenchus nematodes and several Ascomycetous fungal species., [Conclusions] The close phylogenetic relationships and conserved gene structure between the sequences from nematodes and fungi strongly supports the hypothesis that nematode GH45 cellulase genes were acquired via HGT from fungi. The rapid duplication and turnover of these genes within Bursaphelenchus genomes demonstrate that useful sequences acquired via HGT can become established in the genomes of recipient organisms and may open novel niches for these organisms to exploit., This work was supported by JSPS KAKENHI Grant Numbers 20353659 and 23248024. Severe rot of leaves, peduncles and flowers caused by Gibberella zeae (anamorph: Fusarium graminearum) was found on potted plants of hyacinth (Hyacinthus orientalis), a liliaceous ornamental, in greenhouses in Kagawa Prefecture, Japan, in January 2001. This disease was named “Fusarium rot of hyacinth” as a new disease because only the anamorph, F. graminearum, was identified on the diseased host plant.

New perennial canker diseases of Abies veitchii and Acer crataegifolium are described. Pathogenicity of the causal fungus was confirmed on stems of young Abies and Acer trees using two monoascospore isolates from the perithecia from the two tree species. The causal fungus, producing Nectria-state-teleomorph and Cylindrocarpon-anamorph, was identified as Nectria castaneicola. It was then transferred to the genus Neonectria, as N. castaneicola (W. Yamam. et Oyasu) Tak. Kobay. et Hirooka, comb. nov. followed by the recent concept of Nectriaceae.

In Japan, Japanese cedar pollen dispersal is one of the major causes of pollinosis. Sydowia japonica is an ascomycetous fungus that grows exclusively on the male strobili of Japanese cedar, suggesting a possible mechanism for controlling pollen dispersal. To evaluate this possibility, eleven isolates of S. japonica were collected from around Japan and used as an inoculum to male strobili of Japanese cedar. The treatment demonstrated that the fungus infected only the pollen and prevented pollen dispersal. The fungus did not cause any additional symptoms to other parts of Japanese cedar, such as needles, stems, and buds. All S. japonica isolates collected around Japan could serve to control pollen dispersal. Periodic observation of the fungal pathogenesis with stereomicroscope and scanning electron microscope showed that hyphal fragments and conidia of S. japonica germinated on the surface of male strobili, and the germ tube entered pollen sacs through opening microsporophylls. Within the pollen sacs, the hyphae penetrated pollen gradually, such that all pollen was infected by the fungus by approximately one month before the pollen dispersal season. The infected pollen was destroyed due to the fungal infection and was never released. Our data suggests a novel approach of preventing pollen dispersal using pollen-specific fungal infection.

Differences between Neonectria castaneicola, which causes stem and perennial canker of trees, and Neo. rugulosa have not been clearly shown in previous studies. In this study these two species were compared in detail using 17 Japanese isolates consisting of 10 strains of Neo. castaneicola and seven of Neo. rugulosa. Four-spored asci were constantly found in Neo. castaneicola and this species produced larger ascospores and macroconidia than Neo. rugulosa which produced eight-spored asci. The mating system of Neo. castaneicola was homothallic while Neo. rugulosa was heterothallic. Characters in each species, such as the number of ascospores in an ascus and mating system, were constantly transferred to the 3rd generation. Molecular analysis revealed that the 10 isolates of Neo. castaneicola and seven of Neo. rugulosa were differentiated using rDNA sequence data from the nuclear rDNA ITS region. Moreover, Neo. castaneicola and Neo. rugulosa were separated into different clades. From these results, it was concluded that Neo. castaneicola should be maintained as an independent species, separate from Neo. rugulosa. The isolates of Neo. rugulosa used in this study were the first reported in Japan and found on Castanea crenata, Castanopsis sp., Myrica rubra and Quercus acutissima.
