ObjectiveFamine exposure and higher serum calcium levels are related with increased risk of many disorders, including Alzheimer’s disease, atherosclerosis, diabetes, and osteoporosis. Whether famine exposure has any effect on serum calcium level is unclear. Besides, the normal reference range of serum calcium is variable among different populations. Our aims are 1) determining the reference interval of calcium in Chinese adults; 2) exploring its relationship with famine exposure.MethodsData in this study was from a cross-sectional study of the epidemiologic investigation carried out during March-August 2010 in Jiading district, Shanghai, China. Nine thousand and two hundred eleven participants with estimated glomerular filtration rate (eGFR) ≥60ml/min/1.73m2 were involved to calculate reference interval of total calcium from 10569 participants aged 40 years or older. The analysis of famine exposure was conducted in 9315 participants with complete serum biochemical data and birth year information.ResultsAfter rejecting outliers, the 95% reference interval of total serum calcium was 2.122~2.518 mmol/L. The equation of albumin-adjusted calcium was: Total calcium + 0.019* (49-Albumin), with a 95% reference interval of 2.151~2.500 mmol/L. Compared to the age-balanced control group, there was an increased risk of being at the upper quartile of total serum calcium (OR=1.350, 95%CI=1.199-1.521) and albumin-adjusted calcium (OR=1.381, 95%CI=1.234-1.544) in subjects experienced famine exposure in childhood. Females were more vulnerable to this impact (OR= 1.621, 95%CI= 1.396-1.883 for total serum calcium; OR=1.722, 95%CI= 1.497-1.980 for albumin-adjusted calcium).ConclusionsFamine exposure is an important environmental factor associated with the changes in circulating calcium concentrations, the newly established serum calcium normal range and albumin-adjusted calcium equation, together with the history of childhood famine exposure, might be useful in identifying subjects with abnormal calcium homeostasis and related diseases, especially in females.

Fluorescence imaging has been an indispensable tool to provide dynamic information about the localization and quantity of organisms. Meanwhile, due to the intrinsic hollow structure and modularized biofunctionalities, polymer vesicles have been widely applied in biomedical field. However, most polymer vesicles are embedded with organic fluorophores for fluorescence imaging, which have certain drawbacks such as leakage and possible cytotoxicity. Here, we present a biodegradable polypeptide-based vesicle with intrinsic blue fluorescence without introducing any fluorophore for real-time visualization of antibacterial process. Through modular design to integrate multiple functional fragments, poly(e-caprolactone)-block-poly(tryptophan)-block-poly(lysine-stat-phenylalanine) [PCL25-b-PTrp2-b-P(Lys13-stat-Phe4)] was synthesized, where PCL chains form the hydrophobic membrane, P(Lys-stat-Phe) and PTrp provide intrinsic fluorescence and broad-spectrum antibacterial activity. It is noteworthy that the fluorescence emission was shifted from invisible ultraviolet range of amino acids to visible range (emission maximum at 436 nm), which makes it possible to visualize the antibacterial process. In addition, through utilizing the intrinsic fluorescence of vesicles, confocal fluorescent imaging of vesicles with bacteria validated the specific adhesion of vesicle towards bacteria, and the bacterial death through membrane disruption. Overall, we provided a novel approach to developing biodegradable fluorescent polypeptide-based vesicles for real-time visualization of antibacterial process.

Boletus littoreus (Boletaceae, Boletales) is described as a new species from tropical China based on morphological and molecular evidence from the nuclear ribosomal large subunit RNA (28S), the translation elongation factor 1-α gene (TEF1), and the RNA polymerase II second largest subunit gene (RPB2). It is morphologically characterized by white basidiomata, a stipe without reticulations, a pileipellis composed of elliptic-fusiform to subglobose, sometimes filamentous, chains of hyphae, and is associated with trees of Dipterocarpaceae. An illustrated morphological description is provided along with comparison to related and similar species.

Age-related osteoporosis is characterized by the accumulation of senescent osteoblastic cells in bone microenvironment and significantly reduced osteogenic differentiation. Clearing of the senescent cells is helpful to improve bone formation in aged mice. Bone morphogenetic protein 9 (BMP9), a multifunctional protein produced and secreted by liver, was reported to improve osteoporosis caused by estrogen withdrawal. However, the mechanism of BMP9 has not been fully elucidated, and its effect on senile osteoporosis has not been reported. This study reveals that BMP9 significantly increases bone mass and improves bone biomechanical properties in aged mice. Furthermore, BMP9 reduces expression of senescent genes in bone microenvironment, accompanied by decreased senescence-associated secretory phenotypes (SASPs) such as Ccl5, Mmp9, Hmgb1, Nfkb1, and Vcam1. In vitro, Bmp9 treatment inhibits osteoblast senescence through activating Smad1, which suppresses the transcriptional activity of Stat1, thereby inhibits P21 expression and SASPs production. Furthermore, inhibiting the Smad1 signal in vivo can reverse the inhibitory effect of BMP9 on Stat1 and downstream senescent genes, which eliminates the protection of BMP9 on age-related osteoporosis. These findings highlight the critical role of BMP9 on reducing age-related bone loss by inhibiting osteoblast senescence through Smad1-Stat1-P21 axis.

In order to investigate the influence of the auxiliary ligand of the cyanidometal bridge on metal to metal charge transfer (MMCT) in cyanidometal-bridged complexes, two groups of heterotrimetallic cyanidometal-bridged complexes, trans-[Cp*(dppe)Fe-NC-Ru(L)2-CN-Fe(dppe)Cp*][PF6]n (L = bpy, 1(PF6)n; L = 4,4'-dmbpy, 2(PF6)n; n = 2, 3, 4) (Cp* = 1,2,3,4,5-pentamethylcyclopentadiene, dppe = 1,2-bis(diphenylphosphino)ethane, bpy = 2,2'-bipyridine, 4,4'-dmbpy = 4,4'-dimethyl-2,2'-bipyridyl) were synthesized and fully characterized. The MMCT of the one-electron oxidation mixed valence complexes is mainly attributed to RuII and FeII → FeIII MMCT transitions, and the MMCT of the two-electron oxidation complexes is mainly attributed to RuII → FeIII MMCT transitions. The energy of the MMCT of the four complexes decreases with the increase of the electron donating ability of the auxiliary ligand of the cyanidometal bridge. The IR, EPR, and Mössbauer spectra, and the solvent independence of MMCT characterizations indicate that the one-electron oxidation mixed valence complexes may belong to Class II-III systems, and the two-electron oxidation complexes may be localized at low temperature but delocalized at room temperature on the EPR timescale.

PHYTOCHROME INTERACTING FACTORs (PIFs) are a subset of helix-loop-helix (bHLH) transcription factors, which play critical roles in plant growth and development, as well as in adaption to ambient environments. However, PIF members have not been completely identified in apple (Malus domestica), a widely distributed fruit crop with significant economical importance. Here, we characterized MdPIF3, the homolog of AtPIF3, and determined its role in response to abiotic stresses in apple. We first analyzed its gene and protein structure, and found that it contained bHLH domain, active phytochrome B binding (APB) motif, as well as active phytochrome A binding (APA) motif. Yeast-two-hybrid assays indicated that MdPIF3 formed a homodimer by itself and heterodimers with other MdPIFs. Moreover, MdPIF3 was responsive to light and cold treatment at both transcriptional and post-translational levels. Overexpression of MdPIF3 reduced cold tolerance but enhanced drought resistance in both apple callus and Arabidopsis. The bHLH-type protein, MdPIF3, plays a key role in cold and drought tolerance in plants.

Objective To compare the prognostic value of electronic SOFA (eSOFA) scores and simplified SOFA (sSOFA) scores in evaluating the prognosis of patients with sepsis. Methods A retrospective analysis was conducted of clinical data of 245 patients with sepsis admitted to the Department of Emergency and Critical Care Medicine, Shanghai Pudong Hospital from August 2016 to August 2019. Clinical end points were survival status 28 days after hospital admission. eSOFA score, sSOFA score, qSOFA score and SOFA score were evaluated to predict patient prognosis using the area under the receiver operating characteristic curve (AUC). Results AUC of eSOFA score, sSOFA score, qSOFA score and SOFA score for predicting the prognosis of the patients was 0.757 (95% CI 0.686-0.828), 0.721 (95% CI 0.665-0.787), 0.662 (95% CI 0.586-0.738), 0.806 (95% CI 0.747-0.866, respectively. The eSOFA score was superior to the sSOFA score (Z=2.317, P=0.021) in terms of AUC. Conclusions The eSOFA and sSOFA scores have predictive value for the prognosis of sepsis, and eSOFA score is better than sSOFA score. DOI: 10.11855/j.issn.0577-7402.2020.07.11

Background & AimsPrimary hyperparathyroidism(PHPT) has been evolving into a milder asymptomatic disease. No study has assessed the association between famine exposure and such a shift. We aim to explore the effects of China’s Great Famine exposure on the changing pattern of PHPT phenotypes.Methods750 PHPT patients diagnosed from 2000 to 2019 were studied. The clinical presentations were compared between them in recent 10 years (2010-2019) and previous 10 years (2000-2009). Participants were then categorized into fetal, childhood, adolescent, adult exposure, and unexposed groups. Logistic regression was used to estimate the odds ratios (ORs) and confidence intervals (CIs) of famine exposure as factors contributing to the changes in the clinical presentations of PHPT.ResultsSerum levels of PTH, albumin-corrected Ca, tumor size, eGFR, BMDs (all Prd tertile PTH were 2.79(1.34,5.8), 2.07(1.04,4.11), 3.10(1.15,8.38) and 8.85(2.56,30.56) for patients with fetal, childhood, adolescent and adult famine exposure, respectively. The ORs (95%CIs) of upper 3rd tertile albumin-corrected Ca and upper 3rd tertile of tumor size was 4.78(1.39, 16.38) and 4.07(1.12,14.84) for participants with adult famine exposure, respectively. All these associations were independent of age, sex, disease duration and other confounders.ConclusionsThe clinical manifestations of PHPT in China continue to be milder. Exposure to famine is associated with PHPT. Less famine exposure might be responsible for the mile form of PHPT in recent years.

This study aims to investigate the current situation of pelvic floor muscle (PFM) in the second trimester postpartum 42 days by detecting the electrophysiological indexes of PFM on pelvic floor rehabilitation after 2 months.In total, 198 cases of second child puerpera were selected, who were delivered in our hospital between June 1, 2020, and December 10, 2020, and underwent outpatient reexamination 42 days after delivery.Except for the rest post-baseline stage values, the pelvic floor surface electromyography evaluation values significantly differed from each other at the considered time points in group A, P .05; on day 72 in group B, no obvious improvement in the evaluation values was observed as than those on day 42 (P .05) except for the endurance contractions stage values. However, on day 102, all values were markedly different at each considered time point (P .05). On day 102 postpartum, the evaluation values of group A in the rest pre-baseline stage, the time before and after peak of phasic (flick) contractions stage, and the endurance contractions stage were significantly improved to those in group B with all P .05. On day 42 after parturition, each index of the tonic contractions stage was higher after spontaneous labor than that after cesarean; the differences were all significant, P .05, but on day 102 postpartum, all of the values exhibited no difference between the 2 modes. In only the phasic (flick) contractions stage at 42th, were the values of younger mothers obviously higher, P = .025; the other stage values for different ages of women during different time periods were not statistically significant, P .05.In the short term, the effect of biofeedback plus electrical stimulation on the PFM function in second pregnant women was better than that of the Kegel exercise, but with time, there was no significant difference between the 2 training methods on the recovery of the PFM.

Background The factors that predict the progression of Mycoplasma pneumoniae infection remain inconclusive. Therefore, we investigated macrolide resistance prevalence, M pneumoniae genotype, and clinical characteristics of childhood M pneumoniae respiratory tract infections in Taiwan. Methods A total of 295 children hospitalized with respiratory tract infections with positive serological M pneumoniae immunoglobulin M test results were enrolled in this 3-year prospective study. Oropharyngeal swabs were obtained for M pneumoniae cultures and polymerase chain reaction tests. All M pneumoniae specimens were further characterized by P1 typing, multilocus variable-number tandem-repeat analysis (MLVA), and macrolide resistance genotyping. The clinical characteristics and blood cytokine profiles were analyzed accordingly. Results Of 138 M pneumoniae specimens, type I P1 was the predominant (136 of 138, 98.6%). The MLVA type P (4-4-5-7-2) was the leading strain (42 of 138, 30.4%), followed by type J, U, A, and X. The overall macrolide-resistant rate was 38.4% (53 of 138); the resistance rate increased dramatically yearly: 10.6% in 2017, 47.5% in 2018, and 62.5% in 2019 (P, We present a 3-year prospective study on the molecular epidemiology, clinical characteristics, and cytokine profiles of childhood M pneumoniae infection in Taiwan. An increased macrolide resistance rate associated with emergence of MLVA type U was noted.

On the β plane approximation, the two-layer quasigeostrophic mode is used to study the baroclinic instability of the quadric shear basic zonal flows on a uniform bottom topography. The phase speed and growth rate of instability waves are functions of the shear zonal basic flows and bottom topographic slope. The study focus is on the effects of topography and the quadric shear basic zonal flows (the second derivative of basic zonal flows is not zero). The meridional slope destabilise (stabilise) zonal flows, it plays an unstable role in disturbance, moreover the effect of the second derivative of basic zonal flows is to accelerate the instability of disturbance. The zonal slope always destabilises the zonal basic flows through zonal and meridional wavenumber. Moreover, the effect of the second derivative of basic zonal flows is to accelerate the instability of disturbance.

Strigolactones (SLs) are phytohomones that regulate shoot branching and hypocotyl elongation. Here, the strigolactone receptor gene MdD14 in apple was cloned. MdD14 was localized to the nucleus and cytoplasm. GUS staining showed that MdD14 was expressed in various tissues. A promoter analysis revealed that MdD14 contained multiple response elements and its expression levels were induced by various abiotic stress treatment. The post-translational of MdD14 was regulated by the SL analog GR24. Ectopic expression of MdD14 in Arabidopsis is resulted in a phenotype that inhibited shoot branching and hypocotyl elongation, as well as increased tolerance to salt, drought and low temperature stresses. These findings reveal the functions of MdD14 and lay a foundation for studying SLs in apple. The apple MdD14 plays a key role in the SL signal transduction pathway and is involved in various abiotic stress resistance.

An unusual tetra-nuclear linear cyanido-bridged complex [Ru2 (μ-ap)4 -CN-Ru2 (μ-ap)4 ](BPh4 ) (1) (ap=2-anilinopyridinate) has been synthesized and well characterized. The crystallographic data, magnetic measurement, IR, EPR and theoretical calculation results demonstrate that complex 1 is the first example of mixed spin Ru2 5+ -based complex with uncommon electronic configurations of S=1/2 for the cyanido-C bound Ru2 5+ and S=3/2 for the cyanido-N bound Ru2 5+ . This phenomenon can be understood by the theoretical calculation results that from the precursor Ru2 (μ-ap)4 (CN) (S=3/2) to complex 1 the energy gap between π* and δ* orbitals of the cyanido-C bound Ru2 5+ core increases from 0.57 to 1.61 eV due to the enhancement of asymmetrical π back-bonding effect, but that of the cyanido-N bound Ru2 5+ core is essential identical (0.56 eV). Besides, the analysis of UV/Vis-NIR spectra suggests that there exists metal to metal charge transfer (MMCT) from the cyanido-N bound Ru2 5+ (S=3/2) to the cyanido-C bound Ru2 5+ (S=1/2), supported by the TDDFT calculations.

Objective: To compare the molecular epidemiology of methicillin-resistant Staphylococcus aureus (MRSA) isolates between an old, urban hospital and a new, rural hospital over the same time period. Methods: The molecular characteristics of 398 MRSA bloodstream isolates collected between 2007 and 2013 from two hospitals in Taiwan were analyzed retrospectively; 202 isolates were from the old hospital and 196 from the new hospital (opened in 2007). Lgr4, a G-protein-coupled receptor, is associated with various physiological and pathological processes including oncogenesis, energy metabolism, and bone remodeling. However, whether Lgr4 is involved in osteoblasts' metabolism is not clear. Here we uncover that in preosteoblast cell line, lacking Lgr4 results in decreased osteogenic function along with reduced glucose consumption, glucose uptake, and lactate production in the presence of abundant oxygen, which is referred to as aerobic glycolysis. Activating canonical Wnt/β-catenin signaling rescued the glycolytic dysfunction. Lgr4 promotes the expression of pyruvate dehydrogenase kinase 1 (pdk1) and is abolished by interfering canonical Wnt/β-catenin signaling. Mice lacking Lgr4 specifically in osteoblasts (Lgr4osb-/- ) exhibit decreased bone mass and strength due to reduced bone formation. Additionally, glycolysis of osteoblasts is impaired in Lgr4osb-/- mice. Our study reveals a novel function of Lgr4 in regulating the cellular metabolism of osteoblasts. © 2021 American Society for Bone and Mineral Research (ASBMR).

The synthesis and characterization of Class II-III mixed valence complexes have been an interesting topic due to their special intermediate behaviour between localized and delocalized mixed valence complexes. To investigate the influence of the isocyanidometal bridge on metal-to-metal charge transfer (MMCT) properties, a family of new isocyanidometal-bridged complexes and their one-electron oxidation products cis-[Cp(dppe)Fe-CN-Ru(L)2 -NC-Fe(dppe)Cp][PF6 ]n (n=2, 3) (Cp=1,3-cyclopentadiene, dppe=1,2-bis(diphenylphosphino)ethane, L=2,2'-bipyridine (bpy, 1[PF6 ]n ), 5,5'-dimethyl-2,2'-bipyridyl (5,5'-dmbpy, 2[PF6 ]n ) and 4,4'-dimethyl-2,2'-bipyridyl (4,4'-dmbpy, 3[PF6 ]n )) have been synthesized and fully characterized. The experimental results suggest that all the one-electron oxidation products may belong to Class II-III mixed valence complexes, supported by TDDFT calculations. With the change of the substituents of the bipyridyl ligand on the Ru centre from H, 5,5'-dimethyl to 4,4'-dimethyl, the energy of MMCT for the one-electron oxidation complexes changes in the order: 13+ 34+ >24+ . The potential splitting (ΔE1/2 (2)) between the two terminal Fe centres for N[PF6 ]2 are the largest potential splitting for the cyanido-bridged complexes reported so far. This work shows that the smaller potential difference between the bridging and the terminal metal centres would result in the more delocalized mixed valence complex.

Nitrogen is an important factor that affects plant anthocyanin accumulation. In apple, the nitrate-responsive BTB/TAZ protein MdBT2 negatively regulates anthocyanin biosynthesis. In this study, we found that MdBT2 undergoes posttranslational modifications in response to nitrate deficiency. Yeast two-hybrid, protein pull-down, and bimolecular fluorescence complementation (BiFC) assays showed that MdBT2 interacts with MdGRF11, a 14-3-3 protein; 14-3-3 proteins compose a family of highly conserved phosphopeptide-binding proteins involved in multiple physiological and biological processes. The interaction of MdGRF11 negatively regulated the stability of the MdBT2 protein via a 26S proteasome-dependent pathway, which increased the abundance of MdMYB1 proteins to activate the expression of anthocyanin biosynthesis-related genes. Taken together, the results demonstrate the critical role of 14-3-3 proteins in the regulation of nitrate deficiency-induced anthocyanin accumulation. Our results provide a novel avenue to elucidate the mechanism underlying the induction of anthocyanin biosynthesis in response to nitrate deficiency.

Nitrate acts as a vital signal molecule in the modulation of plant growth and development. The phytohormones gibberellin (GA) is also involved in this process. However, the exact molecular mechanism of how nitrate and GA signaling pathway work together in regulating plant growth remains poorly understood. In this study, we found that a nitrate-responsive BTB/TAZ protein MdBT2 participates in regulating nitrate-induced plant growth in apple (Malus × domestica). Yeast two-hybridization, protein pull-down, and bimolecular fluorescence complementation (BiFC) assays showed that MdBT2 interacts with a DELLA protein MdRGL3a, which is required for the ubiquitination and degradation of MdRGL3a proteins via a 26S proteasome-dependent pathway. Furthermore, heterologous expression of MdBT2 partially rescued growth inhibition caused by overexpression of MdRGL3a in Arabidopsis. Taken together, our findings indicate that MdBT2 promotes nitrate-induced plant growth partially through reducing the abundance of the DELLA protein MdRGL3a.

In this paper, the influence of quadric shear basic Zonal flows and β on the downstream development of unstable chaotic baroclinic waves is studied from the two-layer model in wide channel controlled by quasi geostrophic potential vorticity equation. Through the obtained Lorentz equation, we focused on the influence of the quadric shear zonal flow (the second derivative of the basic zonal flow is constant) on the downstream development of baroclinic waves. In the absence of zonal shear flow, chaotic behavior along feature points would occur, and the amplitude would change rapidly from one feature to another, that is, it would change very quickly in space. When zonal shear flow is introduced, it will smooth the solution of the equation and reduce the instability, and with the increase of zonal shear flow, the instability in space will increase gradually. So the quadric shear zonal flow has great influence on the stability in space.

Phytochrome-interacting factors (PIFs), members of the basic helix-loop-helix transcription factor family that have been extensively investigated in Arabidopsis thaliana, play essential roles in plant growth and development. However, PIF members have not been systematically investigated in apples, a worldwide perennial woody crop of economic importance. Here, seven PIF genes were identified from the Malus &times, domestica reference genome. Chromosomal locations, gene structures, and phylogenetic relationships of these members were analyzed. Analysis of cis-acting elements in promoter regions of MdPIF genes indicated that various elements were related to light, abiotic stress, and plant hormone responsiveness. Subsequently, subcellular localization and transcriptional activity analysis revealed that MdPIFs were typical nuclear transcription factors with transcriptional activation ability. Expression analysis demonstrated that MdPIF genes had different gene expression patterns for various abiotic factors. Moreover, overexpressed MdPIF4 reduced the sensitivity of apple calluses to abscisic acid (ABA). Our work lays foundations for further investigation of PIF functions in plant growth and development in apples.

This study identified a new bHLHm1 transcription factor MdSAT1 which functioned in mediating tolerance to salt and drought resistance. Changes in the expression of stress-related genes play crucial roles in response to environmental stress. Basic helix-loop-helix (bHLH) proteins are the largest superfamily of transcription factors and a large number of bHLH proteins function in plant responses to abiotic stresses. We identified a new bHLHm1 transcription factor from apple and named it MdSAT1. β-Glucuronidase (GUS) staining showed that MdSAT1 expressed in various tissues with highly expressed in leaves. Promoter analysis revealed that MdSAT1 contained multiple response elements and its transcription was induced by several environmental cues, particularly salt and drought stresses. Overexpression of MdSAT1 in apple calli and Arabidopsis resulted in a phenotype of increased tolerance to salt and drought. Altering abscisic acid (ABA) treatment increased the sensitivity of MdSAT1-OE Arabidopsis to ABA, and heavy metal stress, osmotic stress, and ethylene did not participate in MdSAT1 mediated plant development. These findings reveal the abiotic stress functions of MdSAT1 and pave the way for further functional investigation.

Bacterial strains of the Bacillus cereus group produce various toxins that cause diarrheal and emetic food poisoning. In this study, five main oyster farming areas and 15 fishing ports in Taiwan were examined for the status of B. cereus group bacteria inhabiting seawater and shellfish. On average, bacteria of the B. cereus group were detected in 32.6% of the seawater samples (n = 89) and 2.5% of the oysters (n = 81) in the oyster farming areas and in 7.9% of the seawater samples (n = 202) and 0.68% of the shellfish products (n = 292) in fishing ports. To trace the potential source of B. cereus group bacteria in intertidal oyster farming areas, we simultaneously explored their terrestrial river basins. In total, 44 B. cereus group strains were purified and cultured from water and shellfish for the analysis of virulence genes, panC gene typing, antibiotic susceptibility testing, and enterobacterial repetitive intergenic consensus PCR (ERIC-PCR) fingerprinting. The enterotoxin-coding genes nheABC, hblCDA, entFM, cytK-1, and cytK-2 were detected in 70.4%, 84.1%, 97.7%, 72.7%, and 75% of the total isolates, among which 40.9% carried all these genes. According to panC gene analysis, the dominant isolates belonged to the panC group IV. In antibiotic susceptibility tests, most B. cereus group isolates were resistant to ampicillin (97.7%) and sulfamethoxazole/trimethoprim (100%). The percentage of multidrug resistant B. cereus group isolates was 34.6%. Finally, the 44 B. cereus group isolates were classified into 43 types and categorized into five clusters using ERIC-PCR fingerprinting. The B. cereus group isolates from different oyster farming areas were concentrated within the two main clusters; however, those from river basins displayed a wide genetic diversity, indicating the presence of multiple sources of B. cereus group bacteria in river basins.
