Abstract As the burgeoning field of Artificial Intelligence (AI) continues to permeate the fabric of healthcare, particularly in the realms of patient surveillance and telemedicine, a transformative era beckons. This manuscript endeavors to unravel the intricacies of recent AI advancements and their profound implications for reconceptualizing the delivery of medical care. Through the introduction of innovative instruments such as virtual assistant chatbots, wearable monitoring devices, predictive analytic models, personalized treatment regimens, and automated appointment systems, AI is not only amplifying the quality of care but also empowering patients and fostering a more interactive dynamic between the patient and the healthcare provider. Yet, this progressive infiltration of AI into the healthcare sphere grapples with a plethora of challenges hitherto unseen. The exigent issues of data security and privacy, the specter of algorithmic bias, the requisite adaptability of regulatory frameworks, and the matter of patient acceptance and trust in AI solutions demand immediate and thoughtful resolution .The importance of establishing stringent and far-reaching policies, ensuring technological impartiality, and cultivating patient confidence is paramount to ensure that AI-driven enhancements in healthcare service provision remain both ethically sound and efficient. In conclusion, we advocate for an expansion of research efforts aimed at navigating the ethical complexities inherent to a technology-evolving landscape, catalyzing policy innovation, and devising AI applications that are not only clinically effective but also earn the trust of the patient populace. By melding expertise across disciplines, we stand at the threshold of an era wherein AI's role in healthcare is both ethically unimpeachable and conducive to elevating the global health quotient.

Harnessing the photoinduced phase transitions in organic crystals, especially the changes in shape and structure across various dimensions, offers a fascinating avenue for exact spatiotemporal control, which is crucial for developing future smart devices. In our study, we report a new photoactive molecular crystal made from (E)-2-(3-phenyl-allylidene)malonate ((E)-PADM). When exposed to ultraviolet (UV) light at 365 nm, this compound experiences an E-to-Z photoisomerization in liquid solution and a crystal-to-liquid phase transition in solid crystals. Remarkably, nanoscopic crystalline rods boost their melting rate and degree compared to bulk crystals, indicating that miniaturization enhances the photoinduced melting effect. Our results demonstrate a simple approach to rapidly drive molecular crystals into liquids via photochemical reactions and phase transitions.

Despite the low solubility of hydrogen isotopes (HIs) in tungsten (W), their concentration can reach up to ∼10 at.% after low-energy plasma irradiation. This is generally attributed to the vacancies that may accommodate excessive HIs. However, the kinetic energy of incident HIs transferred to W is far below the energy threshold to create a Frenkel pair, the underlying mechanism of defect production is still unclear. Here, we investigate the influence of H on the defect production in W using the molecular dynamic (MD) simulations. It is found that the threshold displacement energy (TDE) in bulk W slight decreases with the increasing of H concentration. This is due to the formation of H-vacancy complexes, which prevents the vacancy-interstitial recombination. More importantly, the H effects are significantly magnified in the surface region. On the one hand, the maximum kinetic energy transferred from 400 eV H to W can reach up to ∼21 eV due to the double-hit process, which is two times higher than that predicted by elastic collision model. On the other hand, the momentum transferred to W is completely random, including both the recoil direction upward and downward from the surface. Accordingly, the lowest TDE in W surface is only 15–21 eV at sub-surface layers with the depth of 6.7–11.1 Å, which is lower than the maximum kinetic energy transferred to W. Therefore, the low-energy HIs irradiation can create the defects in W surface directly. Our findings provide deep insight into defect production in W at sub-threshold energy and have wider implications for materials performance under low-energy ions irradiation.

Helium (He)-induced damage is a sensitive concern for the performance of tungsten plasma facing materials (W-PFMs). Recent experiments have revealed that trace impurities in He plasma can effectively prevent the formation of He bubbles and fuzz on W surfaces. To explore its plausibility and underlying mechanism, we performed a multiscale computational study that combines density functional theory calculations and object kinetic Monte Carlo simulations to investigate the effects of a small quantity of beryllium (Be) on the evolution of He bubbles. It is found that there is a strong attractive interaction between He and Be, which can be attributed to the decrease in electron density and the lattice distortion induced by embedded Be atoms. Therefore, the co-implantation of Be continuously introduces trapping centers for He. Due to the low implantation depth and high migration energy of Be, the Be atoms are located close to the surface, leading to the trapping of the majority of He within the near-surface region and the development of a shielding layer for He permeation. The presence of Be facilitates the dispersion of the trapped He, skewing the He clusters into smaller sizes. More importantly, the Be trapping centers bring the He clusters closer to the surface, significantly increasing the probability of bubble bursting and the release of He back to the vacuum. This ultimately leads to a lower retention of He in the case of He + Be co-irradiation, compared with the case of He-only irradiation. Consequently, our findings elucidate the suppressive effect of a low flux of Be atoms on the growth of He bubbles, highlighting the need to focus on synergetic effects between plasma species.

The aggregation behavior of hydrogen (H) isotopes after low-energy plasma irradiation is particularly important for tungsten (W) in future fusion devices. Here, using the first-principles calculation, we demonstrated an interesting strain effect that may drive the planar aggregation of interstitial H atoms in W. Although there are attractive interactions between H atoms, the spontaneous nucleation of these platelet-like H clusters in strain-free W appears impossible even at high H concentrations (up to 0.1 at.%) because of the extremely low configurational entropy. However, applied uniaxial strain significantly increased the binding energy of H platelets and enabled planar H clusters to form. These results suggest that uniaxial strain enhances the nucleation and growth of H platelets in W, regardless of whether it is a compressive or tensile strain. Moreover, the binding energy of one-layer H clusters was lower than that of parallel H platelets, implying that the formation of multi-layer H clusters in W and their stability is also promoted by uniaxial strain. Meanwhile, the presence of planar H clusters dramatically reduced the vacancy formation energy in W, which in turn provided an extra trapping site to accommodate excessive H atoms. These results provide an important reference for understanding the H evolution in W-PFMs.

Electrical impedance tomography (EIT) is a radiation-free and noninvasive medical image reconstruction technique in which a current is injected and the reflected voltage is received through electrodes. EIT electrodes require good connection with the skin for data acquisition and image reconstruction. However, detached electrodes are a common occurrence and cause measurement errors in EIT clinical applications. To address these issues, in this study, we proposed a method for detecting faulty electrodes using the differential voltage value of the detached electrode in an EIT system. Additionally, we proposed the voltage-replace and voltage-shift methods to compensate for invalid data from the faulty electrodes. In this study, we present the simulation, experimental, and in vivo chest results of our proposed methods to verify and evaluate the feasibility of this approach.

Infrared detection technology has been widely applied in many areas. Unlike internal photoemission and the photoelectric mechanism, which are limited by the interface barrier height and material bandgap, the research of the hot carrier effect from nanometer thickness of metal could surpass the capability of silicon-based Schottky devices to detect mid-infrared and even far-infrared. In this work, we investigate the effects of physical characteristics of Cr nanometal surfaces and metal/silicon interfaces on hot carrier optical detection. Based on the results of scanning electron microscopy, atomic force microscopy, and X-ray diffraction analysis, the hot carrier effect and the variation of optical response intensity are found to depend highly on the physical properties of metal surfaces, such as surface coverage, metal thickness, and internal stress. Since the contact layer formed by Cr and Si is the main role of infrared light detection in the experiment, the higher the metal coverage, the higher the optical response. Additionally, a thicker metal surface makes the hot carriers take a longer time to convert into current signals after generation, leading to signal degradation due to the short lifetime of the hot carriers. Furthermore, the film with the best hot carrier effect induced in the Cr/Si structure is able to detect an infrared signal up to 4.2 μm. Additionally, it has a 229 times improvement in the signal-to-noise ratio (SNR) for a single band compared with ones with less favorable conditions.

The plasticity and hardness of metals are largely dependent on how dislocation interacts with solute atoms. Here, taking bcc molybdenum (Mo) as the example, the interaction of interstitial solutes carbon (C) and oxygen (O) with screw dislocation, and their influences on the dislocation motion, have been determined using first-principles calculations and thermodynamic models. Due to the incompact atomic structure and variation of electronic states in the dislocation core, C and O will segregate from the bulk system to the dislocation region. Notably, the presence of C/O at the dislocation induces the reconstruction of the core structure, from an easy-core to hard-core configuration. This originates from the fact that the hard-core structure provides a larger available volume at the interstitial site than the easy-core structure and, thus, facilitates the dissolution of C and O. More importantly, the addition of C/O in the dislocation significantly increases the Peierls stresses and double-kink formation enthalpies of screw dislocation in Mo, from 1.91 GPa and 1.18 eV for C/O-free dislocation to 5.63/4.69 GPa and 1.77/1.58 eV for C/O-saturated dislocation. Therefore, these interstitial solutes have a pinning effect on the dislocation motion, and this effect becomes stronger with higher segregating levels. This work reveals the profound effect of interstitial solutes on the properties of the dislocation core and provides a fundamental factor to account for the interstitial solutes-related phenomena in bcc metals.

Direct exposure to intensive visible light can lead to solar retinopathy, including macular injury. The signs and symptoms include central scotoma, metamorphopsia, and decreased vision. However, there have been few studies examining retinal injury due to intensive light stimulation at the cellular level. Neural network arrangements and gene expression patterns in zebrafish photoreceptors are similar to those observed in humans, and photoreceptor injury in zebrafish can induce stem cell-based cellular regeneration. Therefore, the zebrafish retina is considered a useful model for studying photoreceptor injury in humans. In the current study, the central retinal photoreceptors of zebrafish were selectively ablated by stimulation with high-intensity light. Retinal injury, cell proliferation and regeneration of cones and rods were assessed at 1, 3 and 7 days post lesion with immunohistochemistry and in situ hybridization. Additionally, a light/dark box test was used to assess zebrafish behavior. The results revealed that photoreceptors were regenerated by 7 days after the light-induced injury. However, the regenerated cells showed a disrupted arrangement at the lesion site. During the injury-regeneration process, the zebrafish exhibited reduced locomotor capacity, weakened phototaxis and increased movement angular velocity. These behaviors matched the morphological changes of retinal injury and regeneration in a number of ways. This study demonstrates that the zebrafish retina has a robust capacity for regeneration. Visual impairment and stress responses following high-intensity light stimulation appear to contribute to the alteration of behaviors.

AIM: To investigate the role of tumor necrosis factor-alpha (TNF-α) in zebrafish retinal development and myelination. METHODS: Morpholino oligonucleotides (MO), which are complementary to the translation start site of the wild-type embryonic zebrafish TNF-α mRNA sequence, were synthesized and injected into one- to four-cell embryos. The translation blocking specificity was verified by Western blotting using an anti-TNF-α antibody, whole-mount in situ hybridization using a hepatocyte-specific mRNA probe ceruloplasmin (cp), and co-injection of TNF-α MO and TNF-α mRNA. An atonal homolog 7 (atoh7) mRNA probe was used to detect neurogenesis onset. The retinal neurodifferentiation was analyzed by immunohistochemistry using antibodies Zn12, Zpr1, and Zpr3 to label ganglion cells, cones, and rods, respectively. Myelin basic protein (mbp) was used as a marker to track and observe the myelination using whole-mount in situ hybridization. RESULTS: Targeted knockdown of TNF-α resulted in specific suppression of TNF-α expression and a severely underdeveloped liver. The co-injection of TNF-α MO and mRNA rescued the liver development. Retinal neurogenesis in TNF-α morphants was initiated on time. The retina was fully laminated, while ganglion cells, cones, and rods were well differentiated at 72 hours post-fertilization (hpf). mbp was expressed in Schwann cells in the lateral line nerves and cranial nerves from 3 days post-fertilization (dpf) as well as in oligodendrocytes linearly along the hindbrain bundles and the spinal cord from 4 dpf, which closely resembled its endogenous profile. CONCLUSION: TNF-α is not an essential regulator for retinal neurogenesis and optic myelination.

We investigate the influence of hydrostatic/biaxial strain on the formation, migration, and clustering of vacancy in tungsten (W) using a first-principles method, and show that the vacancy behaviors are strongly dependent on the strain. Both a monovacancy formation energy and a divacancy binding energy decrease with the increasing of compressive hydrostatic/biaxial strain, but increase with the increasing of tensile strain. Specifically, the binding energy of divacancy changes from negative to positive when the hydrostatic (biaxial) tensile strain is larger than 1.5% (2%). These results indicate that the compressive strain will facilitate the formation of monovacancy in W, while the tensile strain will enhance the attraction between vacancies. This can be attributed to the redistribution of electronic states of W atoms surrounding vacancy. Furthermore, although the migration energy of the monovacancy also exhibits a monotonic linear dependence on the hydrostatic strain, it shows a parabola with an opening down under the biaxial strain. Namely, the vacancy mobility will always be promoted by biaxial strain in W, almost independent of the sign of strain. Such unexpected anisotropic strain-enhanced vacancy mobility originates from the Poisson effect. On the basis of the first-principles results, the nucleation of vacancy clusters in strained W is further determined with the object kinetic Monte Carlo simulations. It is found that the formation time of tri-vacancy decrease significantly with the increasing of tensile strain, while the vacancy clusters are not observed in compressively strained W, indicating that the tensile strain can enhance the formation of voids. Our results provide a good reference for understanding the vacancy behaviors in W.

AIM:To investigate the impact of titanium dioxide nanoparticles (TiO2 NPs) on embryonic development and retinal neurogenesis. METHODS:The agglomeration and sedimentation of TiO2 NPs solutions at different dilutions were observed, and the ultraviolet-visible spectra of their supernatants were measured. Zebrafish embryos were experimentally exposed to TiO2 NPs until 72h postfertilization (hpf). The retinal neurogenesis and distribution of the microglia were analyzed by immunohistochemistry and whole mount in situ hybridization. RESULTS: The1 mg/L was determined to be an appropriate exposure dose. Embryos exposed to TiO2 NPs had a normal phenotype. The neurogenesis was initiated on time, and ganglion cells, cones and rods were well differentiated at 72 hpf. The expression of fms mRNA and the 4C4 antibody, which were specific to microglia in the central nervous system (CNS), closely resembled their endogenous profile. CONCLUSION:These data demonstrate that short-term exposure to TiO2 NPs at a low dose does not lead to delayed embryonic development or retinal neurotoxicity.

Abstract Immunotherapy has revolutionized cancer treatment, but the lack of a reliable predictive biomarker for treatment response remains a challenge. Alpha-1,6-Mannosylglycoprotein 6-β-N-Acetylglucosaminyltransferase 5 (MGAT5) is a key regulator of complex N-glycan synthesis, and its dysregulation is associated with cancer progression. The lectin Phaseolus vulgaris leukoagglutinin (PHA-L) specifically binds to mature MGAT5 products. Previous studies have indicated elevated PHA-L staining in head and neck squamous cell carcinoma (HNSCC), which implies increased activity of MGAT5. However, the specific role of MGAT5 in HNSCC remains unclear. In this study, we found significantly higher PHA-L staining and MGAT5 expression in HNSCC tumors compared to adjacent non-tumor tissues. Using a mass spectrometry (MS)-based glycoproteomic approach, we identified 163 potential protein substrates of MGAT5. Functional analysis revealed that protein substrates of MGAT5 regulated pathways related to T cell proliferation and activation. We further discovered that PD-L1 was among the protein substrates of MGAT5, and the expression of MGAT5 protected tumor cells from cytotoxic T lymphocyte (CTL) killing. Treatment of nivolumab alleviated the protective effects of MGAT5 on CTL activity. Consistently, patients with MGAT5-positive tumors showed improved responses to immunotherapy compared to those with MGAT5-negative tumors. Using purified PD-L1 from HNSCC cells and a glycoproteomic approach, we further deciphered that the N35 and N200 sites carry the majority of complex N-glycans on PD-L1. Our findings highlight the critical role of MGAT5-mediated branched N-glycans on PD-L1 in modulating the interaction with the immune checkpoint receptor PD-1. Consequently, we propose that MGAT5 could serve as a biomarker to predict patients’ responses to anti-PD-1 therapy. Furthermore, targeting the branched N-glycans at N35 and N200 of PD-L1 may lead to the development of novel diagnostic and therapeutic approaches.

Mesenchymal stem cells (MSCs) have gained attention as a promising therapeutic approach in both preclinical and clinical osteoarthritis (OA) settings. Various joint cell types, such as chondrocytes, synovial fibroblasts, osteoblasts, and tenocytes, can produce and release extracellular vesicles (EVs), which subsequently influence the biological activities of recipient cells. Recently, extracellular vesicles derived from mesenchymal stem cells (MSC-EVs) have shown the potential to modulate various physiological and pathological processes through the modulation of cellular differentiation, immune responses, and tissue repair. This review explores the roles and therapeutic potential of MSC-EVs in OA and rheumatoid arthritis, cardiovascular disease, age-related macular degeneration, Alzheimer's disease, and other degenerative diseases. Notably, we provide a comprehensive summary of exosome biogenesis, microRNA composition, mechanisms of intercellular transfer, and their evolving role in the highlight of exosome-based treatments in both preclinical and clinical avenues.

Abstract Proteins with intrinsically disordered regions (IDRs) often undergo phase separation to control their functions spatiotemporally. Changing the pH alters the protonation levels of charged sidechains, which in turn affects the attractive or repulsive force for phase separation. In a cell, the rupture of membrane‐bound compartments, such as lysosomes, creates an abrupt change in pH. However, how proteins’ phase separation reacts to different pH environments remains largely unexplored. Here, using extensive mutagenesis, NMR spectroscopy, and biophysical techniques, it is shown that the assembly of galectin‐3, a widely studied lysosomal damage marker, is driven by cation‐π interactions between positively charged residues in its folded domain with aromatic residues in the IDR in addition to π–π interaction between IDRs. It is also found that the sole two negatively charged residues in its IDR sense pH changes for tuning the condensation tendency. Also, these two residues may prevent this prion‐like IDR domain from forming rapid and extensive aggregates. These results demonstrate how cation‐π, π–π, and electrostatic interactions can regulate protein condensation between disordered and structured domains and highlight the importance of sparse negatively charged residues in prion‐like IDRs.

Abstract The purpose of this study was to establish an integrated predictive model that combines clinical features, DVH, radiomics, and dosiomics features to predict RIHT in patients receiving tomotherapy for nasopharyngeal carcinoma. Data from 219 patients with nasopharyngeal carcinoma were randomly divided into a training cohort (n = 175) and a test cohort (n = 44) in an 8:2 ratio. RIHT is defined as serum thyroid-stimulating hormone (TSH) greater than 5.6 μU/mL, with or without a decrease in free thyroxine (FT4). Clinical features, 27 DVH features, 107 radiomics features and 107 dosiomics features were extracted for each case and included in the model construction. The least absolute shrinkage and selection operator (LASSO) regression method was used to select the most relevant features. The eXtreme Gradient Boosting (XGBoost) was then employed to train separate models using the selected features from clinical, DVH, radiomics and dosiomics data. Finally, a combined model incorporating all features was developed. The models were evaluated using receiver operating characteristic (ROC) curves and decision curve analysis. In the test cohort, the area under the receiver operating characteristic curve (AUC) for the clinical, DVH, radiomics, dosiomics and combined models were 0.798 (95% confidence interval [CI], 0.656–0.941), 0.673 (0.512–0.834), 0.714 (0.555–0.873), 0.698 (0.530–0.848) and 0.842 (0.724–0.960), respectively. The combined model exhibited higher AUC values compared to other models. The decision curve analysis demonstrated that the combined model had superior clinical utility within the threshold probability range of 1% to 79% when compared to the other models. This study has successfully developed a predictive model that combines multiple features. The performance of the combined model is superior to that of single-feature models, allowing for early prediction of RIHT in patients with nasopharyngeal carcinoma after tomotherapy.

Coronaviruses (CoVs) are enveloped single-stranded RNA viruses that predominantly attack the human respiratory system. In recent decades, several deadly human CoVs, including SARS-CoV, SARS-CoV-2, and MERS-CoV, have brought great impact on public health and economics. However, their high infectivity and the demand for high biosafety level facilities restrict the pathogenesis research of CoV infection. Exacerbated inflammatory cell infiltration is associated with poor prognosis in CoV-associated diseases. In this study, we used human CoV 229E (HCoV-229E), a CoV associated with relatively fewer biohazards, to investigate the pathogenesis of CoV infection and the regulation of neutrophil functions by CoV-infected lung cells. Induced pluripotent stem cell (iPSC)-derived alveolar epithelial type II cells (iAECIIs) exhibiting specific biomarkers and phenotypes were employed as an experimental model for CoV infection. After infection, the detection of dsRNA, S, and N proteins validated the infection of iAECIIs with HCoV-229E. The culture medium conditioned by the infected iAECIIs promoted the migration of neutrophils as well as their adhesion to the infected iAECIIs. Cytokine array revealed the elevated secretion of cytokines associated with chemotaxis and adhesion into the conditioned media from the infected iAECIIs. The importance of IL-8 secretion and ICAM-1 expression for neutrophil migration and adhesion, respectively, was demonstrated by using neutralizing antibodies. Moreover, next-generation sequencing analysis of the transcriptome revealed the upregulation of genes associated with cytokine signaling. To summarize, we established an in vitro model of CoV infection that can be applied for the study of the immune system perturbations during severe coronaviral disease.

Photo-assisted lithium sulfur batteries (PA-LSBs) provide vital and sustainable protocols for promoting sulfur redox reactions via powerful photoinduced effects. However, precise control of the stepwise adsorption, diffusion and photocatalytic conversion of polysulfides at the surface of photocatalysts is required to accelerate the photo-assisted process. Herein, optical field and built-in electric field synergistically-assisted LSBs are developed with a p-n junction of Co3O4-TiO2 on the carbon cloth, possessing a spontaneously generated built-in electric field and a well-matched energy band structure with sulfur redox reactions. Under light irradiation, the directional migration of soluble polysulfides and the space separation of photogenerated carriers are achieved with the synergistical coupling of the optical field and built-in electric field to precisely regulate the selective deposition of Li2S and inhibit the shuttle effect via an effective photocatalytic-promoted process, leading to a maximum capacity of 1087 mAh g−1 at 2 C and a low capacity attenuation of 0.068% per cycle at 5 C. A high areal capacity of 9.6 mAh cm−2 and a great potential photo-charge process can be realized with light irradiation. Furthermore, the stability of lithium metal anodes is improved accordingly. This work demonstrates a new insight to develop high-performance LSBs with a multifield synergistical coupling protocol.
