Oxytocin (OXT) a neuropeptide synthesized in the hypothalamic nuclei has a variety of function including socio-emotional processes in mammals. While the neural circuits and signaling pathways in central OXT converge in the paraventricular nucleus of the hypothalamus (PVN), we illuminate specific function of discrete PVN OXT circuits, which connect to the medial amygdala (MeA) and the bed nucleus of the stria terminalis (BnST) in mouse models. The OXTPVN→BnST projections are innervated from entire portions of the PVN, while those OXTPVN→MeA projections are asymmetrically innervated from the posterior portion of the PVN. Compared with OXT neurons in B6 wild type mice, BTBR mice that are recognized as a behavior-based autism model exhibited defect in the OXTPVN→BnST projection. We demonstrate that chemogenetic activation of OXTPVN→MeA circuit enhances anxiety-like behavior and facilitates social approach behavior, while activation of OXTPVN→BnST circuit suppresses anxiety-like behavior along with inhibiting social approach. This chemogenetic manipulation on the OXTPVN→BnST circuit proves ineffective in BTBR mice. Accordingly, chemogenetic activation of OXTPVN neurons that stimulate both OXT circuits induces OXT receptor expressions in both MeA and BnST as with those by social encounter in B6 mice. The induction of OXT receptor genes in the BnST was not observed in BTBR mice. These data support the hypothesis that OXT circuits serve as a regulator for OXT signaling in PVN to control socio-emotional approach/avoidance behavior, and a defect of OXTPVN→BnST circuit contributes to autism-like social phenotypes in BTBR mice.

There has long been interest in the effect of stay abroad on pragmatic development in second language (L2) pragmatics research. However, few testing instruments exist to assess pragmatics learning in stay abroad and provide information about learners’ development to help institutions evaluate the success of stay abroad programs. In this study, we describe the adaptation and validation of Roever’s (2005) test of second language routine formulae to be used large-scale with Japanese stay-abroad learners. We follow Kane’s (2006) argument-based approach to validation, evaluating the first four inferences: Domain description, Evaluation, Generalization, and Explanation. We also investigate the test’s practicality within the institutional setting of intended use. We find evidence supporting the use of the test for providing information on pragmatics learning during stay abroad. The test can differentiate between low- and high-ability learners, shows adequate reliability, and reflects the roles of proficiency and exposure in pragmatics learning in stay abroad contexts. The test is also practical and requires few resources. Further steps, such as the investigation of the Extrapolation and Decision inference, are discussed.

In this study, we demonstrated that novel rice-derived bioactive peptides promote the secretion of ghrelin, an endogenous orexigenic hormone secreted from the stomach. The enzymatic digest of rice endosperm protein with subtilisin, a microorganism-derived enzyme, stimulated acylated ghrelin secretion in the ghrelin-releasing cell line MGN3-1 and increased food intake after oral administration in mice. By performing a comprehensive analysis based on structure-activity relationships, we selected candidate peptides from over 30,000 peptides in the rice digest. Among them, we found that QAFEPIRSV and TNPWHSPRQGSF, corresponding to the amino acid sequence of the rice endoplasmic proteins glutelin A1 or A2(52-60) and B1 or B2(31-42), respectively, stimulated acylated ghrelin release in MGN3-1 cells. Landslides and slope failures are often caused by earthquakes. This study proposes a method to map earthquake-induced slope failure hazards that uses the analytic hierarchy process (AHP) and a geographic information system (GIS) for four districts where many slope failures were induced by earthquakes (the 2018 Hokkaido Eastern Iburi, 2016 Kumamoto, 2008 Iwate-Miyagi Nairiku, and 2004 Mid Niigata Prefecture earthquakes). The assessment system, which was based on the National Research Institute for Earth Science and Disaster Resilience landslide distribution maps, was analyzed using the methods of previously published. We considered the relationships between the earthquake-induced slope failure distributions and landslide hazard factors (elevation, slope angle, slope type, catchment degree, geology, and vegetation). These relationships were utilized for pairwise comparisons of the factors in the AHP analysis. The slope angle, slope type, and catchment degree exerted the highest effects on the slope failure distribution in the four districts. The four earthquake-induced slope failure distributions were highly consistent with the slope failure hazard rank. These results provide a practical method for evaluating earthquake-induced slope-failure hazards.

BACKGROUND: Chronic kidney disease (CKD) impairs the anti-inflammatory effects of high-density lipoprotein (HDL) and increases cardiovascular mortality. Though the potential role of dietary interventions to manage HDL is well studied, the clinical trials aimed to increase HDL levels have failed to reduce cardiovascular risk, rendering HDL function to be explored as a more relevant clinical parameter. This study investigates the effects of rice endosperm protein (REP), a plant-based protein, on the anti-inflammatory properties of HDL and renal injury-driven atherosclerosis in comparison with casein, an animal protein. MATERIALS AND METHODS: Ten-week-old apolipoprotein E-deficient hyperlipidemic mice underwent uninephrectomy. The mice (n = 6 each) were pair-fed a normal casein-based diet or a REP-based diet (both with 20.0% protein content) for seven weeks. Atherosclerotic lesions were detected by en face Sudan IV staining of the aorta. RESULTS: The number and sizes of the atherosclerotic lesions were significantly lower in the REP-based diet-fed group than the casein-based diet-fed group (p = 0.038). However, the REP-based diet neither elicited an ameliorative effect on kidney function or histology nor impacted the cholesterol profiles. Furthermore, HDL from the REP-based diet-fed mice significantly suppressed the inflammatory cytokine response of human umbilical vein endothelial cells than that from the casein-based diet-fed mice (MCP-1, p = 0.010; IL-6, p = 0.011; IL-1β, p = 0.028). CONCLUSIONS: The REP-based diet has a higher potential to lessen the atherosclerotic lesions accelerated by renal mass reduction than a casein-based diet, which could be associated with the anti-inflammatory effects of HDL.

It is ideal to ingest bioactive substances from daily foods to stay healthy. Rice is the staple food for almost half of the human population. We found that an orally administered enzymatic digest of rice endosperm protein exhibits antidepressant-like effects in the tail suspension test (TST) using mice. We performed a comprehensive peptide analysis of the digest using liquid chromatography-tandem mass spectrometry, and the tridecapeptide QQFLPEGQSQSQK emerged as a candidate of the antidepressant-like peptide. Oral administration of the chemosynthetic peptide exhibited antidepressant-like effects at a low dose comparable to known drugs in the TST. This also exhibited anti-depressant-like effect in the forced swim test. We named it rice endosperm-derived antidepressant-like peptide (REAP). Intriguingly, intraperitoneal administration had no effect. Orally administered REAP(8–13) but not REAP(1–7) exhibited antidepressant-like activity, suggesting that the C-terminal structure is important for the antidepressant-like effect. We confirmed the presence of REAP, corresponding to rice glutelin type B4(130–142) and B5(130–142), in the digest. The effects of REAP were blocked by either intraperitoneal or intracerebroventricular administration of a dopamine D1 antagonist; however, REAP had no affinity for the receptor. These results suggest that it exerts its antidepressant-like activity through promotion of endogenous dopamine release and activation of central D1 receptors. REAP-induced antidepressant-like effect was at least in part blocked by vagotomy, implying the gut-brain communication is associated with REAP’s effect. Taken together, oral administration of a novel tridecapeptide exhibited antidepressant-like effects via the dopamine D1 system. This is the first report of a rice-derived peptide that exhibits antidepressant-like effects.

We conducted a randomized experiment targeting 322 Japanese high school students to examine the impacts of a newly developed English-language learning program. The treated students were offered an opportunity to communicate for 25 minutes with English-speaking Filipino teachers via Skype several times a week over a 5-month period as an extracurricular activity. The results show that the Skype program increased the interest of the treated students in an international vocation and in foreign affairs. However, the students did not improve their English communication abilities, as measured by standardized tests, probably because of the program's low utilization rate. Further investigation showed that the utilization rate was particularly low among students demonstrating a tendency to procrastinate. These results suggest the importance of maintaining students’ motivation to keep using such information and communication technology-assisted learning programs if they are not already incorporated into the existing curriculum. Having procrastinators self-regulate may be especially crucial.

Carotenoids are naturally synthesized in some species of bacteria, archaea, and fungi (including yeasts) as well as all photosynthetic organisms. Escherichia coli has been the most popular bacterial host for the heterologous production of a variety of carotenoids, including even xanthophylls unique to photosynthetic eukaryotes such as lutein, antheraxanthin, and violaxanthin. However, conversion efficiency of these epoxy-xanthophylls (antheraxanthin and violaxanthin) from zeaxanthin remained substantially low. We here examined several factors affecting their productivity in E. coli. Two sorts of plasmids were introduced into the bacterial host, i.e., a plasmid to produce zeaxanthin due to the presence of the Pantoea ananatis crtE, crtB, crtI, crtY, and crtZ genes in addition to the Haematococcus pluvialis IDI gene, and one containing each of zeaxanthin epoxidase (ZEP) genes originated from nine photosynthetic eukaryotes. It was consequently found that paprika (Capsicum annuum) ZEP (CaZEP) showed the highest conversion activity. Next, using the CaZEP gene, we performed optimization experiments in relation to E. coli strains as the production hosts, expression vectors, and ribosome-binding site (RBS) sequences. As a result, the highest productivity of violaxanthin (231 μg/g dry weight) was observed, when the pUC18 vector was used with CaZEP preceded by a RBS sequence of score 5000 in strain JM101(DE3).

The purpose of this study was to clarify outcomes on the lower limbs in cases of lying and taking Manual Lymph Drainage (MLD) for nurses after an 8-hour shift. The participants were eight females who were not pregnant, over 35 years old and had not had any operations on their bodies. We measured circumference of seven points on their lower limbs by two times on different days. It meant that we measured before and after at the time of lying in bed or taking MLD for 30 minutes. By the lying in bed, four points of the left side and one point of the right side decreased significantly the lower limb’s circumference. However, the circumference of the inguinal region and popliteal fossa did not change. By taking MLD, six points of on the left side and three points of right side decreased significantly the lower limb’s circumference. We found that the circumference of the lower limbs, including the inguinal region and popliteal fossa, were decreased. This result suggested MLD was a useful skill for to hasten blood circulation. To decrease the circumference of the lower limbs, we suggest that it is not only lying, but also add to MLD on their lower limbs.

While entrepreneurs play a key role in industrial development, the managerial capacity of those in developing countries seems limited. A number of randomized controlled trials have been conducted to evaluate the impacts of management training, coaching, or consultation programs. These studies found that the interventions had positive impacts on management knowledge and management practices, but the impacts on business performance—measured in terms of sales revenue, value added, or profit—were often statistically insignificant. Such mixed results may be attributed to experiment designs, including training content and the time elapsed till the follow-up observation. The present study attempts to substantiate this hypothesis by means of a randomized controlled trial of management training in Tanzania that targets 113 small manufacturers. As in some recent studies, it extends the observation period to three years to examine the trajectory of training impacts. Unlike many other studies, it is an impact assessment of training programs that covered quality control and production management as the training topics in addition to entrepreneurship, marketing, and record keeping. The treated firms made adaptive efforts to select useful practices and modify them to fit their business operation. It finds that the training effects on business performance are large and statistically significant, particularly in the medium-run.

Arginine vasopressin (AVP) is a neurohypophysial hormone that coordinatively regulates central socio-emotional behavior and peripheral control of antidiuretic fluid homeostasis. Most mammals, including rodents, utilize exocrine or urine-contained scent marking as a social signaling tool that facilitates social adaptation. The exocrine scent marking behavior is postulated to fine-tune sensory and cognitive abilities to recognize key social features via exocrine/urinary olfactory cues and subsequently control exocrine deposition or urinary marking through the mediation of osmotic fluid balance. AVP is implicated as a major player in controlling both recognition and signaling responses. This review provides constructive hypotheses on the coordinative processes of the AVP neurohypophysial circuits in the systemic regulations of fluid control and social-communicative behavior, via the expression of exocrine scent marking, and further emphasizes a potential role of AVP in a common mechanism underlying social communication in rodents.

Arginine-vasopressin (AVP) is a neurohypophyseal peptide that plays a critical role in the regulation of social behavior in mammals. Neuronal AVP regulates male-specific social signaling processes, such as exocrine urinary scent deposition and marking behavior in mice. In the periphery, AVP is transported to the portal bloodstream and acts as an antidiuretic hormone. These AVP dynamics imply that the central role of AVP in the stimulation of urinary marking is dissociated with the peripheral role of AVP in the retention of osmotic conditions. Using male BALB/c mice as subjects, peripheral injection of AVP decreased urinary marking and urination. In contrast, a central infusion of AVP facilitated urinary marking with no effect on urination, while an antagonist of the AVP 1a receptor inhibited marking. Centrally AVP-injected mice also exhibited typical behaviors, such as hiccough/sneeze-like reactions and flash scratching, particularly when confronted with a stimulus mouse through a wire mesh screen. Significant expression of these typical reactions in these mice resulted in the disruption of marking deposition. Further analysis of AVP synthesis illustrated that AVP levels increased in the midbrain but not in the circulation immediately after the test, particularly when confronted with a stimulus mouse. The central AVP regulates urinary marking and other typical behaviors in a dose- and situation-dependent manner. The sequential process implies that centrally synthesized AVP may be secreted into the circulation following immediate neuronal processes, and then peripheral AVP acts as an antidiuretic hormone on urinary marking behavior.

This study examines the association between social preferences and recycling behavior in a developing country. A conceptual framework that modifies the impure altruism model with heterogeneous preferences and further incorporates the impure public good model is presented to study informal recycling behaviors. Based on survey data collected from 755 households in Hanoi, Vietnam, we find that the majority of sample households recycle all the recyclable materials they consume even though such recycling is not obligatory. Half the recycling households sell their recyclables to informal junk buyers, whereas the other half provide recyclables without monetary compensation. Our finding of voluntary recycling implies that a certain proportion of households have a positive marginal willingness to pay for an increase in the environmental public good. The results of the regression analyses show that recycling behavior is associated with altruism, as measured by a modified version of the self-report altruism scale. Among high-income households, recycling behavior is associated with inequality aversion, as elicited from a hypothetical game. These findings suggest that social preferences are important determinants of the private provision of public goods through the informal sector in a developing country.

This study is the first report on liquid water and ice imaging conducted at a pulsed spallation neutron source facility. Neutron imaging can be utilised to visualise the water distribution inside polymer electrolyte fuel cells (PEFCs). Particularly, energy-resolved neutron imaging is a methodology capable of distinguishing between liquid water and ice, and is effective for investigating ice formation in PEFCs operating in a subfreezing environment. The distinction principle is based on the fact that the cross sections of liquid water and ice differ from each other at low neutron energies. In order to quantitatively observe transient freezing and thawing phenomena in a multiphase mixture (gas/liquid/solid) within real PEFCs with high spatial resolution, a pulsed neutron beam with both high intensity and wide energy range is most appropriate. In the validation study of the present work, we used water sealed in narrow capillary tubes to simulate the flow channels of a PEFC, and a pulsed neutron beam was applied to distinguish ice, liquid water and super-cooled water, and to clarify freezing and thawing phenomena of the water within the capillary tubes. Moreover, we have enabled the observation of liquid water/ice distributions in a large field of view (300 mm × 300 mm) by manufacturing a sub-zero environment chamber that can be cooled down to −30 °C, as a step towards in situ visualisation of full-size fuel cells.

This paper investigates the impact of management training programs on garment clusters in Vietnam and Tanzania. The study found that in the medium run firms showed improvement once they had identified useful practices and adapted them to their operations. Although it takes a few years to experience a significant impact on incomes, management training can increase not just management scores but also incomes or value added.

Obesity and related disorders, which are increasing in adults worldwide, are closely linked to childhood diet and are associated with chronic inflammation. Rice endosperm protein (REP) intake during adulthood has been reported to improve lipid metabolism and suppress the progression of diabetic kidney disease in animal models. However, the effects of REP intake during childhood on adulthood health are unclear. Therefore, we used a mouse model to experimentally investigate the preconditioning effects of REP intake during childhood on the development of obesity and related disorders in adulthood. Male C57BL/6J mice were pair-fed a normal-fat diet containing casein or REP during the juvenile period and then a high-fat diet (HFD) containing casein or REP during adulthood. Mice fed REP during the juvenile period showed better body weight, blood pressure, serum lipid profiles, lipopolysaccharide (LPS)-binding protein levels, and glucose tolerance in adulthood than those fed casein during the juvenile period. HFD-induced renal tubulo-glomerular alterations and hepatic microvesicular steatosis were less evident in REP-fed mice than in casein-fed ones. REP intake during the juvenile period improved HFD-induced dysbiosis (i.e., Escherichia genus proliferation and reduced gut microbiota diversity), thereby suppressing endotoxin-related chronic inflammation. Indeed, REP-derived peptides showed antibacterial activity against Escherichia coli, a major producer of LPS. In conclusion, REP supplementation during the juvenile period may regulate the gut microbiota and thus suppress the development of obesity and related disorders in adulthood in mice.

Angle-resolved photoelectron spectroscopy has been used to follow the valence band and near Fermi edge electronic band structure in the quasi-1D compound NaV 2 O 4 . In this current study we have acquired the very first high-quality, high-resolution ARPES data from this material. Our data clearly reveal two distinct dispersive bands that cross the Fermi level at different k F . This is a clear signature that the electronic properties of this material is affected by the presence of a mixed valence state on the different vanadium chains and possibly also the low-temperature magnetic spin order.

If the middle-income traps were merely equated with prolonged growth slowdowns occurring in the middle-income stage, as it is in some empirical studies, this concept would lose the meaning of existence. This paper proposes a new definition that considers the trap the aggravation of slowdowns due to inadequate responses, such as the adoption of counterproductive policies or the failure to adopt policies conducive to growth. By using data from high-, middle- and low-income economies in East Asia, the paper asks which symptoms we should inspect in order to diagnose the middle-income trap when an economy is having a growth slowdown.

This paper illustrates the sharp contrast in welfare impacts between the rich and the poor caused by typhoon Milenyo in a Philippine village. We find that fish prices dropped sharply due to the damage caused to fish pens near the village, leading to positive net welfare gains among the wealthy. In contrast , the poor do not consume much fish and thus did not gain from the sharp decline in prices. Arylative carboxylation of allenes proceeded in an intramolecular manner to afford the corresponding β,γ-unsaturated carboxylic acids in high yields using PdCl2/PAr3 (Ar = C6H4-p-CF3) and ZnEt2 under 1 atm of CO2. The intermediate of the cyclization/carboxylation sequence is thought to be a nucleophilic η1-allylethylpalladium, which reacts with CO2 at the γ-position of palladium. The products obtained could be efficiently converted into 3-substituted indole-2-carboxylate derivatives. One-pot synthesis of strychnocarpine, a β-carboline alkaloid, from the carboxylated product was also demonstrated.
