14. Redistribution of Histone Marks on Inflammatory Genes Associated With Intracerebral Hemorrhage-Induced Acute Brain Injury in Aging Rats

Abstract

The contribution of histone mark redistribution to the age-induced decline of endogenous neuroprotection remains unclear. In this study, we used an intracerebral hemorrhage (ICH)-induced acute brain injury rat model to study the transcriptional and chromatin responses in 13- and 22-month-old rats. Transcriptome analysis (RNA-seq) revealed that the expression of neuroinflammation-associated genes was systematically upregulated in ICH rat brains, irrespective of age. Further, we found that interferon-γ (IFN-γ) response genes were activated in both 13- and 22-month-old rats. Anti-IFN-γ treatment markedly reduced ICH-induced acute brain injury in 22-month-old rats. At the chromatin level, ICH induced the redistribution of histone modifications in the promoter regions, especially H3K4me3 and H3K27me3, in neuroinflammation-associated genes in 13- and 22-month-old rats, respectively. Moreover, ICH-induced histone mark redistribution and gene expression were found to be correlated. Collectively, these findings demonstrate that histone modifications related to gene expression are extensively regulated in 13- and 22-month-old rats and that anti-IFN-γ is effective for ICH treatment, highlighting the potential of developing therapies targeting histone modifications to cure age-related diseases, including brain injury and neuroinflammation.

15. Carrying load estimation and risk assessment of cultivated land contaminated by livestock and poultry breeding in Huizhou

Abstract

With the rapid development of the livestock and poultry breeding industry in Huizhou, Guangdong Province, improper utilization of livestock and poultry fecal sewage may pollute cultivated land and even affect the drinking water source in the Dongjiang basin through surface runoff. In order to assess the carrying loads and risks of livestock and poultry fecal sewage in the cultivated land of Huizhou, this study collected the latest statistical yearbook data of livestock and poultry breeding in Huizhou and the second national survey of pollution sources. According to these data, the production and emissions of livestock and poultry fecal sewage and major pollutants(COD, NH3-N, TN, and TP) in 2018 were calculated using the method of the production and discharge coefficient. The cultivated land carrying loads and environmental risks of livestock and poultry fecal sewage, TN, and TP were then estimated and assessed based on the actual cultivated land area of each district(county). The results showed that in 2018, the quantity of poultry fecal sewage produced by livestock and poultry breeding in Huizhou was 1.51×106 t with emissions reaching 4.44×105 t, and the contribution proportion and its order for each livestock and poultry animal was pig(50.3%) > broiler(17.2%) > beef(14.3%) > layer(13.4%) > cow(4.8%). Moreover, the production and emissions of major pollutants totaled 2.61×108 kg and 7.66×107 kg, respectively. Among all the districts(counties), Boluo had the largest emission of fecal sewage because of its large scale livestock and poultry breeding, which accounted for 42.9%. The cultivated land carrying loads of livestock and poultry fecal sewage, TN, and TP in Huizhou were 7.69 t×hm-2, 65.38 kg×hm-2, and 15.63 kg×hm-2, respectively. The alarm grading levels for poultry fecal sewage and TN were level Ⅰ, whereas that for TP was level Ⅱ, and the threats caused by them to the surrounding environment were small. Thus, the resource utilization of livestock and poultry waste needs to be strengthened continuously through balance planning for planting and breeding.

Abstract

BACKGROUND Microbial transformation of phytosterols has been applied to produce androst-4-ene-3,17-dione ( AD). However, low substrate solubility and product inhibition are major problems to be solved. Biphasic systems are efficient strategies, and ionic liquids ( ILs), as one of the phases in biphasic systems, are promising alternatives to traditional organic solvents due to environmental constraints. RESULTS In this work, nine water-immiscible ILs were used in phytosterols side-chain cleavage by Mycobacterium sp. MB 3683. The solubilities of phytosterols and AD in these ILs and distribution coefficients of AD between water and ILs were first measured. [ PrMIM][ PF6] was determined to be a suitable candidate to establish a biphasic IL-aqueous system. Under optimal conditions, i.e. inoculum size of 12% with 48 h age, IL addition at 84 h, and phase ratio of 20:1 (v/v, aqueous/ IL), AD production reached 2.23 g L−1 after 5 days of biotransformation with substrate concentration of 5 g L−1 in a scaled-up biphasic system of 200 mL. This bioconversion conducted in a biphasic IL-aqueous system was more efficient than that in a monophasic aqueous system. CONCLUSION Owing to the environmental advantages of ILs and the above positive results, biphasic IL-aqueous systems have the potential to be applied in phytosterols side-chain cleavage. © 2015 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Abstract

Nowadays, the silver is widely used in the biological field and its biological safety catches great attention. It is important to know the distribution of silver ions within the biological organism and the toxic threshold concentration in the tissue. Therefore, a highly sensitive method for measurement of trace amount of silver ion in the medical biological samples is needed. With its high sensitivity for detection of metal ions, inductively coupled plasma mass spectrometry (ICP-MS) method is well suited for quantification of trace amount of silver ion in such samples, but method development is still in its infancy. Consequently, a simple and convenient method for determination of trace amount of silver in the animal serum, tissues or organs was developed, in which the samples were subjected to the microwave digestion, followed by the ICP-MS analysis. To begin with, the samples of serum, muscle, bone marrow, bone, heart, liver, spleen, and kidney were sequently processed in 5 mL of HNO3 and 2 mL of H2O2 solution. Then the samples were completely digested by microwave with the power of 2 000 watts. The temperature was raised gradually by 3-step program. Moreover, the data achieved were reproducible and the method was time saving and especially for large amounts of sample processing. Then the digested solutions were diluted to constant volume. Finally, the concentration of 107Ag in the samples was analyzed by the method of ICP-MS under the optimized conditions. Element yttrium (Y) was used as the internal standard to compensate for matrix suppression effect and improve the accuracy of measurement. For one thing, the analytical results showed that the detection limit of the trace element 107Ag was 0.98 μg · kg(-1), and furthermore, the correlation coefficient of standard curve was 0.999 9. For another thing, the recovery rate of the silver element ranged from 98% to 107%, which was calculated according to measured quantity before adding standard, adding standard and measured quantity after adding standard. At the same time, the relative standard deviation (RSD) of the method was in the range of 2.0%-4.3%. The concentrations of element silver in animal serum, tissues and organs were determined by the aboved method. The obtained results showed that silver ions were mainly accumulated in the liver after they were intaken into the body. The results suggested that the microwave digestion-ICP-MS method could accurately determine the trace element Ag in the body. The method developed has good feasibility and is suitable for the determination of trace element Ag in various types of medical and biological samples, especially for large quantities of biological samples. The process has the advantages of easysample processing and it is simple and convenient. In addition, the accurate results could be obtained in a short time with high sensitivity. Last but not least, the method provides the guidance for the determination of trace elements in other biological samples.
