Abstract

SUMMARY: The significant concentrations of phosphate in wastewater can be considered as an important fertilizer source. Some microalgae uptake phosphate from wastewater and store it in the form of polyphosphate (polyP). Therefore, biological phosphorus recovery processes have been attracting significant attention recently. However, wastewaters are often rich in ammonium, and so microalgae used for phosphorus recovery should be tolerant of this component. In the present study, 151 microalgae were isolated from freshwater, brackish water and seawater. Among them, 11 strains showed higher growth rates in ammonium‐rich media. Of these, the strain SLG4‐06 originating from a marine environment showed the highest polyP accumulation rate and was selected as the most potential alga for phosphorus recovery from wastewater. Based on the morphological and ultrastructural characteristics and phylogenetic analyses of nuclear‐encoded 18S rDNA and chloroplast‐encoded rbcL of strain SLG4‐06, we propose Picochloropsis malayensis gen. et sp. nov. for this strain in the Chlorellales, Trebouxiophyceae. Because P. malayensis can grow in both seawater and freshwater environments, it may be suitable for wastewater treatment in marine aquaculture ponds with widely fluctuating salinity levels. 42. Enhancing Diatom, Cyclotella meneghiniana Growth Using Growth-Promoting Bacteria Isolated From the Phycosphere of Chlorophytes and Chrysophytes.

Abstract

The relationship between microalgae and bacteria in a microenvironment, the phycosphere, has a significant role in enhancing the quality and quantity of microalgal production, which would in turn affect consumers' growth and nutritional quality, such as the zooplankton, which are important live feeds in aquaculture. Thus, selecting and characterising suitable microalgal growth-promoting bacteria (MGPB) for enhancing microalgal production is an important process since not all bacteria promote high growth. In this study, physcosphere bacteria associated with chlorophytes and chrysophytes were isolated, screened for their microalgae-promoting attributes (phosphorus solubilisation, indole-3-acetic acid (IAA) production, and nitrogen fixation) and re-inoculated into microalgal cultures. A total of seven bacterial isolates were recorded to have multiple growth promoting traits, with three strains (CY-2, CY-4, CY-5) showing the greatest (P < 0.05) link for those traits. These seven potential MGPB were molecularly characterised using 16S rRNA approach. The phylogenetic tree of the isolated bacteria demonstrated the dominant bacteria associated with the chlorophytes were in the class bacteroidetes, while the chrysophytes appeared to be associated with Firmicutes bacteria suggesting that the compositions were strictly species-specific to the microalgae host. Enhanced Cyclotella meneghiniana growth by the seven isolated bacterial strains was highly dependent on the growth-promoting traits; especially those demonstrated by Pseudomonas hibiscicola and Ochrobactrum haematophilum. These two bacteria showed the potential to enhance the quality of microalgae, and they could be bioencapsulated and used to improve the quality of zooplankton as one of the main live feeds in the aquaculture industry. 43. Physicochemical Impacts on Bacterial Communities in Putrajaya Lake, Malaysia.

Abstract

This study determines the associations between the bacterial communities and water physicochemical parameters in Putrajaya Lake and Putrajaya Wetlands Park, Malaysia. Bacterial communities were assessed by metagenomics of the 16S rRNA gene from lake water input, central wetlands, and primary lake area. Water samples (n=18) were collected during two different periods: post-high rainfall events (samples collected in May) and dry periods (July). The data revealed that bacterial communities of the three sites were taxonomically distinct and associated with different environmental parameters. However, no significant differences were found between the wet and dry periods. Alpha diversity analyses revealed the highest index in May 2018 in the constructed wetlands (H'= 5.397) than those from water input or primary lake (p<0.05). Overall, 49 phyla, 147 classes, 284 orders, 471 families, 778 genera and 62 species of bacteria were identified. Verrumicrobia and Firmicutes showed a strong positive correlation with ammonianitrogen (r = 0.709). Actinobacteria and Cyanobacteria had a moderate positive correlation with nitrate with r value (r = 0.673) and (r = 0.647), respectively. In this study, the metagenomics of the 16S rRNA gene amplicon by Illumina MiSeq has successfully identified the bacterial community assemblage in Putrajaya Lake and wetlands. Bacterial composition was associated with the availability of physicochemical properties of specific sites. The effectiveness of the engineered wetlands of Putrajaya in bioremediation was demonstrated by the marked decrease in certain nutrient concentrations from lake water input to the primary lake area.
