Abstract Shiga toxin-producing Escherichia coli (STEC) can cause mild diarrhea even severe hemolytic uremic syndrome (HUS). Shiga toxin (Stx) is the primary virulence factor. Two Stx types and several subtypes have been identified. STEC strains encoding stx2f (Stx2f-STECs) are frequently identified from pigeons. Stx2f was initially considered to be associated with mild symptoms, more recently Stx2f-STECs have been isolated from HUS cases, indicating their pathogenic potential. Here, we investigated the prevalence of Stx2f-STECs among domestic pigeons in two regions in China, characterized the strains using whole-genome sequencing (WGS), and assessed the Stx2f transcriptions. Thirty-two Stx2f-STECs (4.36%) were culture-positive out of 734 fecal samples (one strain per sample). No other stx subtype-containing strain was isolated. Four serotypes and two sequence types were determined, and a novel sequence type ST15057 was identified. All strains harbored the E. coli attaching and effacing gene eae. Two types of Stx2f prophages were assigned. Stx2f-STECs showed variable Stx transcription levels induced by mitomycin C. Whole genome single-nucleotide polymorphism (wgSNP) analysis revealed different genetic backgrounds between pigeon-derived strains and those from diarrheal or HUS patients. In contrast, pigeon-derived Stx2f-STECs from diverse regions exhibited genetic similarity. Our study reports the prevalence and characteristics of Stx2f-STECs from pigeons in China. The pigeon-derived strains might pose low public health risk.

Abstract Glutaraldehyde (GTA) crosslinking is commonly used to improve the thermal stability and mechanical strength of collagen casings. The aim of this research was to determine the optimal pH of the crosslinking between GTA and collagen as well as the crosslinking mechanisms. The weakly alkaline environment could facilitate the generation of GTA polymerization through the rapid generation of -C = C-C = O and -N = C-C = C- conjugated structures, and enhance the crosslinking reaction of GTA polymers with collagen amino groups. In the pH range of 8–10, the fibril diameter and d-space value declined significantly in the self-assembled collagen fibril-GTA system. Meanwhile, collagen casing films crosslinked with GTA in weakly alkaline conditions exhibited higher mechanical strength and thermal stability. These results suggest that the crosslinking of collagen casings and GTA can be improved by adjusting the pH. Possible crosslinking mechanisms related to the formation of conjugated long chains have also been proposed. This study could provide guidance on the appropriate use of GTA in the production process of collagen casings. Graphical Abstract

Abstract MicroRNA (miRNA) is a type of endogenous non-coding small RNA, which is abundant in living organisms. miRNAs play an important role in regulating gene expression and myriad cellular processes by binding to target messenger RNAs through complementary base pairing, and cross-species regulation mammalian cells by plant-derived xeno-miRNAs has been described. Here, we examined the miRNA species in two alfalfa (Medicago sativa, lucerne) cultivars commonly grown in Ningxia, China: cv. Zhongmu 1 and cv. Xinyan 52. Both cultivars have good salt and drought resistance. We found that the miRNA profiles were similar between the cultivars, with a slightly higher number of miRNAs present in the newer cv. Xinyan 52, which may contribute to its improved salt and drought tolerance. miRNAs were stable during drying, and some miRNAs were increased in dry versus fresh alfalfa, suggesting some miRNAs may be upregulated during drying. Alfalfa-derived miRNAs could be detected in exosomes from serum and whey collected from dairy cows, confirming the ability of the exogenous miRNAs (xeno-miRNAs) to enter the circulation and reach the mammary epithelium. In vitro studies confirmed that overexpression of mtr-miR156a could downregulate expression of Phosphatase 2 Regulatory Subunit B'gamma ( PPP2R5D) and Phosphoinositide-3-kinase Regulatory Subunit 2 (PIK3R2). Overexpression of mtr-miR156a also modulated PI3K–AKT–mTOR signaling as well as the casein content of milk produced by bovine mammary epithelial cells. Based on the known roles of PPP2R5D and PIK3R2 in regulating the PI3K–AKT–mTOR pathway as well as the effect of PI3K–AKT–mTOR on milk protein content, our findings implicate alfalfa-derived miR156a as a new cross-species regulator of milk quality in dairy cows.

Abstract Objectives Serum/glucocorticoid-inducible kinase 1 (SGK1) gene encodes a serine/threonine protein kinase that plays an essential role in cellular stress response and regulation of multiple metabolic processes. However, its role in bovine adipogenesis remains unknown. In this study, we aimed to clarify the role of SGK1 in bovine lipid accumulation and improvement of meat quality. Methods Preadipocytes were induced to differentiation to detect the temporal expression pattern of SGK1. Heart, liver, lung, spleen, kidney, muscle and fat tissues were collected to detect its tissue expression profile. Recombinant adenovirus and the lentivirus were packaged for overexpression and knockdown. Oil Red O staining, quantitative real-time PCR, Western blot analysis, Yeast two-hybrid assay, luciferase assay and RNA-seq were performed to study the regulatory mechanism of SGK1. Results SGK1 showed significantly higher expression in adipose and significantly induced expression in differentiated adipocytes. Furthermore, overexpression of SGK1 greatly promoted adipogenesis and inhibited proliferation, which could be shown by the remarkable increasement of lipid droplet, and the expression levels of adipogenic marker genes and cell cycle-related genes. Inversely, its knockdown inhibited adipogenesis and facilitated proliferation. Mechanistically, SGK1 regulates the phosphorylation and expression of two critical proteins of FoxO family, FOXO1/FOXO3. Importantly, SGK1 attenuates the transcriptional repression role of FOXO1 for PPARγ via phosphorylating the site S256, then promoting the bovine fat deposition. Conclusions SGK1 is a required epigenetic regulatory factor for bovine preadipocyte proliferation and differentiation, which contributes to a better understanding of fat deposition and meat quality improvement in cattle.

Abstract Background A knowledge of gouty arthritis could help in the primary prevention of the disease development and lead to an early diagnosis if it occurs. This study investigated the knowledge, attitudes, and practices (KAP) toward gouty arthritis in the general population > 30 years old. Methods This web-based cross-sectional study was conducted among the general population > 30 years old between January and March 2023 in Chengdu, Sichuan. The questionnaire was designed by the investigators based on the available guidelines (Cronbach’s α = 0.846). A score above 70% indicated good knowledge, a positive attitude, and proactive practice. Multivariable and structural equation modeling (SEM) analyses were performed to analyze the factors influencing KAP. Results A total of 537 questionnaires were included. The knowledge, attitudes, and practices scores were 13.12 ± 6.41, 25.28 ± 3.97, and 45.25 ± 5.77, respectively. Abstract Objective To study the effects of chemotherapy on cognitive function in breast cancer patients, and to investigate the relationship of MemTrax test of memory and related functions to the FACT-Cog functional self-assessment for the evaluation and management of chemobrain. Methods In this prospective cohort study, clinical information of pathologically confirmed female breast cancer patients who decided to receive chemotherapy were collected in a questionnaire which was developed for this study and provided as a supplementary file. The FACT-Cog self-assessment and MemTrax test were administered before and after the chemotherapy treatments. Patients with chemobrain were identified using published criteria based on FACT-Cog scores, and MemTrax scores from chemobrain patients were analyzed. Results Fifty-six patients participated in this study, of which 41 participants completed 4 or more cycles of chemotherapy and were included in the final analyses here. Using the reported high end of minimal clinical differences (10.6 points) of FACT-Cog before and after chemotherapy, 18 patients suffered from chemobrain in this study. In these 18 chemobrain patients, no cognitive impairments were detected by MemTrax, which paradoxically demonstrated an improvement in the normal cognitive range. Conclusion The cognitive impairment induced by chemotherapy in breast cancer patients is detectable by the FACT-Cog in a Chinese cohort but is not detected by the MemTrax memory test. The fact that the more objective MemTrax could not detect the impairment could alleviate patients’ concerns which in turn would be beneficial for patients’ mental health.

Renal interstitial fibrosis (RIF) is a significant pathological change in diabetic kidney disease (DKD) that can be induced by endothelial-to-mesenchymal transition (EndMT). Lymphangiogenesis, mediated by the vascular endothelial growth factor-C (VEGF-C)/vascular endothelial growth factor receptor-3 (VEGFR-3) pathway, plays a crucial role in the development of RIF in DKD. Although numerous studies have demonstrated the efficacy of empagliflozin in treating renal injury, its effects on lymphangiogenesis in DKD-related RIF and the underlying mechanisms remain unclear. In the present study, significant lymphangiogenesis was assessed in the renal interstitium of patients with DKD. We subsequently explored the relationship between DKD-related RIF and lymphangiogenesis in mouse models, high-glucose (HG)-stimulated renal HK-2 cell lines, and human lymphatic endothelial cells (hLECs). Additionally, we evaluated the effects of empagliflozin on these processes. The results revealed that HG induces lymphangiogenesis, which exacerbates RIF by promoting inflammatory responses. Furthermore, hLECs directly contributed to the progression of DKD-related RIF through EndMT. Further analysis revealed that tubular epithelial cells (TECs) act as effector cells for VEGF-C, with the epithelial-to-mesenchymal transition (EMT) of TECs occurring concurrently with the EndMT of lymphatic vessels. Empagliflozin inhibited RIF in DKD by suppressing the VEGF-C/VEGFR3 pathway and reducing lymphangiogenesis. In conclusion, this study elucidates the interplay between lymphangiogenesis, EndMT, and RIF in DKD and provides new insights into the mechanism by which empagliflozin treats DKD.

Goji berries are a small-fruited shrub with industrial importance whose fruit considered beneficial in both fresh and dried forms. Current germplasms of goji berries include small fruits with a short shelf life, less sweet and bitter taste, and a lack of appropriate genetic information. This study aimed to employ whole genome resequencing to generate an ultra-dense bin linkage map and to elucidate the genetic basis of goji fruit quality and size using quantitative trait loci (QTL) mapping analysis in a cross-pollinated hybrid population. To achieve this goal, human sensory tests were carried out to determine the bitter taste (BT) and sweet taste (ST), and to quantify the soluble solid content (SSC), fruit firmness (FF), and fruit size-related traits of fresh goji fruits over three or four years. The results revealed that the goji bin linkage map based on resequencing spanned a total length of 966.42 cM and an average bin interval of 0.03 cM. Subsequent variant calling and ordering resulted in 3,058 bins containing 35,331 polymorphic markers across 12 chromosomes. A total of 99 QTLs, with individual loci in different environments explaining a phenotypic variance of 1.21-16.95% were identified for the studied traits. Ten major effects, including colocalized QTLs corresponding to different traits, were identified on chromosomes 1, 3, 5, 6, 7, and 8, with a maximum Logarithm of Odds (LOD) of 29.25 and 16.95% of explained phenotypic variance (PVE). In addition, four stable loci, one for FF, one for fruit weight (FW), and two for fruit shape index (FSI), were mainly mapped on chromosomes 5, 6, and 7, elucidating 2.10-16.95% PVE. These findings offer valuable insights into the genetic architecture of goji fruit traits along with identified specific loci and markers to further improve and develop sweeter, less bitter and larger fruited goji berry cultivars with extended shelf life.

Abstract Milk protein content is an important index to evaluate the quality and nutrition of milk. Accumulating evidence suggests that microRNAs (miRNAs) play important roles in bovine lactation, but little is known regarding the cross-kingdom regulatory roles of plant-derived exogenous miRNAs (xeno-miRNAs) in milk protein synthesis, particularly the underlying molecular mechanisms. The purpose of this study was to explore the regulatory mechanism of alfalfa-derived xeno-miRNAs on proliferation and milk protein synthesis in bovine mammary epithelial cells (BMECs). Our previous study showed that alfalfa miR159a (mtr-miR159a, xeno-miR159a) was highly expressed in alfalfa, and the abundance of mtr-miR159a was significantly lower in serum and whey from high-protein-milk dairy cows compared with low-protein-milk dairy cows. In this study, mRNA expression was detected by real-time quantitative PCR (qRT-PCR), and casein content was evaluated by enzyme-linked immunosorbent assay (ELISA). Cell proliferation and apoptosis were detected using the cell counting kit 8 (CCK-8) assay, 5-ethynyl-2′-deoxyuridine (EdU) staining, western blot, and flow cytometry. A dual-luciferase reporter assay was used to determine the regulation of Protein Tyrosine Phosphatase Receptor Type F (PTPRF) by xeno-miR159a. We found that xeno-miR159a overexpression inhibited proliferation of BMEC and promoted cell apoptosis. Besides, xeno-miR159a overexpression decreased β-casein abundance, and increased α-casein and κ-casein abundance in BMECs. Dual-luciferase reporter assay result confirmed that PTPRF is a target gene of xeno-miR159a. These results provide new insights into the mechanism by which alfalfa-derived miRNAs regulate BMECs proliferation and milk protein synthesis.

Abstract Background Hox gene family is an important transcription factor that regulates cell process, and plays a role in the process of adipocytes differentiation and fat deposition. Previous transcriptome sequencing studies have indicated that the Homeobox A9 gene (HOXA9) is a candidate gene for regulating the process of bovine lipid metabolism, but the function and specific mechanism of action remain unclear. Therefore, this study aims to explore the role of HOXA9 in the proliferation, differentiation and apoptosis of bovine preadipocytes through gain-of-function and lose-of-function. Result It found HOXA9 highly expressed in bovine adipose tissue, and its expression level changed significantly during adipocytes differentiation process. It gave a hint that HOXA9 may be involved in the process of bovine lipid metabolism. The results of HOXA9 gain-of-function experiments indicated that HOXA9 appeared to act as a negative regulator not only in the differentiation but also in the proliferation of bovine preadipocytes, which is mainly reflected that overexpression of HOXA9 down-regulate the mRNA and protein expression level of PPARγ, CEBPα and FABP4 (P

Objective It was shown that microRNAs (miRNAs) play an important role in milk protein synthesis. However, the post-transcriptional regulation of casein expression by exogenous miRNA (xeno-miRNAs) in ruminants remains unclear. This study explores the regulatory roles of alfalfa xeno-miR162 on casein synthesis in bovine mammary epithelial cells (bMECs). Methods The effects of alfalfa xenomiR-162 and G protein subunit gamma 11 (GNG11) on proliferation and milk protein metabolism of bMECs were detected by 5-Ethynyl-2′-Deoxyuridine (EdU) staining, flow cytometry, cell counting kit-8 (CCK-8), enzyme-linked immunosorbent assay, quantitative real-time polymerase chain reaction (qRT-PCR), and Western blot. Dual-luciferase reporter assay was used to verify the targeting relationship between GNG11 and xenomiR-162. Results Results showed that over-expression of xenomiR-162 inhibited cell proliferation but promoted apoptosis, which also up-regulated the expression of several casein coding genes, including CSN1S1, CSN1S2, and CSN3, while decreasing the expression of CSN2. Furthermore, the targeting relationship between GNG11 and xenomiR-162 was determined, and it was confirmed that GNG11 silencing also inhibited cell proliferation but promoted apoptosis and reduced the expression of casein coding genes and genes related to the mammalian target of rapamycin (mTOR) pathway. Conclusion Alfalfa xenomiR-162 appears to regulate bMECs proliferation and milk protein synthesis via GNG11 in the mTOR pathway, suggesting that this xeno-miRNA could be harnessed to modulate CSN3 expression in dairy cows, and increase κ-casein contents in milk.

Summary: Constipation and frailty are associated with intestinal dysbiosis. This study aims to identify intestinal microbial signatures that can differentiate between constipated elders accompanied by frailty and those without frailty. We collected stool samples from 61 participants and conducted 16S rRNA gene sequencing. Constipated patients with frailty (Constipation_F) exhibited reduced gut microbial diversities compared to constipated patients without frailty (Constipation_NF) and healthy individuals (C). From differential genera, random forest models identified 14, 8, and 5 biomarkers for distinguishing Constipation_F from Constipation_NF, Constipation_F from C, and Constipation_NF from C, respectively. Functional analysis revealed that pathways (P381-PWY and PWY-5507) related to vitamin B12 synthesis were reduced in Constipation_F, which aligns with the decreased abundances of vitamin-B12-producing Actinomyces and Akkermansia in this group. Our study unveils substantial differences in gut microbiota between constipated elders with frailty and those without, underscoring the diagnostic and therapeutic potential of genera involved in vitamin B12 synthesis.

The transformation of salvianolic acid B brought on by heat treatment recovery of ethanol eluent, which is a difficult problem in pharmaceutical technology, affects the purity of raw material when the medicinal raw material salvianolic acid B is purified by resin. Ultrasonic-assisted nanofiltration separation (UANS) was first employed to improve efficiency of resource utilization by regulating rejection and separating salvianolic acid B and rosmarinic acid from organic pharmaceutical wastewater. The rejection was related to three variables: ultrasonic power, pH, and ethanol concentration. But there were differences in the effects of variables on the rejections of salvianolic acid B and rosmarinic acid. The rejections of rosmarinic acid and salvianolic acid B showed a decreasing trend with an increase in ultrasonic power or a decrease in pH; however, when the concentration of ethanol was increased from 5 % to 35 %, the salvianolic acid B rejection increased from 84.96 % to 96.60 % and the rosmarinic acid rejection decreased from 35.09 % to 17.51 %. On the basis of response surface methodology (RSM), the optimal UANS parameters for solution conditions involving different ethanol concentrations are as follows: 10 % ethanol solution (ultrasonic power 500 W and pH 6.15), 20 % ethanol solution (ultrasonic power 500 W and pH 6.54), and 30 % ethanol solution (ultrasonic power 460 W and pH 6.34). The molecular proportions of salvianolic acid B were 10.75 %, 7.13 %, and 8.27 % in 10 %, 20 %, and 30 % ethanol wastewater, while the molecular proportions of rosmarinic acid were 40.52 %, 33.83 %, and 69.87 %, respectively. And the recoveries of salvianolic acid B in 10 %, 20 %, and 30 % ethanol wastewater were 93.56 %, 95.04 %, and 97.30 %, respectively, while the recoveries of rosmarinic acid were 3.19 %, 2.27 %, and 0.56 %. The molecular proportion and the rejection are correlated exponentially. In comparison with conventional nanofiltration separation (CNS), UANS is able to resolve the conflict between rosmarinic acid and salvianolic acid B in pharmaceutical wastewater, as well as enhance resource recycling and separation efficiency to prevent pollution of the environment from pharmaceutical wastewater. Experiments using UANS at different power intensities suggest that the ultrasonic at a power intensity of 46–50 W/L and the power density of 0.92–1.00 W/cm2 may resolve the separation conflict between rosmarinic acid and salvianolic acid B. This work suggests that UANS may be a significant advancement in the field of ultrasonic separation and has several potential uses in the water treatment industry.

Abstract Background Diabetic kidney disease (DKD) represents a microvascular complication of diabetes mellitus. Shenkang Pills (SKP), a traditional Chinese medicine formula, has been widely used in the treatment of DKD and has obvious antioxidant effect. Ferroptosis, a novel mode of cell death due to iron overload, has been shown to be associated with DKD. Nevertheless, the precise effects and underlying mechanisms of SKP on ferroptosis in diabetic kidney disease remain unclear. Methods The active components of SKP were retrieved from the Traditional Chinese Medicine Systems Pharmacology (TCMSP) database. Protein–protein interaction (PPI) network and Herb-ingredient-targets gene network were constructed using Cytoscape. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analyses were conducted utilizing the Metascape system database. Additionally, an in vivo model of DKD induced by Streptozotocin (STZ) was established to further investigate and validate the possible mechanisms underlying the effectiveness of SKP. Results We retrieved 56 compounds and identified 223 targets of SKP through the TCMSP database. Key targets were ascertained using PPI network analysis. By constructing a Herb-Ingredient-Targets gene network, we isolated the primary active components in SKP that potentially counteract ferroptosis in diabetic kidney disease. KEGG pathway enrichment analysis suggested that SKP has the potential to alleviate ferroptosis through HIF signaling pathway, thereby mitigating renal injury in DKD. In animal experiments, fasting blood glucose, 24 h urine protein, urea nitrogen and serum creatine were measured. The results showed that SKP could improve DKD. Results from animal experiments were also confirmed the efficacy of SKP in alleviating renal fibrosis, oxidative stress and ferroptosis in DKD mice. These effects were accompanied by the significant reductions in renal tissue expression of HIF-1α and HO-1 proteins. The mRNA and immunohistochemistry results were the same as above. Conclusions SKP potentially mitigating renal injury in DKD by subduing ferroptosis through the intricacies of the HIF-1α/HO-1 signaling pathway.

At present, the effects of environmental electromagnetic irradiation on the metabolism of organisms have attracted extensive attention, but the mechanism is still not clear. D-glucose plays an important role in the metabolism of organisms. In this work, the change in the optical rotation of D-glucose solution under an electrostatic field is measured experimentally, so as to explain the mechanism of the electric field-induced biological effect. The experimental results show that the electrostatic field can alter the optical rotation of D-glucose solution at different temperatures. Under the different strengths of electrostatic field, the specific rotation of D-glucose solution increases at different temperatures; the maximum increase can reach 2.07%, but the effect of temperature and electric field strength on the rotation increment is nonlinear and very complex. Further, it turns out that the proportion of α-D-glucose in solution increases by up to 3.25% under the electrostatic field, while the proportion of β-D-glucose decreases by as much as 1.75%. The experimental study confirms that electrostatic field can change the proportion of two conformation molecules (α and β-D-glucose) in D-glucose solution, which can provide a novel explanation for the mechanism of the electric field-induced biological effect.

Organic/Si hybrid solar cells have attracted considerable attention for their uncomplicated fabrication process and superior device efficiency, making them a promising candidate for sustainable energy applications. However, the efficient collection and separation of charge carriers at the organic/Si heterojunction interface are primarily hindered by the inadequate work function of poly (3,4-ethylenedioxythiophene): poly (styrenesulfonate) (PEDOT:PSS). Here, the application of a high-work-function MoO3 film onto the n-Si/PEDOT:PSS surface leads to a notable enhancement in the device’s built-in potential. This enhancement results in the creation of an inversion layer near the n-Si surface and facilitates charge separation at the interface. Simultaneously, it inhibits charge recombination at the heterojunction interface. As a result, the champion PEDOT:PSS/Si solar cell, which incorporates a MoO3 interface layer, demonstrates an efficiency of 16.0% and achieves a high fill factor of 80.8%. These findings provide a straightforward and promising strategy for promoting the collection and transmission of charge carriers at the interface of photovoltaic devices.

Fumaric acid, a naturally occurring preservative with antimicrobial properties, has been widely used in the baking industry. However, its direct addition interferes with yeast activity and negatively impacts the gluten structure. This study investigates the potential of spray-congealing as a method for encapsulating fumaric acid within solid lipid microparticles. The selection of lipid carriers and surfactants is critical, so hydrogenated palm stearin, hydrogenated rapeseed oil, and Compritol ATO 888 (glyceryl behenate) were chosen as lipid carriers, and propylene glycol monostearate and glyceryl monolaurate were utilised as surfactants with varying concentrations. Rheological properties, encapsulation efficiency, particle size, moisture content, and thermal behaviour were assessed, along with the release profiles under different temperature conditions simulating the baking process. The findings indicate that the addition of surfactants significantly impacts the viscosity and stability of the molten mixtures, which in turn affects the spray-congealing process and the release of fumaric acid. The temperature-dependent and time-dependent release profiles demonstrate the potential for customising release kinetics to suit specific applications, such as the baking industry. This study may contribute to the development of a controlled-release system that synchronises with the baking process, thereby optimising fumaric acid’s functionality while preserving the quality of baked goods.

Objective: Shenshuai Yingyang Jiaonang (SSYYJN), a traditional Chinese medicine formula, can ameliorate muscle atrophy associated with chronic kidney disease (CKD). However, its mechanisms of action remain unclear. This study is to investigate the molecular mechanisms involved in the effects of SSYYJN in ameliorating muscle atrophy associated with CKD in rats. Methods: The chemical compounds of SSYYJN were identified by UPLC-Q-Orbitrap HRMS. Considering the dose-response relationship of the identified compounds, male SD rats were randomly divided into Sham, Model, SSYYJN, and α-Keto Acid (KA) groups. Subsequently, we assessed the therapeutic and anti-ferroptotic effects of SSYYJN. Network pharmacology studies were used to predict the molecular mechanism of SSYYJN on ferroptosis and were further verified for accuracy. Results: A total of 42 active compounds were identified from SSYYJN. SSYYJN alleviated muscle atrophy caused by CKD, as evidenced by changes in body weight, serum biochemical indices, mass and histopathology of the skeletal muscle, and the levels of MuRF1. SSYYJN reduced the levels of iron, MDA, and ROS, increased the levels of GSH, NAPDH, and Gpx4. Network pharmacology analysis indicated that SSYYJN exerted anti-ferroptotic effects that were closely related to the HIF-1α signaling pathway. Molecular protein and genetic test results showed that SSYYJN increased HIF-1α protein and increased SLC7A11. Conclusions: SSYYJN attenuates muscle atrophy in CKD by inhibiting ferroptosis through the activation of the HIF-1α/SLC7A11 pathway and might be a promising traditional Chinese medicine for muscle atrophy in CKD.

Abstract Indicine cattle, also referred to as zebu (Bos taurus indicus), play a central role in pastoral communities across a wide range of agro-ecosystems, from extremely hot semiarid regions to hot humid tropical regions. However, their adaptive genetic changes following their dispersal into East Asia from the Indian subcontinent have remained poorly documented. Here, we characterize their global genetic diversity using high-quality whole-genome sequencing data from 354 indicine cattle of 57 breeds/populations, including major indicine phylogeographic groups worldwide. We reveal their probable migration into East Asia was along a coastal route rather than inland routes and we detected introgression from other bovine species. Genomic regions carrying morphology-, immune-, and heat-tolerance-related genes underwent divergent selection according to Asian agro-ecologies. We identify distinct sets of loci that contain promising candidate variants for adaptation to hot semi-arid and hot humid tropical ecosystems. Our results indicate that the rapid and successful adaptation of East Asian indicine cattle to hot humid environments was promoted by localized introgression from banteng and/or gaur. Our findings provide insights into the history and environmental adaptation of indicine cattle.

Objective Water inrush in karst tunnel has a great influence on tunnel safety. Methods Taking the Wufeng Tunnel of Yilai Expressway as the research object, the risk of water inrush in the tunnel was identified though field hydrogeological investigation, borehole water level and indoor rainfall monitoring, numerical simulation and small-scale model tests. Results The test results show that the risk of water inrush in the tunnel is mainly affected by the relative spatial position between the karst pipeline and tunnel, including the water pressure of the pipeline. The influence of seepage on the tunnel can be effectively reduced by increasing the thickness of the overlying soil when the test water pressure is 0.2 MPa. But with the increase in water pressure, the seepage of pipeline water is not only vertical seepage but also includes horizontal seepage. The intermittent cracks in the waterproof layer expand, which finally results in water inrush damage in the tunnel. The numerical simulation results show that the maximum shear force of the Wufeng Tunnel is at the arc and shoulder, which may easily form tensile shear failure along this part under groundwater seepage. The finding is consistent with the test results of the small-scale model. The water inrush in the tunnel is the coupling effect of the shear force and seepage field. Conclusion The primary factor of water inrush in the tunnel is water pressure and is closely related to the thickness of water barrier rock.

Objective The dry-wet cycles can promote the deterioration of the mechanical properties and stability of schist slope rock masses and induce geological disasters such as landslides and collapses. Methods Taking mica quartz schist, which is widely distributed in Shiyan, Hubei Province, as the research object, uniaxial compression tests and electron microscope scanning were carried out on mica quartz schist samples under different dry-wet cycles to reveal the degradation law and mechanism of its compressive property. Results The results show that under the action of dry-wet cycles, the degradation effect of the compressive strength and elastic modulus of mica quartz schist were obvious. The degradation rate was fast and the degradation range was large in the early stage, but the degradation rate was slow and the degradation range was low in the later stage. The macroscopic failure modes were gradually controlled by the schistosity planes, which gradually evolved from styles of "X" and "Y" failure to along the schistosity planes failure. The microstructure was characterized by continuous spalling and granulation of flake mineral particles, and the microcracks as well as pores were gradually developed, expanded and connected. The mica quartz schist produces secondary minerals, ions and rock cuttings and leaves the rock continuously under dry-wet cycles, which promotes the preferential expansion and cracking of schistosity, microcracks and other structural planes. which then degrades the overall mechanical properties of the mica quartz schist. Conclusion The research results can provide a theoretical basis for the stability analysis and treatment design of schist slopes.

The article is devoted to the experimental determination of thermokinetic parameters of oil sludge thermal degradation using the model-free Ozawa-Flynn-Wall method in the presence of a nanocatalyst (nickel, cobalt and iron-supported microsilicate) by calculating Arrhenius kinetic parameters (activation energy and pre-exponential factor). The phase composition of the reflex microsilicate was established – 4.12; 2.51 Å – SiO2, nickel-supported microsilicate reflexes: 2.09; 1.48 Å – NiO, reflexes: 4.25 Å – SiO2 and acid numbers of microsilicate – 64 μmol/g of prepared nanocatalysts. Using the method of Brunauer, Emmett and Teller, the specific surface area of the microsilicate was established – 18.3 ± 0.3 m2/g, the microsilicate with nickel applied – 20.9 ± 0.2 m2/g and the adsorption isotherm of the prepared nanocatalysts (microsilicate with nickel, cobalt and iron). Thermokinetic parameters of thermal decomposition of oil sludge without a catalyst and with a catalyst at an increment of 0.9 are 99.0 and 93.3 kJ/mol nickel-supported microsilicate, 51.9 kJ/mol cobalt-supported microsilicate, 111.3 kJ/mol iron-supported microsilicate and non-metal-supported microsilicate 173.7 kJ/mol, respectively. The study of the kinetic parameters of pyrolysis of oil sludge using various catalysts makes it possible to assess their influence on the process of decomposition of organic components. The results of the experiments showed that the use of catalysts significantly affects the destruction of oil sludge. Dynamic thermal analysis at different heating rates studied the dynamics of oil sludge decomposition. The study of the effect of catalysts on the kinetic parameters of oil sludge pyrolysis is an important step in the development of new methods for the disposal of petroleum products and the reduction of their negative impact on the environment. The obtained experimental data on thermal degradation kinetics of oil sludge will find application in designing a reactor for the process of destructive hydrogenation of heavy hydrocarbon raw materials.

Abstract Background Structural variations (SVs) in individual genomes are major determinants of complex traits, including adaptability to environmental variables. The Mongolian and Hainan cattle breeds in East Asia are of taurine and indicine origins that have evolved to adapt to cold and hot environments, respectively. However, few studies have investigated SVs in East Asian cattle genomes and their roles in environmental adaptation, and little is known about adaptively introgressed SVs in East Asian cattle. Results In this study, we examine the roles of SVs in the climate adaptation of these two cattle lineages by generating highly contiguous chromosome-scale genome assemblies. Comparison of the two assemblies along with 18 Mongolian and Hainan cattle genomes obtained by long-read sequencing data provides a catalog of 123,898 nonredundant SVs. Several SVs detected from long reads are in exons of genes associated with epidermal differentiation, skin barrier, and bovine tuberculosis resistance. Functional investigations show that a 108-bp exonic insertion in SPN may affect the uptake of Mycobacterium tuberculosis by macrophages, which might contribute to the low susceptibility of Hainan cattle to bovine tuberculosis. Genotyping of 373 whole genomes from 39 breeds identifies 2610 SVs that are differentiated along a “north–south” gradient in China and overlap with 862 related genes that are enriched in pathways related to environmental adaptation. We identify 1457 Chinese indicine-stratified SVs that possibly originate from banteng and are frequent in Chinese indicine cattle. Conclusions Our findings highlight the unique contribution of SVs in East Asian cattle to environmental adaptation and disease resistance.

Fungal endophytes exhibit symbiotic relationships with their host plants and convert inorganic selenium to organoselenium and Se0. In order to elucidate how Epichloë sinensis from Festuca sinensis adapts to different concentrations of sodium selenate, the dynamic changes of mycelial enzyme activities and metabolic changes at the transcriptional level were documented over a period of 36 h. The activity of enzymes (superoxide dismutase, glutathione reductase, glutathione S-transferase, cysteine synthetase, and methionine synthesis) in mycelia increased in the presence of increased Se concentrations during the cultivation period. The strain with selenium enrichment showed differential changes in gene expression compared to the strain without selenium enrichment, with more changes observed at higher Se concentrations over time. Notably, genes related to ribosomes or ribosome biogenesis in eukaryotes showed significant expression differences among certain groups, with up-regulation of genes involved in oxidoreductase activity, superoxide dismutase, and siderophore biosynthetic processes, and down-regulation of genes involved in steroid biosynthesis. These findings contribute to a better understanding of the transcriptional response of Epichloë sinensis to selenium.

The coal pillar is an important structure to control the stability of the roadway surrounding rock and maintain the safety of underground mining activities. An unreasonable design of the coal pillar size can result in the failure of the surrounding rock structure or waste of coal resources. The northern Shaanxi mining area of China belongs to the shallow buried coal seam mining in the gully area, and the gully topography makes the bearing law of the coal pillar and the development law of the internal fracture more complicated. In this study, based on the geological conditions of the Longhua Mine 20202 working face, a PFC2D numerical model was established to study the damage characteristics of coal pillars under the different overlying strata base load ratios in the gentle terrain area and the different gully slope sections in the gully terrain area, and the coal pillar design strategy based on the fractal characteristics of the fractures was proposed to provide a reference for determining the width of the coal pillars in mines under similar geological conditions. The results show that the reliability of the mathematical equation between the overlying strata base load ratio and the fractal dimension of the fractures in the coal pillar is high, the smaller the overlying strata base load ratio is, the greater the damage degree of the coal pillar is, and the width of the coal pillar of 15 m under the condition of the actual overlying strata base load ratio (1.19) is more reasonable. Compared with the gentle terrain area, the damage degree of the coal pillar in the gully terrain area is larger, in which the fractal dimension of the fracture in the coal pillar located below the gully bottom is the smallest, and the coal pillar in the gully terrain should be set as far as possible to make the coal pillar located below the gully bottom, so as to ensure the stability of the coal pillar.

Type 2 diabetes mellitus (T2DM) is a burgeoning public health challenge worldwide. Individuals with T2DM are at increased risk for skeletal muscle atrophy, a serious complication that significantly compromises quality of life and for which effective prevention measures are currently inadequate. Emerging evidence indicates that systemic and local inflammation stemming from the compromised intestinal barrier is one of the crucial mechanisms contributing to skeletal muscle atrophy in T2DM patients. Notably, natural plant polysaccharides were found to be capable of enhancing intestinal barrier function and mitigating secondary inflammation in some diseases. Herein, we hypothesized that Grifola frondosa polysaccharide (GFP), one of the major plant polysaccharides, could prevent skeletal muscle atrophy in T2DM via regulating intestinal barrier function and inhibiting systemic and local inflammation. Using a well-established T2DM rat model, we demonstrated that GFP was able to not only prevent hyperglycemia and insulin resistance but also repair intestinal mucosal barrier damage and subsequent inflammation, thereby alleviating the skeletal muscle atrophy in the T2DM rat model. Additionally, the binding free energy analysis and molecular docking of monosaccharides constituting GFP were further expanded for related targets to uncover more potential mechanisms. These results provide a novel preventative and therapeutic strategy for T2DM patients.

Wild desert grasslands are characterized by diverse habitats, uneven plant distribution, similarities among plant class, and the presence of plant shadows. However, the existing models for detecting plant species in desert grasslands exhibit low precision, require a large number of parameters, and incur high computational cost, rendering them unsuitable for deployment in plant recognition scenarios within these environments. To address these challenges, this paper proposes a lightweight and fast plant species detection system, termed YOLOv8s-KDT, tailored for complex desert grassland environments. Firstly, the model introduces a dynamic convolutional KernelWarehouse method to reduce the dimensionality of convolutional kernels and increase their number, thus achieving a better balance between parameter efficiency and representation ability. Secondly, the model incorporates triplet attention into its feature extraction network, effectively capturing the relationship between channel and spatial position and enhancing the model’s feature extraction capabilities. Finally, the introduction of a dynamic detection head tackles the issue related to target detection head and attention non-uniformity, thus improving the representation of the target detection head while reducing computational cost. The experimental results demonstrate that the upgraded YOLOv8s-KDT model can rapidly and effectively identify desert grassland plants. Compared to the original model, FLOPs decreased by 50.8%, accuracy improved by 4.5%, and mAP increased by 5.6%. Currently, the YOLOv8s-KDT model is deployed in the mobile plant identification APP of Ningxia desert grassland and the fixed-point ecological information observation platform. It facilitates the investigation of desert grassland vegetation distribution across the entire Ningxia region as well as long-term observation and tracking of plant ecological information in specific areas, such as Dashuikeng, Huangji Field, and Hongsibu in Ningxia.

Abstract Low‐coverage whole genome sequencing (lcWGS) has great potential to effectively genotype large‐scale population and to provide solid data for imputation; however, the time for imputation needs to be optimized. There is also no publicly available reference panel for whole genome selection in cattle. Here, we proposed a combination of Beagle v5.4 for phasing and GLIMPSE2 for imputation, which is fast and accurate for cattle lcWGS data. Furthermore, we established a multi‐breed reference panel with 61.8 million SNPs based on 2976 worldwide cattle, of which 1766 were bulls, by evaluating diversity and the size of the reference panel. The evaluation of imputation accuracy was conducted using new reference panel for both lcWGS and Bovine BeadChip data. The average concordance rate in Holstein was 99.6%, 99.6%, and 99.5% for 1X, 0.5X, and 0.1X lcWGS data, 99.5% and 99.0% for 777K and 50K chip data, and it was 98.8% for 1X lcWGS data in Simmental. We further investigated the factors affecting the imputation accuracy of lcWGS data and discovered that segmental duplication, structural variant, and guanine‐cytosine content were the top three factors. Interestingly, we found that 10 regions longer than 0.5 Mb showed low imputation accuracy enriched with immune function, such as 96.1% characterized genes in regions of chromosome 10, with more attention being paid on downstream immune‐related analysis. Our study provides the workflow of imputing lcWGS data and establishes the first high‐quality cattle reference panel with free access, which provides a resource to conduct subsequent large‐scale genome‐wide association studies and genomic selection.

Abstract Persistent room temperature phosphorescent materials with unique mechanical properties and robust optical properties have great potential in flexible electronics and photonics. However, developing such materials remains a formidable challenge. Here, we present highly stretchable, lightweight, and multicolored persistent luminescence elastomers, produced by incorporating ionic room temperature phosphorescent polymers and polyvinyl alcohol into a polydimethylsiloxane matrix. These prepared elastomers exhibit high optical transparency in daylight and emit bright persistent luminescence after the removal of 365 nm excitation. The homogeneous distribution of polymers within the matrix has been confirmed by confocal fluorescence microscopy, scanning electron microscopy, and atomic force microscopy. Mechanical property investigations revealed that the prepared persistent luminescence elastomers possess satisfactory stretchability. Impressively, these elastomers maintain robust optical properties even under extensive and repeated mechanical deformations, a characteristic previously unprecedented. These fantastic features make these persistent luminescence elastomers ideal candidates for potential applications in wearable devices, flexible displays, and anti-counterfeiting.

Since the leprosy cases have fallen dramatically, the incidence of leprosy has remained stable over the past years, indicating that multidrug therapy seems unable to eradicate leprosy. More seriously, the emergence of rifampicin-resistant strains also affects the effectiveness of treatment. Immunoprophylaxis was mainly carried out through vaccination with the BCG but also included vaccines such as LepVax and MiP. Meanwhile, it is well known that the infection and pathogenesis largely depend on the host’s genetic background and immunity, with the onset of the disease being genetically regulated. The immune process heavily influences the clinical course of the disease. However, the impact of immune processes and genetic regulation of leprosy on pathogenesis and immunological levels is largely unknown. Therefore, we summarize the latest research progress in leprosy treatment, prevention, immunity and gene function. The comprehensive research in these areas will help elucidate the pathogenesis of leprosy and provide a basis for developing leprosy elimination strategies.

Ultrasonic-assisted activated carbon separation (UACS) was first employed to improve product quality by regulating adsorption rate and removing bacterial endotoxin from salvia miltiorrhizae injection. The adsorption rate was related to three variables: activated carbon dosage, ultrasonic power, and pH. With the increase of activated carbon dosage from 0.05 % to 1.0 %, the adsorption rates of salvianolic acids and bacterial endotoxin increased simultaneously. The adsorption rates at which bacteria endotoxins increased from 52.52 % to 97.16 % were much higher than salvianolic acids. As the ultrasonic power increased from 0 to 700 W, the adsorption rates of salvianolic acids on activated carbon declined to less than 10 %, but bacterial endotoxin increased to more than 87 %. As the pH increased from 2.00 to 8.00, the adsorption rate of salvianolic acid dropped whereas bacterial endotoxin remained relatively stable. On the basis of response surface methodology (RSM), the optimal separation conditions were established to be activated carbon dose of 0.70 %, ultrasonic power of 600 W, and pH of 7.90. The experimental adsorption rates of bacterial endotoxin were 94.15 %, which satisfied the salvia miltiorrhizae injection quality criterion. Meanwhile, salvianolic acids' adsorption rates were 1.92 % for tanshinol, 4.05 % for protocatechualdehyde, 2.21 % for rosmarinic acid, and 3.77 % for salvianolic acid B, all of which were much lower than conventional activated carbon adsorption (CACA). Salvianolic acids' adsorption mechanism on activated carbon is dependent on the component's molecular state. Under ideal separation conditions, the molecular states of the four salvianolic acids fall between 1.13 % and 6.60 %. The quality of salvia miltiorrhizae injection can be improved while maintaining injection safety by reducing the adsorption rates of salvianolic acids to less than 5 % by the use of ultrasound to accelerate the desorption mass transfer rate on the activated carbon surface. When activated carbon adsorption was used in the process of producing salvia miltiorrhizae injection, the pH of the solution was around 5.00, and the proportion of each component's molecular state was tanshinol 7.05 %, protocatechualdehyde 48.93 %, rosmarinic acid 13.79 %, and salvianolic acid B 10.28 %, respectively. The loss of useful components was evident, and the corresponding activated carbon adsorption rate ranged from 20.74 % to 41.05 %. The average variation rate in plasma His and IgE was significant (P 0.05). The ultrasonic at a power intensity of 60 W/L and the power density of 1.20 W/cm2 may resolve the separation contradiction between salvianolic acids and bacterial endotoxin, according to experiments conducted with UACS at different power intensities. According to this study, UACS has a lot of potential applications in the pharmaceutical manufacturing industry and may represent a breakthrough in the field of ultrasonic separation.

Fermented foods have protective effects on body health. In our previously study, we found Lactiplantibacillus plantarum fermentation enhanced antioxidant activity of kiwifruit in vitro digestion. Then, in this work we explored the protective effect of fermented kiwi on intestinal injury induced by acute lipopolysaccharide (LPS) stress. Compared to non-fermented kiwi pulp (KP), Lactiplantibacillus plantarum fermented kiwi pulp (FKP-LP) contained more peptides, hormones and vitamins contents, lesser nucleic acid and carbohydrate contents. FKP-LP could relieve the intestinal injury by improving morphological of tight junction and upregulating tight junction proteins mRNA expression. Fermented kiwi maintained the mitochondrial morphology, mitochondrial respiratory function, and mitochondrial homeostasis, and relieved the LPS induced injury by regulating the contents of energy substances, and the respiratory chain complex enzyme activity through the pathway of AMPK and its downstream factors including PGC-1α, NRF1, NRF2, TFAM, and ULK2.

Dataspace is the transformation of cyberspace from ＆quot;computing centric＆quot; to ＆quot;data centric＆quot;, which contains great technological issues and innovative opportunities.Similar to the internet, which is the main infrastructure of cyberspace, dataspace also needs a new ＆quot;data-centric＆quot; infrastructure, whose core function is to realize the first-class entity of data.From the perspective of dataspace, the supports and shortcomings of mainstream technologies such as the internet, the World Wide Web, and the digital object architecture for the first-class entity of data were analyzed and summarized, and then the basic connotations and technical challenges of dataspace infrastructure were given.Finally, a first-class data substantialization method based on data pragmatics was proposed.Based on this method, a solution called the internet of data by integrating digital object architecture, distributed ledger, smart contract, and other technologies was proposed to support the construction and operation of internetscale dataspace infrastructure.

Accurate analysis of the main chemical components of oil-bearing sludge is an important prerequisite for effective soil remediation and resource reuse. However, precise analysis of the extract components is difficult to achieve because of the mutual interference of saturates, aromatics, and resins, collectively called SAR, and asphaltene that are introduced during chloroform extraction. In this study, SAR was efficiently extracted using n-heptane, while asphaltene components were retained in soil because of their insolubility in n-heptane. The maximum yield of SAR extraction was 27.0%, indicating an increase of 1.75% compared to chloroform extraction. The extracted SAR components were separated by chromatography and the main structural units and components were analyzed. The results show that saturates, aromatics, and resins have a single component, a high content of major components, and contain fewer impurities using n-heptane extraction. Moreover, the solubility of asphaltene was inhibited during the effective extraction of SAR components with n-heptane and did not influence the subsequent analysis of SAR component. Efficient SAR extraction, accurate SAR component analysis, and high efficiency asphaltene separation was achieved using n-heptane-extraction-assisted pyrolysis. This provides a new method for the component analysis of oily sludge and promotes its efficient separation.

Ultrasonic-assisted supercritical fluid separation (USFS) was firstly applied to regulate solubility and remove plasticizers from ganoderma lucidum spores’ oil to improve product safety. Separation efficiency was related with four variables, including temperature, pressure and ultrasonic power. The QD-T6A ultrasonic generator probe, which provided for the study with adjustable ultrasonic power 0 W to 800 W and the ultrasonic frequency was 40 kHz, was fixed at the entrance of the primary separation kettle. The optimal separation conditions were determined to be temperature as 15.0 °C, pressure as 18.0 MPa, and ultrasonic power as 360 W of ultrasonic power on the basis of response surface methodology (RSM). Experimental Di-n-butylphthalate (DBP) and Diethyl phthalate (DEP) content were 0.09 mg and 0.04 mg, respectively, which were below the limits for plasticizers. Meanwhile, the total triterpene and ganoderic acid A contents were 6.89 g and 1.10 g, respectively, comparable to conventional supercritical fluid extraction. The experiments with USFS at different power intensities revealed that ultrasonic at a power intensity of 36 W/L and the power density of 0.20 W/cm2 could resolve the separation contradiction between ganoderma lucidum spores’ oil and plasticizers. This study revealed that USFS could be an innovation in the field of ultrasonic separation, with numerous potentials uses in pharmaceutical manufacturing.

Background: Smilax is a traditional medicine used for the clinical treatment of pelvic inflammatory disease (CPID), and its leaves have long been used as food by the Korean people in Asia. In previous studies, we found that flavonoids extract can improve CPID by inhibiting inflammation, which may be the main effective component of Smilax China, but its mechanism of action has not been fully elucidated. This study explores the mechanism by which Chinese rhizome flavonoids (FSCR) regulate the autophagy pathway of NLRP3 inflammasome and promote macrophage reprogramming. Methods: In the in vivo experiment, the sham surgery group and the CPID group were given physiological saline and FCSR for 7 days, respectively. We determined the expression of NLRP3 inflammasome autophagy pathway genes and proteins by observing the pathological damage of rat uteri through HE and Masson staining, immunofluorescence, RT-PCR, and WB experiments. Molecular docking simulation predicted the binding potential of flavonoids with the potential target VPS34, and knockdown experiments were conducted to validate the knockdown effect of lipopolysaccharide treatment on primary rat endometrial cells to simulate an in vitro CPID model. Results: FCSR significantly reduced serum IL-1β in CPID rats, TNF-α. The concentration of TAX2 reduced the infiltration of inflammatory cells and fibrosis of endometrial cells, promoted M2 polarization of macrophages, upregulated autophagy pathways, and inhibited the activation of NLRP3 inflammasomes. After knocking down the NLRP3 inflammasome autophagy target VPS34, the FSCR effect was eliminated. Conclusions: By targeting VPS34, FCSR potentially promotes autophagic cell reprogramming through the NLRP3 inflammasome related autophagy pathway.

BackgroundHigh-producing dairy cows face varying degrees of metabolic stress and challenges during the late perinatal period, resulting in ruminal bacteria abundance and their fermentative ability occurring as a series of changes. However, the dynamic changes are still not clear.Aims/methodsTen healthy, high-producing Holstein dairy cows with similar body conditions and the same parity were selected, and ruminal fluid from the dairy cows at postpartum 0, 7, 14, and 21 d was collected before morning feeding. 16S rRNA high-throughput sequencing, GC-MS/MS targeted metabolomics, and UPLC-MS/MS untargeted metabolomics were applied in the study to investigate the dynamic changes within 21 d postpartum.ResultsThe results displayed that the structures of ruminal bacteria were significantly altered from 0 to 7 d postpartum (R = 0.486, P = 0.002), reflecting the significantly declining abundances of Euryarchaeota and Chloroflexi phyla and Christensenellaceae, Methanobrevibacter, and Flexilinea genera (P < 0.05) and the obviously ascending abundances of Ruminococcaceae, Moryella, Pseudobutyrivibrio, and Prevotellaceae genera at 7 d postpartum (P < 0.05). The structures of ruminal bacteria also varied significantly from 7 to 14 d postpartum (R = 0.125, P = 0.022), reflecting the reducing abundances of Christensenellaceae, Ruminococcaceae, and Moryella genera (P < 0.05), and the elevating abundances of Sharpea and Olsenella genera at 14 d postpartum (P < 0.05). The metabolic profiles of ruminal SCFAs were obviously varied from 0 to 7 d postpartum, resulting in higher levels of propionic acid, butyric acid, and valeric acid at 7 d postpartum (P < 0.05); the metabolic profiles of other ruminal metabolites were significantly shifted from 0 to 7 d postpartum, with 27 significantly elevated metabolites and 35 apparently reduced metabolites (P < 0.05). The correlation analysis indicated that propionic acid was positively correlated with Prevotellaceae and Ruminococcaceae (P < 0.05), negatively correlated with Methanobrevibacter (P < 0.01); butyric acid was positively associated with Prevotellaceae, Ruminococcaceae, and Pseudobutyrivibrio (P < 0.05), negatively associated with Christensenellaceae (P < 0.01); valeric acid was positively linked with Prevotellaceae and Ruminococcaceae (P < 0.05); pyridoxal was positively correlated with Flexilinea and Methanobrevibacter (P < 0.05) and negatively correlated with Ruminococcaceae (P < 0.01); tyramine was negatively linked with Ruminococcaceae (P < 0.01).ConclusionThe findings contribute to the decision of nutritional management and prevention of metabolic diseases in high-producing dairy cows during the late perinatal period.

Background and objectives: There are concerns about the serological responses to Coronavirus disease 2019 (COVID-19) vaccines in inflammatory bowel disease (IBD) patients, particularly those receiving anti-TNF therapy. This study aimed to systematically evaluate the efficacy of COVID-19 vaccines in IBD patients receiving anti-TNF therapy. Methods: Electronic databases were searched to identify relevant studies. We calculated pooled seroconversion rate after COVID-19 vaccination and subgroup analysis for vaccine types and different treatments were performed. Additionally, we estimated pooled rate of T cell response, neutralization response, and breakthrough infections in this population. Results: 32 studies were included in the meta-analysis. IBD patients receiving anti-TNF therapy had relatively high overall seroconversion rate after complete vaccination, with no statistical difference in antibody responses associated with different drug treatments. The pooled positivity rate of T cell response was 0.85 in IBD patients receiving anti-TNF therapy. Compared with healthy controls, the positivity of neutralization assays was significantly lower in IBD patients receiving anti-TNF therapy. The pooled rate of breakthrough infections in IBD patients receiving anti-TNF therapy was 0.04. Conclusions: COVID-19 vaccines have shown good efficacy in IBD patients receiving anti-TNF therapy. However, IBD patients receiving anti-TNF have a relatively high rate of breakthrough infections and a low level of neutralization response.

Background and Aims: The clinical, prognostic, and therapeutic impact of adrenal insufficiency in acute liver failure (ALF) and acute-on-chronic liver failure (ACLF) remains controversial and exact diagnostic criteria are lacking. We sought to determine the diagnostic and therapeutic value of cortisol measurement and glucocorticoid (GC) treatment in ALF and ACLF. Abstract Circular RNAs (circRNAs) are important transcriptional regulatory RNA molecule that can regulate the transcription of downstream genes by competitive binding of miRNAs or coding proteins or by blocking mRNAs translation. Numerous studies have shown that circRNAs are extensively involved in cell proliferation, differentiation and apoptosis, gene transcription and signal transduction. Fat deposition and muscle development have important effects on beef traits. CircRNAs are involved in regulating bovine fat and muscle cells and are differentially expressed in the tissues composed of these cells, suggesting that circRNAs play an important role in regulating bovine fat formation and muscle development. This review describes differential expression of circRNAs in bovine fat and muscle tissues, research progress in understanding how circRNAs regulate the proliferation and differentiation of bovine fat and muscle cells through competing endogenous RNAs networks, and provide a reference for the subsequent research on the molecular mechanism of circRNAs in regulating fat deposition and muscle development in cattle.

Introduction Hormone replacement treatment (HRT) is the most commonly used endometrial preparation protocol for frozen embryo transfer (FET) in patients with polycystic ovary syndrome (PCOS). However, studies have found that HRT may increase the risk of hypertensive disorders of pregnancy and some obstetric complications. Letrozole is a new first-line ovulation induction drug for PCOS and can effectively induce spontaneous ovulation by reducing oestrogen levels. However, letrozole is still rarely used in FET and has only been reported in a few studies in Asian populations. High-quality, well-powered randomised controlled trials (RCTs) comparing HRT and letrozole-stimulated protocols are lacking. The aim of this study is to compare the efficacy and safety of two protocols in patients with PCOS.Methods and analysis This is a multicentre, open-label RCT in four reproductive medical centres in China. In total, 1078 women with PCOS will be randomised (1:1) to the letrozole-stimulated or HRT group in their first FET cycle and their pregnancy and perinatal outcomes during this cycle will be followed up and analysed. The primary outcome is live birth. Secondary outcomes are cycle cancellation rate, biochemical pregnancy, clinical pregnancy, miscarriage, ectopic pregnancy, obstetric and perinatal complications, neonatal complications and birth weight.Ethics and dissemination Ethical approval was obtained from the Institutional Review Board of Reproductive and Genetic Hospital of CITIC-XIANGYA (LL-SC-2022-001). Written informed consent will be obtained from each participant. The findings will be disseminated through conference presentations and publication in peer-reviewed journals.Trial registration number NCT05227391.

The changes in the composition of intestinal microbiota and metabolites have been linked to digestive disorders in calves, especially neonatal calf diarrhea. Bovine rotavirus (BRV) and bovine coronavirus (BCoV) are known to be the primary culprits behind neonatal calf diarrhea. In this study, we analyzed changes in the fecal microbiota and metabolites of calves with neonatal diarrhea associated with BRV and BCoV infection using high-throughput 16S rRNA sequencing and metabolomics technology. The microbial diversity in the feces of calves infected with BRV and BCoV with diarrhea decreased significantly, and the composition changed significantly. The significant increase of Fusobacterium and the reductions of some bacteria genera, including Faecalibacterium, Bifidobacterium, Ruminococcus, Subdoligranulum, Parabacteroides, Collinsella, and Olsenella, etc., were closely related to diarrhea associated with BRV and BCoV infection. Metabolites in the feces of BRV and BCoV-infected calves with diarrhea were significantly changed. Phosphatidylcholine [PC; 16:1(9 Z)/16:1(9 Z)], lysophosphatidylethanolamine (LysoPE; 0:0/22:0), lysophosphatidylcholine (LysoPC; P-16:0) and LysoPE (0:0/18:0) were significantly higher in the feces of BRV-infected calves with diarrhea. In contrast, some others, such as desthiobiotin, were significantly lower. BRV infection affects glycerophospholipid metabolism and biotin metabolism in calves. Two differential metabolites were significantly increased, and 67 differential metabolites were significantly reduced in the feces of BCoV-infected calves with diarrhea. Seven significantly reduced metabolites, including deoxythymidylic acid (DTMP), dihydrobiopterin, dihydroneopterin triphosphate, cortexolone, cortisol, pantetheine, and pregnenolone sulfate, were enriched in the folate biosynthesis, pantothenate and CoA biosynthesis, pyrimidine metabolism, and steroid hormone biosynthesis pathway. The decrease in these metabolites was closely associated with increased harmful bacteria and reduced commensal bacteria. The content of short-chain fatty acids (SCFAs) such as acetic acid and propionic acid in the feces of BRV and BCoV-infected calves with diarrhea was lower than that of healthy calves, which was associated with the depletion of SCFAs-producing bacteria such as Parabacteroides, Fournierella, and Collinsella. The present study showed that BRV and BCoV infections changed the composition of the calf fecal microbiota and were associated with changes in fecal metabolites. This study lays the foundation for further revealing the roles of intestinal microbiota in neonatal calf diarrhea associated with BRV and BCoV infection.

Currently, facility cultivation produces almost a third of all vegetables in China. The intensive production style has led to serious soil problems that need to be tackled. In this paper, a pot experiment was set up to evaluate the effects of optimized fertilization on vegetable growth and soil properties. Specifically, calcium, magnesium, boron and molybdenum were added on the basis of soil testing. The results showed that the growth of Chinese cabbage was significantly increased by optimized fertilization. The soil pH increased (by 3.82%), and EC decreased (by 8.54%). The abundance of culturable bacteria increased by 33.86%, whereas that of fungi decreased by 70.7%. The optimized fertilization increased the richness but not the evenness of soil microorganisms, increased the relative abundance of Proteobacteria, Firmicutes, Bacillus and Bacteroidetes, and decreased the relative abundance of Actinobacteria, Chloroflexi, and superphyla. Optimized fertilization inhibited the growth of Chytridiomycota and Mortierellomycota, especially the plant pathogen Fusarium. Moreover, balanced fertilization was beneficial in promoting various metabolic processes of soil bacteria. Soil water-soluble Ca, Mg, and available Mo might be the main factors driving the change in microbial groups.

The differences in soil fungal communities in four agricultural areas growing wheat (Triticum aestivum), rapeseed (Brassica napus), and barley (Hordeum vulgare) in the Qinghai Province, namely the Dulan (DL), Gonghe (GH), Huzhu (HZ), and Datong (DT) counties, were investigated using high-throughput sequencing. The region showed highly significant effects on soil pH, organic matter, ammonium nitrogen, nitrate nitrogen, total phosphate, effective phosphate, total sulfur, and effective sulfur (p < 0.01). The crop type resulted in highly significant (p < 0.01) variations in total phosphate and effective phosphate. Principal coordinates analysis and nonmetric multidimensional scaling revealed significant differences in soil fungal diversity and fungal community composition in the soils of three crops or four regions (p < 0.05). Although the soils of the four regions or three crops had similar dominant phyla, classes, and genera, these taxa differed in terms of their relative abundance. Four, 12, 15, and 16 biomarkers with significant linear discriminant analysis effect sizes were identified in the HZ, DL, GH, and DT groups, respectively. A total of 36, 12, and eight significant biomarkers were observed in the wheat, rapeseed, and barley soils, respectively. In addition, altitude and soil physicochemical properties had significant relationships with fungal diversity and community composition (p < 0.05, p < 0.01).

Abstract Maize is a crop that is highly susceptible to the negative effects of low temperature. Low temperature can delay seed germination and cause a decrease in seed vigor, which seriously affects seedling emergence and yield. In this study, 190 maize accessions (inbred lines) with strong germination potential at normal temperature (25°C) were selected from more than 500 accessions to construct a new association mapping panel to further investigate germination under chilling stress (5°C). We re‐sequenced the genomes of the 190 diverse accessions and obtained 4,886,919 high‐quality SNPs. We then used this data to analyze population structure, perform principal components analysis, and construct a phylogenetic tree of the new maize panel. The relative germination rate (RGR) and relative germination index (RGI) are two traits that are significantly related to chilling‐tolerant germination. Genome‐wide association analysis showed that RGR and RGI shared a major QTL, and they also shared the top SNP. There were a total of 26 significant SNPs in common. These SNPs hit directly or indirectly within 37 candidate genes. Among these 37 gene candidates, eight are homologs of genes previously reported to be related to both germination and low‐temperature stress, and another 12 genes related to low‐temperature stress or other abiotic stresses such as drought, salinity, oxidative, and high light stress. In addition, RGR and RGI had another 15 and 26 significant SNPs, respectively, which were associated with 17 and 92 candidate genes, respectively. Further qRT‐PCR analysis using 26 chilling‐tolerant and 22 chilling‐sensitive accessions implied that Zm00001eb272370, Zm00001eb272390, and Zm00001eb272400 associated with the top SNP, may play different roles during cold‐germination. Thus, our study not only established a new association mapping panel suitable for investigation of germination at low temperature but also provided valuable genetic resources for future studies to improve chilling‐tolerant maize varieties.

Background Skeletal muscle is not only an important tissue involved in exercise and metabolism, but also an important part of livestock and poultry meat products. Its growth and development determines the output and quality of meat to a certain extent, and has an important impact on the economic benefits of animal husbandry. Skeletal muscle development is a complex regulatory network process, and its molecular mechanism needs to be further studied. Method We used a weighted co-expression network (WGCNA) and single gene set enrichment analysis (GSEA) to study the RNA-seq data set of bovine tissue differential expression analysis, and the core genes and functional enrichment pathways closely related to muscle tissue development were screened. Finally, the accuracy of the analysis results was verified by tissue expression profile detection and bovine skeletal muscle satellite cell differentiation model in vitro (BSMSCs). Results In this study, Atp2a1, Tmod4, Lmod3, Ryr1 and Mybpc2 were identified as marker genes in muscle tissue, which are mainly involved in glycolysis/gluconeogenesis, AMPK pathway and insulin pathway. The assay results showed that these five genes were highly expressed in muscle tissue and positively correlated with the differentiation of bovine BSMSCs. Conclusions In this study, several muscle tissue characteristic genes were excavated, which may play an important role in muscle development and provide new insights for bovine molecular genetic breeding.

Exosomes are a kind of extracellular vesicles that are produced and secreted by different mammalian cells. They serve as cargo proteins and can transfer different kinds of biomolecules, including proteins, lipids, and nucleic acids, which consequently act on target cells to exert different biological effects. Recent years have witnessed a significant increase in the number of studies on exosomes due to the potential effects of exosomes in the diagnosis and treatment of cancers, neurodegenerative diseases, and immune disorders. Previous studies have demonstrated that exosomal contents, especially miRNAs, are implicated in numerous physiological processes such as reproduction, and are crucial regulators of mammalian reproduction and pregnancy-related diseases. Here, we describe the origin, composition, and intercellular communication of exosomes, and discuss their functions in follicular development, early embryonic development, embryonic implantation, male reproduction and development of pregnancy-related diseases in humans and animals. We believe this study will provide a foundation for revealing the mechanism of exosomes in regulating mammalian reproduction, and providing new approaches and ideas for the diagnosis and treatment of pregnancy-related diseases.
