Chemoresistance is closely related to the therapeutic effect and prognosis in breast cancer patients. Increasing evidences demonstrated that RNA binding proteins (RBPs) have notable roles in regulating cancer cell proliferation, metastasis and chemotherapeutic sensitivity. RNA binding motif single stranded interacting protein 2 (RBMS2), an RBP, has been considered to be a tumor suppressor in several cancers. However, its role of doxorubicin sensitivity in breast cancer patients has not yet been fully revealed. Growing evidence indicates N6-methyladenosine (m6A) has biological function in oncogenesis. METTL3, the catalytic component, is the most important part of methyltransferase complex and plays a crucial role in cancers. However, the biological function of circRNAs derived from METTL3 in breast cancer and the underlying molecular mechanism remains unclear. Herein, we report circMETTL3, which has not been explored in breast cancer, and it is markedly upregulated in breast cancer. Moreover, we uncovered that circMETTL3 could facilitate cell proliferation, migration and invasion in breast cancer. Mechanism investigation showed that circMETTL3 might act as a competing endogenous RNA (ceRNA) of miR-31-5p and upregulate its target cyclin-dependent kinases (CDK1). Moreover, m6A modification of circMETTL3 might affect its expression. Taken together, our results elucidate that circMETTL3 promotes breast cancer progression through circMETTL3/miR-31-5p/CDK1 axis. Moreover, METTL3, the host gene of circMETTL3, may regulate circMETTL3 expression in an m6A-dependent manner, while circMETTL3 has no effect on METTL3 expression, providing a new relationship between the circRNA and the corresponding host gene. Thus, it may serve as a new therapeutic target for breast cancer.

Three new species of the genus Hexarhopalus Fairmaire, 1891, all of the nominate subgenus, are described and figured from China: Hexarhopalus (Hexarhopalus) ferreri sp. nov. from Yunnan, H. (H.) tianbaoyanensis sp. nov. from Fujian, and H. (H.) yeziyangi sp. nov. from Jiangxi.

Background: Triple negative breast cancer (TNBC) is highly malignant and has a worse prognosis, compared with other subtypes of breast cancer due to the absence of therapeutic targets. MKLP1 plays a crucial role in tumorigenesis and cancer progression. However, the role of MKLP1 in triple negative breast cancer and the underlying mechanism remain unknown. The study aimed to elucidate the biological function regulatory mechanism of MKLP1 in triple negative breast cancerMethods: Quantitative real-time PCR and Western blotting were used to determine the MKLP1 expression in breast cancer tissues and cell lines. Then, functional experiments in vitro and in vivo were performed to investigate the effects of MKLP1 on tumor growth and metastasis in triple negative breast cancer. Chromatin immunoprecipitation assay was conducted to illustrate the potential regulatory mechanisms of MKLP1 in triple negative breast cancer.Results: We found that MKLP1 was significantly up-regulated and associated with poor prognosis in triple negative breast cancer. MKLP1 could promote triple negative breast cancer proliferation, migration and invasion in vitro and in vivo. MKLP1 could activate Wnt/β-catenin pathway and promote EMT progression. In addition, FOXM1, upregulated by WDR5 via H3K4me3 modification, directly bound to the promoter of MKLP1 gene to promote its transcription and accelerated TNBC progression via Wnt/β-catenin pathway. Both of small inhibitor of FOXM1 and WDR5 could inhibit TNBC progression. Conclusions: Our findings elucidate WDR5/FOXM1/MKLP1/Wnt/β-catenin axis is associated with TNBC progression and may provide a novel and promising therapeutic target for TNBC treatment.

Background: Recently, studies have shown that kinesins(KIF) play an important role in the occurrence and development of many tumors. However, there is no complete understanding of the role of KIF family in Pan cancer, and its role in the immunity and tumor microenvironment (TME) is unclear.Methods: Based on TCGA database and integrated several R packages, we explored the relationship between the expression of KIF genes and patient survival, immune subtypes, TME, tumor stem cell correlation, and drug sensitivity in cancer.Results: We use nine highly expressed KIF genes(KIF2C, KIF4A, KIF7, KIF11, KIF14, KIF18A, KIF18B, KIF20A, KIF20B) to represent whole KIF family. The change in KIF gene expression was significantly related to overall survival. The nine KIFs' high expression is accompanied by the up-regulation of C1 immune subtype, which is related to cell proliferation and interruption of immune process. Further, KIF gene expression showed significant correlation and cancer cell stemness characteristics. Top25 relevant KIF-drug pairs were displayed according to their P values. We further discussed KIF family influence in Mesothelioma(MESO) and Sarcoma(SARC). The CIBERSORT results manifested that increased level of infiltration of B cells naive, Mast cells resting and NK cells activated could be used as a protective factor to promote survival.Conclusions: Our study supplemented a complete map of the effect of KIF family in Pan cancer. We suggested that KIF family may be a potential target for cancer therapy.

The results indicated that circMETTL3 could promote breast cancer cell proliferation, migration and invasion as well as tumorigenesis in vivo. Mechanistic analysis showed that circMETTL3 might act as a ceRNA (competing endogenous RNA) of miR-31-5p to relieve the repressive effect of miR-31-5p on its target cyclin-dependent kinases (CDK1). Moreover, m6A modification of circMETTL3 might affect its expression.Conclusion: Our findings suggest that circMETTL3 promotes breast cancer progression through circMETTL3/miR-31-5p/CDK1 axis. Moreover, METTL3, the host gene of circMETTL3, may regulates circMETTL3 expression in a m6A-dependent manner, while circMETTL3 has no effect on METTL3 expression, providing a new relationship between the circRNA and the corresponding host gene. The purpose of this paper is to explore the consumer acceptability of Usage Based Insurance (this article is referred to as UBI) in the context of vehicle networking. The research and analysis are carried out by the combination of qualitative and quantitative research. This study research selects two large insurance company management personnel or the project head as the interview object and takes the UBI present situation and the future development as the topic to carry on the semi-structured interview. The visitors think that UBI is the future development trend, but in mainland China market, insurance companies are still in the exploratory stage of UBI. Furthermore, this study uses questionnaires to collect sample data on vehicle owner's acceptance of UBI, monitoring acceptance of funds and good degree of driving habits and apply the linear regression model to find out the significant factors that affect consumers' purchasing tendency of UBI. The results show that, first, the more the record, the lower purchase propensity of UBI that consumers have. Second, compared to a single traditional insurance, personalized UBI is more easily favored by consumers. Third, the higher the acceptance of personal information monitored during driving, the higher purchase propensity of UBI that consumers have. The information security in the big data era highlights its importance.

The Differential Quadrature Method（abbreviated as DQM）is easy to be implemented by computers to solve many problems in engineering mechanics. In the field of pipe vibration methods are mainly finite element method , finite difference calculus and so on. In the formation of coefficient matrix these methods need to calculate a large number of numerical integration , resulting in low efficiency, while we can use matlab to program, taking the advantage of easy programming of DQM, after establishing mathematical model of the pipe conveying fluid .The program designed in this paper can be a very good solution to pipe nature frequency.After obtaining the nature frequency, we have analyzed the relationship between pipe length and nature frequency and have found out the critical value of the pipe length. The method can also be applied to engineering filed related .
