Many marine invertebrate larvae undergo complex morphological and physiological changes during the planktonic—benthic transition (a.k.a. metamorphosis). In this study, transcriptome analysis of different developmental stages was used to uncover the molecular mechanisms underpinning larval settlement and metamorphosis of the mussel, Mytilus coruscus. Analysis of highly upregulated differentially expressed genes (DEGs) at the pediveliger stage revealed enrichment of immune-related genes. The results may indicate that larvae co-opt molecules of the immune system to sense and respond to external chemical cues and neuroendocrine signaling pathways forecast and trigger the response. The upregulation of adhesive protein genes linked to byssal thread secretion indicates the anchoring capacity required for larval settlement arises prior to metamorphosis. The results of gene expression support a role for the immune and neuroendocrine systems in mussel metamorphosis and provide the basis for future studies to disentangle gene networks and the biology of this important lifecycle transformation. info:eu-repo/semantics/publishedVersion

Urban road traffic congestion is caused by various factors, such as dynamic and real-time, complexity, and changeability, and existing methods are subjective, have low accuracy and poor real-time, and unable to automatically identify. This study proposes a multicause automatic real-time identification of urban road traffic congestion on the basis of the Bayesian network. First, the method performs systematic mechanism analysis and simulation verification on the relationship between the dynamic observable variables of urban road traffic and the multiple congestion causes to construct the Bayesian network structure. Second, the obtained measured historical data are used for parameter learning and training to obtain the complete Bayesian network model. Finally, multiple causes of traffic congestion can be automatically and simultaneously identified in real time when inputting the observable variables of traffic under road working conditions into the Bayesian network model. This method has high flexibility that can help express the correlation of nodes better and has strong interpretability, which can fully use expert experience and knowledge and achieve automatization in real time. The results in the research on Quanxiu Street, Quanzhou City show that the construction of the multicause automatic real-time identification of urban road traffic congestion based on the Bayesian network is reasonable. Five congestion causes have higher recognition accuracy rates than contrast methods, which include pedestrian influence, peak traffic, parking occupied road, unreasonable signal timing, and impact of traffic crossing the road.

As an important exploration target in the Sichuan Basin, the Lower Triassic Feixianguan Formation has been previously interpreted to be deposited in the trough-platform margin system according to the genetic model of classic lithofacies paleogeography. However, this model does not apply to the northwestern Sichuan Basin. Accordingly, this study proposed a new genetic model that highlighted the carbonate ramp based on the outcrop sections, drilling and logging data, and seismic sections. A large-scale regional regression was found to occur in Sichuan Basin at the end of the Late Permian, leading to the evolution of the original Kaijiang-Liangping Trough into a very shallow tidal flat/lagoon environment at the beginning of the Early Triassic. The sedimentary pattern during the Early Triassic was different from that during the Late Permian. A total of six long-term cycles (LSC1-LSC6) were identified within the Feixianguan Formation. The deposition of LSC1-LSC3 witnessed the filling of the trough to a large extent, and the depositional periods of LSC4 and LSC5 were the main shoal-forming periods. The Feixianguan Formation in the study area was speculated to be deposited in the continuous continental carbonate ramp. With the intensified uplifting of the Longmenshan island chain and the Kangdian ancient land on the northwestern margin of the Upper Yangtze Platform, a large number of terrigenous materials were input into the study area from west to east, leading to the continuous eastward migration of the carbonate facies belt during the deposition of the Fei 1 and Fei 2 Members. Meanwhile, large-area inner-ramp shoal bodies were formed during the deposition of LSC5. It became possible to find a giant gas reservoir belt in the Feixianguan Formation of the northwestern Sichuan Basin and the results of this study provide a new complement to the existing classic trough-platform margin shoal-forming model.

Background/Aims To investigate the beneficial effect of N-Acetylcysteine (NAC) on pancreatic microvascular perfusion in acute necrotizing pancreatitis (ANP). Methods Fifty-four rats were divided into a control group, an ANP group and an NAC-treated group. The ANP model was established by a retrograde injection of 3% sodium taurocholate into the pancreatic duct. The NAC-treated group received an intravenous infusion of NAC just 2 hours before and 30 minutes after the induction of ANP. The pancreatic microvascular perfusion was measured with laser Doppler flowmetry and pancreatic samples were collected for histological examination. To investigate the risk factors for capillary leak syndrome (CLS) in children with malignant hematologic diseases.Thirty children with hematological malignancies complicated with CLS were analyzed with multiple logistic regression analysis.At the test level of 0.05, hypoxemia and septicemia were found to significantly correlate with CLS in these children, and the number of white blood cells before CLS and severe bone marrow suppression were near the test level.Hypoxemia and septicemia are risk factors for CLS in children with malignant hematologic diseases.

Stage III/IV extranodal natural killer/T cell lymphoma (ENKL) has a poor response and poor survival. Given the sensitivity of ENKL to radiotherapy and the fact that there is no consensus on standard chemotherapy, we conducted a clinical trial of LVDP regimen, combining LVDP chemotherapy (containing etoposide, dexamethasone, L-asparaginase, and cisplatin), followed by radiotherapy as a consolidation therapy regimen, for newly diagnosed patients with stage III/IV ENKL to evaluate the efficacy and safety of this regimen. The primary endpoints were overall response rate (ORR) and survival [overall survival (OS) and progression-free survival (PFS)] at 1 or 2 years, while the secondary endpoints were toxicity and adverse effects. In total, 18 patients were enrolled in this trial from July 2010 to September 2013. The mean completed cycles of chemotherapy was 4.04 (range 1-8 cycles), and the ORR was 50 %. During a mean follow-up of 21.8 months (range 2-51 months), the 1-year OS and PFS rates were 72.2 and 50.0 %, respectively, the 2-year OS and PFS rates were 33.3 and 22.2 %, respectively, and the median OS and PFS were 23.0 and 10.5 months, respectively. Severe adverse effects during therapy included six cases of grade 3/4 bone marrow suppression and one case of grade 3 transaminase increase. Sex, eastern cancer oncology group, performance status, Korean Prognostic Index, International Prognostic index, and bone marrow infiltration may influence the prognosis of advanced-stage ENKL.

It is commonly believed that paragangliomas are rare tumors arising from the neural crest-derived chromaffin cells. Although it has been speculated that paraganglioma is related to stem cell origin, there has been lack of direct evidence demonstrating the presence of (neural) stem cells in these tumor tissues. In this study, we found a subgroup of human paraganglioma from ten clinical samples displayed definitive markers of CD133 and/or nestin, the fundamental features of neural stem cell capable of self-renewal and differentiation. A panel of lineage-specific markers was also manifest in some of these tumors, consistent with the hierarchical and heterogeneous nature of these tumors. These observations strongly suggest that at least some forms of paraganglioma maintain tumor stem-like cells (TSCs) that potentially contribute to the histologic complexity of human paraganglioma. Finally, we found that the genomic DNA structure becomes highly unstable in tumor cells of paraganglioma, indicating the loss of tight control of genomic surveillance system be an important transitory event from normal multi-potent tissue stem cells to TSCs.

To study the relationship between ovarian clear cell adenocarcinoma and DNA damage.14 samples were selected from clinical ovarian cases including 3 cases with normal ovarian tissue, 6 cases with poorly differentiated ovarian tumor, 5 cases with ovarian clear cell adenocarcinoma, treated by X-ray irradiation and frozen sections respectively. DNA damage response was analyzed by immunofluorescence and Western blot.Before X-ray irradiation, compared to normal ovarian tissue, a large number of endogenous damage existed in ovarian clear cell adenocarcinoma, and phosphorylation of histone family 2A variant (H2AX) was abnormally enhanced 1 hour after irradiation treatment, however, DNA repair was normal in ovarian clear cell adenocarcinoma. Phosphorylation of H2AX was dispensable for p53 binding protein 1 (53BP1) activation and couldn't be colocalized in clear-type ovarian cancer tissues.The abnormal DNA damage activation implies that the network of DNA damage signaling pathway may be defective.

PURPOSE: The tetraspanin CD151 acts as a promoter of metastasis and invasion in several tumors. However, the role of CD151 in human gastric cancer (HGC) remains unclear. METHODS: Twenty HGC specimens and matched nontumor samples, human gastric epithelial cells (HGEC), and four gastric cancer cell lines were used to analyze CD151 expression. Short hairpin RNA-mediated downregulation of CD151 expression in HGC cells was performed to examine the role of CD151 in the proliferation and metastasis/invasion of HGC cells in vivo and in vitro. The relationship of CD151 with integrin α3 in HGC cells was investigated by silencing integrin α3 followed by co-immunoprecipitation and immunofluorescence staining. Finally, the prognostic value of CD151 and integrin α3 was evaluated by immunohistochemistry in tissue microarrays of 76 HGC patients. RESULTS: CD151 was expressed at higher levels in HGC tissues and HGC cells than in nontumor tissues and HGEC cells. Down-regulation of CD151 by vshRNA-CD151 impaired metastasis and invasion of HGC-27 cells, but did not affect cell proliferation. CD151 formed a complex with integrin α3 in HGC cells. CD151-cDNA transfection rescued the metastatic potential and invasiveness of HGC-27-vshCD151 cells, but not those of HGC-27-vshintegrin α3 cells in vitro.
