As an important indicator of sexual maturity of hens, age at first egg (AFE) is significantly associated with reproduction performance. In this study, 400 hens were divided into 6 groups based on AFE to analyze the difference of reproduction performance, reproduction hormone levels and the characterization of the hypothalamo–pituitary–gonadal axis in chickens. The results showed that the egg production of the hens in the late-maturing groups was significantly lower than that of the ones in other groups and the precocious hens had a lower egg production rate. The hens in late-maturing group had a lower fertility rate, LH levels and shorter duration of peak of egg production (PEP), the precocious hens had lower PRL levels. In addition, the characterization of the hypothalamo–pituitary–gonadal axis showed that the individuals with normal AFE had higher GNRH, GNRHR, ESR1, KITLG, and CYP11A1 expression levels than late-maturing and precocious individuals, which indicated that the chickens with normal AFE advantages on reproduction regulation system.

Meat quality is closely related to the development of skeletal muscle, in which PITX2 and SIX1 genes play important regulatory roles. The present study firstly provided the data of chronological expression files of PITX2 and SIX1 genes in the post-hatching pectoral muscle and analyzed the association of their polymorphisms with the meat quality traits of Wuliang Mountain Black-bone (WLMB) chickens. The results showed that both PITX2 and SIX1 genes were weakly expressed in the second and third weeks, and then increased significantly from the third week to the fourth week. Furthermore, there was a significant positive correlation between the expression levels of the two genes. Twelve and one SNPs were detected in the chicken PITX2 and SIX1 genes, respectively, of which four SNPs (g.9830C &gt, T, g.10073C &gt, T, g.13335G &gt, A, g.13726A &gt, G) of the PITX2 gene and one SNP (g.564G &gt, A) of the SIX1 gene were significantly associated with chicken meat quality traits. For the PITX2 gene, chickens with the CT genotype of g.9830C &gt, T showed the highest meat color L*, shear force (SF), pH, and the lowest electrical conductivity (EC), and drip loss (DL) (p &lt, 0.05 or p &lt, 0.01), chickens with the CC genotype of g.10073C &gt, T had the lowest L*, pH, and the highest DL (p &lt, 0.01). For the SIX1 gene, chickens with the GG genotype of g.564G &gt, A had the highest (p &lt, 0.05) SF and pH. Furthermore, pH had a significant correlation with all the other meat quality traits. The current study could contribute to the research of regulatory mechanisms of meat quality and lay the foundation for improving meat quality based on marker-assisted selection in chickens.

Simple Summary The production of chickens plays an increasingly important role in people’s daily life because of the high nutritional quality and relatively low cost. Meanwhile, with the improvement of living standards, the requirements for meat quality have also increased significantly. Thus, the improvement of meat quality is becoming more and more important in chicken breeding. Studies have shown that meat quality is closely related to the development of skeletal muscle, which is a complex process regulated by many genes. Paired-like homeodomain transcription factor 2 (PITX2) and SIX homeobox 1 (SIX1) genes are involved in skeletal muscle development, and their polymorphisms were significantly associated with the carcass and body size traits of chickens in our previous studies. The objective of the present study was to detect the chronological expression of them on the pectoral muscles, and evaluate whether their single nucleotide polymorphisms (SNPs) are associated with the meat quality traits in chickens. Sperm motility is one of the most important indicators to evaluate poultry fertility. In order to explore key molecular regulation roles related to sperm motility, we employed testicular RNA sequencing of pigeon. A total of 705 known and 385 novel microRNAs were identified. Compared with the low sperm motility group, four upregulated and two downregulated miRNAs in the high sperm motility group were identified. A total of 3567 target mRNAs were predicted and four target mRNAs were selected to validate by qPCR. The miRNA-mRNA interaction network analysis, indicated that mmu-miR-183-5p /FOXO1 and PC-3p-244994_31/CHDH pairs might affect sperm motility. GO and KEGG annotation analysis showed that target genes of differentially expressed miRNAs were related to serine/threonine kinase activity, ATP binding, Wnt and MAPK signaling pathway. The study provided a global miRNAs transcriptome of pigeon and a novel insight into the expression of the miRNAs in testes that associated with sperm motility.

In this study, we employed RNA sequencing to analyse the testis transcriptome (lncRNA and mRNA) of ten pigeons with high and low sperm motility. In total, 46,117 mRNAs and 17,463 lncRNAs were identified, of which 2673 mRNAs and 229 lncRNAs (P &lt, 0.05) were significantly differentially expressed (DE) between the high and low sperm motility groups. Gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) annotation analysis showed that target genes of DE lncRNAs and DE mRNAs were related to calcium ion binding, ATP binding, and spermatogenesis. Moreover, we found that UBB, a target gene of lncRNA MSTRG.7787.5, was involved in germ cell development. Our study provided a catalogue of lncRNAs and mRNAs associated with sperm motility, and they deserve further study to deepen the understanding of biological processes in the pigeon testis.
