Background Codonopsis pilosula and C. tangshen are both plants widely used in traditional Chinese medicine. Polysaccharides, which are their primary active components, are thought to be important in their extensive use. In this study, two neutral polysaccharide fractions of C. pilosula (CPPN) and C. tangshen (CTPN) were obtained by fractionation on a DEAE-Sepharose column and characterized. Results It was confirmed that the neutral polymers CPPN and CTPN were β-(2,1)-linked inulin-type fructans with non-reducing terminal glucose, and degree of polymerization (DP) of 19.6 and 25.2, respectively. The antioxidant and prebiotic activities in vitro were assayed based on IPEC-J2 cell lines and five strains of Lactobacillus. Results indicated that the effects of CPPN and CTPN were increased antioxidant defense in intestinal epithelial cells through enhanced cell viability, improved expression of total antioxidant capacity, glutathione peroxidase, superoxide dismutase and catalase, and reduced levels of malondialdehyde and lactic dehydrogenase. The prebiotic activity of CPPN and CTPN was demonstrated by the promoting effect on Lactobacillus proliferation in vitro. The different biological activities obtained between the two fractions are probably due to the different DP and thus molecular weights of CPPN and CTPN. Conclusion The inulin fractions from C. pilosula and C. tangshen were natural sources of potential intestinal antioxidants as well as prebiotics, which will be valuable in further studies and new applications of inulin-containing health products. © 2020 Society of Chemical Industry.

Background Codonopsis pilosula possesses properties related to nourishing the spleen and stomach, and tonifying qi of stomach and mind in traditional Chinese medicine. Polysaccharides, which are the primary active components of C. pilosula, are thought to be in charge of their extensive use. Now, a systematic summary structural and bioactivities diversity of polysaccharide from C. pilosula (CPs) are necessary to address their future aspects and application. Scope and approach In this review, the extraction and purification methods, structural features and bioactivities diversity of CPs are summarized and discussed. Structure-activity relationships (SAR) is also analyzed and discussed. Key findings and Conclusions Boiling water extraction, ultrasonic extraction, enzymatic hydrolysis and microwave methods, subcritical water extraction and accelerated solvent extraction are used for extracting CPs. Among them, water extraction and alcohol precipitation method is the most frequently used one. Around 15 neutral fractions and 25 acidic fractions were purified from different species of C. pilosula, while only few of them were well characterized. CPs exhibited various biological activities, including immunomodulation, antitumor, prebiotics, antifatigue, antiviral, antioxidation activity. However, most of bioactivities reported were based on crude CPs; in vivo studies of purified CPs and the potential mechanisms need to be extensively investigated. SAR illustrates that chemical and enzymatic modification of CPs could significantly increase the bioactivity of CPs, but other impact factors on SAR such as water solubility, molecular weight and chain information are still unknown.

The root of Angelica sinensis, has been commonly used in gynecology for centuries, and is normally applied divided into different parts in various clinical applications. At present, the majority of existing studies focus on the volatile oil and ferulic acid extracted from different parts of A. sinensis, but there is a dearth of scientific information on its water-soluble polysaccharides.The structures of polysaccharides from plants, have been reported contributing to multiple pharmacological activities such as anti-oxidative, anti-inflammatory, anti-tumor and liver protection. Therefore, the focus of this study was on its anti-oxidative and anti-inflammatory activities in vitro, which would be based on the various polysaccharides with distinct structures obtained from different parts of the A. sinensis root.Four parts of A. sinensis root were separated according to the Chinese Pharmacopoeia: head, body, tail and whole body. Crude polysaccharides were obtained by water extraction and ethanol precipitation method, and were further fractionated by DEAE Sepharose chromatographic column and gel filtration. The comparison of ASPs from different root parts were performed, including chemical compositions determined by colorimetric analysis, monosaccharide compositions measured by high performance liquid chromatography (HPLC), glycosidic linkage units determined by methylation and gas chromatography-mass spectrometry (GC-MS), organic functional groups determined by FT-IR, molecular weight (Mw) demarcated by gel permeation chromatography, and the viscosities and solubilities were measured according to method published in the previous report with minor modification. In vitro biological activities of APSs were compared on lipopolysaccharide (LPS)-induced inflammatory and oxidative stress models on IPEC-J2 cells.Four purified polysaccharides, ASP-H-AP, ASP-B-AP, ASP-T-AP and ASP-Hb-AP from the root of A. sinensis, were obtained, and consisted of various contents of protein and the polyphenol. They were possibly pectic polysaccharides with a long homogalacturonan region as the main backbone and ramified with rhamnogalacturonan I region, but they were differed by subregions and the relative contents of glycosidic units. The Mw of four pectic polysaccharides were ranged from 67.9-267.7 kDa. The infrared spectrum also showed that the four polysaccharide fractions contained the characteristic peaks of polysaccharides. Their distinct primary structure could lead to a variety of biological activities. In vitro biological assays suggested that four polysaccharide fractions can protect IPEC-J2 cells against the LPS-induced inflammation by down-regulating inflammation factors and related genes on IPEC-J2 cells. These polysaccharides also could alleviate oxidative stress on IPEC-J2 cells by up-regulating the gene and protein expressions of antioxidant enzymes. It was concluded that ASP-H-AP possessed better anti-inflammatory and anti-oxidative effects, while those of ASP-T-AP was relatively poor among the four polysaccharide fractions.All results indicated that the structure of pectic polysaccharides from different root parts of A. sinensis differed, which lead to their distinct anti-inflammatory and anti-oxidative activities. This may also be one of the factors why different parts of A. sinensis showed various pharmacological activities and applied independently in traditional use. In addition, it would be valuable for further studies on structure-activity relationship of polysaccharides obtained by different root parts of A. sinensis.

An inulin (CPPF), isolated from a traditional Chinese herbal medicine

Roots of Aconitum carmichaelii are used in Asian countries due to its content of bioactive alkaloids. In the production of root preparations, tons of leaves are usually discarded, leading to a huge waste of herbal material. The aim of this study is to investigate the polysaccharides in these unutilized leaves. A neutral polysaccharide (AL-N) appeared to be a mixture of heteromannans, and two purified acidic polysaccharides (AL-I-I and AL-I-II) were shown to be pectins containing a homogalacturonan backbone substituted with terminal β-Xylp-units. AL-I-I consisted of a type-I rhamnogalacturonan core, with arabinan and type-II arabinogalactan domains while AL-I-II was less branched. AL-N and AL-I-I were able to modulate the complement system, while AL-I-II was inactive. Interestingly, AL-N, AL-I-I and AL-I-II were shown to exert anti-inflammatory effects on porcine enterocyte IPEC-J2 cells. AL-I-I and AL-I-II were able to down-regulate the expression of toll-like receptor 4 (TLR4) and nucleotide-binding oligomerization domain 1 (NOD1).

Aconitum carmichaelii, belonging to the Ranunculaceae family, is a widely used traditional herbal plant in Asian countries, especially in China. The lateral ("Fuzi") and mother ("Chuanwu") roots are the two main plant parts used in Traditional Chinese Medicine (TCM), where they are used in the treatment of acute myocardial infarction, heart failure, rheumatoid arthritis, and as analgesics.In order to further guide the research direction and application of A. carmichaelii, this study aims to give a systematic and in-depth overview on the phytochemical and pharmacological studies of non-alkaloid natural products with focus on polysaccharides and phenolic compounds.A comprehensive search in the literature was conducted based on the databases Google Scholar, SciFinder (American Chemical Society), Springer Link, PubMed Science, Science Direct and China National Knowledge Internet, Wanfang Data, in addition to books, doctoral and master's dissertations, and official website. The main keywords were: "Aconitum carmichaelii", "Aconiti Lateralis Radix Praeparata", "Fuzi", "Chuanwu", "Aconiti Radix", "monkshood" and "Bushi".A. carmichaelii is known for the use of its different root parts, including "Fuzi" and "Chuanwu". Different types of polysaccharides, both neutral and acidic, and 39 phenolic compounds like flavonoids, phenylpropanoids, lignans, neolignans, and benzoic acid derivatives have been isolated and identified from the roots. Pharmacological studies of the isolated polysaccharides have demonstrated various biological effects such as hypoglycemic, hypolipidemic, cardiovascular, immunomodulatory, anti-tumor, and neuropharmacological activities. Studies on pharmacological effects of the phenolic compounds isolated from the roots are however limited.This review shows that polysaccharides could be one of the active components in the roots of A. carmichaelii, and they are promising for future applications due to their pharmacological properties. In addition, polysaccharides are generally non-toxic, biocompatible, and biodegradable. This review also sheds light on new research directions for A. carmichaelii. A more detailed structural characterization of polysaccharides from different root parts of A. carmichaelii, and their structure-activity relationships are required. Additionally, their pharmacological properties as immunomodulators in the intestinal system should be investigated. Further, more knowledge about the pharmacological effects and molecular mechanisms of the phenolic compounds that have been identified are needed.

Platycodonis Radix is widely used as homology of medicine and food in China; polysaccharides are thought to be one of its functional constituents. In this study, a pectic polysaccharide, PGP-I-I, was obtained from the root of the traditional medicine plant Platycodon grandiflorus through ion exchange chromatography and gel filtration. This was characterized being mainly composed of 1,5-α-L-arabinan and both arabinogalactan type I (AG-I) and II chains linked to rhamnogalacturonan I (RG-I) backbone linked to longer galacturonan chains. In vitro bioactivity study showed that PGP-I-I could restore the intestinal cellular antioxidant defense under the condition of hydrogen peroxide (H2O2) treatment through promoting the expressions of cellular antioxidant genes and protect against oxidative damages.

Purpose A bibliometric study was conducted to identify and analyze the most influential 100 publications in vertebral augmentation research.Methods Databases of Web of Science and Pubmed were searched using the key words of ‘percutaneous vertebroplasty’, ‘percutaneous kyphoplasty’, ‘balloon kyphoplasty’, ‘vertebroplasty’, ‘kyphoplasty’ and ‘vertebral augmentation’. The searched publications were arranged by the times of cited per year after excluding the papers not associated with vertebral augmentation closely. The top 100 publications were defined as the most influential papers in this field and analyzed. Collecting data included the journal, the year of publication, the nation of the Corresponding author, the total times of citation, the citation times per year since publication, and the study type of the papers. The evidence levels of the clinical studies were graded between I to V. The systemic reviews and meta-analyses were combined as a specific type.Results The total citation times of the 100 publications were 27222 and they were cited 18.3 times per year in average. These papers were published in 31 journals in which 13 kinds of journals published more than 2 papers. Corresponding authors came from 17 different nations in which the American authors were the most. The level IV evidence studies were the most as 41 and only 3 papers reached the standards of level I evidence study. The number of the publications decreased obviously after 2009, in which year the two level I evidence publications were published.Conclusion There are still contrary opinions about vertebral augmentation now even after the publications of the two authoritative studies of level I evidence. New studies should be developed with more deliberated design and longer follow-up, meanwhile the patients should be selected more cautious and the evaluation factors should be included more extensively.

Purification, characterization and biological activities of polysaccharides from Lycium barbarum pedicel were investigated in this study. Two polysaccharides, PLBP-I-I and PLBP-II-I, were obtained from water extracts by anion exchange chromatography and gel filtration. Structural elucidation based on IR, 1H NMR, and 13C NMR spectra indicated that these two fractions were typical pectic polysaccharides, with homogalacturonan and rhamnogalacturonan type I regions and arabinogalactan side chains, and some of the galacturonic acid units were methyl esterified. Both fractions exhibited potent complement fixating activity and pro-antioxidant defense capacity, and those two fractions showed different activities. The higher complement fixation activity was obtained in fraction PLBP-I-I, while the higher pro-antioxidant defense capacity was obtained in fraction PLBP-II-I, which may be due to the structural differences between those two fractions. Thus, the pedicel of L. barbarum could be used as a potential source for natural immunomodulator and antioxidant.

The extensive prospects of polysaccharides in medicine and pharmaceuticals have elicited much interest. Ligusticum chuanxiong polysaccharides, previously extracted as neutral polysaccharides, exhibit antioxidant, anticancer, andantibacterial capacities in vitro. Here we show one pectic polysaccharide extracted from rhizome of L. chuanxiong(LCP-II-I). Structural analysis indicated its long homogalacturonan region is interrupted by rhamnogalacturonan type I, with arabinan and arabinogalactan type I as side chains. Intake of the LCP-II-I significantly promoted the expression of antioxidant enzymes and their master regulator PGC-1α, resulting in higher antioxidant capacity in jejunum and cecum of aged mice. Consistently, treatment with LCP-II-I promotes antioxidant enzymes expression, and protects SW480 cells against H2O2 induced oxidative stress. Our work isolates and identifies a pectic polysaccharide, LCP-II-I, and reveals its potential application in antioxidant defense, which will be important for oxidative stress related disorders treatment in aging intestine.

Two polysaccharides, GCP-I-I and GCP-II-I, were obtained from 100°C water extracts of Gentiana crassicaulis roots by DEAE anion exchange chromatography and gel filtration. The results from methanolysis, methylation, FT-IR and NMR, indicated that these two fractions are typical pectic polysaccharides, with HG and RG-I regions and AG-I/AG-II side chains, and some of the galacturonic acid units of fraction GCP-I-I were methyl esterified. Fractions GCP-I-I and GCP-II-I, both exhibited potent complement fixation, and fraction GCP-I-I was more potent than positive control BPII. The higher complement fixation activity obtained in fraction GCP-I-I may be due to the higher Mw and/or higher amount of AG-II present in fraction GCP-I-I than fraction GCP-II-I. The polysaccharides from G. crassicaulis could be used as a potential natural immunomodulator.

The inulin-type fructan was obtained by DEAE anion exchange chromatography from C. pilosula Nannf. var. modesta (Nannf.) L. T. Shen, after optimized extract condition, which was established by response surface methodology, designed using Box-Behnken factorial design and the optimum condition were: extracting 2.5 h with ratio of solvent to material 40 mL/g at 100 °C, twice. The maximum extraction yield was 20.6 ± 0.2%. It was confirmed as β-(2-1) linkage fructan, with terminal glucose, and with a degree of polymerization of 2–17 (DPav = 6), shown by the results of methanolysis, methylation, nuclear magnetic resonance and molecular weight determination. The prebiotic activity was proven on account of stimulation effect on Lactobacillus and pH reduction of medium in vitro. The results indicated that the inulin from C. pilosula could be used as a potential natural source of prebiotics.

Sijunzi Decoction (SJZD) is a formula used for the treatment of spleen deficiency and gastrointestinal diseases in Traditional Chinese Medicine. Polysaccharides are reported to be the main components of SJZD responsible for its bio-functions. However, highly purified and clearly characterized polysaccharides from SJZD are not well described. Here we obtained a purified polysaccharide (SJZDP-II-I) from SJZD using ion exchange chromatography and gel filtration. Structure analysis by FT-IR and NMR identified SJZDP-II-I as a typical pectic polysaccharide with homogalacturonan and rhamnogalacturonan type I regions and arabinogalactan type I and II as side chains. In vitro studies indicated that SJZDP-II-I treatment could significantly enhance the total antioxidant capacity of SW480 cells, resulting from the promoted expressions of antioxidant enzymes and their master regulator PGC-1α, which would be valuable for further research and applications.

We have carried out the investigation on a sponge-derived fungus,which was identified as Emericella variecolor from the south sea of China. Two new chemical constituents,(+)-2-acetyl-dihydroterrein (1) and (+)-3-acetyl-dihydroterrein (2),with four known compounds,anditomin (3),andilesin A (4),andilesin C (5) and andilesin B (6),were isolated from this fungus by column chromatography over silica gel, Sephadex LH-20, and ODS. The structures of 1 and 2 were elucidated by spectroscopic methods including NMR,HR-ESI-MS,and CD.

This paper presents a new calibration method for binocular vision system, based on CPSO-BP neural network. Firstly, the training set of the back propagation (BP) neural network is formed by the image feature point extracted from the binocular vision system. Then the cooperate particle swarm optimization (CPSO) algorithm is introduced to optimize the weights of the BP neural network, making the network with a stronger ability of the global optimization. Experimental results demonstrate that the proposed CPSO-BP-based algorithm has a higher calibration precision than the traditional BP-based calibration method.

In this study, an acidic polysaccharide from Codonopsis pilosula Nannf. var. modesta (Nannf.) L. T. Shen (WCP-I) and its main fragment, WCP-Ia, obtained after pectinase digestion, were structurally elucidated and found to consist of a rhamnogalacturonan I (RG-I) region containing both arabinogalactan type I (AG-I) and type II (AG-II) as sidechains. They both expressed immunomodulating activity against Peyer&rsquo, s patch cells. Endo-1,4-&beta, galactanase degradation gave a decrease of interleukine 6 (IL-6) production compared with native WCP-I and WCP-Ia, but exo-&alpha, l-arabinofuranosidase digestion showed no changes in activity. This demonstrated that the stimulation activity partly disappeared with removal of &beta, d-(1&rarr, 4)-galactan chains, proving that the AG-I side chain plays an important role in immunoregulation activity. WCP-Ia had a better promotion effect than WCP-I in vivo, shown through an increased spleen index, higher concentrations of IL-6, transforming growth factor-&beta, (TGF-&beta, ), and tumor necrosis factor-&alpha, (TNF-&alpha, ) in serum, and a slight increment in the secretory immunoglobulin A (sIgA) and CD4+/CD8+ T lymphocyte ratio. These results suggest that &beta, 4)-galactan-containing chains in WCP-I play an essential role in the expression of immunomodulating activity. Combining all the results in this and previous studies, the intestinal immune system might be the target site of WCP-Ia.

Platycodon grandiflorus is a plant widely used in traditional Chinese medicine, of which polysaccharides are reported to be the main components responsible for its bio-functions. In this work, the inulin-type fructan (PGF) was obtained by DEAE anion exchange chromatography from the water extracted from P. grandifloras. Characterization was performed with methanolysis, methylation, and NMR and the results showed that PGF is a &beta, (2-1) linked fructan, with terminal glucose and with a degree of polymerization of 2&ndash, 10. In order to study its biofunctions, the prebiotic and immunomodulation properties were assayed. We found that PGF exhibited good prebiotic activity, as shown by a promotion on six strains of lactobacillus proliferation. Additionally, the PGF also displayed direct immunomodulation on intestinal epithelial cells and stimulated the expressions of anti-inflammatory factors. These results indicated that the inulin from P. grandiflorus is a potential natural source of prebiotics as well as a potential intestinal immunomodulator, which will be valuable for further studies and new applications.

Based on the condition of No.1-2 coal seam of Shang Wan Coal Mine Shendong mining area, the movement of the overlying strata of the 7.0m large mining height workface have been studied by similar simulation test, which geometric similarity ratio is 1:50.The results show that: the immediate roof strata caves stratified to loose blocks, and the goaf can’t be filled with these loose blocks; the basic roof strata caves stratified too, the lower slice caves to loose blocks, it evolves into the immediate roof and widen the immediate roof; there is the periodic weighting phenomenon of size alternating in the large mining height workface of No.1-2 coal seam ; the immediate roof and the basic roof and the overlying strata of basic roof have different characteristics of caving; the large mining height workface has a greater sphere of supporting pressure than the traditional small height workface, the peak supporting pressure of the large mining height workface is more depth to the coalwall than the average mining height workface, and this led to workface roof strata fracture further ahead.

Based on the high mining height of large cutting height workface in shallow thick coal seam and the few falling waste rock in goaf, the key roof can not be supported effectively, the facture mechanical model of key roof was established. The theoretical calculation formula of key stratum fracture step and working resistance of support were obtained by using fracture mechanics. The results show that the fracture step of key roof relate to not only the mechanical character of key roof and the load of overlaying rock seam, but also the working resistance of support and horizontal pressure in key roof. The reasonable working resistance of support and the step of roof fracture were analyzed in 1-2coalmine 51104 face of a mine in Dongsheng area. The theoretical results are well agreeable with the field measured results.

Rhizome of Ligusticum chuanxiong is an effective medical plant, which has been extensively applied for centuries in migraine and cardiovascular diseases treatment in China. Polysaccharides from this plant have been shown to have interesting bioactivities, but previous studies have only been performed on the neutral polysaccharides. In this study, LCP-I-I, a pectic polysaccharide fraction, was obtained from the 100 °C water extracts of L. chuangxiong rhizomes and purified by diethylaminethyl (DEAE) sepharose anion exchange chromatography and gel filtration. Monosaccharide analysis and linkage determination in addition to Fourier transform infrared (FT-IR) spectrometer and Nuclear magnetic resonance (NMR) spectrum, indicated that LCP-I-I is a typical pectic polysaccharide, with homo-galacturonan and rhamnogalacturonan type I regions and arabinogalactan type I and type II (AG-I/AG-II) side chains. LCP-I-I exhibited potent complement fixation activity, ICH50 of 26.3 ± 2.2 µg/mL, and thus has potential as a natural immunomodulator.

Patients in a vegetative state due to nervous system injury loss cognitive ability, and have no autonomous behavior ability.They lie in bed perennial, unable to turn over, unconscious painless, and can't put the discomfort message to others.Their partial body because of long time compression, poor blood circulation, which cause cell necrosis, and lead to decubitus. Especially the closed bedsore, not easy to find,and seriously affect the treatment and rehabilitation of vegetables, even threaten the life of patients. We should closely observe the patient nursing daily, to reduce the occurrence of complications by careful nursing. The article summarized the nursing of elderly patients in 1 cases, and provided experience for clinical nursing.
