Abstract Allogeneic platelet-rich plasma (al-PRP) is gaining attention in clinical practice for treating chronic refractory wounds, though research results remain controversial. To assess the clinical efficacy of al-PRP for chronic refractory wounds. Databases including PubMed, Cochrane Library, Embase, CNKI, SinoMed, VIP, and WFPD were searched for randomized controlled trials comparing al-PRP with conventional treatments up to October 2023. Two researchers independently screened studies, extracted data, and assessed quality. Statistical analysis was conducted using RevMan 5.4, and potential publication bias was assessed and corrected using funnel plots and Egger’s test. Twelve studies with 717 cases were included. Abstract Total knee arthroplasty (TKA) is the most cost-effective, and potent method for the treatment of end-stage knee osteoarthritis. Acute normovolemic haemodilution (ANH) can effectively replace the need for allogeneic transfusions due to the high amount of bleeding during TKA. However, more studies are needed to prove the efficacy and safety of ANH and to clarify its indications in the field of knee replacement. Medical records from June 1, 2019 to June 1, 2021 were searched and grouped according to inclusion and exclusion criteria. PART I: 58 patients with ANH during TKA were selected as the ANH group (n = 58), and 58 patients with allogeneic transfusion were chosen as the control group (n = 58). PART II: Patients with anaemia were divided into the ANH group (n = 18) and the control group (n = 12). PART I: The postoperative inflammatory index and serum albumin in the ANH group were significantly lower than those in the control group. No significant difference was observed in the theoretical loss of red blood cells, postoperative renal function, liver function, cardiac function and biochemical ion index between the two groups. The effective rate of ANH in the normal haemoglobin group was significantly lower than that in the anaemia group. PART II: In patients with anaemia, the theoretical loss of red blood cells in patients with ANH was less than that in the control group. The postoperative inflammation, renal function, liver function and cardiac function in the ANH group were better than those in the control group, and no significant difference was noted in biochemical ions and nutritional status indicators. This paper shows that ANH not only can replace allogeneic transfusion in TKA, especially in patients with anaemia, but also has lower inflammatory indicators than allogeneic transfusion. From a security perspective, the body’s tolerance to ANH is within the body’s compensation range.

Lacking self-repair abilities, injuries to articular cartilage can lead to cartilage degeneration and ultimately result in osteoarthritis. Tissue engineering based on functional bioactive scaffolds are emerging as promising approaches for articular cartilage regeneration and repair. Although the use of cell-laden scaffolds prior to implantation can regenerate and repair cartilage lesions to some extent, these approaches are still restricted by limited cell sources, excessive costs, risks of disease transmission and complex manufacturing practices. Acellular approaches through the recruitment of endogenous cells offer great promise for in situ articular cartilage regeneration. In this study, we propose an endogenous stem cell recruitment strategy for cartilage repair. Based on an injectable, adhesive and self-healable o-alg-THAM/gel hydrogel system as scaffolds and a biophysio-enhanced bioactive microspheres engineered based on hBMSCs secretion during chondrogenic differentiation as bioactive supplement, the as proposed functional material effectively and specifically recruit endogenous stem cells for cartilage repair, providing new insights into in situ articular cartilage regeneration.

C-terminal binding protein (CtBP), a transcriptional co-repressor, significantly influences cellular signaling, impacting various biological processes including cell proliferation, differentiation, apoptosis, and immune responses. The CtBP family comprises two highly conserved proteins, CtBP1 and CtBP2, which have been shown to play critical roles in both tumorigenesis and the regulation of viral infections. Elevated CtBP expression is noted in various tumor tissues, promoting tumorigenesis, invasiveness, and metastasis through multiple pathways. Additionally, CtBP’s role in viral infections varies, exhibiting differing or even opposing effects depending on the virus. This review synthesizes the advances in CtBP’s function research in viral infections and virus-associated tumorigenesis, offering new insights into potential antiviral and anticancer strategies.

Functional tissue engineering strategies provide innovative approach for the repair and regeneration of damaged cartilage. Hydrogel is widely used because it could provide rapid defect filling and proper structure support, and is biocompatible for cell aggregation and matrix deposition. Efforts have been made to seek suitable scaffolds for cartilage tissue engineering. Here Alg-DA/Ac-β-CD/gelatin hydrogel was designed with the features of physical and chemical multiple crosslinking and self-healing properties. Gelation time, swelling ratio, biodegradability and biocompatibility of the hydrogels were systematically characterized, and the injectable self-healing adhesive hydrogel were demonstrated to exhibit ideal properties for cartilage repair. Furthermore, the new hydrogel design introduces a pre-gel state before photo-crosslinking, where increased viscosity and decreased fluidity allow the gel to remain in a semi-solid condition. This granted multiple administration routes to the hydrogels, which brings hydrogels the ability to adapt to complex clinical situations. Pulsed electromagnetic fields (PEMF) have been recognized as a promising solution to various health problems owing to their noninvasive properties and therapeutic potentials. PEMF treatment offers a better clinical outcome with fewer, if any, side effects, and wildly used in musculoskeletal tissue repair. Thereby we propose PEMF as an effective biophysical stimulation to be 4th key element in cartilage tissue engineering. In this study, the as-prepared Alg-DA/Ac-β-CD/gelatin hydrogels were utilized in the rat osteochondral defect model, and the potential application of PEMF in cartilage tissue engineering were investigated. PEMF treatment were proven to enhance the quality of engineered chondrogenic constructs in vitro, and facilitate chondrogenesis and cartilage repair in vivo. All of the results suggested that with the injectable self-healing adhesive hydrogel and PEMF treatment, this newly proposed tissue engineering strategy revealed superior clinical potential for cartilage defect treatment.

The development of open-source geometric constraint solvers is a pressing research topic, as commercially available solvers may not meet the research requirements. In this paper, we examine the use of numerical methods in PlaneGCS, an open-source geometric constraint solver within the FreeCAD CAD software. Our study focuses on PlaneGCS's constraint solving algorithms and the three built-in single-subsystem solving methods: BFGS, LM, and Dogleg. Based on our research results, the DFP method was implemented in PlaneGCS and was successfully verified in FreeCAD. To evaluate the performance of the algorithms, we used the solving state of the constraint system as a test criterion, and analysed their solving time, adaptability, and number of iterations. Our results highlight the performance differences between the algorithms and provide empirical guidance for selection of constraint solving algorithms and research based on open-source geometric constraint solvers.

The eastern Sichuan Basin in China holds vast potential for oil and gas exploration in the Lower-Middle Jurassic strata. However, the geological characteristics and hydrocarbon accumulation patterns of this region remain largely unclear. During the deposition period of the Lower-Middle Jurassic strata, the eastern Sichuan is characterized by the formation of multiple sets of source, reservoir, and caprock assemblages through depositing lake-delta-fluvial deposits, which have great exploration potential. The Jurassic source rocks in eastern Sichuan are mainly developed in the Dongyuemiao Member, Da’anzhai Member, and Liangshan Formation. These source rocks have a total organic carbon (TOC) content greater than 1 and a varying range of organic matter maturity, with a Ro value of 0.8–2.0. The kerogen in these source rocks is primarily type II, with a smaller proportion being type III. A range of reservoir rocks can be found in the Jurassic strata of eastern Sichuan, with sandstone reservoirs being predominantly found in the Liangshan Formation, Shaximiao Formation, and Zhenzhuchong Member. Shale reservoirs are mostly present in the Dongyuemiao, Da’anzhai, Liangshan, and Maanshan Members, and there is a limited distribution of limestone reservoirs in the Da’anzhai Member and Dongyuemiao Formation. The arrangement of source rocks and reservoir rocks in eastern Sichuan has led to the formation of three types of reservoir-forming combinations, including lower generation and upper storage, self-generation and self-storage, and composite. Sandstone reservoirs are typically of lower generation and upper storage, shale reservoirs are primarily of self-generation and self-storage, and limestone reservoirs are mostly composite. The exploration of Jurassic oil and gas in eastern Sichuan should prioritize “layer and area selections.” The Da’anzhai, Dongyuemiao, and Liangshan shale reservoirs should be the primary exploration targets, with the semi-deep lake deposits in the syncline area being the most favorable. The degree of fracture development in the exploration area also has a significant impact on the shale oil and gas content. The Liangshan Formation and Shaximiao Formation sandstone reservoirs can serve as secondary exploration targets, with anticline areas that have better sealing conditions being more favorable. Limestone reservoirs have limited distribution, and exploration areas with high and steep fractures are relatively more advantageous.

Coronal mass ejections (CMEs) constitute the major source of severe space weather events, with the potential to cause enormous damage to humans and spacecraft in space. It is becoming increasingly important to detect and track CMEs, since there are more and more space activities and facilities. We have developed a new algorithm to automatically derive a CME’s kinematic parameters based on machine learning. Our method consists of three steps: recognition, tracking, and the determination of parameters. First, we train a convolutional neural network to classify images from Solar and Heliospheric Observatory Large Angle Spectrometric Coronagraph observations into two categories, containing CME(s) or not. Next, we apply the principal component analysis algorithm and Otsu’s method to acquire binary-labeled CME regions. Then, we employ the track-match algorithm to track a CME’s motion in time-series images and finally determine the CME’s kinematic parameters, e.g., velocity, angular width, and central position angle. The results of four typical CME events with different morphological characteristics are presented and compared with a manual CME catalog and several automatic CME catalogs. Our algorithm shows some advantages in the recognition of CME structure and the accuracy of the kinematic parameters. This algorithm can be helpful for real-time CME warnings and predictions. In the future, this algorithm is capable of being applied to CME initialization in magnetohydrodynamic simulations to study the propagation characteristics of real CME events and to provide more efficient predictions of CMEs’ geoeffectiveness.

Osteoarthritis (OA) is the most common joint disease; thus, understanding the pathological mechanisms of OA initiation and progression is critical for OA treatment. MicroRNAs (miRNAs) have been shown to be involved in the progression of osteoarthritis, one candidate is microRNA-378 (miR-378), which is highly expressed in the synovium of OA patients during late-stage disease, but its function and the underlying mechanisms of how it contributes to disease progression remain poorly understood. In this study, miR-378 transgenic (TG) mice were used to study the role of miR-378 in OA development. miR-378 TG mice developed spontaneous OA and also exaggerated surgery-induced disease progression. Upon in vitro OA induction, miR-378 expression was upregulated and correlated with elevated inflammation and chondrocyte hypertrophy. Chondrocytes isolated from articular cartilage from miR-378 TG mice showed impaired chondrogenic differentiation. The bone marrow mesenchymal stem cells (BMSCs) collected from miR-378 TG mice also showed repressed chondrogenesis compared with the control group. The autophagy-related protein Atg2a, as well as chondrogenesis regulator Sox6, were identified as downstream targets of miR-378. Ectopic expression of Atg2a and Sox6 rescued miR-378-repressed chondrocyte autophagy and BMSC chondrogenesis, respectively. Anti-miR-378 lentivirus intra-articular injection in an established OA mouse model was shown to ameliorate OA progression, promote articular regeneration, and repress hypertrophy. Atg2a and Sox6 were again confirmed to be the target of miR-378 in vivo. In conclusion, miR-378 amplified OA development via repressing chondrocyte autophagy and by inhibiting BMSCs chondrogenesis, thus indicating miR-378 may be a potential therapeutic target for OA treatments.

Chronic skin wounds impose immense suffers and economic burdens. Current research mainly focuses on acute wound management which exhibits less effective in chronic wound healing. Growth differentiation factor 11 (GDF11) has profound effects on several important physiological processes related to chronic wound healing, such as inflammation, cell proliferation, migration, angiogenesis, and neurogenesis. This review summarizes recent advances in biology of chronic wounds and the potential role of GDF11 on wound healing with its regenerative effects, as well as the potential delivery methods of GDF11. The challenges and future perspectives of GDF11-based therapy for chronic wound care are also discussed.The Translational Potential of this Article: This review summarized the significance of GDF11 in the modulation of inflammation, vascularization, cell proliferation, and remodeling, which are important physiological processes of chronic wound healing. The potential delivery methods of GDF11 in the management of chronic wound healing is also summarized. This review may provide potential therapeutic approaches based on GDF11 for chronic wound healing.

Aims: Treatment for delayed wound healing resulting from peripheral vascular diseases and diabetic foot ulcers remains a challenge. A novel surgical technique named ‘tibial cortex transverse transport’ (TTT) has been developed for treating peripheral ischaemia, with encouraging clinical effects. However, its underlying mechanisms remain unclear. In the present study, we explored the potential biological mechanisms of TTT surgery using various techniques in a rat TTT animal model. Methods: A novel rat model of TTT was established with a designed external fixator, and effects on wound healing were investigated. Laser speckle perfusion imaging, vessel perfusion, histology, and immunohistochemistry were used to evaluate the wound healing processes. Results: Gross and histological examinations showed that TTT technique accelerated wound closure and enhanced the quality of the newly formed skin tissues. In the TTT group, haematoxylin and eosin (H&E) staining demonstrated a better epidermis and dermis recovery, while immunohistochemical staining showed that TTT technique promoted local collagen deposition. The TTT technique also benefited to angiogenesis and immunomodulation. In the TTT group, blood flow in the wound area was higher than that of other groups according to laser speckle imaging with more blood vessels observed. Enhanced neovascularization was seen in the TTT group with double immune-labelling of CD31 and α-Smooth Muscle Actin (α-SMA). The number of M2 macrophages at the wound site in the TTT group was also increased. Conclusion: The TTT technique accelerated wound healing through enhanced angiogenesis and immunomodulation. Due to the industry knowledge barrier, general computer aided design (CAD) software cannot do everything in digital manufacturing by itself, and industry CAD, therefore, occupies a crucial position in the CAD industry. To develop industry CAD smoothly, open-source is the best choice. We analyzed recent examples of industry CAD development and divided the development methods into four types: development based on the graphics development environment, development based on geometric modelling kernel, secondary development based on general CAD, and hybrid development. We analyzed the characteristics of various methods and believe that the method based on the hybrid development of the geometric modelling kernel and the graphics development environment is the best open-source industry CAD development method. We proposed a system architecture of open-source industry CAD for reference and conducted a preliminary exploration of the reference architecture to verify its feasibility.

Sand bodies in the Jurassic lacustrine-fluvial sedimentary system in the Sichuan Basin are realistic objects of shallow tight oil and gas exploration and development. Due to the influence of sedimentary microfacies, sand bodies have geological characteristics such as type diverse, variable lithology, significant thickness variation, and strong heterogeneity. Therefore, the fine description and prediction of different types of sand bodies is the current bottleneck for exploration and development. Based on the classification of various sand group types of the 1st member of Jurassic Shaximiao Formation, reservoir analysis was carried out, logging and geophysical response characteristics were concluded, and fine identification and spatial prediction were made by time-sharing window. Results show that: (1) The 1st member of Shaximiao Formation can be divided into 5 sand groups, among which the 1st sand group is mainly littoral-shallow lacustrine sheet sand and beach bar sand, and the 2nd to 5th sand groups are delta front facies channel-plain channel; (2) The average porosity of channel sand is 4.68%, which is significantly higher than that of bottom sheet sand (3.1%). The physical properties of the Ⅰ channel are the best, followed by the Ⅳ, Ⅱ, and Ⅲ channels. (3) The prediction standard of channel sand is: natural gamma < 65 API, and the prediction standard of sheet sand locates between 65 and 75 API. In sand body prediction, for sheet sands (beach bars), the study time window is extracted to be within 0-20 ms, and for river channels, the study time window is in the range of 20-100 ms upwards from the bottom of the Shaximiao Formation. Finally, a set of methods suitable for fine description and prediction of different types of sand bodies in the lacustrine-fluvial sedimentary system was established.

Background In mainland China, HPV vaccines have been available to the public. However, only a few related studies among health care providers, as the key information providers, were reported although public concerns on HPV vaccines still exist. In this study, we aim to assess the knowledge of HPV, its vaccines, and attitudes toward HPV vaccines among the three most important groups of health care providers in Western China. Method This was a cross-sectional questionnaire-based study. Health care providers including obstetrician-gynecologists (OB-GYNs), pediatricians, and immunization service providers in Western China were investigated regarding their knowledge of HPV and its vaccines and their attitudes toward HPV vaccines. Results Of 1079 health care providers completing the survey, 1015 (94.1%) knew HPV infection is the primary cause of cervical cancer. However, lower knowledge levels of other HPV-related diseases were also found (43.2%). About three-quarters (74.1%) of practitioners interviewed would be willing to recommend HPV vaccination, which was found to be lower among the OB-GYNs (69.6%) and the pediatricians (73.2%). “Lack of relevant knowledge,” “concerns on safety and efficacy” and price were the three most important concerns surrounding HPV vaccination. Conclusion The interviewed practitioners did not have adequate knowledge of HPV and its vaccines in depth. Education interventions are highly recommended to the health care providers, especially for OB-GYNs and pediatricians, to increase the coverage of HPV vaccination among the population. For the currently high price of vaccines, a future co-sharing mechanism between the government, the providers, and the individuals might be a solution.

The Jurassic Da’anzhai Member in Sichuan Basin is an important target for shale oil exploration. Whole rock X-ray diffraction analysis, thin section, SEM and TOC testing, as well as major and trace element analysis are used to analyze the relationship of lithofacies and pore types with the sedimentary environment of the Da’anzhai Member in the Central Sichuan Basin. The results show the Da’anzhai Member in the Well Ren’an 1 (RA1) is divided into six types of lithofacies, based on a three-level division method of mineral composition-TOC-mineral structure— namely, massive mud-bearing shell limestone lithofacies (F), thick argillaceous shell limestone lithofacies (E), laminated clay silt shale lithofacies (D), thin shell calcareous shale lithofacies (C), laminated shell-bearing shale lithofacies (B), and thick clay shale lithofacies (A). The pore spaces include intergranular pores, intergranular dissolution pores, authigenic calcite intergranular pores, authigenic quartz intercrystalline pores, intergranular pores of clay minerals or pyrite nodules, as well as organic pores. The Da’anzhai Member was deposited in a semi-deep to shallow lake sedimentary environment, with relatively low salinity. This strata experienced moderate-to-strong chemical weathering as a result of the semi-arid, warm and humid paleoclimate environmental conditions that persisted during its deposition; low weathering and dry paleoclimate conditions occurred periodically. Paleoenvironment is an important influence on the lithofacies and pore types of the Da’anzhai Member. The persistence of arid climate conditions contributed to a decline in water depth, relatively high salinity, low organic productivity, and the deposition of carbonate rocks that characterize lithofacies E and F; mostly intercrystalline pores formed in these lithofacies in response to the prevailing arid climate conditions. Conversely, warm, and humid climate conditions increased the inflow of fresh water, terrigenous debris, and nutrients into the lacustrine basin that led to the deposition of fine-grained sedimentary rocks of lithofacies A, B, and C. These lithofacies are characterized by abundant nano pores, such as clay intercrystalline pores and organic pores.

Aims: Distraction osteogenesis (DO) is a useful orthopaedic procedure employed to lengthen and reshape bones by stimulating bone formation through controlled slow stretching force. Despite its promising applications, difficulties are still encountered. Our previous study demonstrated that pulsed electromagnetic field (PEMF) treatment significantly enhances bone mineralization and neovascularization, suggesting its potential application. The current study compared a new, high slew rate (HSR) PEMF signal, with different treatment durations, with the standard Food and Drug Administration (FDA)-approved signal, to determine if HSR PEMF is a better alternative for bone formation augmentation. Methods: The effects of a HSR PEMF signal with three daily treatment durations (0.5, one, and three hours/day) were investigated in an established rat DO model with comparison of an FDA-approved classic signal (three hrs/day). PEMF treatments were applied to the rats daily for 35 days, starting from the distraction phase until termination. Radiography, micro-CT (μCT), biomechanical tests, and histological examinations were employed to evaluate the quality of bone formation. Results: All rats tolerated the treatment well and no obvious adverse effects were found. By comparison, the HSR signal (three hrs/day) treatment group achieved the best healing outcome, in that endochondral ossification and bone consolidation were enhanced. In addition, HSR signal treatment (one one hr/day) had similar effects to treatment using the classic signal (three three hrs/day), indicating that treatment duration could be significantly shortened with the HSR signal. Conclusion: HSR signal may significantly enhance bone formation and shorten daily treatment duration in DO, making it a potential candidate for a new clinical protocol for patients undergoing DO treatments. Abstract Achieving robust underwater adhesion by bioadhesives remains a challenge due to interfacial water. Herein a coacervate‐to‐hydrogel strategy to enhance interfacial water repulsion and bulk adhesion of bioadhesives is reported. The polyethyleneimine/thioctic acid (PEI/TA) coacervate is deposited onto underwater substrates, which can effectively repel interfacial water and completely spread into substrate surface irregularities due to its liquid and water‐immiscible nature. The physical interactions between coacervate and substrate can further enhance interfacial adhesion. Furthermore, driven by the spontaneous hydrophobic aggregation of TA molecules and strong electrostatic interaction between PEI and TA, the coacervate can turn into a hydrogel in situ within minutes without additional stimuli to develop enhanced matrix cohesion and robust bulk adhesion on diverse underwater substrates. Molecular dynamics simulations further reveal atomistic details of the formation and wet adhesion of the PEI/TA coacervate via multimode physical interactions. Lastly, it is demonstrated that the PEI/TA coacervate‐derived hydrogel can effectively repel blood and therefore efficiently deliver the carried growth factors at wound sites, thereby enhancing wound healing in an animal model. The advantages of the PEI/TA coacervate‐derived hydrogel including body fluid‐immiscibility, strong underwater adhesion, adaptability to fit irregular target sites, and excellent biocompatibility make it a promising bioadhesive for diverse biomedical applications.

Osteoarthritis (OA) is a degenerative disease associated with joint inflammation, articular cartilage degeneration and subchondral hypertrophy. Small molecules which both ameliorate chondrocyte OA phenotype and activate bone marrow-derived mesenchymal stem cells (BMSCs) chondrogenesis under inflammatory conditions have the therapeutical potential for OA treatment. In this study, we characterized a novel small molecule which could ameliorate OA progression via novel regulating mechanisms. Docosahexaenoic acid (DHA), a bioactive molecule, was screened from a small molecule library and showed anti-inflammatory and chondroprotective effects in OA chondrocytes, as well as ameliorated IL-1β impaired BMSCs chondrogenesis in Wnt/β-catenin and NF-κB signaling dependent manners. Furthermore, Malat1 was found to be the key mediator of DHA-mediating anti-inflammation chondroprotection and chondrogenesis. DHA also rescued cartilage loss and damage in a surgery-induced OA mice model. The elevation of serum Malat1 levels caused by OA was also downregulated by DHA treatment. Taken together, our findings demonstrated that DHA, with a dual-signaling repression property, exerted its anti-inflammation, chondroprotection and chondrogenesis function possibly via regulating Malat1 level, suggesting that it may be a possible drug candidate for OA patients with elevated MALAT1 expression levels.

Osteoporosis, a common systematic bone homeostasis disorder related disease, still urgently needs innovative treatment methods. Several natural small molecules were found to be effective therapeutics in osteoporosis. In the present study, quercetin was screened out from a library of natural small molecular compounds by a dual luciferase reporter system. Quercetin was found to upregulate Wnt/β-catenin while inhibiting NF-κB signaling activities, and thereby rescuing osteoporosis-induced tumor necrosis factor alpha (TNFα) impaired BMSCs osteogenesis. Furthermore, a putative functional lncRNA, Malat1, was shown to be a key mediator in quercetin regulated signaling activities and TNFα-impaired BMSCs osteogenesis, as mentioned above. In an ovariectomy (OVX)-induced osteoporosis mouse model, quercetin administration could significantly rescue OVX-induced bone loss and structure deterioration. Serum levels of Malat1 were also obviously rescued in the OVX model after quercetin treatment. In conclusion, our study demonstrated that quercetin could rescue TNFα-impaired BMSCs osteogenesis in vitro and osteoporosis-induced bone loss in vivo, in a Malat1-dependent manner, suggesting that quercetin may serve as a therapeutic candidate for osteoporosis treatment.

Background Healthcare workers (HCWs) play a key role in the recommendation of HPV vaccination. Our study aimed to understand to what extent a structured health intervention could change the knowledge and attitudes toward HPV and its vaccines among HCWs in Western China. Methods This was a multi-center, questionnaire-based interventional study conducted across 12 cities of seven provinces in Western China, from November 2018 to July 2019. Participants were recruited from local health systems by e-invitation. Questionnaires were administered to participants before and after the intervention. Results A total of 1448 HCWs attended the educational lectures and 1354 participants completed both pre- and post-study questionnaires. Abstract Background Colposcopy diagnosis and directed biopsy are the key components in cervical cancer screening programs. However, their performance is limited by the requirement for experienced colposcopists. This study aimed to develop and validate a Colposcopic Artificial Intelligence Auxiliary Diagnostic System (CAIADS) for grading colposcopic impressions and guiding biopsies. Methods Anonymized digital records of 19,435 patients were obtained from six hospitals across China. These records included colposcopic images, clinical information, and pathological results (gold standard). The data were randomly assigned (7:1:2) to a training and a tuning set for developing CAIADS and to a validation set for evaluating performance. Results The agreement between CAIADS-graded colposcopic impressions and pathology findings was higher than that of colposcopies interpreted by colposcopists (82.2% versus 65.9%, kappa 0.750 versus 0.516, p 0.05). The CAIADS also demonstrated a superior ability in predicting biopsy sites, with a median mean-intersection-over-union (mIoU) of 0.758. Conclusions The CAIADS has potential in assisting beginners and for improving the diagnostic quality of colposcopy and biopsy in the detection of cervical precancer/cancer.

Summary: Background: Distraction Osteogenesis (DO) is a widely used surgical procedure for limb lengthening, deformity correction, and management of segmental bone defects. However, delayed bone consolidation during DO remains an unsolved clinical problem, which significantly and negatively affects the quality of healing outcome. Pulsed electromagnetic field (PEMF) therapy has been shown to promote osteogenesis and exhibit beneficial effects on the management of delayed union and nonunion fractures. Thus, we hypothesize that PEMF could have beneficial effects on DO. Methods: In this study, specialized PEMF devices employing a novel high slew rate signal were used to firstly investigate effects of PEMF on osteogenesis of MSCs in vitro. MSCs cultured in osteogenic inductive medium treated with or without PEMF (3hrs/day) were subject to qPCR test, Alizarin Red S and ALP staining on day 7. Secondly, an in vivo study was undertaken using a DO model with fifty-six SD-rats randomly divided into two groups. The PEMF group received daily PEMF treatment, 3hr/day, and the control group received no treatment. Histology and IHC staining, radiographic exams, μCT, and biomechanical tests were used to evaluate the quality of bone healing. Immunofluorescence staining and vessel perfusion following by μCT were used to evaluate the effect of PEMF on angiogenesis. Results: Significantly more bone volume and mineral density were observed in the PEMF group compared with the control group, along with better biomechanical properties. Histological results showed that better quality callus formation with less cartilage in callus in PEMF group than control group. Immunofluorescence staining and vessel perfusion results showed that more blood vessels at both early and late stage of DO in PEMF group compared with control group. Conclusion: Our study showed that new high slew rate PEMF signal could promote osteogenesis and angiogenesis in a rat model of DO, which provides insight into the development of new noninvasive mean to accelerate bone formation in the DO process. The translation potential of this article: This study provides supporting evidence for the use of high slew rate PEMF signal to promote bone formation in DO.

Summary: Background: Long duration of bone consolidation in distraction osteogenesis (DO) has been regarded as a major disadvantage. Porous bone substitutes have been long used in aiding bone formation. However the effects of geometric tubular structure of the porous scaffolds on bone formation in DO remains unknown. This study investigated the effects of porous scaffolds with three different geometric structures on bone regeneration in a rabbit DO model. Methods: β-tricalcium phosphate (β-TCP) was fabricated into no tubular, single tubular, or multi-tubular structure constructs. The microstructure of the three types of β-TCP constructs was determined by scanning electron microscope (SEM). Unilateral tibial osteotomy was made at the midshaft with 1 cm tibial shaft removed, fixed with an external lengthener in adulkt New Zealand white rabbits. After acute tibial shortening of 0.5 cm, the remaining 0.5 cm defect was implanted with β-TCP constructs with no tubular, single tubular, or multi-tubular structure. Five days after surgery, the tibia was lengthened at 0.5 mm/12 h for 5 days. After further 28 days of consolidation, the animals were terminated. Bilateral tibial specimens were collected for X-ray, micro-computed tomography examination, four-point bending mechanical test and histological analysis. Results: SEM results showed that pore sizes and pore interconnectivity in all three types of β-TCP constructs were identical. 38 days after surgery (4 weeks after lengthening), the new bone formation and mechanical properties were significantly enhanced in the groups implanted with multi-tubular and single tubular constructs, comparing to the group implanted with no tubular constructs. Furthermore, the group implanted with multi-tubular constructs had significantly better mechanical properties comparing to the group implanted with single tubular constructs. Histological results confirmed that group implanted with multi-tubular constructs had the most advanced bone formation and remodelling in the defect gap. Conclusion: The current study confirmed that β-TCP constructs with multi-tubular structure was the best choice as scaffold aid for bone regeneration in DO, indicating a novel fabrication approach of multi-tubular constructs for rapid bone formation together with DO procedure. The translational potential of this article: Porous β-TCP scaffold with a multi-tubular structure used in combination with DO procedure can significantly shorten the bone consolidation duration. The tubular shaped porous β-TCP scaffold may be used to fill bone defect and to aid bone formation, consolidaiton and remodelling in DO.

Osteonecrosis of the femoral head (ONFH) is a leading cause of mobility impairment which may lead to a total hip replacement. Recent studies have found tendon derived stem cells (TDSCs) might be an ideal cell source for musculoskeletal tissue regeneration. And our previous study has shown Sox11 could promote osteogenesis of bone marrow-derived MSCs. However, the effect of TDSCs or Sox11 over-expressing TDSCs (TDSCs-Sox11) on bone regeneration in ONFH has not been investigated. In the present study, TDSCs were infected with AAV carrying Sox11 or empty vector. We showed that Sox11 could promote the proliferation and osteogenic differentiation of TDSCs, as well as angiogenesis in vitro. The western blot analysis showed that Sox11 could activate the PI3K/Akt signaling pathway to promote osteogenesis of TDSCs. Finally, using a rabbit model of hormone-induced ONFH, our result demonstrated that local administration of TDSCs or TDSCs overexpressing Sox11 could accelerate bone regeneration in necrotic femoral heads, and TDSCs overexpressing Sox11 showed better effects. TDSCs over-expressing Sox11 might be a promising cell source for stem cell therapy to promote bone regeneration, such as ONFH, fracture, bone defect, and so on.

In order to deeply study the heat conduction of nanoporous aerogel, a model of gas-solid heat conduction was established based on the microstructure of aerogel. The model was divided into two subdomains with uniform mesh because of the different gas-solid characteristics, and simulation was performed on each domain using the lattice Boltzmann method. The value of temperature on the boundaries of subdomains was determined by interpolation. Finally, the temperature distribution and the thermal conductivity were maintained. It can be concluded that when the gas-phase scale was fixed, the temperature distribution of the solid phase became more uniform when the scale increased; when the solid-phase scale was fixed, the temperature jump on the gas-solid interface decreased with the increase in the gas-phase scale; and the thermal conductivity of gas-solid coupling varied with the scale of the gas phase or solid phase, showing a scale effect in varying degrees.

Abstract Stretchable electronics‐based surface electromyography (sEMG) evaluation devices are expected to play a big role in clinical diagnosis. However, the stability and quality of the signals collected by existing stretchable electronics are too poor, especially when muscle movement is involved, making them inappropriate for high standard clinical diagnosis. Here, a physiology‐based design method for stretchable electronics and a novel airbag‐type stretchable electrode array (ASEA) device for assessment of the complex female pelvic floor muscle (PFM) is proposed. Clinical trials show that the ASEA device is able to provide stable contact interface and multi‐channel accurate data acquisition. The stability and quality of the sEMG signal are much better than those obtained by the existing stretchable electronics‐based PFM electrode devices. Furthermore, a muscle‐unit evaluation method (MUEM) to assess the PFM complex state is proposed, especially its cross‐interaction between muscles. Clinical trials show that MUEM can accurately and comprehensively assess PFM state and the correlations between main muscles, which unveils the mechanisms of some special muscle states that are not possible using traditional methods. This proof of concept research holds the promise for the development of new diagnostic strategies for muscle pathological research, and has the potential for clinical application and general implication.

Osteoporotic fracture has been regarded as one of the most common bone disorders in the aging society. The natural herb-derived small molecules were revealed as potential treatment approaches for osteoporotic fracture healing. Sesamin is a member of lignan family, which possesses estrogenic activity and plays a significant role in modulating bone homeostasis. Our previous study reported the promoting effect of sesamin on postmenopausal osteoporosis treatment. However, the role of sesamin in osteoporotic fracture healing has not been well studied yet. In this study, we further investigated the putative treatment effect of sesamin on osteoporotic fracture healing. Our study indicated that sesamin could activate bone morphogenetic protein 2 (BMP2) signaling pathway and further promotes in vitro chondrogenesis and angiogenesis activities. This promoting effect was abolished by the treatment of ERα inhibitor. In the osteoporotic bone fracture model, we demonstrated that sesamin markedly improves the callus formation and increases the cartilaginous area at the early-stage, as well as narrowing the fracture gap, and expands callus volume at the late-stage fracture healing site of the OVX mice femur. Furthermore, the angiogenesis at the osteoporotic fracture site was also significantly improved by sesamin treatment. In conclusion, our research illustrated the therapeutic potential and underlying regulation mechanisms of sesamin on osteoporotic fracture healing. Our studies shed light on developing herb-derived bioactive compounds as novel drugs for the treatment of osteoporotic fracture healing, especially for postmenopausal women with low estrogen level.

As one of the leading causes of bone fracture in postmenopausal women and in older men, osteoporosis worldwide is attracting more attention in recent decades. Osteoporosis is a common disease mainly resulting from an imbalance of bone formation and bone resorption. Pharmaceutically active compounds that both activate osteogenesis, while repressing osteoclastogenesis hold the potential of being therapeutic medications for osteoporosis treatment. In the present study, sesamin, a bioactive ingredient derived from the seed of Sesamum Indicum, was screened out from a bioactive compound library and shown to exhibit dual-regulating functions on these two processes. Sesamin was demonstrated to promote osteogenesis by upregulating Wnt/β-catenin, while repressing osteoclastogenesis via downregulating NF-κB signaling . Furthermore, DANCR was found to be the key regulator in sesamin-mediated bone formation and resorption . In an ovariectomy (OVX)-induced osteoporotic mouse model, sesamin could rescue OVX-induced bone loss and impairment. The increased serum level of DANCR caused by OVX was also downregulated upon sesamin treatment. In conclusion, our results demonstrate that sesamin plays a dual-functional role in both osteogenesis activation and osteoclastogenesis de-activation in a DANCR-dependent manner, suggesting that it may be a possible medication candidate for osteoporotic patients with elevated DNACR expression levels.

Using the staged sowing experiment data and rice quality data of the specialized forage rice "Zhongzao 39" (hereinafter referred to as "Z-39") conducted in Nanning, Guangxi, combined with climate and geographic information data, the meteorological factors that play a key role in the formation of rice quality for the specialized forage rice "Z-39" are screened and determined. Based on the actual planting of specialize forage rice in Guangxi and the critical disaster causing indicators of "Z-39" specialize forage rice, the climate quality zoning index of "Z-39" is determined. GIS fine grid spatial simulation analysis method is used for refined spatial simulation analysis on key meteorological factors. The zoning indicator method and path analysis method are fully used for refined level zoning of the climate quality of Guangxi specialized forage rice "Z-39". It includes three levels: optimal, high-quality, and suboptimal. Ultimately, a refined zoning map for the climate quality of "Z-39" is developed, and the results are in line with the actual situation of forage rice cultivation in Guangxi. The aim is to provide a scientific basis for the rational planning and sustainable development of the specialized forage rice "Z-39" in Guangxi. [ABSTRACT FROM AUTHOR]
