Pseudomonas plecoglossicida is one of the main pathogens causing visceral white spot disease of the large yellow croaker (Pseudosciaena crocea). Although antibiotics are used to control P. plecoglossicida infections, the long-term and large-scale use of antibiotics can lead to the development of drug-resistant bacteria as well as environmental pollution. Therefore, it is necessary to develop novel antibacterial agents to treat P. plecoglossicida infections. In this study, we first developed a two-step-centrifugation method to isolate live and dead P. plecoglossicida cells. Subsequently, we used the Systematic Evolution of Ligands by Exponential Enrichment (SELEX) screening based on inhibition rate to directly isolate antibacterial aptamers from the random library without post-processing. We isolated five antibacterial aptamers, namely B1, B2, B4, B8, and B109, which showed good inhibitory effects against P. plecoglossicida. Among these, B4 showed the highest inhibitory effect (62.40 ± 4.17 %). Further analysis revealed no positive correlation between the inhibitory effect of aptamers and their affinities with the target bacterium. The B4- and B109-binding proteins were isolated from P. plecoglossicida by magnetic separation and sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE). Mass spectrometry analysis revealed that the 26 kDa ribosomal protein L2 was the probable B4-binding protein, while the 26 kDa ribosomal protein S3 and 75 kDa ribosomal protein S1 or succinate dehydrogenase flavoprotein subunit were the probable B109-binding proteins. Structural and subcellular localization analyses of these potential B4- and B109-binding proteins were also conducted. Our findings suggest that the inhibitory activity of the antibacterial aptamers against P. plecoglossicida may be mediated via their interaction with the ribosomal proteins, which can interfere with the protein synthesis process in the bacterium, affecting its growth. These findings provide the scientific basis for the development of functional antibacterial aptamers and the elucidation of their mechanisms of action.

This work systematically reviews the complex mechanisms of CO2-water-rock interactions, microscopic simulations of reactive transport (dissolution, precipitation and precipitate migration) in porous media, and microscopic simulations of CO2-water-rock system. The work points out the key issues in current research and provides suggestions for future research. After injection of CO2 into underground reservoirs, not only conventional pressure-driven flow and mass transfer processes occur, but also special physicochemical phenomena like dissolution, precipitation, and precipitate migration. The coupling of these processes causes complex changes in permeability and porosity parameters of the porous media. Pore-scale microscopic flow simulations can provide detailed information within the three-dimensional pore and throat space and explicitly observe changes in the fluid-solid interfaces of porous media during reactions. At present, the research has limitations in the decoupling of complex mechanisms, characterization of differential multi-mineral reactions, precipitation generation mechanisms and characterization (crystal nucleation and mineral detachment), simulation methods for precipitation-fluid interaction, and coupling mechanisms of multiple physicochemical processes. In future studies, it is essential to innovate experimental methods to decouple “dissolution–precipitation–precipitate migration” processes, improve the accuracy of experimental testing of minerals geochemical reaction-related parameters, build reliable characterization of various precipitation types, establish precipitation-fluid interaction simulation methods, coordinate the boundary conditions of different physicochemical processes, and, finally, achieve coupled flow simulation of “dissolution−precipitation−precipitate migration” within CO2-water-rock systems.

Pseudomonas plecoglossicida can infect Pseudosciaena crocea and cause visceral white spot disease, resulting in substantial economic losses to the P. crocea industry. Pseudomonas plecoglossicida is a temperate-sensitive bacterium and only infects P. crocea below 22 ℃. Aptamers are oligonucleotide molecules selected by the systematic evolution of ligands by exponential enrichment (SELEX) and can identify all kinds of targets. In the present study, five aptamers (M17, M19, M48, M56 and M81) for P. plecoglossicida cultured at 18 ℃ were selected by SELEX and based on comparative analysis of the high frequency sequences. The five aptamers had significantly higher affinities for the pathogenic P. plecoglossicida at 18 ℃ than for the nonpathogenic P. plecoglossicida at 28 ℃ and the other six bacteria (Escherichia coli, Edwardsiella tarda, Aeromonas hydrophila, Vibiro harveryi, Vibiro alginolyticus, Pseudomonas aeruginosa) (P

Deoxyribozyme is a kind of DNA molecule with catalytic activity. It can be selected by systematic evolution of ligands by exponential enrichment (SELEX). This review focuses on the classification, selection and main applications of deoxyribozymes. According to the catalysis, deoxyribozymes can be classified into cleaved, ligated, phosphorylated and other types. Nowadays the cleaved and ligated types account for a large proportion, and nucleic acids are the main substrates of deoxyribozymes. Biotin-streptavidin method or polyacrylamide gel electrophoresis is often used to separate the catalytic DNA in selection of deoxyribozymes. Deoxyribozymes have good application prospect in metal ion detection, gene therapy and DNA molecular encryption system.

In order to solve the problem that the interference signal changes rapidly in the high dynamic environment and moves out of the null of the beam,resulting in the reduced performance of the anti-interference jamming algorithm,a robust beam forming algorithm based on null broadening and deepening was proposed.First set multiple virtual interference in the interference interval instead of a single interference,and then modified the covariance matrix by the covariance matrix forward and backward spatial smoothing technique.Simulation results show that the proposed algorithm can not only effectively widen and deepen the null in the direction of interference signal,which makes the array has a better output signal-to-noise ratio,but also maintain strong robustness when the interference changes rapidly.In the high dynamic environment,has a high array output when interference moves rapidly,the proposed algorithm has better performance and good robustness than other algorithms.It can provide a theoretical reference for strong interference suppression in the high dynamic environment.

The natural 15N abundance of ammonia (δ15N-NH3) emitted from livestock waste within a specific area (δ15N-NH3-LW) can be used to estimate its contribution to atmospheric NH3 through an isotope mixing model. However, the limited availability of reports on δ15N-NH3-LW hinders the accuracy of regional NH3 source partitioning. In this study, we conducted research in the suburbs of Shenyang, China, and combined our results (n = 71) with data from published literature (n = 56) to develop a δ15N-NH3 spectrum emitted from cattle, pig, sheep, and poultry waste. We also explored the national and global spatial distribution of δ15N-NH3-LW based on the proportion of NH3 emissions from these four types of livestock waste. Our results showed that the δ15N-NH3 values ranged from −59.9‰ to −7.7‰, with a mean value of −27.3‰. This value was significantly lower than that from non-agricultural sources, such as coal combustion and motor vehicle exhaust, but overlapped with values from fertilizer N volatilization. Specifically, the δ15N-NH3 values emitted from cattle, pig, sheep, and poultry waste were −32.1 ± 15.9‰, −20.7 ± 7‰, −24.3 ± 5.9‰, and −19.1 ± 4.9‰, respectively. Considering regional differences in livestock farming structures, δ15N-NH3-LW exhibited a gradual decreasing trend from Southeast to Northwest across China. For example, Fujian Province, located on the southeastern coast, exhibited the highest δ15N-NH3-LW value of −19.8‰, owing to its high proportion of poultry farming, while the Tibet Autonomous Region, in the western inland, displayed the lowest δ15N-NH3-LW value of −30.8‰, due to its high proportion of cattle farming. Globally, the δ15N-NH3-LW value was highest in Southeast Asia (−23.4‰) and lowest in East Africa (−30.4‰). Overall, our study revealed significant spatial variation in δ15N-NH3-LW at both national and global scales. To more accurately assess the regional contributions of NH3 emissions from livestock waste, it is essential to use region-specific δ15N-NH3-LW values for future NH3 source partitioning.

There is limited knowledge regarding the impact of nitrification inhibitors (NIs) and straw application on fertilizer N retention and in-season release. We conducted a trial to study the transformation of 15N-labeled urea in soils during the growing season of maize. To facilitate multiple destructive samplings throughout the season, we utilized a larger plot (25 m2) and a lower abundance 15N-fertilizer (1.193%) than usual. Soil extractable mineral N, mineral fixed ammonium, and organic N (ON) recovered 20 ± 21% (mean ± standard deviation), 6 ± 5%, and 25 ± 6% of the applied fertilizer N across three sampling stages of the growing season. On average, the bioavailability of fertilizer N in extractable mineral form was four times higher than that of mineral fixed ammonium. In contrast, fertilizer-derived ON represented a relatively stable N pool, maintaining high content throughout the growing period and becoming the major form (82%–93%) in the pool of total soil 15N at the physiological maturity stage of maize. Moreover, the co-application of nitrapyrin (a type of NI) significantly promoted fertilizer N storage in the ON form while the effect of straw was not significant. In conclusion, the NI-induced promotion of fertilizer-derived ON likely plays a critical role in storing fertilizer N for subsequent cultivations, rather than providing N nutrients for crop uptake during the current season.

Reducing fertilizer-N rate, applying a nitrification inhibitor (NI), and incorporating straw are widely recommended to improve N use efficiency of crops and decrease N losses. A field 15N tracer study was conducted to compare their effectiveness on fertilizer-N fates during the maize growing season in Northeast China. The following six treatments were used: (1) no N fertilization (control); (2) 200 kg urea-N ha−1 (100%N); (3) 200 kg urea-N ha−1 and straw (100%N + S); (4) 160 kg urea-N ha−1 (80%N); (5) 160 kg urea-N ha−1 and NI (Nitrapyrin in this study) (80%N + NI); and (6) 160 kg urea-N ha−1, NI, and straw (80%N + NI + S). The results showed that the five N fertilization treatments yielded 16–25% more grain and 39–60% more crop N uptake than the control, but the differences among the five treatments were not statistically significant. Compared with the 100%N, 20% fertilizer-N reduction (80%N) decreased the 15N concentration in topsoil and plant pools but increased the proportion of plant 15N recovery at harvesting (NUE15N, 60% vs. 50%). Compared with the 80%N, NI co-application (80%N + NI) delayed soil nitrification and increased soil 15N retention at harvesting (52% vs. 36%), thereby decreasing NUE15N significantly. Straw incorporation decreased fertilizer-N retention in soil compared with NI co-application because it promoted NUE15N significantly. In conclusion, the results demonstrate that NI and straw additions are efficient strategies for stabilizing fertilizer-N in soils and potentially minimizing N loss; however, their effects on NUE15N vary and the related mechanism must be further clarified in long-term trials.

Deforestation is one of the most serious environmental problems facing humankind. It continues to escalate rapidly across many regions of the world, thereby deteriorating the forest soil quality. This has prompted a large number of field-based studies aimed at understanding the impacts of deforestation on soil properties. However, the lack of comprehensive meta-analyses that utilized these studies has limited our deeper understanding of how different soil properties, including the soil organic carbon (SOC) pool, respond to deforestation. To address this critical knowledge gap, we conducted a meta-analysis of 144 studies to explore the impacts of deforestation on soil chemical, physical, and biological properties, with special emphasis on the long-term changes in SOC, such as concentrations, stocks, and sequestration. The results revealed that deforestation significantly decreased soil organic matter, electrical conductivity, and base saturation by 52%, 50%, and 98%, respectively. While deforestation increased soil total nitrogen content and decreased available phosphorus content by 51% and 99%, respectively, it resulted in slight decreases in some chemical properties, including soil pH (1%) and base cations (1%–13%). Deforestation significantly increased bulk density by 27% and soil erosion by 47%, but significantly decreased soil aggregate stability by 39% and saturated hydraulic conductivity by 63%. Soil microbial biomass C and N concentrations and enzyme activities were significantly decreased as a consequence of deforestation. Soil biological properties were much more affected by deforestation than soil physical and chemical properties. Regarding the SOC, the land use conversion from forest to pasture significantly increased SOC concentrations, stocks, and sequestration rates (11%–13%), whereas the land use conversions from forest to both plantation and cropland significantly decreased SOC concentrations, stocks, and sequestration rates (10%–43%). This observed decline in SOC accumulations decreased with increasing years after deforestation. The SOC dynamics following deforestation were predominantly regulated by microbial biomass concentrations, dehydrogenase activity, soil erosion, saturated hydraulic conductivity, aggregate stability, as well as concentrations of total organic carbon, total nitrogen, total phosphorus and organic matter. The present meta-analytical study provides compelling evidence that deforestation can induce profound changes in soil characteristics, including soil C contents, and has significant implications for soil health sustainability and climate change mitigation.

Abstract Denitrification and leaching nitrogen (N) losses are poorly constrained in Earth System Models (ESMs). Here, we produce a global map of natural soil 15N abundance and quantify soil denitrification N loss for global natural ecosystems using an isotope-benchmarking method. We show an overestimation of denitrification by almost two times in the 13 ESMs of the Sixth Phase Coupled Model Intercomparison Project (CMIP6, 73 ± 31 Tg N yr−1), compared with our estimate of 38 ± 11 Tg N yr−1, which is rooted in isotope mass balance. Moreover, we find a negative correlation between the sensitivity of plant production to rising carbon dioxide (CO2) concentration and denitrification in boreal regions, revealing that overestimated denitrification in ESMs would translate to an exaggeration of N limitation on the responses of plant growth to elevated CO2. Our study highlights the need of improving the representation of the denitrification in ESMs and better assessing the effects of terrestrial ecosystems on CO2 mitigation.

Societal Impact Statement Increasing atmospheric nitrogen (N) deposition represents a major global change factor, but its long‐term effect on tree growth and carbon (C) sequestration remains uncertain. Our manipulation experiment and meta‐analysis reveal that N deposition in temperate and boreal forests promoted tree growth and the allocation of more C into wood than into leaves in China and worldwide. Thus, N deposition may increase forest C sequestration through enhanced wood production and distribution of C into stable sinks. In the context of achieving “Carbon Neutrality,” understanding how N deposition affect long‐term forest C sinks will help us with mitigation strategies under climate change. Summary Increased nitrogen (N) deposition is driving many temperate and boreal forests in the Northern Hemisphere towards N saturation. However, it is uncertain how long‐term N deposition affects tree growth and carbon (C) allocation in these forests. To investigate this, we treated temperate larch and mixed forests in northeastern China with N additions for 8 years. In addition, we collected data from 25 N‐addition experiments in temperate and boreal forests worldwide to reveal the overall effects of N on tree growth and C allocation. Nitrogen additions significantly promoted total biomass increment by 24% in both study forests, with on average additional 8 kg C per kg N gain into woody biomass over the study period. Nitrogen additions increased the ratio of woody biomass increment to foliage litterfall production in the larch forest (by 34%). Literature data analysis also revealed greater N promotion on wood (24%) over foliage (9%) production. However, the positive effect on foliage diminished over time. These results combined imply that N deposition may promote tree growth in temperate and boreal regions and drive proportionally more photosynthate allocation into wood than leaves, thus may enhance forest C sequestration in the long run.

Abstract Background Forest succession is an important ecological process and has been studied for more than a century. However, changes in nitrogen (N) availability during succession remain unclear as they may lead to either N saturation or N limitation. Here, we propose a conceptual model to illustrate changes in N availability during four stages of secondary succession using the natural abundance of 15N in plant leaves (foliar δ15N). We predicted that N availability would decline in the early stages of succession and then increase in late stages, coinciding with the changes in foliar δ15N, with the inflection point varying in different climate zones. Data on foliar δ15N from 16 succession sequences were synthesized to explore changes in N availability during forest succession. Results The compiled data were consistent with the proposed conceptual model. Foliar δ15N in boreal and temperate forests decreased significantly in the first two stages of succession (estimated to last at least 66 years in temperate forests), at a rate of 0.18‰ and 0.38‰ per decade, respectively, and decreased slightly in tropical forests in the first 23 years. Foliar δ15N is projected to increase in later stages in all forests, which is supported by observations in both temperate and tropical forests. The inflection points of N availability when N limitation peaked during succession were different in different climate zones, implying different ecosystem N turnovers. Conclusions Our study reconciles the controversies regarding changes in N availability during forest secondary succession. Our findings are also useful for predicting the recovery of N and carbon accumulation during succession. Nonetheless, studies on forest secondary succession using foliar δ15N have thus far been limited, and more research should be conducted to further verify the conceptual model proposed here.

A study using paired 15N tracers shows atmospheric N deposited in oxidized form is more likely retained by trees, while the reduced form is retained in soil. The authors argue that this is a greater contribution of deposited N to the global forest C sink than previously reported.

Abstract Conversion of natural and secondary mixed forests to plantation monocultures can alter soil physical, chemical, and biological properties, which may lead to changes in ecosystem nitrogen (N) status by altering soil N availability. However, assessing and comparing N status between forest types within and across sites remain challenging because different indicators often yield contradicting conclusions. We developed a multiple‐indicator approach to evaluate N status across sites and between larch monocultures and mixed secondary forests paired at three sites (Daxinganling, Maoershan, and Qingyuan) in northeastern China. The selected indicators were foliar N content (%N), soil carbon‐to‐N ratio (C/N), soil inorganic N, foliar 15N natural abundance (δ15N), and δ15N of bulk soil, ammonium, and nitrate in the soil profiles. Our results, as shown mainly by C/N of soil organic layer and the fraction of nitrate in soil inorganic N, showed that N availability increased from north to south in the order Daxinganling

Abstract Background Conifers partition different N forms from soil, including ammonium, nitrate, and dissolved organic N (DON), to sustain plant growth. Previous studies focused on inorganic N sources and specific amino acid forms using 15N labelling, but knowledge of the contribution of DON to mature conifers’ N uptake is still scarce. Here, we quantified the contribution of different N forms (DON vs. NH4 + vs. NO3 −) to total N uptake, based on 15N natural abundance of plant and soil available N, in four mature conifers (Pinus koraiensis, Pinus sylvestris, Picea koraiensis, and Larix olgensis). Results DON contributed 31%, 29%, 28%, and 24% to total N uptake by Larix olgensis, Picea koraiensis, Pinus koraiensis, and Pinus sylvestris, respectively, whereas nitrate contributed 42 to 52% and ammonium contributed 19 to 29% of total N uptake for these four coniferous species. Conclusions Our results suggested that all four conifers could take up a relatively large proportion of nitrate, while DON was also an important N source for the four conifers. Given that DON was the dominant N form in study soil, such uptake pattern of conifers could be an adaptive strategy for plants to compete for the limited available N sources from soil so as to promote conifer growth and maintain species coexistence.

As nitrous oxide (N2O) is one of the most important greenhouse gases, N2O emission pathways and regulation techniques in soils with different vegetation types have become a research focus. Currently, a diverse array of research exists on the N2O emissions from soils of different vegetation types, e.g., forest, grassland, and agriculture. Few studies have investigated the microbial processes of N2O emissions from lawn soils. Fertilization levels in lawn soils are often similar to or much higher than those in agricultural ecosystems, thus fertilized lawn is an important source of atmospheric N2O. In the study, we employed the 15N-nitrate labelling method combined with the nitrification inhibition technique to distinguish microbial processes and their contribution to N2O emissions in long-term nitrogen fertilised lawns. We found that the N2O emission rate from the control treatment was 1.0 nmol g−1 h−1 over the incubation, with autotrophic nitrification contributing 60%. The N2O emission rate increased to 1.4 nmol g−1 h−1 from the soil treated with long-term N fertilization, and the contribution of autotrophic nitrification increased to 69%. N fertilization did not significantly increase the contribution of denitrification (24–26%) in the total N2O emissions. However, N fertilization substantially decreased the contribution of heterotrophic nitrification from 13 to 0.4% in the total N2O emissions. Co-denitrification to N2O was detected but the overall contribution was of minor importance (3–5%). The correlation analysis revealed that soil NO3− levels were the main influencing factors in the N2O producing microbial processes. Our results suggest that N fertilization altered both N2O production rates and the contribution pattern of microbial processes, and indicate the autotrophic nitrification and heterotrophic nitrification are more sensitive to N fertilization than denitrification and co-denitrification.

Abstract Nitrogen-containing species are major components in atmospheric aerosols. However, little is known about the sources of N-containing aerosols over high mountainous regions, especially for organic nitrogen (ON). This study aims to reveal the emission sources of both inorganic and organic nitrogen in tropospheric aerosols atop Mt. Tai, China, and to improve our understanding of the N cycle imbalance in the North China Plain (NCP). Total suspended particle (TSP) samples were collected on a daytime/nighttime basis in spring 2017 and were investigated for the concentrations and stable N isotopic compositions of total nitrogen, NH4 +, NO3 − and ON. Our results show that the concentrations of N-containing compounds were higher in daytime than nighttime, mainly resulting from mountain–valley breezes and the changes of planetary boundary layer height. However, no significant day/nighttime changes were found for their corresponding δ15N values, indicating similar contributions from different N sources between day and night. The MixSIAR Bayesian stable isotope mixing model results suggest that the most important emission source of NH3 for aerosol NH4 + was agriculture, followed by fossil fuel-related sources, human waste and biomass burning. Aerosol NO3 − was mainly formed from combustion and mobile emitted NO x . Interestingly, the isotopes of ON suggest that ON were very likely firstly of primary origin. Our study reveals the characteristics of reactive N emission sources and helps understand the regional transport of tropospheric N-containing aerosols in the NCP.

Massive stores of carbon and nutrients in permafrost could be released by global warming. Here the authors show that though warming across the Tibetan alpine permafrost region accelerates nitrogen liberation, contrary to expectations the elevated nutrients do not alleviate plant nitrogen limitation.

Excess nitrogen deposition has significant impacts on water eutrophication, soil acidification, eleveted greenhouse gas emissions, and biodiversity loss. These impacts mostly derive from conventional manipulative experiments in the field by adding nitrogen solution directly onto grassland or forest floors. For forest ecosystems, previous field experiments have usually ignored the nitrogen cycles in the canopy, which are important in responses to airborne nitrogen input. Although whole-forest canopy nitrogen fertilization has recently been conducted to promote our understanding of nitrogen deposition processes, spraying nitrogen solution onto plants still largely ignores the dry deposition of ammonia (as well as other gaseous reactive nitrogen species). To date, there have been a limited number of field studies that have investigated the bi-directional exchange of ammonia between the atmosphere and plants, not to mention the impacts of ammonia on natural ecosystems. Due to the increasing trend of atmospheric ammonia concentrations worldwide and its dominant role in nitrogen deposition and haze pollution, the next generation of experiments should mimick nitrogen deposition on natural ecosystems by further considering the dry deposition of ammonia.

Abstract Assessment of nitrogen (N) saturation of forests is critical to the evaluation of the manner in which ecosystems will respond to current and future global changes such as N deposition. However, quantifying N saturation remains a challenge. We developed a conceptual model of N saturation stages in forest ecosystems based on (1) a hypothetical relative rate of ammonification, nitrification, and denitrification, (2) concentrations of ammonium and nitrate in the soil, and (3) 15N enrichment pattern of bulk soil N, ammonium, and nitrate in the soil profile. We tested the hypotheses using data from forests located at five sites across eastern Asia. The fraction of nitrate in total inorganic N indicated that the sites represent an N saturation gradient with one boreal forest at stage 1 (least saturated), three temperate forests at stage 2, and one tropical forest at stage 3 (most saturated). The δ15N of bulk soil N increased from topsoil to subsoil more sharply at N‐limited sites than at the N‐rich sites along the N deposition gradient. We also found distinct 15N enrichment patterns of bulk soil N, ammonium, and nitrate in the soil profile across the study sites. At stage 1, nitrate was more 15N‐depleted than ammonium only in the organic soil horizon, indicating limited nitrification, while the 15N depletion of nitrate to ammonium was observed in the deeper mineral soil at stages 2 and 3. Furthermore, ammonium was more 15N‐depleted than bulk soil N at stages 1 and 2 but more 15N‐enriched than bulk soil N at stage 3. Our study suggests that soil profile patterns of δ15N of bulk soil N, ammonium, and nitrate provide information about the relative rates of mineralization, nitrification, and denitrification and thus can be an additional measure of N saturation of forest ecosystems across broad environmental gradients.

Forests worldwide store large quantities of carbon (C), particularly in soils as soil organic C (SOC). In northeastern China, two dominant forest types, secondary mixed forest (MF) and larch plantation forest (LF), cover extensive areas. However, we lack an understanding of the patterns and the mechanisms of SOC storage and stabilization in MF and LF, especially in deep soil layers. This research aims to illustrate the vertical distribution and mineral protection of SOC over soil depth; we also used δ13C values of soil fractions to evaluate SOC stability. Samples from the surface litter (Oi), organic layer (Oa+e), and 0–40 cm mineral soils were collected from both MF and LF plots. We used two different methods to separate bulk soils into distinguished fractions: (1) macro- and micro-aggregates and the non-aggregated fraction, and (2) particulate organic matter (POM) and mineral-associated organic matter (MAOM). The C concentrations, C stocks, and δ13C of all soil fractions were determined. Our findings were as follows: (1) SOC was mainly stored in mineral soils and was 13.6% lower in LF (8609 ± 1180 g C m−2) than MF (9969 ± 2084 g C m−2). (2) In both MF and LF, the SOC stock was mainly stored in aggregates (averaged 92.7%); macroaggregates dominated in the surface layers (Oa+e layer and 0–10 cm) but microaggregates dominated in the deep layers (10–20 cm and 20–40 cm). In mineral soils, MAOM was the dominant fraction of the C stock (averaged 81.6%). (3) The proportion of C distributed in microaggregates and MAOM increased from Oa+e to the 20–40 cm layer. (4) The C/N ratios and δ13C values of MAOM were smaller and heavier compared to those of POM. Our study demonstrated that in both forests, aggregate formation and mineral association predominantly contributed to SOC storage, and large stocks of SOC were distributed in the deep soil. The increasing proportion of SOC in microaggregates and MAOM along the soil depth, most likely derived from microbial turnover and microbial necromass, influenced SOC stability in both forest types.

Abstract Nitrogen stable isotope composition (δ15N) of nitrate (NO3−) deposition can aid in source apportionment of its precursor emissions, nitrogen oxides (NOx), with implications in mitigation policy to address serious air pollution. However, potential δ15N fractionation during atmospheric NO3− formation may hinder accurate quantification of NOx contributions. Previously, NOx photochemical reactions have been suggested to be the dominant δ15N fractionation process in NO3− formation. Here, we have quantified the potential fractionation effects associated with NOx photochemical reactions based upon δ15N‐NO3− measured in 1 year of daily‐based bulk deposition samples in Shenyang, a megacity with distinct seasonal fossil fuel combustion in northeastern China. The mean δ15N was 0.9 ± 4.0‰ and ranged from −4.9 to 8.3‰, with the lowest and highest values observed during summer and winter, respectively. Calculated NOx photochemical equilibrium fractionation may account for up to 42% of the observed seasonal δ15N change in NO3− deposition. However, the relative NOx source contribution trends, estimated based upon a δ15N Bayesian mixing model, were insensitive to NOx photochemical fractionation considerations. Overall, NOx source partitioning based upon averaged year‐long δ15N of bulk NO3− deposition estimated the following relative trend in NOx emission sources: coal combustion > biomass burning = vehicle emissions ≫ soil emissions. Seasonally, the increase of coal combustion emissions from summer to winter drives the seasonality of δ15N in NO3− deposition, indicating a necessity to control NOx emissions from coal combustion to improve wintertime air quality.

Abstract Nitrogen (N) fertilization has boosted grain production during the past century, while excess N fertilization with declining N use efficiency (NUE) has led to severe pollution in many regions. To achieve the goal of sustainable food production, Technologies and Management Practices (TMPs, e.g., optimum N application rate and methods, N‐transformation inhibitors) have been developed to improve crop yield and NUE and to reduce N losses. However, it remains unclear how N fate has been changed by environmental factors and TMPs. Here, we compiled a dataset of 366 field 15N tracer observations from 74 publications worldwide and conducted a meta‐analysis to examine how environmental conditions and management practices influence the fertilizer‐N fate one growing season after the fertilization of maize. We show that the proportion of 15N taken up by aboveground biomass (NUE15N), was significantly lower in China (33%) than that in North America (42%) and the European Union (54%). Soil organic carbon was the most critical environmental factor positively correlated with NUE15N. Among the nine selected categories of TMPs, deep placement of fertilizer and split application increased the grain yield and decreased the fertilizer‐N loss consistently among studies. Reducing the fertilizer‐N rate could increase the NUE15N but presents risks of yield reduction in some regions. This study demonstrates the importance of SOC and varying impacts of TMPs on the efficiency of fertilizer use in cropping systems globally and provides critical information for farmers and policymakers to improve N management for higher productivity and less pollution.

Rainfed agriculture is one of the most common farming practices in the world and is vulnerable to global climate change. However, only limited studies have been conducted on rainfed agriculture, mainly using low-frequency manual techniques, which caused large uncertainties in estimating annual N2O emissions. In this study, we used a fully automated system to continuously measure soil N2O emissions for two years (April 2017 to March 2019) in a typical rainfed maize field in Northeast China. The annual N2O emissions were 2.8 kg N ha−1 in year 1 (April 2017 to March 2018) and 1.8 kg N ha−1 in year 2 (April 2018 to March 2019), accounting for 1.9 and 1.2% of the nitrogen fertilizer applied, respectively. The inter-annual variability was mainly due to different weather conditions encountered in years 1 and 2. A severe drought in year 1 reduced plant N uptake, leaving high mineral N in the soil, and the following moderate rainfalls promoted a large amount of N2O emissions. The seasonal pattern of N2O fluxes was mainly controlled by soil temperature and soil nitrate concentration. Both soil moisture and the molar ratio of NO/N2O indicate that N2O and NO were mainly derived from nitrification, resulting in a significant positive correlation between N2O and NO flux in the intra-rows (where nitrogen fertilizer was applied). Moreover, we observed that the N2O emissions during the freeze–thaw periods were negligible in this region for rainfed agriculture. Our long-term and high-resolution measurements of soil N2O emissions suggest that sampling between LST 9:00 and 10:00 is the best empirical sampling time for the intermittent manual measurements.

The stable isotopes of nitrogen in nitrate archived in polar ice have been interpreted as reflecting a shift in reactive nitrogen sources or changes in atmospheric chemical reactivity. Here, we present a novel concentration and isotopic record of nitrate (δ15N-NO3−) from a central Tibetan Plateau ice core over the last ~200 years. We find that nitrate concentration increased from 6.0 ± 2.3 μeq/L (mean ± 1σ) in the preindustrial period (prior to 1900s) to 7.3 ± 2.7 μeq/L in post-1950. Over the same time period, the δ15N-NO3− decreased from 8.7 ± 3.7‰ to 4.2 ± 3.1‰, with much larger interannual variation in δ15N-NO3− during the preindustrial period. We present a useful framework for quantifying the sensitivity of the isotopic composition of atmospheric nitrate to changes in both sources and chemistry (gas and aerosol phase). After 1950, nitrogen deposition is primarily driven by fertilizer use, leading to significant increases in concentration and decreases in δ15N-NO3−. The large interannual variability of ice core δ15N-NO3− in the preindustrial reflects natural processes, namely the El Niño Southern Oscillation (ENSO) and dust events. Our results highlight a new connection between the nitrogen cycle and ENSO, and the overprinting of natural climate signals by recent anthropogenic increases in reactive nitrogen release. In China, nitrate (NO3−) becomes the main contributor to fine particles (PM2.5) because the emissions of its precursor, nitrogen oxides (NOx), were not recognized and controlled well in recent years. In this work, sources, conversion, and geographical origin of NOx were interpreted combining the isotopic information (δ15N and δ18O) of NO3− and dual modelling at five Chinese megacities (Beijing, Shanghai, Guangzhou, Wuhan and Chengdu) during 2013–2014. Results showed that the δ15N-NO3− values (n = 512) ranged from −12.3‰ to +22.9‰, and the average δ18O-NO3− value was +83.4‰ ± 17.2‰. The isotopic compositions both had a rising tendency as ambient temperature dropped, attributing largely to the source changes. Fundamental questions of how plant species within secondary forests and plantations in northeast China partition limited nitrogen (N) resource remain unclear. Here we conducted a 15N tracer greenhouse study to determine glycine, ammonium, and nitrate uptake by the seedlings of two coniferous species, Pinus koraiensis (Pinus) and Larix keampferi (Larix), and two broadleaf species, Quercus mongolica (Quercus) and Juglans mandshurica (Juglans), that are common in natural secondary forests in northeast China. Glycine contributed 43% to total N uptake of Pinus, but only 20, 11, and 21% to N uptake by Larix, Quercus, and Juglans, respectively (whole plant), whereas nitrate uptake was 24, 74, 88, and 68% of total uptake for these four species, respectively. Retention of glycine carbon versus nitrogen in Pinus roots indicated that 36% of glycine uptake was retained intact. Nitrate was preferentially used by Larix, Quercus, and Juglans, with nitrate uptake constituting 68∼88% of total N use by these three species. These results demonstrated that these dominant tree species in secondary forests in northeast China partitioned limited N resource by varying uptake of glycine, ammonium and nitrate, with all species, except Pinus, using nitrate that are most abundant within these soils. Such N use pattern may also provide potential underlying mechanisms for the higher retention of deposited nitrate than ammonium into aboveground biomass in these secondary forests.

Due to its significant ecological and climate consequences, atmospheric nitrogen (N) deposition is a growing global concern, especially in the severely N-polluted regions such as the Pearl River Delta (PRD) region of southern China. One-year measurements of reactive N species, including ammonium nitrogen ( -N), nitrate nitrogen (-N) and total organic nitrogen (ON) in dry and wet deposition, were conducted using an automated wet–dry sampler incorporated with a DDAS (dry deposition on aqueous surface) sampling device at Dinghushan (DHS), a natural forest site in the northwest of PRD and at Hengmen (HM), an estuary site in the south of PRD during 2006–2007. Total deposition fluxes of N at DHS and HM were up to 48.2 and 37.8 kg ha−1 yr−1, respectively, with most of the deposition occurring in the rainy season. Wet deposition was the dominant form, contributing 65–70% to the total deposition. -N was the largest contributor to the total N deposition at DHS (47%) due to significant influence of agriculture emissions. ON was the most important N component at HM (41%), which is probably attributed to the marine sources. However, -N deposition is increasing rapidly recently and is expected to be more important in the near future. The current N deposition level in PRD is much higher than those in Europe and North America. Great challenges exist in reducing reactive N emission in this region. Thus, a scenario of rising N deposition in PRD in the near future cannot be ruled out. The environmental consequences due to elevated N deposition should therefore be paid more attention in the future.
