The root of Astragalus membranaceus (Fisch.) Bunge is one of the most frequently used herbs in traditional Chinese medicine (TCM) formulae for fighting COVID-19 infections, due to the presence of isoflavonoids and astragalosides associated with antiviral and immune-enhancing activities. For the first time, the exposure of A. membranaceus hairy root cultures (AMHRCs) to different colors of LED lights i.e., red, green, blue, red/green/blue (1/1/1, RGB), and white, was conducted to promote the root growth and accumulation of isoflavonoids and astragalosides. LED light treatment regardless of colors was found beneficial for root growth, which might be a result of the formation of more root hairs upon light stimulation. Blue LED light was found most effective for enhancing phytochemical accumulation. Results showed that the productivity of root biomass in blue-light grown AMHRCs with an initial inoculum size of 0.6% for 55 days was 1.40-fold higher than that in dark (control), and yields of high-value isoflavonoids and astragalosides including calycosin, formononetin, astragaloside IV, and astragaloside I increased by 3.17-fold, 2.66-fold, 1.78-fold, and 1.52-fold relative to control, respectively. Moreover, the photooxidative stress together with transcriptional activation of biosynthesis genes might contribute to the enhanced accumulation of isoflavonoids and astragalosides in blue-light grown AMHRCs. Overall, this work offered a feasible approach for obtaining higher yields of root biomass and medicinally important compounds in AMHRCs via the simple supplementation of blue LED light, which made blue-light grown AMHRCs industrially attractive as plant factory in controlled growing systems.

Strawberry is a nutritious food that is susceptible to mechanical injury and microbiological infection. Traditional coatings for strawberry packaging provide resistance against microbial infection but not against mechanical damage. In this study, a soft and elastic cellulose sponge modified with silver nanoparticles (AgNPs@CS-1:1) was prepared as strawberry packaging material, and it provided effective protection against mechanical damage. In addition, after 1000 cyclic compression, AgNPs@CS-1:1 presented only 16.80% unrecoverable deformation and still had elasticity, suggesting its fatigue resistance and durable protection for strawberry against damage caused by repeated vibrations during transportation. In addition, AgNPs@CS-1:1 had good antibacterial (E. coli and S. aureus) and antifungal (Rhizopus stolonifer) abilities. The storage time of strawberries packaged by AgNPs@CS-1:1 was extended to 12 days without microbial invasion. Thus, AgNPs@CS-1:1 provided dual protection at the physical and microbial levels. This study proposes a new method for the preservation of strawberries based on the utilization of cellulose.

Astragaloside IV/β-cyclodextrin inclusion complex(AsIV/β-CD IC) was successfully prepared by low-temperature sedimentation technology. Under the preparation conditions inclusion temperature 60℃, inclusion time 3h, core-wall material molar ratio 1:1, and inclusion rate 750 rpm, ­­­the optimal encapsulation efficiencyreached 81.63%. It was characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffractometer (XRD) and thermogravimetric (TGA). Physicochemical characterization of the AsIV/β-CD IC indicated that the complexation of AsIV/β-CD was successful, and the thermal stability of AsIV wasimproved clearly. Through granulation technology, astragalus saponin granules (ASG) with uniform size from AsIV/β-CD IC were obtained. ASG possessed significant water-solubility and storage stability beside fine taste. In addition, ASG demonstrated beneficial bioactivity in antioxidant and antibacterial functions. Antioxidative stress detection showed that ASG could inhibit the increase of MDA content and the decrease of SOD content in cells caused by LPS. In addition, in vitro antibacterial experiments of ASG showed that ASG has obvious antibacterial activity against 6 strains, especially the MIC value of Escherichia coli and Bacillus subtilis reached 12.5 mg/mL.

Germinated pigeon pea seeds (GPPSs) are good dietary supplements with satisfactory nutritional and medicinal values. Chlorogenic acid (CGA) is a polyphenol compound with health functions. In order to enrich and purify CGA from mulberry leaf extracts efficiently, three-dimensional porous cellulose-based microspheres modified with ionic liquid functional groups (CMs-ILs) was developed. In terms of adsorption properties, compared with three different commercial macroporous resins (D101, NKA-9, and AB-8), CMs-ILs exhibited a greater adsorption capacity (103.6 mg/g) and a faster adsorption rate to reach equilibrium (40 min). The density functional theory (DFT) calculations were implemented to study the adsorption mechanism. It was showed that hydrogen bonding and π-π stacking were the main interactions between CGA and ILs. The results of a contact angle test and water drop diffusion experiments indicated that the super hydrophilic surface and porous structures of CMs-ILs accelerating the mass transfer rate, leading to a fast adsorption rate. Moreover, the relative adsorption capacity was still over 85% of the original after reusing for 5 times. Therefore, CMs-ILs would be a promising material for enrichment of CGA from natural plants.

Flavonoids are important metabolites of pigeon pea in relation to its stress resistance. However, the molecular basis and regulatory mechanisms of flavonoids in pigeon pea remain unclear. Methyl jasmonate (MeJA) is a signaling molecule associated with biosynthesis of flavonoids. In this study, after exogenous treatment of 10 mg/L MeJA, infection of pathogenic fungi to pigeon pea was alleviated and the content of flavonoids was increased. Results of gene expression and metabolic changes that were respectively analyzed by transcriptome sequencing and high performance liquid chromatography (HPLC) showed that (1) concentrations of various flavonoids, such as genistein, apigenin, vitexin and biochanin A were significantly up-regulated; (2) 13675 differentially expressed genes were produced, mainly enriched in signal transduction and isoflavone biosynthesis pathways: (3) the expression levels of key synthase genes (CcI2′H, CcHIDH, Cc7-IOMT) in the flavonoid biosynthesis pathway were significantly up-regulated; (4) Overexpression of CcbHLH35 significantly induced upregulation of flavonoid synthase genes and accumulation of genistein, vitexin and apigenin. Our findings reveals the pivotal roles of MeJA in synthesis and functioning of flavonoids in pigeon pea, which provide a basis for further studies on flavonoid-mediated defense responses.

Numerous plant secondary metabolites have remarkable impacts on both food supplements and pharmaceuticals for human health improvement. However, higher plants can only generate small amounts of these chemicals with specific temporal and spatial arrangements, which are unable to satisfy the expanding market demands. Cyanobacteria can directly utilize CO2, light energy, and inorganic nutrients to synthesize versatile plant-specific photosynthetic intermediates and organic compounds in large-scale photobioreactors with outstanding economic merit. Thus, they have been rapidly developed as a "green" chassis for the synthesis of bioproducts. Flavonoids, chemical compounds based on aromatic amino acids, are considered to be indispensable components in a variety of nutraceutical, pharmaceutical, and cosmetic applications. In contrast to heterotrophic metabolic engineering pioneers, such as yeast and Escherichia coli, information about the biosynthesis flavonoids and their derivatives is less comprehensive than that of their photosynthetic counterparts. Here, we review both benefits and challenges to promote cyanobacterial cell factories for flavonoid biosynthesis. With increasing concerns about global environmental issues and food security, we are confident that energy self-supporting cyanobacteria will attract increasing attention for the generation of different kinds of bioproducts. We hope that the work presented here will serve as an index and encourage more scientists to join in the relevant research area.

BACKGROUND A palatal radicular groove is an unusual developmental deformity of the tooth, which may serve as a channel linking the periodontal and periapical inflammation, and yet no literature could be obtained analyzing microbiota within the palatal radicular grooves. CASE SUMMARY Four patients diagnosed with palatal radicular groove and concomitant periodontal-endodontic deformity in permanent maxillary lateral incisors were enrolled in this work. Twelve bacterial samples from 4 patients were collected from different parts of the palatal radicular groove during intentional replantation surgery. Illumina sequencing was performed to analyze the taxonomical composition and microbiome structure inside the palatal grooves, and 1162 operational taxonomic units were obtained. The phyla of Firmicutes and Proteobacteria predominated in most of the samples. An unknown genus from the Bacillaceae family, Lactococcus, and Porphyromonas were the most abundant genera identified. There was no difference in the microbiota richness and diversity in three sections of the groove. CONCLUSION The unique ecological niches inside the palatal grooves harbored bacterial communities that shared some component features of both the endodontic and periodontal infections. The existence of palatal groove may play an interaction bridge between the root apex and tooth cervix and thus impair the outcome of traditional therapeutic methods such as root canal treatment and periodontal management.

Background Inonotus obliquus, a famous dark brown white-rot fungus on birch trees, has remarkable anti-cancer activity due to its special triterpenpoid components. The aims of this study were to develop an efficient and integrated strategy for preparative separation of triterpenoids from Inonotus obliquus and to provide ideas for the preparation of bioactive ingredients in other complex materials. Methods In the present study, an integrated technology of negative pressure cavitation extraction (NPCE) and high-speed counter-current chromatography (HSCCC) separation of four main triterpenoids in Inonotus obliquus was developed for the first time. Response surface methodology was used to optimize the negative pressure cavitation extraction parameters of triterpenoids from Inonotus obliquus. For the rapid and effective preparative separation of triterpenoids, the HSCCC method with Hex-ACN (1:1; v/v) system was established. Significant Findings The extraction yield of total triterpenoids reached 17.41 mg/g under the optimal NPCE extraction conditions. The specific four triterpenoids including 3β-hydroxy-lanosta-8, 24-diene-21-al (0.84 mg/g), Inoterpene F (1.09 mg/g), Inotodiol (2.93 mg/g), Lanosterol (1.42 mg/g) were obtained by one step separation with the purities of more than 95%. This is the first report on the preparative separation of triterpenoids from Inonotus obliquus by NPCE-HSCCC.

Cellulose sponges with compressibility and resilience are an ideal packaging material for fruits with fragile skin. Here, a soft and elastic all-cellulose sponge (CS) with a hierarchical cellular structure was fabricated, where the long molecular chain cellulose constructed major pores, the cellulose at nanoscale acted as an elastic nanofiller to fill the gaps of long molecular chain cellulose fibers and constructed minor pores. With these two kinds of pores, this structure can absorb strain hierarchically. The sponge can protect fruits from mechanical damage when dropped or repeated vibration. Furthermore, the CS modified with chlorogenic acid (C-CGAS) had excellent antibacterial and antifungal abilities. Therefore, C-CGAS could extend the storage time of strawberries to 18 days without any microbial invasion, which is the longest storage time reported thus far. This study provides a new idea for the preparation of polymer sponges and a new design for the development of antimicrobial packaging materials.

A microcrystalline cellulose-based temperature sensitivity paclitaxel molecular imprinted hydrogel (MCC-TSMIHs-PTX) was successfully prepared by temperature-sensitive monomer N-isopropylacrylamide, functional monomer 4-vinylpyridine, cross-linking agent N, N'-methylenebisacrylamide and microcrystalline cellulose. They showed imprinting effective responses to the temperature changes. The results of adsorption kinetics, adsorption equilibrium, thermodynamics, selectivity and reusability showed the successful formation of a grafting thermosensitivity hydrogel with higher adsorption capacity and specific recognition. When the temperature reached 308 K, imprinting effect of hydrogel cavities would be most effective and conducive to capture template molecules. When the temperature reached 288 K, the lowest imprinting effect would facilitate the desorption of PTX. Finally, the MCC-TSMIHs-PTX was applied to enrich the paclitaxel in Taxus × media extracts samples, the relative contents of PTX in the samples were increased greatly from 7.23% to 78.32%, indicating the MCC-TSMIHs-PTX was a stable adsorption capacity for efficient separation and enrichment of PTX in Taxus × media extracts.

Calcineurin B-like (CBL)-interacting protein kinases (CIPKs) play a central role in Ca2+ signalling and promote drought tolerance in plants. The CIPK gene family in pigeon pea (Cajanus cajan L.), a major food crop affected by drought, has not previously been characterised. Here, we identified 28 CIPK genes in the pigeon pea genome. Five CcCIPK genes were strongly upregulated in roots upon drought treatment and were selected for further characterisation. Overexpression of CcCIPK13 and CcCIPK14 increased survival rates by two- to three-fold relative to controls after 14 days of drought. Furthermore, the three major flavonoids, genistin, genistein and apigenin, were significantly upregulated in the same transgenic plants. Using CcCIPK14 as bait, we performed a yeast two-hybrid screen and identified six interactors, including CcCBL1. CcCIPK14 exhibited autophosphorylation and phosphorylation of CcCBL1 in vitro. CcCBL1-overexpressed plants displayed higher survival rates upon drought stress as well as higher expression of flavonoid biosynthetic genes and flavonoid content. CcCIPK14-overexpressed plants in which CcCBL1 transcript levels were reduced by RNA interference had lower survival rates, which indicated CcCBL1 in the same pathway as CcCIPK14. Together, our results demonstrate a role for the CcCIPK14-CcCBL1 complex in drought stress tolerance through the regulation of flavonoid biosynthesis in pigeon pea.

A novel ternary deep eutectic solvent (DES), consisted of choline chloride, oxalic acid and ethylene glycol, was developed as a green, low-cost and recyclable pretreatment system for multi-stage utilization of Eucommia ulmoides seed shells. Under optimum conditions, 79.7 % hemicellulose and 65.6 % lignin were quickly removed while 84.0 % cellulose was retained. After DES pretreatment, the yield and purity of gutta-percha achieved 85.1 mg/g and 96.2 %, which increased 1.4 and 1.8 folds higher than that of un-treatment ones. Meanwhile, 69.1 % enzymatic digestibility of cellulose was obtained, that was 2.3 folds higher than that of raw substrates. Moreover, 53.6 % low-condensation lignin with aromatic structures and valuable aryl-ether linkages was well collected. Importantly, the DES that has been recycled five runs can still remove 73.9 % hemicellulose and 58.0 % lignin. Overall, the DES was determined to efficiently promote the separation and conversion of high-quality gutta-percha, value-added lignin and high-yield glucose from Eucommia ulmoides seed shells.

Due to the growing global population, reduction in arable land and effects of climate change, incongruity between food supply and demand has become increasingly severe. Nowadays, with awareness of the elementary nutrients required for human growth, increasing attention is being paid to the health and medical functions of food. Along with increased food production achieved by modern agricultural techniques, underutilised functional foods are an important strategy for solving food security problems and maintaining the nutritional quality of the human diet. Rosa roxburghii Tratt (RRT) is a natural fruit that contains unique functional and nutritional constituents, which are characterised by a high anti-oxidant potential. This review summarises the biological characteristics, chemical composition, health-promoting properties and development status of RRT products to inspire investigations on the use of RRT fruit as a functional food, dietary supplement and pharmaceutical additive. The nutrients and functional ingredients of RRT fruit are described in detail to provide more reference information for nutritionists and pharmacists.

Exploring the fragmentation and loss of soil aggregates is of great significance for revealing the mechanism of water erosion. To determine the effect of raindrop size on the distribution of soil aggregate fractions induced by shallow runoff, a variable-needle raindrop-generating device and a slope runoff scouring device (length 90 cm × width 10 cm × height 10 cm) were employed in the study. Five raindrop sizes (2.67 to 4.05 mm) and three runoff depths (3.82, 4.86, and 7.98 mm) were selected to conduct the runoff alone (F) test and the rainfall + runoff (RF) test. The results showed that for the RF test under the raindrop size >3.05 mm, the soil loss rate was the highest at runoff depth 4.86 mm and was 104.30 to 421.52, 69.86 to 169.81, and 26.80 to 89.28 times higher than that of the F test under the three runoff depths, respectively. The rainfall disturbance coefficient (the ratio of soil loss caused by the RF test to that caused by the F test) was closely related to the raindrop size and runoff depth and had a significant power function relationship with both. Under the F test condition, the soil loss rate increased with the decreasing aggregate fraction distribution, and the loss rate of each aggregate fraction increased with increasing runoff depth. Similarly, for each RF test, the soil loss rate also increased with the decrease in the aggregate fraction distribution. The maximum soil loss rate was observed in the 1 mm aggregate fraction by shallow runoff. However, rainfall mainly enhanced the transportation of 1 to 0.053 mm aggregate fractions as the raindrop size increased to greater than 3.05 mm.

In the present study, an integrated process for preparation of anthocyanins components from blue honeysuckle fruits using deep eutectic solvent-based negative pressure cavitation extraction (DES-NPCE) was explored rationally. Single factor optimization combined with Box-Behnken Design (BBD) were applied to confirm the influential parameters. The maximum extraction yields of total anthocyanins and main constituent cyanidin-3-glucoside (C3G) reached 6.601 mg/g and 4.839 mg/g, which were 1.197–1.565 folds of other conventional methods. Moreover, the anthocyanins products exhibited higher stability from DES-NPCE extraction than that of conventional extraction solvents, which was conducive to the preservation and utilization of anthocyanins. After enrichment with macroporous resin D101, the contents of total anthocyanins and C3G reached 25.72% and 19.05%, and the recovery yields were 81.35% and 82.24%, respectively. Furthermore, the anthocyanins products exhibited more powerful chemical and cellular antioxidant activities, which were similar or stronger than that of positive control Vc or BHT. These results revealed that the proposed process was definitely an efficient and sustainable strategy for the stabilization preparation of thermo sensitive biologically active natural products. The developed method has also proved to have sufficient potential for application in food, nutrition and pharmaceutical industries.

Various kinds of bioactive compounds contribute to versatile health-promoting properties of Eucommia ulmoides Oliver (E. ulmoides). In present study, we developed a UPLC-QqQ-MS/MS method for simultaneous quantification of fourteen characteristic active compounds, including 3 lignans, 4 iridoids, 3 flavonoids and 4 phenolics in E. ulmoides and its tea product for the first time. The running time of the method is 6.5 min. It has good linearity, sensitivity, precision, accuracy, and stability. Using this high-throughput method, the distributions of fourteen characteristic active compounds in E. ulmoides and its tea product were clarified. Also, it was found that E. ulmoides tea exhibited superiority in contents of chlorogenic acid as compared with natural resources. Overall, the study provided a rapid, reliable, and efficient analysis method, which could be applied for the quality evaluation of E. ulmoides natural resources and their relative products in the field of food and medicine.

Phenolic compounds in pigeon pea possess various biological properties beneficial to human health. In this study, pigeon pea hairy root cultures (PPHRCs) were developed as an effective in vitro platform for the production of phenolic compounds. A high-productive hairy root line was screened and characterized, and its culture conditions were optimized in terms of biomass productivity and phenolic yield. The comparative profiling of 10 phenolic compounds in PPHRCs and pigeon pea natural resources (seeds, leaves, and roots) was achieved by ultra-high-performance liquid chromatography-tandem mass spectrometry analysis. The total phenolic yield in PPHRCs (3278.44 μg/g) was much higher than those in seeds (68.86 μg/g) and roots (846.03 μg/g), and comparable to leaves (3379.49 μg/g). Notably, PPHRCs exhibited superiority in the yield of the most important health-promoting compound cajaninstilbene acid (2996.23 μg/g) against natural resources (4.42-2293.31 μg/g). Overall, PPHRCs could serve as promising potential alternative sources for the production of phenolic compounds with nutraceutical/medicinal values.

Antioxidant additives are commonly used to prevent biodiesel oxidation. Most antioxidant studies have focused on the effects of chemosynthetic antioxidants, but there has been limited research on natural antioxidants. In this study, a cheap and readily available natural antioxidant, Pyrola extract, was applied for biodiesel protection. An immersion test that exposes biodiesel to copper chips was used to compare the antioxidant activities between Pyrola extract and the commercial antioxidants, pyrogallol and BHT. Pyrola extract performs much better than pyrogallol (PY) and BHT on all of the quality characteristics of biodiesel, including acid value, iodine value, peroxide value, and corrosion rate of the coppers. Analysis by gas chromatography also indicates that Pyrola extract helps to maintain the original components of the biodiesel. Due to the simple extraction process and low cost raw materials, Pyrola extract may be a potential antioxidant for protecting biodiesel.

In the present study, modified polyporous magnetic cellulose beads (PMCBs) were synthesized by several organic amines (EDA, DETA, TETA and AST) grafting PMCBs and then utilized as a new kind of support to immobilize Candida antarctica lipase B (CALB) for biodiesel production. The immobilized lipase catalysts based on PMCBs were characterized by Fourier transform infrared and scanning electron microscopy. The results showed that PMCBs-DETA was a more suitable immobilization support and the catalytic activity of immobilized lipases could reach 1.0 U/mg. Then the synthesized catalyst of CALB-PMCBs-DETA was used to produce biodiesel from Yellow horn seed oil via transesterification reaction. A high biodiesel yield (92.3%) was obtained under the optimal reaction conditions and the main compositions of the prepared biodiesel were tested by GC-MS. Moreover, the catalyst of CALB-PMCBs-DETA could be removed easily by a magnet and recycled at least 5 times. As a new kind of support, the modified PMCBs could be a promising alternative for the carriers of immobilizing enzymes used for the green and efficient production of biodiesel.

Growth-regulating factor (GRF) is a transcription factor unique to plants that plays a crucial role in the growth, development and stress adaptation of plants. However, information on the GRFs related to salt stress in Populus davidiana × P. bolleana is lacking. In this study, we characterized the activity of PdbGRF1 in transgenic Populus davidiana × P. bolleana under salt stress. qRTPCR analyses showed that PdbGRF1 was highly expressed in young leaves and that the pattern of PdbGRF1 expression was significantly changed at most time points under salt stress, which suggests that PdbGRF1 expression may be related to the salt stress response. This study explored an integrated strategy for extraction and preparative separation of four anthocyanins from Ribes nigrum L. using DES-based microwave assisted extraction and flash chromatography. The extraction yield of anthocyanins reached 2.03 mg/g after optimization. An efficient preparative separation process of four main anthocyanins by macroporous resin and flash chromatography was established afterwards. The yields of D3G, D3R, C3G and C3R reached 0.32 mg/g, 0.47 mg/g, 0.18 mg/g and 0.43 mg/g with the purities of more than 95%, respectively. The developed procedure represents an efficient and convenient alternative for the systematic utilization of natural products from plant materials.

Fig (Ficus carica L.) leaves are produced each year and often disposed, resulting in a waste of resources. Fig waste leaves are rich in flavonoids, which have strong antioxidant activity; however, the variety and chemical structure of antioxidants in fig leaves have not been reported in detail. To take full advantage of fig waste leaves, antioxidant capacity of different extracts (petroleum ether, ethyl acetate, and water) was evaluated by 1, 1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic) acid (ABTS), and ferric-ion-reducing antioxidant power (FRAP) methods. The results showed that flavonoids in ethyl acetate extraction had the highest content (83.92 ± 0.01 mg/g), maximum DPPH scavenging activity (IC50 0.54 mg/mL), highest ABTS scavenging rate (80.28%), and FRAP (3.46 mmol/g). Furthermore, an HPLC-QTOF-MS/MS-DPPH method was developed to identify 11 flavonoids in fig waste leaves. This rapid and efficient method can not only be used for screening the antioxidant components in fig waste leaves, but also can be combined with mass spectrometry to identify the compounds with antioxidant capacity. There are three flavonoids with significant antioxidant capacity, which are 3-O-(rhamnopyranosyl-glucopyranosyl)-7-O-(glucopyranosyl)-quercetin, isoschaftoside, and rutin. The results confirmed that fig waste leaves contain a variety of antioxidant components, which contributed to increase the value of fig waste leaves as antioxidants.

UV-B radiation is an ideal elicitation strategy for promoting phytochemical accumulation in plant in vitro cultures, associated with various advantages of easy manipulation, cost-effectiveness, no residue, and instantaneous termination. For the first time, UV-B radiation was used to enhance the production of bioactive phenolic compounds (flavonoids and stilbenes) in pigeon pea hairy root cultures (PPHRCs). The total yield of eight flavonoids (414.95 ± 50.68 μg/g DW) in 42-day-old PPHRCs exposed to 4 h of UV-B radiation increased by 1.49-fold as against control, whereas the yield of cajaninstilbene acid (6566.01 ± 702.14 μg/g DW) in PPHRCs undergoing 10 h of UV-B radiation significantly increased by 2.31-fold over control. UV-B radiation was found to induce the oxidative stress in PPHRCs and cause the tissue damage to hairy roots, which improved the levels of endogenous salicylic acid thus triggering the expression of genes involved in phenylpropanoid biosynthesis pathway. And, a regulation competition in metabolic flow dominated by CHS and STS was responsible for the difference in accumulation trends of flavonoids and cajaninstilbene acid. Results of this study not only provide a feasible and simple UV-B supplementation strategy for the enhanced production of bioactive phenolic compounds (especially the high-value cajaninstilbene acid) in PPHRCs, but also contributed to the understanding of photobiological responses related to secondary metabolism.
