Abstract The epithelial–mesenchymal transition (EMT) is a genetic reprogramming that tumor cells utilize for metastasis. Epsin-3 (EPN3) is an endocytic adapter protein involved in clathrin-mediated endocytosis and had been previously linked to EMT in breast cancer and glioma metastasis. In this study, identified the role of epsin-3 in lung adenocarcinoma and metastasis and epsin-3 levels identified using an expression profile analysis of patient data indicated the protein was abnormally overexpressed in lung adenocarcinoma patients and this was directly linked to disease severity. Gene knockdowns of EPN3 in human adenocarcinoma cell line A549 and the non-small cell lung carcinoma cell line H1299 decreased the levels of mesenchymal markers, including vimentin (VIM), N-cadherin (NCAD) and embryonic transcription factors like zinc finger E-box binding homeobox 1(ZEB1), snail, and the key molecules of Wnt pathway such as β-catenin and resulted in increased expression of the epithelial marker E-cadherin (ECAD). Our data links EPN3 to the EMT process in lung cancer and inhibition of its expression reduced the metastatic and invasive ability of lung adenocarcinoma cells by inhibiting the EMT process.

Abstract Background The detailed transcriptomic profiles during human serotonin neuron (SN) differentiation remain elusive. The establishment of a reporter system based on SN terminal selector holds promise to produce highly-purified cells with an early serotonergic fate and help elucidate the molecular events during human SN development process. Methods A fifth Ewing variant (FEV)-EGFP reporter system was established by CRISPR/Cas9 technology to indicate SN since postmitotic stage. FACS was performed to purify SN from the heterogeneous cell populations. RNA-sequencing analysis was performed for cells at four key stages of differentiation (pluripotent stem cells, serotonergic neural progenitors, purified postmitotic SN and purifed mature SN) to explore the transcriptomic dynamics during SN differentiation. Results We found that human serotonergic fate specification may commence as early as day 21 of differentiation from human pluripotent stem cells. Furthermore, the transcriptional factors ZIC1, HOXA2 and MSX2 were identified as the hub genes responsible for orchestrating serotonergic fate determination. Conclusions For the first time, we exposed the developmental transcriptomic profiles of human SN via FEV reporter system, which will further our understanding for the development process of human SN. Abstract Corporate social responsibility (CSR) could be an effective risk-alleviating tool for companies. However, an important issue is whether CSR plays a risk-alleviating role in large negative shocks such as the COVID-19 pandemic. This study investigated the impact of corporate social responsibility on stock performance in China during the COVID-19 pandemic, focusing on the differing effects of strategic and responsive CSR. By analysing daily data on newly confirmed COVID-19 cases and the stock performance of A-share listed companies during the lockdown period in 2020, we find that CSR can significantly mitigate the negative effects of the pandemic on stock returns and recovery. Specifically, responsive CSR has a significant short-term moderating effect, whereas strategic CSR has a significant long-term moderating effect. The moderating effects of CSR are more pronounced among non-state-owned firms, firms with lower financial leverage, and large companies. These findings suggest that companies can reduce short- and long-term risks by strategically using responsive and strategic CSR in response to negative shocks in global economies.

To study the impact of cutting blasting on the surface, a vibration waveform prediction function was constructed, and a method of dividing the affected area was proposed. Based on the equivalent spherical charge theory, it is possible to establish a connection between the fitting coefficient and the engineering parameters in the equivalent source intensity function. Furthermore, a blasting vibration waveform function suitable for engineering can be constructed. Secondly, the reliability of the method introduced is verified through the data monitored on-site. Finally, the affected partitions of blasting vibration are divided based on the peak particle velocity and vibration displacement as standards. The results show that the vibration waveform prediction system introduced can restore the vibration waveform corresponding to cutting blasting. In addition, the zoning method can reasonably divide the scope of the affected area.

Abstract Improving the resolution of computed tomography (CT) medical images can help doctors more accurately identify lesions, which is important in clinical diagnosis. In the absence of natural paired datasets of high resolution and low resolution image pairs, we abandoned the conventional Bicubic method and innovatively used a dataset of images of a single resolution to create near‐natural high–low‐resolution image pairs by designing a deep learning network and utilizing noise injection. In addition, we propose a super‐resolution generative adversarial network called KerSRGAN which includes a super‐resolution generator, super‐resolution discriminator, and super‐resolution feature extractor to achieve a 4× super‐resolution of CT images. The results of an experimental evaluation show that KerSRGAN achieved superior performance compared to the state‐of‐the‐art methods in terms of a quantitative comparison of non‐reference image quality evaluation indicators on the generated 4× super‐resolution CT images. Moreover, in terms of an intuitive visual comparison, the images generated by the KerSRGAN method had more precise details and better perceptual quality.

ABSTRACT Enterohemorrhagic Escherichia coli (EHEC) can colonize the gastrointestinal tract and cause bloody diarrhea in children. Previous studies showed that its pathogenesis could be mediated by metabolites from both the host and microbiota. L-Glutamine (Gln) was found to be depleted in intestinal tissues as the main energy source according to previous studies. Hence, we aimed to determine the effects of Gln on EHEC infection and its underlying mode of action. In this study, a Gln-limited signal was found to activate the type 3 secretion system (T3SS), which is crucial for EHEC infection via perturbation of central metabolism. By shifting the phosphorylation of NtrC, a key regulator in bacterial nitrogen metabolism, Gln stimulates ler transcripts in a σS-PchA-dependent manner. Our in vivo experiments further demonstrated that Gln supplementation can reduce EHEC colonization in the gastrointestinal tract by repressing T3SS. Moreover, Gln could further attenuate bacterial infection by boosting host defense, which might be dependent on multiple pathways. Besides, our experiments demonstrated that Gln did not induce Shiga-like toxin (Stx) production or cause impairment of gut flora. In conclusion, our study presented evidence that Gln could act against EHEC infection by reducing bacterial virulence and strengthening host defense. Therefore, Gln serves as a promising therapeutic agent for EHEC infection. IMPORTANCE The type 3 secretion system (T3SS) was obtained in many Gram-negative bacterial pathogens, and it is crucial for their pathogenesis. Environmental signals were found to be involved in the expression regulation of T3SS, which was vital for successful bacterial infection in the host. Here, we discovered that L-glutamine (Gln), the most abundant amino acid in the human body, could repress enterohemorrhagic Escherichia coli (EHEC) T3SS expression via nitrogen metabolism and therefore had potential as an antivirulence agent. Our in vitro and in vivo evidence demonstrated that Gln could decline EHEC infection by attenuating bacterial virulence and enhancing host defense simultaneously. We repurpose Gln as a potential treatment for EHEC infection accordingly.

Societal Impact Statement Genetic engineering is used to introduce new genes into plants to obtain crops with novel traits. However, the unintended effects of genetically engineered (GE) crops on nontarget organisms—such as beneficial insects—are a topic of much concern. We evaluate the potential unintended effects of pollen from three GE maize and seven commercialized conventional maize lines on the ladybird beetle Propylea japonica, by combining omics approaches with feeding assays. Our results suggest that unintended changes caused by genetic engineering in maize pollen may not lead to biologically relevant effects on P. japonica. Meanwhile our study provides a useful strategy to assess the biological impacts of genetic engineering on nontarget organisms. Summary The potential effects caused by the inserted traits in genetically engineered (GE) plants on nontarget organisms (NTOs) have been well assessed. However, whether the process of genetic engineering itself causes unintended changes that go beyond the natural variation of the crop and further poses any biological effects to NTOs is still under debate. Here, we evaluated the potential unintended effects of pollen from three GE maize and seven commercialized conventional maize lines on the NTO Propylea japonica by combining omics approaches with feeding assays. The results showed that genetic breeding indeed brought somewhat differences at both proteome and metabolome levels in maize pollen, although such differences were far more common in conventionally crossbred plants. Feeding experiments indicated that the changes in proteins and metabolites caused by genetic breeding did not lead to unintended effects on the NTOs that go beyond those measured for the conventional crossbred lines. Together, our results suggest that the differences detected by omics experiments may not cause any biological relevant effects on NTOs and the combination of omics approaches and NTOs feeding assays provide a valid approach to assess the biological relevance of compositional effects caused by genetic breeding.

While transgenic Bacillus thuringiensis (Bt) maize provides pest resistance and a reduced application of chemical pesticides, a comprehensive environmental risk assessment is mandatory before its field release. This research determined the concentrations of Bt protein in plant tissue and in arthropods under field conditions in Gongzhuling City, northeastern China, to provide guidance for the selection of indicator species for non-target risk assessment studies. Bt maize expressing Cry1Ab/2Aj and non-transformed near-isoline were grown under identical environmental and agricultural conditions. Cry1Ab/2Aj was detected in plant tissues and arthropods collected from Bt maize plots during pre-flowering, flowering, and post-flowering. The expression of Cry1Ab/2Aj varied across growth stages and maize tissues, as well as in the collected arthropods at the three growth stages. Therefore, representative species should be chosen to cover the whole growing season and to represent different habitats and ecological functions. Dalbulus maidis (Hemiptera: Cicadellidae), Rhopalosiphum padi (Hemiptera: Aphididae), Heteronychus arator (Coleoptera: Scarabaeidae), and Somaticus angulatus (Coleoptera: Tenebrionidae) are suitable non-target herbivores. Propylea japonica (Coleoptera: Coccinellidae), Paederus fuscipes (Coleoptera: Staphylinidae), Chrysoperla nipponensis (Neuroptera: Chrysopidae), and spiders are suggested predators. Apis cerana and Apis mellifera ligustica (both Hymenoptera: Apidae) represent pollinators and Folsomia candida (Collembola: Isotomidae) decomposers.

To optimize the parameters of slicing ferrite with high precision diamond ring saw, an orthogonal test is designed with the fabricating surface accuracy and the surface roughness as evaluation indicators and the spindle speed, the feed speed and the tension force as factors. Based on the grey theory, the data analysis and the comprehensive evaluation of the multiple process targets are carried out to obtain an optimized process parameter combination, namely the spindle speed 1 000 r/min, the feed speed 1.0 mm/min and the tension force 90 N. The slicing test results show that the optimized parameter combination can obtain a surface accuracy of PV 7.37 μm and a surface roughness Ra of 0.882 μm, and the slicing surface quality is improved, which verifies the effectiveness and practicability of this method in the optimization of ferrite slicing process parameters.

Abstract Background Non-small cell lung cancer (NSCLC) is the most prevalent form of lung cancer globally, with average age of cancer patients becoming younger gradually. It is of significance to gain a comprehensive understanding of molecular mechanism underlying NSCLC. Methods Quantitative polymerase chain reaction (qPCR) and western blot were applied to measure RNA and protein levels separately. Functional assays and western blot were performed to determine the effects of miR-489-3p and USP48 on cell growth, migration and epithelial-mesenchymal transition (EMT) in NSCLC. TOP/FOP flash luciferase reporter assay was carried out to detect the activity of Wnt pathway. Besides, qPCR, RNA pulldown and luciferase reporter assays were conducted to probe into the target gene of miR-489-3p. Immunoprecipitation-western blot (IP-western blot) analysis was implemented to assess the effect of USP48 on the ubiquitination of β-catenin. Results miR-489-3p hampers NSCLC cell proliferation, migration and EMT in vitro and NSCLC tumorigenesis and metastasis in vivo. Additionally, miR-489-3p inactivates Wnt/β-catenin signaling pathway and regulates USP48 to inhibit the ubiquitination of β-catenin. Moreover, USP48 propels the development of NSCLC cells. Conclusions The current study demonstrated that miR-489-3p promotes the malignant progression of NSCLC cells via targeting USP48, which might offer a new perspective into NSCLC treatment. PurposeLung cancer is the leading cause of death from cancer and the number of operable elderly lung cancer patients is increasing, with advanced age being associated with a poorer prognosis. However, there is no easy and comprehensive prognostic assessment method for these patients.MethodsClinicopathological data of patients aged 65 years or older with TNM stage I-II lung cancer from 2004 to 2018 were downloaded from the SEER database. Patients from 2004 to 2015 were randomized into a training group (n = 16,457) and a validation group (n = 7,048). Data from 2016 to 2018 (n = 6,231) were used for external validation. Two nomogram prognostic models were created after independent prognostic factors connected to both overall survival (OS) and cancer-specific survival (CSS) in the training set by using univariate and multivariate Cox proportional hazards regression analysis. In turn, overall survival (OS) and cancer-specific survival (CSS) were predicted for patients at 1, 3, and 5 years. Based on the concordance index (C-index), calibration curves, area under the receiver operating characteristics (ROC) curve (AUC), the time-dependent area under the ROC curve, the validity, accuracy, discrimination, predictive ability, and clinical utility of the models were evaluated. Decision curve analysis (DCA) was used to assess the clinical value of the models.ResultsA total of 29,736 patients were included. Univariate and multivariate analyses suggested that age, race, gender, marriage, disease grade, AJCC stage, T-stage, surgery, radiotherapy, chemotherapy, and tumor size were independent risk factors for patient prognosis. These 11 variables were included in nomogram to predict OS and CSS of patients. C-indexes of OS for the training, validation and external validation sets were 0.730 (95% CI, 0.709–0.751), 0.734 (95% CI, 0.722–0.746), and 0.750 (95% CI, 0.734–0.766), respectively. The AUC results for the training and validation sets indicated good accuracy for this nomogram. The calibration curves demonstrated a high degree of concordance between actual and anticipated values, and the DCA demonstrated that the nomograms had better clinical application than the traditional TNM staging approach.ConclusionThis study identified risk factors for survival in operable elderly lung cancer patients and established a new column line graph for predicting OS and CSS in these patients. The model has good clinical application and can be a good clinical decision-making tool for physicians and patients.

BackgroundIn recent years, teenage students’ internet gaming disorder has raised widespread concern in the society. The current study aims to explore how perceptions of school climate, maladaptive cognition, and belief in a just world impact teenage students’ internet gaming disorder and seek the suitable intervention to prevent teenage students’ internet gaming disorder tendency.MethodsA total of 1,164 teenage students (age: 19.62 ± 3.14 years) were evaluated using the Perceived School Climate Scale (PSCS), Maladaptive Cognition Scale (MCS), Belief in a Just World Scale (BJWS), and Internet Gaming Disorder Scale (IGDS).Results(1) Perceptions of school climate could predict teenage students’ internet gaming disorder [β = –0.15, p < 0.001, 95% CI = (–0.20, –0.11)]. (2) Teenage students’ maladaptive cognition exerts a partial mediating effect between their perceptions of school climate and internet gaming disorder [ab = –0.13, boot SE = 0.01, 95% CI = (–0.16, –0.11)]. (3) The direct effect of perceptions of school climate on internet gaming disorder [β = –0.07, p < 0.01, 95% CI = (–0.11, –0.02)], the first half of the mediation model “perceptions of school climate → maladaptive cognition” [β = –0.08, p < 0.01, 95% CI = (–0.12, –0.03)], and the second half of the mediation model “maladaptive cognition → internet gaming disorder” [β = 0.10, p < 0.001, 95% CI = (0.06, 0.14)] are all moderated reciprocally by teenage students’ belief in a just world.ConclusionSchools should create a good climate and shape belief in a just world to reduce the maladaptive cognition of teenagers, to effectively prevent teenage students’ internet gaming disorder tendency.

RNA interference (RNAi) that is triggered by small or short RNAs has shown enormous potential in the development of pest control strategies. Two microRNAs (miRNAs), Csu-novel-260 and Csu-miR-14, were used in insect-resistant genetically engineered (IRGE) rice lines to confer resistance to Chilo suppressalis. However, a risk assessment of RNAi-based products is essential to determine the safety of a biopesticide or IRGE crop for commercialization. The non-target organism Folsomia candida, which plays an important ecological role as a soil decomposer in agricultural ecosystems, was used to assess the risk of miRNAs Csu-novel-260 and Csu-miR-14. In this study, a dietary miRNA toxicity assay system was established in F. candida. The expression levels of target genes, survival rate, fecundity and body size were investigated to evaluate the effects of the miRNAs on F. candida under the worst-case scenario. The results showed that the dietary miRNA toxicity assay system could be used for risk assessment of miRNA in F. candida. The target genes of miRNAs were influenced by miRNA at some time points. However, no significant differences were observed in the life-table parameters in F. candida fed with a diet containing miRNAs. The dietary effects of two miRNAs on F. candida are neutral.

Employees’ voice and innovation behaviors are an important source of organizational competitiveness. Scholars in the field of organizational behaviors have discussed how to increase the willingness of employees to engage in voice and innovation behaviors from a diversity of perspectives. Innovation has always been a strategic goal of organizations. To motivate employees to offer valuable advice and innovative ideas, organizations have to provide various incentive, feedback and supportive programs. Combined with the social exchange and social cognitive theories, this study presents an argument that the effective strategic human resource management can gradually improve the self-efficacy, psychological contract, voice behaviors and innovation behaviors of employees, and further verifies the relationship among them. A sample of 553 employees was used and analyzed via structure equation modeling. This study adopted PLS-SEM to verify structural model and examine the mediating effect of psychological mechanism. The results showed that strategic human resource management has a significant and positive impact on self-efficacy, psychological contract, voice behavior and innovation behavior; self-efficacy has a significant and positive impact on psychological contract, voice behavior and innovation behavior; psychological contract only has a significant and positive impact on innovation behavior, but not on voice behavior. Given the above research findings, this study gives some practical implications in the end.

Abstract Background Clear cell renal cell carcinoma (ccRCC) is the most common subtype of renal cell carcinoma and accounts for cancer-related deaths. Survival rates are very low when the tumor is discovered in the late-stage. Thus, developing an efficient strategy to stratify patients by the stage of the cancer and inner mechanisms that drive the development and progression of cancers is critical in early prevention and treatment. Results In this study, we developed new strategies to extract important gene features and trained machine learning-based classifiers to predict stages of ccRCC samples. The novelty of our approach is that (i) We improved the feature preprocessing procedure by binning and coding, and increased the stability of data and robustness of the classification model. (ii) We proposed a joint gene selection algorithm by combining the Fast-Correlation-Based Filter (FCBF) search with the information value, the linear correlation coefficient, and variance inflation factor, and removed irrelevant/redundant features. Then the logistic regression-based feature selection method was used to determine influencing factors. (iii) Classification models were developed using machine learning algorithms. This method is evaluated on RNA expression value of clear cell renal cell carcinoma derived from The Cancer Genome Atlas (TCGA). The results showed that the result on the testing set (accuracy of 81.15% and AUC 0.86) outperformed state-of-the-art models (accuracy of 72.64% and AUC 0.81) and a gene set FJL-set was developed, which contained 23 genes, far less than 64. Furthermore, a gene function analysis was used to explore molecular mechanisms that might affect cancer development. Conclusions The results suggested that our model can extract more prognostic information, and is worthy of further investigation and validation in order to understand the progression mechanism.

Context Fructus Meliae toosendan extracts (FMTE) have a good therapeutic effect on coccidiosis, but there is no relevant research on its prophylactic effect on coccidiosis. Objective This study comprehensively evaluates the anticoccidial effect of FMTE. Materials and methods In vitro, the unsporulated oocysts were treated with serial dilutions of FMTE and incubated for 7 d, and the sporulated oocysts were counted for calculating the median lethal concentration (LC50) of FMTE. In vivo, 180 10-day-old broiler chickens free of coccidiosis were weighted and randomly distributed into six groups: normal group, untreated group, 4 protective groups (positive group and three FMTE groups). From day 10 to day 21, chickens in the three FMTE groups were pre-treated with FMTE at the dosage of 2.5, 5 and 10 g/kg/d, respectively, and chickens in the positive group were pre-treated with qiuliling (10 g/kg/d). On day 14, chickens in all groups except the normal group were orally infected with 1.5 × 104 sporulated oocysts. The clinical symptoms were observed from day 10 to day 21, the anticoccidial index (ACI), tissue lesions, and intestinal microflora were determined on day 21. Results FMTE showed anti-sporulation effect against E. tenella and the LC50 value was 245.83 µg/mL in vitro. In vivo, FMTE at the dosage of 10 g/kg/d was effective against E. tenella infection, and its ACI value was 162.56, which was higher than the value of positive drug qiuliling (128.81). Discussion and conclusions: FMTE have potent anticoccidial effects, and it presents an alternative anticoccidial agent for avian coccidiosis control.

Lysozymes are naturally occurring enzymes present in a variety of biological organisms, such as bacteria, fungi, and animal bodily secretions and tissues. It is also the main ingredient of many ethnomedicines. It is well known that lysozymes and lysozyme-like enzymes can be used as anti-bacterial agents by degrading bacterial cell wall peptidoglycan that leads to cell death, and can also inhibit fungi, yeasts, and viruses. In addition to its direct antimicrobial activity, lysozyme is also an important component of the innate immune system in most mammals. Increasing evidence has shown the immune-modulatory effects of lysozymes against infection and inflammation. More recently, studies have revealed the anti-cancer activities of lysozyme in multiple types of tumors, potentially through its immune-modulatory activities. In this review, we summarized the major functions and underlying mechanisms of lysozymes derived from animal and plant sources. We highlighted the therapeutic applications and recent advances of lysozymes in cancers, hypertension, and viral diseases, aiming toseeking alternative therapies for standard medical treatment bypassing side effects. We also evaluated the role of lysozyme as a promising cancer marker for prognosis to indicate the outcomes recurrence for patients.

Recently, issues of human resource management gradually attract a lot of attention from organizational behavior scholars, thus how to effectively improve service employees’ job attitude and performance to meet the needs of stakeholders is one of the key issues in internal marketing. Based on the perspective of internal marketing, the study transforms the relevant factors applied to maintaining external customer relations into internal employee-oriented factors, so as to increase the understanding of the relationship between internal service recovery and internal relationship quality (IRQ). This study aims to explore (1) whether internal service recovery enhances IRQ; (2) whether internal relationship investment (IRI) positively moderates relationship between internal service recovery and IRQ; and (3) whether effectiveness of internal service recovery differentiates under different exchange relationship (high/low quality leader-member exchange). In this study, a total of 206 Mainland China and 250 Taiwanese participants were collected. In this study, a variance-based structural equation modeling (PLS-SEM) was performed to test the proposed hypothesizes and conduct comparative analysis. Empirical results in both samples show that internal service recovery has positive and significant effects on IRQ; internal relationship investment and leader-member exchange (LMX) positively and significantly moderate the relationship between internal service recovery and IRQ. Finally, based on the results, this study provides some discussions, suggestions and managerial implications for future studies in organizational management.

Eucalyptus plantations play an important role in the timber supply and global warming mitigation around the world. Forest age is a critical factor for evaluating and modeling forest structure (e.g., diameter at breast height (DBH), height (H), aboveground carbon stocks (ACS)) and their dynamics. Recently, the spatial distribution of forest age at different scales based on time series remote sensing data has been widely investigated. However, it is unclear whether such data can effectively support the simulation and assessment of forest structure, especially in fast-growing plantation forests. In this study, the physiological principles in predicting growth (3-PG) model was firstly optimized and calibrated using survey and UAV lidar data at the sample plot (SP) scale, and was then applied at the forest sub-compartment (FSC) scale by designing different simulation scenarios driven by different forest age data sources and adjustments. The sensitivity of the simulated forest structure parameters to forest age was assessed at the SP and FSC levels. The results show that both the survey forest age data and the remote-sensing-derived forest age data could accurately estimate the DBH, H, and ACS of eucalyptus plantations with the coefficients of determination (R2) ranging from 0.87 to 0.94, and the relative root mean square error (RRMSE) below 20% at SP level. At the FSC level, the simulation results based on remotely sensed forest age data are significantly better than FSC forest age data from surveys by forestry bureaus, with R2 of ACS 0.7, RMSE 9.12 Mg/ha, and RRMSE 28.24%. The results of the sensitivity analysis show that the DBH, H, and ACS show different degrees of variation under different adjusted forest ages at SP and FSC level. The maximum difference in ACS is 82.91% at the SP scale if the forest age decreases 12 months and 41.23% at the FSC scale if the forest age increases 12 months. This study provides an important reference for future studies using forest age data obtained by remote sensing to drive the forest carbon model in a large spatial scale.

An 80%Ar-10%CO2-10%He ternary gas mixture was used as the shielding gas during the narrow-gap welding of thick Q690E high-strength steel plates. Complete and defect-free welded joints were obtained, and the microstructure and mechanical properties of the welded joint were investigated. The weld zone consists of a solidification area and interlayer zone, and the heat-affected zone consists of a coarse-grain heat-affected zone (CG-HAZ) and a fine-grain heat-affected zone (FG-HAZ). The microstructures of the weld zone are mainly lath bainite (LB), acicular ferrite (AF), and granular bainite (GB). The microstructure of the CG-HAZ is lath martensite (LM) and the microstructure of FG-HAZ is GB. Methods with different heat inputs were used to study their effects on the mechanical properties of the welded joint. It was found that the microstructure and mechanical properties of the welded joints are better with lower heat input. With tandem wire narrow-gap GMA welding, the tensile strength of the joints declined from 795.3 to 718.3 MPa and the impact toughness at −40 °C resulted in a weak position in the weld zone, which declined from 76~81 J to 55~69 J, when the welding speed reduced from 350 to 250 mm/min. With oscillating-arc narrow-gap GMA welding, the tensile strength achieved 853.4 MPa and the impact toughness at −40 °C was around 69~87 J. The results indicated that, under the appropriate heat input, the tensile strength of the joint exceeds 770 MPa and the low temperature impact toughness at −40 °C exceeds 69 J. A 155 mm-thick Q690E steel welded joint was obtained and the mechanical properties of the welded joint meets the requirements of the offshore drilling platforms.

In this study, a nickel ferrite (NiFe2O4) system was constructed to purify a phenol solution in water. During the process, the influences of several critical operating parameters including the NiFe2O4 amount, PS dosage, MW power, initial pH value, and different natural water anions were systematically studied. The results indicated that the constructed system performed excellently regarding the removal efficiency (97.74%) of phenol within 30 min. Meanwhile, the influence of co-existing anions such as Cl−, NO3−, H2PO4−, and HCO3− was also studied, which displayed an inhibiting action on phenol degradation, while HA facilitated it. To explore the reaction mechanism of this system, major free radical quenching experiments were conducted, and it was confirmed that both SO4•− and HO• were primary radicals. Moreover, stability experiments confirmed the apt stability of the NiFe2O4 system. Besides, the mineralization and toxicity analysis performed during phenol degradation also confirmed the superiority of the as-constructed system. Furthermore, the possible degradation mechanism of phenol was proposed. Hence, this system could be applied in advanced wastewater treatment.

Transgenic Bacillus thuringiensis (Bt) maize has broad prospects for application in China. Before commercialization, it is necessary to assess possible ecological impacts, including impacts on non-target arthropods (NTAs) in the field. In the present study, transgenic Bt maize expressing cry1Ab/2Aj and its corresponding non-transformed near isoline were planted under the same environmental and agricultural conditions, and arthropods in the field were collected during the three main growth stages of maize. In a one year trial, the results showed the composition of NTA communities in the transgenic and control maize fields were similar. There were no significant differences for community-level parameters of species richness (S), Shannon–Wiener diversity index (H′), evenness index (J) and Simpson’s dominant concentration (C) between the two types of maize fields. Likewise, a Bray–Curtis dissimilarity and distance analysis showed that Cry1Ab/2Aj toxin exposure did not increase community dissimilarities between Bt and non-Bt maize plots and that the structure of the NTAs community was similar on the two maize varieties. Furthermore, planting of the transgenic cry1Ab/2Aj maize did not affect the density or composition of non-target decomposers, herbivores, predators, parasitoids and pollinator guilds. In summary, our results showed that planting of Bt maize producing Cry1Ab/Cry2Aj proteins do not adversely affect population dynamics and diversity of NTAs.

Traditional forest inventories are based on field surveys of established sample plots, which involve field measurements of individual trees within a sample plot and the selection of proper allometric equations for tree volume calculation. Thus, accurate field measurements and properly selected allometric equations are two crucial factors for providing high-quality tree volumes. One key problem is the difficulty in accurately acquiring tree height data, resulting in high uncertainty in tree volume calculation when the diameter at breast height (DBH) alone is used. This study examined the uncertainty of tree height measurements using different means and the impact of allometric models on tree volume estimation accuracy. Masson pine and eucalyptus plantations in Fujian Province, China, were selected as examples; their tree heights were measured three ways: using an 18-m telescopic pole, UAV Lidar (unmanned aerial vehicle, light detection and ranging) data, and direct measurement of felled trees, with the latest one as a reference. The DBH-based and DBH–height-based allometric equations corresponding to specific tree species were used for the calculations of tree volumes. The results show that (1) tree volumes calculated from the DBH-based models were lower than those from the DBH–height-based models. On average, tree volumes were underestimated by 0.018 m3 and 0.117 m3 for Masson pine and eucalyptus, respectively, while the relative root-mean-squared errors (RMSEr) were 24.04% and 33.90%, respectively, when using the DBH-based model; (2) the tree height extracted from UAV Lidar data was more accurate than that measured using a telescopic pole, because the pole measurement method generally underestimated the tree height, especially when the trees were taller than the length of the pole (18 m in our study); (3) the tree heights measured using different methods greatly impacted the accuracies of tree volumes calculated using the DBH–height model. The telescopic-pole-measured tree heights resulted in a relative error of 9.1–11.8% in tree volume calculations. This research implies that incorporation of UAV Lidar data with DBH field measurements can effectively improve tree volume estimation and could be a new direction for sample plot data collection in the future.

Abstract Background Ladybird beetles (Coleoptera: Coccinellidae) are abundant predatory species in many agroecosystems, are valued for their biological pest control functions, and have been recommended as test species for studies supporting the assessment of non-target effects of insect-resistant Bt crops. Although insecticidal Bt proteins are known to be highly specific against target pests, some recent laboratory studies reported putative toxic effects of Bt proteins on ladybird species. While such studies have been criticised because of methodological shortcomings or inconsistencies, they cast doubt on the insecticidal spectrum of activity of some Bt proteins. Performing a systematic review that synthesises all existing evidence on this controversial topic may help to resolve the remaining scientific uncertainties. The review question to be addressed by the systematic review is the following: Are ladybird beetles (Coleoptera: Coccinellidae) affected by Bt proteins expressed in genetically modified insect-resistant crops? The systematic review will focus on studies performed under controlled environmental conditions. Methods An extensive literature search will be conducted to identify the articles relevant to the review question. A wide range of electronic bibliographic databases, the internet search engine Google Scholar, and websites of specialized organizations will be searched. Citation searching, reference list-checking and searching of key journals will also be performed. The relevance of the identified articles will be assessed against a set of pre-defined eligibility criteria, following a two-step approach. In the first step, title and abstract (or summary) will be screened, whilst in the second step the full text of all remaining articles will be assessed by two members of the review team. All relevant studies will be subjected to an appraisal of external (generalisability) and internal (risk of bias) validity. Data from the selected studies will be extracted and synthesised in a narrative report. If a sufficient number of datasets generated with comparable experimental setup is available, statistical meta-analyses will be conducted on a range of comparisons and including sensitivity analyses.

BackgroundThe effects of up-regulated CircCHST15 on lung cancer remained unclear. In this study, the role of CircCHST15 in lung cancer was investigated.MethodsDual-luciferase reporter verified the bioinformatics prediction that CircCHST15 targeted miR-155-5p and miR-194-5p. The correlation between CircCHST15 and PD-L1 was analyzed by Pearson analysis. CCK-8 and colony formation was performed to determine the viability and proliferation of lung cancer cells. After the lung cancer (subcutaneous-xenotransplant) model was established in mice, the T cell subtype and related cytokines in mouse tumor tissues were detected by flow cytometry and ELISA. Moreover, the expressions of CircCHST15, miR-155-5p, miR-194-5p, immune-related, and proliferation-related factors of the lung cancer cells or mice tumor tissues were detected by immunohistochemistry, RT-qPCR, or Western blot.ResultsCircCHST15 and PD-L1 were high-expressed in lung cancer, and the two was positively correlated. CircCHST15 targeted miR-155-5p and miR-194-5p, the later further targeted PD-L1. Lung cancer cell viability and proliferation were increased by miR-155-5p and inhibited by miR-194-5p. CircCHST15 located in the cytoplasm promoted tumor growth, down-regulated the expressions of miR-155-5p and miR-194-5p, and up-regulated the expressions of PD-L1, Ki-67, PCNA, CCL17, CCL22, IFN-γ, TNF-β, and IL-10. Also, CircCHST15 decreased the CD8+ cells in mouse blood and tumor, but increased the Tregs in mouse tumor. PD-L1 inhibitor showed an opposite effect to CircCHST15 on mouse tumors.ConclusionCircCHST15 sponged miR-155-5p and miR-194-5p to promote the PD-L1-mediated immune escape of lung cancer cells.

Plants typically release large quantities of volatiles in response to herbivory by insects. This benefits the plants by, for instance, attracting the natural enemies of the herbivores. We show that the brown planthopper (BPH) has cleverly turned this around by exploiting herbivore-induced plant volatiles (HIPVs) that provide safe havens for its offspring. BPH females preferentially oviposit on rice plants already infested by the rice striped stem borer (SSB), which are avoided by the egg parasitoid Anagrus nilaparvatae, the most important natural enemy of BPH. Using synthetic versions of volatiles identified from plants infested by BPH and/or SSB, we demonstrate the role of HIPVs in these interactions. Moreover, greenhouse and field cage experiments confirm the adaptiveness of the BPH oviposition strategy, resulting in 80% lower parasitism rates of its eggs. Besides revealing a novel exploitation of HIPVs, these findings may lead to novel control strategies against an exceedingly important rice pest.

The Bacillus thuringiensis toxin Cry1F has been used to develop insect-resistant genetically engineered crops. There has been great interest in evaluating its potential risk to non-target organisms (NTOs). However, the majority of previous risk assessments only examined one generation of NTOs using several physiological indicators, which cannot comprehensively detect some potential sub-lethal effects at the molecular level. In this study, we conducted a laboratory-based, multi-generational risk assessment of Cry1F for the collembolan Folsomia candida, an important representative of soil arthropods in terms of survival, reproduction, and differentially expressed genes (DEGs) identified from whole transcriptome profiles. Our results demonstrated that Cry1F was continuously ingested by collembolans over three consecutive generations, but it did not affect the survival or reproduction of F. candida. There were no significant differences in the global gene expression between F. candida—fed diets with and without Cry1F, and no consistent co-expressed DEGs over three generations. In addition, Cry1F did not obviously alter the expression profiles of seven sensitive biological markers. Our composite data indicates that Cry1F had no long-term harmful effects on collembolan F. candida.

The adsorption removal of quinoline from model diesel by using alumina, diatomite, silica gel, and allochroic silica gel as adsorbents was investigated. The experiment results indicated that the adsorption denitrogenation performance of allochroic silica gel was more superior to other three adsorbents. The silica gel and allochroic silica gel were confirmed by characterization with X-ray diffraction, nitrogen adsorption–desorption, and ammonia temperature programmed desoption (NH 3 -TPD). X-ray diffraction results indicated that both the samples were amorphous structures. The average pore diameters of silica gel and allochroic silica gel were 18.46 and 1.80 nm, the Brunauer–Emmett–Teller surface areas were 437.86 and 623.39 m 2 /g, and the pore volumes were 0.9724 and 0.3442 cm 3 /g, respectively. The results of TPD showed that the acidity of allochroic silica gel was far stronger than that of silica gel which greatly enhanced its adsorption denitrogenation performance. The adsorption denitrogenation performance of allochroic silica gel for quinoline, aniline, and pyridine from model diesel was as follows: aniline, pyridine, and quinoline. Adsorption temperature, particle size, and arenes added in model diesel had little impact on the removal of aniline and pyridine except quinoline. The adsorbent-to-oil ratio had significant effects on adsorption denitrogenation, especially for quinoline. The N–Co bond between Co in allochroic silica gel and N atom in the basic nitrogen compounds molecule played a significant role. Furthermore, the allochroic silica gel could be easily regenerated by its adsorption denitrogenation performance for quinoline and pyridine by using calcination once or several times, except aniline. The adsorption isotherm results revealed that the adsorption of pyridine and aniline belonged to the Langmuir–Freundlich binding model, but the adsorption of quinoline belonged to Freundlich model.

Interactions between plants and insect herbivores are important determinants of plant productivity in cultivated and natural agricultural fields. The rice leaf folder (Cnaphalocrocis medinalis) causes tremendous damage to rice production in Asian countries. However, little information is available about how rice plants defend themselves against this destructive pest at molecular and biochemical levels. Here, we observed the transcriptomic and metabolomic differences in rice leaves after 0, 1, 6, 12, and 24 h of being fed by C. medinalis using RNA sequencing and metabolome profiling. Transcriptional analyses showed that gene expression responds rapidly to leaf folder infestation, with the most significant transcriptional changes occurring within 6 h after the initiation of feeding. Metabolite abundance changed more slowly than gene expression. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses indicated that the rice transcriptional response to infestation involved genes encoding protein kinases, transcription factors, biosynthesis of secondary metabolites, photosynthesis, and phytohormone signaling. Moreover, the jasmonic acid-dependent signaling pathway triggered by leaf folder herbivory played a vital role in rice defense against this pest. Taken together, our results provide comprehensive insights into the defense system of rice to this species and may inform the development of insect-resistant rice varieties.

A mango ETHYLENE RESPONSE1 (ETR1) gene, designated MiETR1b, was isolated from the cotyledon of mango (Mangifera indica L. ‘Zihua’) using RT-PCR, and the 5′ and 3′ rapid amplification of cDNA ends. The full-length cDNA was 2 530 bp, with an open reading frame of 2 220 bp, and it encoded a putative protein of 739 amino acids. The genomic DNA sequence of MiETR1b was 4 116 bp in length, having a 3 305 bp sequence from the start to terminator codon, containing six exons and five introns. The deduced amino acids possessed conserved domains of the GAF and HATPase_c superfamilies. A phylogenetic tree analysis indicated that MiETR1b had the highest similarity to MiETR1 from M. indica and a high similarity to CsERS1, DlETR1, TcERS1 and PtETR1. Quantitative real-time PCR showed that MiETR1b was expressed in the proximal and distal cut surfaces throughout the adventitious root formation period. Meanwhile, the expression of MiETR1b in the distal cut surface was significantly up-regulated within 6–48 h. Pre-treatments with indole-3-butyric acid and 2,3,5-triiodobenzoic acid significantly down-regulated MiETR1b expression at 1 d and 6 h, respectively. However, more ethylene was produced from 12 to 24 h, while ethylene production decreased after 4 days of culturing. In conclusion, MiETR1b might play an important role during the adventitious root formation of mango cotyledon segments, which is related to ethylene production.

Coccidiosis is one of the most economically important diseases affecting the poultry industry. Currently, anticoccidial drugs used in veterinary clinics show many deficiencies, and new control measures are urgently needed. This study presents an anticoccidial herbal powder “Shi Yin Zi”, which consists of Cnidium monnieri (L.) Cuss, Taraxacum mongolicum Hand.-Mazz., and sodium chloride. In chickens infected with Eimeria tenella, supplementation with “Shi Yin Zi” powder for 3 d prior to infection or treatment with “Shi Yin Zi” powder after infection could improve the survival rate and relative growth rate and alleviate the pathological changes in the cecum, liver, and kidney. “Shi Yin Zi” powder could recover the levels of alanine aminotransferase, creatinine, albumin, and triglycerides in serum. The hemorrhage occurrence and total number of oocysts in feces were reduced. The anti-coccidial indexes reached 165 for the prophylactic effect and 144 for the therapeutic effect. The anti-coccidial effects were equal to positive controls (monensin and sulfamlopyrazine). These results suggest that “Shi Ying Zi” powder possesses a potent anticoccidial effect and exhibits the potential to control E. tenella infection.

Winter wheat expressing the sucrose transporter HvSUT1 from barley (HOSUT) has an increased yield potential. Genetic engineering should improve cultivars without increasing susceptibility to biotic stresses or causing negative impacts on ecosystem services. We studied the effects of HOSUT wheat on cereal aphids that feed on the sugar-rich phloem sap. Three HOSUT winter wheat lines, their conventional parental cultivar Certo, and three conventional cultivars were used. Clip cage experiments in the greenhouse showed no differences in life-table parameters of Rhopalosiphum padi and Sitobion avenae (Hemiptera: Aphididae) on transgenic lines compared to Certo, except higher fecundity of S. avenae on one HOSUT line. Population development of both aphid species over three weeks on caged flowering tillers did not reveal differences between the HOSUT lines and Certo. When aphids were monitored in a Swiss field study over two years, no differences between HOSUT lines and Certo were observed. We conclude that HOSUT wheat did not have consistent effects on aphids compared to the parental cultivar and measured parameters were generally in the range observed for the conventional winter wheat cultivars. Thus, HOSUT wheat is unlikely to suffer from increased aphid damage.

DNA methylation changes can occur in some loci during callus culture, resulting in somaclonal variations (SVs). In the present study, we applied whole genome bisulfite sequencing to analyze context-specific DNA methylation changes in the pineapple genome between the cutting seedings and 5 SV plants. In general, SV plants exhibited methylation patterns analogous to those of cutting seedlings (CK). A total of 27.98% of the genomic cytosines of CK were methylcytosines, which was higher than that of 5 SV plants. Moreover, mCG and mCHG was hypermethylated, whereas mCHH was hypomethylated among the 5 SV plants genomic when compared with the CK. Most of the variation of DNA methylation was distributed in gene bodies, thus suggesting that phenotypic differences are probably perturbed by genes methylated from callus culture. In addition, the methylated genes were highly enriched for the Gene Ontology (GO) categories of binding and catalytic activity, cell part and organelle, cellular process, abiotic stimulus, and DNA modification. These results suggest that methylation mediates these pathways in the callus culture of pineapple. The results also suggested that the callus culture induced DNA methylation may result in the SV.

Assessing the potential effects of insect-resistant genetically engineered (GE) plants on collembolans is important because these common soil arthropods may be exposed to insecticidal proteins produced in GE plants by ingestion of plant residues, crop pollen, or root exudates. Laboratory studies were conducted to evaluate the potential effects of two Bacillus thuringiensis (Bt)-rice lines expressing Cry1C and Cry2A in pollen and leaves and of their non-Bt conventional isolines on the fitness of the collembolan Folsomia candida and on the activities of its antioxidant-related enzymes, superoxide dismutase and peroxidase, and of its detoxification-related enzymes, glutathione reductase and glutathione S-transferase. Survival, development, reproduction, and the intrinsic rate of increase (rm) were not significantly reduced when F. candida fed on the Bt rice pollen or leaf powder than on the non-Bt rice materials; these parameters, however, were significantly reduced when F. candida fed on non-Bt rice pollen or non-Bt leaf-based diets containing the protease inhibitor E-64 at 75 μg/g. The activities of the antioxidant-related and detoxification-related enzymes in F. candida were not significantly affected when F. candida fed on the Bt rice materials, but were significantly increased when F. candida fed on the non-Bt rice materials containing E-64. The results demonstrate that Cry1C and Cry2A are not toxic to F. candida, and also indicate the absence of unintended effects on the collembolan caused by any change in plant tissue nutritional composition due to foreign gene transformation.

The ladybird Propylea japonica, adults of the green lacewing Chrysoperla nipponensis and the honey bee Apis mellifera are common pollen feeders in many crop systems. They could therefore be directly exposed to Cry proteins in Bacillus thuringiensis (Bt)-transgenic crop fields by ingestion of pollen. They, or closely related species, are therefore often selected as surrogate test species in non-target risk assessment of Bt plants. In the current study, we evaluated the potential effects of the ingestion of Bt maize pollen containing the Cry1Ab/Cry1Ac fusion protein on various life-table parameters of the three pollen-feeding non-target species in laboratory feeding assays. The results showed that pupation rate and male adult fresh weight of P. japonica were significantly increased when fed pollen from Bt maize compared to control maize pollen, but other test life-table parameters were not affected. For the other two species, none of the tested life-table parameters (survival, pre-oviposition period, fecundity and adult fresh weight for C. nipponensis; survival and mean acinus diameter of hypopharyngeal glands for A. mellifera) differed between non-Bt and Bt maize pollen treatments. ELISA measurements confirmed the stability and uptake of the Cry protein by all three species during the feeding bioassays. In addition, a sensitive insect bioassay confirmed the bioactivity of the Cry1Ab/Cry1Ac protein in the Bt maize pollen used. Overall, the results suggested that the three pollen feeders are not sensitive to the Cry1Ab/Cry1Ac protein, and planting of the Bt maize variety will pose a negligible risk to P. japonica, adult C. nipponensis and adult A. mellifera.

In fields of genetically modified, insect-resistant rice expressing Bacillus thuringiensis (Bt) proteins, frogs are exposed to Bt Cry proteins by consuming both target and non-target insects, and through their highly permeable skin. In the present study, we assessed the potential risk posed by transgenic cry1Ca rice (T1C-19) on the development of a frog species by adding purified Cry1Ca protein or T1C-19 rice straw into the rearing water of Xenopus laevis tadpoles, and by feeding X. laevis froglets diets containing rice grains of T1C-19 or its non-transformed counterpart MH63. Our results showed that there were no significant differences among groups receiving 100 μg L-1 or 10 μg L-1 Cry1Ca and the blank control in terms of time to completed metamorphosis, survival rate, body weight, body length, organ weight and liver enzyme activity after being exposed to the Cry1Ca (P > 0.05). Although some detection indices in the rice straw groups were significantly different from those of the blank control group (P < 0.05), there was no significant difference between the T1C-19 and MH63 rice straw groups. Moreover, there were no significant differences in the mortality rate, body weight, daily weight gain, liver and fat body weight of the froglets between the T1C-19 and MH63 dietary groups after 90 days, and there were no abnormal pathological changes in the stomach, intestines, livers, spleens and gonads. Thus, we conclude that the planting of transgenic cry1Ca rice will not adversely affect frog development.

Propylea japonica (Thunberg) (Coleoptera: Coccinellidae) are prevalent predators and pollen feeders in East Asian maize fields. They are therefore indirectly (via prey) and directly (via pollen) exposed to Cry proteins within Bt-transgenic maize fields. The effects of Cry1Ie-producing transgenic maize pollen on the fitness of P. japonica was assessed using two dietary-exposure experiments in the laboratory. In the first experiment, survival, larval developmental time, adult fresh weight, and fecundity did not differ between ladybirds consuming Bt or non-Bt maize pollen. In the second experiment, none of the tested lethal and sublethal parameters of P. japonica were negatively affected when fed a rapeseed pollen-based diet containing Cry1Ie protein at 200 μg/g dry weight of diet. In contrast, the larval developmental time, adult fresh weight, and fecundity of P. japonica were significantly adversely affected when fed diet containing the positive control compound E-64. In both experiments, the bioactivity of the Cry1Ie protein in the food sources was confirmed by bioassays with a Cry1Ie-sensitive lepidopteran species. These results indicated that P. japonica are not affected by the consumption of Cry1Ie-expressing maize pollen and are not sensitive to the Cry1Ie protein, suggesting that the growing of Bt maize expressing Cry1Ie protein will pose a negligible risk to P. japonica.

A rape seed pollen-based diet was developed and found to be suitable for use in a dietary exposure assay for Propylea japonica. Using the diet, we established and validated a dietary exposure assay by using the protease inhibitor E-64 as positive control. Dose-dependent responses were documented for all observed life-table parameters of P. japonica including survival, pupation and eclosion rates, development time and adult weight. Results suggested that the dietary assay can detect the effects of insecticidal compounds on the survival and development of P. japonica. Using the established dietary assay, we subsequently tested the toxicity of Cry1Ab, Cry1Ac and Cry1F proteins that are expressed by transgenic maize, cotton or rice plants to P. japonica larvae. The diet containing E-64 was included as a positive control. Survival and development of P. japonica larvae were not adversely affected when the diet contained purified Cry1Ab, Cry1Ac, or Cry1F at 500 µg/g diet representing a worst-case exposure scenario. In contrast, P. japonica larvae were adversely affected when the diet contained E-64. The bioactivity and stability of the Cry proteins in the diet and Cry protein uptake by the ladybird larvae were confirmed by bioassay with a Cry-sensitive insect species and by ELISA. The current study describes a suitable experimental system for assessing the potential effects of gut-active insecticidal compounds on ladybird beetle larvae. The experiments with the Cry proteins demonstrate that P. japonica larvae are not sensitive to Cry1Ab, Cry1Ac and Cry1F.

Soybean tissue and arthropods were collected in Bt soybean fields in China at different times during the growing season to investigate the exposure of arthropods to the plant-produced Cry1Ac toxin and the transmission of the toxin within the food web. Samples from 52 arthropod species/taxa belonging to 42 families in 10 orders were analysed for their Cry1Ac content using enzyme-linked immunosorbent assay (ELISA). Among the 22 species/taxa for which three samples were analysed, toxin concentration was highest in the grasshopper Atractomorpha sinensis and represented about 50% of the concentration in soybean leaves. Other species/taxa did not contain detectable toxin or contained a concentration that was between 1 and 10% of that detected in leaves. These Cry1Ac-positive arthropods included a number of mesophyll-feeding Hemiptera, a cicadellid, a curculionid beetle and, among the predators, a thomisid spider and an unidentified predatory bug belonging to the Anthocoridae. Within an arthropod species/taxon, the Cry1Ac content sometimes varied between life stages (nymphs/larvae vs. adults) and sampling dates (before, during, and after flowering). Our study is the first to provide information on Cry1Ac-expression levels in soybean plants and Cry1Ac concentrations in non-target arthropods in Chinese soybean fields. The data will be useful for assessing the risk of non-target arthropod exposure to Cry1Ac in soybean.

By consuming mulberry leaves covered with pollen from nearby genetically engineered, insect-resistant rice lines producing Cry proteins derived from Bacillus thuringiensis (Bt), larvae of the domestic silkworm, Bombyx mori (Linnaeus) (Lepidoptera: Bombyxidae), could be exposed to insecticidal proteins. Laboratory experiments were conducted to assess the potential effects of Cry1C- or Cry2A-producing transgenic rice (T1C-19, T2A-1) pollen on B. mori fitness. In a short-term assay, B. mori larvae were fed mulberry leaves covered with different densities of pollen from Bt rice lines or their corresponding near isoline (control) for the first 3 d and then were fed mulberry leaves without pollen. No effect was detected on any life table parameter, even at 1800 pollen grains/cm(2) leaf, which is much higher than the mean natural density of rice pollen on leaves of mulberry trees near paddy fields. In a long-term assay, the larvae were fed Bt and control pollen in the same way but for their entire larval stage (approximately 27 d). Bt pollen densities ≥ 150 grains/cm(2) leaf reduced 14-d larval weight, increased larval development time, and reduced adult eclosion rate. ELISA analyses showed that 72.6% of the Cry protein was still detected in the pollen grains excreted with the feces. The low exposure of silkworm larvae to Cry proteins when feeding Bt rice pollen may be the explanation for the relatively low toxicity detected in the current study. Although the results demonstrate that B. mori larvae are sensitive to Cry1C and Cry2A proteins, the exposure levels that harmed the larvae in the current study are far greater than natural exposure levels. We therefore conclude that consumption of Bt rice pollen will pose a low to negligible risk to B. mori.

A class of stochastic set differential equations (SSDEs) with non-Lipschitzian coefficients is investigated. We first give the preliminaries on the stochastic set differential equations. Then the nonexplosion of solutions to the SSDEs is discussed. Moreover, the existence and uniqueness of the solutions to SSDEs are proven. Finally, the continuous dependence of the solutions to SSDEs is studied.

Rice and maize are important cereal crops that serve as staple foods, feed, and industrial material in China. Multiple factors constrain the production of both crops, among which insect pests are an important one. Lepidopteran pests cause enormous yield losses for the crops annually. In order to control these pests, China plays an active role in development and application of genetic engineering (GE) to crops, and dozens of GE rice and GE maize lines expressing insecticidal proteins from the soil bacterium Bacillus thuringiensis (Bt) have been developed. Many lines have entered environmental release, field testing, and preproduction testing, and laboratory and field experiments have shown that most of the Bt rice and Bt maize lines developed in China exhibited effective control of major target lepidopteran pests on rice (Chilo suppressalis, Scirpophaga incertulas, and Cnaphalocrocis medinalis) and maize (Ostrinia furnacalis), demonstrating bright prospects for application. However, none of these Bt lines has yet been commercially planted through this writing in 2016. Challenges and perspectives for development and application of Bt rice and maize in China are discussed. This article provides a general context for colleagues to learn about research and development of Bt crops in China, and may shed light on future work in this field.

Crystal (Cry) proteins derived from Bacillus thuringiensis (Bt) have been widely used in transgenic crops due to their toxicity against insect pests. However, the distribution and metabolism of these toxins in insect tissues and organs have remained obscure because the target insects do not ingest much toxin. In this study, several Cry1Ac-resistant strains of Helicoverpa armigera, fed artificial diets containing high doses of Cry1Ac toxin, were used to investigate the distribution and metabolism of Cry1Ac in their bodies. Cry1Ac was only detected in larvae, not in pupae or adults. Also, Cry1Ac passed through the midgut into other tissues, such as the hemolymph and fat body, but did not reach the larval integument. Metabolic tests revealed that Cry1Ac degraded most rapidly in the fat body, followed by the hemolymph, peritrophic membrane and its contents. The toxin was metabolized slowly in the midgut, but was degraded in all locations within 48 h. These findings will improve understanding of the functional mechanism of Bt toxins in target insects and the biotransfer and the bioaccumulation of Bt toxins in arthropod food webs in the Bt crop ecosystem.

DNA microarray analysis is an effective method to detect unintended effects by detecting differentially expressed genes (DEG) in safety assessment of genetically modified (GM) crops. With the aim to reveal the distribution of DEG of GM crops under different conditions, we performed DNA microarray analysis using transgenic rice Huahui 1 (HH1) and its non-transgenic parent Minghui 63 (MH63) at different developmental stages and environmental conditions. Considerable DEG were selected in each group of HH1 under different conditions. For each group of HH1, the number of DEG was different; however, considerable common DEG were shared between different groups of HH1. These findings suggested that both DEG and common DEG were adequate for investigation of unintended effects. Furthermore, a number of significantly changed pathways were found in all groups of HH1, indicating genetic modification caused everlasting changes to plants. To our knowledge, our study for the first time provided the non-uniformly distributed pattern for DEG of GM crops at different developmental stages and environments. Our result also suggested that DEG selected in GM plants at specific developmental stage and environment could act as useful clues for further evaluation of unintended effects of GM plants.

The ladybird beetle, Coleomegilla maculata (DeGeer), is a common and abundant predator in many cropping systems. Its larvae and adults are predaceous, feeding on aphids, thrips, lepidopteran larvae and plant tissues, such as pollen. Therefore, this species is exposed to insecticidal proteins expressed in insect-resistant, genetically engineered cotton expressing Cry proteins derived from Bacillus thuringiensis (Bt). A tritrophic bioassay was conduced to evaluate the potential impact of Cry2Ab- and Cry1Ac-expressing cotton on fitness parameters of C. maculata using Bt-susceptible and -resistant larvae of Trichoplusia ni as prey. Coleomegilla maculata survival, development time, adult weight and fecundity were not different when they were fed with resistant T. ni larvae reared on either Bt or control cotton. To ensure that C. maculata were not sensitive to the tested Cry toxins independent from the plant background and to add certainty to the hazard assessment, C. maculata larvae were fed artificial diet incorporated with Cry2Ab, Cry1Ac or both at >10 times higher concentrations than in cotton tissue. Artificial diet containing E-64 was included as a positive control. No differences were detected in any life-table parameters between Cry protein-containing diet treatments and the control diet. In contrast, larvae of C. maculata fed the E-64 could not develop to the pupal stage and the 7-d larval weight was significantly negatively affected. In both feeding assays, the stability and bioactivity of Cry proteins in the food sources were confirmed by ELISA and sensitive-insect bioassays. Our results show that C. maculata is not affected by Bt cotton and is not sensitive to Cry2Ab and Cry1Ac at concentrations exceeding the levels in Bt cotton, thus demonstrating that Bt cotton will pose a negligible risk to C. maculata. More importantly, this study demonstrates a comprehensive system for assessing the risk of genetically modified plants on non-target organisms.

Adults of the common green lacewing, Chrysoperla carnea (Stephens) (Neuroptera: Chrysopidae), are prevalent pollen-consumers in maize fields. They are therefore exposed to insecticidal proteins expressed in the pollen of insect-resistant, genetically engineered maize varieties expressing Cry proteins derived from Bacillus thuringiensis (Bt). Laboratory experiments were conducted to evaluate the impact of Cry3Bb1 or Cry1Ab-expressing transgenic maize (MON 88017, Event Bt176) pollen on fitness parameters of adult C. carnea. Adults were fed pollen from Bt maize varieties or their corresponding near isolines together with sucrose solution for 28 days. Survival, pre-oviposition period, fecundity, fertility and dry weight were not different between Bt or non-Bt maize pollen treatments. In order to ensure that adults of C. carnea are not sensitive to the tested toxins independent from the plant background and to add certainty to the hazard assessment, adult C. carnea were fed with artificial diet containing purified Cry3Bb1 or Cry1Ab at about a 10 times higher concentration than in maize pollen. Artificial diet containing Galanthus nivalis agglutinin (GNA) was included as a positive control. No differences were found in any life-table parameter between Cry protein containing diet treatments and control diet. However, the pre-oviposition period, daily and total fecundity and dry weight of C. carnea were significantly negatively affected by GNA-feeding. In both feeding assays, the stability and bioactivity of Cry proteins in the food sources as well as the uptake by C. carnea was confirmed. These results show that adults of C. carnea are not affected by Bt maize pollen and are not sensitive to Cry1Ab and Cry3Bb1 at concentrations exceeding the levels in pollen. Consequently, Bt maize pollen consumption will pose a negligible risk to adult C. carnea.
