1. SLC35D3 promotes white adipose tissue browning to ameliorate obesity by NOTCH signaling

Abstract

Abstract White adipose tissue browning can promote lipid burning to increase energy expenditure and improve adiposity. Here, we show that Slc35d3 expression is significantly lower in adipose tissues of obese mice. While adipocyte-specific Slc35d3 knockin is protected against diet-induced obesity, adipocyte-specific Slc35d3 knockout inhibits white adipose tissue browning and causes decreased energy expenditure and impaired insulin sensitivity in mice. Mechanistically, we confirm that SLC35D3 interacts with the NOTCH1 extracellular domain, which leads to the accumulation of NOTCH1 in the endoplasmic reticulum and thus inhibits the NOTCH1 signaling pathway. In addition, knockdown of Notch1 in mouse inguinal white adipose tissue mediated by orthotopic injection of AAV8-adiponectin-shNotch1 shows considerable improvement in obesity and glucolipid metabolism, which is more pronounced in adipocyte-specific Slc35d3 knockout mice than in knockin mice. Overall, in this study, we reveal that SLC35D3 is involved in obesity via NOTCH1 signaling, and low adipose SLC35D3 expression in obesity might be a therapeutic target for obesity and associated metabolic disorders.

Abstract

Abstract Insulin resistance is associated with many pathological conditions, and an in-depth understanding of the mechanisms involved is necessary to improve insulin sensitivity. Here, we show that ZFYVE28 expression is decreased in insulin-sensitive obese individuals but increased in insulin-resistant individuals. Insulin signaling inhibits ZFYVE28 expression by inhibiting NOTCH1 via the RAS/ERK pathway, whereas ZFYVE28 expression is elevated due to impaired insulin signaling in insulin resistance. While Zfyve28 overexpression impairs insulin sensitivity and causes lipid accumulation, Zfyve28 knockout in mice can significantly improve insulin sensitivity and other indicators associated with insulin resistance. Mechanistically, ZFYVE28 colocalizes with early endosomes via the FYVE domain, which inhibits the generation of recycling endosomes but promotes the conversion of early to late endosomes, ultimately promoting phosphorylated insulin receptor degradation. This effect disappears with deletion of the FYVE domain. Overall, in this study, we reveal that ZFYVE28 is involved in insulin resistance by promoting phosphorylated insulin receptor degradation, and ZFYVE28 may be a potential therapeutic target to improve insulin sensitivity.

Abstract

The electro-hydraulic power source with an electro-hydraulic variable pump driven by a servo motor is suitable for electrified construction machinery. To achieve better energy efficiency in different working conditions, the multi-mode control scheme was proposed for the electro-hydraulic power source. The control scheme includes pressure control, flow control, and torque control modes. The switching rule among the three control modes was formulated based on the minimum pump pressure. The fuzzy PID controller was designed, and a composite flow regulation strategy was formulated, including the load-sensitive adaptive displacement regulation and servo motor variable speed regulation. The AMESim-Simulink co-simulation model of multi-mode control was established. The test platform was built, and the experimental study was carried out. The results indicate that the fuzzy PID control has a shorter response time and a more stable control effect compared with PID control. Additionally, the composite flow regulation strategy improves the flow regulation range by 36% and reduces the flow overshoot by 20% compared with the load-sensitive adaptive displacement regulation. As the main control valve received an opening step signal, the full flow regulation (7~81 L/min) of the power source took approximately 0.5 s to rise and 0.2 s to fall. The relative error of pressure difference for the main control valve was 0.63%. When receiving the pressure and torque step signal, the pump pressure and pump input torque both took approximately 0.45 s to rise and 0.2 s to fall. The relative errors of pump pressure and torque control were 0.2% and 0.16%, respectively. In the multi-mode control, the electro-hydraulic power source could switch smoothly between flow control mode, pressure control mode, and torque control mode. These results provide a reference for the multi-mode control of an electro-hydraulic power source with an electro-hydraulic variable pump driven by a servo motor.

Abstract

The physics potential of detecting ^8 B solar neutrinos will be exploited at the Jiangmen Underground Neutrino Observatory (JUNO), in a model-independent manner by using three distinct channels of the charged current (CC), neutral current (NC), and elastic scattering (ES) interactions. Due to the largest-ever mass of ^13 C nuclei in the liquid scintillator detectors and the expected low background level, ^8 B solar neutrinos are observable in the CC and NC interactions on ^13 C for the first time. By virtue of optimized event selections and muon veto strategies, backgrounds from the accidental coincidence, muon-induced isotopes, and external backgrounds can be greatly suppressed. Excellent signal-to-background ratios can be achieved in the CC, NC, and ES channels to guarantee the observation of the ^8 B solar neutrinos. From the sensitivity studies performed in this work, we show that JUNO, with 10 yr of data, can reach the 1 σ precision levels of 5%, 8%, and 20% for the ^8 B neutrino flux, ${\sin }^{2}{\theta }_{12}$ , and ${\rm{\Delta }}{m}_{21}^{2}$ , respectively. Probing the details of both solar physics and neutrino physics would be unique and helpful. In addition, when combined with the Sudbury Neutrino Observatory measurement, the world's best precision of 3% is expected for the measurement of the ^8 B neutrino flux.

Abstract

Abstract Main goal of the JUNO experiment is to determine the neutrino mass ordering using a 20 kt liquid-scintillator detector. Its key feature is an excellent energy resolution of at least 3% at 1 MeV, for which its instruments need to meet a certain quality and thus have to be fully characterized. More than 20,000 20-inch PMTs have been received and assessed by JUNO after a detailed testing program which began in 2017 and elapsed for about four years. Based on this mass characterization and a set of specific requirements, a good quality of all accepted PMTs could be ascertained. This paper presents the performed testing procedure with the designed testing systems as well as the statistical characteristics of all 20-inch PMTs intended to be used in the JUNO experiment, covering more than fifteen performance parameters including the photocathode uniformity. This constitutes the largest sample of 20-inch PMTs ever produced and studied in detail to date, i.e. 15,000 of the newly developed 20-inch MCP-PMTs from Northern Night Vision Technology Co. (NNVT) and 5000 of dynode PMTs from Hamamatsu Photonics K. K.(HPK).

6. A genome-wide association study identifies a novel association between SDC3 and apparent treatment-resistant hypertension

Abstract

Abstract Background Compared with patients who require fewer antihypertensive agents, those with apparent treatment-resistant hypertension (aTRH) are at increased risk for cardiovascular and all-cause mortality, independent of blood pressure control. However, the etiopathogenesis of aTRH is still poorly elucidated. Methods We performed a genome-wide association study (GWAS) in first cohort including 586 aTRHs and 871 healthy controls. Next, expression quantitative trait locus (eQTL) analysis was used to identify genes that are regulated by single nucleotide polymorphisms (SNPs) derived from the GWAS. Then, we verified the genes obtained from the eQTL analysis in the validation cohort including 65 aTRHs, 96 hypertensives, and 100 healthy controls through gene expression profiling analysis and real-time quantitative polymerase chain reaction (RT-qPCR) assay. Results The GWAS in first cohort revealed four suggestive loci (1p35, 4q13.2-21.1, 5q22-23.2, and 15q11.1-q12) represented by 23 SNPs. The 23 significant SNPs were in or near LAPTM5, SDC3, UGT2A1, FTMT, and NIPA1. eQTL analysis uncovered 14 SNPs in 1p35 locus all had same regulation directions for SDC3 and LAPTM5. The disease susceptible alleles of SNPs in 1p35 locus were associated with lower gene expression for SDC3 and higher gene expression for LAPTM5. The disease susceptible alleles of SNPs in 4q13.2-21.1 were associated with higher gene expression for UGT2B4. GTEx database did not show any statistically significant eQTLs between the SNPs in 5q22-23.2 and 15q11.1-q12 loci and their influenced genes. Then, gene expression profiling analysis in the validation cohort confirmed lower expression of SDC3 in aTRH but no significant differences on LAPTM5 and UGT2B4, when compared with controls and hypertensives, respectively. RT-qPCR assay further verified the lower expression of SDC3 in aTRH. Conclusions Our study identified a novel association of SDC3 with aTRH, which contributes to the elucidation of its etiopathogenesis and provides a promising therapeutic target.

Abstract

Abstract We study damping signatures at the Jiangmen Underground Neutrino Observatory (JUNO), a medium-baseline reactor neutrino oscillation experiment. These damping signatures are motivated by various new physics models, including quantum decoherence, ν 3 decay, neutrino absorption, and wave packet decoherence. The phenomenological effects of these models can be characterized by exponential damping factors at the probability level. We assess how well JUNO can constrain these damping parameters and how to disentangle these different damping signatures at JUNO. Compared to current experimental limits, JUNO can significantly improve the limits on τ 3/m 3 in the ν 3 decay model, the width of the neutrino wave packet σ x , and the intrinsic relative dispersion of neutrino momentum σ rel.

Abstract

ObjectiveThis study aims to investigate the composition and function of the gut microbiome in long-term depression using an 8-week chronic unpredictable mild stress (CUMS) rat model.Materials and methodsAnimals were sacrificed after either 4 weeks or 8 weeks under CUMS to mimic long-term depression in humans. The gut microbiome was analyzed to identify potential depression-related gut microbes, and the fecal metabolome was analyzed to detect their functional metabolites. The correlations between altered gut microbes and metabolites in the long-term depression rats were explored. The crucial metabolic pathways related to long-term depression were uncovered through enrichment analysis based on these gut microbes and metabolites.ResultsThe microbial composition of long-term depression (8-week CUMS) showed decreased species richness indices and different profiles compared with the control group and the 4-week CUMS group, characterized by disturbance of Alistipes indistinctus, Bacteroides ovatus, and Alistipes senegalensis at the species level. Additionally, long-term depression was associated with disturbances in fecal metabolomics. D-pinitol was the only increased metabolite in the 8-week CUMS group among the top 10 differential metabolites, while the top 3 decreased metabolites in the long-term depression rats included indoxyl sulfate, trimethylaminen-oxide, and 3 alpha,7 alpha-dihydroxy-12-oxocholanoic acid. The disordered fecal metabolomics in the long-term depression rats mainly involved the biosynthesis of pantothenate, CoA, valine, leucine and isoleucine.ConclusionOur findings suggest that the gut microbiome may participate in the long-term development of depression, and the mechanism may be related to the regulation of gut metabolism.

Naturally occurring oxides could react with zinc oxide (ZnO) nanoparticles (NPs) and then change its transformation and toxicity to ecological receptors. Abstract

Abstract The OSIRIS detector is a subsystem of the liquid scintillator filling chain of the JUNO reactor neutrino experiment. Its purpose is to validate the radiopurity of the scintillator to assure that all components of the JUNO scintillator system work to specifications and only neutrino-grade scintillator is filled into the JUNO Central Detector. The aspired sensitivity level of $$10^{-16}\hbox { g/g}$$ 10 - 16 g/g of $$^{238}\hbox {U}$$ 238 U and $$^{232}\hbox {Th}$$ 232 Th requires a large ( $$\sim 20\,\hbox {m}^3$$ ∼ 20 m 3 ) detection volume and ultralow background levels. The present paper reports on the design and major components of the OSIRIS detector, the detector simulation as well as the measuring strategies foreseen and the sensitivity levels to U/Th that can be reached in this setup.

Abstract

Abstract JUNO is a massive liquid scintillator detector with a primary scientific goal of determining the neutrino mass ordering by studying the oscillated anti-neutrino flux coming from two nuclear power plants at 53 km distance. The expected signal anti-neutrino interaction rate is only 60 counts per day (cpd), therefore a careful control of the background sources due to radioactivity is critical. In particular, natural radioactivity present in all materials and in the environment represents a serious issue that could impair the sensitivity of the experiment if appropriate countermeasures were not foreseen. In this paper we discuss the background reduction strategies undertaken by the JUNO collaboration to reduce at minimum the impact of natural radioactivity. We describe our efforts for an optimized experimental design, a careful material screening and accurate detector production handling, and a constant control of the expected results through a meticulous Monte Carlo simulation program. We show that all these actions should allow us to keep the background count rate safely below the target value of 10 Hz (i.e. ∼1 cpd accidental background) in the default fiducial volume, above an energy threshold of 0.7 MeV.

Abstract

Abstract Atmospheric neutrinos are one of the most relevant natural neutrino sources that can be exploited to infer properties about cosmic rays and neutrino oscillations. The Jiangmen Underground Neutrino Observatory (JUNO) experiment, a 20 kton liquid scintillator detector with excellent energy resolution is currently under construction in China. JUNO will be able to detect several atmospheric neutrinos per day given the large volume. A study on the JUNO detection and reconstruction capabilities of atmospheric $$\nu _e$$ ν e and $$\nu _\mu $$ ν μ fluxes is presented in this paper. In this study, a sample of atmospheric neutrino Monte Carlo events has been generated, starting from theoretical models, and then processed by the detector simulation. The excellent timing resolution of the 3” PMT light detection system of JUNO detector and the much higher light yield for scintillation over Cherenkov allow to measure the time structure of the scintillation light with very high precision. Since $$\nu _e$$ ν e and $$\nu _\mu $$ ν μ interactions produce a slightly different light pattern, the different time evolution of light allows to discriminate the flavor of primary neutrinos. A probabilistic unfolding method has been used, in order to infer the primary neutrino energy spectrum from the detector experimental observables. The simulated spectrum has been reconstructed between 100 MeV and 10 GeV, showing a great potential of the detector in the atmospheric low energy region.

Abstract

Abstract We present the calibration strategy for the 20 kton liquid scintillator central detector of the Jiangmen Underground Neutrino Observatory (JUNO). By utilizing a comprehensive multiple-source and multiple-positional calibration program, in combination with a novel dual calorimetry technique exploiting two independent photosensors and readout systems, we demonstrate that the JUNO central detector can achieve a better than 1% energy linearity and a 3% effective energy resolution, required by the neutrino mass ordering determination.

Abstract

High-spin states in 84Rb are studied by using the 70Zn(18O, p3n)84Rb reaction at a beam energy of 75 MeV. Three high-lying negative-parity bands are established, whose level spacings are very regular, i.e., there is no signature splitting. The dipole character of the transitions of these three bands is assigned by the γ-γ directional correlations of oriented states (DCO) intensity ratios and the multipolarity M1 is suggested by analogy with multiparticle excitations in neighboring nuclei. Strong M1 and weak or no E2 transitions are observed. All these characteristic features show they are magnetic rotational bands.

Abstract

We study the production of entropy in the context of a nonequilibrium chiral phase transition. The dynamical symmetry breaking is modeled by a Langevin equation for the order parameter coupled to the Bjorken dynamics of a quark plasma. We investigate the impact of dissipation and noise on the entropy and explore the possibility of reheating for crossover and first-order phase transitions, depending on the expansion rate of the fluid. The relative increase in S/N is estimated to range from 10% for a crossover to 100% for a first-order phase transition at low beam energies, which could be detected in the pion-to-proton ratio as a function of beam energy. 17. Large-Nc operator analysis of hyperon–nucleon interactions in SU(3) chiral effective field theory

Abstract

We study octet–octet baryon (JP=12+) contact interactions in SU(3) chiral effective field theory by using large-Nc operator analysis. By applying the 1/Nc expansion of the Hartree Hamiltonian, we find 15 operators in the octet–octet baryon potential where 4 operators for the leading order (LO) and 11 for the next-to-next-to-leading order (NNLO). The large-Nc operator analysis of octet–octet baryon matrix elements reduces the number of free parameters from 15 to 6 at the LO of the 1/Nc expansion. The application of large-Nc sum rules to the Jülich model of hyperon–nucleon (YN) interactions at the LO of the chiral expansion reduces the model parameters to 3 from 5 at the LO of 1/Nc expansion. We find that the values of LECs fitted to YN scattering data in Ref. [21] in the relativistic covariant ChEFT (EG) approach are more consistent with the predictions of the large-Nc analysis than in the heavy baryon (HB) formalism approach.

Abstract

The material, δ-MnO2, has exhibited superior performance on the removal of methylene blue (MB), but the process is significantly impacted by pH, and the impacting mechanism is still unclear. In this study, the effects of pH on the removal mechanism of MB using synthesized δ-MnO2 were investigated by distinguishing the adsorption and oxidation of MB by δ-MnO2 during the removal process in the dark. The results show that the total removal efficiency of MB by δ-MnO2 decreased significantly with an increase in the pH. MB could be removed by δ-MnO2 via an adsorption mechanism and oxidation mechanism, and the proportion of adsorptive removal and oxidative removal was different under different pH conditions. With an increase in the initial pH from 2.00 to 8.05, the redox potential of δ-MnO2 decreased, and its oxidation ability for the removal of MB also gradually decreased. In contrast, the surface negative charges of δ-MnO2 increased with an increase in the pH, and the adsorption ability towards positively charged MB also gradually increased. This indicates that the effects of pH on the removal of MB by δ-MnO2 are primarily dominated by its influence on the oxidation ability of δ-MnO2. In addition, it is further proved that the pH value has a significant effect on the oxidation and adsorption of MB on δ-MnO2. Moreover, the significant effects of pH on the oxidation of MB by δ-MnO2 are further demonstrated by observing the changes in Mn2+ and the UV-Vis spectra of intermediate products during the reaction, as well as the changes in the FTIR and XPS characterizations of δ-MnO2 after the reaction.

Abstract

With the intensification of human activities, arsenite (As(III)) pollutant from the soil and ground water has been a threat to human health, and the problem gradually becomes the focus of attention. In this study, the effects of several environment factors on As(III) removal ability of complex minerals are determined through the analysis of mineralogical characteristics of the complex synthesis of hematite and kaolin, using X-ray diffraction, Fourier transform infrared, and specific surface area. In the results, the XRD patterns of hematite and kaolin complexes indicate that the loading covers up some characteristic peaks of minerals in kaolin, which can be that hematite loading decrease the order of structure for some minerals. With increasing Fe content, the hydroxyl sites gradually increase, therefore strengthening the As(III) removal ability of complexes. With increasing the As(III) concentration, the removal ratios of kaolin on As(III) almost keep unchanged and that of complexes show no obvious regularity. However, with the pH increasing, the removal ratios of all samples keep increasing. Furthermore, the increasing of As(III) concentration and pH both improve As(III) removal amount significantly, especially for As(III) concentration. In addition, there is no forming of new mineral through it as observed by XRD. Therefore, hematite loading can promote the As(III) removal on kaolin through adsorption in different environments, which can provide a better method for the remediation of arsenic pollution.

Abstract

Iron-based heterogeneous Fenton systems have been widely proposed for recalcitrant contaminants degradation. However, low efficiency of H2O2 decomposition and Fe(II)/Fe(III) recycling quite limited their applications. In this work, Co-doped iron sulfide (FeS) was successfully prepared through a modified hydrothermal method, and characterized by XRD, FESEM, HRTEM, XPS and BET. With Co(II) substitution, FeS structure transformed from pyrrhotite to mackinawite. The 10% Co-FeS was highly effective in H2O2 activation toward Rhodamine B (RhB) degradation. The pseudo first-order kinetic constant of 10% Co-FeS was 0.323 min−1, which was 31.4 and 85 times higher than that of CoS2 and FeS, respectively. Quenching experiments and EPR analysis illustrated that •OH was the dominant radicals for RhB degradation and the role of Co(II) in H2O2 direct decomposition was negligible. Batch experiment suggested that the excellent catalytic performance of 10% Co-FeS were ascribed to: (i) Co incorporation into FeS structure facilitated electron transfer between S species and Fe3+, thus accelerating Fe(II)/Fe(III) conversion on the surface, and (ii) more active sites were exposed. The Co-doped FeS exhibited high stability with low Fe/Co leaching and great recyclability. This novel strategy brought potential insights to develop highly efficient catalyst like metal sulfides for wastewater treatment.

Abstract Recently, the wide application of CuO nanoparticles (NPs) in engineering field inevitably leads to its release into various geologic settings, which has aroused great concern about the geochemical behaviors of CuO NPs due to its high surface reactivity and impact on the fate of co-existing contaminants. However, the redox transformation of pollutants mediated by CuO NPs and the underlying mechanism still remain poorly understood. Here, we studied the interaction of CuO NPs with As(III), and explored the reaction pathways using batch experiments and multiple spectroscopic techniques. The results of in situ quick scanning X-ray absorption spectroscopy (Q-XAS) analysis verified that CuO NPs is capable of catalytically oxidize As(III) under dark conditions efficiently at a wide range of pHs. As(III) was firstly adsorbed on CuO NPs surface and then gradually oxidized to As(V) with dissolved O2 as the terminal electron acceptor. As(III) adsorption increased to the maximum at a pH close to PZC of CuO NPs (~ pH 9.2), and then sharply decreased with increasing pH, while the oxidation capacity monotonically increased with pH. X-ray photoelectron spectroscopy and electron paramagnetic resonance characterization of samples from batch experiments indicated that two pathways may be involved in As(III) catalytic oxidation: (1) direct electron transfer from As(III) to Cu(II), followed by concomitant re-oxidation of the produced Cu(I) by dissolved O2 back to Cu(II) on CuO NPs surface, and (2) As(III) oxidation by reactive oxygen species (ROS) produced from the above Cu(I) oxygenation process. These observations facilitate a better understanding of the surface catalytic property of CuO NPs and its interaction with As(III) and other elements with variable valence in geochemical environments.

Abstract

Breast carcinoma is the most common female cancer with considerable metastatic potential. Discovery of new therapeutic approaches for treatment of metastatic breast cancer is still needed. Here, we reported our finding with niclosamide, an FDA approved anthelmintic drug. The potency of niclosamide on breast cancer was assessed in vitro and in vivo. In this investigation, we found that niclosamide showed a dramatic growth inhibition against breast cancer cell lines and induced apoptosis of 4T1 cells in a dose-dependent manner. Further, Western blot analysis demonstrated the occurrence of its apoptosis was associated with activation of Cleaved caspases-3, down-regulation of Bcl-2, Mcl-1 and Survivin. Moreover, niclosamide blocked breast cancer cells migration and invasion, and the reduction of phosphorylated STAT3(Tyr705), phosphorylated FAK(Tyr925) and phosphorylated Src(Tyr416) were also observed. Furthermore, in our animal experiments, intraperitoneal administration of 20 mg/kg/d niclosamide suppressed 4T1 tumor growth without detectable toxicity. Histological and immunohistochemical analyses revealed a decrease in Ki67-positive cells, VEGF-positive cells and microvessel density (MVD) and an increase in Cleaved caspase-3-positive cells upon niclosamide. Notably, niclosamide reduced the number of myeloid-derived suppressor cells (MDSCs) in tumor tissues and blocked formation of pulmonary metastases. Taken together, these results demonstrated that niclosamide may be a promising candidate for breast cancer.

Abstract

The sperm-associated antigen 5 (SPAG5) is an important protein in mitosis and cell cycle checkpoint regulation, with more attention as a novel oncogene in various cancers. High level of SPAG5 expression has been detected in our clinical gastric cancer (GC) samples and The Cancer Genome Atlas GC data. However, the bio-function and potential mechanism of SPAG5 in GC remain unclear. In this study, we investigated the role of SPAG5 in GC development and the correlation between SPAG5 and 5-fluorouracil (5-FU) treatment. SPAG5 expression was increased in GC samples compared with that in normal tissues (80.8% vs. 22.0%), which was apparently associated with a worse outcome. Biological experiments showed that knockdown of SPAG5 induced apoptosis and suppressed proliferation in cells and animal models. Downregulation of SPAG5 enhanced the sensitivity of 5-FU in GC cells. Gene microarray chip identified 856 upregulated and 787 downregulated genes in SPAG5 silencing cells. Furthermore, 12 significant genes, including CDKN1A, CDKN1B, EIF4E, MAPK1, and HSP90B1, belonged to the PI3K/AKT signaling pathway using ingenuity pathway analysis. Meanwhile, real-time PCR and Western blotting results showed that knockdown of SPAG5 inhibited PI3K/AKT signaling pathway. Collectively, SPAG5 promotes the growth of GC cells by regulating PI3K/AKT signaling pathway, which could be the promising target gene in GC therapy.

Abstract

Hypoxia-inducible factor (HIF-1α), a core transcription factor responding to changes in cellular oxygen levels, is closely associated with a wide range of physiological and pathological conditions. However, its differential impacts on vascular cells types and molecular programs modulating human vascular homeostasis and regeneration remain largely elusive. Here, we applied CRISPR/Cas9-mediated gene editing of human embryonic stem cells (hESCs) and directed differentiation to generate HIF-1α-deficient human vascular cells including vascular endothelial cells (VECs), vascular smooth muscle cells (VSMCs), and mesenchymal stem cells (MSCs), as a platform for discovering cell type-specific hypoxia-induced response mechanisms. Through comparative molecular profiling across cell types under normoxic and hypoxic conditions, we provide insight into the indispensable role of HIF-1α in the promotion of ischemic vascular regeneration. We found human MSCs to be the vascular cell type most susceptible to HIF-1α deficiency, and that transcriptional inactivation of ANKZF1, an effector of HIF-1α, impaired pro-angiogenic processes. Altogether, our findings deepen the understanding of HIF-1α in human angiogenesis and support further explorations of novel therapeutic strategies of vascular regeneration against ischemic damage.

The $Λ$ baryon spectrum is studied within the SU(3) flavor symmetry in a constituent quark model. We found that it is rather difficult to accommodate some negative-parity $Λ$ resonances as single $q^2s$ ($q = u,\,d$ quarks) states in the conventional three-quark picture. The ground $q^3s\bar q$ pentaquark mass spectrum is evaluated and a possible interpretation is proposed in the work: the observed $Λ(1405)1/2^{-}$, $Λ(1670)1/2^{-}$ and $Λ(1800)1/2^{-}$ are three-state mixtures of two $p$-wave $q^2s$ states and one ground $q^3s\bar q$ pentaquark state, so are the $Λ(1520)3/2^{-}$, $Λ(1690)3/2^{-}$ and $Λ(2050)3/2^{-}$ resonances., 6 pages, 5 tables

Abstract

Thermoacoustic heat pumps are a promising heating technology that utilizes medium/low-grade heat to reduce reliance on electricity. This study proposes a single direct-coupled configuration for a thermoacoustic heat pump, aimed at minimizing system complexity and making it suitable for domestic applications. Numerical investigations were conducted under typical household heating conditions, including performance analysis, exergy loss evaluation, and axial distribution of key parameters. Results show that the proposed thermoacoustic heat pump achieves a heating capacity of 5.7 kW and a coefficient of performance of 1.4, with a heating temperature of 300 °C and a heat-sink temperature of 55 °C. A comparison with existing absorption heat pumps reveals favorable adaptability for large temperature lift applications. Abstract Direct reduction of unactivated alkyl halides for C(sp3)-N couplings under mild conditions presents a significant challenge in organic synthesis due to their low reduction potential. Herein, we introduce an in situ formed pyridyl-carbene-ligated copper (I) catalyst that is capable of abstracting halide atom and generating alkyl radicals for general C(sp3)-N couplings under visible light. Control experiments confirmed that the mono-pyridyl-carbene-ligated copper complex is the active species responsible for catalysis. Mechanistic investigations using transient absorption spectroscopy across multiple decades of timescales revealed ultrafast intersystem crossing (260 ps) of the photoexcited copper (I) complexes into their long-lived triplet excited states (>2 μs). The non-Stern-Volmer quenching dynamics of the triplets by unactivated alkyl halides suggests an association between copper (I) complexes and alkyl halides, thereby facilitating the abstraction of halide atoms via inner-sphere single electron transfer (SET), rather than outer-sphere SET, for the formation of alkyl radicals for subsequent cross couplings.

Abstract

Rationale: Hypertension is the most important risk factor for cardiovascular and cerebrovascular diseases. Getting deep insight into the pathogenesis of hypertension is necessary. Objective: To investigate the role of ANK (ankyrin) repeat domain 36 ( ANKRD36 ) in hypertension. Methods and Results: We first recruited an essential hypertension cohort and then performed genome-wide transcriptome analysis with peripheral blood mRNA. ANKRD36 was found to be significantly lower expressed in hypertension. The anchorin repeat domain mediates a variety of protein-protein interactions. The ENaC (epithelial sodium channel) genes expression was found upregulated in human umbilical vein endothelial cells with ANKRD36 knockdown by using Affymetrix expression profile chip. In human embryonic kidney epithelial cells and HEK293T (human embryonic kidney cells), ANKRD36 overexpression significantly downregulated expression of ENaC genes, and ANKRD36 knockdown upregulated their expression. The chromosome immunoprecipitation assay and yin yang 1 ( YY1 ) knockdown showed the expression of ENaC was regulated by ANKRD36 via YY1, a dual-function transcription factor ubiquitously expressed in human tissues. Immunocoprecipitation and fluorescence resonance energy transfer assay confirmed the interaction between ANKRD36 and YY1. The nucleo-cytoplasmic ratio of YY1 decreased when ANKRD36 was overexpressed and also increased when ANKRD36 was knocked down. ANK2 domain of ANKRD36 was critical to its interaction with YY1. Ankrd36 knockout mice showed higher blood pressure levels and Na + reabsorption, especially when fed with high-salt diet. Higher expression of ENaC genes was observed in renal tubular epithelial cells from the knockout mice, and Yy1 knockdown mitigated the alteration. Ankrd36 knockout mice also showed more sensitive response to ENaC inhibitor amiloride treatment. Conclusions: We identified that ANKRD36 was involved in blood pressure regulation by interacting with YY1 and then altering expression of ENaC genes. Lower expressed ANKRD36 in hypertension might be a potential therapeutic target, and the application of ENaC inhibitors on hypertension treatment might be extended when serum K + levels are closely monitored.

Abstract

Abnormal thyroid hormone (TH) function has been observed in all components of metabolic syndrome (MetS), but the mechanisms remain unclear. Altered genomic methylation status is closely related to MetS. Our aim was to determine whether methylation regulation in TH function–related genes is involved in MetS. In a small strictly selected cohort, low TH function was observed in MetS group, as well as lower THRB promoter methylation levels in peripheral blood leukocytes in a genome-wide methylation screening by Illumina 450K beadchip. The results of beadchip assay were then confirmed by Sequenom MassARRAY. Low THRB promoter methylation levels and low TH function in MetS were confirmed in another big-size validation cohort. Lower methylation levels were associated with higher THRB expression in peripheral blood leukocytes, and altered THRB promoter methylation status influenced its promoter activity and expression. In the MetS rat models constructed by high fat and high fructose diet, lower TH function was also observed, as well as lower Thrb promoter methylation levels. Furthermore, systematic inflammation observed in MetS was found to induce decreased THRB promoter methylation levels as well as corresponding THRB expression. Additionally, oral treatment with a physiological T3 dose mitigated hypertension and insulin resistance and partially alleviated hepatic steatosis and adipocyte hypertrophy in MetS rats. Low methylation levels of THRB promoter in peripheral blood leukocytes induced by systematic inflammation were involved in low TH function in MetS, whereas low TH function deteriorates MetS. This might serve as a novel therapeutic target of MetS.

Abstract

Hair loss affects millions of people at some time in their life, and safe and efficient treatments for hair loss are a significant unmet medical need. We report that topical delivery of quercetin (Que) stimulates resting hair follicles to grow with rapid follicular keratinocyte proliferation and replenishes perifollicular microvasculature in mice. We construct dynamic single-cell transcriptome landscapes over the course of hair regrowth and find that Que treatment stimulates the differentiation trajectory in the hair follicles and induces an angiogenic signature in dermal endothelial cells by activating HIF-1α in endothelial cells. Skin administration of a HIF-1α agonist partially recapitulates the pro-angiogenesis and hair-growing effects of Que. Together, these findings provide a molecular understanding for the efficacy of Que in hair regrowth, which underscores the translational potential of targeting the hair follicle niche as a strategy for regenerative medicine, and suggest a route of pharmacological intervention that may promote hair regrowth.

Abstract

With the intensification of human activities, arsenite (As(III)) pollutant from the soil and ground water has been a threat to human health, and the problem gradually becomes the focus of attention. In this study, the effects of several environment factors on As(III) removal ability of complex minerals are determined through the analysis of mineralogical characteristics of the complex synthesis of hematite and kaolin, using X-ray diffraction, Fourier transform infrared, and specific surface area. In the results, the XRD patterns of hematite and kaolin complexes indicate that the loading covers up some characteristic peaks of minerals in kaolin, which can be that hematite loading decrease the order of structure for some minerals. With increasing Fe content, the hydroxyl sites gradually increase, therefore strengthening the As(III) removal ability of complexes. With increasing the As(III) concentration, the removal ratios of kaolin on As(III) almost keep unchanged and that of complexes show no obvious regularity. However, with the pH increasing, the removal ratios of all samples keep increasing. Furthermore, the increasing of As(III) concentration and pH both improve As(III) removal amount significantly, especially for As(III) concentration. In addition, there is no forming of new mineral through it as observed by XRD. Therefore, hematite loading can promote the As(III) removal on kaolin through adsorption in different environments, which can provide a better method for the remediation of arsenic pollution.

49. Channel HCN4 mutation R666Q associated with sporadic arrhythmia decreases channel electrophysiological function and increases protein degradation

Abstract

Mutations in the hyperpolarization-activated nucleotide-gated channel 4 (HCN4) are known to be associated with arrhythmias in which QT prolongation (delayed ventricular repolarization) is rare. Here, we identified a HCN4 mutation, HCN4-R666Q, in two sporadic arrhythmia patients with sinus bradycardia, QT prolongation, and short bursts of ventricular tachycardia. To determine the functional effect of the mutation, we conducted clinical, genetic, and functional analyses using whole-cell voltage-clamp, qPCR, Western blot, confocal microscopy, and co-immunoprecipitation. The mean current density of HEK293T cells transfected with HCN4-R666Q was lower in 24 to 36 h after transfection and was much lower in 36 to 48 h after transfection relative to cells transfected with wildtype HCN4. Additionally, we determined that the HCN4-R666Q mutant was more susceptible to ubiquitin-proteasome system-mediated protein degradation than wildtype HCN4. This decreased current density for HCN4-R666Q could be partly rescued by treatment with a proteasome inhibitor. Therefore, we conclude that HCN4-R666Q had an effect on HCN4 function in two aspects, including decreasing the current density of the channel as a biophysical effect and weakening its protein stability. Our findings provide new insights into the pathogenesis of the HCN4-R666Q mutation.
