Abstrct Background Apoptotic vesicles (ApoVs), which are extracellular vesicles released by apoptotic cells, have been reported to exhibit substantial therapeutic potential for inflammatory diseases and tissue regeneration. While extensive research has been dedicated to mesenchymal stem cells (MSCs), the investigation into immune cell-derived ApoVs remains limited, particularly regarding the function and fate of macrophage-derived ApoVs in the context of periodontitis (PD). Results Our study corroborates the occurrence and contribution of resident macrophage apoptosis in Porphyromonas gingivalis (Pg)-associated PD. The findings unveil the pivotal role played by apoptotic macrophages and their derived ApoVs in orchestrating periodontal bone remodeling. The enrichments of diverse functional miRNAs within these ApoVs are discerned through sequencing techniques. Moreover, our study elucidates that the macrophage-derived ApoVs predominantly deliver miR-143-3p, targeting insulin-like growth factor-binding protein 5 (IGFBP5), thereby disrupting periodontal bone homeostasis. Conclusions Our study reveals that macrophages in Pg-associated PD undergo apoptosis and generate miR-143-3p-enriched ApoVs to silence IGFBP5, resulting in the perturbation of osteogenic-osteoclastic balance and the ensuing periodontal bone destruction. Accordingly, interventions targeting miR-143-3p in macrophages or employment of apoptosis inhibitor Z-VAD hold promise as effective therapeutic strategies for the management of PD.

Abstract This study employed Environmental DNA (eDNA) barcoding technology to delve into the influence of the tributaries and mainstem on fish diversity and spatiotemporal distribution in a hotspot fish conservation area in the upper Yangtze River. A total of 123 fish species were detected, belonging to 7 orders, 19 families, and 77 genera. The composition of fish species in tributaries is similar to that in mainstem, with higher fish community diversity in tributaries during the spring and summer. Exploration of fish ecotypes revealed significant differences between mainstem and tributaries. The fish community is mainly influenced by key environmental factors such as water temperature, dissolved oxygen, electrical conductivity, and ammonia nitrogen, with a higher impact of these factors on tributaries than on mainstem. In conclusion, while tributaries and mainstem in the Jiangjin section exhibit similarities in fish community composition, there are notable differences in community structure and diversity. Therefore, the protection of not only mainstem but also tributaries and their associated fish habitats is crucial for promoting the overall health and sustainability.

To investigate the effect of sulfated modification on the structure and function of coconut haustorium polysaccharide (CHP), three kinds of sulfated coconut haustorium polysaccharides (SCHP-2, SCHP-3, and SCHP-4) were prepared using sulfation reagents containing different ratios of chlorosulfonic acid and pyridine (v/v, mL/mL: 1/2, 1/3, and 1/4) after extracting CHP from the raw materials of coconut haustorium, and their basic chemical compositions, structural characteristics and antioxidant activity in vitro were compared and analyzed. Results showed that all three sulfation reagents were able to modify CHP (characteristic peaks of S=O and C-O-S were observed near 1100~1250 cm−1 and 820 cm−1 in infrared spectra, respectively), and the degree of substitution (DS) was the highest (0.75) at the ratio of chlorosulfonic acid and pyridine for 1/2. The total sugar, uronic acid content, and molecular weight of the three SCHP decreased, and the molar content of monosaccharides also changed when comparing with that of CHP. The thermal stability had slightly weakened as well (the degradation temperature of polysaccharides decreased from 211.17 ℃ and 282.01 ℃ of CHP to 194.86~207.70 ℃ and 233.35~262.46 ℃ of SCHP, respectively). The results of antioxidant experiments in vitro showed that the scavenging activity of SCHP on DPPH and hydroxyl radicals was lower than that of CHP, and the larger the DS was, the more obvious the decrease in activity was. Abstract Background Idiopathic pulmonary fibrosis (IPF) is a severe lung condition, and finding better ways to diagnose and treat the disease is crucial for improving patient outcomes. Our study sought to develop an artificial neural network (ANN) model for IPF and determine the immune cell types that differed between the IPF and control groups. Methods From the Gene Expression Omnibus (GEO) database, we first obtained IPF microarray datasets. To conduct protein-protein interaction (PPI) networks and enrichment analyses, differentially expressed genes (DEGs) were screened between tissues of patients with IPF and tissues of controls. Afterward, we identified the important feature genes associated with IPF using random forest (RF) analysis, and then constructed and validated a prediction ANN mode. In addition, the proportions of immune cells were quantified using cell-type identification by estimating relative subsets of RNA transcripts (CIBERSORT) analysis, which was performed on microarray datasets based on gene expression profiling. Results A total of 11 downregulated and 36 upregulated DEGs were identified. PPI networks and enrichment analyses were carried out; the immune system and extracellular matrix were the subjects of the enrichments. Using RF analysis, the significant feature genes LRRC17, COMP, ASPN, CRTAC1, POSTN, COL3A1, PEBP4, IL13RA2, and CA4 were identified. The nine feature gene scores were integrated into the ANN to develop a diagnostic prediction model. The receiver operating characteristic (ROC) curves demonstrated the strong diagnostic ability of the ANN in predicting IPF in the training and testing sets. An analysis of IPF tissues in comparison to normal tissues revealed a reduction in the infiltration of natural killer cells resting, monocytes, macrophages M0, and neutrophils; conversely, the infiltration of T cells CD4 memory resting, mast cells, and macrophages M0 increased. Conclusion LRRC17, COMP, ASPN, CRTAC1, POSTN, COL3A1, PEBP4, IL13RA2, and CA4 were determined as key feature genes for IPF. The nine feature genes in the ANN model will be extremely important for diagnosing IPF. It may be possible to use differentiated immune cells from IPF samples in comparison to normal samples as targets for immunotherapy in patients with IPF.

Abstract Purpose To evaluate the predictive capabilities of MRI-based radiomics for detecting lymphovascular space invasion (LVSI) in patients diagnosed with endometrial carcinoma (EC). Materials and methods A retrospective analysis was conducted on 160 female patients diagnosed with EC. The radiomics model including T2-weighted and dynamic contrast-enhanced MRI (DCE-MRI) images was established. Additionally, a conventional MRI model, which incorporated MRI-reported FIGO stage, deep myometrial infiltration (DMI), adnexal involvement, and vaginal/parametrial involvement, was established. Finally, a combined model was created by integrating the radiomics signature and conventional MRI characteristics. The predictive performance was validated by the area under the curve (AUC) of the receiver operating characteristic (ROC) curves. A stratified analysis was conducted to compare the differences between the three models by Delong test. Results In predicting LVSI, the radiomics model outperformed the clinical model in the training cohort (AUC: 0.899 vs. 0.8862) but not in the test cohort (AUC: 0.812 vs. 0.8758). The combined model demonstrated superior performance in both the training and test cohorts (training cohort: AUC = 0.934, 95% CI: 0.8807–0.9873; testing cohort: AUC = 0.905, 95% CI: 0.7679-1). Conclusions The combined model exhibited utility in preoperatively predicting LVSI in patients with EC, offering potential benefits for clinical decision-making.

Oxy-combustion is a promising carbon-capture technology, but atmospheric-pressure oxy-combustion has a relatively low net efficiency, limiting its application in power plants. In pressurized oxy-combustion (POC), the boiler, air separation unit, flue gas recirculation unit, and CO2 purification and compression unit are all operated at elevated pressure; this makes the process more efficient, with many advantages over atmospheric pressure, such as low NOx emissions, a smaller boiler size, and more. POC is also more promising for industrial application and has attracted widespread research interest in recent years. It can produce high-pressure CO2 with a purity of approximately 95%, which can be used directly for enhanced oil recovery or geo-sequestration. However, the pollutant emissions must meet the standards for carbon capture, storage, and utilization. Because of the high oxygen and moisture concentrations in POC, the formation of acids via the oxidation and solution of SOx and NOx can be increased, causing the corrosion of pipelines and equipment. Furthermore, particulate matter (PM) and mercury emissions can harm the environment and human health. The main distinction between pressurized and atmospheric-pressure oxy-combustion is the former’s elevated pressure; thus, the effect of this pressure on the pollutants emitted from POC—including SOx, NOx, PM, and mercury—must be understood, and effective control methodologies must be incorporated to control the formation of these pollutants. This paper reviews recent advances in research on SOx, NOx, PM, and mercury formation and control in POC systems that can aid in pollutant control in such systems.

Abstract Multi‐agent reinforcement learning relies on reward signals to guide the policy networks of individual agents. However, in high‐dimensional continuous spaces, the non‐stationary environment can provide outdated experiences that hinder convergence, resulting in ineffective training performance for multi‐agent systems. To tackle this issue, a novel reinforcement learning scheme, Mutual Information Oriented Deep Skill Chaining (MioDSC), is proposed that generates an optimised cooperative policy by incorporating intrinsic rewards based on mutual information to improve exploration efficiency. These rewards encourage agents to diversify their learning process by engaging in actions that increase the mutual information between their actions and the environment state. In addition, MioDSC can generate cooperative policies using the options framework, allowing agents to learn and reuse complex action sequences and accelerating the convergence speed of multi‐agent learning. MioDSC was evaluated in the multi‐agent particle environment and the StarCraft multi‐agent challenge at varying difficulty levels. The experimental results demonstrate that MioDSC outperforms state‐of‐the‐art methods and is robust across various multi‐agent system tasks with high stability.

Pheretimoid earthworms, Amynthas carnosus, were collected from Northeast and North China. An update on the distribution and systematics of the A. carnosus complex in eastern Asia using both morphological and molecular data is provided. Three subspecies, A. carnosus carnosus, A. carnosus naribunji, and A. carnosus roki, are confirmed. Comparisons of morphological characters between the subspecies of A. carnosus are provided. Our results support the subspecies assignment with an intraspecific K2P genetic distance of not greater than 10% using the mitochondrial cytochrome c oxidase subunit I (COI). In addition, a re-description of the morphology of A. carnosus naribunji is presented here.

ABSTRACTThis study aimed to provide data for the clinical features of invasive pneumococcal disease (IPD) and the molecular characteristics of Streptococcus pneumoniae isolates from paediatric patients in China. We conducted a multi-centre prospective study for IPD in 19 hospitals across China from January 2019 to December 2021. Data of demographic characteristics, risk factors for IPD, death, and disability was collected and analysed. Serotypes, antibiotic susceptibility, and multi-locus sequence typing (MLST) of pneumococcal isolates were also detected. A total of 478 IPD cases and 355 pneumococcal isolates were enrolled. Among the patients, 260 were male, and the median age was 35 months (interquartile range, 12–46 months). Septicaemia (37.7%), meningitis (32.4%), and pneumonia (27.8%) were common disease types, and 46 (9.6%) patients died from IPD. Thirty-four serotypes were detected, 19F (24.2%), 14 (17.7%), 23F (14.9%), 6B (10.4%) and 19A (9.6%) were common serotypes. Pneumococcal isolates were highly resistant to macrolides (98.3%), tetracycline (94.1%), and trimethoprim/sulfamethoxazole (70.7%). Non-sensitive rates of penicillin were 6.2% and 83.3% in non-meningitis and meningitis isolates. 19F-ST271, 19A-ST320 and 14-ST876 showed high resistance to antibiotics. This multi-centre study reports the clinical features of IPD and demonstrates serotype distribution and antibiotic resistance of pneumococcal isolates in Chinese children. There exists the potential to reduce IPD by improved uptake of pneumococcal vaccination, and continued surveillance is warranted.

Background The microbiomes on the surface of unclean removable prostheses are complex and yet largely underexplored using metagenomic sequencing technology.Objectives To characterize the microbiome of removable prostheses with different levels of cleanliness using Type IIB Restriction-site Associated DNA for Microbiome (2bRAD-M) sequencing and compare the Microbial Index of Pathogenic Bacteria (MIP) between clean and unclean prostheses.Materials and Methods Ninety-seven removable prostheses were classified into ‘clean’ and ‘unclean’ groups. All prosthesis plaque samples underwent 2bRAD metagenomic sequencing to characterize the species-resolved microbial composition. AbstractHorizontal platelet-rich fibrin (H-PRF) contains a variety of bioactive growth factors and cytokines that play a key role in the process of tissue healing and regeneration. The blood collection tubes used to produce Solid-PRF (plasmatrix (PM) tubes) have previously been shown to have a great impact on the morphology, strength and composition of the final H-PRF clot. Therefore, modification to PM tubes is an important step toward the future optimization of PRF. To this end, we innovatively modified the inner wall surface of the PM tubes with plasma and adjusted the gas environment inside the PM tubes to prepare super-hydrophilic anaerobic plasmatrix tubes (SHAP tubes). It was made anaerobic for the preparation of H-PRF with the aim of improving mechanical strength and bioactivity. The findings demonstrated that an anaerobic environment stimulated platelet activation within the PRF tubes. After compression, the prepared H-PRF membrane formed a fibrous cross-linked network with high fracture strength, ideal degradation characteristics, in addition to a significant increase in size. Thereafter, the H-PRF membranes prepared by the SHAP tubes significantly promoted collagen synthesis of gingival fibroblast and the mineralization of osteoblasts while maintaining excellent biocompatibility, and advantageous antibacterial properties. In conclusion, the newly modified PRF tubes had better platelet activation properties leading to better mechanical strength, a longer degradation period, and better regenerative properties in oral cell types including gingival fibroblast and alveolar osteoblasts. It also improves the success rate of H-PRF preparation in patients with coagulation dysfunction and expands the clinical application scenario.

The rich and special biodiversity in mountain rivers is experiencing a continuous decline due to the influence of climate change and anthropogenic factors. To explore the alterations in the distribution of aquatic biodiversity in mountain rivers in the context of cascade dam development, the environmental DNA (eDNA) metabarcoding technique was used to examine the aquatic biodiversity of the Wujiang River, a representative mountain river with an 11-stage cascade dam. In this study, a comprehensive analysis of aquatic organisms in the Wujiang River mainstem was conducted, and 17 phyla and 38 classes were detected in the 36 water samples. The most abundant group is the benthic organisms, and the least abundant group is the amphibians. The results of the diversity analysis showed a clear longitudinal distribution pattern of aquatic organisms along the altitudinal gradient, with higher richness of aquatic organism communities the further to the lower reaches. Meanwhile, RDA analysis revealed that altitude, dissolved oxygen, reservoir length, and reservoir construction time were the key environmental factors influencing the distribution of aquatic organisms in the Wujiang River mainstem. The findings of this research also showed the applicability of the eDNA method in detecting aquatic biodiversity.

IntroductionOsteoarthritis (OA) is a prevalent joint disease that severely impacts patients’ quality of life. Due to its unclear pathogenesis and lack of effective therapeutic targets, discovering new biomarkers for OA is essential. Recently, the role of chondrocyte subpopulations in OA progression has gained significant attention, offering potential insights into the disease. This study aimed to explore the role of fibrocartilage chondrocytes (FC) in the progression of OA and identify key biomarkers related to FC.MethodsWe analyzed single-cell ribonucleic acid sequencing (scRNA-seq) data from samples of OA and normal cartilage, focusing on FC. Microarray data were integrated to identify differentially expressed genes (DEGs). We conducted functional-enrichment analyses, including Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO), and used weighted gene co-expression network analysis (WGCNA) and the least absolute shrinkage and selection operator (LASSO) algorithm to select biomarkers. A novel risk model for OA was constructed using these biomarkers. We then built a transcription factor (TF)–gene interaction network and performed immunohistochemistry (IHC) to validate protein expression levels of these biomarkers in cartilage samples.ResultsThe study identified 545 marker genes associated with FC in OA. GO and KEGG analyses revealed their biological functions; microarray analysis identified 243 DEGs on which functional-enrichment analysis were conducted. Using WGCNA and LASSO, we identified six hub genes, on the basis of which we constructed a risk model for OA. In addition, correlation analysis revealed a close association between Forkhead Box (FoxO)-mediated transcription and these these biomarkers. IHC showed significantly lower protein levels of ABCA5, ABCA6 and SLC7A8 in OA samples than in normal samples.ConclusionThis study used a multi-omics approach to identify six FC-related OA biomarkers (BCL6, ABCA5, ABCA6, CITED2, NR1D1, and SLC7A8) and developed an exploratory risk model. Functional enrichment analysis revealed that the FoxO pathway may be linked to these markers, particularly implicating ABCA5 and ABCA6 in cholesterol homeostasis within chondrocytes. These findings highlight ABCA family members as novel contributors to OA pathogenesis and suggest new therapeutic targets.

ObjectiveAbdominal aortic aneurysm (AAA) is a life-threatening vascular condition. This study aimed to discover new indicators for the early detection of AAA and explore the possible involvement of immune cell activity in its development.MethodsSourced from the Gene Expression Omnibus, the AAA microarray datasets GSE47472 and GSE57691 were combined to generate the training set. Additionally, a separate dataset (GSE7084) was designated as the validation set. Enrichment analyses were carried out to explore the underlying biological mechanisms using Disease Ontology, Kyoto Encyclopedia of Genes and Genomes, and Gene Ontology. We then utilized weighted gene co-expression network analysis (WGCNA) along with 3 machine learning techniques: least absolute shrinkage and selection operator, support vector machine-recursive feature elimination, and random forest, to identify feature genes for AAA. Moreover, data were validated using the receiver operating characteristic (ROC) curve, with feature genes defined as those having an area under the curve above 85% and a p-value below 0.05. Finally, the single sample gene set enrichment analysis algorithm was applied to probe the immune landscape in AAA and its connection to the selected feature genes.ResultsWe discovered 72 differentially expressed genes (DEGs) when comparing healthy and AAA samples, including 36 upregulated and 36 downregulated genes. Functional enrichment analysis revealed that the DEGs associated with AAA are primarily involved in inflammatory regulation and immune response. By intersecting the result of 3 machine learning algorithms and WGCNA, 3 feature genes were identified, including MRAP2, PPP1R14A, and PLN genes. The diagnostic performance of all these genes was strong, as revealed by the ROC analysis. A significant increase in 15 immune cell types in AAA samples was observed, based on the analysis of immune cell infiltration. In addition, the 3 feature genes show a strong linkage with different types of immune cells.ConclusionThree feature genes (MRAP2, PPP1R14A, and PLN) related to the development of AAA were identified. These genes are linked to immune cell activity and the inflammatory microenvironment, providing potential biomarkers for early detection and a basis for further research into AAA progression.

Background: This study aims to provide an estimated dataset of lens zonule balance tension (LZBT) measurements in cataractous lenses among Chinese patients and to conduct a statistical analysis of anterior segment parameters. Methods: This is a cross-sectional study. We included a total of 833 eyes from 833 Chinese participants aged 23–91 years who underwent cataract surgery. Anterior segment parameters were measured using swept-source anterior segment optical coherence tomography (SS-AS OCT) to calculate lens gravity (LG) and crystalline lens volume (VOL). Axial length (AL) was measured using IOL-Master 700. LZBT was calculated using force decomposition and synthesis methods. Results: The mean LZBT in the horizontal direction was 8.48E-05 ± 3.23E-05 N across all eyes. The LZBT in the subtemporal-supraspinal and supraspinal-temporal directions was 2 times greater than in the vertical direction. The balanced force on the suspensory ligament in the horizontal direction was twice as strong as in the vertical direction. Additionally, anterior segment parameters such as lens equivalent diameter (LE-dia), radius of the anterior lens surface curvature (RAL), radius of the posterior lens surface curvature (RPL), anterior chamber depth (ACD), iris area, iris volume, and iris thickness showed positive correlations with AL. Conclusions: Accounting for gravitational effects, we concluded that zonule force is asymmetrically distributed. Importantly, this study establishes the normal range of LZBT across different meridians using force synthesis and decomposition, offering new insights and feasibility for studying suspensory ligament biomechanics.

Abstract The brine‐seawater interface (BSI) is a unique type of groundwater‐seawater interface (GSI) characterized by the higher density of underground brine compared to seawater. This study focuses on characterizing the bay‐scale BSI morphology and identifying submarine‐groundwater discharge windows using a comprehensive in‐situ geophysical detection on the south bank of Laizhou Bay. Our findings reveal that the BSI forms an extensive mixing zone (15–20 km) without distinct contours between waters of varying salinities. The discharge windows for underground brine are located in nearshore areas with fine sand distribution and offshore pockmark areas. Hydraulic and salinity gradients drive the underground brine discharge through these windows. The aquitard window is the primary area for shallow and deep brine exchange, likely evolved from paleochannels, ancient tidal creeks, or ancient underwater barriers. These findings provide crucial modeling support for analyzing environmental evolution mechanisms and theoretical basis for planning the underground brine mining in similar coastal regions.

Biofilms are complex bacterial communities characterized by a high persister prevalence, which contributes to chronic and relapsing infections. Historically, persister formation in biofilms has been linked to constraints imposed by their dense structures. However, we observed an elevated persister frequency accompanying the stage of cell adhesion, marking the onset of biofilm development. Subsequent mechanistic studies uncovered a comparable type of toxin-antitoxin (TA) module (TA-like system) triggered by cell adhesion, which is responsible for this elevation. In this module, the toxin HipH acts as a genotoxic deoxyribonuclease, inducing DNA double strand breaks and genome instability. While the second messenger c-di-GMP functions as the antitoxin, exerting control over HipH expression and activity. The dynamic interplay between c-di-GMP and HipH levels emerges as a crucial determinant governing genome stability and persister generation within biofilms. These findings unveil a unique TA system, where small molecules act as the antitoxin, outlining a biofilm-specific molecular mechanism influencing genome stability and antibiotic persistence, with potential implications for treating biofilm infections.

Abstract Background and aims The prognosis of hepatocellular carcinoma (HCC) with macrovascular invasion(MaVI)is poor, and the treatment is limited. This study aims to explore the efficacy and safety of hepatic arterial infusion chemotherapy (HAIC), combined with lenvatinib and programmed cell death-1(PD-1) inhibitor in the first-line treatment of HCC with MaVI. Methods From July 2020 to February 2022, we retrospectively analyzed consecutive patients with HCC with MaVI who received hepatic arterial infusion FOLFOX(oxaliplatin, 5-fluorouracil, and leucovorin)combined with lenvatinib and PD-1 inhibitor. The efficacy was evaluated by RECIST 1.1. Kaplan-Meier was used to explore the overall survival and progression-free survival (PFS), and the COX regression model was used to analyze the risk factors of PFS. Adverse events (AEs) were evaluated according to CTCAE5.0. Results Thirty-two patients with HCC complicated with MaVI were recruited from the Second Affiliated Hospital of Nanchang University. Among the patients treated with HAIC combined with lenvatinib and PD-1 inhibitor, ten patients (31.25%) got partial response, eighteen patients (56.25%) maintained stable disease and four patients (12.50%) suffered progressive disease during follow-up; and objective response rate was 31.25%, and disease control rate was 87.5%. The median PFS was 179 days. Univariate and multivariate Cox analysis showed that the extrahepatic metastases and Child-Pugh score were independent prognostic factors of PFS. Twenty-two (68.75%) patients suffered adverse reactions. The main AEs were elevated transaminase (46.87%), thrombocytopenia (40.63%), hypoalbuminemia (28.13%), nausea and vomiting (21.88%), leukopenia (18.76%), abdominal pain (15.63%), hypertension (15.63%) and fever (15.63%). There were seven cases (21.88%) that had grade 3 or above AEs; Among them, two cases with elevated transaminase (6.25%), leukopenia, thrombocytopenia, nausea and vomiting, abdominal pain, and diarrhea occurred in one case respectively. Moreover, no treatment-related death was observed. Conclusions Hepatic arterial infusion of FOLFOX combined with lenvatinib and PD-1 inhibitor as the first-line treatment for HCC complicated with MaVI is effective, and adverse reactions are tolerable.

id="st1" To explore which pattern recognition receptors (PRRs) play a key role in the development of hand, foot, and mouth disease (HFMD) by analyzing PRR-associated genes. id="st2" We conducted a comparative analysis of PRR-associated gene expression in human peripheral blood mononuclear cells (PBMCs) infected with enterovirus 71 (EV-A71) which were derived from patients with HFMD of different severities and at different stages. A total of 30 PRR-associated genes were identified as significantly upregulated both over time and across different EV-A71 isolates. Subsequently, ELISA was employed to quantify the expression of the six most prominent genes among these 30 identified genes, specifically, BST2, IRF7, 1FI16, TRIM21, MX1, and DDX58. id="st3" Compared with those at the recovery stage, the expression levels of BST2 (P=0.027), IFI16 (P=0.016), MX1 (P=0.046) and DDX58 (P=0.008) in the acute stage of infection were significantly upregulated, while no significant difference in the expression levels of IRF7 (P=0.495) and TRIM21 (P=0.071) was found between different stages of the disease. The expression levels of BST2, IRF7, IFI16 and MX1 were significantly higher in children infected with single pathogen than those infected with mixed pathogens, and BST2, IRF7, IFI16 and MX1 expression levels were significantly lower in coxsackie B virus (COXB) positive patients than the negative patients. Expression levels of one or more of BST2, IRF7, IFI16, TRIM21, MX1 and DDX58 genes were correlated with PCT levels, various white blood cell counts, and serum antibody levels that reflect disease course of HFMD. Aspartate aminotransferase was correlated with BST2, MX1 and DDX58 expression levels. id="st4" PRR-associated genes likely initiate the immune response in patients at the acute stage of HFMD.

Despite providing significant assistance to human society, cascade dams can also have negative impacts on river ecosystems. As the crucial components of river ecosystem, the responses of phytoplankton and zooplankton to cascade dams have rarely been studied simultaneously, and thus, lacking the understanding of the difference in succession between them. Here, we investigated the phytoplankton and zooplankton communities in a river-way with cascade dams using an environmental DNA metabarcoding technology. Along the reservoir areas separated by dams, we found an obvious downward trend in diversity of plankton communities with significant variations in their compositions. The relative abundances of Bacillariophyta and Chlorophyta continued to decrease while Intramacronucleata increased along the river-way. Results of association analyses recognized temperature and flow rate as potential factors resiling the impacts of cascade dams. Beta diversity decomposition indicated species replacement as the main mechanism for variations in plankton communities with higher contribution for phytoplankton. Additionally, we detected wider environmental adaptation (broader environmental breadth, phylogenetic single, and niche breadth) and stronger dispersal ability in phytoplankton than in zooplankton. Environmental variables showed a stronger effect for variations in phytoplankton than zooplankton. Furthermore, we observed that community assembly of phytoplankton and zooplankton was, based on the null model, by heterogeneous selection and drift, respectively. These results suggested differences in phytoplankton and zooplankton response to cascade dams and highlighted the stronger environmental filtering in phytoplankton.

PurposeAttention, a complex cognitive process, is linked to the functional activities of the brain’s dorsal attention network (DAN) and default network (DN). This study aimed to investigate the feasibility, safety, and blinding efficacy of a transcranial direct current stimulation (tDCS) paradigm designed to increase the excitability of the DAN while inhibiting the DN (DAN+/DN-tDCS) on attention function in healthy young adults.MethodsIn this randomized controlled experiment, participants were assigned to either the DAN+/DN-tDCS group or the sham group. A single intervention session was conducted at a total intensity of 4 mA for 20 min. Participants completed the Attention Network Test (ANT) immediately before and after stimulation. Blinding efficacy and adverse effects were assessed post-stimulation.ResultsForty participants completed the study, with 20 in each group. Paired-sample t-test showed a significant post-stimulation improvement in executive effect performance (t = 2.245; p = 0.037) in the DAN+/DN-tDCS group. The sham group did not exhibit any significant differences in ANT performance. Participants identified the stimulation type with 52.50% accuracy, indicating no difference in blinding efficacy between groups (p = 0.241). ObjectiveThis study investigates the association between convalescent plasma therapy and the negative conversion rate in patients with persistent COVID-19 test positivity.MethodA retrospective analysis was conducted on patients with severe or mild to moderate COVID-19 whose viral nucleic acid tests remained positive for over 30 days. Patients were categorized into two groups: those who administered convalescent plasma therapy and those who were not. Data collected included information on therapy strategies used (convalescent plasma, corticosteroids, interferons, etc.), patients’ demographic characteristics, comorbidities, therapeutic medications, and nucleic acid testing results. Patients in the convalescent plasma therapy group were matched 1:2 ratio with those in the non-convalescent plasma therapy group. Cumulative negative conversion rates on the fifth, tenth, and fifteenth days post-therapy initiation were analyzed as dependent variables. Independent variables included therapy strategies, demographic characteristics, comorbidities, and therapeutic medication usage. Univariate analysis was conducted, and factors with a p-value (P) less than 0.2 were included in a paired Cox proportional hazards model.ResultsThere was no statistically significant difference in the cumulative negative conversion rate between the convalescent plasma therapy group and the non-convalescent plasma therapy group on the fifth, tenth, and fifteenth days. Specifically, on day the fifth, the negative conversion rate was 41.46% in the convalescent plasma therapy group compared to 34.15% in the non-convalescent plasma therapy group (HR: 1.72, 95% CI: 0.82–3.61, P = 0.15). On the tenth day, it was 63.41% in the convalescent plasma therapy group and 63.41% in the non-convalescent plasma therapy group (HR: 1.25, 95% CI: 0.69∼2.26, P = 0.46). On the fifteenth day, the negative conversion rate was 85.37% in the convalescent plasma therapy group and 75.61% in the non-convalescent plasma therapy group (HR: 1.19, 95% CI: 0.71–1.97, P = 0.51).ConclusionOur finding does not support the hypothesis that convalescent plasma therapy could accelerate the time to negative conversion in patients who consistently test positive for COVID-19.

Alport syndrome (AS), a hereditary kidney disease with a high risk for renal failure, is attributed to pathogenic variants in genes COL4A3, COL4A4, and COL4A5 that encode type IV collagen. Next-generation sequencing (NGS) is increasingly applied to the diagnosis of AS, but complex genotype–phenotype correlation, that is, identifying the significance of variants, is still a huge clinical challenge. In this study, we reported the case of a 27-year-old Chinese woman with a family history of hematuria and proteinuria. Notably, the proband is the only one in her family with renal insufficiency. NGS was performed in this family, and it was revealed that the proband was a compound heterozygote for two variants in the COL4A3 gene: c.2990G>A inherited from her father and c.4981C>T inherited from her mother. We modeled the spatial structure of the corresponding protein and assumed that structural abnormalities led to the breakdown of type IV collagen networks, a major component of the glomerular basement membrane. Thus, the proband was diagnosed with autosomal recessive AS, characterized by severe defects of the glomerular basement membrane. Hence, the proband showed a loss of renal function. This case presentation emphasizes the importance of NGS for AS diagnosis and introduces a novel genotype of AS.

The combination of the steel-FRP composite bar (SFCB) and engineered cementitious composite (ECC) holds favorable applications in terms of durable resilient structures. However, a clear understanding of the interfacial bond performance of this novel combination is currently lacking. Therefore, the bond behavior and mechanism of the SFCB-ECC interface were investigated through direct pullout tests. A total of 48 cube specimens were fabricated, with the test variables including bar type, bar diameter, embedment length, and matrix type. The experimental results indicated that the bond failure mode of SFCBs and FRP bars differed from that of deformed steel bars, attributed to the damage of resin-rich ribs in relatively high-strength ECC. The bond-slip curves revealed both the fundamental differences and similarities among steel bars, SFCBs, and FRP bars. The Poisson effect and shear lag effect increased with a decrease in bar stiffness and an increase in diameter. The combined impact of the interaction between the bar stiffness and diameter resulted in varying bond strengths for steel bars, SFCBs, and GFRP bars, despite their similar surface geometries. A theoretical model for the bond strength at the SFCB-ECC interface was derived using the thick-walled cylinder principle and subsequently validated with test data. Finally, a unified empirical model framework for predicting the bond strength of various bar-matrix combinations was proposed based on the theoretical model. The accuracy of this method was evaluated using a collected database.
