Abstract Background Individuals with sensory processing sensitivity (SPS) tend to be overreactive in response to negative environmental stimuli. More is known about the positive relationship between SPS and quality of life (QoL); nevertheless, less is known regarding the roles of resilience and social determinants in this association. This research aimed to investigate the potential mediation effect of resilience and the moderation effect of social determinants on the relationship between SPS and QoL in a large sample of Chinese cancer patients. Methods We used the most recent datasets from an ongoing project conducted in southwest China. A two-stage random sampling strategy with a probability proportionate to sample size (PPS) design was adopted. The associations between resilience, SPS, and QoL were evaluated using a linear regression model. Path analysis was adopted to examine the mediation of resilience. Results Resilience was positively associated with quality of life, while increased sensory processing sensitivity was negatively associated with quality of life. The restricted cubic spline analysis revealed that as resilience increased, the coefficients of quality of life rapidly increased across all domains. Conversely, the coefficients for quality of life gradually decreased with the escalation of sensory processing sensitivity. Resilience was a significant mediator, accounting for 21.88% of the total SPS-QoL association. The mediation effect of resilience varied across ethnicity and sex. Conclusion Sensory processing sensitivity was significantly associated with quality of life in cancer patients, and promoting resilience could mitigate this negative impact. However, the effect of resilience varies across sex and ethnicity. Therefore, targeted resilience promotion interventions, especially those integrating social characteristics, should be considered for implementation.

BackgroundThere is still a lack of clinically validated biomarkers to screen lung cancer patients suitable for programmed dead cell-1 (PD-1)/programmed dead cell receptor-1 (PD-L1) immunotherapy. Detection of PD-L1 expression is invasively operated, and some PD-L1-negative patients can also benefit from immunotherapy; thus, the joint modeling of both deep learning images and clinical features was used to improve the prediction performance of PD-L1 expression in non-small cell lung cancer (NSCLC).MethodsRetrospective collection of 101 patients diagnosed with pathology in our hospital who underwent 18F FDG PET/CT scans, with lung cancer tissue Tumor Propulsion Score (TPS) ≥1% as a positive expression. Lesions were extracted after preprocessing PET/CT images, and using deep learning 3D DenseNet121 to learn lesions in PET, CT, and PET/CT images, 1,024 fully connected features were extracted; clinical features (age, gender, smoking/no smoking history, lesion diameter, lesion volume, maximum standard uptake value of lesions [SUVmax], mean standard uptake value of lesions [SUVmean], total lesion glycolysis [TLG]) were combined for joint modeling based on the structured data Category Embedding Model.ResultsArea under a receiver operating characteristic (ROC) curve (AUC) and accuracy of predicting PD-L1 positive for PET, CT, and PET/CT test groups were 0.814 ± 0.0152, 0.7212 ± 0.0861, and 0.90 ± 0.0605, 0.806 ± 0.023, 0.70 ± 0.074, and 0.950 ± 0.0250, respectively. After joint clinical feature modeling, the AUC and accuracy of predicting PD-L1 positive for PET/CT were 0.96 ± 0.00905 and 0.950 ± 0.0250, respectively.ConclusionThis study combines the features of 18F-FDG PET/CT images with clinical features using deep learning to predict the expression of PD-L1 in NSCLC, suggesting that 18F-FDG PET/CT images can be conducted as biomarkers for PD-L1 expression.

Background This study endeavored to develop a nicotinamide adenine dinucleotide (NAD+) metabolism-related biomarkers in gastric cancer (GC), which could provide a theoretical foundation for prognosis and therapy of GC patients. Methods In this study, differentially expressed genes (DEGs1) between GC and paraneoplastic tissues were overlapped with NAD+ metabolism-related genes (NMRGs) to identify differentially expressed NMRGs (DE-NMRGs). Then, GC patients were divided into high and low score groups by gene set variation analysis (GSVA) algorithm for differential expression analysis to obtain DEGs2, which was overlapped with DEGs1 for identification of intersection genes. These genes were further analyzed using univariate Cox and least absolute shrinkage and selection operator (LASSO) regression analyses to obtain prognostic genes for constructing a risk model. Enrichment and immune infiltration analyses further investigated investigate the different risk groups, and qRT-PCR validated the prognostic genes. Results Initially, we identified DE-NMRGs involved in NAD biosynthesis, with seven (DNAJB13, CST2, THPO, CIDEA, ONECUT1, UPK1B and SNCG) showing prognostic significance in GC. Subsequent, a prognostic model was constructed in which the risk score, derived from the expression profiles of these genes, along with gender, emerged as robust independent predictors of patient outcomes in GC. Enrichment analysis linked high-risk patients to synaptic membrane pathways and low-risk to the CMG complex pathway. Tumor immune infiltration analysis revealed correlations between risk scores and immune cell abundance, suggesting a relationship between NAD+ metabolism and immune response in GC. The prognostic significance of our identified genes was validated by qRT-PCR, which confirmed their upregulated expression in GC tissue samples. Conclusion In this study, seven NAD+ metabolism-related markers were established, which is of great significance for the development of prognostic molecular biomarkers and clinical prognosis prediction for gastric cancer patients.

Abstract Action recognition has been applied in fields such as smart homes, gaming, traffic management, and security monitoring. Motion recognition is helpful for biomechanical analysis, auxiliary training systems, table tennis robots, motion-sensing games, virtual reality and other fields. In our study, we collected data on table tennis skill motion, created the TTMD6 dataset, and analyzed the characteristics of table tennis paddle trajectories. We propose a motion recognition algorithm to recognize paddle trajectories. Other research has used multijoint data to identify actions, while we use only the paddle trajectory to recognize table tennis skill motions, accelerating the speed of motion recognition. Therefore, it is feasible to use paddle trajectories to recognize table tennis skill motions.

Members of Cytospora include saprobes, endophytes and important plant pathogens, which are widely distributed on various wood hosts and have a wide global distribution. In this study, the species definitions were conducted, based on multigene phylogeny (ITS, act, rpb2, tef1-α and tub2 genes) and comparisons of morphological characters. A total of 22 representative isolates obtained from 21 specimens in Fengtai District of Beijing City were identified as seven species of Cytospora, including four known species (C. albodisca, C. ailanthicola, C. euonymina, C. haidianensis) and three novel species (C. fengtaiensis, C. pinea, C. sorbariae). The results provide an understanding of the taxonomy of Cytospora species associated with canker and dieback diseases in Fengtai District, Beijing, China.

In the context of large-scale rural construction in China, scientific and effective cognition and evaluation of the visual quality of public space in traditional villages is very important for improving the spatial quality of traditional villages. This study builds a cognitive and evaluation mechanism about the visual quality of public space in traditional villages by focusing on eye tracking techniques in ergonomic analysis, combining the results of subjective questionnaires and empirical judgments about the spatial perception of traditional villages. Lixiang Village in Nanjing is selected as a case to explore a scientific analysis of the visual quality of public spaces in traditional villages by using eye tracking technology to collect the subjects’ observation of different types of public spaces in the village and combining the results of the subjective questionnaire. The results of the study showed that the correlations between subjects’ subjective ratings and eye movement data are not as strong, but only moderate for some of these indicators. Moreover, buildings and structures are the most important factors in shaping the richness of public space. Finally, relevant design suggestions and strategies are proposed: in traditional village public spaces, the visual quality of structures and surrounding enclosing buildings should be focused on enhancing, while the focus should be on optimizing the orientation of paths, and also on shaping the unique visual impression of each public space.

Microplastics (MPs) and benzo[a]pyrene (B[a]P) are prevalent environmental pollutants. Numerous studies have extensively reported their individual adverse effects on organisms. However, the combined effects and mechanisms of exposure in mammals remain unknown. Thus, this study aims to investigate the potential effects of oral administration of 0.5μm polystyrene (PS) MPs (1 mg/mL or 5 mg/mL), B[a]P (1 mg/mL or 5 mg/mL) and combined (1 mg/mL or 5 mg/mL) on 64 male SD rats by gavage method over 6-weeks. The results demonstrate that the liver histopathological examination showed that the liver lobules in the combined (5 mg/kg) group had blurred and loose boundaries, liver cord morphological disorders, and significant steatosis. The levels of AST, ALT, TC, and TG in the combined dose groups were significantly higher than those in the other groups, the combined (5 mg/kg) group had the lowest levels of antioxidant enzymes and the highest levels of oxidants. The expression of Nrf2 was lowest and the expression of P38, NF-κB, and TNF-α was highest in the combined (5 mg/kg) group. In conclusion, these findings indicate that the combination of PSMPs and B[a]P can cause the highest levels of oxidative stress and elicit markedly enhanced toxic effects, which cause severe liver damage.

Solar photovoltaic (PV) is one of the most environmental-friendly and promising resources for achieving carbon peak and neutrality targets. Despite their ecological fragility, China’s vast desert regions have become the most promising areas for PV plant development due to their extensive land area and relatively low utilization value. Artificial ecological measures in the PV plants can reduce the environmental damage caused by the construction activity and promote the ecological condition of fragile desert ecosystems, therefore yield both ecological and economic benefits. However, the understanding of the current status and ecological benefits of this approach in existing desert PV plants is limited. Here we surveyed 40 PV plants in northern China’s deserts to identify the ecological construction modes and their influencing factors. We quantified the ecosystem service value (ESV) provided by these PV plants using remote sensing data and estimated the potential for ESV enhancement. Our results show that PV plant construction in desert regions can significantly improve the ecosystem, even with natural restoration measures (M1) alone, resulting in a 74% increase in average fractional vegetation cover (FVC) during the growing season, although the maximum average FVC of only about 10%. The integrated mode M4, which combined artificial vegetation planting M2 and sand control measures M3, further enhance the average growing season FVC to 14.53%. Currently, 22.5% of plants lack ecological measures, 40% employ only a single measure, but 92% of new plants since 2017 have adopted at least one ecological construction mode. The main influencing factors include surface type, policy support, water resources, ecological construction costs, and scientific management guidance. If artificial ecological construction were incorporated, a significant ESV could be achieved in these PV plants, reaching $8.9 million (a 7.7-fold increase) if assuming a targeted 50% vegetation coverage. This study provides evidence for evaluating the ecological benefit and planning of large-scale PV farms in deserts.

Abstract Background Immunogenic cell death (ICD) stimulates adaptive immunity and holds significant promise in cancer therapy. Nevertheless, the influence of ICD-associated long non-coding RNAs (lncRNAs) on the prognosis of patients with lung squamous cell carcinoma (LUSC) remains unexplored. Methods We employed data from the The Cancer Genome Atlas (TCGA)database to identify ICD-related lncRNAs associated with the prognosis of LUSC using univariate Cox regression analysis. Subsequently, we utilized the LOSS regression model to construct a predictive risk model for assessing the prognosis of LUSC patients based on ICD-related lncRNAs. Our study randomly allocated187 TCGA patients into a training group and 184 patients for testing the predictive model. Furthermore, we conducted quantitative polymerase chain reaction (qPCR) analysis on 43 tumor tissues from LUSC patients to evaluate lncRNA expression levelsPearson correlation analysis was utilized to analyze the correlation of risk scores with positron emission tomography/computed tomography (PET/CT) parameters among LUSC patients. Results The findings from the univariate Cox regression revealed 16 ICD-associated lncRNAs linked to LUSC prognosis, with 12 of these lncRNAs integrated into our risk model utilizing the LOSS regression. Survival analysis indicated a markedly higher overall survival time among patients in the low-risk group compared to those in the high-risk group. The area under the Receiver operating characteristic (ROC) curve to differentiate high-risk and low-risk patients was 0.688. Additionally, the overall survival rate was superior in the low-risk group compared to the high-risk group. Correlation analysis demonstrated a positive association between the risk score calculated based on the ICD-lncRNA risk model and the maximum standard uptake value (SUVmax) (r = 0.427, P = 0.0043) as well as metabolic volume (MTV)of PET-CT (r = 0.360, P = 0.0177) in 43 LUSC patients. Conclusion We have successfully developed a risk model founded on ICD-related lncRNAs that proves effective in predicting the overall survival of LUSC patients.

Background:The mechanical properties of the aorta are particularly important in clinical medicine and forensic science, serving as basic data for further exploration of aortic disease or injury mechanisms.Objective:To study the influence of various factors (age, gender, test direction, anatomical location, and pathological characteristics) on the mechanical properties and thickness of the aorta.Methods:In this study, a total of 24 aortas (age range: 54–88 years old) were collected, one hundred and seventy-four dog-bone-shaped samples were made, and then the uniaxial tensile test was run, finally, pathological grouping was performed through histological staining.Results:Atherosclerotic plaques were mainly distributed near the openings of blood vessel branches. The distribution was most severe in the abdominal aorta, followed by the aortic arch. Aortic atherosclerosis was a more severe trend in the male group. In the comparison of thickness, there were no significant differences in age (over 50 years) and test direction, the average thickness of the aorta was greater in the male group than the female group and decreased progressively from the ascending aorta to the abdominal aorta. Comparing the mechanical parameters, various parameters are mainly negatively correlated with age, especially in the circumferential ascending aorta (εp “Y = −0.01402*X + 1.762, R2 = 0.6882”, εt “Y = −0.01062*X + 1.250, R2 = 0.6772”); the parameters of males in the healthy group were larger, while the parameters of females were larger in atherosclerosis group; the aorta has anisotropy, the parameters in the circumferential direction were greater than those in the axial direction; the parameters of the ascending aorta were the largest in the circumferential direction, the ultimate stress [σp “1.69 (1.08,2.32)”] and ultimate elastic modulus [E2“8.28 (6.67,10.25)”] of the abdominal aorta were significantly larger in the axial direction; In the circumferential direction, the stress [σp “2.2 (1.31,3.98)”, σt “0.13 (0.09,0.31)”] and ultimate elastic modulus (E2 “14.10 ± 7.21”) of adaptive intimal thickening were greater than those of other groups, the strain (εp “0.82 ± 0.17”, εt “0.53 ± 0.14”) of pathological intimal thickening was the largest in the pathological group.Conclusion:The present study systematically analyzed the influence of age, sex, test direction, anatomical site, and pathological characteristics on the biomechanical properties of the aorta, described the distribution of aortic atherosclerosis, and illustrated the characteristics of aortic thickness changes. At the same time, new insights into the grouping of pathological features were presented.

Abstract Cell-adhesive factors mediate adhesion of cells to substrates via peptide motifs such as the Arg–Gly–Asp (RGD) sequence. With the onset of sustainability issues, there is a pressing need to find alternatives to animal-derived cell-adhesive factors, especially for cell-cultivated food applications. In this paper, we show how data mining can be a powerful approach toward identifying fungal-derived cell-adhesive proteins and present a method to isolate and utilize these proteins as extracellular matrices (ECM) to support cell adhesion and culture in 3D. Screening of a protein database for fungal and plant proteins uncovered that ~5.5% of the unique reported proteins contain RGD sequences. A plot of fungi species vs RGD percentage revealed that 98% of the species exhibited an RGD percentage > = 1%. We observed the formation of protein particles in crude extracts isolated from basidiomycete fungi, which could be correlated to their stability towards particle aggregation at different temperatures. These protein particles were incorporated in 3D fiber matrices encapsulating mouse myoblast cells, showing a positive effect on cell alignment. We demonstrated a cell traction stress on the protein particles (from Flammulina velutipes) that was comparable to cells on fibronectin. A snapshot of the RGD-containing proteins in the fungal extracts was obtained by combining SDS-PAGE and mass spectrometry of the peptide fragments obtained by enzymatic cleavage. Therefore, a sustainable source of cell-adhesive proteins is widely available in the fungi kingdom. A method has been developed to identify candidate species and produce cell-adhesive matrices, applicable to the cell-cultivated food and healthcare industries.

This study adopted a quasi-experimental design to explore the effects of fundamental movement skill intervention on the acquisition and retention of physical activity levels and health-related fitness in children with autism spectrum disorder (ASD). In the experiments, 11 children received fundamental motor skill training (12 weeks, 60 min/session, 4 times/week), and 10 children maintained traditional physical activity. Assessments were performed using an ActiGraph GT3X+ accelerometer, health-related fitness pre–post intervention, and 1-month follow-up tests. The sedentary time during physical activity was significantly decreased (p = 0.01), and there were large changes in health-related physical fitness indicators, including significantly improved body composition (body mass index, F(1,19) = 8.631, p = 0.03, partial η2 = 0.312), muscle strength and endurance (sit-ups, F(1,19) = 3.376, p = 0.02, partial η2 = 0.151 and vertical jumps, F(1,19) = 5.309, p = 0.04, partial η2 = 0.218), and flexibility (sit and reach, F(1,19) = 36.228, p = 0.02, partial η2 = 0.656). Moreover, the follow-up tests showed that the children’s sedentary time continued to reduce, and the muscle strength and endurance (sit-ups, F(1,19) = 4.215, p = 0.01, partial η2 = 0.426) improved continuously after the intervention. Based on this study, actionable and regular fundamental movement skill programs can be provided in the future as an effective way to achieve the healthy development goals of children with ASD.

With the widespread adoption of battery technology in electric vehicles, there has been significant attention drawn to the increasing frequency of battery fire incidents. However, the jetting behavior and expansion force during the thermal runaway (TR) of batteries represent highly dynamic phenomena, which lack comprehensive quantitative description. This study addresses this gap by employing an enhanced experimental setup that synchronizes the video timing of cameras with a signal acquisition system, enabling the multidimensional quantification of signals, such as images, temperature, voltage, and pressure. It also provides a detailed description of the jetting behavior and expansion force characteristics over time for Li(Ni0.8Co0.1Mn0.1)O2 batteries undergoing thermal runaway in an open environment. The results from three experiments effectively identify key temporal features, including the timing of the initial jetting spark, maximum jetting velocity, jetting duration, explosion duration, and patterns of flame volume variation. This quantitative analytical approach proves effective across various battery types and conditions. The findings could offer scientific foundations and experimental strategies for parameter identification in fire prevention and thermal runaway model development.

Pheromones play an important role in mate choice in insects, and pheromone synthesis pathways are potential targets for the control of harmful insects, among which desaturation is of great significance in pheromone structural diversity. However, little is known about the desaturase genes regulating pheromone synthesis in Coleoptera. In this study, taking the internationally significant pest Anoplophora glabripennis as a research object, we identified 6 AglaFAD genes, all of which were mapped to the endoplasmic reticulum and shared a highly similar distribution of conserved domains. A phylogenetic analysis showed that AglaFAD1–2 and AglaFAD3–6 exerted desaturation at different positions of the acyl chain, respectively. In regard to the expression levels of these six AglaFADs in both sexes, six tissues and three developmental stages were analyzed by qPCR. Combined with the chemical composition of the female pheromones that have been identified, two candidate genes, AglaFAD2 and AglaFAD5, which are specifically expressed in females, were screened, showing higher expression levels before mating and significantly decreasing after mating. It is speculated that they may be involved in the biosynthesis of contact pheromones in females. These results provide a basis for detailed functional studies of candidate genes in insect pheromone synthesis.

Abstract Due to commonalities in pathophysiology, age-related macular degeneration (AMD) represents a uniquely accessible model to investigate therapies for neurodegenerative diseases, leading us to examine whether pathways of disease progression are shared across neurodegenerative conditions. Here we use single-nucleus RNA sequencing to profile lesions from 11 postmortem human retinas with age-related macular degeneration and 6 control retinas with no history of retinal disease. We create a machine-learning pipeline based on recent advances in data geometry and topology and identify activated glial populations enriched in the early phase of disease. Examining single-cell data from Alzheimer’s disease and progressive multiple sclerosis with our pipeline, we find a similar glial activation profile enriched in the early phase of these neurodegenerative diseases. In late-stage age-related macular degeneration, we identify a microglia-to-astrocyte signaling axis mediated by interleukin-1β which drives angiogenesis characteristic of disease pathogenesis. We validated this mechanism using in vitro and in vivo assays in mouse, identifying a possible new therapeutic target for AMD and possibly other neurodegenerative conditions. Thus, due to shared glial states, the retina provides a potential system for investigating therapeutic approaches in neurodegenerative diseases.

Pre-harvest yield prediction of ratoon rice is critical for guiding crop interventions in precision agriculture. However, the unique agronomic practice (i.e., varied stubble height treatment) in rice ratooning could lead to inconsistent rice phenology, which had a significant impact on yield prediction of ratoon rice. Multi-temporal unmanned aerial vehicle (UAV)-based remote sensing can likely monitor ratoon rice productivity and reflect maximum yield potential across growing seasons for improving the yield prediction compared with previous methods. Thus, in this study, we explored the performance of combination of agronomic practice information (API) and single-phase, multi-spectral features [vegetation indices (VIs) and texture (Tex) features] in predicting ratoon rice yield, and developed a new UAV-based method to retrieve yield formation process by using multi-temporal features which were effective in improving yield forecasting accuracy of ratoon rice. The results showed that the integrated use of VIs, Tex and API (VIs & Tex + API) improved the accuracy of yield prediction than single-phase UAV imagery-based feature, with the panicle initiation stage being the best period for yield prediction (R2 as 0.732, RMSE as 0.406, RRMSE as 0.101). More importantly, compared with previous multi-temporal UAV-based methods, our proposed multi- temporal method (multi-temporal model VIs & Tex: R2 as 0.795, RMSE as 0.298, RRMSE as 0.072) can increase R2 by 0.020–0.111 and decrease RMSE by 0.020–0.080 in crop yield forecasting. This study provides an effective method for accurate pre-harvest yield prediction of ratoon rice in precision agriculture, which is of great significance to take timely means for ensuring ratoon rice production and food security.

China’s rapid development of aquaculture has focused attention on its environmental impacts and the need for a comprehensive method that will enable environmental managers to estimate and evaluate aquaculture pollutant emissions. Few reports have estimated the environmental quality load of aquaculture conducted under different culture modes at the township level, and even fewer have studied the methods used to evaluate pollutant emissions. Here, we present a novel joint method based on an inventory analysis and an improved entropy weight method supplemented by remote sensing and an analysis of emission intensity. The method was applied to estimate and evaluate the environmental mass load of aquaculture in Zhuhai, Guangdong Province, China. The results showed that the main pollutant in areas dominated by freshwater aquaculture was chemical oxygen demand, whereas in areas dominated by mariculture the main pollutant was total nitrogen. Negative emissions occurred in Jinwan and Gaolan Port Districts, whereas the highest environmental quality load under different aquaculture modes was found in Baijiao Town, located in Doumen District. This study identified the excessive aquaculture density in Zhuhai. More generally, it provides a reference method for the environmental management of aquaculture in coastal cities.

Objective: To systematically evaluate the efficacy of moxibustion in diarrhea-predominant irritable bowel syndrome (IBS-D) model rats.Methods: A comprehensive search was conducted in the China National Knowledge Infrastructure, WanFang Data, VIP, PubMed, Embase, and Web of Science databases from their inception to June 30, 2023. Relevant animal experiments investigating moxibustion for treating IBS-D in model rats were included. Two independent researchers screened the literature, extracted data, and evaluated the risk of bias in the selected studies. The meta-analysis was performed using RevMan 5.3 software.Results: In total, 21 animal studies comprising 680 model rats were included. The meta-analysis results demonstrated that moxibustion enhanced the threshold capacity of the abdominal withdrawal reflex (AWR) [standardized mean difference (SMD) = 1.84; 95% confidence interval (CI): 1.09, 2.60; p < 0.00001], ameliorated the rate of loose stool (SMD = −4.03; 95% CI: –5.76, −2.30; p < 0.00001), and decreased the colon 5-hydroxytryptamine (SMD = −3.67; 95% CI: –5.33, −2.01; p < 0.00001), serum interleukin-1β (SMD = −3.24, 95% CI: –4.06, −2.41; p < 0.00001), serum tumor necrosis factor-α (SMD = −2.35, 95% CI: –4.12, −0.58; p < 0.00001), and serum substance P (SMD = −5.14, 95% CI: –8.45, −1.83; p = 0.002) concentrations. Moxibustion did not affect the blood calcitonin gene-related peptide level compared to the blank model group (p = 0.15).Conclusion: Moxibustion modulated the brain-gut interaction, reduced visceral hypersensitivity, inhibited intestinal inflammation, and regulated the immune balance, improving the rate of loose stool and increasing the AWR threshold capacity in IBS-D model rats, achieving good analgesic and antidiarrheal effects. However, these conclusions require further validation due to limitations in the quantity and quality of the included studies.

Abstract Background Chestnut blight, one of the most serious branch diseases in Castanea caused by Cryphonectria parasitica, which has ravaged across American chestnut and most of European chestnut since the early twentieth century. Interestingly, the Chinese chestnut is strongly resistant to chestnut blight, shedding light on restoring the ecological status of Castanea plants severely affected by chestnut blight. To better explore the early defense of Chinese chestnut elicited in response to C. parasitica, the early stage of infection process of C. parasitica was observed and RNA sequencing-based transcriptomic profiling of responses of the chestnut blight-resistant wild resource ‘HBY-1’ at 0, 3 and 9 h after C. parasitica inoculation was performed. Results First, we found that 9 h was a critical period for Chinese chestnut infected by C. parasitica, which was the basis of further study on transcriptional activation of Chinese chestnut in response to chestnut blight in the early stage. In the transcriptome analysis, a total of 283 differentially expressed genes were identified between T9 h and Mock9 h, and these DEGs were mainly divided into two clusters, one of which was metabolism-related pathways including biosynthesis of secondary metabolites, phenylpropanoid biosynthesis, amino sugar and nucleotide sugar metabolism, and photosynthesis; the other was related to plant-pathogen interaction and MAPK signal transduction. Meanwhile, the two clusters of pathways could be connected through junction among phosphatidylinositol signaling system, phytohormone signaling pathway and α-Linolenic acid metabolism pathway. It is worth noting that genes associated with JA biosynthesis and metabolic pathway were significantly up-regulated, revealing that the entire JA metabolic pathway was activated in Chinese chestnut at the early stage of chestnut blight infection. Conclusion We identified the important infection nodes of C. parasitica and observed the morphological changes of Chinese chestnut wounds at the early stage of infection. In response to chestnut blight, the plant hormone and MAPK signal transduction pathways, plant-pathogen interaction pathways and metabolism-related pathways were activated at the early stage. JA biosynthesis and metabolic pathway may be particularly involved in the Chinese chestnut resistance to chestnut blight. These results contributes to verifying the key genes involved in the resistance of Chinese chestnut to C. parasitica.

Physical fatigue is one of the important problems that plague public health. Our objective is to investigate the anti-fatigue effect of a botanical beverage with ginseng extract and pueraria extract, including ginseng, wolfberry, Polygonatum, Pueraria, and niacin. Sixty ICR male mice were randomly divided into 6 groups, namely blank control group, experimental control group (swimming group), positive control group (caffeine group), low-dose group (3.33 g/kg BW), medium-dose group (6.67 g/kg BW), and high-dose group (13.33 g/kg BW) of botanical beverage. Then we performed behavioral examination, including grip and turn-club test, and investigated fatigue-related biochemical indexes and behavioral tests after consecutive gavage administration for 30 days.. Results showed that the fatigue rotarod time and forelimb pulling force of the mice are significantly increased in the middle and high dose groups of botanical beverage and the caffeine group, compared with the blank control group. The contents of blood sugar level, liver glycogen, and muscle glycogen of the mice in the middle dose group increased by 53.58%, 18.75%, and 45.00%, respectively, compared with the swimming group. While the contents of blood lactate and urea nitrogen and the activities of lactate dehydrogenase and creatine kinase in the middle-dose group decreased by 5.56%, 18.78% and 27.18%, 9.45%, respectively. It’s concluded that botanical beverage with ginseng extract and pueraria extract had the effect on relieving physical fatigue.

In this paper, the protective effect of a botanical beverage with pueraria extract and Hovenia dulcis Thunb. extract on acute alcoholic liver injury in mice was investigated. The acute alcoholic liver injury model was established by a one-time gavage of 50% alcohol, and then the relevant biochemical indicators and liver tissue morphology were detected to evaluate the hepatoprotective effect of the botanical beverage. Compared with the model control group, the middle-dose of plant beverage with pueraria extract and Hovenia dulcis Thunb. extract can significantly reduce the levels of ALT, AST, TC, TG and LDL in the serum of mice with acute alcoholic liver injury to (19.17±6.22) U/L, (14.81±4.61) U/L, (4.77±0.33) mmol/L, (1.92±0.24) mmol//L, (0.22±0.09) mmol/L, increase the activities of SOD, GSH-Px and GSH in liver, and decrease MDA content to (0.80±0.09) nmol/mg pro. This plant drink can also significantly improve the pathological changes of liver tissue to a certain extent, and reduce steatosis and inflammatory cell infiltration. The experimental results showed that the plant beverage with pueraria extract and Hovenia dulcis Thunb. extract had a certain protective effect on the liver tissue of mice after acute alcohol exposure.

ObjectiveAggressive angiomyxoma (AAM) is a rare kind of soft tissue tumor. The purpose of this study is to summarize the clinical manifestations and treatment strategy of AAM in women.MethodWe searched the case reports on AAM in EMBASE, Web of Science and PubMed, China biomedical database, Wanfang database, VIP database, and China National Knowledge Internet from the start of database construction to November 2022 without any language restrictions in place. Then, the obtained case data were extracted, summarized, and analyzed.ResultA total of 74 articles were retrieved involving 87 cases. The age ranges of onset were 2–67 years. The median age at onset was 34 years. The size of the tumor varied greatly among individuals, and about 65.5% of them were asymptomatic. MRI, ultrasound, and needle biopsy were used for diagnosis. Surgery was the primary mode of treatment, but it was prone to relapse. Gonadotropin-releasing hormone agonist (GnRH-a) might be used to reduce the tumor size before the operation and prevent recurrence after the operation. For patients who are unwilling to receive surgical treatment, GnRH-a alone could be attempted.ConclusionDoctors should consider the possibility of AAM in women with genital tumors. For surgery, it must be ensured that the negative surgical margin is recommended and achieved for preventing recurrence, but we should not ignore the impact of the excessive pursuit for a negative margin on the patient’s reproductive function protection and postoperative recovery. Long-term follow-up is necessary regardless of whether patients receive medical treatment or surgical treatment.

(1) Lipases are catalysts widely applied in industrial fields. To sustain the harsh treatments in industries, optimizing lipase activities and thermal stability is necessary to reduce production loss. (2) The thermostability of Thermomyces lanuginosus lipase (TLL) was evaluated via B-factor analysis and consensus-sequence substitutions. Five single-point variants (K24S, D27N, D27R, P29S, and A30P) with improved thermostability were constructed via site-directed mutagenesis. (3) The optimal reaction temperatures of all the five variants displayed 5 °C improvement compared with TLL. Four variants, except D27N, showed enhanced residual activities at 80 °C. The melting temperatures of three variants (D27R, P29S, and A30P) were significantly increased. The molecular dynamics simulations indicated that the 25-loop (residues 24–30) in the N-terminus of the five variants generated more hydrogen bonds with surrounding amino acids; hydrogen bond pair D254-I255 preserved in the C-terminus of the variants also contributes to the improved thermostability. Furthermore, the newly formed salt-bridge interaction (R27…E56) in D27R was identified as a crucial determinant for thermostability. (4) Our study discovered that substituting residues from the 25-loop will enhance the stability of the N-terminus and C-terminus simultaneously, restrict the most flexible regions of TLL, and result in improved thermostability.

Neural circuits are characterized as interconnecting neuron networks connected by synapses. Some kinds of gene expression and/or functional changes of neurons and synaptic connections may result in aberrant neural circuits, which has been recognized as one crucial pathological mechanism for the onset of many neurological diseases. Gradual advances in single-cell sequencing approaches with strong technological advantages, as exemplified by high throughput and increased resolution for live cells, have enabled it to assist us in understanding neuronal diversity across diverse brain regions and further transformed our knowledge of cellular building blocks of neural circuits through revealing numerous molecular signatures. Currently published transcriptomic studies have elucidated various neuronal subpopulations as well as their distribution across prefrontal cortex, hippocampus, hypothalamus, and dorsal root ganglion, etc. Better characterization of brain region-specific circuits may shed light on new pathological mechanisms involved and assist in selecting potential targets for the prevention and treatment of specific neurological disorders based on their established roles. Given diverse neuronal populations across different brain regions, we aim to give a brief sketch of current progress in understanding neuronal diversity and neural circuit complexity according to their locations. With the special focus on the application of single-cell sequencing, we thereby summarize relevant region-specific findings. Considering the importance of spatial context and connectivity in neural circuits, we also discuss a few published results obtained by spatial transcriptomics. Taken together, these single-cell sequencing data may lay a mechanistic basis for functional identification of brain circuit components, which links their molecular signatures to anatomical regions, connectivity, morphology, and physiology. Furthermore, the comprehensive characterization of neuron subtypes, their distributions, and connectivity patterns via single-cell sequencing is critical for understanding neural circuit properties and how they generate region-dependent interactions in different context.

Irritable bowel syndrome (IBS) is a common digestive disorder observed in clinics. Current studies suggest that the pathogenesis of the disease is closely related to abnormal brain–gut interactions, hypokinesia, visceral sensory hypersensitivity in the gastrointestinal tract, and alterations in the intestinal microenvironment. However, it is difficult for a single factor to explain the heterogeneity of symptoms. The Rome IV criteria emphasized the holistic biologic-psycho-social model of IBS, suggesting that symptoms of the disease are closely related to neurogastroenterology and various abnormalities in brain–gut interaction. This study comprehensively reviewed the relationship between the brain–gut axis and IBS, the structure of the brain–gut axis, and the relationship between the brain–gut axis and intestinal microenvironment, and discussed the relationship between the abnormal regulation of the nervous system, endocrine system, and immune system and the incidence of IBS on the basis of brain–gut axis. In terms of treatment, acupuncture therapy can regulate the neuroendocrine-immune system of the body and improve the intestinal microenvironment, and it has the advantages of safety, economy, and effectiveness. We study the pathogenesis of IBS from local to global and micro to macro, and review the use of acupuncture to treat the disease as a whole so as to provide new ideas for the treatment of the disease.

The abandoned mines along the Yangtze River in Jiangsu Province is the key area along the Yangtze River Economic Belt, which need to be restored and governed. Among the ecological restoration projects, it has been one of the difficulties that how to quickly and intuitively track the progress of the distribution and restoration of a great many abandoned mines in the region. In this paper, the author takes the 10 km area on both sides of the Belt as study object. And selects high-resolution remote sensing images in each year from 2017 to 2020 as the data source. By human-computer interactive interpretation on the ArcGIS platform and field verification, the author obtains the information such as the distribution of open-pit abandoned mines, geological environment problems and ecological restoration in the study area. This paper mainly focus on the geological environment problems existing in abandoned mines and summarizes the ecological restoration measures. It is shown that it ’s a wide area need to be restored in Zhenjiang and Nanjing. The area of open-pit abandoned mines mainly focus on the quarries and transit sites of the mines, with construction limestone and construction sand mines as the main types of mines involved. The environmental problems mainly include topography and geomorphological destruction, geological disasters, ecological environment damage and soil and water pollution, etc. And the part rehabilitated and being rehabilitated of the study area show an increasing trend from 2018 to 2020. As of October 2020, the restoration area of abandoned mines has been effective, with 67.08% of the area restored. The main ecological restoration measures include natural restoration, soil backfilling, land leveling, slope cutting and integrated land management. It is recommended that the original landform of the mining area should be protected to the maximum extent and the land resources fully utilized when carrying out restoration and treatment. The results and suggestions provide technical reference for the ecological restoration and treatment of open-pit abandoned mines in the same type of area.

Devices for continuous in-vivo testing (CIVT) can detect target substances in real time, thus providing a valuable window into a patient's condition, their response to therapeutics, metabolic activities, and neurotransmitter transmission in the brain. Therefore, CIVT devices have received increased attention because they are expected to greatly assist disease diagnosis and treatment and research on human pathogenesis. However, CIVT has been achieved for only a few markers, and it remains challenging to detect many key markers. Therefore, it is important to summarize the key technologies and methodologies of CIVT, and to examine the direction of future development of CIVT. We review recent progress in the development of CIVT devices, with consideration of the structure of these devices, principles governing continuous detection, and nanomaterials used for electrode modification. This detailed and comprehensive review of CIVT devices serves three purposes: (1) to summarize the advantages and disadvantages of existing devices, (2) to provide a reference for development of CIVT equipment to detect additional important markers, and (3) to discuss future prospects with emphasis on problems that must be overcome for further development of CIVT equipment. This review aims to promote progress in research on CIVT devices and contribute to future innovation in personalized medical treatments.

Background: Tamoxifen (TMX) is one of the most widely used drugs to treat breast cancer (BC). However, acquired drug resistance is still a major obstacle to its application, rendering it crucial to explore the mechanisms of TMX resistance in BC. This aims of this study were to identify the mechanisms of TMX resistance and construct ceRNA regulatory networks in breast cancer.Methods: GEO2R was used to screen for differentially expressed mRNAs (DEmRNAs) leading to drug resistance in BC cells. MiRTarbase and miRNet were used to predict miRNAs and lncRNAs upstream, and the competing endogenous RNA (ceRNA) regulatory network of BC cell resistance was constructed by starBase. We used the Kaplan–Meier plotter and Gene Expression Profiling Interactive Analysis (GEPIA) to analyze the expression and prognostic differences of genes in the ceRNA network with core axis, and qRT-PCR was used to further verify the above conclusions.Results: We found that 21 DEmRNAs were upregulated and 43 DEmRNA downregulated in drug-resistant BC cells. DEmRNAs were noticeably enriched in pathways relevant to cancer. We then constructed a protein-protein interaction (PPI) network based on the STRING database and defined 10 top-ranked hub genes among the upregulated and downregulated DEmRNAs. The 20 DEmRNAs were predicted to obtain 113 upstream miRNAs and 501 lncRNAs. Among them, 7 mRNAs, 22 lncRNAs, and 11 miRNAs were used to structure the ceRNA regulatory network of drug resistance in BC cells. 4 mRNAs, 4 lncRNAs, and 3 miRNAs were detected by GEPIA and the Kaplan–Meier plotter to be significantly associated with BC expression and prognosis. The differential expression of the genes in BC cells was confirmed by qRT-PCR.Conclusion: The ceRNA regulatory network of TMX-resistant BC was successfully constructed and confirmed. This will provide an important resource for finding therapeutic targets for TMX resistance, where the discovery of candidate conventional mechanisms can aid clinical decision-making. In addition, this resource will help discover the mechanisms behind this type of resistance.

Objective: This study aimed to analyze the accuracy of determination results of five kinds of carbamate pesticide residues in apple. Methods: Five kinds of carbamate pesticide residues in apples were detected according to NY/T 761—2008, and the uncertainty of the determination process was evaluated and analyzed. Results: The contents of carbosulfur, methomyl, 3-hydroxycarbamate, aldicarb and isoprocarb in apples are expressed in the form of expanded uncertainty as （0.105 7±0.007 8）, （0.107 7±0.006 2）, （0.109 2±0.005 8）, （0.103 9±0.008 4）, （0.106 1±0.007 6） mg/kg （k=2）. Conclusion: The main sources of uncertainty in the experiment are standard material, recovery rate, sample pretreatment, and detection equipment, and the overall expression is Urel（Standard）＞Urel（Recovery Rate）＞Urel （Pretreatment）＞Urel （Instrument）.

Ticks were identified as arthropods that are pathogenic vectors. Dermacentor nuttalli is one of the dominant tick species in Inner Mongolia, and it carries and transmits a wide range of pathogenic microorganisms. However, at present, only the detection of D. nuttalli adult ticks and D. nuttalli different developmental stages carrying one specific pathogen, or the next-generation sequencing of D. nuttalli adult ticks were available. In this study, we investigated the microbial community structures of D. nuttalli in different growth stages under laboratory artificial feeding conditions. Total DNA was extracted from seven growth stages (female adult ticks, eggs, larval ticks, engorged larval ticks, nymphal ticks, engorged nymphal ticks, and second-generation adult ticks) obtained from laboratory artificial feeding of engorged D. nuttalli female ticks in Inner Mongolia. Then, the 16S rDNA V3–V4 hypervariable region was amplified to construct an Illumina PE250 library. Finally, 16S rRNA sequencing was performed on Illumina Novaseq 6000 platform. The sequencing data were analyzed using molecular biology software and platforms. The Illumina PE250 sequencing results showed that the egg stage had the highest diversity and number of species (28.74%, 98/341), while the engorged nymph stage had the lowest diversity and number of species (9.72%, 21/216). A total of 387 genera of 22 phyla were annotated in D. nuttalli, with 9 phyla and 57 genera found throughout all 7 growth stages. The dominant phylum was Proteobacteria; the dominant genera were Arsenophonus and Rickettsia; and the genera with the highest relative abundance in the 7 growth stages were Pseudomonas, Paenalcaligenes, Arsenophonus, Arsenophonus, Pseudomonas, Arsenophonus, and Rickettsia, respectively. Among the 23 exact species annotated, Brucella melitensis exhibits pathogeny that poses a serious threat to humans and animals. In this study, the microbial community composition at different growth stages of D. nuttalli was comprehensively analyzed for the first time.

Melophagus ovinus (sheep ked) is one of the common ectoparasites in sheep. In addition to causing direct damage to the host through biting and sucking blood, sheep ked is a potential vector of helminths, protozoa, bacteria, and viruses. Sheep M. ovinus samples from three regions in Tibet were selected for DNA extraction. The 16S rDNA V3-V4 hypervariable region was amplified, after genomic DNA fragmentation, Illumina Hiseq libraries were constructed. The 16S rRNA sequencing and viral metagenomics sequencing were separately conducted on the Illumina Novaseq 6000 platform and molecular biology software and platforms were employed to analyze the sequencing data. Illumina PE250 sequencing results demonstrated that the dominant bacteria phylum in M. ovinus from Tibet, China was Proteobacteria, where 29 bacteria genera were annotated. The dominant bacterial genera were Bartonella, Wolbachia, and Arsenophonus; Bartonella chomelii, Wolbachia spp., and Arsenophonus spp. were the dominant bacterial species in M. ovinus from Tibet, China. We also detected Kluyvera intermedia, Corynebacterium maris DSM 45190, Planomicrobium okeanokoites, and Rhodococcus erythropolis, of which the relative abundance of Kluyvera intermedia was high. Illumina Hiseq sequencing results demonstrated that 4 virus orders were detected in M. ovinus from Tibet, China, and 3 samples were annotated into 29 families, 30 families, and 28 families of viruses, respectively. Virus families related to vertebrates and insects mainly included Mimiviridae, Marseilleviridae, Poxviridae, Ascoviridae, Iridoviridae, Baculoviridae, Hytrosaviridae, Nudiviridae, Polydnaviridae, Adomaviridae, Asfarviridae, Hepeviridae, Herpesviridae, and Retroviridae; at the species level, the relative abundance of Tupanvirus_soda_lake, Klosneuvirus_KNV1, and Indivirus_ILV1 was higher. African swine fever virus and many poxviruses from the family Poxviridae were detected, albeit their relative abundance was low. The dominant bacterial phylum of M. ovinus from Tibet, China was Proteobacteria, and the dominant bacterial genera were Bartonella, Wolbachia, and Arsenophonus, where 23 out of 29 annotated bacteria genera were first reported in M. ovinus. Kluyvera intermedia, Corynebacterium maris DSM 45190, Planomicrobium okeanokoites, and Rhodococcus erythropolis were detected for the first time. All DNA viruses detected in this study have been reported in M. ovinus for the first time.

Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders in gynecology. Traditional Chinese medicine (TCM) is widely used for the treatment of PCOS in China. The Bu Shen Hua Zhuo formula (BSHZF), a TCM decoction, has shown great therapeutic efficacy in clinical practice. However, the mechanism underlying the BSHZF function in PCOS remains unclear. This study aimed to identify the potential mechanisms of action of BSHZF in the treatment of PCOS. PCOS-model rats treated with letrozole were administered different doses of BSHZF, metformin, and 1% carboxymethylcellulose. Serum sex hormones, fasting blood glucose, and fasting insulin levels were measured, and the morphology of the ovaries was observed in each group, including the normal group. The structure and abundance of the gut microbiota in rats were measured using 16S ribosomal RNA gene sequencing. Toll-like receptor 4 (TLR4) and phospho-NF-κB p65 levels in the ovarian tissue of the rats were detected using Western blotting. Furthermore, the levels of lipopolysaccharide (LPS) and inflammatory cytokines TNF-α, IL-6, and IL-8 in the serum of rats were detected by ELISA. The results showed that BSHZF administration was associated with a decrease in body weight, fasting blood glucose, fasting insulin, and testosterone and changes in ovarian morphology in PCOS-model rats. Moreover, BSHZF was associated with an increase in the α-diversity of gut microbiota, decrease in the relative abundance of Firmicutes, and increase in Lactobacillus and short chain fatty acid–producing bacteria (Allobaculum, Bacteroides, Ruminococcaceae_UCG-014). Furthermore, BSHZF may promote carbohydrate and protein metabolism. In addition, BSHZF was associated with a decrease in the serum level of LPS and TLR4 expression, thereby inhibiting the activation of the NF-κB signaling–mediated inflammatory response in ovarian tissue. Therefore, the beneficial effects of BSHZF on PCOS pathogenesis are associated with its ability to normalize gut microbiota function and inhibit PCOS-related inflammation.

Autism spectrum disorder (ASD) is characterized by social communicative abnormalities. Deficits and delays in gestural communication are among the early deficits of ASD and also a major social modality in early caregiver-toddler interaction. Caregiver gestures have an important role in the cognitive and social development of children with ASD. Thus, it is urgent to further explore the role of caregiver gestures in early caregiver-toddler interaction. In this cross-sectional study, we observed the caregivers' gestures and responses of toddlers aged between 18 and 24 months during play (ASD = 44, TD = 29) and dining activities (ASD = 34, TD = 27). By observing the different frequencies and patterns of gestures by the caregiver-child interaction and the different proportions of children's responses to the caregiver's gestures, we found that, compared to caregivers of typically developing toddlers, caregivers of toddlers with ASD had fewer synchronized gestures and more unsynchronized gestures in the play activity and more supplementary gestures in dining activity. Toddlers with ASD produced more social responses to caregivers' synchronized gestures, whereas the use of synchronized gestures by the caregivers in caregiver-toddler interaction had a positive influence on social responses to toddlers with ASD. The findings suggest that effective use of gestures by caregivers during caregiver-toddler activities can improve children's social responses.

Boron (B) is essential to plant development. However, excessive B is toxic to plants. This research was performed to evaluate the effects of B toxicity on cell wall architecture of Chinese chestnut (Castanea mollissima Blume) pollen tubes with emphasis on the relationship among pectins, cellulose, and callose. Results showed that 0.8 mM H3BO3 inhibited pollen germination and led to abnormal morphology of the pollen tubes. B toxicity also affected the distribution of cell wall components of the pollen tube. In control pollen tube, esterified and acid pectins were distributed unevenly, with the former mainly at the tip and the latter on the distal region. Cellulose was distributed uniformly on the surface with less at the tip; callose reduced gradually from base to sub-tip of the pollen tubes and no callose at the tip of the tube was detected. B toxicity led to the deposition of esterified and acid pectins, cellulose, and callose at the tip of the pollen tube. Results from scanning electron microscopy and transmission electron microscopy showed that B toxicity also altered pollen tube wall ultrastructure. The results from enzymatic treatment illustrated that there existed a close relationship among pectins, cellulose, and callose. B toxicity also altered the relationship. In a word, B toxicity altered deposition and relationship of pectins, cellulose, and callose of pollen tube wall.

Computational simulations can accelerate the design and modelling of origami robots and mechanisms. This paper presents a computational method using algorithms developed in Python to generate different tessellated origami crease patterns simultaneously. This paper aims to automate this process by introducing a system that automatically generates origami crease patterns in Scalable Vector Graphics format. By introducing different parameters, variations of the same underlying tessellated crease pattern can be obtained. The user interface consists of an input file where the user can input the desired parameters, which are then processed by an algorithm written in Python to generate the respective origami 2D crease patterns. These origami crease patterns can serve as inputs to current origami design software and algorithms to generate origami design models for faster and easier visual comparison. This paper utilizes a basic biomimetic inspiration origami pattern to demonstrate the functionality by varying underlying crease pattern parameters that give rise to symmetric and asymmetric spring origami 3D structures. Furthermore, this paper conducts a qualitative analysis of the origami design outputs of an origami simulator from the input crease patterns and the respective manual folding of the origami structure.

However, whether Wii Fit exercise improves the balance and lower limb muscle strength of older adults remains uncertain.ObjectiveTo evaluate the current literature by using meta-analyses to assess whether Wii Fit exercise is associated with improved balance and lower limb muscle strength in older adults.MethodsWe searched PubMed, EMBASE, Web of Science, China National Knowledge Infrastructure, and Wanfang Data from inception to February 16, 2022, for relevant studies and conducted a manual search of the literature. Data extraction of the included studies were independently performed by two authors. The methodologic quality of each included study was assessed using the Physical Therapy Evidence Database (PEDro) scale. Meta-analyses were conducted using Review Manager 5.3.ResultsOf 991 articles initially identified, 10 articles (379 participants; aged ≥65 years, range 65–92 years; 188 participants in Wii Fit exercise groups, and 191 participants in control groups), including 8 randomized controlled trials, were assessed in this meta-analysis. The methodologic quality of the included studies evaluated with the PEDro average score was 6.4 and ranged from 5 to 8 points, indicating moderate quality. Compared with no exercise, Wii Fit exercise was associated with significant improved scores on the Berg Balance Scale, which evaluates functional balance (mean difference, 1.38; 95% CI, 0.61–2.16; I2 = 31%; P = 0.0005) and on the Timed Up and Go test, which evaluates static and dynamic balance (standardized mean difference, −0.51; 95% CI,−0.88 to −0.14; I2 = 52%; P = 0.007). However, for the three studies assessed, Wii Fit exercise did not significantly improve scores on the 30-s chair stand test, which evaluates the functional strength and endurance of the lower limb muscles (mean difference, 0.82; 95% CI, −0.14 to 1.77; z = 1.68; P = 0.09).ConclusionsOur findings indicated that Wii Fit exercise is an effective approach to improve functional, static, and dynamic balance among older adults. Additional meta-analyzes with higher numbers of randomized controlled trials are suggested to confirm the benefits of Wii Fit exercise on balance in older adults, to establish whether functional strength and endurance of lower limb muscles are improved, and to explore the relationship between improved balance and fall prevention in this population.

This study aimed to investigate the gap between adaptive functioning and cognitive functioning, especially verbal and nonverbal intelligence quotient (IQ) in Chinese children with ASD. We systematically explored cognitive functioning, ASD severity, early signs of developmental abnormalities, and socioeconomic factors as mediating factors of adaptive functioning. We enrolled 151 children (age: 2.5?6 years) with ASD and categorized them into one group with IQ [greater than or equal to] 70 and another with IQ < 70. The two groups were calibrated for age, age at diagnosis, and IQ, and the relationship of adaptive skills with vocabulary acquisition index (VAI) and nonverbal index (NVI) were separately analyzed. Results show that the gap between IQ and adaptive functioning was significant in children with ASD having IQ [greater than or equal to] 70, with both VAI and NVI showing statistically significant differences (all P < 0.001). VAI correlated positively with scores for overall adaptive skills and specific domains, whereas NVI had no significant correlations with adaptive skill scores. Age of first walking unaided had an independent positive correlation (all P < 0.05) with scores of adaptive skills and specific domains. IQ-adaptive functioning gap is significant in children with ASD having IQ [greater than or equal to] 70, suggesting that defining "high-functioning autism" merely on the basis of IQ is not appropriate. Verbal IQ and early signs of motor development are specific and possible predictors of adaptive functioning in children with ASD, respectively.

Sun, Wang et al. report an origami paper-based immunosensor for the electrochemical detection of the VEGF-C biomarker in blood serum. The immunosensor is made by modifying the surface of working electrodes with new methylene blue, amino-functional single-walled carbon nanotubes, and gold nanoparticles and demonstrates excellent performance with a limit of detection in the range of picograms per milliliter.

Doxorubicin (DOX) is the most clinically important antibiotic in cancer treatment, but its severe cardiotoxicity and other side effects limit its clinical use. Therefore, monitoring DOX concentrations during therapy is essential to improve efficacy and reduce adverse effects. Here, we fabricated a sensitive electrochemical aptasensor for DOX detection. The sensor used gold wire as the working electrode and was modified with reduced graphene oxide (rGO)/gold nanoparticles (AuNPs) to improve the sensitivity. An aptamer was used as the recognition element for the DOX. The 5′ end of the aptamer was modified with a thiol group, and thus immobilized to the AuNPs, and the 3′ end was modified with methylene blue, which acts as the electron mediator. The combination between the aptamer and DOX would produce a binding-induced conformation, which changes the electron transfer rate, yielding a current change that correlates with the concentration of DOX. The aptasensor exhibited good linearity in the DOX concentration range of 0.3 μM to 6 μM, with a detection limit of 0.1 μM. In addition, the aptasensor was used for DOX detection in real samples and results, and showed good recovery. The proposed electrochemical aptasensor will provide a sensitive, fast, simple, and reliable new platform for detecting DOX.

Spherical roller bearings (SRBs) are widely used under self-aligning operating conditions, such as rotor bending or an angular misalignment between inner and outer rings due to their self-aligning function. However, the characterization of SRBs’ self-aligning function is often ignored in the present models. The reason for this is that the self-aligning condition is essentially a fault condition, and many scholars have assumed SRBs are in an ideal operating condition. Although there is nothing wrong with this analysis theoretically, it is incapable of characterizing SRBs’ service behavior comprehensively. In this work, the Lagrange equation was introduced to model the relationship among the rollers and the inner and outer rings. The contact region in particular was characterized in detail in order to solve the problems of undetermined contact status (UCS) and the varying law of the self-aligning contact angle (SAC angle). For the experiment, a novel SRBs pedestal with a self-aligning operating condition was designed, and the relevant self-aligning function testing was carried out. A good agreement was shown between the theoretical and experimental results. The results pointed out that, if taking no account of the self-aligning function, SRBs can be regarded as angular contact ball bearings or cylinder roller bearings. The amplitude of the inner-ring motion orbit is determined by the external load, but the shape is influenced by the direction and magnitude of the SAC angle. In the example of this paper, the values of the main frequency equal 136.8 Hz. Some additional frequencies are clearly aroused under the self-aligning operating condition, whose value is approximately equal to 8.3 Hz or its integer multiples. The dynamic performance of SRBs will be substantially improved by a light axial load plus an anticlockwise self-aligning contact angle rather than a large axial preload.

ObjectiveMinimally invasive puncture and drainage (MIPD) of hematomas was the preferred option for appropriate patients with hypertensive intracerebral hemorrhage (HICH). The goal of our research was to introduce the MIPD surgery using mixed reality holographic navigation technology (MRHNT).MethodWe provided the complete workflow for hematoma puncture using MRHNT included three-dimensional model reconstruction by preoperative CT examination, puncture trajectory design, immersive presentation of model, and real environment and hematoma puncture using dual-plane navigation by wearing special equipment. We collected clinical data on eight patients with HICH who underwent MIPD using MRHNT from March 2021 to August 2021, including the hematoma evacuation rate, operation time, deviation in drainage tube target, postoperative complications, and 2-week postoperative GCS.ResultThe workflow for hematoma puncture using MRHNT were performed in all eight cases, in which the average hematoma evacuation rate was 47.36±9.16%, the average operation time was 82.14±15.74 min, and the average deviation of the drainage tube target was 5.76±0.80 mm. There was no delayed bleeding, acute ischemic stroke, intracranial infection, or epilepsy 2 weeks after surgery. The 2-week postoperative GCS was improved compared with the preoperative GCS.ConclusionThe research concluded it was feasible to perform the MIPD by MRHNT on patients with HICH. The risk of general anesthesia and highly professional holographic information processing restricted the promotion of the technology, it was necessary for technical innovation and the accumulation of more case experience and verification of its superiority.

Chinese chestnut (Castanea mollissima), a species with recalcitrant seeds, is an important source of nuts and forest ecosystem services. The germination rate of recalcitrant seeds is low in natural habitats and decreases under conditions of desiccation and low temperature. The germination rate of cultivated Chinese chestnut seeds is significantly higher than that of wild seeds. To explore the reasons for the higher germination rate of cultivated seeds in Chinese chestnut, 113,524 structural variants (SVs) between the wild and cultivated Chinese chestnut genomes were detected through genome comparison. Genotyping these SVs in 60 Chinese chestnut accessions identified allele frequency changes during Chinese chestnut domestication, and some SVs are overlapping genes for controlling seed germination. Transcriptome analysis revealed downregulation of the abscisic acid synthesis genes and upregulation of the beta-amylase synthesis genes in strongly selected genes of cultivated seeds. On the other hand, hormone and enzyme activity assays indicated a decrease in endogenous ABA level and an increase in beta-amylase activity in cultivated seeds. These results shed light on the higher germination rate of cultivated seeds. Moreover, phosphatidic acid synthesis genes are highly expressed in seed germination stages of wild Chinese chestnut and may play a role in recalcitrant seed germination. These findings provide new insight into the regulation of wild seed germination and promote natural regeneration and succession in forest ecosystems.

ObjectiveEctopic adrenocorticotropic hormone (ACTH) syndrome (EAS) is a condition of hypercortisolism caused by non-pituitary tumors secreting ACTH. Appendiceal neuroendocrine tumor as a rare cause of ectopic ACTH syndrome was reported scarcely. We aimed to report a patient diagnosed with EAS caused by an appendiceal neuroendocrine tumor and summarized characteristics of these similar cases reported before.Case Report and Literature ReviewWe reported a case with Cushing’s syndrome who was misdiagnosed as pituitary ACTH adenoma at first and accepted sella exploration. Serum and urinary cortisol decreased, and symptoms were relieved in the following 4 months after surgery but recurred 6 months after surgery. The abnormal rhythm of plasma cortisol and ACTH presented periodic secretion and seemingly rose significantly after food intake. EAS was diagnosed according to inferior petrosal sinus sampling (IPSS). Appendiceal mass was identified by 68Ga-DOTA-Tyr3-octreotate (DOTATATE)-PET-CT and removed. The pathological result was consistent with appendiceal neuroendocrine tumor with ACTH (+). The literature review demonstrated 7 cases diagnosed with EAS caused by appendiceal neuroendocrine tumor with similarities and differences.ConclusionThe diagnosis of an ectopic ACTH-producing tumor caused by the appendiceal neuroendocrine tumor can be a challenging procedure. Periodic ACTH and cortisol secretion may lead to missed diagnosis and misdiagnosis. IPSS is crucial in the diagnosis of EAS and 68Ga-DOTATATE-PET-CT plays an important role in the identification of lesions.

Among the three existing targeted gene editing technologies, zinc-finger nucleases, transcription activator-like effector nucleases, and clustered regularly interspaced short palindromic repeats-CRISPR-associated 9 (CRISPR-Cas9), the latter is widely used owing to its simplicity, efficiency, and low cost. Here, we routinely infected A172 and U251 cells with lentiviral vectors, in which aquaporin-8 (AQP8) was knocked out using CRISPR/Cas9. Our results indicated that cryopreservation did not significantly alter the viral infection efficiency, but influenced AQP8 expression in the infected cells at both protein and mRNA levels compared with the non-cryopreserved samples. Further, AQP8 expression at protein and mRNA levels in recovered cryopreserved infected cells did not significantly differ from those in the blank and negative controls, indicating that the lentivirus was still infectious at low temperatures. However, it failed to release the AQP8-targeting guide RNA in the infected cells, or the guide RNA was released, but underwent changes that caused it to malfunction in the cells with CRISPR/Cas9-mediated AQP8 knock-out. Our findings possibly provide some insights into the reliability of lentiviruses as CRISPR/Cas9 vectors.

In this paper, we study the basket CDS pricing with two defaultable counterparties based on the reduced-form model. The default jump intensities of the reference firms and counterparties are all assumed to follow the mean-reverting constant elasticity of variance (CEV) processes. Taking the Vasicek process which is a special case of CEV process as an example, the approximate analytic solutions of the joint survival probability density, the probability densities of the first default and the first two defaults can be solved by using PDE method. In addition, we also extend the Vasciek process to the Vasciek process with cojumps and obtain the approximate closed-form solutions of the relevant default probability densities. Then with the expressions of the probability densities, we can get the formula of the basket CDS price with two defaultable counterparties. In the numerical analysis, we do sensitivity analysis and compare the basket CDS prices under our model with that with only one defaultable counterparty. The numerical results show that our model can be applied into practice.

In this paper, we study the valuation of vulnerable European options incorporating the reduced-form approach, which models the credit default of the counterparty. We provide an analytical pricing model in which the components of the state processes, including the dynamics of the underlying asset value and the intensity process corresponding to the default event, are cross-exciting and they could facilitate the description of complex structure of events dependence. To illustrate how our model works, we present an application when the state variables follow specific affine jump-diffusion processes. Semi-analytical pricing formulae are obtained through a system of matrix Riccati equations. The derived formula can be implemented numerically, and we give numerical analysis to investigate the impact of the dynamic correlation between jump risk of the underlying asset value and default risk of the counterparty.

Somatic embryogenesis (SE) is an effective approach of in vitro regeneration that depends on plant cell totipotency. However, largely unknown of molecular mechanisms of SE in woody plants such as Chinese chestnut (Castanea mollissima Blume), limits the development of the woody plant industry. Here, we report the MADS-box transcription factor CmAGL11 in Chinese chestnut. CmAGL11 transcripts specifically accumulated in the globular embryo. Overexpression of CmAGL11 in chestnut callus enhanced its SE capacity, and the development of somatic embryos occurred significantly faster than in the control. RNA-seq results showed that CmAGL11 affects the expression of several genes related to the gibberellin, auxin, and ethylene pathways. Moreover, the analysis of DNA methylation status indicated that the promoter methylation plays a role in regulation of CmAGL11 expression during SE. Our results demonstrated that CmAGL11 plays an important role in the SE process in Chinese chestnut, possibly by regulating gibberellin, auxin, and ethylene pathways. It will help establish an efficient platform to accelerate genetic improvement and germplasm innovation in Chinese chestnut.

Background: Endometriosis is a hormone- and organ-dependent disease with unclear pathogenesis. Danggui Shaoyao powder (DSP) has been used for the treatment of endometriosis for many years and has been proven to be effective, but its underlying molecular mechanisms remain unclear. In this study, we explored the molecular targets and pathways of DSP in the treatment of endometriosis mainly from the perspective of network pharmacology, and provided some novel ideas for managing this disease. Methods: In the present study, we focused on the underlying mechanisms between DPS and endometriosis, via oral bioavailability screening, drug similarity assessment, target identification, network analysis, drug relocation, and molecular docking using network pharmacology. Results: By bioinformatics, 54 active components of DSP were predicted from the 534 components provided by the TCMSP database. We then found 85 validated targets and 6059 predicted targets for herbal components, and 2241 targets for endometriosis. After mapping, there were 255 targets in common. After that, network analysis was performed to screen key networks and core targets. Ninety-nine indirect targets were collected from 14 direct ones.
