6. Anti-EGFL7 antibodies inhibit rat prolactinoma MMQ cells proliferation and PRL secretion

Abstract

Prolactinoma is the most frequently diagnosed pituitary tumors. Dopamine agonists (DAs) are recognized as first-line therapy; however, approximately 10% patients will develop resistance to DAs therapy. Consequently, a large number of investigations have been carried out to identify novel therapeutic targets. Recently, studies have suggested that epidermal growth factor-like domain 7 (EGFL7) can promote tumor growth, invasion, and angiogenesis. We previously reported that overexpression of EGFL7 might play a crucial role in hormone-producing pituitary adenomas. In the present study, we now demonstrated a significantly higher protein expression of EGFL7 in prolactinoma compared with the normal pituitary gland. However, inhibition of EGFL7 with anti-EGFL7 antibodies significantly reduced the proliferation and PRL secretion of rat prolactinoma MMQ cells. Notably, in vitro administration of anti-EGFL7 antibodies significantly induced MMQ cells apoptosis in a dose-dependent manner. In conclusion, our finding suggests that EGFL7 is significantly overexpressed in prolactinoma and inhibition of EGFL7 with antibodies promoted MMQ cells apoptosis and inhibited PRL secretion. Thus, EGFL7 may serve as a potential novel therapeutic target for prolactinomas.

11. Lesions causing central sleep apnea localize to one common brain network

Abstract

ObjectivesTo characterize the specific brain regions for central sleep apnea (CSA) and identify its functional connectivity network.MethodsWe performed a literature search and identified 27 brain injuries causing CSA. We used a recently validated methodology termed “lesion network mapping” to identify the functional brain network subtending the pathophysiology of CSA. Two separate statistical approaches, the two-sample t-test and the Liebermeister test, were used to evaluate the specificity of this network for CSA through a comparison of our results with those of two other neurological syndromes. An additional independent cohort of six CSA cases was used to assess reproducibility.ResultsOur results showed that, despite lesions causing CSA being heterogeneous for brain localization, they share a common brain network defined by connectivity to the middle cingulate gyrus and bilateral cerebellar posterior lobes. This CSA-associated connectivity pattern was unique when compared with lesions causing the other two neurological syndromes. The CAS-specific regions were replicated by the additional independent cohort of six CSA cases. Finally, we found that all lesions causing CSA aligned well with the network defined by connectivity to the cingulate gyrus and bilateral cerebellar posterior lobes.ConclusionOur results suggest that brain injuries responsible for CSA are part of a common brain network defined by connectivity to the middle cingulate gyrus and bilateral cerebellar posterior lobes, lending insight into the neuroanatomical substrate of CSA.

23. Natural Polyphenol Delivered Methylprednisolone Achieve Targeted Enrichment for Acute Spinal Cord Injury Therapy.

Abstract

The strong anti-inflammatory effect of methylprednisolone (MP) is a necessary treatment for various severe cases including acute spinal cord injury (SCI). However, concerns have been raised regarding adverse effects from MP, which also severely limits its clinical application. Natural polyphenols, due to their rich phenolic hydroxyl chemical properties, can form dynamic structures without additional modification, achieving targeted enrichment and drug release at the disease lesion, making them a highly promising carrier. Considering the clinical application challenges of MP, a natural polyphenolic platform is employed for targeted and efficient delivery of MP, reducing its systemic side effects. Both in vitro and SCI models demonstrated polyphenols have multiple advantages as carriers for delivering MP: (1) Achieved maximum enrichment at the injured site in 2 h post-administration, which met the desires of early treatment for diseases; (2) Traceless release of MP; (3) Reducing its side effects; (4) Endowed treatment system with new antioxidative properties, which is also an aspect that needs to be addressed for diseases treatment. This study highlighted a promising prospect of the robust delivery system based on natural polyphenols can successfully overcome the barrier of MP treatment, providing the possibility for its widespread clinical application., (© 2024 Wiley‐VCH GmbH.)
