In the construction and maintenance of particle accelerators, all the accelerator elements should be installed in the same coordinate system, only in this way could the devices in the actual world be consistent with the design drawings. However, with the occurrence of the movements of the reinforced concrete cover plates at short notice or building deformations in the long term, the control points upon the engineering structure will be displaced, and the fitness between the subnetwork and the global control network may be irresponsible. Therefore, it is necessary to evaluate the deformations of the 3D alignment control network. Different from the extant investigations, in this paper, to characterize the deformations of the control network, all of the congruent models between the points measured in different epochs have been identified, and the congruence model with the most control points is considered as the primary or fundamental model, the remaining models are recognized as the additional ones. Furthermore, the discrepancies between the primary S-transformation parameters and the additional S-transformation parameters can reflect the relative movements of the additional congruence models. Both the iterative GCT method and the iterative combinatorial theory are proposed to detect multiple congruence models in the control network. Considering the actual work of the alignment, it is essential to identify the competitive models in the monitoring network, which can provide us a hint that, even the fitness between the subnetwork and the global control network is good, there are still deformations which may be ignored. The numerical experiments show that the suggested approaches can describe the deformation of the 3D alignment control network roundly.

Roses (Rosa spp.) are widely used as ornamental plants and cut flowers and in perfumes and cosmetics; therefore, they have extremely high ornamental and economic value. Improving ornamental qualities (color, aroma, flower shape, plant architecture, petal senescence), agronomic traits (disease resistance, stress tolerance), and other traits would add value to cultivated roses. However, the lack of an efficient genetic transformation system limits functional genetic studies of roses. Therefore, we aimed to develop a simple, high-throughput Agrobacterium rhizogenes-mediated hairy root transformation system to analyze gene function in rose. We explored the factors affecting the induction and transformation of hairy root cultures in rose by screening different types of explants, Rosa genotypes, Agrobacterium strains, media, and infection and co-cultivation times for genetic transformation under aseptic conditions. We established an A. rhizogenes-mediated hairy root transformation system in rose using A. rhizogenes strains MSU440 and Ar Qual, successfully obtained transgenic hairy roots, and conducted preliminary experiments to examine their regeneration by employing the ultraviolet light (UV)-visible reporter eYGFPuv as a marker. Finally, we established a simple hairy root transformation process under non-aseptic conditions. The rapid and efficient transformation system developed here will enable efforts to investigate gene function in roses and engineer improved rose varieties. SUMMARY: The petals of rose (Rosa sp.) flowers determine the ornamental and industrial worth of this species. The number of petals in roses was previously shown to be subject to fluctuations in ambient temperature. However, the mechanisms by which rose detects and responds to temperature changes are not entirely understood. In this study, we identified short interstitial telomere motifs (telo boxes) in the second intron of AGAMOUS (RcAG) from China rose (Rosa chinensis) that play an essential role in precise temperature perception. The second intron of RcAG harbors two telo boxes that recruit telomere repeat binding factors (RcTRBs), which interact with CURLY LEAF (RcCLF) to compose a repressor complex. We show that this complex suppresses RcAG expression when plants are subjected to low temperatures via depositing H3K27me3 marks (trimethylation of lysine 27 on histone H3) over the RcAG gene body. This regulatory mechanism explains the low‐temperature‐dependent decrease in RcAG transcript levels, leading to the production of more petals under these conditions. Our results underscore an interesting intron‐mediated regulatory mechanism governing RcAG expression, enabling rose plants to perceive temperature cues and establish petal numbers. Abstract Background Suicide and major depressive disorders (MDD) are strongly associated, and genetic factors are responsible for at least part of the variability in suicide risk. We investigated whether variation at the tryptophan hydroxylase-2 (TPH2) gene rs7305115 SNP may predispose to suicide attempts in MDD. Methods We genotyped TPH2 gene rs7305115 SNP in 215 MDD patients with suicide and matched MDD patients without suicide. Differences in behavioral and personality traits according to genotypic variation were investigated by logistic regression analysis. Results There were no significant differences between MDD patients with suicide and controls in genotypic (AG and GG) frequencies for rs7305115 SNP, but the distribution of AA genotype differed significantly (14.4% vs. 29.3%, p < 0.001). The G-allele frequency was significantly higher in cases than control group (58.1% vs.45.6%, p < 0.001), but the A-allele carrier indicated a decreased trend in MDD with suicide behaviors than control group (41.9% vs.54.4%, p < 0.001). The multivariate logistic regression analysis indicated that TPH2 rs7305115 AA (OR 0.33, 95% CI 0.22-0.99), family history of suicide (OR 2.98, 95% CI 1.17-5.04), negative life events half year ago (OR 6.64, 95% CI 2.48-11.04) and hopelessness (OR 7.68, 95% CI 5.79-13.74) were significantly associated with the suicide behaviors in MDD patients. Conclusions The study suggested that hopelessness, negative life events and family history of suicide were risk factors of attempted suicide in MDD while the TPH2 rs7305115A remained a significant protective predictor of suicide attempts.
