1. Relaxin 2 carried by magnetically directed liposomes accelerates rat midpalatal suture expansion and subsequent new bone formation

Abstract: Relaxin (RLN) is an insulin-like peptide hormone that enables softening and lengthening of the pubic symphysis and uterine cervix. Here, we analyzed the effects of RLN2 on the expansion of rat midpalatal suture (MPS) using a magnetically directed liposome-based drug delivery system. Thirty-six male rats were divided into three groups: control (MPS was not expanded), lipo (expanded for 1 week with vehicle liposomes encapsulating ferric oxide and Cy5.5), and RLN-lipo (expanded for 1 week with the liposomes coated with RLN2). Rats were sacrificed after 1 week of expansion or after 2 weeks of retention. To accumulate RLN2-liposomes, a magnetic sheet was fixed to the palatal mucosa of the MPS. In vivo imaging showed magnetically controlled accumulation of liposomes in the MPS for 72 h. Immunohistochemistry revealed RLN2 expression in the MPS after expansion and relaxin receptor (RXFP) 2 expression at the osteogenic front (OF) in the RLN-lipo group; all groups expressed RXFP1 in the MPS. MPS expansion and bone formation were significantly accelerated at the OF in RLN-lipo group compared with the other groups. In the RLN-lipo group, significantly accelerated serrate bone deposition and elevated periostin (POSTN), iNOS, and MMP-1 levels were observed in the MPS. Sclerostin (SOST) expression was significantly reduced in newly formed bone in the RLN-lipo group. Our data revealed that RLN2 enhanced suture expansion via MMP-1 and iNOS secretion in the sutural fibroblasts and new bone formation via POSTN expression in osteoblasts at the OF. These properties may be useful for developing a new less-invasive orthopedic treatment aiming at sutural modification of cranio- and maxillofacial deformity patients. 3. Changes in superoxide dismutase 3 (SOD3) expression in periodontal tissue during orthodontic tooth movement of rat molars and the effect of SOD3 on in vitro hypoxia-exposed rat periodontal ligament cells

Abstract: Summary Background/Objectives Hypoxia during orthodontic tooth movement (OTM) induces reactive oxygen species (ROS) production in periodontal tissues. Superoxide dismutase 3 (SOD3) is an anti-inflammatory enzyme that protects cells from ROS. This study investigated the expression and function of SOD3 during rat OTM and in hypoxia-exposed rat periodontal ligament (PDL) cells. Materials/Methods OTM of right maxillary first molars were performed in 8-week-old male Sprague–Dawley rats using closed-coil spring for 1 and 14 days (n = 6 per group). SOD3 and hypoxia-inducible factor 1-alpha (HIF-1α) protein expression was evaluated by immunohistochemistry. The effects of SOD3 on cell viability and proliferation, ROS production, and mRNA expression of Hif1-α, receptor activator of nuclear factor kappa-Β ligand (Rankl), and osteoprotegerin (Opg) in PDL cells and osteoclast differentiation were investigated under normal and hypoxic conditions. Results SOD3 expression in PDL tissues significantly decreased on the compression side on day 1 and on both sides on day 14 of OTM. HIF-1α levels significantly increased on the compression side on day 14. Cell viability, cell proliferation, and Opg mRNA expression decreased, whereas ROS production and Hif1-α and Rankl mRNA expression increased in the PDL cells upon SOD3 silencing. Hypoxia reduced Sod3 and Opg mRNA expression and increased ROS, Rankl mRNA expression, and osteoclast formation; SOD3 treatment attenuated these effects. Conclusion/Implications SOD3 plays a role in periodontal tissue remodelling during OTM and in hypoxia-exposed PDL cells through ROS, HIF-1α, and RANKL/OPG pathways. Moreover, SOD3 treatment could attenuate the negative effects of hypoxia on the PDL cells.

4. Regulation of Calvarial Osteogenesis by Concomitant De-repression of GLI3 and Activation of IHH Targets

Abstract: Loss-of-function mutations in GLI3 and IHH cause craniosynostosis and reduced osteogenesis, respectively. In this study, we show that Ihh ligand, the receptor Ptch1 and Gli transcription factors are differentially expressed in embryonic mouse calvaria osteogenic condensations. We show that in both Ihh−/− and Gli3Xt−J/Xt−J embryonic mice, the normal gene expression architecture is lost and this results in disorganized calvarial bone development. RUNX2 is a master regulatory transcription factor controlling osteogenesis. In the absence of Gli3, RUNX2 isoform II and IHH are upregulated, and RUNX2 isoform I downregulated. This is consistent with the expanded and aberrant osteogenesis observed in Gli3Xt−J/Xt−J mice, and consistent with Runx2-I expression by relatively immature osteoprogenitors. Ihh−/− mice exhibited small calvarial bones and HH target genes, Ptch1 and Gli1, were absent. This indicates that IHH is the functional HH ligand, and that it is not compensated by another HH ligand. To decipher the roles and potential interaction of Gli3 and Ihh, we generated Ihh−/−;Gli3Xt−J/Xt−J compound mutant mice. Even in the absence of Ihh, Gli3 deletion was sufficient to induce aberrant precocious ossification across the developing suture, indicating that the craniosynostosis phenotype of Gli3Xt−J/Xt−J mice is not dependent on IHH ligand. Also, we found that Ihh was not required for Runx2 expression as the expression of RUNX2 target genes was unaffected by deletion of Ihh. To test whether RUNX2 has a role upstream of IHH, we performed RUNX2 siRNA knock down experiments in WT calvarial osteoblasts and explants and found that Ihh expression is suppressed. Our results show that IHH is the functional HH ligand in the embryonic mouse calvaria osteogenic condensations, where it regulates the progression of osteoblastic differentiation. As GLI3 represses the expression of Runx2-II and Ihh, and also elevates the Runx2-I expression, and as IHH may be regulated by RUNX2 these results raise the possibility of a regulatory feedback circuit to control calvarial osteogenesis and suture patency. Taken together, RUNX2-controlled osteoblastic cell fate is regulated by IHH through concomitant inhibition of GLI3-repressor formation and activation of downstream targets.

5. Detection of very-high energy gamma rays from RS Ophiuchi nova outburst with the Large-Sized Telescope of the Cherenkov Telescope Array

Abstract: The first Large-Sized Telescope (LST-1) of the future Cherenkov Telescope Array (CTA) is located in the northern site of CTA, the Roque de los Muchachos Observatory in the island of La Palma. The LST-1 started its commissioning phase in November 2019 and has been collecting data since then. In August 2021 an outburst from the recurrent nova RS Ophiuchi, a binary system consisting of a red giant and a white dwarf, was reported in optical and high energy (HE) gamma rays. The LST-1 observed RS Ophiuchi during several nights coincident with the outburst and detected a signal of very-high energy (VHE) gamma rays together with the current generation of gamma-ray observatories. This is the first time a nova is detected in the VHE range. In this contribution the results from the LST-1 observations will be presented.

Abstract: The periodontal ligament (PDL) plays an important role in orthodontic tooth movement; however, the underlying molecular mechanism remains unclear. We have previously reported that the Mohawk homeobox (Mkx), a tendon-specific transcription factor, is expressed in the PDL and regulates its homeostasis. In the present study, we examined the role of Mkx in orthodontic tooth movement via bone remodeling induced by mechanical stimulation in Mkx-deficient rats, which are widely used as experimental animals for orthodontic force application. Orthodontic tooth movement of the maxillary first molar was performed in 7-week-old male Mkx-deficient rats (n = 4) and wild-type Wistar rats (n = 4) using coil springs for 14 days. Hematoxylin and eosin (H&E) staining and tartrate-resistant acid phosphatase (TRAP) staining were performed to evaluate morphological changes and osteoclasts. Furthermore, changes in the expression of receptor activator nuclear factor-kappa B ligand (RANKL) were demonstrated using immunostaining. The amount of tooth movement was significantly lower in Mkx-deficient rats than in wild-type rats. The number of TRAP-positive cells was suppressed in Mkx-deficient rats on the compression side. Orthodontic tooth movement experiments in Mkx-deficient rats suggested that Mkx is involved in osteoclast induction at the alveolar bone surface on the compression side. This study reveals the possibility that Mkx plays a mechanosensory role in orthodontic tooth movement by inducing RANKL expression and osteoclastogenesis.

Abstract: Background Apert syndrome is an autosomal, dominant inherited disorder characterized by craniosynostosis and syndactyly caused by gain‐of‐function mutations in the fibroblast growth factor receptor 2 (FGFR2) gene. Wnt/β‐catenin signaling plays critical roles in regulating the skeletal development. Here, we analyzed the role of this pathway in the developing coronal sutures (CS) of a murine Apert syndrome model (Fgfr2S252W/+). Results We observed aberrantly increased mRNA expression of Lrp5 and Lrp6 in CS of Fgfr2S252W/+ mice, whereas both wild type (WT) and Fgfr2S252W/+ mice showed similar expression of other Wnt/β‐catenin‐related genes, such as Wnt3, Wnt3a, Fzd4, Fzd6, Axin2, and Dkk1 as evidenced by in situ hybridization. Significantly increased Lrp5 and Lrp6 mRNA expression was observed by quantitative PCR analysis of cultured cells isolated from CS of Fgfr2S252W/+ mice. Phospho‐LRP5, phospho‐LRP6, and non‐phospho‐β‐catenin were upregulated in Fgfr2S252W/+ CS compared with that in WT CS. Short‐interfering RNA targeting Lrp5 and Lrp6 significantly reduced runt‐related transcription factor 2, collagen type 1 alpha 1, and osteocalcin mRNA expression, and alkaline phosphatase activity in cultured cells. Conclusions The Wnt/β‐catenin pathway was activated in the CS of Fgfr2S252W/+ mice during craniofacial development, suggesting the involvement of the Wnt/β‐catenin pathway in the pathogenesis of CS synostosis in Fgfr2S252W/+ mice. Abstract: Summary Background and objectives Mutations in the fibroblast growth factor receptor 2 (FGFR2) gene are responsible for both Apert syndrome (AS) and Crouzon syndrome (CS). These diseases share phenotypic characteristics, including midfacial hypoplasia and premature fusion of the calvarial suture(s). Given the extensive range of craniofacial growth and developmental abnormalities, management of these patients requires a multidisciplinary approach. This study aimed to compare craniofacial, oral, and cervical morphological characteristics in Japanese orthodontic patients with AS or CS. Subjects and methods Lateral cephalograms, orthopantomograms, dental casts, medical interview records, facial photographs, and intraoral photographs of 7 AS patients and 12 CS patients on initial visits were used in this study. Cephalometric analyses were performed, and standard scores were calculated based on age- and sex-matched Japanese standard values. Results Cephalometric analysis revealed that AS patients had significantly more severe maxillary hypoplasia in two dimensions and increased clockwise mandibular rotation. Additionally, cleft of the soft palate, anterior open bite, severe crowding in the maxillary dental arch, and congenitally missing teeth occurred more frequently among AS patients. Multiple fusions between cervical vertebrae C2, C3, C5, and C6 were observed in the AS patients. Limitations Small sample size. Conclusions/implications Our study shows that AS patients have more severe craniofacial and maxillofacial deformities than CS patients.

Abstract: The realtime follow-up of neutrino events is a promising approach to search for astrophysical neutrino sources. It has so far provided compelling evidence for a neutrino point source: the flaring gamma-ray blazar TXS 0506+056 observed in coincidence with the high-energy neutrino IceCube-170922A detected by IceCube. The detection of very-high-energy gamma rays (VHE, $\mathrm{E} > 100\,\mathrm{GeV}$) from this source helped establish the coincidence and constrained the modeling of the blazar emission at the time of the IceCube event. The four major imaging atmospheric Cherenkov telescope arrays (IACTs) - FACT, H.E.S.S., MAGIC, and VERITAS - operate an active follow-up program of target-of-opportunity observations of neutrino alerts sent by IceCube. This program has two main components. One are the observations of known gamma-ray sources around which a cluster of candidate neutrino events has been identified by IceCube (Gamma-ray Follow-Up, GFU). Second one is the follow-up of single high-energy neutrino candidate events of potential astrophysical origin such as IceCube-170922A. GFU has been recently upgraded by IceCube in collaboration with the IACT groups. Abstract: The origin of Galactic Cosmic rays (GCRs), whose spectrum extends to PeV energies, is one of the longest-standing problems in astroparticle physics. One of the main sources of GCRs are regarded to be Supernova remnants (SNRs). While SNRs are known to accelerate protons, so far there is no evidence that SNRs can accelerate CRs to PeV energies. Providing that ~10% of the parent Cosmic ray energy is converted to gamma rays, the gamma-ray spectrum extending up to ~100 TeV would be a signature of a so-called Galactic PeVatron, an object responsible for the production of protons up to the knee of the Cosmic ray spectrum. The current multi-wavelength data indicate that HESS J1809-193 is one of the most promising Galactic PeVatron candidates. So far, no firm identification on the source nature has been established as there are several possible counterparts at lower energies; one of them being SNR G11.0−0.0. We report here the results of an observational campaign performed by the MAGIC telescopes on HESS J1809-193 since 2019 in the very-high-energy gamma-ray domain (E>100 GeV). The data were obtained with the Very Large Zenith Angle (VLZA) technique, which increased the collection area significantly to about one square kilometer. Abstract: X-ray and gamma-ray binaries are systems consisting of a compact object and normally a non-degenerate companion star. Most of these sources have been shown to emit radiation in a broad frequency range, from radio up to X-rays and sometimes gamma rays. We report on recent results in very high-energy gamma rays above 100 GeV obtained by the MAGIC Collaboration for the Galactic X-ray binaries MAXI J1820+070 and 1A 0535+262, and the gamma-ray binary HESS J0632+057. Multiwavelength data at lower energies are also provided for a better contextualisation of the sources.

Abstract: QSO B0218+357 is currently the only gravitationally lensed source from which very-high-energy (VHE, &100GeV) gamma-ray emission has been detected. We report the multiwavelength monitoring observations of this source performed between 2016 and 2020 in radio interferometry, optical, X-ray and gamma-ray bands. Abstract: Blazars, together with other active galactic nuclei, are the most luminous persistent sources in our universe; and therefore a prime candidate for very-high-energy (>0.2 TeV, VHE) gamma-ray observations. For the two MAGIC telescopes, the Mrk501 galaxy is among the brightest observed blazars due to its proximity. We report a multi-wavelength and multi-messenger study of Mrk501 with data from 2017 to 2020, when Mrk501 showed a VHE flux typically below 10% that of the Crab Nebula. During this time, we performed three long observations with NuSTAR, which characterized the hard X-ray emission during three different low-activity flux levels. This Mrk501 dataset provided the unprecedented opportunity to study multi-wavelength variability and correlations with sensitive instruments during historically low X-ray and VHE gamma-ray emission (below 5% of the Crab Nebula flux in the VHE range), which could be considered as the baseline emission of Mrk501. We complemented the broadband spectral energy distributions (SED) of the identified historically low X-ray and VHE gamma-ray flux with data published by IceCube, in order to evaluate the potential existence of a hadronic component that is stable (or slowly variable), and less visible than the leptonic component that may dominate the emission during typical and flaring activity. Abstract: The high-peaked BL Lac object 1ES 0647+250 is one of the few distant blazars detected at very-high-energy (VHE, E>100 GeV) γ rays during non-flaring activity. Its redshift is still uncertain, with a recently proposed lower limit of z>0.29. This blazar was first detected by the MAGIC telescopes between 2009 and 2011 during its low state, displaying a flux of around 2% of the Crab Nebula above 100 GeV, but it has shown several periods of high activity, where the VHE γ-ray flux increased by more than one order of magnitude. The VHE spectra of four observed periods are characterized and the redshift of this source is estimated using the HE and VHE spectral shapes. Abstract: We present the first search for dark matter (DM) spectral lines in the Galactic centre (GC) region with the MAGIC telescopes. The MAGIC telescopes, located on the Canary island of La Palma (Spain), are sensitive to gamma rays in the energy range from 50 GeV to 50 TeV. MAGIC has performed indirect DM searches in various astrophysical targets, such as dwarf spheroidal galaxies and clusters of galaxies. Observations at high zenith angles significantly increase the telescopes' collection area and sensitivity for gamma rays in the TeV regime. We present the results obtained with more than 200 hours of high-zenith angle observations of the GC region with MAGIC, which allow us to probe promising heavy SUSY models, and to obtain competitive limits to the DM annihilation cross-section at high DM particle mass, compared to existing constraints. 17. Three-dimensional analysis of the palatal morphology in growing patients with Apert syndrome and Crouzon syndrome

Abstract: Patients with Apert syndrome or Crouzon syndrome present with severe defects in oral-maxillofacial growth and development. In this study, we conducted a quantitative three-dimensional (3D) analysis of the palatal morphology of patients with Apert syndrome and Crouzon syndrome. Four patients with Apert syndrome (average age, 11.0 ± 0.8 years) and five with Crouzon syndrome (average age, 10.1 ± 1.6 years) were investigated. The participants' maxillary dental casts were scanned and analyzed using 3D imaging. Palatal width, depth, cross-sectional area, and palatal angle (PW, PD, PCA, and PA, respectively) were measured, and standard scores were calculated based on sex- and age-matched Japanese standard values; the actual palatal surface areas (PSA) and palatal volumes (PV) were also measured. Our results show that patients with Apert syndrome and Crouzon syndrome had a very narrow PW (standard score: -3.79 and - 0.47, respectively). 3D analysis revealed that patients with Apert syndrome had a significantly shallower PD (standard score: -1.35) than those with Crouzon syndrome (standard score: 2.47), resulting in a smaller PCA (standard score: -5.13), PSA (5.49 cm

Abstract: We report the detection of pulsed emission from the Geminga pulsar (PSR J0633+1746) with the MAGIC Telescopes. After the Crab and Vela pulsars, Geminga is the third one detected in the very high energy domain, and its estimated age of ~340 ky makes it the oldest one. The spectrum derived by MAGIC extends from ~15 GeV to 75 GeV and can be well modeled with a simple, soft power-law function. For energies below 40 GeV it overlaps with Fermi-LAT measurements. Joint fits to MAGIC and Fermi-LAT data disfavour the existence of a sub-exponential cut-off in this energy range. Our results are discussed in the framework of the outer gap accelerator model. Such power-law emission can be interpreted as the transition from curvature radiation to inverse Compton (IC) scattering of charges accelerated in the northern outer gap. The IC component is expected to continue towards higher energies. The model fails to fit the overall shape of the spectrum, indicating that a major review is required. The MAGIC Telescopes are two IACTs on the Canary Island of La Palma. In recent years they have significantly improved their performance below 100 GeV with the introduction of a novel trigger system, the Sum-Trigger-II, which halves the energy threshold of the system.

Abstract: OT 081 is a luminous blazar well known for its variability in many energy bands. The very-high-energy (VHE; E >100 GeV) gamma-ray emission from the source was discovered by MAGIC and H.E.S.S. during flaring activity in July 2016, after a trigger from the Large Area Telescope (LAT) onboard the Fermi satellite. By analysing the multiwavelength light curves andthe broadband spectral energy distribution (SED), we study the activity of the source and investigate four individual states of activity in the window from MJD 57575 to 57602. The intrinsic gamma-ray spectrum can be described by a power law with spectral indices of 3.27 ± 0.44(MAGIC)and 3.39 ± 0.58(H.E.S.S.) over energy ranges 60–300 GeV and 120–500 GeV, respectively. The combined contemporaneous high-energy (HE;E >100 MeV) through VHE SED shows curvatureand can be described by a log-parabola shape. A simple one-zone synchrotron self-Compton (SSC)model is not sufficient to describe the broadband SED. The presence of broad emission lines in the optical spectrum of the source challenges the categorisation of OT 081 as a BL Lac and, together with the emission scenarios tested, points to the possibility that the source is transitional in nature between a BL Lac and a flat-spectrum radio quasar.

Abstract: The discovery of the electromagnetic counterpart AT2017gfo and the GRB 170817A, associated to the binary neutron star merger GW170817, was one of the major advances in the study of gamma- ray bursts (GRBs) and the hallmark of the multi-messenger astronomy with gravitational waves. Another breakthrough in GRB physics is represented by the discovery of the highly energetic, teraelectronvolt (TeV) component in the GRB 190114C, possibly an universal component in all GRBs. This conclusion is also suggested by the hint of TeV emission in the short GRB 160821B and a few more events reported in the literature. The missing observational piece is the joint detection of TeV emission and gravitational waves from a short GRB and its progenitor. MAGIC observed the counterpart AT2017gfo as soon as the visibility conditions allowed it, namely from January to June 2018. These observations correspond to the maximum flux level observed in the radio and X-ray bands. The upper limits derived from TeV observations are compared with the modelling of the late non-thermal emission using the multi-frequency SED.

Abstract: On 14th January 2019, the MAGIC collaboration achieved the first significant detection at TeV energies of a gamma-ray burst (GRB), namely GRB 190114C. This observation sets the first experimental proof of very high energy (VHE, >~100 GeV) gamma-ray emission in GRBs, after more than 50 years from the first GRB detection and many searches with Cherenkov telescopes in the last decades. The data collected by MAGIC and by more than 20 other ground-based and space-borne instruments, spanning 17 orders of magnitude in energy, revealed a new GeV-TeV emission component in the GRB afterglow. This unprecedented multi-wavelength dataset, including VHE data for the first time, allowed a detailed study of the broadband emission. A one-zone synchrotron-self Compton scenario with internal $\gamma$-$\gamma$ absorption could be used to describe the broadband emission, using parameters compatible with those found in previous studies of GRB afterglows below the GeV energy range. This detection opened a new era in the studies of GRBs, leading to new questions such as the universality of TeV emission in different types of GRBs. In this contribution, we will present the GRB follow-up program performed by the MAGIC collaboration, which started more than 15 years ago. We will highlight the results on GRB 190114C, discuss the implications for GRB physics, and report the latest developments and the prospects for future observations of GRBs with the MAGIC telescopes.

Abstract: The Large-Sized Telescope (LST) prototype of the future Cherenkov Telescope Array (CTA) is located at the Northern site of CTA, on the Canary Island of La Palma. It is designed to provide optimal performance in the lowest part of the energy range covered by CTA, observing gamma rays down to energies of tens of GeV. The LST prototype started performing astronomical observations in November 2019 during the commissioning of the telescope and it has been taking data since then. In this contribution, we will present the tuning of the characteristics of the telescope in the Monte Carlo (MC) simulations to describe the data obtained, the estimation of its angular and energy resolution, and an evaluation of its sensitivity, both with simulations and with observations of the Crab Nebula.

Abstract: Dwarf spheroidal galaxies (dSphs) are among the best candidates to perform indirect searches for DM, having the highest known mass-to-light ratio and being free of astrophysical gamma-ray emitting sources. The Major Atmospheric Gamma Imaging Cherenkov (MAGIC) telescopes, located on the Canary Island of La Palma, have observed a fair amount of optimal dSphs in the recent years. This is the outcome of diversifying the observation strategy in order to avoid possible biases in target selection and to improve previous results. In this contribution we will report on new MAGIC results obtained from 52.1 hours of observation of the Draco dSph in 2018 and 49.5 hours of the Coma Berenices dSph in 2019. We will also present the results of a joint analysis of Draco and Coma Berenices dSphs with other dSphs observed by MAGIC so far. The selected dataset accounts for 354.4 hours of good quality data, resulting in one of the largest dSphs samples ever collected by an array of Cherenkov telescopes. This allows us to derive the most constraining limits from dSphs, among Cherenkov telescopes, on the WIMP annihilation cross section for different annihilation channels in the WIMP mass range 70 GeV to 100 TeV.

Abstract: The flat-spectrum radio quasar QSO B1420+326 underwent an enhanced $\gamma$-ray flux state seen by Fermi-LAT at the turn of 2019/2020. Compared to the low state both the position and luminosity of the two spectral energy distribution peaks changed by at least two orders of magnitude. The high state resulted in the discovery of the very-high-energy (>100 GeV) $\gamma$-ray emission from the source by the MAGIC telescopes. The organized multiwavelength campaign allow us to trace the broadband emission of the source through different phases of the flaring activity. The source was observed by 20 instruments in radio, near-infrared, optical, ultra-violet, X-ray and $\gamma$-ray bands. We use dedicated optical spectroscopy results to estimate the accretion disc and the dust torus luminosity. The optical spectroscopy shows a prominent FeII bump with flux evolving together with the continuum emission and a MgII line with varying equivalent width. The $\gamma$-ray flare was accompanied by a rotation of the optical polarization vector and emission of a new superluminal radio knot. We model spectral energy distributions in different flare phases in the framework of combined synchrotron-self-Compton and external Compton scenario in which the shape of the electron energy distribution is determined from cooling processes.

26. Therapeutic effect of nanogel-based delivery of soluble FGFR2 with S252W mutation on craniosynostosis.

Abstract: Apert syndrome is an autosomal dominantly inherited disorder caused by missense mutations in fibroblast growth factor receptor 2 (FGFR2). Surgical procedures are frequently required to reduce morphological and functional defects in patients with Apert syndrome; therefore, the development of noninvasive procedures to treat Apert syndrome is critical. Here we aimed to clarify the etiological mechanisms of craniosynostosis in mouse models of Apert syndrome and verify the effects of purified soluble FGFR2 harboring the S252W mutation (sFGFR2IIIcS252W) on calvarial sutures in Apert syndrome mice in vitro. We observed increased expression of Fgf10, Esrp1, and Fgfr2IIIb, which are indispensable for epidermal development, in coronal sutures in Apert syndrome mice. Purified sFGFR2IIIcS252W exhibited binding affinity for fibroblast growth factor (Fgf) 2 but also formed heterodimers with FGFR2IIIc, FGFR2IIIcS252W, and FGFR2IIIbS252W. Administration of sFGFR2IIIcS252W also inhibited Fgf2-dependent proliferation, phosphorylation of intracellular signaling molecules, and mineralization of FGFR2S252W-overexpressing MC3T3-E1 osteoblasts. sFGFR2IIIcS252W complexed with nanogels maintained the patency of coronal sutures, whereas synostosis was observed where the nanogel without sFGFR2S252W was applied. Thus, based on our current data, we suggest that increased Fgf10 and Fgfr2IIIb expression may induce the onset of craniosynostosis in patients with Apert syndrome and that the appropriate delivery of purified sFGFR2IIIcS252W could be effective for treating this disorder.

Abstract: With the improved sensitivity with respect to the previous generation, current space-borne and ground-based gamma-ray telescopes have made the number of γ-ray sources detected at GeV- TeV energies increase many folds over the last decade. Many of the detected extended gamma-ray sources are not associated with any known sources at other wavelengths. Understanding the nature of these sources and the origin of the observed high energy gamma-ray emission remains a great challenge. Using the MAGIC telescopes, we have observed one such unassociated gamma-ray source, named HESS J1841-055 at TeV energies. We investigate the physical nature and origin of the γ-ray emission from this extended source. In this paper, we present the results of our detailed investigation on this source using MAGIC data and other multi-waveband information on nearby sources. The results included in this proceeding and shown in this presentation were published in ref. [1]. The details about the analysis, interpretation and modelling can be found in the aforementioned reference.

Abstract: Gamma-ray bursts (GRBs) are the most energetic phenomena in the Universe. Many aspects of GRB physics are still under debate, such as the origin of their gamma-ray emission above the GeV energy range. In 2019, MAGIC detected TeV gamma rays from the long GRB 190114C, whose emission can be well explained by synchrotron-self Compton emission by relativistic electrons. However, it is still unclear whether such a process is common in GRBs, given the reduced number of GRBs detected until now at the very high energies (VHE). GRB 201216C is a long GRB and is the second one detected by MAGIC in this energy range. After receiving the alert provided by Swift-BAT, MAGIC automatically slewed to the GRB position, starting observations 56 seconds after the GRB onset. In the offline analyses of the collected data, we confirmed the detection of gamma-ray emission with a significance above 5 sigma. Following measurements from optical facilities, the redshift of this GRB was estimated to be 𝑧� = 1.1. This makes GRB 201216C the most distant object ever detected by ground-based gamma-ray telescopes. In this contribution we will show the analysis results of the MAGIC data, also in comparison with past detected GRBs in the same energy range. Abstract: In February 2019, a flaring state of the extreme blazar candidate TXS 1515-273 was registered by the Fermi-LAT, which triggered observations with the MAGIC telescopes and the X-ray satellites Swift, XMM-Newton and NuStar. The observations led to the discovery of the source at very-high-energy (VHE, 100 GeV < E < $ 100 TeV) gamma-ray energies and the detection of short time scales of variability (1 h) in several X-ray bands. The analysis of the observed variability helped us to constrain the physical parameters of the emission region. Thanks to the high-quality X-ray data, the synchrotron peak location was determined. The source was classified as a high synchrotron peaked source during the flaring activity. We constructed the broadband spectral energy distribution from radio to TeV. We interpreted it assuming leptonic emission and taking into account the constraints from the X-ray variability. We tested two scenarios: a simple one-zone model and a two-component model. Both models were found to describe the data well from X-ray to VHE gamma ray, but the two-zone model allows for a more accurate modelling of the emission at radio and optical energies.

Abstract: Imaging Atmospheric Cherenkov telescopes (IACTs) are designed to detect cosmic gamma rays. As a by-product, IACTs detect Cherenkov flashes generated by millions of hadronic air showers every night. We present the proton energy spectrum from several hundred GeV to several hundred TeV, retrieved from the hadron induced showers detected by the MAGIC telescopes. The protons are discriminated from He and other heavy nuclei by means of using machine learning classification. The energy estimation is based on a specially developed deep neural network regressor. In the last decade, Deep Learning methods gained much interest in the scientific community for their ability to extract complex relations in data and process large datasets in a short time. The proton energy spectrum obtained in this work is compared to the spectra obtained by dedicated cosmic ray experiments.

Abstract: Silicon photomultipliers (SiPMs) have become the baseline choice for cameras of the small-sized telescopes (SSTs) of the Cherenkov Telescope Array (CTA). On the other hand, SiPMs are relatively new to the field and covering large surfaces and operating at high data rates still are challenges to outperform photomultipliers (PMTs). The higher sensitivity in the near infra-red and longer signals compared to PMTs result in higher night sky background rate for SiPMs. However, the robustness of the SiPMs represents a unique opportunity to ensure long-term operation with low maintenance and better duty cycle than PMTs. The proposed camera for large size telescopes will feature 0.05 degree pixels, low power and fast front-end electronics and a fully digital readout. In this work, we present the status of dedicated simulations and data analysis for the performance estimation. The design features and the different strategies identified, so far, to tackle the demanding requirements and the improved performance are described.

Abstract: The well-known supernova remnant (SNR) W 44 is observed in high-energy gamma rays and widely studied to investigate cosmic ray (CR) acceleration. Several analyses of the W 44 surroundings showed the presence of gamma-ray emission offset from the radio SNR shell. This emission is thought to originate from escaped high-energy CRs. We present a detailed analysis of the W 44 region as seen by Fermi-LAT, focusing on the spatial and spectral characteristics of both W44 SNR and its surroundings. The spatial analysis was limited to energies above 1 GeV in order to exploit the improved angular resolution of the instrument, deriving a detailed description of the region morphology. Observations of the north-western region of W 44, also known as SRC-1 from previous works, were conducted with the MAGIC telescopes in the very high-energy gamma-ray band. We analysed MAGIC data exploiting the spatial information derived with the Fermi-LAT analysis at GeV energies. Here we show the results of both analyses and the combined Fermi-LAT and MAGIC spectra, thus obtaining constraining information on the diffusion of the escaped CRs.

Abstract: The measurement of an astrophysical flux of high-energy neutrinos by IceCube is an important step towards finding the long-sought sources of cosmic rays. Nevertheless, the long exposure neutrino sky map shows no significant indication of point sources so far. The real-time followup of neutrino events turned out to be the most successful approach in neutrino point-source searches. It brought, among others, the most compelling evidence for a neutrino point source: the flaring gamma-ray blazar TXS 0506+056 in coincidence with a single high-energy neutrino from IceCube (IceCube-170922A). The fast multiwavelength(MWL) follow-up of this alert was key for establishing this coincidence and constraining the subsequent theoretical modeling for this event. In the long run, accurate and contemporaneous MWL spectral measurements are essential ingredients in investigating the physical processes leading to particle acceleration and emission of radiation. A deeper understanding of those processes allows us to put constraints on the potential neutrino emission. Here we present the light curves and simultaneous spectral energy distributions from November 2017 till February 2021 of MAGIC and MWL monitoring of TXS 0506+056. The more than the two year-long rise and high state of the radio light curve of TXS 0506+056, which started near the time of the IceCube neutrino detection, seems to have ended, as indicated by a steep decrease in the first half of 2021. Abstract: The periodontal ligament (PDL) plays an important role in orthodontic tooth movement; however, the underlying molecular mechanism remains unclear. We have previously reported that the Mohawk homeobox (Mkx), a tendon-specific transcription factor, is expressed in the PDL and regulates its homeostasis.In the present study, we examined the role of Mkx in orthodontic tooth movement via bone remodeling induced by mechanical stimulation in Mkx-deficient rats, which are widely used as experimental animals for orthodontic force application. Orthodontic tooth movement of the maxillary first molar was performed in 7-week-old male Mkx-deficient rats (n = 4) and wild-type Wistar rats (n = 4) using coil springs for 14 days. Hematoxylin and eosin (HE) staining and tartrate-resistant acid phosphatase (TRAP) staining were performed to evaluate morphological changes and osteoclasts. Furthermore, changes in the expression of receptor activator nuclear factor-kappa B ligand (RANKL) were demonstrated using immunostaining.The amount of tooth movement was significantly lower in Mkx-deficient rats than in wild-type rats. The number of TRAP-positive cells was suppressed in Mkx-deficient rats on the compression side.Orthodontic tooth movement experiments in Mkx-deficient rats suggested that Mkx is involved in osteoclast induction at the alveolar bone surface on the compression side. This study reveals the possibility that Mkx plays a mechanosensory role in orthodontic tooth movement by inducing RANKL expression and osteoclastogenesis.

Abstract: The Cherenkov Telescope Array (CTA) is the next-generation ground-based very-high-energy gamma-ray observatory. The Large-Sized Telescope (LST) of CTA is designed to detect gamma rays between 20 GeV and a few TeV with a 23-meter diameter mirror. We have developed the focal plane camera of the first LST, which has 1855 photomultiplier tubes (PMTs) and the readout system which samples a PMT waveform at GHz with switched capacitor arrays, Domino Ring Sampler ver4 (DRS4). To measure the precise pulse charge and arrival time of Cherenkov signals, we developed a method to calibrate the output voltage of DRS4 and the sampling time interval, as well as an analysis method to correct the spike noise of DRS4. Since the first LST was inaugurated in 2018, we have performed the commissioning tests and calibrated the camera. Abstract: The Hippo pathway is essential for determining organ size by regulating cell proliferation. Previous reports have shown that impairing this pathway causes abnormal tooth development. However, the precise expression profile of the members of the transcriptional enhanced associate domain family (Tead), which are key transcription factors mediating Yap function, during tooth development is unclear. In this study, among the four isoforms of Tead (Tead1 - 4), only the expression of Tead1 mRNA was observed using semiquantitative RT- PCR in murine developing tooth germ at E16.5. The expression level of Tead1 mRNA in the excised murine mandibular molar tooth germ was significantly higher at E16.5 than at other developmental stages, as determined using quantitative PCR. We found that the mRNA expression of connective tissue growth factor (Ctgf), which is one of the Yap target genes directly controlling cell growth, changed consistently with that of Tead1 in developing molars. Fluorescent immunostaining revealed that Tead1 protein was expressed in both epithelial cells and mesenchymal cells of the dental lamina and dental epithelium, including the primary enamel knot during the cap stage. During the early bell stage (E16.5), Tead1 was expressed intensely in the inner and outer enamel epithelium, including the secondary enamel knot and the neighboring mesenchymal cells. Tead1 then specifically localized to the inner and outer enamel epithelium, which is responsible for enamel formation during the bell stage. These expression patterns were consistent with those of Yap, Taz, and Ctgf protein in developing molars. These results suggest that Tead1 acts as a mediator of the biological functions of Yap, such as the morphogenesis of cusp formation, during tooth development.

Abstract: The Cherenkov Telescope Array (CTA) is the next-generation ground-based very-high-energy gamma-ray observatory. By using three types of telescopes CTA can cover a wide energy range (20 GeV–300 TeV) with an order of magnitude higher sensitivity than the current telescopes. The Large-Sized Telescope (LST) is designed to detect 20 GeV–1 TeV gamma rays thanks to the large light collection area, sensitive photosensors, a fast trigger system, and readout electronics. The camera readout system must have a high signal-to-noise ratio and a linear signal sampling with a large dynamic range in order to efficiently detect dim and low-energy atmospheric showers. To meet this requirement we use the Domino Ring Sampler version 4 (DRS4), which also enables ultra-fast sampling with low power consumption. Some of the intrinsic characteristics of DRS4 chips require software corrections. These procedures lower the effect of non-Gaussian noise contribution and improve the timing resolution of the system. In this contribution we discuss the calibration algorithms and the resulting performance.

Abstract: Relaxin (RLN) is an insulin-like peptide hormone that enables softening and lengthening of the pubic symphysis and uterine cervix. Here, we analyzed the effects of RLN2 on the expansion of rat midpalatal suture (MPS) using a magnetically directed liposome-based drug delivery system. Thirty-six male rats were divided into three groups: control (MPS was not expanded), lipo (expanded for 1 week with vehicle liposomes encapsulating ferric oxide and Cy5.5), and RLN-lipo (expanded for 1 week with the liposomes coated with RLN2). Rats were sacrificed after 1 week of expansion or after 2 weeks of retention. To accumulate RLN2-liposomes, a magnetic sheet was fixed to the palatal mucosa of the MPS. In vivo imaging showed magnetically controlled accumulation of liposomes in the MPS for 72 h. Immunohistochemistry revealed RLN2 expression in the MPS after expansion and relaxin receptor (RXFP) 2 expression at the osteogenic front (OF) in the RLN-lipo group; all groups expressed RXFP1 in the MPS. MPS expansion and bone formation were significantly accelerated at the OF in RLN-lipo group compared with the other groups. In the RLN-lipo group, significantly accelerated serrate bone deposition and elevated periostin (POSTN), iNOS, and MMP-1 levels were observed in the MPS. Sclerostin (SOST) expression was significantly reduced in newly formed bone in the RLN-lipo group. Our data revealed that RLN2 enhanced suture expansion via MMP-1 and iNOS secretion in the sutural fibroblasts and new bone formation via POSTN expression in osteoblasts at the OF. These properties may be useful for developing a new less-invasive orthopedic treatment aiming at sutural modification of cranio- and maxillofacial deformity patients., Highlights • In vivo Magnetically localization of RLN2 carried by liposome at rat midpalatal suture (MPS) was originally performed. • RLN2 promoted efficiency of the MPS expansion with secretion of Mmp1 and iNos in the mid-sutural fibroblasts. • During expansion period, RLN2 increased the number and differentiation of osteoblast cells in the MPS. • RLN2 enhanced newly bone formation at the MPS during expansion and retention period through Rxfp2. • Sinus-like bone formation and Postn localization at the expanded MPS was observed by RLN2 administration.

Abstract: RELAXIN (RLN) is a polypeptide hormone of the insulin-like hormone family; it facilitates birth by softening and widening the pubic symphysis and cervix in many mammals, including humans. The role of RLN in bone metabolism was recently suggested by its ability to induce osteoclastogenesis and activate osteoclast function. RLN binds to RELAXIN/INSULIN-LIKE FAMILY PEPTIDE 1 (RXFP1) and 2 (RXFP2), with varying species-specific affinities. Young men with mutated RXFP2 are at high risk for osteoporosis, as RXFP2 influences osteoblast metabolism by binding to INSULIN-LIKE PEPTIDE 3 (INSL3). However, there have been no reports on RLN function in osteoblast differentiation and mineralization or on the functionally dominant receptors for RLN in osteoblasts. We previously described Rxfp1 and 2 expression patterns in developing mouse oral components, including the maxillary and mandibular bones, Meckel's cartilage, tongue, and tooth primordia. We hypothesized that Rln/Rxfp signaling is a key mediator of skeletal development and metabolism. Here, we present the gene expression patterns of Rxfp1 and 2 in developing mouse calvarial frontal bones as determined by in situ hybridization. In addition, RLN enhanced osteoblastic differentiation and caused abnormal mineralization and extracellular matrix metabolism through Rxfp2, which was predominant over Rxfp1 in MC3T3-E1 mouse calvarial osteoblasts. Our data suggest a novel role for Rln in craniofacial skeletal development and metabolism through Rxfp2.

Abstract: Beckwith-Wiedemann syndrome (BWS) is a congenital growth disorder. Children born with BWS develop enlarged organs, including the tongue, a large body, and other signs. A woman with BWS was treated and followed for 30 years. Treatment consisted of tongue reduction, orthopedic and orthodontic treatment, orthognathic surgery, and retention. The patient was first treated when she was 5 years old. Her original orthodontic problems included macroglossia, anterior open bite, anterior crossbite, and a skeletal Class III jaw relationship caused by significant mandibular protrusion. The jaw-base relationships did not improve in the early preadolescent period after phase 1 of orthodontic treatment with a vertical chincap. With the growth spurt accompanying puberty, she developed a severe skeletal Class III jaw relationship and a constricted maxillary arch. Surgically assisted rapid maxillary expansion was performed at 23 years of age to correct the severe discrepancy between the maxillary and mandibular dental arch widths. Then, at 26 years, a LeFort I osteotomy, a horseshoe osteotomy, a bilateral sagittal split ramus osteotomy, and genioplasty were performed after presurgical orthodontic treatment with extraction of the mandibular first molars. Both the facial profile and the occlusion were stable after 6 years of retention. This case report discusses the result of long-term observation of a patient with BWS who underwent tongue reduction, early orthodontic treatment, and surgical-orthodontic treatment.

Abstract: Objective Relaxin is a pleiotropic hormone of the insulin-like peptide hormone family that plays an important role in reproductive physiology as well as in fibrosis, angiogenesis, and bone remodelling. It binds to the relaxin family peptide receptors 1 and 2 (Rxfp1 and Rxfp2) and can, in addition and independently, bind and activate the glucocorticoid receptor Nr3c1. Despite the wide-ranging effect of relaxin, the expression patterns of Rxfp1 and 2 during facial development have not been examined. In this study, we aimed to identify the mRNA expression patterns of Rxfp1, Rxfp2, and Nr3c1 in oral tissues during late mouse facial development in order to pinpoint the structures that could be sensitive to relaxin signalling during this period. Design Rxfp1, Rxfp2, and Nr3c1 mRNAs were identified by in situ hybridization using digoxigenin-labelled riboprobes on coronal sections of mouse heads from embryonic days 13.5 to 18.5. Results We found that Rxfp1, Rxfp2, and Nr3c1 mRNAs were expressed on the developing maxilla and mandible, Meckel's cartilage, tongue, and tooth primordia between embryonic days 13.5–18.5. Conclusions Receptors that bind relaxin were present in developing oral tissues of mice. This finding suggests that relaxin may be involved in the prenatal development of the face.
