Reactive oxygen species (ROS) generation to induce cell death is an effective strategy for cancer therapy. In particular, chemodynamic therapy (CDT), using Fenton-type reactions to generate highly cytotoxic hydroxyl radical (•OH), is a promising treatment modality. The extracellular matrix (ECM) is a “nest” established by cells in their surroundings. It is a microenvironment that supports cellular function and survival, as well as their collaboration with one another. In native organs, the ECM provides the desired spatial architecture, biomolecular composition, and biomechanical cues to guide cellular organization and activities that are unique to each organ. Recapitulating these essential features of the extracellular environment holds the key to enabling effective biofabrication of functional tissues, thus, becoming the central goal of engineering biomaterials in regenerative medicine. Accordingly, native ECM biomaterials have emerged as robust and versatile contenders for supporting tissue repair and regeneration, with growing interest in techniques that enhance their regenerative potential. In particular, chemoselective ECM functionalization is an emerging research direction that endows the native ECM with desirable new features without altering the overall material properties. Further, the ECM is constantly undergoing remodeling, especially following implantation and during regeneration, which is critical for the long-term stability and functionality of the engineered tissues. Here, we present a timely review of the unique material properties and applications of ECM biomaterials in tissue engineering, highlighting recent advances in ECM manufacture, functionalization, and remodeling characterization, while also providing an outlook of potential research directions that will move the field forward. In native organs, the extracellular matrix (ECM) provides a wide array of essential biological, mechanical, and physiological signals to guide cellular activities such as adhesion, survival, proliferation, differentiation, and interaction with other cells. ECM biomaterials extracted from native organs are gaining popularity in tissue engineering. In particular, recent advancements in whole-organ decellularization and 3D bioprinting are expediting the progress in bioengineering organs with physiologically relevant tissue architecture and biomolecular composition. This review summarizes key concepts in using native ECM biomaterials for tissue engineering and highlights the conventional and emerging strategies for further modification of such ECM materials to endow them with the desired biomimetic functions to facilitate tissue regeneration. We will also briefly cover the current understanding of constructive ECM remodeling during regeneration as well as related investigatory tools.

Nanoscale extracellular vesicles (EVs) represent a unique cellular derivative that reflect the therapeutic potential of mesenchymal stem cells (MSCs) toward tissue engineering and injury repair without the logistical and safety concerns of utilizing living cells. However, upon systemic administration in vivo,EVs undergo rapid clearance and typically lack controlled targeted delivery, thus reducing their effectiveness in therapeutic regenerative therapies. Here, we describe a strategy that enables long-term in vivo spatial EV retention by chemoselective immobilization of metabolically incoporated azido ligand-bearing EVs (azido-EVs) within a dibenzocyclooctyne-modified collagen hydrogel. MSC-derived azido-EVs exhibit comparable morphological and functional properties as their non-labeled EV counterparts and, when immobilized within collagen hydrogel implants via click chemistry, they elicited more robust host cell infiltration, angiogenic and immunoregulatory responses including vascular ingrowth and macrophage recruitment compared to ten times the higher dose required by non-immobilized EVs. We envision this technology will enable a wide range of applications to spatially promote vascularization and host integration relevant to tissue engineering and regenerative medicine applications.

As the material transportation system of the human body, the vascular network carries the transportation of materials and nutrients. Currently, the construction of functional microvascular networks is an urgent requirement for the development of regenerative medicine andin vitrodrug screening systems. How to construct organs with functional blood vessels is the focus and challenge of tissue engineering research. Here in this review article, we first introduced the basic characteristics of blood vessels in the body and the mechanism of angiogenesisin vivo, summarized the current methods of constructing tissue blood vesselsin vitroandin vivo, and focused on comparing the functions, applications and advantages of constructing different types of vascular chips to generate blood vessels. Finally, the challenges and opportunities faced by the development of this field were discussed.

Decellularized extracellular matrix (ECM) biomaterials derived from native tissues and organs are widely used for tissue engineering and wound repair. To boost their regenerative potential, ECM biomaterials can be functionalized via the immobilization of bioactive molecules. To enable ECM functionalization in a chemoselective manner, we have recently reported an effective approach for labeling native organ ECM with the click chemistry-reactive azide ligand via physiologic post-translational glycosylation. Here, using the rat lung as a model, we provide a detailed protocol for in vivo and ex vivo metabolic azide labeling of the native organ ECM using N-Azidoacetylgalactosamine-tetraacylated (Ac(4)GalNAz), together with procedures for decellularization and labeling characterization. Our approach enables specific and robust ECM labeling within three days in vivo or within one day during ex vivo organ culture. The resulting ECM labeling remains stable following decellularization. With our approach, ECM biomaterials can be functionalized with desired alkyne-modified biomolecules, such as growth factors and glycosaminoglycans, for tissue engineering and regenerative applications.

A total of 432 lung adenocarcinoma patientswith pleural metastasis (metastasis group) and 424patients with lung adenocarcinoma but without pleuralmetastasis (nonmetastasis group) were enrolled in this study. Thus, the present study aimed to investigate whether -216G/T (rs712829), a functional polymorphism of the EGFR promoter that is able to induce EGFR activation and overexpression, is associated with the pleural metastasis of lung adenocarcinoma. The study subjects were comprised of 326 patients with primary lung adenocarcinoma and 312 matched cases with pleural metastasis. The -216G/T genotypes were determined in all subjects by PCR amplification and direct DNA sequencing, and EGFR expression was also evaluated by immunohistochemical staining in the primary tumor tissues with various -216G/T genotype backgrounds. Due to quantum interference, light can transmit through dense atomic media, a phenomenon known as electromagnetically induced transparency (EIT). We propose that EIT is not limited to light transmission and there is an electronic analog where resonant transparency in charge transport in an opaque structure can be induced by electromagnetic radiation. A triple-quantum-dots system with Λ-type level structure is generally opaque due to the level in the center dot being significantly higher and therefore hopping from the left dot to the center dot is almost forbidden. We demonstrate that an electromagnetically induced electron transparency (EIET) in charge of transport can indeed occur in the Λ-type system. The direct evidence of EIET is that an electron can travel from the left dot to the right dot, while the center dot apparently becomes invisible. We analyze EIET and the related shot noise in both the zero and strong Coulomb blockade regimes. It is found that the EIET (position, height, and symmetry) can be tuned by several controllable parameters of the radiation fields, such as the Rabi frequencies and detuning frequencies. The result offers a transparency/opaque tuning technique in charge transport using interfering radiation fields.

Phase relaxation of electrons transferring through an electromechanical transistor is studied using the Aharonov-Bohm interferometer. With the approach of quantum master equation, the phase properties of an electron are numerically analyzed based on the interference fringes. Coherence of electron is partially destroyed by its scattering on excited levels of the local nanomechanical oscillator. Transmission amplitudes with respect to two adjacent mechanical vibrational levels have a phase difference of $\pi$. The result offers a resonant transport tuning technique using radiation fields.
