Abstract

See Hansen and Møller (doi:10.1093/brain/awz318) for a scientific commentary on this article. Using polygenic risk scores from a genome-wide association study in generalized and focal epilepsy, Leu et al. reveal a significantly higher genetic burden for epilepsy in multiple cohorts of people with epilepsy compared to population controls. Quantification of common variant burden may be valuable for epilepsy prognosis and treatment., Rare genetic variants can cause epilepsy, and genetic testing has been widely adopted for severe, paediatric-onset epilepsies. The phenotypic consequences of common genetic risk burden for epilepsies and their potential future clinical applications have not yet been determined. Using polygenic risk scores (PRS) from a European-ancestry genome-wide association study in generalized and focal epilepsy, we quantified common genetic burden in patients with generalized epilepsy (GE-PRS) or focal epilepsy (FE-PRS) from two independent non-Finnish European cohorts (Epi25 Consortium, n = 5705; Cleveland Clinic Epilepsy Center, n = 620; both compared to 20 435 controls). One Finnish-ancestry population isolate (Finnish-ancestry Epi25, n = 449; compared to 1559 controls), two European-ancestry biobanks (UK Biobank, n = 383 656; Vanderbilt biorepository, n = 49 494), and one Japanese-ancestry biobank (BioBank Japan, n = 168 680) were used for additional replications. Across 8386 patients with epilepsy and 622 212 population controls, we found and replicated significantly higher GE-PRS in patients with generalized epilepsy of European-ancestry compared to patients with focal epilepsy (Epi25: P = 1.64×10−15; Cleveland: P = 2.85×10−4; Finnish-ancestry Epi25: P = 1.80×10−4) or population controls (Epi25: P = 2.35×10−70; Cleveland: P = 1.43×10−7; Finnish-ancestry Epi25: P = 3.11×10−4; UK Biobank and Vanderbilt biorepository meta-analysis: P = 7.99×10−4). FE-PRS were significantly higher in patients with focal epilepsy compared to controls in the non-Finnish, non-biobank cohorts (Epi25: P = 5.74×10−19; Cleveland: P = 1.69×10−6). European ancestry-derived PRS did not predict generalized epilepsy or focal epilepsy in Japanese-ancestry individuals. Finally, we observed a significant 4.6-fold and a 4.5-fold enrichment of patients with generalized epilepsy compared to controls in the top 0.5% highest GE-PRS of the two non-Finnish European cohorts (Epi25: P = 2.60×10−15; Cleveland: P = 1.39×10−2). We conclude that common variant risk associated with epilepsy is significantly enriched in multiple cohorts of patients with epilepsy compared to controls—in particular for generalized epilepsy. As sample sizes and PRS accuracy continue to increase with further common variant discovery, PRS could complement established clinical biomarkers and augment genetic testing for patient classification, comorbidity research, and potentially targeted treatment.

Abstract

This panel seeks to initiate a discussion within the modeling and simulation community about the way we teach conceptual modeling for simulation with the view of bringing about improvements. The challenge addressed is how to educate and equip the novice analyst to become a professional rather than letting him become an artist - being very much the current practice. The need for professionalism is related to good quality research and education in a straightforward way. Emerging insights from literature on the relevance of conceptual modeling for project success, increasing system complexity, and stakeholders taking up an active role in conceptual modeling, further stress this need. This paper highlights key observations motivating the panel, and presents "position papers" on panelists' views on the way forward for education in conceptual modeling. The paper concludes with some themes from the viewpoints in the format of a SWOT (Strengths, Weaknesses, Opportunities and Threats) analysis.

11. Aggregate Modeling for Flow Time Prediction of an End-of-Aisle Order Picking Workstation with Overtaking

Abstract

An aggregate modeling methodology is proposed to predict flow time distributions of an end-of-aisle order picking workstation in parts-to-picker automated warehouses with overtaking. The proposed aggregate model uses as input an aggregated process time referred to as the effective process time in combination with overtaking distributions and decision probabilities, which we measure directly from product arrival and departure data. Experimental results show that the predicted flow time distributions are accurate, with prediction errors of the flow time mean and squared coefficient of variation less than 4% and 9%, respectively. As a case study, we use data collected from a real, operating warehouse and show that the predicted flow time distributions resemble the flow time distributions measured from the data.

Abstract

In recent years several simulation-based serious games have been developed for mastering new business concepts in operations management. This indicates the high potential of simulation use for pedagogical purposes. Unfortunately, this potential is hardly reflected in simulation methodology. We consider this issue by identifying alternative demands game use of simulation sets for model building and application. Moreover, we propose a framework for conceptual modelling for simulation-based serious gaming, which addresses relevant issues in a systematic, step-wise manner. Use of the framework is illustrated by two case examples, highlighting simulation use for training and education respectively.

With a single exception, research on transshipments overlooks replenishment lead times. The only approach for two-location inventory systems with non-negligible lead times could not be generalized to a multi-location setting, and the proposed heuristic method cannot guarantee to provide optimal solutions. This paper uses simulation optimization by combining an LP/network flow formulation with infinitesimal perturbation analysis to examine the multi-location transshipment problem with positive replenishment lead times, and demonstrates the computation of the optimal base stock quantities through sample path optimization. From a methodological perspective, this paper deploys an elegant duality-based gradient computation method to improve computational efficiency. In test problems, our algorithm was also able to achieve better objective values than an existing algorithm.

Abstract

In this paper we present a multi-agent simulation model to investigate purchasing activities in an organizational environment. The starting point is the observation that the majority of purchasing activities in organizations are usually performed without any involvement of the organization's purchasing department. The purpose of the experiments is to investigate if and how certain factors determine the degree to which purchasing professionals become involved in the purchasing of non-product related (NPR) items and services. Among the factors investigated are: corporate purchasing policies, available information, and the nature of the various purchasing activities. Preliminary results show that the behavior of the multi-agent simulation model is an acceptable representation of reality. Furthermore, the results show the limits of a purchasing department's added value and the important role of organizational learning in that respect. The paper provides directions for further research.

Abstract

Agent-based simulations are models where multiple entities sense and stochastically respond to conditions in their local environments, mimicking complex large-scale system behavior. We provide an overview of some important issues in the modeling and analysis of agent-based systems. Examples are drawn from a range of fields: biological modeling, sociological modeling, and industrial applications, though we focus on recent results for a variety of military applications. Based on our experiences with various agent-based models, we describe issues that simulation analysts should be aware of when embarking on agent-based model development. We also describe a number of tools (both graphical and analytical) that we have found particularly useful for analyzing these types of simulation models. Abstract

A reinforcement learning agent has been developed to determine optimal operating policies in a multi-part serial line. The agent interacts with a discrete event simulation model of a stochastic production facility. This study identifies issues important to the simulation developer who wishes to optimise a complex simulation or develop a robust operating policy. Critical parameters pertinent to 'tuning' an agent quickly and enabling it to rapidly learn the system were investigated.

Abstract

Permanent hypoparathyroidism is a serious clinical situation. Allotransplantation of the parathyroid cells is relatively new approach to treatment. Non-cultivated allotransplantation in rabbits is not tried before. In this research parathyroidectomy was performed in six female New Zealand white rabbits. After division of surgically removed tissues into two, cryopreservation after cell isolation was done. Non-cultivated cross allotransplantation was performed under immunosuppression. Serum calcium and phosphorus levels were observed 15 days and histopathological analyses of the transplanted parathyroid tissues were studied. Significant changes in serum calcium and phosphorus levels during the experiment were observed (p=0.001 for both). Calcium levels which were significantly dropped to 6.66±0.7 mg/dL after parathyroidectomy and progressively increased up to 15.98±1.25 mg/dL at the end of the experiment (p=0.004). Phosphorus levels which were increased to 9.38±0.63 mg/dL after parathyroidectomy and stabilized to 4.46±1.06 mg/dL at the end of the experiment (p=0.007). All allotransplanted parathyroid tissues showed normal tissue architecture without evidence of cellular rejection. In conclusion allotransplantation of the parathyroid tissues without cultivation may be considered as an alternative and safe approach for the treatment of permanent hypoparathyroidism.
