Leaf senescence is the final stage of leaf development, where the nutrients and energy of senescent leaves are redistributed to developing tissues or organs for plant growth, reproduction, and defense. Outer leaves are photosynthetic organs that usually senesce at the late heading stage in Chinese cabbage, and premature leaf senescence often reduces leafy head yield and quality. In this study, 11 premature leaf senescence mutants were screened from an ethyl methanesulfonate-mutagenized population of the double haploid line ‘FT’ in Chinese cabbage. At the early heading stage, the mutants exhibited edge yellowing within its outer leaves, and at the mature stage, its leafy head weight decreased significantly. Genetic analysis revealed that the mutated trait of all 11 mutants corresponds to single gene recessive inheritance. Semi-diallel cross tests showed that 5 of the 11 were allelic mutants. MutMap and Kompetitive Allele Specific PCR genotyping revealed that BraA01g001400.3C was the candidate gene, which is orthologous of Arabidopsis SUPPRESSOR OF rps4-RLD 1, encoding an immune regulator, so we named it as BrSRFR1. All the BrSRFR1 in the five allelic mutants exhibited single nucleotide polymorphisms at different positions on their exons and led to premature translation termination, which confirmed that defect in BrSRFR1 led to premature leaf senescence. These results verify the role of BrSRFR1 on leaf senescence and provide a new insight into the mechanisms of leaf senescence in Chinese cabbage, which reveals a novel function of SRFR1 in plant development.

Tripterygium wilfordii Hook F (TWHF) is a traditional Chinese medicine widely used in the treatment of systemic lupus erythematosus (SLE), with triptolide (TP) as its main active ingredient. However, its side effects also induced by TP, especially hepatotoxicity and reproductive toxicity, largely limit its application in a subset of patients. Monoclonal antibodies (mAbs) developed for the treatment of SLE that deplete B cells by targeting B cell-expressing antigens, such as CD19, have failed in clinical trials, partly due to their poor efficacy in consuming B cells. Here, we report the development of a rationally designed antibody‒drug conjugate (ADC), CD19 mAb-TP conjugate, to alleviate the side effects of TWHF and simultaneously improve the therapeutic efficacy of CD19 mAb. The CD19 mAb-TP conjugate, which was named ADC-TP, selectively depleted B cell subsets both in vitro and in vivo and effectively alleviated disease symptoms in mouse lupus models with enhanced therapeutic efficacy than CD19 mAb and fewer side effects than TP. Our present study proposes a CD19 mAb‒TP conjugate strategy to mitigate the toxicity of TWHF while also enhancing the therapeutical efficacy of CD19 mAbs for the treatment of SLE, providing a feasible method for improving the current agents used for treating SLE.

Abstract Systemic lupus erythematosus (SLE) is an autoimmune disorder that commonly affects the skin, kidneys, joints, and various other systemic tissues, with its development intricately linked to the process of immunosenescence. Quercetin (QC), a phytochemical that occurs naturally, demonstrates many different biological capabilities, such as antibacterial, antioxidant, and anti-inflammatory activities. Our investigation found that QC effectively reduced kidney damage and relieved mesenteric lymph nodes (mLNs) swelling in MRL/lpr lupus mice. Moreover, QC has been found to decrease the number of senescent follicular helper T (Tfh) cells, a pivotal kind of T cells that contribute to the progression of SLE. In vitro, QC exhibited the capacity to modulate mRNA expression levels, with the downregulation of IL-6, IL21-AS1, IL-27, BCL6, and BCL2L12, and the upregulation of FOXP1 and BIM. This modulation resulted in the suppression of Tfh cells differentiation and the enhancement of apoptosis in senescent CD4+ T cells. In addition, the HuProtTM Human Proteome Microarray revealed that QC can directly bind to BCL-2 protein and therefore promote the apoptosis of senescent CD4+ T cell. As a result, our investigative elucidate the potent inhibitory action of QC on the ontogeny of Tfh cells, along with its capacity to abrogate the immunosenescent phenotype. This positions QC as a promising therapeutic strategy for treating SLE.

Highlights MIL-88C (Fe) with varying aspect ratios as a precursor was synthesized by regulating oil bath conditions, followed by one-step thermal decomposition to obtain carbon-coated iron-based composites. High-efficiency microwave absorption properties were achieved with RL min value of -67.4 dB (2.13 mm) and wide effective absorption bandwidth (EAB) of 5.52 GHz (1.90 mm) under the low filler loading. A symmetric gradient honeycomb structure was constructed utilizing the high-frequency structure simulator, achieving an EAB of 14.6 GHz and a RL min of -59.0 dB.

Maize plant architecture influences planting density and, in turn, grain yield. Most of the plant architecture-related traits can be described as organ size. We describe a miniature maize mutant, Tiny plant 4 (Tip4), which exhibits reduced size of multiple organs and exhibits a semi-dominant monofactorial inheritance characteristic. Positional cloning confirmed that a 4-bp deletion in the NAC TF with transmembrane motif 1-Like (NTL) gene ZmNTL2, denoted as ZmNTL2Δ, confers the Tip4 mutation. qRT-PCR showed that ZmNTL2 was expressed in all tested tissues. ZmNTL2 functions as a transcriptional activator and is located in both the nucleus and biomembranes. The mutation does not affect the mRNA abundance of ZmNTL2 locus, but it does result in the loss of transmembrane domain and confines the ZmNTL2Δ protein to the nucleus. Knocking out ZmNTL2 has no effect on maize organ size development, indicating that the 4-bp deletion might be a gain-of-function mutation in organ size regulation. Combining transcriptome sequencing with cytokinin and auxin content determination suggests that the decreased organ size may be possibly mediated by changes in hormone homeostasis.

The frequency of heatwaves in the Arctic is on the rise under global warming. These occurrences not only profoundly impact the local ecological environment but also exert remote effects on East Asia and even the global climate. Yet, there exists a noticeable dearth of research focus on Arctic compound daytime-nighttime heatwaves, limiting our comprehension of Arctic climate dynamics. We investigated the occurrence and extinction mechanism for compound daytime-nighttime heatwaves in the Barents–Kara Sea (BKS) during the boreal autumn and explored their association with the sea ice variability. Our results show that a significant dipole pattern appears in the geopotential height two days before the occurrence of compound daytime-nighttime heatwaves in the BKS during autumn, characterized by a negative anomaly centered over Greenland and a positive anomaly centered over the BKS. A robust southerly anomaly in the middle of this dipole pattern facilitates the continuous inflow of warm, moist air from the Atlantic Ocean to the BKS. Both the strong intrusion of moisture and the transport of heat (positive temperature advection) driven by the large-scale atmospheric circulation increase downward latent heat flux, sensible heat flux and net longwave radiation. These factors collectively increase the near-surface temperature over the BKS, ultimately leading to the occurrence of compound daytime-nighttime heatwaves in this region of the Arctic. The extinction of compound daytime-nighttime heatwaves in the BKS is a result of the weakening of the transport of heat and intrusion of water vapor caused by changes in the large-scale circulation. The intrusion of water vapor and the transport of heat significantly reduce the sea ice concentration in most of the BKS. This reduction in sea ice persists for an additional day after the termination of compound daytime-nighttime heatwaves in the BKS. A process of positive atmospheric temperature feedback on a sub-monthly scale may potentially influence the maintenance of compound daytime-nighttime heatwaves in the BKS during the boreal autumn.

Abstract Background The meticulous selection of appropriate vaccine adjuvants is crucial for optimizing immune responses. Traditionally, pemphigus vulgaris (PV), an autoimmune disorder, has been modelled using complete Freund's adjuvant (CFA). In this study, we aimed to discern potential variations in immune responses elicited by Toll‐like receptor (TLR) ligands as compared to CFA. Methods A comprehensive investigation was conducted, comparing the effects of these adjuvants in conjunction with ovalbumin or desmoglein‐3. Flow cytometry was employed to analyse distinct cell subsets, while enzyme‐linked immunosorbent assay quantified antigen‐specific antibodies and cytokine levels. Histological examination of harvested skin tissues and transcriptome analysis of skin lesions were performed to identify differentially expressed genes. Results TLR ligands demonstrated efficacy in inducing PV‐like symptoms in wild‐type mice, in contrast to CFA. This underscored the substantial impact of the adjuvant on self‐antigen tolerance. Furthermore, we proposed an enhanced method for establishing a PV model through adoptive transfer, substituting CFA with TLR ligands. Our results revealed that in contrast to the perception that CFA being the most potent immunopotentiator reported, CFA promoted regulatory T cells (Treg), follicular regulatory T cells and IL‐10‐producing neutrophils, whereas TLR ligands downregulated CCL17 and IL‐10. This suggested potential implications for the recruitment and activation of Treg subsets. While B cell and CD8+ T cell responses exhibited similarity, CFA induced less activation in dendritic cell subsets. A novel mouse model of PV and systemic comparison of immunostimulatory effects of adjuvants were provided by this study. Conclusions The systematic comparison of CFA and TLR ligands shed light on the distinctive properties of these adjuvants, presenting innovative mouse models for the investigation of pemphigus. This study significantly contributes to adjuvant research and advances our understanding of PV pathogenesis. Key points/highlights Immunization with desmoglein 3 and Toll‐like receptor (TLR) ligands effectively induces pemphigus symptoms in wild‐type mice, whereas complete Freund's adjuvant (CFA) fails. TLR ligands heightened the autoreactivity of donor cells in the adoptive transfer pemphigus model. CFA promoted regulatory T cells and IL‐10‐producing neutrophils, whereas TLR ligands downregulated CCL17 and IL‐10, leading to more effective immune responses.

Abstract Triboelectric nanogenerators (TENGs), a promising strategy for harvesting distributed low‐quality power sources, face inevitable bottlenecks regarding long‐term abrasion and poor durability. Herein, both issues are addressed by selecting an earthworm‐inspired self‐replenishing bionic film (ERB) as the tribo‐material of sliding‐freestanding TENGs (SF‐TENGs), it consists of an interconnected 3D porous network structure capable of storing and releasing lubricant under cyclic mechanical stimuli. Thanks to the superiority of self‐replenishing property, there is no need for periodic replenishment and accurate content control of lubricant over the interfacial‐lubricating SF‐TENGs based on dense tribo‐layers. Additionally, an SF‐TENG based on ERB film (ERB‐TENG) demonstrates remarkable output stability with only a slight attenuation of 1% after continuous operation for 100 000 cycles. Moreover, the ERB‐TENG displays a distinguished anti‐wear property, exhibiting no distinct abrasion with an ultra‐low coefficient of friction (0.077) and maintaining output stability over a prolonged period of 35 days. Furthermore, integration with an energy management circuit enables the ERB‐TENG to achieve a 39‐fold boost in charging speed. This work proposes a creative approach to enhance the durability and extend the lifespan of TENG devices, which is also successfully applied to wind energy harvesting and intelligent sports monitoring.

Abstract Noncanonical pyroptosis is triggered by Caspase 4/5/11, which cleaves Gasdermin D (GSDMD), leading to cell lysis. While GSDMD has been studied previously in systemic lupus erythematosus (SLE), the role of pyroptosis in SLE pathogenesis remains unclear and contentious, with limited understanding of Caspase 11‐mediated pyroptosis in this condition. In this study, we explored the level of Caspase 11‐mediated pyroptosis in SLE, identifying both the upstream pathways and the interaction between pyroptosis and adaptive immune responses. We observed increased Caspase 5/11 and GSDMD‐dependent pyroptosis in the macrophages/monocytes of both lupus patients and mice. We identified serum lipopolysaccharide (LPS), released from the gut due to a compromised gut barrier, as the signal that triggers Caspase 11 activation in MRL/lpr mice. We further discovered that pyroptotic macrophages promote the differentiation of mature B cells independently of T cells. Additionally, inhibiting Caspase 11 and preventing LPS leakage proved effective in improving lupus symptoms in MRL/lpr mice. These findings suggest that elevated serum LPS, resulting from a damaged gut barrier, induces Caspase 11/GSDMD‐mediated pyroptosis, which in turn promotes B cell differentiation and enhances autoimmune responses in SLE. Thus, targeting Caspase 11 could be a viable therapeutic strategy for SLE.

Coupling the San Diego gas phase reaction mechanism and the Moss Brookes soot model using FLUENT14.0 software, the effect of adding H2/CO on the fuel side on soot formation in an ethylene/air laminar diffusion flame was studied. A specific analysis was conducted on the effects of H2, CO and its chemical effects on flame temperature, soot volume fraction, mole fractions of important intermediate products OH, H, and C2H2, as well as rate of soot mass nucleation, surface growth, and oxidation. In the numerical calculation, the virtual substances FH2 and FCO are set to separate the chemical effect of H2 and CO and to analyze the chemical effect of adding H2 and CO on soot formation. The results show that the flame temperature increases slightly, and the soot volume fraction decreases monotonically with adding H2 and CO. The chemical effect of H2 increases the temperature, the mole fraction of C2H2 and H, the soot nucleation rate, and the surface growth rate, and finally, it promotes soot formation. The chemical effect of CO increases the temperature and H mole fraction, reduces the OH mole fraction, and then increases the soot surface growth rate and reduces the soot oxidation rate. The higher soot nucleation, surface growth rate, and lower oxidation rate jointly promote soot formation.

Abstract In the three‐phase grid‐connected current‐source inverters (CSIs), the resonance result from the AC‐side CL filter and the quality of the grid‐current waveform under the unbalanced and harmonic grid voltage conditions are two issues deserving attention. To solve the two problems, a continuous control set‐model predictive control (CCS‐MPC) method based on the optimization theory is proposed in the two‐phase synchronous coordinate frame in this paper. The constructed cost function takes grid‐connected current and output current of the inverter as control target, and obtains its optimal solution under the least‐squares method, where the optimal inverter‐side current reference is composed of inductor‐current proportional feedback, capacitor‐voltage proportional feedback, and inverter‐side current steady‐state value feedforward. The proposed method can easily realize the normal operation of three‐phase grid‐connected CSIs because the method can be determined by only one parameter under the unbalanced and distorted grids. Furthermore, in the proposed method, the proportional feedback of capacitor voltage can effectively suppress the CL filter resonance. Finally, simulation and experimental results verify the effectiveness of the proposed control strategy.

IntroductionAlthough it is widely acknowledged that biodiversity maintains plant community assembly processes, exploring the patterns and drivers of beta-diversity (β-diversity; species variation among local plant communities) has received much less attention compared to alpha-diversity (α-diversity; species variation within a local plant community). Here, we aim to examine the patterns and spatial–environmental drivers of taxonomic and phylogenetic β-diversity, and their components such as species turnover and nestedness, in large-scale Leymus chinensis grassland communities.MethodsWe collected plant community data from 166 sites across widely distributed L. chinensis communities in northern China, and then calculated the taxonomic and phylogenetic β-diversity indices (overall, turnover and nestedness) using a pairwise dissimilarity approach. To assess the effects and to explain the variation in the patterns of β-diversity, we collected data on geospatial, climate and soil conditions. We applied descriptive statistics, Mental correlations, and multiple linear regression models to assess the patterns and spatial–environmental drivers of β-diversity.ResultsThe β-turnover, as compared to β-nestedness, exhibited a predominant influence, constituting 92.6% of the taxonomic β-diversity and 80.4% of the phylogenetic β-diversity. Most of the spatial–environmental variables were significantly positively correlated with the overall taxonomic and phylogenetic β-diversity and β-turnover, but not with β-nestedness. Climatic factors such as MAP and MAT were the strongest predictors of both taxonomic and phylogenetic β-diversity and β-turnover. The variance partitioning analysis showed that the combined effects of spatial and environmental factors accounted for 19% and 16% of the variation in the taxonomic and phylogenetic β-diversity (overall), 17% and 12% of the variation in the β-turnover, and 7% and 1% of the variation in the β-nestedness, respectively, which were higher than independent effects of either spatial or environmental factors.DiscussionAt larger spatial scales, the turnover component of β-diversity may be associated with the species complementarity effect, but dominant or functionally important species can vary among communities due to the species selection effect. By incorporating β-diversity into grassland management strategies, we can enhance the provision of vital ecosystem services that bolster human welfare, serving as a resilient barrier against the adverse effects of climate change at regional and global scales.

Abstract Background The Gibberellic Acid-Stimulated Arabidopsis (GASA) gene family is widely involved in the regulation of plant growth, development, and stress response. However, information on the GASA gene family has not been reported in Chinese cabbage (Brassica rapa L. ssp. pekinensis). Results Here, we conducted genome-wide identification and analysis of the GASA genes in Chinese cabbage. In total, 15 GASA genes were identified in the Chinese cabbage genome, and the physicochemical property, subcellular location, and tertiary structure of the corresponding GASA proteins were elucidated. Phylogenetic analysis, conserved motif, and gene structure showed that the GASA proteins were divided into three well-conserved subfamilies. Synteny analysis proposed that the expansion of the GASA genes was influenced mainly by whole-genome duplication (WGD) and transposed duplication (TRD) and that duplication gene pairs were under negative selection. Cis-acting elements of the GASA promoters were involved in plant development, hormonal and stress responses. Expression profile analysis showed that the GASA genes were widely expressed in different tissues of Chinese cabbage, but their expression patterns appeared to diverse. The qRT-PCR analysis of nine GASA genes confirmed that they responded to salt stress, heat stress, and hormonal triggers. Conclusions Overall, this study provides a theoretical basis for further exploring the important role of the GASA gene family in the functional genome of Chinese cabbage.

Abstract Background Perioperative neurocognitive disorders (PND), such as delirium and cognitive impairment, are commonly encountered complications in aged patients. The inhibitory neurotransmitter γ-aminobutyric acid (GABA) is aberrantly synthesized from reactive astrocytes following inflammatory stimulation and is implicated in the pathophysiology of neurodegenerative diseases. Additionally, the activation of NOD-like receptor protein 3 (NLRP3) inflammasome is involved in PND. Herein, we aimed to investigate whether the NLRP3–GABA signaling pathway contributes to the pathogenesis of aging mice’s PND. Methods 24-month-old C57BL/6 and astrocyte-specific NLRP3 knockout male mice were used to establish a PND model via tibial fracture surgery. The monoamine oxidase-B (MAOB) inhibitor selegiline (1 mg/kg) was intraperitoneally administered once a day for 7 days after the surgery. PND, including impulsive-like behaviors and cognitive impairment, was evaluated by open field test, elevated plus maze, and fear conditioning. Thereafter, pathological changes of neurodegeneration were explored by western blot and immunofluorescence assays. Results Selegiline administration significantly ameliorated TF-induced impulsive-like behaviors and reduced excessive GABA production in reactive hippocampal astrocytes. Moreover, astrocyte-specific NLRP3 knockout mice reversed TF-induced impulsive-like and cognitive impairment behaviors, decreased GABA levels in reactive astrocytes, ameliorated NLRP3-associated inflammatory responses during the early stage, and restored neuronal degeneration in the hippocampus. Conclusions Our findings suggest that anesthesia and surgical procedures trigger neuroinflammation and cognitive deficits, which may be due to NLRP3–GABA activation in the hippocampus of aged mice.

This paper mainly discusses the pricing problems of power-barrier option in uncertain financial market. The fluctuation of stock price is regarded as an uncertain process and the interest rate is floating. The uncertain differential equation is invoked to simulate this fluctuation in an uncertain environment. Then, the clear pricing formulas of power-barrier option are given. Finally, the corresponding numerical examples and a real data example are put forward to illustrate the method.

Difenoconazole (DFZ) is a classical triazole fungicide that causes immunosuppression in non-target organisms. Ferulic acid (FA) is a polyphenolic molecule found in nature that has antioxidant and anti-inflammatory activities. The purpose of this investigation was to see if FA could prevent DFZ-induced immunosuppression and to identify the potential mechanisms. Carp were exposed to 1/10 LC50 of DFZ as well as fed normal feed or feed containing dietary additive FA for 30 d. It was found that DFZ-induced immunosuppression could be improved by FA, as evidenced by upregulation of Hb, C3 and IgM and downregulation of LDH. It was then investigated that FA could ameliorate DFZ-induced splenic injury through p53-mediated apoptosis. At the same time, enhancing the levels of CAT, GSH and T-AOC in spleen and transcription levels Nrf2 signaling pathway related genes indicated that FA reduced oxidative damage caused by DFZ by blocking the Nrf2 signaling pathway. In addition, FA inhibited the inflammatory response triggered by TRAF/TAK1/NF-κB signaling pathway, downregulated the transcript levels of pro-inflammatory factors (il-1β, tnf-α, il-6) and the level of NLRP3 inflammasome (NRLP3, ASC, Caspase 1), and upregulated the transcript levels of anti-inflammatory factors (tgf-β1, il-10). In conclusion, the above results suggested that FA mediated TRAF/TAK1/NF-κB, Nrf2, and p53 pathways to attenuate DFZ-induced inflammation, oxidative stress, and apoptosis thereby enhancing the immune capacity of carp.

Flue gas stream generated from combusted natural gas, petroleum, and coal in power plants contains enormous CO2 with detrimental effects on the environment. CO2 capture systems must be integrated into these power plants to prevent emissions of CO2 into the open atmosphere. This work uses rate-based process modelling and a parametric design approach to optimally design large-scale amine-based PCC and compression systems that can be integrated into real 550 MW coal-fired and 555MW NGCC fired power plants. A comparative analysis of energy and economy is also conducted. Based on monoethanolamine (MEA), the process parameters of the post-combustion capture and compression system for CO2 were used as the study baseline. Different amines and alternative flow scheme optimization of the post-combustion capture with compression models are developed for coal and natural gas combined cycle (NGCC) cases and their energy consumption performances compared. Also, the operating and capital costs of the CO2 Capture and Storage (CCS) plant are determined for the overall process of economic evaluation concerning technical and economic performance parameters. The results show that rich solvent cooled recycle (RSR) and lean vapour compression (LVC) process modification under the integration case of CO2 capture and compressed Coal, energy savings of 20.4%, 41.8% and 34.8% for MEA, piperazine activated methyldiethanolamine solution (MDEA+PZ) and piperazine activated monoethanolamine (MEA+PZ) respectively, is possible when compared with the basic situation of MEA. RSR+LVC process modification under the integration case of CO2 capture and compressed NGCC, energy savings of 18.9%, 35.4% and 34.4% for MEA, MDEA+PZ and MEA+PZ, respectively, is possible when compared with the basic situation of MEA. Economic performance: the total annual operating costs of MDEA+PZ, RSR+LVC technological process optimized for the coal case was reduced by approximately 30% compared with the basic situation of MEA, while that for the NGCC case was reduced by 19%. The cost of CO2 avoided for the coal used MDEA+PZ into RSR+LVC technological process optimized capture is decreased by 13% compared with coal captured in MEA basic situation. For the NGCC cases, the cost of CO2 avoided for the NGCC of MDEA+PZ, RSR+LVC process optimized capture is decreased by 7% compared with NGCC with MEA basic case capture.

G protein-coupled receptor 183 (GPR183) has been indicated to mediate the migration and localisation of immune cells in T cell-dependent antibody responses. Systemic lupus erythematosus (SLE) is a canonical autoimmune disease involving B cell-mediated tolerance destruction and excessive pathogenic autoantibody production, in which multiple GPCRs play a role. To date, there has been no systematic study regarding the expression of GPR183 in lymphocyte subsets of SLE patients. In this research, firstly, we observed the expression trends of GRP183 in various T and B cell subsets in human tonsil tissues. These lymphocyte subsets include CD4+, CD8+, naïve T, effector T, Tfh, activated Tfh, Th1, Th2, Th17, Treg, CD19+CD27-, CD19+CD27+, naïve B, germinal centre B, memory B, and plasma cells. Further, compared with healthy controls (HCs), GPR183 expression levels in above peripheral blood lymphocyte subsets of patients with SLE were reduced overall. The differential expression of GPR183 expression between inactive and active SLE patients indicates that GPR183 expression may be concerned with the disease activity of SLE. This was further confirmed through the strong negative correlation with SLEDAI score and positive correlation with serum complement protein C3, C4 and C1q levels. Further receiver operating characteristic (ROC) curve analysis revealed that GPR183 expression in circulating CD27-IgD+ B cells may be beneficial in distinguishing between inactive and active SLE patients. In addition, type I interferon stimulation could down-regulate the expression of GPR183 in peripheral blood T and B cell subsets. Aberrant expression of GPR183 may provide some novel insights into disease activity prediction and underlying pathogenesis of SLE.

Host defense against helminth infection is mediated by mucosal type 2 immunity. Using gain- and loss-of-function mouse models, and mouse intestinal organoids, Xiong et al. show that SIRT6 modulates tuft and goblet cell expansion in intestinal epithelium by activating STAT6 to maintain type 2 mucosal immunity in response to helminth infection.

This paper reports an experimental study on the effects of O2 concentration and CO2 concentration on soot volume fraction and primary particle size of flame centre in ethylene laminar co-diffusion flame. The incandescent light was induced by a two-color band method and laser-excited by wavelength 1064 nm. Previous studies have shown that the oxygen concentration affects the soot volume fraction in the flame. However, the effects of O2 concentration on the primary particle size under CO2 atmosphere are unclear and need to be further discussed. Seven flames were optically detected, in which the oxygen concentration of the accompanying gas varied from 21% to 50% by volume. One primary flame was burned in co-flowing ethylene/air. Six ethylene flames were burned in an oxygen-rich O2/N2 or O2/CO2 atmosphere with an oxygen mole fraction from 30% to 50%. The C2H4 flow rate remained constant in all operating conditions. The results show that the SVF observed by the LII method and spectroscopy in the oxygen-enriched flame are in good agreement. Furthermore, the soot's primary particle diameter measured by the time-resolved LII method is in good agreement with the transmission electron microscope image analysis.With the increase of oxygen concentration, the flame height becomes shorter, the luminosity becomes brighter, and the soot yield increases. Compared with an N2 atmosphere with the same oxygen concentration, the soot yield in a CO2 atmosphere was significantly inhibited. In all the flames studied, the SVF along the flame axis showed a similar distribution: it first increased to the maximum value and then decreased rapidly until the flame tip. The maximum SVF on the axis appears between 0.64 and 0.78 based on the normalized height of the visible flame. In all ethylene/(O2/N2) flames, the maximum primary particle diameter and average primary particle diameter increase with oxygen content. For ethylene/(O2/CO2) flame, the maximum diameter of primary particles decreases with the increased oxygen content. The uncertainty of primary diameter is mainly caused by the thermal adaptation coefficient, the lower LII signal of smaller particles and the uncertainty of local flame temperature.

Abstract Conductive hydrogel has a vital application prospect in flexible electronic fields such as electronic skin and force sensors. Developing conductive hydrogel with significant toughness and high sensitivity is urgently needed for application research. In this work, a strong and sensitive strain sensor based on conductive hydrogel is demonstrated by introducing MXene (Ti3C2Tx) into the micelle crosslinked polyacrylic acid (PAA)/poly(3,4‐ethylenedioxythiophene):poly(styrene‐sulfonate) (PEDOT:PSS) hydrogel network. The functional polymer micelle crosslinkers can dissipate external stress by deformation, endowing the hydrogel with high strength. The combination of MXene both improves the polymer network structure and the conductive pathways, further enhancing the mechanical properties and sensing performance. Resultantly, the flexible strain sensor base on PAA/PEDOT:PSS/MXene conductive hydrogel exhibits excellent sensing performance with a high gauge factor of 20.86, a large strain detection range of 1000%, as well as good adhesion on different interfaces. Thus, it can be used to monitor various movements of the human body and identify all kinds of handwriting, showing great potential into wearable electronics.

Ginkgo biloba L. has a unique evolutionary status. Owing to its high medicinal and ornamental value, ginkgo has also recently become a research hotspot. However, the large genome and long juvenile period, as well as the lack of an effective genetic transformation system, have hindered gaining a full understanding of the comprehensive functions of ginkgo genes. At present, heterologous expression of genes in model plants is the primary method used in ginkgo-related research; however, these distant plant model relatives limit reliable interpretation of the results for direct applications in ginkgo breeding. To overcome these limitations, in this study, an efficient isolation and transient expression system for ginkgo protoplasts was established. A large number of intact and homogeneous ginkgo mesophyll protoplasts were isolated using 2% cellulase and 0.25% pectinase in 0.4 M mannitol. The activity of these protoplasts remained above 90% even after 24 h. Furthermore, when the concentration of the polyethylene glycol 4000 solution was 30%–40% (w/v), the transformation efficiency of the protoplasts reached 40%. Finally, the reliability of the system was verified using subcellular localization, transient overexpression, and protein interaction experiments with ginkgo genes, thereby providing a technical platform for the identification and analysis of ginkgo gene functions. The proposed method partially compensates for the limitations associated with the lack of a genetic transformation system and provides technical support to expand research on elucidating the functions of ginkgo genes.

Flavonoids are important secondary metabolites with extensive pharmacological functions. Ginkgo biloba L. (ginkgo) has attracted extensive attention because of its high flavonoid medicinal value. However, little is understood about ginkgo flavonol biosynthesis. Herein, we cloned the full-length gingko GbFLSa gene (1314 bp), which encodes a 363 amino acid protein that has a typical 2-oxoglutarate (2OG)-Fe(II) oxygenase region. Recombinant GbFLSa protein with a molecular mass of 41 kDa was expressed in Escherichia coli BL21(DE3). The protein was localized to the cytoplasm. Moreover, proanthocyanins, including catechin, epicatechin, epigallocatechin and gallocatechin, were significantly less abundant in transgenic poplar than in nontransgenic (CK) plants. In addition, dihydroflavonol 4-reductase, anthocyanidin synthase and leucoanthocyanidin reductase expression levels were significantly lower than those of their CK counterparts. GbFLSa thus encodes a functional protein that might negatively regulate proanthocyanin biosynthesis. This study helps elucidate the role of GbFLSa in plant metabolism and the potential molecular mechanism of flavonoid biosynthesis.

IntroductionNeutrophil extracellular trap (NET) is a novel defense strategy of neutrophils and found to be induced by Porphyromonas gingivalis (P. gingivalis) lipopolysaccharide (LPS) or high glucose. The aim of this study was to investigate the roles and mechanisms of NET formation in high glucose inflammatory microenvironment.MethodsNETs induced by 1 μg/ml P. gingivalis LPS and/or 25 mM glucose were visualized using a fluorescence microscopy and the levels of extracellular DNA were determined by a microplate reader. The bactericidal efficiency of NETs was assessed by quantifying the survival P. gingivalis in neutrophils. The levels of NLRP3 and IL-1β in THP-1 derived-macrophages, and the expressions of p-PKC βII, p-MEK1/2, p-ERK1/2, ORAI1 and ORAI2 in neutrophils were detected by Western blot. Moreover, levels of intracellular Ca2+ and reactive oxygen species (ROS) in neutrophils were explored by flow cytometry.ResultsP. gingivalis LPS enhanced the formation of NETs and increased the levels of extracellular DNA in high glucose microenvironment (p < 0.05). Compared with normal glucose inflammatory microenvironment, quantities of extra- and intracellular viable P. gingivalis in neutrophils exposed to NETs induced in high glucose inflammatory one were increased (p < 0.05) and the expressions of NLRP3 and IL-1β were dramatically increased in macrophages co-cultured with NETs from high glucose inflammatory microenvironment (p < 0.05). In addition, levels of ROS, intracellular Ca2+, p-PKC βII, p-MEK1/2, p-ERK1/2, ORAI1 and ORAI2 were increased in neutrophils stimulated with both high glucose and P. gingivalis LPS compared with the single stimulus groups (p < 0.05).DiscussionIn high glucose inflammatory microenvironment, formation of NETs was enhanced via oxidative stress, which failed to reverse the decreased bactericidal capacity in high glucose microenvironment, and instead aggravated the subsequent inflammatory responses.

BackgroundThe incidence of magnet ingestion by children has recently increased in China. Magnet ingestion is associated with an extremely high risk of gastrointestinal damage because loops of bowel can become trapped and squeezed between multiple magnets in different locations. However, the lack of imaging sensitivity makes clinical decision-making difficult.ObjectiveThis study was conducted to investigate the performance of ultrasound in diagnosing gastrointestinal magnet ingestion in children.MethodsFrom April 2017 to February 2021, all children with a history of magnet ingestion or a diagnosis of gastrointestinal magnet as shown by x-ray or ultrasound in our hospital were included as study candidates. Patients who were lost to follow-up or had known malformations of the gastrointestinal tract were excluded. Eligible patients were those with surgical or endoscopic confirmation of gastrointestinal magnet, those who passed the magnet out of the alimentary tract without assistance, and those with confirmed absence of the magnet on abdominal x-ray examination after 1 month of conservative treatment. All eligible patients' ultrasound and x-ray examination data were evaluated. The sensitivity, specificity, and area under the curve (AUC) of ultrasound was calculated for diagnosing magnet ingestion, locating the magnet (stomach, small intestine, or colon), and confirming the phenomenon of wall entrapment.ResultsOf 112 patients, 107 had a magnetic foreign body and 5 did not. Magnets were correctly detected by ultrasound in 97 patients, with an observed sensitivity of 90.65% and specificity of 100%. Satisfactory sensitivity was obtained for ultrasound localization of gastric magnets (96.30%) and small intestinal magnets (100.00%), but sensitivity for ultrasound localization of colonic magnets was relatively poor (73.33%). The discrimination of wall entrapment by ultrasound was good (AUC = 0.93), with an observed sensitivity and specificity of 92.00% and 93.62%, respectively.ConclusionsUltrasound can be used to locate gastrointestinal magnets (in the stomach, small intestine, or colon) with good clinical efficacy in identifying wall entrapment.

Soil microbiomes are characterized by their composition and networks, which are linked to soil nitrogen (N) availability. In nature, inorganic N dominates at one end and organic N dominates at the other end along soil N gradients; however, little is known about how this shift influences soil microbiome composition and co-occurrence networks, as well as their controls. To this end, we conducted an experiment with the host plant Solidago canadensis, which was subject to three N regimes: inorganic N-dominated, co-dominated by inorganic and organic N (CIO), and organic N-dominated. Organic N dominance exhibited stronger effects on the composition and co-occurrence networks of soil microbiomes than inorganic N dominance. The predominant control was plant traits for bacterial and fungal richness, and soil pH for keystone species. Relative to the CIO regime, inorganic N dominance did not affect fungal richness and increased keystone species; organic N dominance decreased fungal richness and keystone species. Pathogenic fungi and arbuscular mycorrhizal fungi were suppressed by organic N dominance but not by inorganic N dominance. These findings suggest that the shift from soil inorganic N-dominance to soil organic N-dominance could strongly shape soil microbiome composition and co-occurrence networks by altering species diversity and topological properties.

IntroductionAlthough the relationships between species diversity and aboveground biomass (AGB) are highly debated in grassland ecosystems, it is not well understood how climatic factors influence AGB directly and indirectly via plant coverage and species diversity in large-scale grasslands along a topographic gradient. In doing so, we hypothesized that climatic factors would regulate plant coverage, species diversity and AGB due to maintaining plant metabolic and ecological processes, but the relationship of plant coverage with AGB would be stronger than species diversity due to covering physical niche space. MethodsTo test the proposed hypothesis, we collected data for calculations of species richness, evenness, plant coverage and AGB across 123 grassland sites (i.e., the mean of 3 plots in each site) dominated by Leymus chinensis in northern China. We used a structural equation model for linking the direct and indirect effects of topographic slope, mean annual precipitation and temperature on AGB via plant coverage, species richness, and evenness through multiple complex pathways. ResultsWe found that plant coverage increased AGB, but species evenness declined AGB better than species richness. Topographic slope influenced AGB directly but not indirectly via plant coverage and species diversity, whereas temperature and precipitation increased with increasing topographic slope. Regarding opposing mechanisms, on the one hand, precipitation increased AGB directly and indirectly via plant coverage as compared to species richness and evenness. On the other hand, temperature declined AGB indirectly via plant coverage but increased via species evenness as compared to species richness, whereas the direct effect was negligible. DiscussionOur results show that niche complementarity and selection effects are jointly regulating AGB, but these processes are dependent on climatic factors. Plant coverage promoted the coexistence of species but depended greatly on precipitation and temperature. Our results highlight that precipitation and temperature are two key climatic drivers of species richness, evenness, plant coverage and AGB through complex direct and indirect pathways. Our study suggests that grasslands are sensitive to climate change, i.e., a decline in water availability and an increase in atmospheric heat. We argue that temperature and precipitation should be considered in grassland management for higher productivity in the context of both plant coverage and species diversity which underpin animals and human well-being.

Deep vein thrombosis is a common clinical peripheral vascular disease that occurs frequently in orthopaedic patients and may lead to pulmonary embolism (PE) if the thrombus is dislodged. pulmonary embolism can be prevented by placing an inferior vena cava filter (IVCF) to intercept the dislodged thrombus. Thus, IVCFs play an important role in orthopaedics. However, the occurrence of complications after inferior vena cava filter placement, particularly recurrent thromboembolism, makes it necessary to carefully assess the risk-benefit of filter placement. There is no accepted statement as to whether IVCF should be placed in orthopaedic patients. Based on the problems currently displayed in the use of IVCFs, an ideal IVCF is proposed that does not affect the vessel wall and haemodynamics and intercepts thrombi well. The biodegradable filters that currently exist come close to the description of an ideal filter that can reduce the occurrence of various complications. Currently available biodegradable IVCFs consist of various organic polymeric materials. Biodegradable metals have shown good performance in making biodegradable IVCFs. However, among the available experimental studies on degradable filters, there are no experimental studies on filters made of degradable metals. This article reviews the use of IVCFs in orthopaedics, the current status of filters and the progress of research into biodegradable vena cava filters and suggests possible future developments based on the published literature by an electronic search of PubMed and Medline databases for articles related to IVCFs searchable by October 2022 and a manual search for citations to relevant studies.

Frostbite in Southwestern China has been overlooked due to its low incidence, relatively mild temperature and lack of literature published before. However, it needs to be further studied for religious diversity and distinct geomorphology. In this article, we reported an 18-year-old Tibetan girl who suffered from blizzard attack during pilgrimage. Her feet and several fingers showed mummified gangrene upon physical examination with poor movement. She was diagnosed with 3rd to 4th degree of frostbite. The girl was given oral ibuprofen, debridement and other regular treatment daily, but she was eventually amputated due to insufficient thrombolytic management in primary hospital, delayed informing consent in the referral hospital and ethnic conflict between religion and guidelines. This case enriched the experience of managing complex frostbite in Tibetan population and alarms that efforts should be integrated to protect pilgrims and mountaineers in the Tibetan region.

The NAC (NAM-ATAF1/2-CUC) transcription factor family is one of the largest plant-specific transcription factor families, playing an important role in plant growth and development and abiotic stress response. As a short-rotation woody plant, Salix integra (S. integra) has high lead (Pb) phytoremediation potential. To understand the role of NAC in S. integra Pb tolerance, 53 SiNAC transcripts were identified using third-generation and next-generation transcriptomic data from S. integra exposed to Pb stress, and a phylogenetic analysis revealed 11 subfamilies. A sequence alignment showed that multiple subfamilies represented by TIP and ATAF had a gene that produced more than one transcript under Pb stress, and different transcripts had different responses to Pb. By analyzing the expression profiles of SiNACs at 9 Pb stress time points, 41 of 53 SiNACs were found to be significantly responsive to Pb. Short time-series expression miner (STEM) analysis revealed that 41 SiNACs had two significant Pb positive response patterns (early and late), both containing 10 SiNACs. The SiNACs with the most significant Pb response were mainly from the ATAF and NAP subfamilies. Therefore, 4 and 3 SiNACs from the ATAF and NAP subfamilies, respectively, were selected as candidate Pb-responsive SiNACs for further structural and functional analysis. The RT-qPCR results of 7 transcripts also confirmed the different Pb response patterns of the ATAF and NAP subfamilies. SiNAC004 and SiNAC120, which were randomly selected from two subfamilies, were confirmed to be nuclear localization proteins by subcellular localization experiments. Functional prediction analysis of the associated transcripts of seven candidate SiNACs showed that the target pathways of ATAF subfamily SiNACs were “sulfur metabolism” and “glutathione metabolism”, and the target pathways of NAP subfamily SiNACs were “ribosome” and “phenylpropanoid biosynthesis”. This study not only identified two NAC subfamilies with different Pb response patterns but also identified Pb-responsive SiNACs that could provide a basis for subsequent gene function verification.

The Malus crabapple is an important woody ornamental plant. The fading of petals during its development significantly affects their ornamental value. Petal color is related to anthocyanin content and miRNAs play an important role in the post-transcriptional regulation of anthocyanin synthesis. However, the mechanisms underlying miRNA regulation of petal fading have rarely been studied. Transcriptome and small RNA sequencing of petals from the blooming phases of Malus. ‘Indian Summer’ varieties S1 (small bud), S2 (initial-flowering), and S3 (late-flowering) allowed us to identify 230 known miRNAs and 17 novel miRNAs, including 52 differentially expressed miRNAs which targeted 494 genes and formed 823 miRNA–target pairs. Based on the target gene annotation results, miRNA–target pairs were screened that may be involved in the fading process of Malus crabapple petals through three different pathways: anthocyanin synthesis, transport, and degradation, involving mcr-miR858-MYB1\MYB5 and mcr-miR396-McCHI inhibiting anthocyanin synthesis; mcr-miR167, mcr-miR390, mcr-miR535, and mcr-miR858 inhibiting anthocyanin transport from the cytoplasm to the vacuole by targeting ABC transporter genes (ABCB, ABCC, ABCD, and ABCG); and mcr-miR398 targeting the superoxide dismutase genes (CZSOD2 and CCS) to accelerate anthocyanin degradation. These findings offer a novel approach to understanding the mechanism of petal fading and serve as a reference for other plants with floral fading.

Abstract Alcohol-related liver disease (ALD), a condition caused by alcohol overconsumption, occurs in three stages of liver injury including steatosis, hepatitis, and cirrhosis. DEP domain-containing protein 5 (DEPDC5), a component of GAP activities towards Rags 1 (GATOR1) complex, is a repressor of amino acid-sensing branch of the mammalian target of rapamycin complex 1 (mTORC1) pathway. In the current study, we found that aberrant activation of mTORC1 was likely attributed to the reduction of DEPDC5 in the livers of ethanol-fed mice or ALD patients. To further define the in vivo role of DEPDC5 in ALD development, we generated Depdc5 hepatocyte-specific knockout mouse model (Depdc5-LKO) in which mTORC1 pathway was constitutively activated through loss of the inhibitory effect of GATOR1. Hepatic Depdc5 ablation leads to mild hepatomegaly and liver injury and protects against diet-induced liver steatosis. In contrast, ethanol-fed Depdc5-LKO mice developed severe hepatic steatosis and inflammation. Pharmacological intervention with Torin 1 suppressed mTORC1 activity and remarkably ameliorated ethanol-induced hepatic steatosis and inflammation in both control and Depdc5-LKO mice. The pathological effect of sustained mTORC1 activity in ALD may be attributed to the suppression of peroxisome proliferator activated receptor α (PPARα), the master regulator of fatty acid oxidation in hepatocytes, because fenofibrate (PPARα agonist) treatment reverses ethanol-induced liver steatosis and inflammation in Depdc5-LKO mice. These findings provide novel insights into the in vivo role of hepatic DEPDC5 in the development of ALD.

Abstract Background Extracellular vesicles (EVs) derived from tumor cells are implicated in the progression of malignancies through the transfer of molecular cargo microRNAs (miRNAs or miRs). We aimed to explore the role of EVs derived from breast cancer cells carrying miR-182-5p in the occurrence and development of breast cancer. Methods Differentially expressed miRNAs and their downstream target genes related to breast cancer were screened through GEO and TCGA databases. miR-182-5p expression was examined in cancer tissues and adjacent normal tissues from patients with breast cancer. EVs were isolated from breast cancer cell line MDA-MB-231 cells and identified. The gain- and loss-of function approaches of miR-182-5p and CKLF-like MARVEL transmembrane domain-containing 7 (CMTM7) were performed in MDA-MB-231 cells and the isolated EVs. Human umbilical vein endothelial cells (HUVECs) were subjected to co-culture with MDA-MB-231 cell-derived EVs and biological behaviors were detected by CCK-8 assay, flow cytometry, immunohistochemical staining, Transwell assay and vessel-like tube formation in vitro. A xenograft mouse model in nude mice was established to observe the tumorigenesis and metastasis of breast cancer cells in vivo. Results miR-182-5p was highly expressed in breast cancer tissues and cells, and this high expression was associated with poor prognosis of breast cancer patients. miR-182-5p overexpression was shown to promote tumor angiogenesis in breast cancer. Moreover, our data indicated that miR-182-5p was highly enriched in EVs from MDA-MD-231 cells and then ultimately enhanced the proliferation, migration, and angiogenesis of HUVECs in vitro and in vivo. Moreover, we found that CMTM7 is a target of miR-182-5p. EVs-miR-182-5p promotes tumorigenesis and metastasis of breast cancer cells by regulating the CMTM7/EGFR/AKT signaling axis. Conclusions Taken altogether, our findings demonstrates that EVs secreted by breast cancer cells could carry miR-182-5p to aggravate breast cancer through downregulating CMTM7 expression and activating the EGFR/AKT signaling pathway.

Abstract In recent years, flexible stress sensors capable of monitoring diverse body movements and physiological signals have been attracting great attention in the fields of healthcare systems, human–machine interfaces, and wearable electronics. Inspired by the structure of natural eggshell inner membrane (ESIM), we developed a pressure sensor based on MXene (Ti3C2Tx)/Ag NWs (silver nanowires) composite electrodes and the micro-structured dielectric layer to meet the application requirements of wide detection range and long-term stability for the sensors. In the light of the nanoscale-microarray of the dielectric layer and the rough surface of electrode materials, this pressure sensor is expected to allow great and persistent deformation during the loading process. As a result, the device is characterized by an improved sensitivity, fast response (in the millisecond range), wide detection range (0–600 kPa), and long-term stability. The outstanding performance of the proposed sensor makes it possible to detect various human activities, such as speaking, air blowing, clenching, walking, finger/knee/elbow bending, and striking, demonstrating its good application prospects in wearable and flexible electronic devices.

PurposeTo assess the prevalence of refractive errors (REs) in school children aged 6–18 years in urban and rural settings in Dalian, Northeast of China.MethodsThis is a school-based cross-sectional survey using multi-stage randomization technique. Six- to eighteen-year-old school children from elementary schools, junior and senior high schools from a rural area and an urban area in Dalian were included in December 2018. All subjects underwent a comprehensive questionnaire and eye examination.ResultsA total of 4,522 school children with 6–18 years of age were investigated. The age, gender-adjusted prevalence of myopia, and anisometropia were 82.71 and 7.27% among the urban students as compared to 71.76% and 5.41% among the rural ones (OR = 1.80, 95 % CI = 1.53 - 2.11, P < 0.001; OR = 1.29, 95 % CI = 1.00–1.67, P = 0.049), respectively. The hyperopia was less common in urban students than in rural ones (5.63 vs. 10.21%; OR = 0.54, 95 % CI: 0.43–0.67, P < 0.001). However, there was no significant difference in prevalence of astigmatism between urban (46.07%) and rural (44.69%) participants (OR = 0.96, 95 % CI: 0.84–1.10, P = 0.559). The differences on prevalence of REs were attributed to different social-demographic and physiologic factors.ConclusionsThe students from urban settings are more likely to have myopia and anisometropia but less likely to have hyperopia than their rural counterparts. Although considerable attention had been paid to controlling REs, it is necessary to further consider the urban-rural differences in REs.

Abstract Background Micronaire is a comprehensive index reflecting the fineness and maturity of cotton fiber. Micronaire is one of the important internal quality indicators of the cotton fiber and is closely related to the value of the cotton fiber. Understanding the genetic basis of micronaire is required for the genetic improvement of the trait. However, the genetic architecture of micronaire at the genomic level is unclear. The present genome-wide association study (GWAS) aimed to identify the genetic mechanism of the micronaire trait in 83 representative upland cotton lines grown in multiple environments. Results GWAS of micronaire used 83 upland cotton accessions assayed by a Cotton 63 K Illumina Infinium single nucleotide polymorphism (SNP) array. A total of 11 quantitative trait loci (QTLs) for micronaire were detected on 10 chromosomes. These 11 QTLs included 27 identified genes with specific expression patterns. A novel QTL, qFM-A12–1, included 12 significant SNPs, and GhFLA9 was identified as a candidate gene based on haplotype block analysis and on strong and direct linkage disequilibrium between the significantly related SNPs and gene. GhFLA9 was expressed at a high level during secondary wall thickening at 20∼25 days post-anthesis. The expression level of GhFLA9 was significantly higher in the low micronaire line (Msco-12) than that in the high micronaire line (Chuangyou-9). Conclusions This study provides a genetic reference for genetic improvement of cotton fiber micronaire and a foundation for verification of the functions of GhFLA9.

Leaf flattening plays a vital role in the establishment of plant architecture, which is closely related to plant photosynthesis and, thus, influences the product yield and quality of Chinese cabbage. In this study, we used the doubled haploid line ‘FT’ of Chinese cabbage as the wild type for ethyl methanesulfonate (EMS) mutagenesis and obtained a mutant cwm with stably inherited compact and wrinkled leaves. Genetic analysis revealed that the mutated trait was controlled by a single recessive nuclear gene, Brcwm. Brcwm was preliminarily mapped to chromosome A07 based on bulked segregant RNA sequencing (BSR-seq) and fine-mapped to a 205.66 kb region containing 39 genes between Indel12 and Indel21 using SSR and Indel analysis. According to the whole-genome re-sequencing results, we found that there was only one nonsynonymous single nucleotide polymorphism (SNP) (C to T) within the target interval on exon 4 of BraA07g021970.3C, which resulted in a proline to serine amino acid substitution. The mutated trait co-segregated with the SNP. Quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) revealed that BraA07g021970.3C expression was dramatically higher in ‘FT’ leaves than that in cwm leaves. BraA07g021970.3C is homologous to AT3G55000 encoding a protein related to cortical microtubule organization. A similar phenotype of dwarfism and wrinkled leaves was observed in the recessive homozygous mutant cwm-f1 of AT3G55000, and its T3 transgenic lines were restored to the Arabidopsis wild-type phenotype through ectopic overexpression of BraA07g021970.3C. These results verified that BraA07g021970.3C was the target gene essential for leaf flattening in Chinese cabbage.

Recent years have seen increasing school safety events along with growing numbers of students in China. In this paper, more than 400 serious school safety events between 2000 and 2018 in China were collected. The causes and characteristics of these events, taking into account the occurrence years, months, regions, education stages and types, were investigated. The results indicate that the number of school safety events has generally increased annually from 2000 to 2018 and can significantly vary each month, showing a higher frequency of occurrence during the “First Semester” (generally from September to December in China). Moreover, spatial distribution of school safety events is normally related to regional economic development; it was found that Guangdong, Jiangsu and Shandong is a first-level occurrence hotspots, followed by Zhejiang, Henan, Hebei and Sichuan, which are secondary occurrence hotspots. Furthermore, statistical analysis shows that the number of school safety events that occurred in kindergartens, primary schools and middle schools are approximately equal (around 1/3). Finally, it was found that the school safety events caused by “Accident” (such as school bus accidents, school fires, crowded stampedes and the collapses of school buildings) occupy a large proportion (57%).

Breast cancer (BC) has a high morbidity and mortality rate. It is imperative to explore the pathogenesis of BC in order to find potential prognostic biomarkers and therapeutic targets. This study screened and verified the differential expression of circ_0001142 in BC tissues and cell lines through bioinformatics and qRT-PCR. Perform dual luciferase reporter gene assay and pull-down detection to verify the correlation between circ_0001142 and miRNA-361–3p and between miR-361–3p and PIK3CB. QRT-PCR, flow cytometry and ELISA were used to study the regulatory effects and regulatory mechanisms of different treatment groups on macrophage polarization. The role of exosomes circ_0001142 in the tumor microenvironment and its influence on BC growth, metastasis and macrophage polarization were investigated through in vivo and in vitro studies. We first found that circ_0001142 is highly expressed in BC. In addition, ERs promote the secretion of tumor exosomes, enhance the entry of circ_0001142 into macrophages and interfere with the process of autophagy and polarization. Finally, it was found that the circ_0001142/miR-361–3p/PIK3CB pathway plays an important role in the polarization of macrophages.

Cotton is one of the most economically important crops worldwide. Seed size is a vital trait for plants connected with yield and germination. GW2 encodes a RING_Ubox E3 ubiquitin ligase that controls seed development by affecting cell growth. Here, are few reports on GW2-like genes in cotton, and the function of GW2 in cotton is poorly understood. In the present study, a genome-wide analysis identified 6 and 3 GW2-like genes in each of the two cultivated tetraploids (Gossypium hirsutum and G. barbadense) and each of their diploid ancestral species (G. arboreum, G. raimondii), respectively. GhGW2-2D has the same functional domain and high sequence similarity with AtDA2 in Arabidopsis. Overexpression of GhGW2-2D in Arabidopsis significantly reduced seed and seedling size, suggesting GhGW2-2D is a potential target for regulating cotton seed size. These results provided information on the genetic and molecular basis of GW2-like genes in cotton, thus establishing a foundation for functional studies of cotton seeds.

ObjectiveTo study the safety and feasibility of transurethral bladder lesion mucosal en bloc resection with 2 μm laser for cystitis glandularis.MethodsFrom July 2018 to July 2019, 58 patients with cystitis glandularis received surgical treatment were selected. All patients in this study were randomly divided into experimental group (transurethral 2 μm laser bladder lesion mucosal en bloc resection) and control group (traditional transurethral resection of bladder lesion mucosal). By analyzing the perioperative and follow-up clinical data of these two operation procedures, we discuss the efficacy and safety of transurethral 2 μm laser bladder lesion mucosal en bloc resection in the treatment of cystitis glandularis.ResultsPatients of two groups received operation successfully without serious complications such as bladder perforation. Compared with the experimental group, the laser treated group hadless bleeding in operation, shorter post-operative catheter indwelling time. These differences were statistically significant. No significant difference existed between two groups in terms of operative time, Bladder flushing time, irritation symptoms of bladder.ConclusionTransurethral 2 μm laser bladder lesion mucosal en bloc resection is safe and effective for the treatment of cystitis glandularis, and it is worthy of further clinical application.

Purpose: The objective of this study is to evaluate the in vivo corneal biomechanical response to three laser refractive surgeries.Methods: Two hundred and twenty-seven patients who submitted to transepithelial photorefractive keratectomy (tPRK), femtosecond laser-assisted in-situ keratomileusis (FS-LASIK), or small-incision lenticule extraction (SMILE) were included in this study. All cases were examined with the Corvis ST preoperatively (up to 3 months) and postoperatively at 1, 3, and 6 months, and the differences in the main device parameters were assessed. The three groups were matched in age, gender ratio, corneal thickness, refractive error corrections, optical zone diameter, and intraocular pressure. Seed size and shape are key agronomic traits affecting seedcotton yield and seed quality in cotton (Gossypium spp.). However, the genetic mechanisms that regulate the seed physical traits in cotton are largely unknown. In this study, an interspecific backcross inbred line (BIL) population of 250 BC1F7 lines, derived from the recurrent parent Upland CRI36 (Gossypium hirsutum) and Hai7124 (Gossypium barbadense), was used to investigate the genetic basis of cotton seed physical traits via quantitative trait locus (QTL) mapping and candidate gene identification. The BILs were tested in five environments, measuring eight seed size and shape-related traits, including 100-kernel weight, kernel length width and their ratio, kernel area, kernel girth, kernel diameter, and kernel roundness. Based on 7,709 single nucleotide polymorphic (SNP) markers, a total of 49 QTLs were detected and each explained 2.91–35.01% of the phenotypic variation, including nine stable QTLs mapped in at least three environments. Based on pathway enrichment, gene annotation, genome sequence, and expression analysis, five genes encoding starch synthase 4, transcription factor PIF7 and MYC4, ubiquitin-conjugating enzyme E27, and THO complex subunit 4A were identified as candidate genes that might be associated with seed size and shape. Our research provides valuable information to improve seed physical traits in cotton breeding.

Abstract No current in vitro tumor model replicates a tumor’s in vivo microenvironment. A culturing technique that better preserves a tumor’s pathophysiological conditions is needed for some important clinical applications, including personalized drug-sensitivity/resistance assays. In this study, we utilized autologous serum or body fluid to build a 3D scaffold and grow a patient’s tumor. We named this technique “3D-ACM” (autologous culture method). Forty-five clinical samples from biopsies, surgically removed tumor tissues and malignant body fluids were cultured with 3D-ACM. Traditional 3D-FBS (fetal bovine serum) cultures were performed side-by-side for comparison. The results were that cells cultured in 3D-ACM rebuilt tissue-like structures, and retained their immuno-phenotypes and cytokine productions. In contrast, the 3D-FBS method promoted mesenchymal cell proliferation. In preliminary chemo drug-sensitivity assays, significantly higher mortality was always associated with FBS-cultured cells. Accordingly, 3D-ACM appears to more reliably preserve a tumor’s biological characteristics, which might improve the accuracy of drug-testing for personalized cancer treatment.

MADS-box genes encode transcription factors that affect plant growth and development. Camellia chekiangoleosa is an oil tree species with ornamental value, but there have been few molecular biological studies on the developmental regulation of this species. To explore their possible role in C. chekiangoleosa and lay a foundation for subsequent research, 89 MADS-box genes were identified across the whole genome of C. chekiangoleosa for the first time. These genes were present on all the chromosomes and were found to have expanded by tandem duplication and fragment duplication. Based on the results of a phylogenetic analysis, the 89 MADS-box genes could be divided into either type I (38) or type II (51). Both the number and proportion of the type II genes were significantly greater than those of Camellia sinensis and Arabidopsis thaliana, indicating that C. chekiangoleosa type II genes experienced a higher duplication rate or a lower loss rate. The results of both a sequence alignment and a conserved motif analysis suggest that the type II genes are more conserved, meaning that they may have originated and differentiated earlier than the type I genes did. At the same time, the presence of extra-long amino acid sequences may be an important feature of C. chekiangoleosa. Gene structure analysis revealed the number of introns of MADS-box genes: twenty-one type I genes had no introns, and 13 type I genes contained only 1~2 introns. The type II genes have far more introns and longer introns than the type I genes do. Some MIKCC genes have super large introns (≥15 kb), which are rare in other species. The super large introns of these MIKCC genes may indicate richer gene expression. Moreover, the results of a qPCR expression analysis of the roots, flowers, leaves and seeds of C. chekiangoleosa showed that the MADS-box genes were expressed in all those tissues. Overall, compared with that of the type I genes, the expression of the type II genes was significantly higher. The CchMADS31 and CchMADS58 genes (type II) were highly expressed specifically in the flowers, which may in turn regulate the size of the flower meristem and petals. CchMADS55 was expressed specifically in the seeds, which might affect seed development. This study provides additional information for the functional characterization of the MADS-box gene family and lays an important foundation for in-depth study of related genes, such as those involved in the development of the reproductive organs of C. chekiangoleosa.

The writing performance of the easy-cone magnetic tunnel junction (MTJ) and perpendicularly magnetized MTJ (pMTJ) under various temperatures was investigated based on the macrospin model. When the temperature is changed from 273 K to 373 K, the switching current density of the pMTJ changes by 56%, whereas this value is only 8% in the easy-cone MTJ. Similarly, the temperature-induced variation of the switching delay is more significant in the pMTJ. This indicates that the easy-cone MTJ has a more stable writing performance under temperature variations, resulting in a wider operating temperature range. In addition, these two types of MTJs exhibit opposite temperature dependence in the current overdrive and write error rate. In the easy cone MTJ, these two performance metrics will reduce as temperature is increased. The results shown in this work demonstrate that the easy-cone MTJ is more suitable to work at high temperatures compared with the pMTJ. Our work provides a guidance for the design of STT-MRAM that is required to operate at high temperatures.

The root system is the only vital organ for plants to connect with soil moisture and nutrients and obtain feedback information. This research aimed to explore the response of different spike type winter wheat varieties to plant and row spacing configurations. Multi-spike and large-spike winter wheat varieties were used as materials. By setting different plant row configurations and planting densities, the spatial and temporal distribution of root length density, root diameter, root dry weight density, and the main control factors of root growth and development of winter wheat during the whole growth period were studied. The results showed that the root system was the most widely distributed and the root diameter was the largest in the 0–40 cm soil depth, with an average root system diameter of more than 0.5 mm. The root length density and root diameter peaked at the heading stage, decreased at the maturity stage, and the root dry weight density peaked at the jointing stage. The jointing stage and heading stage are the most vigorous periods of root growth in winter wheat, when the center of gravity of root growth in winter wheat is gradually moving down. Therefore, the rapid growth and elongation time of a root system can be effectively prolonged at the jointing stage and heading stage, and the root growth rate can be improved. Promoting root thickening can effectively meet the needs for water and nutrients, for the formation and filling of aboveground plants and grains, in the later stage, which is conducive to the formation of aboveground dry matter production and final yield. The root distribution was greatest in the 0–60 cm soil depth, accounting for 95.13–97.84% of the total root length. After the heading stage occurs, the upper roots begin to decline in large quantities. Thus, the jointing stage and heading stage require fertilization and other farmland management operations to increase root nutrients for the ground parts and dry matter accumulation to provide sufficient nutrients so that the number of effective panicles, grain weight, and the number of spike grains coordinate to achieve the highest grain yield. Results showed that the highest yield can be achieved with the planting pattern X2M1. A comprehensive analysis showed that the genetic characteristics of winter wheat varieties were different, and there were some differences in the correlation between wheat yield and root system at the different growth stages. The correlation between the root parameters and yield of multi-spike winter wheat during the overwintering-jointing stage was obvious. For large-spike type winter wheat in the jointing stage, the yield correlation is most obvious.

Searchable symmetric encryption (SSE) has been applied in constructing secure cloud storage for its simplicity, high-efficiency, and practicality. Recent works show that forward privacy has become a fundamental property required for SSE, and several forward secure dynamic SSE schemes supporting single-keyword search have been proposed. However, to the best of our knowledge, there is no forward secure searchable encryption scheme supporting conjunctive-keyword search. In this paper, we propose two forward secure SSE schemes with conjunctive-keyword search functionality. Technically, in our basic scheme, we introduce a bloom filter on the client side to check the existence of keywords in a file. This also makes it easy to delete a file in locality rather than on the server side. In order to avoid the false positive probability of the bloom filter and reduce the redundant search results, we further propose a construction by applying secret-key inner-product encryption. Both of our schemes achieve sub-linear efficiency and one-round communication in the search protocol. Moreover, our methods are generic, as they can be applied to any forward secure single-keyword searchable encryption scheme to obtain a conjunctive-keyword one.
