Abstract The fast expansion of photon detection technology has fertilized the rapid growth of single-photon sensing and imaging techniques. While promising significant advantages over their classical counterparts, they suffer from ambient and quantum noises whose effects become more pronounced at low light levels, limiting the quality of the acquired signal. Here, we study how photon-counting noises degrade a single-pixel optical classifier via compressive sensing, and how its performance can be restored by using quantum parametric mode sorting. Using modified National Institute of Standards and Technology (MNIST) handwritten digits as an example, we examine the effects of detector dark counts and in-band background noises and demonstrate the effectiveness of mode filtering and upconversion detection in addressing those issues. We achieve 94% classification accuracy in the presence of 500 times stronger in-band noise than the signal received. Our results suggest a robust and efficient approach to single photon sensing in a practical environment, where sunlight, ambient, and multiscattering noises can easily dominate the weak signal.

Hormones are important signaling molecules controlling physiological homeostasis. ELISA kits are commonly used to measure hormones; however, few ELISA kits are multiplex, not all species-specific ELISA kits are commercially available, and ELISA kits typically require a significant volume of biological fluids. Pigs resemble humans in digestive physiology, making them an excellent model in preclinical research of nutrition and metabolism. In this study, we developed and validated a simple liquid–liquid extraction procedure and LC-MS/MS method for the simultaneous quantification of insulin, cortisol, glucagon-like peptide-1 (GLP-1) (7-37) and (7-36), acyl and des-acyl ghrelin, and carboxylated osteocalcin in pig serum. The proposed method is specific, highly sensitive (LOQ in ng/mL and pg/mL), reasonably accurate (more than 76.2% of all quality control samples within 20% error from nominal values), and precise (intra-day CV ≤ 10% and inter-day CV ≤ 23.1%). The recoveries of all analytes and corresponding internal standards ranged from 83.7 to 116.0%. The method also requires a low serum volume of 50–100 μL, which is invaluable when sample volume is limited. These methods could be easily extended for use in other mammalian species.

This work investigates the composition of whey protein phospholipid concentrate (WPPC), an underutilized dairy stream, and reveals that it is a source of many bioactive compounds that can benefit the immune system and gut health. Our glycoproteomics approach uncovered that proteins derived from the milk fat globule membrane (MFGM) represent 23% of the total protein relative abundance and identified 85 N-glycans. Released sialic acid, an additional marker of glycosylation, ranged from 1.2 to 2% of the total weight. Xanthine oxidase, a glycosylated marker of MFG bioactivity, was found in high abundance and displayed higher antimicrobial activity than bovine milk, despite its similar fat and solids content. An average MFG diameter of 2.64 ± 0.01 µm was found in liquid WPPC, compared to 4.78 ± 0.13 µm in bovine milk, which likely explains the unusually high presence of glycosylated membrane-bound proteins and phospholipids, whose total fatty acids accounted for 20% of the WPPC total fatty acid pool. Free and bound oxylipins (mainly derived from linoleic acid) were also identified, together with other less abundant anti-inflammatory lipid mediators derived from eicosapentaenoic acid and docosahexaenoic acid. Our study demonstrates that WPPC represents a promising starting material for bioactive compound extraction and a functional vehicle for the delivery of small MFGs.

Almonds (Prunus dulcis) are one of the most consumed tree nuts worldwide and have been recognized as a healthy and nutritious food. Nevertheless, almonds are also a source of allergenic proteins that can trigger several mild to life-threatening allergic reactions. The effects of selected extraction conditions (aqueous vs. protease-assisted aqueous extraction) on the protein profile determined by proteomics analysis of excised SDS-PAGE gel bands, in vitro protein digestibility, and immunoreactivity of almond protein extracts, were evaluated. Proteolysis altered almond protein sequential and conformational characteristics thus affecting digestibility and antigenicity. Proteomics analysis revealed that enzymatic extraction resulted in the reduction of allergen proteins and epitopes. While complete hydrolysis of Prunin 1 and 2 α-chain was observed, Prunin 1 and 2 β-chains were more resistant to hydrolysis. Protein in vitro digestibility increased from 79.1 to 88.5% after proteolysis, as determined by a static digestion model. The degree of hydrolysis (DH) and peptide content of enzymatically extracted proteins during gastric and duodenal digestion were significantly higher than the ones from unhydrolyzed proteins. Proteolysis resulted in a 75% reduction in almond protein immunoreactivity as determined by a sandwich enzyme-linked immunosorbent assay and a reduction in IgE and IgG reactivities using human sera. The present study shows that moderated hydrolysis (7% DH) using protease can be used as a strategy to improve almond protein digestibility and reduce antigenicity. This study's findings could further enhance the potential use of almond protein hydrolysates in the formulation of hypoallergenic food products with improved nutritional quality and safety.

Oxygen A-band measurements can provide important information about cloud top height, cloud physical and optical thickness, and surface atmospheric pressure. So far, O2 A-band measurements are typically made with spectrometers at a spectral resolution of 40 p.m. (such as in the OCO-2 satellite) or a coarser resolution. This paper reports a new CubeSat measurement concept with higher spectral resolution over the O2 A-band using integrated photonic circuits and solar photon counting techniques. An integrated Micro-ring filter (MRR) chip with 10 p.m. resonant linewidth with an extinction ratio of 25 dB or higher is designed, fabricated, and used for precise measurements of the atmospheric oxygen A-band absorption line-shapes around 770 nm. With solar-photon counting and using a narrow-band filter made of an integrated, fast-swept MRR on lithium niobate on insulator (LNOI), we have demonstrated a high-resolution measurement of the O2 A-band absorption spectrum, exhibiting good agreement with the HITRAN database.

Most lidars used for cloud observations have the range resolution of about 10 m, so they are incapable of resolving submeter-scale processes that are crucial to cloud evolution. This article describes a prototype of a ground-based, vertically pointing, time-gated, time-correlated single-photon-counting lidar (referred to as the T2 lidar) developed to explore atmospheric clouds at range resolution two orders of magnitude finer than traditional atmospheric lidars. The T2 lidar emits green-light pulses (532 nm) at a repetition rate of 20.6 kHz and a pulse width of ∼650 ps, which enables the observation of aerosol and cloud layers at heights from a few hundred meters to 7.28 km above the ground level at range resolution down to 10 cm. In addition, a digital delay pulse generator controls the detector to only receive photons for a short period after each laser pulse. This time-gated technique blocks photons arriving from regions outside the target zone, thus significantly reducing the noise level and allowing observation even inside clouds. Initial observations show that the T2 lidar can detect sharp cloud boundaries and fine structures near the cloud base. Such refined measurements of cloud structure could lead to an improved understanding of microphysical processes such as droplet activation, entrainment and mixing, and precipitation.

Imagers capable of reconstructing three-dimensional scenes in the presence of strong background noise are desirable for many remote sensing and imaging applications. Here, the authors report an imager operating in photon-starved and noise-polluted environments through quantum parametric mode sorting.

Reverse-phase solid-phase extraction (SPE) is regularly used for separating and purifying food-derived oligosaccharides and peptides prior to liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. However, the diversity in physicochemical properties of peptides may prevent the complete separation of the two types of analytes. Peptides present in the oligosaccharide fraction not only interfere with glycomics analysis but also escape peptidomics analysis. This work evaluated different SPE approaches for improving LC-MS/MS analysis of both oligosaccharides and peptides through testing on peptide standards and a food sample of commercial interest (proteolyzed almond extract). Compared with conventional reverse-phase SPE, mixed-mode SPE (reverse-phase/strong cation exchange) was more effective in retaining small/hydrophilic peptides and capturing them in the high-organic fraction and thus allowed the identification of more oligosaccharides and dipeptides in the proteolyzed almond extract, with satisfactory MS/MS confirmation. Overall, mixed-mode SPE emerged as the ideal method for simultaneously improving the identification of food-derived oligosaccharides and small peptides using LC-MS/MS analysis.

BackgroundPes planus (flatfoot) is a common deformity characterized by the midfoot arch collapses during walking. As the midfoot is responsible for shock absorption, persons with flatfoot experience increased risk of injuries such as thumb valgus, tendinitis, plantar fasciitis, metatarsal pain, knee pain, lower-back pain with prolonged uphill, downhill, and level walking, depriving them of the physical and mental health benefits of walking as an exercise.MethodsFifteen female college students with flatfoot were recruited. A wireless plantar-pressure system was used to measure the stance time, cadence, plantar pressure, and contact area. Parameters were compared between wearing flat and arch-support insoles using a two-way repeated measures ANOVA with on an incline, decline, and level surface, respectively. The significance level α was set to 0.05. The effect size (ES) was calculated as a measure of the practical relevance of the significance using Cohen's d.ResultsOn the level surface, the stance time in the arch-support insole was significantly shorter than in the flat insole (pConclusionsThese results imply that wearing an arch-support insole provides benefits in the shortened stance time and generation of propulsion force to the big toe while walking on uphill and level surfaces and to the metatarsals 2-4 while walking on the level surface. More evenly distributed contact areas across the midfoot may help absorb shock during uphill, downhill and level walking.

Abstract Selective detection of signal over noise is essential to measurement and signal processing. Time-frequency filtering has been the standard approach for the optimal detection of non-stationary signals. However, there is a fundamental tradeoff between the signal detection efficiency and the amount of undesirable noise detected simultaneously, which restricts its uses under weak signal yet strong noise conditions. Here, we demonstrate quantum parametric mode sorting based on nonlinear optics at the edge of phase matching to improve the tradeoff. By tailoring the nonlinear process in a commercial lithium-niobate waveguide through optical arbitrary waveform generation, we demonstrate highly selective detection of picosecond signals overlapping temporally and spectrally but in orthogonal time-frequency modes as well as against broadband noise, with performance well exceeding the theoretical limit of the optimized time-frequency filtering. We also verify that our device does not introduce any significant quantum noise to the detected signal and demonstrate faithful detection of pico-second single photons. Together, these results point to unexplored opportunities in measurement and signal processing under challenging conditions, such as photon-starving quantum applications.

The effect of hot air (HA, 45°C, 3.5 h) treatment on reducing gray mold caused by Botrytis cinerea in strawberry fruit and the possible mechanisms were investigated. The results showed that HA treatment significantly reduced lesion diameter and enhanced activities of chitinase (CHI), β-1,3-glucanase and phenylalanine ammonia-lyase (PAL) in strawberry fruit. Total phenolic contents were also increased by HA treatment. The activities of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) were higher in HA treated strawberry fruit than those in control. Expression of three defense related genes such as CAT, CCR-1 allele and PLA6 was greatly induced in HA treated strawberry fruit with or without inoculation by B. cinerea. In addition, the in vitro experiment showed that HA treatment inhibited spore germination and tube growth of B. cinerea. These results suggested that HA treatment directly activated disease resistance against B. cinerea in strawberry fruit without priming response and directly inhibiting growth of B. cinerea.

AIM:To investigate the corneal endothelial cell density and morphology and central corneal thickness in the Guangxi Maonan and Han adolescent students of China.METHODS:Noncontact specular microscope (Topcon SP3000P, Tokyo, Japan) was performed in 133 adolescent students of Maonan nationality (M:F 54:79) and 105 adolescent students of Han nationality (M:F 50:55), 5 to 20y of age, who were randomly selected from 3 schools in Huanjiang Maonan Autonomous County of Guangxi Zhuang Autonomous Region of China. Parameters studied included endothelial cell density, mean cell area, coefficient of variation in cell size, percentage hexagonality and central corneal thickness.RESULTS:Endothelial cell density, mean cell area, coefficient of variation in cell size, percentage hexagonality and central corneal thickness in the study population were (2969.50±253.93) cells/mm2, (339.23±29.44) µm2, (29.96±4.07) %, (64.58±9.41) % and (523.71±32.82) µm in Maonan and (2998.26±262.65) cells/mm2, (336.11±30.07) µm2, (29.89±5.03) %, (64.91±11.64) % and (524.39±33.15) µm in Han, respectively. No significant differences were observed in endothelial cell density, mean cell area, coefficient of variation in cell size, percentage hexagonality and central corneal thickness between Maonan and Han (P=0.615, 0.659, 0.528, 0.551, 0.999). In Maonan and Han, we found age was negatively correlated with endothelial cell density and percentage hexagonality and positively correlated with mean cell area and coefficient of variation in cell size. Negative correlation was also found between central corneal thickness and age in Han, whereas no correlation was found in Maonan.CONCLUSION:There were no differences between Maonan and Han in corneal endothelial cell density and morphology and central corneal thickness. In these two nationalities, there were statistically significant decrease in endothelial cell density and percentage hexagonality with increasing age and statistically significant increase in cell area and coefficient of variation in cell size with increasing age. Central corneal thinned with increasing age in Han, whereas difference did not attain statistical significance in Maonan.

Leaderless translation is prevalent in haloarchaea, with many of these leaderless transcripts possessing short 5'-untranslated regions (UTRs) less than 10 nucleotides. Whereas, little is known about the function of this very short 5'-UTR. Our previous studies determined that just four nucleotides preceded the start codon of hsp70 mRNA in Natrinema sp. J7, with residues -3A and +4G, relative to the A of the ATG start codon, acting as the preferred bases around the start codon of all known haloarchaeal hsp70 genes. Here, we examined the effects of nucleotides flanking the start codon on gene expression. The results revealed that shortening and deletion of the short 5'-UTR enhanced transcript levels; however, it led to significant reductions in overall translational efficiency. AUG was efficiently used as start codons, in both the presence and absence of short 5'-UTRs. GUG also could initiate translation, even though it was so inefficient that it would not be detected without considerably elevated transcript. Nucleotide substitutions at position -4 to +6 were shown to affect gene expression by transcript and/or translational levels. Notably, -3A and A/U nucleotides at position +4~+6 were more optimal for gene expression. Nucleotide transversions of -3A to -3C and +4G to +4T with hsp70 promoter from either Haloferax volcanii DS70 or Halobacterium salinarum NRC-1 showed the same effects on gene expression as that of Natrinema sp. J7. Taken together, our results suggest that the nucleotides flanking the start codon in hsp70 mRNAs with very short 5'-UTRs play an important role in haloarchaeal gene expression.

Quantum-mechanically indistinguishable photons produced by independent (or equivalently, mutually phase incoherent) light sources are essential for distributed quantum information processing applications. We demonstrate heralded generation of such photons in two spatially separate telecom-fiber spools, each driven by pulsed pump waves that are measured to have no mutual phase coherence. Through Hong–Ou–Mandel experiments, we measure the quantum interference visibility of those photons to be $76.4\pm 4.2%$ . Our experimental results are well predicted by a quantum multimode theory we developed for such systems without the need for any fitting parameter.

We demonstrate sum-frequency generation between a telecom wavelength and the Rb D2 line, achieved through natural phase matching in a nonlinear whispering gallery mode resonator. Due to the strong optical field confinement and ultra high Q of the cavity, the process saturates already at sub-mW pump peak power, at least two orders of magnitude lower than in existing waveguide-based devices. The experimental data are in agreement with the nonlinear dynamics and phase matching theory based on spherical geometry. Our experimental and theoretical results point toward a new platform for manipulating the color and quantum states of light waves for applications such as atomic memory based quantum networking and logic operations with optical signals.
