Abstract

In this paper, we study the stabilizer-free weak Galerkin methods on polytopal meshes for a class of second order elliptic boundary value problems of divergence form and with gradient nonlinearity in the principal coefficient. With certain assumptions on the nonlinear coefficient, we show that the discrete problem has a unique solution. This is achieved by showing that the associated operator satisfies certain continuity and monotonicity properties. With the help of these properties, we derive optimal error estimates in the energy norm. We present several numerical examples to verify the error estimates.

2. A Wide-Band High-Resolution Transmitter for Optical Isolation Amplifier

Abstract

A wide-band and high-resolution transmitter of optical isolation amplifier is proposed for switching power supply isolation and servo motor drive applications. The transmitter is based on the chopper-stabilized technique and sigma delta modulator. A built-in common-mode voltage circuit of switched capacitor is proposed to ensure the stability of input DC common mode. Meanwhile, the feedback capacitor is used to improve the driving ability, which helps to avoid a buffer with large input swing. The circuit is tapeout with GF CMOS 0.18 μm 1P6M process with 5 V power supply. The test results show that in 5 V supply voltage, the input swing of transmitter is 2 V and the effective signal bandwidth is 110 kHz. And the output resolution achieves 11 bit.

Abstract

Morroniside is known to improve osteoporosis by promoting osteoblastogenesis. The activation of PI3K/Akt/mTOR signaling is a significant mechanism in morroniside-promoted osteoblastogenesis. It is well known that protective autophagy is an important factor in osteoblastogenesis. However, the activation of mTOR signaling can inhibit autophagy. This study aimed to investigate the relationship between mTOR signaling and autophagy in morroniside-regulated osteoblastogenesis. In this study, we investigated the effect of morroniside on the autophagic activity (LC3 conversion rate, LC3-puncta formation, and autophagosome number) of differentiated osteoblast precursors (MC3T3-E1 cells). Then, we identified the roles of mTOR knockdown in morroniside-regulated alterations of autophagy and osteogenic parameters in MC3T3-E1 cells. Next, mTOR knockdown and overexpression were used to observe the roles of mTOR in morroniside-regulated alterations of autophagic molecules (Atg7, Atg13, and Beclin1). Subsequently, the additional value of the above autophagic molecules on morroniside-regulated osteogenic parameters in MC3T3-E1 cells was analyzed based on lentiviral transduction. Finally, combined with morroniside and TAT-Beclin1, the roles of Beclin1 upregulation in the in vivo effects of morroniside was investigated. Our experimental data showed that morroniside promoted both the mTOR activity and autophagy in MC3T3-E1 cells. Morroniside-upregulated autophagic activity and Atg13 or Beclin1 protein level in MC3T3-E1 cells were enhanced by mTOR knockdown. Furthermore, Morroniside-upregulated Atg13 and Beclin1 expression was reversed by mTOR overexpression. Importantly, autophagy upregulation with overexpression of the autophagic gene, Atg13 or BECN1 (gene form of Beclin1), significantly promoted osteoblastogenesis regulated by morroniside. The promotional effect of morroniside on bone microarchitecture, bone mass, and bone parameters (including trabecular bone area and OCN expression in trabecular bone) in ovariectomized (OVX) mice was enhanced by TAT-Beclin1 administration. In conclusion, the autophagy-enhancing drugs related to Beclin1 or Atg13 may be an effective adjuvant therapy in the treatment of osteoporosis with morroniside.

Abstract

RANKL induces osteoclastogenesis via JNK1 signal that exerts an anti-apoptotic effect during osteoclastogenesis. However, the classic downstream c-Jun/AP-1 pathway is not included in anti-apoptosis of JNK1. Thus, the detail mechanism underlying JNK1-resisted apoptosis remains unknown during RANKL-induced osteoclastogenesis. RANKL-induced autophagy results in the degradation of the osteoclastogenesis-inhibitor TRAF3, and TRAF3 is thought as a regulator of apoptosis. Given the key effect of JNK1 in mediating autophagy, our study aims to investigate the significance of TRAF3 in bridging JNK1-mediated autophagy and apoptotic resistance during osteoclastogenesis. In this study, by using Bone Marrow-derived macrophages (BMMs) as osteoclast precursors (OCPs), we found that RANKL-induced TRAF3 degradation was significantly suppressed by JNK inhibitor (SP600125), which was restored by overexpression of Beclin1 (key autophagic protein). Nevertheless, TRAF3 silencing partially reversed the reduced osteoclastogenesis under SP600125 intervention. Besides, OCP apoptosis was positively regulated by TRAF3 overexpression, regardless of the application of autophagy inhibitor or SP600125. Remarkably, the enhanced apoptosis caused by the pharmacological inhibition of Beclin1 was reversed by TRAF3 silencing. Together, these results suggest that JNK1-mediated autophagy could promote RANKL-induced osteoclastogenesis via enhancing TRAF3 degradation. Importantly, JNK1 could prevent OCP apoptosis through autophagy-TRAF3 signaling, which provides more potential targets for clinical treatment of pathological bone loss.

Abstract

We consider a linear algebra approach to establishing a discrete comparison principle for a nonmonotone class of quasilinear elliptic partial differential equations. In the absence of a lower order term, we require local conditions on the mesh to establish the comparison principle and uniqueness of the piecewise linear finite element solution. We consider the assembled matrix corresponding to the linearized problem satisfied by the difference of two solutions to the nonlinear problem. Monotonicity of the assembled matrix establishes a maximum principle for the linear problem and a comparison principle for the nonlinear problem. The matrix analysis approach to the discrete comparison principle yields sharper constants and more relaxed mesh conditions than does the argument by contradiction used in previous work., 17 pages; 1 figure

Abstract

Objectives Oestrogen is known to inhibit osteoclastogenesis, and numerous studies have identified it as an autophagic activator. To date, the role of oestrogen in the autophagy of osteoclast precursors (OCPs) during osteoclastogenesis remains unclear. This study aimed to determine the effect of autophagy regulated by the biologically active form of oestrogen (17β‐estradiol) on osteoclastogenesis. Materials and methods After treatment with 17β‐estradiol in OCPs (from bone marrow‐derived macrophages, BMMs) and ovariectomy (OVX) mice, we measured the effect of 17β‐estradiol on the autophagy of OCPs in vitro and in vivo. In addition, we studied the role of autophagy in the OCP proliferation, osteoclast differentiation and bone loss regulated by 17β‐estradiol using autophagic inhibitor or knock‐down of autophagic genes. Results The results showed that direct administration of 17β‐estradiol enhanced the autophagic response of OCPs. Interestingly, 17β‐estradiol inhibited the stimulatory effect of receptor activator of nuclear factor‐κB ligand (RANKL) on the autophagy and osteoclastogenesis of OCPs. Moreover, 17β‐estradiol inhibited the downstream signalling of RANKL. Autophagic suppression by pharmacological inhibitors or gene silencing enhanced the inhibitory effect of 17β‐estradiol on osteoclastogenesis. In vivo assays showed that the autophagic inhibitor 3‐MA not only inhibited the autophagic activity of the OCPs in the trabecular bone of OVX mice but also enhanced the ability of 17β‐estradiol to ameliorate bone loss. Conclusions In conclusion, our study showed that oestrogen directly enhanced the autophagy of OCPs, which inhibited its anti‐osteoclastogenic effect. Drugs based on autophagic inhibition may enhance the efficacy of oestrogen on osteoporosis.

Abstract

Arnold, Falk, and Winther [Bull. Amer. Math. Soc. 47 (2010), 281--354] showed that mixed variational problems, and their numerical approximation by mixed methods, could be most completely understood using the ideas and tools of Hilbert complexes. This led to the development of the Finite Element Exterior Calculus (FEEC) for a large class of linear elliptic problems. More recently, Holst and Stern [Found. Comp. Math. 12:3 (2012), 263--293 and 363--387] extended the FEEC framework to semi-linear problems, and to problems containing variational crimes, allowing for the analysis and numerical approximation of linear and nonlinear geometric elliptic partial differential equations on Riemannian manifolds of arbitrary spatial dimension, generalizing surface finite element approximation theory. In this article, we develop another distinct extension to the FEEC, namely to parabolic and hyperbolic evolution systems, allowing for the treatment of geometric and other evolution problems. Our approach is to combine the recent work on the FEEC for elliptic problems with a classical approach to solving evolution problems via semi-discrete finite element methods, by viewing solutions to the evolution problem as lying in time-parameterized Hilbert spaces (or Bochner spaces). Building on classical approaches by Thomee for parabolic problems and Geveci for hyperbolic problems, we establish a priori error estimates for Galerkin FEM approximation in the natural parametrized Hilbert space norms. In particular, we recover the results of Thomee and Geveci for two-dimensional domains and lowest-order mixed methods as special cases, effectively extending their results to arbitrary spatial dimension and to an entire family of mixed methods. We also show how the Holst and Stern framework allows for extensions of these results to certain semi-linear evolution problems., Comment: 25 pages. The search for the novel anti-OS agent is extremely urgent and important. Our previous study has proposed that salinomycin is a novel anti-OS agent. Here we characterized DNA-dependent protein kinase catalytic subunit (DNA-PKcs) as a primary salinomycin resistance factor in OS cells. DNA-PKcs inhibitors (NU7026, NU7441 and LY294002) or DNA-PKcs shRNA knockdown dramatically potentiated salinomycin-induced death and apoptosis of OS cells (U2OS and MG-63 lines). Further, forced-expression of microRNA-101 (“miR-101”) downregulated DNA-PKcs and augmented salinomycin’s cytotoxicity against OS cells. Reversely, over-expression of DNA-PKcs in OS cells inhibited salinomycin’s lethality. For the mechanism study, we show that DNA-PKcs is required for salinomycin-induced pro-survival autophagy activation. DNA-PKcs inhibition (by NU7441), shRNA knockdown or miR-101 expression inhibited salinomycin-induced Beclin-1 expression and autophagy induction. Meanwhile, knockdown of Beclin-1 by shRNA significantly sensitized salinomycin-induced OS cell lethality. In vivo , salinomycin administration suppressed U2OS xenograft tumor growth in severe combined immuno-deficient (SCID) mice, and its anti-tumor activity was dramatically potentiated with co-administration of the DNA-PKcs inhibitor NU7026. Together, these results suggest that DNA-PKcs could be a primary resistance factor of salinomycin in OS cells. DNA-PKcs inhibition or silence may thus significantly increase salinomycin’s sensitivity in OS cells.

Abstract

Clinical and radiographic outcomes of multilevel cervical myelopathy (MCM) patients treated with a hybrid technique combining cervical anterior corpectomy and fusion (ACF) with cervical artificial disc replacement (C-ADR) were evaluated. A total of 23 patients including 14 females and 9 males (mean age, 48.3 years) were treated with the hybrid technique and they were followed up for an average time of 35 months (range from 24 to 40 months). Arm- and neck-pain visual analogue scale (VAS) scores, neck disability index (NDI) scores and C2-7 range of motion (ROM) preoperation and 6 weeks, 3 and 24 months postoperation were assessed and compared. Fusion success and system failure based on anteroposterior (AP), lateral and flexion/extension X-rays were examined by an independent reviewer. In this paper, we extend some of the multilevel convergence results obtained by Xu and Zhu in [Xu and Zhu, M3AS 2008], to the case of second order linear reaction-diffusion equations. Specifically, we consider the multilevel preconditioners for solving the linear systems arising from the linear finite element approximation of the problem, where both diffusion and reaction coefficients are piecewise-constant functions. We discuss in detail the influence of both the discontinuous reaction and diffusion coefficients to the performance of the classical BPX and multigrid V-cycle preconditioners., 25 pages, 7 figures. Abstract

Recent studies have identified that constitutively active phosphatidylinositol 3-kinase (PI3K)/Akt/mammalian target of rapamycin (mTOR) signaling is an important feature of osteosarcoma, where it promotes cell proliferation, survival, and chemo-resistance. Here, we studied the therapeutic potential of NVP-BEZ235, a novel dual PI3K/mTOR dual inhibitor, on osteosarcoma cells in vivo and in vitro. NVP-BEZ235 was cytotoxic and cytostatic to a panel of osteosarcoma lines (MG-63, U2OS and SaOs-2), where it induce apoptosis and cell-cycle arrest. At the molecular level, NVP-BEZ235 inhibited PI3K-AKT-mTORC1 activation and downregulated cyclin D1/cyclin B1 expressions, while increasing MEK/Erk phosphorylation in osteosarcoma cells. MEK/Erk inhibitors PD98059 and MEK-162 increased NVP-BEZ235 activity on osteosarcoma cells. In vivo, oral NVP-BEZ235 inhibited U2OS xenograft in SCID mice, and its anti-tumor efficiency was further enhanced by MEK-162 co-administration. Taken together, our findings indicate that dual inhibition of PI3K and mTOR with NVP-BEZ235, either alone or in combination with MEK/Erk inhibitors, may be an efficient treatment for osteosarcoma.

Abstract

Comparison principles are developed for discrete quasilinear elliptic partial differential equations. We consider the analysis of a class of nonmonotone Leray-Lions problems featuring both nonlinear solution and gradient dependence in the principal coefficient, and a solution dependent lower-order term. Sufficient local and global conditions on the discretization are found for piecewise linear finite element solutions to satisfy a comparison principle, which implies uniqueness of the solution. For problems without a lower-order term, our analysis shows the meshsize is only required to be locally controlled, based on the variance of the computed solution over each element. We include a discussion of the simpler semilinear case where a linear algebra argument allows a sharper mesh condition for the lower order term., 20 pages

Here, we aim to provoke AMPK activation via microRNA silencing its phosphatase Ppm1e (protein phosphatase Mg 2+ /Mn 2+ -dependent 1e). We showed that microRNA-135b-5p (“miR-135b-5p”), the anti-Ppm1e microRNA, was significantly downregulated in human osteoblastoma tissues. It was correlated with Ppm1e upregulation and AMPKα1 de-phosphorylation. Forced-expression of miR-135b-5p in human osteoblastoma cells (MG-63 and U2OS lines) silenced Ppm1e, and induced a profound AMPKα1 phosphorylation (at Thr-172). Osteoblastoma cell proliferation was inhibited after miR-135b-5p expression. Intriguingly, Ppm1e shRNA knockdown similarly induced AMPKα1 phosphorylation, causing osteoblastoma cell proliferation. Reversely, AMPKα1 shRNA knockdown or dominant negative mutation almost abolished miR-135b-5p’s actions in osteoblastoma cells. Further in vivo studies demonstrated that U2OS tumor growth in mice was dramatically inhibited after expressing miR-135b-5p or Ppm1e shRNA. Together, our results suggest that miR-135b-induced Ppm1e silence induces AMPK activation to inhibit osteoblastoma cell proliferation.

Abstract

The search for novel and more efficient chemo-agents against malignant osteoblastoma is important. In this study, we examined the potential anti-osteoblastoma function of bufotalin, and studied the underlying mechanisms. Our results showed that bufotalin induced osteoblastoma cell death and apoptosis in dose- and time-dependent manners. Further, bufotalin induced endoplasmic reticulum (ER) stress activation in osteoblastoma cells, the latter was detected by the induction of C/EBP homologous protein (CHOP), phosphorylation of inositol-requiring enzyme 1 (IRE1) and PKR-like endoplasmic reticulum kinase (PERK), as well as caspase-12 activation. Conversely, the ER stress inhibitor salubrinal, the caspase-12 inhibitor z-ATAD-fmk as well as CHOP depletion by shRNA significantly inhibited bufotalin-induced osteoblastoma cell death and apoptosis. Finally, by using a mice xenograft model, we demonstrated that bufotalin inhibited U2OS osteoblastoma cell growth in vivo. In summary, our results suggest that ER stress contributes to bufotalin-induced apoptosis in osteoblastoma cells. Bufotalin might be investigated as a novel anti-osteoblastoma agent.

Abstract

We describe a new method of identifying night-time clouds over the Pierre Auger Observatory using infrared data from the Imager instruments on the GOES-12 and GOES-13 satellites. We compare cloud. identifications resulting from our method to those obtained by the Central Laser Facility of the Auger Observatory. Using our new method we can now develop cloud probability maps for the 3000 km(2) of the Pierre Auger Observatory twice per hour with a spatial resolution of similar to 2.4 km by similar to 5.5 km. Our method could also be applied to monitor cloud cover for other ground-based observatories and for space-based observatories. (C) 2013 Elsevier B.V. Abstract

We introduce and analyze two-level and multi-level preconditioners for a family of Interior Penalty (IP) discontinuous Galerkin (DG) discretizations of second order elliptic problems with large jumps in the diffusion coefficient. Our approach to IPDG-type methods is based on a splitting of the DG space into two components that are orthogonal in the energy inner product naturally induced by the methods. As a result, the methods and their analysis depend in a crucial way on the diffusion coefficient of the problem. The analysis of the proposed preconditioners is presented for both symmetric and non-symmetric IP schemes; dealing simultaneously with the jump in the diffusion coefficient and the non-nested character of the relevant discrete spaces presents extra difficulties in the analysis which precludes a simple extension of existing results. However, we are able to establish robustness (with respect to the diffusion coefficient) and nearly-optimality (up to a logarithmic term depending on the mesh size) for both two-level and BPX-type preconditioners. Following the analysis, we present a sequence of detailed numerical results which verify the theory and illustrate the performance of the methods. Abstract

We report a case of primary empty sella syndrome (ESS) resulting in osteoporotic fractures and persistent non-fusion of the hand epiphyses, and discuss the potential pathogenesis of this disease. A 41-year-old man presented with pain in the right hand and back after a fall. X-radiographs revealed persistent epiphyses and severe osteoporosis. Serum phosphorus and prolactin levels were above normal levels, and free triiodothyronine, free thyroxine and testosterone levels were below normal limits. Magnetic resonance imaging of the head revealed empty sella. A lumbar bone mineral density examination indicated severe osteoporosis. ESS caused a systemic hormone disorder in this patient, resulting in osteoporotic fractures and persistent non-fusion of the hand epiphyses. Possible causes of this anomaly are chronic or congenital abnormities of the pituitary gland.

In this article, we consider two-grid finite element methods for solving semilinear interface problems in d space dimensions, for d = 2 or d = 3. We consider semilinear problems with discontinuous diffusion coefficients, which includes problems containing subcritical, critical, and supercritical nonlinearities. We establish basic quasioptimal a priori error estimates for Galerkin approximations. We then design a two-grid algorithm consisting of a coarse grid solver for the original nonlinear problem, and a fine grid solver for a linearized problem. We analyze the quality of approximations generated by the algorithm and show that the coarse grid may be taken to have much larger elements than the fine grid, and yet one can still obtain approximation quality that is asymptotically as good as solving the original nonlinear problem on the fine mesh. © 2013 Wiley Periodicals, Inc. Abstract

The goal of this paper is to design optimal multilevel solvers for the finite element approximation of second order linear elliptic problems with piecewise constant coefficients on bisection grids. Local multigrid and BPX preconditioners are constructed based on local smoothing only at the newest vertices and their immediate neighbors. The analysis of eigenvalue distributions for these local multilevel preconditioned systems shows that there are only a fixed number of eigenvalues which are deteriorated by the large jump. The remaining eigenvalues are bounded uniformly with respect to the coefficients and the meshsize. Therefore, the resulting preconditioned conjugate gradient algorithm will converge with an asymptotic rate independent of the coefficients and logarithmically with respect to the meshsize. SUMMARY In this paper, we present a multigrid V-cycle preconditioner for the linear system arising from piecewise linear nonconforming Crouzeix–Raviart discretization of second-order elliptic problems with jump coefficients. The preconditioner uses standard conforming subspaces as coarse spaces. We showed that the convergence rates of the (multiplicative) two-grid and multigrid V-cycle algorithms will deteriorate rapidly because of large jumps in coefficient. However, the preconditioned systems have only a fixed number of small eigenvalues depending on the large jump in coefficient, and the effective condition numbers are independent of the coefficient and bounded logarithmically with respect to the mesh size. As a result, the two-grid or multigrid preconditioned conjugate gradient algorithm converges nearly uniformly. We also comment on some major differences of the convergence theory between the nonconforming case and the standard conforming case. Numerical experiments support the theoretical results. Abstract

The surface topography plays an important role in bone formation, which may present specific physical cues to cells and tissues and further improve the bioactivity or osteoconduction of the implants. In this report, nano-hydroxyapatite/silk fibroin (n-HA/SF) composite were prepared via a simplified coprecipitation method. Then these nanocrystals were deposited onto Ti alloy substrates via a template-assisted electrostatic spraying deposition (ESD) technique. The output voltage and flow rate were investigated to produce a cone-jet spraying of n-HA/SF suspensions. By using different types of precisely-designed templates (stripe, grid, and ring), the Ti alloy substrates with different micropatterns (stripe, square, and ring) composed of n-HA/SF composite were created. Our results demonstrated that the precisely-designed micropatterns of n-HA/SF composite onto Ti and its alloy substrates could be easily achieved by using template-assisted ESD.

Abstract

Mesoporous hydroxyapatite (meso-HA) thin films were fabricated by a sol–gel method using cetyltrimethyl ammonium bromide as the template. The phase, surface morphology, and mesoporous structure of the meso-HA films were characterized by X-ray diffraction, scanning electron microscopy, and transmission electron microscopy. The uniform thin films consisted of meso-HA spherical particles with different pore size (2.0 or 3.1 nm) were prepared on the glass substrate at different pH value (pH 3.0 or 7.0). The introduction of mesopores would enhance the surface area of HA. Water contact angle was also measured on the non-mesoporous and meso-HA thin films, revealing the promotion of surface wettability in the mesoporous ones. In vitro cytocompatibility of HA films were evaluated by cell adhesion and proliferation tests using MC3T3-E1 cells. After 3 days of culture on the samples, the cells spread in an elongated shape and were well adhered to the surface of the meso-HA films. Moreover, the cells proliferation on the meso-HA films was higher than that on the non-mesoporous films. Abstract

We develop an efficient and reliable adaptive finite element method (AFEM) for the nonlinear Poisson-Boltzmann equation (PBE). We first examine the regularization technique of Chen, Holst, and Xu; this technique made possible the first a priori pointwise estimates and the first complete solution and approximation theory for the Poisson-Boltzmann equation. It also made possible the first provably convergent discretization of the PBE, and allowed for the development of a provably convergent AFEM for the PBE. However, in practice the regularization turns out to be numerically ill-conditioned. In this article, we examine a second regularization, and establish a number of basic results to ensure that the new approach produces the same mathematical advantages of the original regularization, without the ill-conditioning property. We then design an AFEM scheme based on the new regularized problem, and show that the resulting AFEM scheme is accurate and reliable, by proving a contraction result for the error. This result, which is one of the first results of this type for nonlinear elliptic problems, is based on using continuous and discrete a priori pointwise estimates to establish quasi-orthogonality. To provide a high-quality geometric model as input to the AFEM algorithm, we also describe a class of feature-preserving adaptive mesh generation algorithms designed specifically for constructing meshes of biomolecular structures, based on the intrinsic local structure tensor of the molecular surface. The stability advantages of the new regularization are demonstrated using an FETK-based implementation, through comparisons with the original regularization approach for a model problem. Abstract

STUDY DESIGN A case report and review of previous literature are presented. OBJECTIVE The objective of this manuscript was to report a case of cervical myelopathy caused by invaginated anomalous laminae of the axis in a spina bifida occulta patient with hypoplasia of the arch of the atlas and to discuss the etiology of this disease. SUMMARY OF BACKGROUND DATA To the authors' knowledge, few cases of cervical myelopathy due to invaginated anomalous laminae of the axis have been reported, none of which is combined with hypoplasia of the arch of atlas. Treatment was surgical removal of the invaginated laminae. METHODS The patient's history, clinical examination, imaging findings, and treatment were reported, and the etiology was discussed. RESULTS Characteristic findings were revealed from imaging studies and multiplane reconstruction of the computed tomography images. The patient was treated with a posterior decompressive operation based on the images. A rapid improvement was observed after the surgery, and the patient's neurology was completely restored 1 month later. CONCLUSION We reported a rare characteristic anomaly of the laminae of the axis with hypoplasia of the posterior arch of atlas. A multiplane reconstruction of the computed tomography images was very necessary for treatment of this case. Possible causes of this anomaly may be the failure of ossification or fusion of the embryological term, whereas invagination of the osteophyte may be associated with the traction of the dense fibrous band during growth and development. Surgical removal of the laminae could result in a satisfactory outcome.

This paper gives a solution to an open problem concerning the performance of various multilevel preconditioners for the linear finite element approximation of second-order elliptic boundary value problems with strongly discontinuous coefficients. By analyzing the eigenvalue distribution of the BPX preconditioner and multigrid V-cycle preconditioner, we prove that only a small number of eigenvalues may deteriorate with respect to the discontinuous jump or meshsize, and we prove that all the other eigenvalues are bounded below and above nearly uniformly with respect to the jump and meshsize. As a result, we prove that the convergence rate of the preconditioned conjugate gradient methods is uniform with respect to the large jump and meshsize. We also present some numerical experiments to demonstrate the theoretical results.

Abstract

A flux of neutrons from an astrophysical source in the Galaxy can be detected in the Pierre Auger Observatory as an excess of cosmic-ray air showers arriving from the direction of the source. To avoid the statistical penalty for making many trials, classes of objects are tested in combinations as nine “target sets”, in addition to the search for a neutron flux from the Galactic Center or from the Galactic Plane. Within a target set, each candidate source is weighted in proportion to its electromagnetic flux, its exposure to the Auger Observatory, and its flux attenuation factor due to neutron decay. These searches do not find evidence for a neutron flux from any class of candidate sources. Tabulated results give the combined p-value for each class, with and without the weights, and also the flux upper limit for the most significant candidate source within each class. Abstract

We derive lower bounds on the density of sources of ultra-high energy cosmic rays from the lack of significant clustering in the arrival directions of the highest energy events detected at the Pierre Auger Observatory. The density of uniformly distributed sources of equal intrinsic intensity was found to be larger than similar to (0.06 – 5) x 10(-4) Mpc(-3) at 95% CL, depending on the magnitude of the magnetic defections. Abstract

In this article we study adaptive finite element methods (AFEM) with inexact solvers for a class of semilinear elliptic interface problems.We are particularly interested in nonlinear problems with discontinuous diffusion coefficients, such as the nonlinear Poisson-Boltzmann equation and its regularizations. The algorithm we study consists of the standard SOLVE-ESTIMATE-MARK-REFINE procedure common to many adaptive finite element algorithms, but where the SOLVE step involves only a full solve on the coarsest level, and the remaining levels involve only single Newton updates to the previous approximate solution.

Abstract

Atmospheric conditions at the site of a cosmic ray observatory must be known for reconstructing observed extensive air showers. The Global Data Assimilation System (GDAS) is a global atmospheric model predicated on meteorological measurements and numerical weather predictions. GDAS provides altitude-dependent profiles of the main state variables of the atmosphere like temperature, pressure, and humidity. The original data and their application to the air shower reconstruction of the Pierre Auger Observatory are described. By comparisons with radiosonde and weather station measurements obtained on-site in Malargüe and averaged monthly models, the utility of the GDAS data is shown., Facultad de Ciencias Exactas

Abstract

The Pierre Auger Observatory is exploring the potential of the radio detection technique to study extensive air showers induced by ultra-high energy cosmic rays. The Auger Engineering Radio Array (AERA) addresses both technological and scientific aspects of the radio technique. A first phase of AERA has been operating since September 2010 with detector stations observing radio signals at frequencies between 30 and 80 MHz. In this paper we present comparative studies to identify and optimize the antenna design for the final configuration of AERA consisting of 160 individual radio detector stations. The transient nature of the air shower signal requires a detailed description of the antenna sensor. As the ultra-wideband reception of pulses is not widely discussed in antenna literature, we review the relevant antenna characteristics and enhance theoretical considerations towards the impulse response of antennas including polarization effects and multiple signal reflections. On the basis of the vector effective length we study the transient response characteristics of three candidate antennas in the time domain. Abstract

In this article we develop a convergence theory for goal-oriented adaptive finite element algorithms designed for a class of second-order semilinear elliptic equations. We briefly discuss the target problem class, and introduce several related approximate dual problems that are crucial to both the analysis as well as to the development of a practical numerical method. We then review some standard facts concerning conforming finite element discretization and error-estimate-driven adaptive finite element methods (AFEM). We include a brief summary of a priori estimates for this class of semilinear problems, and then describe some goal-oriented variations of the standard approach to AFEM (GOAFEM). Following the recent approach of Mommer-Stevenson and Holst-Pollock for increasingly general linear problems, we first establish a quasi-error contraction result for the primal problem. We then develop some additional estimates that make it possible to establish contraction of the combined primal-dual quasi-error, and subsequently show convergence with respect to the quantity of interest. Finally, a sequence of numerical experiments are then carefully examined. It is observed that the behavior of the implementation follows the predictions of the theory., Comment: 22 pages, 7 Figures. In this paper, we present a multigrid preconditioner for solving the linear system arising from the piecewise linear nonconforming Crouzeix-Raviart discretization of second order elliptic problems with jump coefficients. The preconditioner uses the standard conforming subspaces as coarse spaces. Abstract

In this paper, we construct an auxiliary space preconditioner for Maxwell’s equations with interface, and generalize the HX preconditioner developed in [9] to the problem with strongly discontinuous coefficients. For the H(curl) interface problem, we show that the condition number of the HX preconditioned system is uniformly bounded with respect to the coefficients and meshsize.

Abstract

This paper is devoted to study of an auxiliary spaces preconditioner for H(div) systems and its application in the mixed formulation of second order elliptic equations. Extensive numerical results show the efficiency and robustness of the algorithms, even in the presence of large coefficient variations. For the mixed formulation of elliptic equations, we use the augmented Lagrange technique to convert the solution of the saddle point problem into the solution of a nearly singular H(div) system. Numerical experiments also justify the robustness and efficiency of this scheme.

Abstract

We introduce and analyze two-level and multilevel preconditioners for a family of Interior Penalty (IP) discontinuous Galerkin (DG) discretizations of second order elliptic problems with large jumps in the diffusion coefficient. Our approach to IPDG-type methods is based on a splitting of the DG space into two components that are orthogonal in the energy inner product naturally induced by the methods. As a result, the methods and their analysis depend in a crucial way on the diffusion coefficient of the problem. The analysis of the proposed preconditioners is presented for both symmetric and non-symmetric IP schemes; dealing simultaneously with the jump in the diffusion coefficient and the non-nested character of the relevant discrete spaces presents additional difficulties in the analysis, which precludes a simple extension of existing results. However, we are able to establish robustness (with respect to the diffusion coefficient) and near-optimality (up to a logarithmic term depending on the mesh size) for both two-level and BPX-type preconditioners, by using a more refined Conjugate Gradient theory. Useful by-products of the analysis are the supporting results on the construction and analysis of simple, efficient and robust two-level and multilevel preconditioners for non-conforming Crouzeix-Raviart discretizations of elliptic problems with jump coefficients.
