Background Central precocious puberty (CPP) is a common endocrine disease in children, which shows an increasing trend year by year in recent years. It can be divided into fast-progressing central precocious puberty (RP-CPP) and slow-progressing central precocious puberty (SP-CPP) through pubertal development. RP-CPP has great harm, but it is difficult to distinguish it from SP-CPP in early clinical stage, mainly relying on the progress of adolescent development and bone age during follow-up. At present, there is a lack of effective laboratory indicators to predict RP-CPP. Objective To investigate the relationship between luteinizing hormone (LH) /follicle-stimulating hormone (FSH) ratio and pubertal development. Methods CPP girls aged 4-10 years old (n=380) admitted to the Children's Hospital Affiliated to Zhengzhou University from January 2020 to May 2022 were regression selected and divided into RP-CPP group (n=130) and SP-CPP group (n=250 cases) according to indicators such as puberal development process. Clinical characteristics of the two groups were compared and analyzed. Univariate and multivariate Logistic regression analysis of the influencing factors of RP-CPP was performed, and ROC curve of LH/FSH ratio on the predictive value of RP-CPP was drawn. The traditional microstructure detecting methods such as metallography and electron backscatter diffraction are destructive to the sample and time-consuming and they cannot meet the needs of rapid online inspection. In this paper, a random forest regression microstructure characterization method based on a laser ultrasound technique is investigated for evaluating the microstructure of a titanium alloy (Ti-6Al-4V). Based on the high correlation between the longitudinal wave velocity of ultrasonic waves, the average grain size of the primary α phase, and the volume fraction of the transformed β matrix of the titanium alloy, and with the longitudinal wave velocity as the input feature and the average grain size of the primary α phase and the volume fraction of the transformed β matrix as the output features, prediction models for the average grain size of the primary α phase and the volume fraction of the transformed β matrix were developed based on a random forest regression. The results show that the mean values of the mean relative errors of the predicted mean grain size of the native α phase and the volume fraction of the transformed β matrix for the six samples in the two prediction models were 11.55% and 10.19%, respectively, and the RMSE and MAE obtained from both prediction models were relatively small, which indicates that the two established random forest regression models have a high prediction accuracy. Objective: To analyze a child with 11β hydroxylase deficiency (11β-OHD) due to CYP11B2/CYP11B1 chimeric gene., Methods: Clinical data of the child who was admitted to Henan Children's Hospital on August 24, 2020 were retrospectively analyzed. Peripheral blood samples of the child and his parents were collected and subjected to whole exome sequencing (WES). Candidate variant was verified by Sanger sequencing. RT-PCR and Long-PCR were carried out to verify the presence of chimeric gene., Results: The patient, a 5-year-old male, had featured premature development of secondary sex characteristics and accelerated growth, and was diagnosed with 21 hydroxylase deficiency (21-OHD). WES revealed that he has harbored a heterozygous c.1385T>C (p.L462P) variant of the CYP11B1 gene, in addition to a 37.02 kb deletion on 8q24.3. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), the c.1385T>C (p.L462P) was rated as a likely pathogenic variant (PM2_Supporting+PP3_Moderate+PM3+PP4). The results of RT-PCR and Long-PCR suggested that CYP11B1 and CYP11B2 genes have recombined to form a CYP11B2 exon 1~7/CYP11B1 exon 7~9 chimeric gene. The patient was diagnosed as 11β-OHD and effectively treated with hydrocortisone and triptorelin. A healthy fetus was delivered following genetic counseling and prenatal diagnosis., Conclusion: 11β-OHD may be misdiagnosed as 21-OHD due to the potential CYP11B2/CYP11B1 chimeric gene, which will require multiple methods for the detection.
