With economic development and changes in the industrial structure of cities in China, the transformation of industries from agricultural to non-agricultural has brought about significant changes in the living spaces, economic growth, ideology, and cultural attitudes of rural people. Placing people first and responding to rural residents’ wishes in the process of rapid development could become an important issue. This study focuses on the relationship between the well-being experience of relocated residents in rural areas and the public realm of new communities, and constructs a framework for understanding the relationship between the two. By doing so, it aims to resolve the conflict between the design of new settlements and the residents’ needs, thus providing a reference for similar designs in the future.

ABSTRACTGray sufu is a traditional Chinese fermented soybean curd, and its flavor is significantly influenced by bacterial community. In this study, volatile flavor compounds (VFCs) were examined by headspace solid‐phase microextraction (HS-SPME) in conjunction with gas chromatography and mass spectrometry (GC‐MS). Additionally, bacterial community was analyzed using high-throughput sequencing. The results showed that, a total of 156 VFCs were tentatively identified, in which 8 esters, 6 S/N-containing compounds, 3 alcohols, 3 aldehydes, 1 ketone and 3 other compounds were recognized as the main characteristic VFCs. The abundant S/N-containing compounds are the most important flavor characteristics of gray sufu. A total of 4 phyla and 12 genera were revealed as dominant bacteria. Firmicutes and Proteobacteria were the most abundant phylum, whereas Bacillus, Tetragenococcus and Lactococcus were the prevailing genera. According to the KEGG pathway annotation, amino acid metabolism (26.98%) was the highest metabolic pathway. Among them, aromatic amino acids metabolism accounted for the highest proportion, up to 34.94%. Correlation analysis showed that Lactobacillus, Tetragenococcus, Bacillus, Pseudomonas and Halanaerobium were the top five positive flavor-producing bacteria. The results will be helpful for understanding the fermentation mechanism of gray sufu and improving flavor quality by developing microbial regulation strategies.

Background: The biochemical and genetic characteristics of four very-long-chain acyl-coenzyme A dehydrogenase deficiency (VLCADD) patients, clarifying their pathogenic genetic factors and evaluating the application value of genetic diagnosis in the early diagnosis of VLCADD, are reported and discussed in this article.Methods: Patients underwent blood tandem mass spectrometry (MS/MS), urine gas chromatography (GC/MS), and high-throughput sequencing technology. New variants were analyzed for pathogenicity using bioinformatics software. Swiss-PdbViewer software was used to predict the effect of variants on the structure of the very-long-chain acyl-CoA dehydrogenase (VLCAD) protein.Result: A total of four VLCADD patients were diagnosed. They revealed elevated levels of C14, C14:1, C14:2, C14:1/C2, C14:1/C10, and C14:1/C12:1. Two patients were early-onset neonatal cases and died during infancy and the neonatal period, respectively. Seven kinds of variants were detected, including four novel variants. Bioinformatics software revealed that the variants were harmful, and the Swiss-PdbViewer results suggest that variation affects protein conformation.Conclusion: This study identified four novel ACADVL gene variants. These findings contribute to the understanding of the genetic basis and pathogenesis of VLCADD. Meanwhile, the study enriches the genetic mutation spectrum and the correlation between genotypes and phenotypes of VLCADD, indicating that genetic diagnosis plays an essential role in the early diagnosis and treatment of VLCADD.

Abstract Objective Mucinous breast cancer (MBC) is a kind of breast cancer (BC), which is rare in clinic, mainly for women, because of the low incidence rate, so there is no unified standard treatment protocol. Elderly patients have a poor prognosis due to their combined comorbidities. This study aims to investigate the effect of surgery and chemoradiotherapy on the prognosis of elderly female MBC patients and construct nomograms for predicting the OS and CSS in elderly female MBC patients. Methods Data for female MBC patients over 65 years are obtained from the Surveillance, Epidemiology and End Results (SEER) database, patients were divided into two groups: the training set and the validation set. External validation data of the prediction model were provided by Kunming Hospital of Traditional Chinese Medicine. We used Cox regression modeling, which was used to identify independent risk factors affecting patient prognosis. After avoiding confounding bias according to the multifactorial Cox regression model, we used these screened statistically significant results to construct column‐line plots. The performance of the model was tested using the consistency index (c‐index), the calibration curve, and the area under the operating characteristic curve of the receiver (AUC). Subsequently, we used decision curve analysis (DCA) to examine the potential clinical value of our nomograms. Results A total of 8103 elderly MBC female patients were extracted from the database SEER and were assigned to the training and validation set, randomly. A total of 83 patients from Kunming Hospital of Traditional Chinese Medicine were used in the external verification set. After multifactorial Cox regression analysis, we found that age, race, T‐stage, M‐stage, surgical approach, radiotherapy, and tumor size were independent risk factors for OS in elderly MBC patients. Similarly, independent risk factors of CSS included age, marital status, N stage, M stage, surgical approach, chemotherapy, and tumor size. The C‐index for the OS training, validation, and external verification set were 0.731 (95%CI 0.715–0.747), 0.738 (95%CI 0.724–0.752), and 0.809 (95%CI 0.731–0.8874). The C‐index of the training set, the validation set, and external verification set for CSS were 0.786 (95%CI 0.747–0.825), 0.776 (95%CI 0.737–0.815), and 0.84 (95%CI0.754–0.926), respectively. The AUC, calibration curves and DCA also showed good accuracy. Conclusions In this study, we construct a new nomogram to predict the prognosis of elderly patients with MBC. The nomograms have undergone internal and external validation and have been confirmed to have good clinical applicability. At the same time, we found that for elderly female MBC patients, surgery and radiotherapy significantly benefit their survival, but chemotherapy is not conducive to patient survival.

Abstract Introduction This study evaluates the effectiveness of a combined regimen involving injectable hydrogels for the treatment of experimental myocardial infarction. Patient concerns Myocardial infarction is an acute illness that negatively affects quality of life and increases mortality rates. Experimental models of myocardial infarction can aid in disease research by allowing for the development of therapies that effectively manage disease progression and promote tissue repair. Diagnosis Experimental animal models of myocardial infarction were established using the ligation method on the anterior descending branch of the left coronary artery (LAD). Interventions The efficacy of intracardiac injection of hydrogels, combined with cells, drugs, cytokines, extracellular vesicles, or nucleic acid therapies, was evaluated to assess the functional and morphological improvements in the post-infarction heart achieved through the combined hydrogel regimen. Outcomes A literature review was conducted using PubMed, Web of Science, Scopus, and Cochrane databases. A total of 83 papers, including studies on 1332 experimental animals (rats, mice, rabbits, sheep, and pigs), were included in the meta-analysis based on the inclusion and exclusion criteria. The overall effect size observed in the group receiving combined hydrogel therapy, compared to the group receiving hydrogel treatment alone, resulted in an ejection fraction (EF) improvement of 8.87% [95% confidence interval (CI): 7.53, 10.21] and a fractional shortening (FS) improvement of 6.31% [95% CI: 5.94, 6.67] in rat models, while in mice models, the improvements were 16.45% [95% CI: 11.29, 21.61] for EF and 5.68% [95% CI: 5.15, 6.22] for FS. The most significant improvements in EF (rats: MD = 9.63% [95% CI: 4.02, 15.23]; mice: MD = 23.93% [95% CI: 17.52, 30.84]) and FS (rats: MD = 8.55% [95% CI: 2.54, 14.56]; mice: MD = 5.68% [95% CI: 5.15, 6.22]) were observed when extracellular vesicle therapy was used. Although there have been significant results in large animal experiments, the number of studies conducted in this area is limited. Conclusion The present study demonstrates that combining hydrogel with other therapies effectively improves heart function and morphology. Further preclinical research using large animal models is necessary for additional study and validation. Graphical abstract

Abstract Asthma represents a significant global challenge that affects individuals across all age groups and imposes substantial social and economic burden. Due to heterogeneity of the disease, not all patients obtain benefit with current treatments. The objective of this study was to explore the impact of MD2 on the progression of asthma using L6H21, a novel MD2 inhibitor, to identify potential targets and drug candidates for asthma treatment. To establish an asthma-related murine model and evaluate the effects of L6H21, ovalbumin (OVA) was used to sensitize and challenge mice. Pathological changes were examined with various staining techniques, such as H&E staining, glycogen staining, and Masson staining. Inflammatory cell infiltration and excessive cytokine secretion were evaluated by analyzing BALF cell count, RT-PCR, and ELISA. The TLR4/MD2 complex formation, as well as the activation of the MAPK and NF-кB pathways, was examined using western blot and co-IP. Treatment with L6H21 demonstrated alleviation of increased airway resistance, lung tissue injury, inflammatory cell infiltration and excessive cytokine secretion triggered by OVA. In addition, it also ameliorated mucus production and collagen deposition. In the L6H21 treatment group, inhibition of MAPK and NF-кB activation was observed, along with the disruption of TLR4/MD2 complex formation, in contrast to the model group. Thus, L6H21 effectively reduced the formation of the MD2 and TLR4 complex induced by OVA in a dose-dependent manner. This reduction resulted in the attenuation of MAPKs/NF-κB activation, enhanced suppression of inflammatory factor secretion, reduced excessive recruitment of inflammatory cells, and ultimately mitigated airway damage. MD2 emerges as a crucial target for asthma treatment, and L6H21, as an MD2 inhibitor, shows promise as a potential drug candidate for the treatment of asthma.

The aim of this study was to explore the formation of volatile lipid oxidation products by the lipoxygenase (LOX)-hydroperoxide lyase (HPL)-mediated pathway in oat, barley and soy bean. LOX activity was found only in barley and soy bean samples, but the lipase and HPL activity was detected in all samples. HPL showed particularly high activity with 13-hydroperoxides, while the activity was quite low when using 9-hydroperoxides, especially in the oat and barley. The optimum pH for HPL in different samples was similar, i.e., pH 6–7. In this condition, the volatile compounds formed dramatically with aldehydes and furans as the dominant products. Furthermore, a remarkable enzymatic degradation of lipids occurred during the preparation of food models with highly refined rapeseed oil (RO) and rapeseed oil fatty acid (ROFA) emulsions, where the ROFAs were more prone to oxidation than RO. This study shows the significance of lipid-degrading enzymes in plant-food flavour formation.

This study aimed to elevate the sensory value of Chinese steamed bread (CSB) by incorporating a blend of oats and corn. Through meticulous formulation, we achieved an optimal amalgamation for oat and corn Chinese steamed bread (OCCSB), comprising 28 % Yunnan corn flour (with 36 % amylose), 6 % Sichuan corn flour (with 26 % amylose), and 2 % oat flour. Employing a comprehensive array of analytical techniques encompassing rheology, microscopy, color assessment, and sensory quality rating, we conducted a comparative study involving OCCSB, meticulously optimized CSB formulations, and commercially available corn-enriched CSBs (comprising 20 % or 25 % corn flour). OCCSB containing 34 % corn flour and 2 % oat flour could be a promising candidate, with the percentage of the corn flour content in the bread being about 1.51 times that of the commercial corn CSBs. OCCSB also had a higher gas porosity, spread ratio (1.38) and quality score (68.13), compared with the commercial corn CSBs. Its appealing visual profile was highlighted by a vivid yellow hue in both the crust and crumb. Furthermore, its unparalleled quality score outshone those of commercial alternatives. These noteworthy findings illuminate the potential of corn and oat as staple food ingredients, propelling advancements in the remarkable processing and practical application.

Abstract Background Intrahepatic cholangiocarcinoma (ICC) is poorly treated due to the presence of an inhibitory immune microenvironment. Tumor-associated macrophages (TAM) are an important component of TME. ALOX5 is an important lipid metabolism enzyme in cancer progression, but the mechanism by which it regulates TAM to promote ICC progression is unknown. The aim of this study was to investigate the potential mechanism of TAM regulation by ALOX5 and the translational effect of targeting ALOX5. Methods In this study, we investigated the association between the spatial localization of epithelial cells and TAMs by combining scRNA-seq analysis with multiplex immunofluorescence analysis. Through bulk sequencing analysis and spatial analysis, lipid metabolism genes closely related to TAM infiltration were screened. In vitro co-culture model was constructed to verify that ALOX5 and its downstream metabolite LTB4 promote M2 macrophage migration. Bulk sequencing after co-culture combined with single-cell analysis was performed to identify key pathways for up-regulation of M2 macrophage migration. Finally, the effect of CSF1R inhibitor (PLX3397) combined with ALOX5 inhibitor (Zileuton) in vivo was investigated by by xenograft tumor formation experiment in nude mice. Results ALOX5 in ICC cells was a key lipid metabolism gene affecting the infiltration of M2 macrophages in TME. Mechanically, LTB4, a metabolite downstream of ALOX5, recruited M2 macrophages to migrate around tumor cells by binding to BLT1/BLT2 and activating the PI3K pathway, which ultimately lead to the promotion of ICC progression. Targeting CSF1R in combination with ALOX5 inhibitor effectively reduced tumor volume and M2 macrophage infiltration abundance. Conclusion In ICC, LTB4, a metabolite secreted by ALOX5 of epithelial cells, binded to BLT1/BLT2 on TAM surface to activate PI3K pathway and promote TAM migration, thus promoting ICC progression. Targeting CSF1R in combination with ALOX5 inhibitor for ICC is a promising combination therapy modality.

This paper seeks contemporary cultural heritage conservation principles by reviewing its history, starting from the 18th century, in practices, international documents, and the literature on this topic. It intends to lay a foundation to avoid damaging cultural heritages by misconducting conservation. This study first found that the conservation objects of cultural heritage include particularly important ones and general ones that are closely related to daily life, and they involve tangible and intangible aspects. Second, cultural heritage conservation involves document-based restoration when necessary as well as identifying the value of them to play their role nowadays. Third, integrating cultural heritage conservation within the context of sustainable development goals is essential for successfully balancing the relationship between the built and natural environments. Fourth, mobilising the public to participate in cultural heritage conservation enables the implementation of conservation to meet the expectations of the public, and may have a positive influence on people’s consciousness. Fifth, as a treasure, cultural heritage conservation is a global responsibility that involves conjoint cooperation. Sixth, more cultural heritage conservation interdisciplinary methods have been developed and could be applied, but this should be limited in order to not destruct their authenticity and integrity.

Abstract Interacting electrons in one dimension (1D) are governed by the Luttinger liquid (LL) theory in which excitations are fractionalized. Can a LL-like state emerge in a 2D system as a stable zero-temperature phase? This question is crucial in the study of non-Fermi liquids. A recent experiment identified twisted bilayer tungsten ditelluride (tWTe2) as a 2D host of LL-like physics at a few kelvins. Here we report evidence for a 2D anisotropic LL state down to 50 mK, spontaneously formed in tWTe2 with a twist angle of ~ 3o. While the system is metallic-like and nearly isotropic above 2 K, a dramatically enhanced electronic anisotropy develops in the millikelvin regime. In the anisotropic phase, we observe characteristics of a 2D LL phase including a power-law across-wire conductance and a zero-bias dip in the along-wire differential resistance. Our results represent a step forward in the search for stable LL physics beyond 1D.

Inter- and intra-specific variations in phenotype are common and can be associated with genomic mutations as well as epigenomic variation. Profiling both genomic and epigenomic variants is at the core of dissecting phenotypic variation. However, an efficient targeted genotyping and epigenotyping system is lacking. We describe a new multiplex targeted genotyping and epigenotyping system called improved bulked-PCR sequencing (iBP-seq). We employed iBP-seq for the detection of genotypes and methylation levels of dozens of target regions in mixed DNA samples. iBP-seq can be adapted for the construction of linkage maps, fine mapping of quantitative-trait loci, and detection of genome editing mutations at a cost as low as $0.016 per site per sample. We developed an automated bioinformatics pipeline, including primer design, a series of bioinformatic analyses for genotyping and epigenotyping, and visualization of results. iBP-seq and its bioinformatics pipeline, available at http://zeasystemsbio.hzau.edu.cn/tools/ibp/, can be adapted to a wide variety of species.

Abstract The conventional understanding that chimeric RNAs are unique to carcinoma and are the products of chromosomal rearrangement is being challenged. However, experimental evidence supporting the function of chimeric RNAs in normal physiology is scarce. We decided to focus on one particular chimeric RNA, CTNNBIP1-CLSTN1. We examined its expression in various tissues and cell types and compared it quantitatively among cancer and noncancer cells. We further investigated its role in a panel of noncancer cells and investigated the functional mechanism. We found that this fusion transcript is expressed in almost all tissues and a wide range of cell types, including fibroblasts, epithelial cells, stem cells, vascular endothelial cells, and hepatocytes. In addition, the CTNNBIP1-CLSTN1 expression level in noncancerous cell lines was not evidently different from that in cancer cell lines. Furthermore, in at least three cell types, silencing CTNNBIP1-CLSTN1 significantly reduced the cell proliferation rate by inducing G2/M arrest and apoptosis. Importantly, rescue experiments confirmed that cell cycle arrest was restored by exogenous expression of the chimera but not the wild-type parental gene. Further evidence is provided that CTNNBIP1-CLSTN1 regulates cell proliferation through SERPINE2. Thus, CTNNBIP1-CLSTN1 is an example of a new class of fusion RNAs, dubbed “housekeeping chimeric RNAs”.

Abstract Microglia, the major resident immune cells in the central nervous system, serve as the frontline soldiers against cerebral ischemic injuries, possibly along with metabolic alterations. However, signaling pathways involved in the regulation of microglial immunometabolism in ischemic stroke remain to be further elucidated. In this study, using single‐nuclei RNA sequencing, a microglial subcluster up‐regulated in ischemic brain tissues is identified, with high expression of Igf1 and Trem2, neuroprotective transcriptional signature and enhanced oxidative phosphorylation. Microglial depletion by PLX3397 exacerbates ischemic brain damage, which is reversed by repopulating the microglia with high Igf1 and Trem2 phenotype. Mechanistically, Igf1 serves as one of the major down‐stream molecules of Trem2, and Trem2‐Igf1 signaling axis regulates microglial functional and metabolic profiles, exerting neuroprotective effects on ischemic stroke. Overexpression of Igf1 and supplementation of cyclocreatine restore microglial glucometabolic levels and cellular functions even in the absence of Trem2. These findings suggest that Trem2‐Igf1 signaling axis reprograms microglial immunometabolic profiles and shifts microglia toward a neuroprotective phenotype, which has promising therapeutic potential in treating ischemic stroke.

Source attribution has traditionally involved combining epidemiological data with different pathogen characterisation methods, including 7-gene multi locus sequence typing (MLST) or serotyping, however, these approaches have limited resolution. In contrast, whole genome sequencing data provide an overview of the whole genome that can be used by attribution algorithms. Here, we applied a random forest (RF) algorithm to predict the primary sources of human clinical Salmonella Typhimurium (S. Typhimurium) and monophasic variants (monophasic S. Typhimurium) isolates. To this end, we utilised single nucleotide polymorphism diversity in the core genome MLST alleles obtained from 1,061 laboratory-confirmed human and animal S. Typhimurium and monophasic S. Typhimurium isolates as inputs into a RF model. The algorithm was used for supervised learning to classify 399 animal S. Typhimurium and monophasic S. Typhimurium isolates into one of eight distinct primary source classes comprising common livestock and pet animal species: cattle, pigs, sheep, other mammals (pets: mostly dogs and horses), broilers, layers, turkeys, and game birds (pheasants, quail, and pigeons). When applied to the training set animal isolates, model accuracy was 0.929 and kappa 0.905, whereas for the test set animal isolates, for which the primary source class information was withheld from the model, the accuracy was 0.779 and kappa 0.700. Subsequently, the model was applied to assign 662 human clinical cases to the eight primary source classes. In the dataset, 60/399 (15.0%) of the animal and 141/662 (21.3%) of the human isolates were associated with a known outbreak of S. Typhimurium definitive type (DT) 104. All but two of the 141 DT104 outbreak linked human isolates were correctly attributed by the model to the primary source classes identified as the origin of the DT104 outbreak. A model that was run without the clonal DT104 animal isolates produced largely congruent outputs (training set accuracy 0.989 and kappa 0.985; test set accuracy 0.781 and kappa 0.663). Overall, our results show that RF offers considerable promise as a suitable methodology for epidemiological tracking and source attribution for foodborne pathogens.

IntroductionThe gut microbiota plays an important role in the development of non-alcoholic steatohepatitis (NASH), but the underlying mechanism is unclear. It has been found that the transcription factor XBP1s plays an important role in regulating inflammation and lipid metabolism and maintaining the integrity of intestinal barrier. However, whether XBP1s modulates the development of NASH by regulating the integrity of the intestinal barrier and altering the composition of the gut microbiota remains unknown.MethodsMice fed with a fat-, fructose-, cholesterol-rich (FFC) diet for 24 weeks successfully established the NASH model, as demonstrated by significant hepatic steatosis, inflammation, hepatocyte injury and fibrosis. The profile of gut microbiota dynamically changed with the different stages of NAFLD via 16S rDNA sequencing the feces from mice fed with FFC diet for 0, 12, or 24 weeks or NASH mice treated with siRNA-loaded folic acid-modified TPGS (hereafter named FT@XBP1).ResultsNASH mice had significantly higher abundance of Firmicutes, Blautia and Bacteroides, and lower abundance of Bifidobacterium and GCA-900066575. FT@XBP1 supplementation had a significantly attenuated effect on FFC diet-induced weight gain, hepatic fat accumulation, dyslipidemia, inflammatory cytokines, ER stress and fibrosis. In particularly, FT@XBP1 modulates the composition of the intestinal flora; for example, NASH mice demonstrated higher abundance of Blautia and Bacteroides, and lower abundance of Actinobacteriota, Muribaculaceae and Bifidobacterium, which were partially restored by FT@XBP1 treatment. Mechanistically, FT@XBP1 increased the expression of ZO-1 in the intestine and had the potential to restore intestinal barrier integrity and improve antimicrobial defense to alleviate enterogenic endotoxemia and activation of inflammatory signaling pathways.DiscussionRegulation of the key transcription factor XBP1s can partially restore the intestinal microbiota structure, maintain the integrity of intestinal mucosal barrier, and prevent the progression of NASH, providing new evidence for treating NASH.

Recently emerged S. Infantis strains carrying resistance to several commonly used antimicrobials have been reported from different parts of the globe, causing human cases of salmonellosis and with occurrence reported predominantly in broiler chickens. Here, we performed phylogenetic and genetic clustering analyses to describe the population structure of 417 S. Infantis originating from multiple European countries and the Americas collected between 1985 and 2019. Of these, 171 were collected from 56 distinct premises located in England and Wales (E/W) between 2009 and 2019, including isolates linked to incursions of multidrug-resistant (MDR) strains from Europe associated with imported poultry meat. The analysis facilitated the comparison of isolates from different E/W sources with isolates originating from other countries. There was a high degree of congruency between the outputs of different types of population structure analyses revealing that the E/W and central European (Germany, Hungary, and Poland) isolates formed several disparate groups, which were distinct from the cluster relating to the United States (USA) and Ecuador/Peru, but that isolates from Brazil were closely related to the E/W and the central European isolates. Nearly half of the analysed strains/genomes (194/417) harboured the IncFIB(pN55391) replicon typical of the “parasitic” pESI-like megaplasmid found in diverse strains of S. Infantis. The isolates that contained the IncFIB(pN55391) replicon clustered together, despite originating from different parts of the globe. This outcome was corroborated by the time-measured phylogeny, which indicated that the initial acquisition of IncFIB(pN55391) likely occurred in Europe in the late 1980s, with a single introduction of IncFIB(pN55391)-carrying S. Infantis to the Americas several years later. Most of the antimicrobial resistance (AMR) genes were identified in isolates that harboured one or more different plasmids, but based on the short-read assemblies, only a minority of the resistance genes found in these isolates were identified as being associated with the detected plasmids, whereas the hybrid assemblies comprising the short and long reads demonstrated that the majority of the identified AMR genes were associated with IncFIB(pN55391) and other detected plasmid replicon types. This finding underlies the importance of applying appropriate methodologies to investigate associations of AMR genes with bacterial plasmids.

Introduction: Elderly patients are more prone to develop acute kidney injury during infections and polymyxin B (PMB)-associated nephrotoxicity than young patients. The differential response to PMB between the elderly and young critically ill patients is unknown. We aimed to assess PMB exposure in elderly patients compared with young critically ill patients, and to determine the covariates of PMB pharmacokinetics in critically ill patients.Methods: Seventeen elderly patients (age ≥ 65 years) and six young critically ill patients (age < 65 years) were enrolled. Six to eight blood samples were collected during the 12 h intervals after at least six doses of intravenous PMB in each patient. PMB plasma concentrations were quantified by high-performance liquid chromatography-tandem mass spectrometry. The primary outcome was PMB exposure as assessed by the area under the concentration-time curve over 24 h at steady state (AUCss, 0–24 h).Results and Discussion: The elderly group had lower total body weight (TBW) and higher Charlson comorbidity scores than young group. Neither AUCss, 0–24 h nor normalized AUCss, 0–24 h (adjusting AUC for the daily dose in mg/kg of TBW) was significantly different between the elderly group and young group. The half-life time was longer in the elderly patients than in young patients (11.21 vs 6.56 h respectively, p = 0.003). Age and TBW were the covariates of half-life time (r = 0.415, p = 0.049 and r = −0.489, p = 0.018, respectively). TBW was the covariate of clearance (r = 0.527, p = 0.010) and AUCss, 0–24 h (r = −0.414, p = 0.049). Patients with AUCss, 0–24 h ≥ 100 mg·h/L had higher baseline serum creatinine levels and lower TBW than patients with AUCss, 0–24 h < 50 mg·h/L or patients with AUCss, 0–24 h 50–100 mg·h/L. The PMB exposures were comparable in elderly and young critically ill patients. High baseline serum creatinine levels and low TBW was associated with PMB overdose.Trial registration: ChiCTR2300073896 retrospectively registered on 25 July 2023.

Chemoproteomic profiling of sulfhydryl-containing proteins has consistently been an attractive research hotspot. However, there remains a dearth of probes that are specifically designed for sulfhydryl-containing proteins, possessing sufficient reactivity, specificity, distinctive isotopic signature, as well as efficient labeling and evaluation capabilities for proteins implicated in the regulation of redox homeostasis. Here, the specific selenium-containing probes (Se-probes) in this work displayed high specificity and reactivity toward cysteine thiols on small molecules, peptides and purified proteins and showed very good competitive effect of proteins labeling in gel-ABPP. We identified more than 6000 candidate proteins. In TOP-ABPP, we investigated the peptide labeled by Se-probes, which revealed a distinct isotopic envelope pattern of selenium in both the primary and secondary mass spectra. This unique pattern can provide compelling evidence for identifying redox regulatory proteins and other target peptides. Furthermore, our examiation of post-translational modification (PTMs) of the cysteine site residues showed that oxidation PTMs was predominantly observed. We anticipate that Se-probes will enable broader and deeper proteome-wide profiling of sulfhydryl-containing proteins, provide an ideal tool for focusing on proteins that regulate redox homeostasis and advance the development of innovative selenium-based pharmaceuticals.

Abstract Background Acute type A aortic dissection (ATAAD) is a catastrophic disease with high morbidity and mortality. Although open surgery is still the gold standard for the treatment of ATAAD, some patients, with advanced age and multiple comorbidities, can only receive medical management alone. Nowadays, thoracic aortic endovascular repair (TEVAR) provides a potential treatment option for the patient with ATAAD, but traditional stent grafts (SGs), which are not designed for the ATAAD, are inapplicable to the unique anatomy of the aortic arch. Therefore, we innovatively created the BRIDGE system (Chuangxin Medical, Shenzhen, China), a complete endovascular reconstruction system designed to treat ATAAD. This study aimed to evaluate the feasibility and safety of the novel Stanford A aortic dissection complete endovascular reconstruction system in a porcine model. Method The BRIDGE system consists of the type A stent system and the type C stent system. Between November 2020 and March 2021, three white swine were utilized in the study. The BRIDGE system was deployed via the transcatheter approach under angiographic guidance. The swine(n = 3) treated with our system were evaluated using angiography before sacrifice 1-month after implantation, which was followed by gross specimen evaluation and histological examination of harvested tissues. Result The acute procedure success rate was 100% (3/3). The immediate post-procedural angiography showed that both type A SGs and type C SGs were deployed in satisfactory locations, with patency of the supra-aortic trunk and no endoleak. The cumulative mortality of 30-day was 0% without any adverse events. No device migration or leakage was observed angiographically, before sacrifice. The gross observation confirmed a type A SG covered part of the entry of anonyma. Favorable endothelialization, no thrombogenesis, and slight inflammatory infiltration of the tissues around the device were confirmed by microscopic examinations in all pigs. Conclusion It was feasible and secure to use Stanford A aortic dissection complete endovascular reconstruction system to implement a transcatheter endovascular repair in a porcine model. With this novel system, treating acute type A aortic dissection may be more efficient and secure in human.

The porosity characteristics of coal seams serve as a pivotal factor in assessing the development potential of coalbed methane (CBM) resources, significantly influencing the adsorption and permeability capabilities of coal reservoirs, as well as the accumulation, entrapment, and preservation of CBM. In this study, we focused on the coal seams of the Xishanyao Formation in the western part of the southern Junggar Basin (NW China). By leveraging the complementarity of nuclear magnetic resonance (NMR), low-temperature liquid nitrogen experiments, and high-pressure mercury intrusion porosimetry (MIP) in spatial exploration range and precision, we conducted a comprehensive analysis to achieve a fine description of porosity characteristics. Furthermore, we explored the coal petrology factors controlling the pore characteristics of the Xishanyao Formation, aiming to provide geological evidence for the selection of favorable areas and the development potential evaluation of CBM in the study area. The results indicate the following: (1) The total pore volume of the coal samples is 6.318 × 10−3 cm3/g on average, and the micropore volume accounts for a relatively high proportion (averaging 44.17%), followed by the fine pores (averaging 39.41%). The average porosity is approximately 3.87%, indicating good gas storage and connectivity of the coal seams, albeit with some heterogeneity. The coal reservoir is dominated by micropores and fine pores with diameters less than 100 nm, and the pore structure is characterized by low pore volume and high pore area. (2) The pore structure is influenced by both the coalification degree and the coal maceral. Within the range of low coalification, porosity increases with the increase in coalification degree. Building upon this, an increase in the vitrinite content promotes the development of micropores and fine pores, while an increase in the inertinite content promotes the development of meso–macropores. The clay mineral content exhibits a negative correlation with the adsorption pore volume ratio and a positive correlation with the seepage pore volume ratio.

In this paper, the roasting, leaching, purification and enrichment and vanadium precipitation technology were reviewed. The research work and methods of vanadium separation by solvent extraction were summarized, and the extraction of vanadium by tertiary amine extraction agent and its development status at home and abroad were discussed emphatically.

This is a paper in the field of metallurgical engineering. Organophosphorus class extractant D2EHPA (P204) is very widely used in vanadium extraction. Firstly, the form of vanadium present in the leach solution affects the effect of extraction, V(IV) is mainly stable in solution in the form of VO2+ in a strongly acidic solution; when the pH value gradually increases, V(V) mostly exists in the form of H2V10O28 4- and HV10O28 5-. Secondly, based on the research of organophosphorus extractant P204 in recent years, this paper analyzes the conformational relationship of organophosphorus extractant P204 and finds that the spatial site resistance effect is equally important as a secondary factor compared with the type of phosphoryl adjacent groups, based on the hydrogen bonding effect, the extractant molecules will produce dimers or even multimers between them, thus changing the structure and properties of the extractant molecules. Finally, the progress of the development and application of P204 extractive vanadium and the outlook on the future of the related fields are presented, pointing out the future research directions of P204 extractive agents.

Abstract Neuromyelitis optica spectrum disorder (NMOSD) is an inflammatory demyelinating disorder of the central nervous system (CNS) triggered by autoimmune mechanisms. Microglia are activated and play a pivotal role in response to tissue injury. Triggering receptor expressed on myeloid cells 2 (TREM2) is expressed by microglia and promotes microglial activation, survival and phagocytosis. Here, we identify a critical role for TREM2 in microglial activation and function during AQP4-IgG and complement-induced demyelination. TREM2-deficient mice had more severe tissue damage and neurological impairment, as well as fewer oligodendrocytes with suppressed proliferation and maturation. The number of microglia clustering in NMOSD lesions and their proliferation were reduced in TREM2-deficient mice. Moreover, morphology analysis and expression of classic markers showed compromised activation of microglia in TREM2-deficient mice, which was accompanied by suppressed phagocytosis and degradation of myelin debris by microglia. These results overall indicate that TREM2 is a key regulator of microglial activation and exert neuroprotective effects in NMOSD demyelination. Graphical Abstract

This study investigated the effect of electron beam irradiation (EBI) on the lipid stability of oat and barley during long-term storage. Results showed that the initial free fatty acid content in oat was higher than that in barley. This may mean that lipid hydrolysis started under the function of lipase when oat and barley were milled into flours. Both storage and EBI factors influenced lipid-degrading enzyme activity and promoted lipid oxidation in oat and barley. However, it seemed that storage had higher impacts because the DPPH scavenging activity decreased greatly, and the contents of both malondialdehyde and volatile lipid oxidation products increased in all samples. Thus, the antioxidant capacity and level of lipid oxidation after EBI treatment should be considered when producing oat and barley foods. Overall, this study shows the high potential of EBI for use as a non-thermal technique in stabilising the storage quality of oat and barley.

l-Heptopyranoses are important components of bacterial polysaccharides and biological active secondary metabolites like septacidin (SEP), which represents a group of nucleoside antibiotics with antitumor, antifungal, and pain-relief activities. However, little is known about the formation mechanisms of those l-heptose moieties. In this study, we deciphered the biosynthetic pathway of the l,l-gluco-heptosamine moiety in SEPs by functional characterizing four genes and proposed that SepI initiates the process by oxidizing the 4′-hydroxyl of l-glycero-α-d-manno-heptose moiety of SEP-328 (2) to a keto group. Subsequently, SepJ (C5 epimerase) and SepA (C3 epimerase) shape the 4′-keto-l-heptopyranose moiety by sequential epimerization reactions. At the last step, an aminotransferase SepG installs the 4′-amino group of the l,l-gluco-heptosamine moiety to generate SEP-327 (3). An interesting phenomenon is that the SEP intermediates with 4′-keto-l-heptopyranose moieties exist as special bicyclic sugars with hemiacetal-hemiketal structures. Notably, l-pyranose is usually converted from d-pyranose by bifunctional C3/C5 epimerase. SepA is an unprecedented monofunctional l-pyranose C3 epimerase. Further in silico and experimental studies revealed that it represents an overlooked metal dependent-sugar epimerase family bearing vicinal oxygen chelate (VOC) architecture.

Although robots have been widely used in a variety of fields, the idea of enabling them to perform multiple tasks in the same way that humans do remains a difficulty. To solve this, we investigate the learning from demonstration (LFD) system with our independently designed symmetrical humanoid dual-arm robot. We present a novel action feature matching algorithm. This algorithm accurately transforms human demonstration data into task models that robots can directly execute, considerably improving LFD’s generalization capabilities. In our studies, we used motion capture cameras to capture human demonstration actions, which included combinations of simple actions (the action layer) and a succession of complicated operational tasks (the task layer). For the action layer data, we employed Gaussian mixture models (GMM) for processing and constructing an action primitive library. As for the task layer data, we created a “keyframe” segmentation method to transform this data into a series of action primitives and build another action primitive library. Guided by our algorithm, the robot successfully imitated complex human tasks. Results show its excellent task learning and execution, providing an effective solution for robots to learn from human demonstrations and significantly advancing robot technology.

This study systematically investigates the impact of corn starch molecular structures on the quality attributes of surimi gel products. Employing molecular analyses to characterize corn starch, three amylopectin fractions (A, B1, and B2), categorized by the degree of polymerization ranges (6 < X ≤ 12, 12 < X ≤ 24, and 24 < X ≤ 36, respectively) were specifically focused on. The surimi gel quality was comprehensively assessed through texture profile analysis, nuclear magnetic resonance, scanning electron microscopy, stained section analysis, and Fourier transform infrared spectroscopy. Results indicated the substantial volume expansion of corn amylopectin upon water absorption, effectively occupying the surimi gel matrix and fostering the development of a more densely packed protein network. Starch gels with higher proportions of A, B1, and B2 exhibited improved hardness, chewiness, and bound water content in the resultant surimi gels. The weight-average molecular weight and peak molecular weight of corn starch showed a strong positive correlation with surimi gel hardness and chewiness. Notably, the secondary structure of proteins within the surimi gel was found to be independent of corn starch’s molecular structure. This study provides valuable insights for optimizing formulations in surimi gel products, emphasizing the significance of elevated A, B1, and B2 content in corn starch as an optimal choice for crafting dense, chewy, water-retaining surimi gels.

Background: Multiparous women are at a higher risk of gestational diabetes mellitus (GDM) than primiparas. Physical activity during pregnancy has been shown to be beneficial for GDM, but there is little evidence on the association between physical activity and glycemic control among women with GDM, whether primiparas or multiparas. Thus, the objective of the present study was to examine the association between physical activity and glycemic control in women with GDM and to determine what, if any, effects result from number of parous events. Methods: A prospective cohort of 1162 women with GDM was recruited, with 604 multiparas (51.98%). The general linear model was used to calculate the risk difference and its 95% confidence interval (95%CI) to quantify the impact of parous events on glycemic control in pregnancy as well as the association between physical activity time and glycemic control. Results: Among 1162 women with GDM, the median daily activity time was 65 min (interquartile range (IQR): 45–90 min), and the abnormal plasma glucose (PG) percentage, calculated as number of abnormal PG tests divided by the total number of PG tests, was 40.00% (IQR: 22.22%–66.67%). The percentage of abnormal PG was stabilized and statistically lower with daily physical activity time exceeding 60 min among primiparas (IQR: 30.89%–44.43%) and exceeding 90 min among multiparas (ranged from 27.76% to 38.82%). After adjusting for potential confounders, primiparas tended to have a lower percentage of abnormal PG than do multiparas (rate difference = –0.39, 95%CI: –3.61 to 2.84). The same amount of physical activity time was significantly less effective for multiparas than for primiparas (trend p-value < 0.01). Conclusion: In women with GDM, being multiparous is associated with less effective glycemic control through physical activity, such that multiparas need more physical activity to achieve glycemic control at a similar level to primiparas.

Abstract Background Eccrine sweat glands (ESGs) and hair follicles (HFs) are the prominent skin appendages regulating human body temperature. C57BL/6 mice and Sprague–Dawley (SD) rats are the most commonly used model animals for studying ESGs and HFs. Previous studies have shown the distribution of ESGs and HFs in volar hindfeet of C57BL/6 mice, but there are few or no reports on the distribution of ESGs and HFs in volar forefeet of C57BL/6 mice and volar feet of SD rats. Here, we investigated the differential distribution and genetic determination of ESGs and HFs in the volar skin of C57BL/6 mice and SD rats through gross observation, iodine-starch sweat test, double staining with Nile Blue A and Oil Red O, hematoxylin and eosin (HE) staining, double immunofluorescence staining of LIM Homeobox 2 (LHX2)/Na+-K+-ATPase α1(NKA) or LHX2/Na+-K+-2Cl－ cotransporter 1 (NKCC1), and qRT-PCR detection of ESG-related gene Engrailed 1 (En1) and HF-related gene LHX2. Results The results showed ESGs but no HFs in the footpads of C57BL/6 mice and SD rats, both ESGs and HFs in the inter-footpads (IFPs) of C57BL/6 mice, and neither ESGs nor HFs in the IFPs of SD rats. The relative quantitative change in En1 was consistent with the differential distribution of ESGs, and the relative quantitative change of LHX2 was consistent with the differential distribution of HFs. Conclusion C57BL/6 mice and SD rats had their own characteristics in the distribution of ESGs and HFs in the volar skin, and researchers should choose mice or rats, and even forefeet or hindfeet as their research object according to different purposes. The study provides a basis for selection of optimal animal models to study development, wound healing and regeneration of skin appendages.

Background: Two common techniques used in canteen hygiene supervision, are the coliform paper assay, which is the standard method, and the adenosine triphosphate (ATP) bioluminescence method. The coliform paper assay requires the incubation of the sample, which is time-consuming and does not provide a real-time assessment. Meanwhile, the ATP bioluminescence assay can provide real-time kitchenware cleanliness data. Objective: This study aimed to compare these two methods for evaluating the sanitary condition of kitchenware and explore whether the ATP bioluminescence assay can be used as a standard method in sanitary inspection. Methods: In this study, the cluster random sampling method was used to sample kitchenware from six canteens in the Hebei province, China. Samples were, assessed through the coliform paper test and ATP bioluminescence assay. Results: Kitchenware negative rates for the coliform paper method and the ATP test were 64.39% and 49.07%, respectively. The Escherichia coli positive detection rate grew steadily as the relative light units (RLU) value for the ATP technique increased. The kappa coefficient for the two methods was 0.549, indicating that the two methods yield relatively consistent results. Conclusion: Although currently not considered a standard method, simply using ATP detection is advantageous for quick on-site detection in catering unit hygiene supervision.

To solve error propagation and exorbitant computational complexity of signal detection in wireless multiple-input multiple-output-orthogonal frequency division multiplexing (MIMO-OFDM) systems, a low-complex and efficient signal detection with iterative feedback is proposed via a constellation point feedback optimization of minimum mean square error-ordered successive interference cancellation (MMSE-OSIC) to approach the optimal detection. The candidate vectors are formed by selecting the candidate constellation points. Additionally, the vector most approaching received signals is chosen by the maximum likelihood (ML) criterion in formed candidate vectors to reduce the error propagation by previous erroneous decision, thus improving the detection performance. Under a large number of matrix inversion operations in the above iterative MMSE process, effective and fast signal detection is hard to be achieved. Then, a symmetric successive relaxation iterative algorithm is proposed to avoid the complex matrix inversion calculation process. The relaxation factor and initial iteration value are reasonably configured with low computational complexity to achieve good detection close to that of the MMSE with fewer iterations. Simultaneously, the error diffusion and complexity accumulation caused by the successive detection of the subsequent OSIC scheme are also improved. In addition, a method via a parallel coarse and fine detection deals with several layers to both reduce iterations and improve performance. Therefore, the proposed scheme significantly promotes the MIMO-OFDM performance and thus plays an irreplaceable role in the future sixth generation (6G) mobile communications and wireless sensor networks, and so on.

The increasing frequency of extreme rainfall is leading to a rise in groundwater levels in coastal areas, significantly affecting high-speed railway operations. To address this concern, this study developed a 2.5-dimensional finite element model of a coupled track-embankment-ground system based on Biot’s porous media theory to analyze the effect of groundwater level rise on the critical velocity of high-speed railways and vibration responses. The findings reveal a consistent decrease in the critical velocity of high-speed railways with rising groundwater levels. Particularly, the increase in groundwater levels within the embankment significantly influences the critical velocity compared to a similar rise in the foundation’s groundwater level. Furthermore, deformations induced by passing trains significantly increase as groundwater levels rise. Specifically, when the groundwater level rises from the foundation bottom to the subgrade surface, subgrade surface deformation increases by approximately 55%. As trains approach the critical velocity, significant vibration phenomena, known as the “Mach effect,” occur at the foundation surface. Importantly, as groundwater levels rise, the “Mach effect” intensifies. Analyzing the vibrating frequency spectrum of the displacement response demonstrates a substantial increase in vibration amplitude, particularly in the high-frequency region, as groundwater levels rise. This study highlights that the rise in groundwater level not only amplifies vibrations but also extends the propagation of high-frequency vibrations, underscoring the importance of effective embankment waterproofing in controlling track vibrations.

Abstract Background Soluble triggering receptor expressed on myeloid cells 2 (sTREM2) in cerebrospinal fluid (CSF) has been described as a biomarker for microglial activation, which were observed increased in a variety of neurological disorders. Objective Our objective was to explore whether genetically determined CSF sTREM2 levels are causally associated with different neurological diseases by conducting a two-sample Mendelian randomization (MR) study. Methods Single nucleotide polymorphisms significantly associated with CSF sTREM2 levels were selected as instrumental variables to estimate the causal effects on clinically common neurological diseases, including stroke, Alzheimer’s diseases, Parkinson’s diseases, amyotrophic lateral sclerosis, multiple sclerosis, and epilepsy and their subtypes. Summary-level statistics of both exposure and outcomes were applied in an MR framework. Results Genetically predicted per 1 pg/dL increase of CSF sTREM2 levels was associated with higher risk of multiple sclerosis (OR = 1.038, 95%CI = 1.014–1.064, p = 0.002). Null association was found in risk of other included neurological disorders. Conclusions These findings provide support for a potential causal relationship between elevated CSF sTREM2 levels and higher risk of multiple sclerosis.

Abstract Paleogeographic reconstruction of Precambrian terranes reworked by Phanerozoic orogens (e.g., the Tibetan Plateau) results in complex lithotectonic relations due to intracrustal reworking by tectonothermal events. Detrital zircon rare earth element (REE) databases at global (global major river sands) and regional (the Gangdese Mountains, southern Tibet) scales reveal trends in LREEN‾/HREEN‾ and Eu/Eu* that effectively record the crustal evolution of the source, including crustal thickness and redox state of the magma that generated the zircons. Regional comparisons of these chemical markers provide a new approach for paleogeographic reconstructions that we apply to study the origin of the Lhasa terrane, southern Tibet. Using Precambrian to early Paleozoic sedimentary and igneous rocks in the Lhasa terrane and compiling detrital zircon analyses from the northern margin of Gondwana, we show that the Lhasa terrane had an African affinity in the Rodinia–Gondwana supercontinent cycles (ca. 1.4–0.4 Ga).

BackgroundRegulatory T cells (Treg) have been identified as a key modulator of neuroinflammation in stroke. However, little is known about the association of Treg subpopulations with clinical outcome in patients with acute ischemic stroke (AIS).MethodsPatients within 1 week from stroke onset were prospectively enrolled in this study. Healthy controls were sex-and age-matched 1:1 to AIS patients. The frequencies of Treg and Treg subsets were analyzed by flow cytometry and compared with nonstroke control. Univariate and multivariate logistic regression analysis was performed to investigate the prognostic value of Treg subsets in stroke outcomes.ResultsA total of 328 patients and 328 controls were included in the study. Compared with controls, patients with AIS had higher levels of Treg frequency and memory Treg (mTreg) frequency, but lower levels of naïve Treg (nTreg) frequency and nTreg/mTreg ratio. One hundred twenty-six (38.4%) patients experienced unfavorable outcome (modified Rankin score 2–6). Multivariate regression analysis showed that nTreg/mTreg ratio was negatively associated with unfavorable 90-day outcome (the highest tertile versus the lowest tertile: odds ratio 0.13, 95% confidential interval [CI] 0.05–0.35). The risk estimation of unfavorable 90 day outcome can be significantly improved by adding nTreg/mTreg ratio to the conventional clinical parameters (continuous net reclassification improvement 91.26, 95% CI 69.04–113.5%, p

Background: Maple syrup urine disease (MSUD) is a rare autosomal recessive amino acid metabolic disease. This study is to identify the pathogenic genetic factors of six cases of MUSD and evaluates the application value of high-throughput sequencing technology in the early diagnosis of MUSD.Methods: Clinical examination was carried out for patients and used blood tandem mass spectrometry (MS/MS), urine gas chromatography-mass spectrometry (GC/MS), and the application of high-throughput sequencing technology for detection. Validate candidate mutations by polymerase chain reaction (PCR)—Sanger sequencing technology. Bioinformatics software analyzed the variants’ pathogenicity. Using Swiss PDB Viewer software to predict the effect of mutation on the structure of BCKDHA and BCKDHB proteins.Result: A total of six MSUD patients were diagnosed, including four males and two females. Nine variants were found in three genes of six MSUD families by high-throughput sequencing, including four missense mutations: c.659C>T(p.A220V), c.818C>T(p.T273I), c.1134C>G(p.D378E), and c.1006G>A(p.G336S); two non-sense mutations: c.1291C>T(p.R431*) and c.331C>T(p.R111*); three deletion mutations: c.550delT (p.S184Pfs*46), c.718delC (p.P240Lfs*14), and c.795delG (p.N266Tfs*64). Sanger sequencing’s results were consistent with the high-throughput sequencing. The bioinformatics software revealed that the mutations were harmful, and the prediction results of Swiss PDB Viewer suggest that variation affects protein conformation.Conclusion: This study identified nine pathogenic variants in the BCKDHA, BCKDHB, and DBT genes in six MSUD families, including two novel pathogenic variants in the BCKDHB gene, which enriched the genetic mutational spectrum of the disease. High-throughput sequencing is essential for the MSUD’s differential diagnosis, early treatment, and prenatal diagnosis.

Abstract Polymer electrolytes (PEs) have been long recognized as the key materials to enable energy‐dense batteries and render flexible energy devices practically viable, owing to their chemical and mechanical reliability. However, much of their promise is yet to be realized. The room‐temperature ion conductivity of existing PEs still falls short of the implementation criterion of 10−4 S cm−1 on the promise of acceptable mechanical properties, thereby precluding their practical application. The twin but inversely related duties of polymers, that is, functioning as both an ion‐conducting medium and a structural backbone, underlie this issue but are less elucidated systematically. The polyacrylate (PA) family is among promising polymer matrices on account of ester polarity, electrode compatibility, chemical tunability, and mechanical durability. The extensive applicability of PA in plasticized gels, dry solids, and emerging composites makes PA‐based PEs representative to illustrate the trade‐off between ion conduction and mechanical strength. We herein seek to outline the stated long‐standing conflict exemplified by PA‐based PEs, focusing on crucial strategies toward balancing and reconciling the two mutually exclusive properties, with the intention of offering designing guidelines for next‐generation PEs.

Traffic congestion has been a hot topic of research in the field of intelligent transportation, which can be alleviated by efficient route navigation. Most of the existing route planning methods are non-negotiated algorithms, which do not take into account the route conflicts and collaborative relationships between multiple vehicles. Also, most negotiated algorithms have not been comprehensively considered dynamic route collaboration between vehicles, large-scale efficient computation, environmental pollution, etc. Therefore, an ecological multivehicle real-time route selection model (EMR2SM) for urban road networks is firstly proposed in this paper, which combines real-time traffic conditions of the road network with travel time, distance, and exhaust emissions as optimization indicators. In order to solve the large-scale computation problem of traditional negotiated algorithms, an adaptive multiswarm bee colony (AMSBC) algorithm is designed, which efficiently solves the multivehicle dynamic route selection problem. AMSBC searches the optimal route for each vehicle in parallel through multiple population division and self-adaption mechanism, to make multivehicle route selection reach Nash equilibrium. Compared with three non-negotiated optimization algorithms based on swarm technology, EMR2SM is verified by experiments that it improves the efficiency and accuracy of the optimal route selection for multiple vehicles and reduces vehicle emissions, which can effectively reduce traffic congestion and environmental pollution.

Taxi sharing is a promising method to save resource consumption and alleviate traffic congestion while satisfying people’s commuting needs. Existing research methods include taxi dispatching methods based on intelligent algorithms, single vehicle route recommendation algorithms, and route recommendation algorithms based on vehicle traffic history. However, these studies either focus on how to efficiently dispatch satisfactory vehicles for passengers, ignoring the effect of efficient routes on vehicle travel efficiency, or let vehicles follow the shortest detour distance recommended by the system, ignoring the traffic congestion caused by the influx of vehicles into the same road. To address the abovementioned problems, a noncooperative game for a taxi sharing model (NCG-TSM) in urban road networks is proposed in this paper combined with the traffic conditions of the optional routes, and a distribution estimation algorithm for the shared taxi game is designed to make multivehicle route selections reach Nash equilibrium. The effectiveness of NCG-TSM is verified through simulation experiments. When the number of vehicles reaches the congestion capacity of the road segment, compared to the three common frameworks, the travel time cost and fuel consumption cost can be reduced by 5.8% to 9.1% and 3.5% to 8.9%, respectively. Besides, the occupancy rate has been improved, especially compared to the BMP framework, by 5.5% to 40%.

IntroductionHypermucoviscous Klebsiella pneumoniae (HmKp) poses an emerging and highly pathogenic global health threat. This study aimed to investigate the clinical and genomic characteristics of HmKp isolates to better understand the virulence mechanisms of the hypermucoviscous (HMV) phenotype.MethodsFrom May 2018 to August 2021, 203 non-repeat K. pneumoniae isolates causing invasive infections were collected from a hospital in Beijing, China. Isolates were divided into HmKp (n=90, 44.3%) and non-HmKp (n=113, 55.7%) groups according to string test results.ResultsMultivariate regression showed that diabetes mellitus (odds ratio [OR]=2.20, 95% confidence interval (CI): 1.20-4.05, p=0.010) and liver abscess (OR=2.93, CI 95%:1.29-7.03, p=0.012) were associated with HmKp infections. K. pneumoniae was highly diverse, comprising 87 sequence types (STs) and 54 serotypes. Among HmKp isolates, ST23 was the most frequent ST (25/90, 27.8%), and the most prevalent serotypes were KL2 (31/90, 34.4%) and KL1 (27/90, 30.0%). As a life-essential coenzyme, nicotinamide adenine dinucleotide (NAD+) has been explored for more than a century. In Saccharomyces, the natural NAD+de novo biosynthetic pathway initiating from tryptophan has been well elucidated. To bypass this stringently controlled natural pathway in yeast, an economical C3N pathway that was developed in Escherichia coli previously was constructed in Saccharomyces as a short detour for de novo NAD+ biosynthesis. After the functional expressions of the C3N genes were identified in Saccharomyces cerevisiae BY4741 by in vitro enzymatic assays, the C3N module was introduced into an NAD+ auxotrophic S. cerevisiae strain BY01, in which the BNA2 gene encoding tryptophan 2,3-dioxygenase was inactivated. The efficient NAD+ synthesis via the C3N pathway was confirmed by both plate assays and fermentation analysis. The applicability of the C3N pathway in cofactor engineering was tested by introducing it into S. cerevisiae BY4741, which improved the cellular NAD(H) level considerably. Consequently, this study proved that the de novo NAD+ biosynthetic pathway can be replaced by an artificial pathway in yeast, which paves a way to design more promising schemes in eukaryotes for rational manipulation of the cellular NAD(H) levels.

This paper proposes an improved frequency domain turbo equalization (IFDTE) with iterative channel estimation and feedback to achieve both a good performance and low complexity in underwater acoustic communications (UWACs). A selective zero-attracting (SZA) improved proportionate normal least mean square (SZA-IPNLMS) algorithm is adopted by utilizing the sparsity of the UWAC channel to estimate it using a training sequence. Simultaneously, a set-membership (SM) SZA differential IPNLMS (SM SZA-DIPNLMS) with variable step size is adopted to estimate the channel status information (CSI) in the iterative channel estimation with soft feedback. In this way, the computational complexity for iterative channel estimation is reduced effectively with minimal performance loss. Different from traditional schemes in UWACs, an IFDTE with expectation propagation (EP) interference cancellation is adopted to estimate the a posteriori probability of transmitted symbols iteratively. A bidirectional IFDTE with the EP interference cancellation is proposed to further accelerate the convergence. THe simulation results show that the proposed channel estimation obtains 1.9 and 0.5 dB performance gains, when compared with those of the IPNLMS and the l0-IPNLMS at a bit error rate (BER) of 10−3. The proposed channel estimation also effectively reduces the unnecessary updating of the coefficients of the UWAC channel. Compared with traditional time-domain turbo equalization and FDTE in UWACs, the IFDTE obtains 0.5 and 1 dB gains in the environment of SPACE’08 and it obtains 0.5 and 0.4 dB gains in the environment of MACE’04 at a BER of 10−3. Therefore, the proposed scheme obtains a good BER performance and low complexity and it is suitable for efficient use in UWACs.

As a significant component of rural settlements, residential architecture is a record of historical changes containing considerable research value. In the study of residential architectural continuity, the focus is on the inheritance and innovation of traditional residential architectural “genes” in contemporary new residential buildings. Based on a systematic review of the literature, the purpose of this study is to analyze the research trends, categories, and variables relating to architectural continuity in residential buildings, and to build a systematic and comprehensive framework for assessing the architectural continuity of residential buildings in rural settlements based on prior research. This study provides guidance and references for evaluating the design of new residential buildings in rural settlements and for formulating regional planning principles. Using the PRISMA guidelines as the basis for the review method, we filtered the literature from three databases: Web of Science (WoS), Scopus, and EI, and studied the 40 articles selected at the end. As can be seen from the results, the literature on architectural continuity in rural settlements has focused more on functional and typological levels and less on the archetypal level of architectural continuity (user behavior). Still, the archetypal level is the most important, because the behavior of housing users directly influences the degree to which housing is continuous in terms of its function and type. The most important finding of this review is that the resident behavior of housing users has a significant influence on the assessment of the continuity of housing architecture, and thus, the continuity of housing architecture in rural settlements must be assessed in conjunction with user behavior.

A treetop trail is an elevated linear green open space that plays a key role in forming a scientifically rational urban space and meeting the growing leisure needs of the people. Taking the Mountains-to-Sea Trail in Xiamen, China as a case, and through 426 questionnaires, this study explores the dimensions of the perceived restorative environment components of greenway recreationists and impacts on behavioral intentions. The demographic factors lead us to the following three conclusions. First, from an age perspective, restorative environmental perceptions are strongest among those aged 60 and above and weakest among those aged 18–30. Second, in terms of place of permanent residence, local visitors have stronger restorative environmental perceptions than other city users. Third, in relation to the number of accompanying travelers, individuals who embark on solo journeys experience the most robust perception, while that diminishes as the count reaches three or more companions. A structural equation model (SEM) is used to present the quantitative relationship among avoidance motivation, treetop trail environmental quality, restorative environmental perception, place attachment, and loyalty. The results showed that users’ escape motivation has a direct and indirect positive correlation with restorative environmental perceptions, and environmental perceptions have a significant positive correlation with restorative environmental perceptions. Furthermore, their place attachment to the restorative nature of the treetop trails positively affected their loyalty. This study provides essential factors to consider when constructing treetop trails in high-density cities.

Accurate prediction of urban residential rents is of great importance for landlords, tenants, and investors. However, existing rents prediction models face challenges in meeting practical demands due to their limited perspectives and inadequate prediction performance. The existing individual prediction models often lack satisfactory accuracy, while ensemble learning models that combine multiple individual models to improve prediction results often overlook the impact of spatial heterogeneity on residential rents. To address these issues, this paper proposes a novel prediction model called GERPM, which stands for Geographically Weighted Stacking Ensemble Learning-Based Urban Residential Rents Prediction Model. GERPM comprehensively analyzes the influencing factors of residential rents from multiple perspectives and leverages a geographically weighted stacking ensemble learning approach. The model combines multiple machine learning and deep learning models, optimizes parameters to achieve optimal predictions, and incorporates the geographically weighted regression (GWR) model to consider spatial heterogeneity. By combining the strengths of deep learning and machine learning models and taking into account geographical factors, GERPM aims to improve prediction accuracy and provide robust predictions for urban residential rents. The model is evaluated using housing data from Nanjing, a major city in China, and compared with representative individual prediction models, the equal weight combination model, and the ensemble learning model. The experimental results demonstrate that GERPM outperforms other models in terms of prediction performance. Furthermore, the model’s effectiveness and robustness are validated by applying it to other major cities in China, such as Shanghai and Hangzhou. Overall, GERPM shows promising potential in accurately predicting urban residential rents and contributing to the advancement of the rental market.

Bromhidrosis has a great negative impact on personal occupation and social psychology. It is not yet clear whether bromhidrosis is caused by apocrine sweat glands or the co-action of apocrine sweat glands and eccrine sweat glands. To distinguish between apocrine sweat glands and eccrine sweat glands, specific antigen markers for apocrine sweat glands and eccrine sweat glands must be found first. In the study, we detected the expression of K7, K18, K19, Na+-K+-2Cl- cotransporter 1 (NKCC1), carbonic anhydrase II (CAII), Forkhead transcription factor a1 (Foxa1), homeobox transcription factor engrailed homeobox1 (En1), gross cystic disease fluid protein-15 (GCDFP-15), mucin-1 (MUC-1), cluster of differentiation 15 (CD15) and apolipoprotein (APOD) in eccrine sweat glands and apocrine sweat glands by immunofluorescence staining. The results showed that K7, K18, K19, Foxa1, GCDFP-15 and MUC-1 were expressed in both apocrine and eccrine sweat glands, CD15 and APOD were only expressed in apocrine sweat glands, and CAII, NKCC1 and En1 were only expressed in eccrine sweat glands. We conclude that CD15 and APOD can serve as specific markers for apocrine sweat glands, while CAII, NKCC1 and En1 can serve as specific markers for eccrine sweat glands to differentiate the two sweat glands.
