While exosomes derived from human umbilical cord mesenchymal stem cells (HucMSC-Exo) have emerged as a promising therapeutic tool, their exact mechanisms of action remain partly elusive. We hypothesize that HucMSC-Exo treatment may modulate neutrophil phenotypes, thereby significantly influencing wound healing outcomes.Methods: HucMSC-Exo were isolated via ultracentrifugation and subsequently administered through subcutaneous injection into full-thickness cutaneous wounds in mice. To determine the impact of host neutrophils on the healing effects of HucMSC-Exo in skin injuries, strategies including neutrophil depletion and adoptive transfer were employed. Flow cytometry was used to evaluate the proportion of N2 subtype neutrophils in both normal and diabetic wounds, and the effect of HucMSC-Exo on this proportion was assessed. Furthermore, the mitochondrial metabolic reprogramming driven by HucMSC-Exo during N2 polarization was investigated through JC1 staining, ATP quantification, fatty acid uptake assays, and assessment of FAO-related genes (Cpt1b, Acadm, and Acadl).Results: Depleting host neutrophils strikingly dampened prohealing effect of HucMSC-Exo on skin injury, while adoptive transfer of bone marrow neutrophils rescued this process. During normal healing process, some neutrophils expressed N2 markers, in contrast, diabetic wounds exhibited a reduced expression of N2 markers. After treatment with HucMSC-Exo, most neutrophils increased the phosphorylation of STAT6, leading to mitochondrial metabolic reprogramming and thus acquired an N2 phenotype. These N2 neutrophils, polarized by HucMSC-Exo, boosted the release of proangiogenic factors, particularly BV8, a myeloid cell-derived proangiogenic factor, and induced angiogenesis thereby favoring tissue restoration.Conclusion: This research uniquely demonstrates the identification of N2 neutrophils in skin injury and shows that HucMSC-Exo could skew neutrophils toward N2 phenotype, enhancing our insight into how cells react to HucMSC-Exo. However, the underlying pathogenesis remains unclear and there is a dearth of reliable diagnostic biomarkers.Objective: The aim of this study was to identify diagnostic biomarkers for SLs and reveal its immunological features.Methods: In this study, gene expression profiling datasets (GSE192564 and GSE192565) of SLs were obtained from the GEO database. The GSE192564 was used as the training group for screening of differentially expressed genes (DEGs) and subsequent depth analysis. Gene set enrichment analysis (GSEA) was employed to explore the biological states associated with SLs. The weighted gene co-expression network analysis (WGCNA) was employed to identify the significant modules and hub genes. Then, the feature genes were further screened by the overlapping of hub genes and up-regulated differential genes. Subsequently, an artificial neural network was constructed for identifying SLs samples. The GSE192565 was used as the test group for validation of feature genes expression level and the model’s classification performance. Furthermore, we conducted immune cell infiltration analysis to reveal the immune infiltration landscape of SLs.Results: The 9 feature genes were identified as diagnostic biomarkers for SLs in this study. And an artificial neural network based on diagnostic biomarkers was successfully constructed for identification of SLs. GSEA highlighted potential role of immune system in pathogenesis of SLs. SLs samples had a higher proportion of several immune cells, including activated CD8 T cell, dendritic cell, myeloid-derived suppressor cell and so on. And diagnostic biomarkers exhibited a strong relationship with the infiltration of most immune cells.Conclusion: Our study identified diagnostic biomarkers for SLs and explored its immunological features, enhancing the comprehension of its pathogenesis.Keywords: solar lentigines, photoaging, diagnostic biomarkers, immune infiltration, artificial neural network

It means that this property of platelets can be used in anti-tumor therapy. However, traditional platelet-based antitumor drugs often cause autologous platelet damage due to lack of targeting, resulting in serious side effects. Therefore, the researchers designed a variety of anti-tumor drug delivery systems based on platelets by targeting platelets or platelet membrane coating. The drug delivery systems have special response modes, which is crucial in the design of nanoparticles. These modes enhance the targeting and improve the anti-tumor effect. The patients were randomly assigned to two groups: the control group receiving routine care and the observation group receiving seamless care based on action research. The random assignment was conducted using a simple random sampling technique, ensuring an equal allocation of participants to each group at a 1:1 ratio, resulting in 40 cases in each group. Outcome measures included imaging examination duration, mean nursing duration, examination cognition, and negative emotion scores.Results: Seamless care provided shorter imaging examination duration and nursing duration, and better ensured uneventful examinations than routine care (P< 0.05). Patients given seamless care exhibited higher examination cognition versus those receiving routine care (P< 0.05). Seamless care offered more mitigation of negative emotions for patients than routine care (P< 0.05).Conclusion: Action research-based seamless care effectively improves imaging efficiency and patients’ awareness of imaging examinations and contributes to alleviating patients’ adverse events.Keywords: action research, seamless care, imaging, anxiety

The effects of different fungal agents on the bacterial community structure and biodiversity in the rhizosphere soil of wheat (Triticum aestivum) infected with cereal cyst nematodes (Heterodera avenae) were analyzed. Field experiments were conducted on plots with severe wheat cyst nematode disease. The control group (X1) was not treated with bacteria. The fungi Trichoderma viride, Paecilomyces lilacinus, Aspergillus niger and T. spinosum were applied to the experimental treatments (X2~X4). The experimental group (X6) was treated with the nematicide abamectin. Illumina MiSeq high-throughput sequencing technology was used to monitor the changes in bacterial community structure and metabolic types in the wheat rhizosphere soil in the six experimental groups. The results of high-throughput sequencing showed that the wheat rhizosphere soil bacteria that are pathogens were primarily concentrated in 13 phyla, 26 classes and 98 genera. Among them, the dominant bacteria primarily included Proteus, Actinomyces, Acetobacter and Bacteroides. The analysis of bacterial community metabolic type showed that the bacteria in the wheat rhizosphere soil could fix nitrogen, perform nitrification, reduce nitrate, and photosynthesize when the four groups of fungal agents were added at much higher levels than those of the control group. These metabolic activities contributed to promoting the cycle between nitrate nitrogen and ammonium nitrogen to increase soil fertility. [ABSTRACT FROM AUTHOR]

The urban three-dimensional (3-D) model, which can truly and vividly express the 3-D spatial information of ground objects, was an essential link in the process of smart city construction. The backpack 3-D laser scanning systems had gradually become an important technical means to build urban 3-D models because of their high efficiency and high fineness. In this paper, the error source of backpack point cloud data was analysed based on the composition of backpack 3-D laser scanning system. The distribution of point cloud error was statistically analysed by designing experiments. It was found that there was a systematic error in the backpack 3-D laser scanning point cloud. There were many factors influencing the system errors and they were superimposed on each other. In order to solve this problem, an error compensation model with additional system parameters based on a multiple linear regression algorithm was constructed. The results showed that this model can weaken the influence of system errors on the backpack point cloud, effectively improve the accuracy of the backpack point cloud, and provide reliable point cloud data for subsequent processing of the building point cloud. Targeted inhibition of Hedgehog signaling at the cell membrane has been associated with anticancer activity in preclinical and early clinical studies. Hedgehog signaling involves activation of Gli transcription factors that can also be induced by alternative pathways. In this study, we identified an interaction between Gli proteins and a transcription coactivator TBP-associated factor 9 (TAF9), and validated its functional relevance in regulating Gli transactivation. We also describe a novel, synthetic small molecule, FN1-8, that efficiently interferes with Gli/TAF9 interaction and downregulate Gli/TAF9-dependent transcriptional activity. More importantly, FN1-8 suppresses cancer cell proliferation in vitro and inhibits tumor growth in vivo. Our results suggest that blocking Gli transactivation, an important control point of multiple oncogenic pathways, may be an effective anticancer strategy.

There are numerous potential causes, with food allergy being the leading cause in children and the focus of this review. Most reactions involve an IgE-mediated mechanism, although non-IgE-mediated and nonimmunologic reactions can occur. Various cofactors to be discussed can place certain individuals at an increased risk of severe or fatal anaphylaxis. The clinical manifestations of anaphylaxis are broad and may involve multiple body systems. Diagnosis of food-related anaphylaxis is primarily based on signs and symptoms and supported, wherever possible, by identification and confirmation of a culprit food allergen. First-line treatment of anaphylaxis is intramuscular administration of epinephrine. Long-term management is generally focused on strict allergen avoidance and more recently on food desensitization using immunotherapy.
